Injection of caffeine into adult male Ephestia kühniella induced a reduction in the amount of neurosecretory material in the resorcin fuchsin stainable (A-) cells of the pars intercerebralis. Both hemel (hexamethyl melamine) and tepa (tris (1-aziridinyl) phosphine oxide) treatments resulted in an increase in stainable neurosecretory products in the cytoplasm of the A-cells. The increase after hemel treatment was largely a result of increased synthesis of neurosecretory material as indicated by increased nuclear volume and nuclear-cytoplasmic ratio. No changes were induced by hemel in the corpora cardiaca and allata of the adult male E.kühniella. This disruption in neuroendocrine function is believed to be involved in the observed mating aberrations (permanent copulation and sterile matings) by adult males after hemel treatment.  相似文献

Summary Membrane-limited electron-dense inclusions designated as elementary neurosecretory granules have a characteristic distribution in cerebropleural ganglia of the nudibranch snail Tritonia diomedia. They occur in the neuropile and also in individual nerve fibres, connectives and commissures. These granules have been found neither in perikarya of nerve cells nor in proximal segments of their processes.Specific fluorescence obtained in Tritonia preparations with Sterba's pseudoisocyanin method for neurosecretory products has the same pattern of location.The distribution of stainable material in preparations prepared with ordinary neurosecretory procedures (chrome haematoxylin-phloxin after Gomori-Bargmann and paraldehydefuchsin after Gomori-Gabe) is similar to that described by different authors in other gastropods, but strongly differs from the locationof elementary neurosecretory granules and of pseudoisocyanin-positive material. Summary Neuronal pathways in the retrocerebral complex and thoracico-abdominal ganglionic mass of the blowflyCalliphora vomitoria have been identified immunocytochemically with antisera against the extended-enkephalins, Met-enkephalin-Arg6-Phe7 (Met-7) and Met-enkephalin-Arg6-Gly7-Leu8 (Met-8). Neurons of the hypocerebral ganglion, immunoreactive to Met-8, have axons in the crop duct nerve and terminals in muscles of the crop and its duct. Certain neurons of the hypocerebral ganglion are also immunoreactive to Met-7, and axons from these cells innervate the heart. Met-8 immunoreactive nerve terminals invest the cells of the corpus allatum. The source of this material is believed to ve a single pair of lateral neurosecretory cells in the brain. There is no Met-7 immunoreactive material in the corpus allatum. In the corpus cardiacum neither Met-7 nor Met-8 immunoreactivity is present in the cells. However, in the neuropil of the gland certain fibres, with their origins elsewhere, do contain Met-8 immunoreactivity. The most prominent neurons in the thoracic ganglion are the Met-7 immunoreactive ventral thoracic neurosecretory cells, axons from which project to neurohaemal areas in the dorsal neural sheath and also, via the ventral connective, to the brain. Co-localisation studies show that the perikarya of these cells are immunoreactive to antisera raised against several vertebrate-type peptides, such as Met-7, gastrin/cholecystokinin and pancreatic polypeptide. However, their axons and terminals show varying amounts of the peptides, suggesting differential transport and utilisation. Only a few cells in the thoracic ganglion are immunoreactive to Met-8 antisera. These lie close to the nerve bundles suppling the legs. In the abdominal ganglion, Met-8 immunoreactive neurons project to the muscles of the hindgut. This study suggests that the extended enkephalin-like peptides ofCalliphora may have a variety of different roles: as neurotransmitter or neuromodulator substances; in the direct innervation of effector organs; and as neurohormones.  相似文献

