Crystal proteins from Bacillus thuringiensis serovar. medellin   总被引:1,自引:0,他引:1  
Colombian strains 163-131 and 24-726 of the Bacillus thuringiensis serovar. medellin (Btmed), serotype H-30, are very toxic to mosquito larvae. Strain 24-726 was serologically and biochemically characterized. It is almost identical to the reference-strain 163-131. The parasporal inclusion of Btmed strain 163-131 was analysed by electron microscopy. The crystal protein matrix was very similar to that observed in B. thuringiensis serovar. israelensis (Bti). Aedes aegypti, Anopheles albimanus and Culex quinquefasciatus larvae were exposed to 500× the half-lethal concentration (LC50) of Btmed strains, Bti strain 1884 and B. thuringiensis serovar. morrisoni (Btm) strain PG-14. Mortality of Aedes aegypti occurred within approx. 60 min with the four strains, whereas C. quinquefasciatus mortality was three times slower with Btmed than with strains 1884 and PG14. The onset mortality of Anopheles albimanus starts when other species are already dead. The thermolabilities of the mosquitocidal activities of the crystal proteins were tested by incubation of cultures of 20 min at various temperatures. Btmed lost all mosquitocidal activity at 73°C, and 1884 and PG-14 at 79°C. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis analysis of the crystals purified from strain 163-131 shows polypeptides at 100 kDa, multiple bands at 80, 75, 70, 67, and 65 kDa, and two doubles at 40–41 and 28–30 kDa. Immunodetection with antibodies raised against Bti toxins shows cross-reaction between the 30-kDa and to a lesser extent the 28-kDa polypeptides of Btmed crystals and Cyt A of Bti. Bacillus thuringiensis serovar israelensis , an entomopathogen for mosquito larvae, was demonstrated to be lysogenized by temperate phage SU-11 whose genome was located extrachromosomally in the cell. The prophage SU-11 was cured at high frequency from the parental strain by continuous sub-culture at high temperature, but the ability to produce δ-endotoxin remained in the prophage cured strain. Moreover, phage induction was found to occur after mating of serovar israelensis with its prophage cured strain, as well as with B. thuringiensis serovar thuringiensis , B. cereus and B. subtilis .  相似文献

Bacillus thuringiensis is a gram-positive, spore-forming bacterium that forms parasporal crystals at the onset of the sporulation phase of its growth. Here, we report the complete genome sequence of B. thuringiensis serovar finitimus strain YBT-020, whose parasporal crystals consist of Cry26Aa and Cry28Aa crystal proteins and are located between the exosporium and the spore coat and remain adhering to the spore after sporulation.  相似文献

The spotted wing drosophila (SWD), Drosophila suzukii, is an invasive species to the USA, and Europe and biological control methods are urgently sought for. In this study, the potential of commercial microbial control products based on the Dipteran‐specific B. thuringiensis serovar. israelensis (B.t.i.) were evaluated in laboratory experiments. These products were tested on SWD larvae and adults but neither one showed more than 10% mortality. A repellent effect of the products to SWD adults was also ruled out. We conclude that B.t.i. products are not suitable for SWD control.  相似文献

In order to detect and identify the most toxic Bacillus thuringiensis strains against pests, we isolated a B. thuringiensis strain (Bn1) from Balaninus nucum (Coleoptera: Curculionidae), the most damaging hazelnut pest. Bn1 was characterized via morphological, biochemical, and molecular techniques. The isolate was serotyped, and the results showed that Bn1 was the B. thuringiensis serovar, kurstaki (H3abc). The scanning electron microscopy indicated that Bn1 has crystals with cubic and bipyramidal shapes. The Polymerase Chain Reactions (PCRs) revealed the presence of the cry1 and cry2 genes. The presence of Cry1 and Cry2 proteins in the Bn1 isolate was confirmed via SDS-PAGE, at approximately 130 kDa and 65 kDa, respectively. The bioassays conducted to determine the insecticidal activity of the Bn1 isolate were conducted with four distinct insects, using spore-crystal mixtures. We noted that Bn1 has higher toxicity as compared with the standard B. thuringiensis subsp. kurstaki (HD-1). The highest observed mortality was 90% against Malacosoma neustria and Lymantria dispar larvae. Our results show that the B. thuringiensis isolate (Bn1) may prove valuable as a significant microbial control agent against lepidopteran pests.  相似文献