Summary Neuronal pathways immunoreactive to antisera against the extended-enkephalins, Met-enkephalin-Arg6-Phe7 (Met-7) and Met-enkephalin-Arg6-Gly7-Leu8 (Met-8), have been identified in the brain of the blowfly Calliphora vomitoria. Co-localisation with other enkephalins in certain neurons suggests that a precursor similar to preproenkephalin A exists in insects and that differential enzymatic processing occurs as in vertebrates. Co-localisations of the extended-enkephalin-like peptides with other vertebrate-type peptides, including cholecystokinin and pancreatic polypeptide, also occur. The enkephalinergic pathways are specific, comprising a few groups of highly characteristic neurons and areas of neuropil. Of special interest is the finding that parts of the antennal chemosensory and the optic lobe visual systems contain Met-8 immunoreactive neurons. Within the median neurosecretory cell groups, some of the giant neurons show immunoreactivity to Met-8 and others to both Met-8 and Met-7. Fibres from these cells project to the corpus cardiacum and also to the suboesophageal ganglion, where arborisations occur in the tritocerebral neuropil. Co-localisation studies of these cells have shown that at certain terminals, one particular type of peptide is the dominant neuroregulator, whilst at other terminals, within the same cell, a different co-synthesised peptide predominates. Several groups of lateral neurosecretory cells show clearly defined enkephalinergic pathways, most of which have connections with the central body. The complex patterns of immunoreactivity seen in terminals in the different parts of the central body, suggest an important role for the enkephalin-like peptides in the integration of multimodal sensory inputs. The physiological functions of the extended-enkephalin-like peptides in the brain of Calliphora is still unknown, but the anatomical evidence suggests they may have a role similar to that in mammals, where they are thought to control aspects of feeding behaviour.  相似文献

Summary Ca++-ATPase activity (cf. Ando et al. 1981) was examined both light- and electron-microscopically in the neurohypophysis of the guinea pig. Apart from a strong activity within the walls of the blood vessels, in the parenchyma of the neurohypophysis the reaction product of the Ca++-ATPase activity was restricted to the plasmalemma of the pituicytes. This reaction was completely dependent upon Ca++ and the substrate, ATP; the reaction was inhibited by 0.1 mM quercetin, an inhibitor of Ca++-ATPase. A reduction of the enzyme activity occurred by 1) adding Mg++ to the standard incubation medium, and 2) substituting Ca++ with Mg++ at varing concentrations. In all experiments the neurosecretory fibers were devoid of Ca++-ATPase activity. The function of the Ca++-ATPase activity in the plasmalemma of the pituicytes is discussed in connection with the regulation of the extracellular Ca++ concentration, which seems to be important with respect to the discharge of secretory material from the neurosecretory fibers.Fellow of the Alexander von Humboldt Foundation, Bonn, Federal Republic of Germany.  相似文献

Summary To assess the role of the hypothalamic neurosecretory system in the mechanism of photoperiodic control of testicular growth in Spizella arborea, we have examined the effect of (1) photostimulation on the density of paraldehyde-fuchsin-stainable (PAF+) material in the neurovascular zone of the median eminence, and (2) interruption of the hypothalamo-hypophysial (neurosecretory) tract on the photoperiodic testicular response. Birds were captured during midwinter and retained on 8-hr daily photoperiods several months prior to the experiment. Interruption of the neurosecretory tract was effected by a small knife fashioned from stainless steel wire; radiography was employed to facilitate stereotaxic placement of the knife assembly.Extending the daily photoperiod from 8 to 20 hrs induced rapid testicular growth in photosensitive Spizella arborea, but had no consistent effect on the density of PAF+ material in the palisade layer of the anterior median eminence. Whereas some terminal controls showed moderate depletion after 21 days' exposure to 20-hr daily photoperiods, others retained as much or more stainable material than initial controls in which testicular growth was not induced. Moreover, testicular growth was induced at essentially identical rates whether the PAF+ innervation of the anterior median eminence was intact or eliminated to varying degrees. The finding that the rate of photoperiodically induced testicular growth is independent of the density or pattern of accumulation of PAF+material in the neurovascular zone of the median eminence, coupled with the inconstant effect of photostimulation on the density of stainable material therein, strongly suggests that the eminential component of the hypothalamic neurosecretory system is not a part of the mechanism that controls photoperiodic testicular growth in S. arborea.Contribution No. 980, Division of Biology, Kansas Agricultural Experiment Station, Manhattan 66502. Neurosecretory cells of only one type (A, sub type A2) are seen in adult Melanoplus. Two groups of about 400 cells each are located dorsally in the pars intercerebralis medialis; four cells are located deep within the protocerebrum. We found no neurosecretory cells in other parts of the central or sympathetic nervous systems. In about 10% of the specimens, there was marked asymmetry in the location of the dorsal cell groups, with both of these groups and their axons located in one lobe of the protocerebrum. The nervi corporis cardiaci 1 cross-over in the corpus cardiacum, with the result that material produced by neurosecretory cells on one side of the brain is transported along axons that undergo two chiasmata to the corpus cardiacum of the same side. Stainable secretory material could be traced clearly from the cerebral cells to the corpus cardiacum, and even into the oesophageal nerves from the hypocerebral ganglion. However, stainable neurosecretory material is never present in the corpus allatum or along any of the nerves to this gland.  相似文献