The parasporal inclusion proteins of the type strain of Bacillus thuringiensis serovar higo (H44), that have moderate mosquitocidal activity, were characterized. The purified parasporal inclusions, spherical in shape, were examined for activity against the two mosquito species, Culex pipiens molestus and Anopheles stephensi and the moth-fly, Telmatoscopus albipunctatus . The LC50 values of the inclusion for the two mosquitoes were 3·41 and 0·15 μg ml−1, respectively. No mortality was shown for T. albipunctatus larvae by the inclusions at concentrations up to 1 mg ml−1. Solubilized parasporal inclusions exhibited no haemolytic activity against sheep erythrocytes. Parasporal inclusions consisted of eight proteins with molecular masses of 98, 91, 71, 63, 59, 50, 44 and 27 kDa. Of these, the 50 and 44 kDa proteins were the major components. Analysis with immunoblotting revealed that, among several inclusion proteins of B. thuringiensis serovar israelensis, only two proteins of 130 kDa and 110 kDa reacted weakly with antibodies against higo proteins. N-terminal amino acid sequences of the 98, 91, and 71 kDa proteins showed 85–100% identity to those of the two established Cry protein classes, Cry4A and Cry10A.  相似文献

AIMS: To characterize the mosquitocidal activity of parasporal inclusions of the Bacillus thuringiensis serovar sotto strain 96-OK-85-24, for comparison with two well-characterized mosquitocidal strains. METHODS AND RESULTS: The strain 96-OK-85-24 significantly differed from the existing mosquitocidal B. thuringiensis strains in: (1) lacking the larvicidal activity against Culex pipiens molestus and haemolytic activity, and (2) SDS-PAGE profiles, immunological properties and N-terminal amino acid sequences of parasporal inclusion proteins. CONCLUSIONS: It is clear from the results that the strain 96-OK-85-24 synthesizes a novel mosquitocidal Cry protein with a unique toxicity spectrum. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first report of the occurrence of a mosquitocidal B. thuringiensis strain with an unusual toxicity spectrum, lacking the activity against the culicine mosquito.  相似文献

The LC50 value of alkali-solubilized parasporal inclusion proteins of a Diptera-specific strain, belonging to Bacillus thuringiensis serovar canadensis, was 2.4 μg/ml for larvae of the mosquito, Aedes aegypti. A significant loss in larvicidal activity occurred when solubilized inclusion proteins were treated with A. aegypti larval gut extract, silkworm (Bombyx mori) larval gut juice, and the proteinase K. Approximately 90% of the larvicidal activity was destroyed upon treatment with proteases in 30 min. The parasporal inclusion was composed of major proteins of 65, 53, and 28 kDa and some other minor proteins. Proteolysis profiles showed that the 65-kDa major protein is highly sensitive to proteases. Purification experiments with DEAE-Toyopearl column chromatography revealed that the 65-kDa protein is responsible for the mosquitocidal activity of this strain. Endogenous chitinase plays a positive role in the pathogenicity of Bacillus thuringiensis to insect pests. The chitinase gene was cloned from B. thuringiensis serovar alesti strain HD-16, and the deduced 676 amino acid sequence showed a high degree of similarity with other Bacillus chitinases. Additionally, the deduced amino acid sequence showed that the protein contained an amino terminus signal peptide and consisted of a catalytic domain, a fibronectin type III domain and a chitin-binding domain. All three domains showed conserved sequences when compared to other bacterial chitinase or cellulase sequences.  相似文献

The type strain Bacillus thuringiensis var. bolivia (serotype H63), isolated from the Bolivian high valleys, has been characterized at different levels. Its parasporal crystal has an unusual shape and it is composed of a protein of 155 kDa which shows two bands of 75 and 80 kDa after activation. Analysis by PCR shows the presence of cry1 genes, and amplification with specific primers gave products for cry1 E, cry1 D, cry4 A and cry4 B with sizes different to those expected. Immunoblotting tests showed positive reaction for Cry1 E, Cry3 A, Cry4 A and Cry11 A crystal proteins. The plasmid pattern revealed two large and two small plasmids. Toxicity tests were performed against 14 insects and a slight toxicity was found against Plutella xylotella and Trichoplusia ni.  相似文献