Summary Neuronal circuits in the brain and retrocerebral complex of the cockroach Diploptera punctata have been mapped immunocytochemically with antisera directed against the extended enkephalin, Met-enkephalin-Arg6-Gly7-Leu8 (Met-8). The pathways link median and lateral neurosecretory cells with the corpus cardiacum/corpus allatum complex. In females, nerve fibres penetrate the corpora allata and varicosities or terminals, immunoreactive to Met-8, surround the glandular cells. Males differ in having almost no Met-8 immunoreactivity in the corpora allata. The corpora cardiaca of both males and females are richly supplied with Met-8 immunoreactive material, in particular in the cap regions immediately adjacent to the corpora allata. A similarity in the amino-acid sequences of Met-8 and the C-terminus of the recently characterised allatostatins of D. punctata suggests that the pathways identified with the Met-8 antisera may be the same as those by which the allatostatins are transported from the brain to the corpus allatum. In comparative studies on the blowfly Calliphora vomitoria, similar neuronal pathways have been identified except that no sexual dimophism with respect to amounts of immunoreactive material within the corpus allatum has been observed. These results suggest a possible homology in the neuropeptide regulation of the gland.  相似文献

Gross morphology, staining characteristics and mapping of the diversity of the neurosecretory cell types in the brain and subesophageal ganglion of the scorpion Heterometrus swammerdami are reported. Special neurosecretory cell groups whose product is stainable with orange-G, acid fuchsin and Heidenhain's hematoxylin are present in the brain. In many of the living isolated neurosecretory cells, the secretory material appears luminous when viewed with dark ground illumination and granular when observed with phase contrast microscope. In the subesophageal ganglion the metameric arrangement of neurosecretory cells is distinct. Neurosecretory product accumulating in specific regions of subesophageal ganglion, and its axonal transport into the dorsal nerves and their termination in cephalic blood vessels apparently representing a storage and release organ of neurosecretion is reported.  相似文献

The posterior neurosecretory cell (PNC) group in the brain of Rhodnius prolixus is composed of five ultrastructurally identical cells. The PNC were examined in the unfed fifth instar and at seven stages (from 15 min to 14 days) after activation was initiated by feeding. Each stage examined revealed successive changes in morphology which can be related to the synthesis, maturation, storage and transport of neurosecretory material. It is suggested, in particular, that the lysosomal system (dense bodies and multivesicular bodies) may play a role in the maturation of the secretory granules.  相似文献

Summary The neurosecretory activity of azadirachtin A-treated Locusta migratoria that failed to mature was compared with that of rapidly maturing control females by means of histological techniques. High resolution drymount autoradiography was used to localize [22,23-3H2]dihydroazadirachtin A in the brain and corpus cardiacum (CC). The neurosecretory system of the locusts treated with this insecticide was accompanied by an unusually high accumulation of paraldehyde fuchsin (PAF)-stainable neurosecretory material (NSM) in the brain fibers and in the storage lobes of the CC. The control females never had such an intense accumulation of NSM. The accumulation of NSM, resulting from its poor release, may influence its synthesis which is controlled by feed-back regulation. [22,23-3H2]Dihydroazadirachtin A is localized mainly at the peripheral blood-brain barrier and does not penetrate into the brain effectively, whereas it completely covers the CC. It is concluded that azadirachtin A affects the endocrine activity of the brain in an indirect way.  相似文献

Summary The fate of 35S-cysteine incorporated into the hypothalamic-hypophyseal neurosecretory system was followed in normal rats as a function of time from 30 min up to 30 hrs after intraperitoneal administration (11 different times, three rats each). Autoradiography and grain counting were employed to determine quantitatively the activities present in: supraoptic nucleus (SON), paraventricular nucleus (PVN), three different sites in the hypothalamic-hypophyseal tract (HHT), corresponding to its initial, middle and late portions, and neurohypophysis (NH). In SON and PVN, the kinetic behaviour of 35S-cysteine was compatible with a two-compartment model, implying decline of tracer activity in accordance with two exponential components. Of these, the slow throughput component had its closely equivalent, apparently independent, counterparts in the HHT and NH. Replicas of the rapidly abating activity peak were seen, superimposed on the slow component, in the HHT with such timing of its appearance at the various sites, as to be consistent with the concept of travel through the HHT, at substantially constant speed and without delay, of material rich in 35S-cysteine that has been primarily synthesized in the SON and PVN (with the transport theory of neurosecretion). This flow was calculated to have a velocity in the order of 0.6 mm per hour. The kinetic phenomena and their association with the supposed sites of synthesis, and transport, of the neurosecretory material are discussed with reference to earlier literature.This work was supported by a grant from the Sigrid Jusélius Foundation, Helsinki  相似文献

In vivo treatment of maize (Zea mays L.) coleoptile segments with auxin (indole-3-acetic acid; IAA) and fusicoccin (FC) followed by plasma-membrane isolation was used to characterize the effects of these treatments on the plasma-membrane H+-ATPase. Both IAA and FC increased H+ extrusion and elongation rate of the coleoptile segments, FC more strongly than IAA. Plasma membranes isolated after in-vivo treatment with FC showed a twofold stimulation of ATP hydrolysis and a several-fold stimulation of H+ pumping, whereas no effect was observed after IAA treatment, irrespective of whether the plasma membranes were prepared by two-phase partitioning or sucrose-gradient centrifugation. A more detailed investigation of the kinetic properties and pH dependence of the enzyme showed that FC treatment led to a twofold increase in V max, a decrease in K m for ATP from 1.5 mM to 0.24 mM, and a change in pH dependence resulting in increased activity at physiological pH levels. Again, IAA treatment showed no effects. Quantitation of the H+-ATPase by immunostaining using four different antibodies revealed no difference between IAA-and FC-treated material, and controls. Summary The complicated architectural pattern of the preoptico-neurohypophysial neurosecretory system of Clarias batrachus is described using bulk staining procedure. The nucleus preopticus is U-shaped; its limbs constitute the PMC and PPC. The difference in staining intensities noticed among the PMC and PPC might suggest a possible functional difference within the system. Axons of the PMC diagonally pass through the PPC and emerge along with those of the latter to form the right and the left main tracts. The folds and recurrent curves of the neurosecretory axonal tracts, the formation of several pairs of lateral tracts and their union to form median tracts might be devices to increase the storing and releasing capacity for the neurosecretory material. The presence of prominent Herring bodies, and diffuse areas in the neurosecretory tract suggest possible sites of release of neurohormones along the highly vascular hypothalamic region. The common neurosecretory tract is divided into several smaller branches at the level of the pars distalis prior to entering the pars intermedia. Branches do penetrate into the pars distalis, but those entering the rostral component of the pituitary are relatively scarce.Regeneration of the anterior stump of the severed hypophysial stalk to form a neurohypophysis-like organ is also demonstrated in toto with the aid of the bulk staining procedure. A demonstrable increase of stainable neurosecretory material was noticed all along the hypothalamo-neurohypophysial complex of some specimens subjected to complete darkness. Total staining methods could be advantageously used to map the course of recurrent tracts and extra-hypophysial axonal endings. Since the neurosecretory system seems to exhibit a varied structural pattern in different fishes, extension of this study to a larger number of species may throw additional light on the function and the evolution of this important system.I am indebted to Dr. K. Summary The paramedian neurosecretory cells (PNC) (A-type) in the suboesophageal ganglion of the cricket, Teleogryllus commodus (Walk.), have been studied by electron microscopy. In control animals (10 day-old virgin females) three different cell stages could be distinguished: Stage 1 shows a variable content of elementary granules and characteristics of actively synthesising cells. Stage 2 is characterised by the presence of numerous fusion bodies, which are formed by the coalescence of elementary granules, presumably for storage purposes. In stage 3 granules are degraded in lysosomes (?). While the production of material rich in cysteine increases after ovariectomy (Dürnberger et al., 1978), the fine structure of the cells is essentially unchanged. The only noted differences are an increased lysosomal synthetic activity and the existence of stages intermediate between 2 and 3, which were never found in control animals. Certain mutant alleles of the otu locus in Drosophila melanogaster produce abnormal nurse cells in the ovaries. These cells are called pseudonurse cells (PNC), since they generate polytene chromosomes instead of endopolyploid ones and do not normally have an oocyte to nurse. The banding pattern of polytene chromosome 3 from the salivary glands (SG) and from PNCs of homozygous otu 1 females was compared and a detailed photomap of PNC chromosomes with different degrees of polyteny is presented. The banding pattern was found to be strikingly similiar in the two tissues. The puffing pattern of the PNC chromosomes was also studied and the function of the PNC chromosomes is discussed. No constrictions or breaks were found in the PNC chromosomes which seems to indicate that these sites, which are known to be underreplicated in the SG chromosomes, are equally replicated along with the rest of the chromosomes in the PNC nuclei.  相似文献