Mosquito larvicides like Bacillus sphaericus and Bacillus thuringiensis serovar. israelensis have been widely and effectively used in mosquito control programs, but the industrial production of these bacilli is expensive. Here we have attempted to develop three cost-effective media, based on cheap sources, potato, common sugar and bengalgram. Growth and production of the insecticidal proteins from these bacteria were satisfactory. Bioassay studies with different mosquito larvae showed considerable toxicity. Therefore the investigation suggests that potato-based culture media are more economical for the industrial production of B. sphaericus and B. thuringiensis serovar. israelensis.  相似文献

A cry2A-type gene, designated as cry2(SKW), was cloned from Bacillus thuringiensis serovar sotto SKW01-10.2-06, and some unique features of the gene were revealed. The cry2(SKW) gene encoded a polypeptide of 635 residues with a predicted molecular mass of 71,137 Da. Cry2(SKW) had 95.4% identity with Cry2Aa in amino acid sequence and was two residues longer than Cry2Aa. Two open reading frames (ORFs), designated as orf1 and orf2, were present upstream of the cry2(SKW) and showed high homology with the corresponding ORFs in the cry2Aa operon. The Orf2 from SKW01-10.2-06 contained a region of repeated sequences. However, unlike Cry2Aa, Cry2(SKW) formed the cuboidal crystalline inclusions when the cry2(SKW) gene was expressed in an acrystalliferous B. thuringiensis strain in the absence of the upstream ORFs. Furthermore, Cry2(SKW) was less toxic to a lepidopteran species, Bombyx mori, than Cry2Aa in spite of high homology between the two proteins. A wettable powder formulation (ABG-6108) of Bacillus thuringiensis serovar. israelensis was tested against chironomid larvae in experimental ponds at 1, 2, 3, 4 and 10 kg/ha (0.25–2.5 ppm), and in a golf pond at 3 kg/ha (0.5 ppm). Significant reductions (18–88%) in populations of Chrinomini and Tanytarsini experimental ponds, and 27–65% reduction of midges in the golf pond occurred during the 4-week evaluation period. The highest dose in experimental ponds had no adverse effects on rotifers, Cyclops spp., Daphnia sp., ostracods, Baetis sp., corixids and notonectids, and Coleoptera. The nontarget invertebrates in the golf pond also remained unaffected.  相似文献

The δ-endo toxin proteins from Bacillus thuringiensis which kill the larvae of various scarabaeid beetles such as Anomala cuprea, A. rufocuprea and Popillia japonica were purified by DEAE ion exchange chromatography. A protein with a molecular size of 130 kDa was purified. During the purification a minor peak was also detected which was estimated to be 67 kDa by SDS-PAGE. Both 130 and 67 kDa proteins showed larvicidal activity against A. cuprea. The lethal concentration of the 130 kDa protein which killed 50% of the larvae tested (LC50) against A. cuprea was 2 μg g1 compost. A comparison by SDS-PAGE of the V8 protease digestion pattern of the 130 and 67 kDa larvicidal proteins showed that proteolytic resistant core peptides of approximately 60 kDa molecular size were resulted. The N -terminus amino acid sequence of the 130 and 67 kDa proteins was determined to be NH2-XXPNNQNEYEIIDAL and NH2-XSRNPGTFI, respectively, which is not identical to the sequence of CryIA, CryIB, CryIC and CryIII proteins.  相似文献

A spore-forming bacterium isolated from the soil of Japan was assigned to Bacillus thuringiensis serovar japonensis (flagellar antigen 23). Parasporal inclusions of this isolate were spherical to ovoid in shape and exhibited high larvicidal activity against coleopterous scarabaeid beetles, the cupreous chafer. Anomala cuprea , the soybean beetle, Anomala rufocuprea , and the Japanese beetle. Popillia japonica. No toxicity was shown by this isolate against larvae of Lepidoptera, Diptera, and Orthoptera, and adults of a chrysomelid coleopteran.  相似文献

Two novel crystal protein genes from a highly mosquitocidal Bacillus thuringiensis serovar medellin strain were cloned and sequenced. The corresponding proteins, Cry29A and Cry30A, were nontoxic when tested individually against the mosquito species bioassayed (Aedes aegypti, Culex pipiens and Anopheles stephensi). However, Cry29A synergized the toxicity of Cry11Bb against Aedes aegypti by a four-fold factor.  相似文献