In otu mutants of Drosophila melanogaster ovarian tumours develop because of the high mitotic activity of the mutant cystocytes; the latter are normally endopolyploid. In certain alleles of otu, however, a varying proportion of the mutant ovarian cystocytes undergo polyteny. Mutant cystocytes with polytene chromosomes are termed pseudonurse cells (PNC). Polytene chromosome morphology and banding patterns in PNC of otu 1/otu3 flies were cytologically analysed. Extensive variability was noted in the quality of the banding pattern of the PNC chromosomes which ranged from highly condensed (condensed PNC chromosomes) to those with a banding pattern (banded PNC chromosomes) similar to that in larval salivary gland cells (SGC). Both the condensed and banded PNC chromosomes frequently enter into a diffuse state characterised by weakened synapsis of the polytene chromatids and alterations in their banding pattern (diffuse PNC chromosomes). Analysis of DNA synthesis patterns in the various morphological forms of PNC polytene chromosomes by 3H-thymidine autoradiography revealed a basic similarity to the pattern seen in polytene nuclei of larval SGC. Independently replicating sites, however, could be unambiguously identified only in banded PNC chromosomes. Comparison of late replicating sites in such PNC chromosomes with those of larval SGC showed a remarkable similarity in the two cell types. These results suggest a close correlation between the polytene chromosome banding pattern and its replicative organization.  相似文献

The annual killifish Austrofundulus limnaeus inhabits rainwater pools in the Maracaibo basin of Venezuela. This species persists in ephemeral habitats by producing diapausing embryos that are resistant to the stresses imposed by the drying of their aquatic habitat. Embryos of A. limnaeus are likely exposed to a highly variable osmotic environment during development, but their tolerance of osmotic stress has not been characterized. We investigated the capacity of these embryos to survive in hypersaline environments and evaluated the possible mechanisms used to support osmoregulation. Diapausing embryos of A. limnaeus defend their internal osmolality of around 290 mOsmol kg−1 H2O−1 against salt stress as high as 50 ppt salinity. We find that diapausing embryos of A. limnaeus have a permeability to water that is orders of magnitude lower than other teleost fish embryos. The activity of ion motive ATPases that may be important in the extrusion of ions via mitochondrial rich cells do not appear to be playing a large role in osmoregulation of A. limnaeus embryos. We conclude that for the duration of embryonic development the unique properties of the enveloping cell layer of A. limnaeus embryos acts as a permeability barrier to water and ions and supports osmoregulation in this species in response to a broad range of osmotic environments.
