Novel hydroxy-cis-enynoic acids have been prepared. Those of type
C(CH2)6CO2H have been found to cyclise to furanoid acids on treatment with alkali, or on chromatography on argentous silica gel. The hydroxy-cis-enynoic acids stimulate smooth muscle, the furanoid acids are not similarly active.  相似文献

Methyl 2,5-disubstituted C18 furanoid fatty ester (viz. methyl 9,12-epoxyoctadeca-9,11-dienoate) was readily converted to methyl 9,12-dioxostearate using mineral or maleic acid. Conversion of the naturally occurring 2,3,5-trisubstituted furanoid fatty ester (viz. methyl 10,13-epoxy-11-methyloctadeca-10,12-dienoate) to the corresponding methyl 10,13-dioxo-11-methylstearate was much slower in rate under similar reaction conditions. The case of separating the dioxo derivatives from a mixture of other common fatty esters was demonstrated and the cyclodehydration of the isolated dioxo derivatives to the parent furanoid ester was rapidly achieved using dilute BF3-methanol complex.  相似文献

The fatty acids of the triacylglycerol fraction of the latex of the rubber plant consists of 97% of a C18 furanoid fatty acid, 10,13-epoxy-11-methyloctadeca-10,12-dienoic. The free fatty acid fraction is composed of a more equally distributed mixture of 16:0, 18:0, 18:1, 18:2 and the furanoid acid. A novel dioxo fatty acid, 10,13-dioxo-11-methyloctadecanoic, was also isolated and characterized.  相似文献

In this study we describe the development of peptidomimetic analogs of the potent vasoactive intestinal peptide receptor binding inhibitor, Leu(1) -Met(2) -Tyr(3) -Pro(4) -Thr(5) -Tyr(6) -Leu(7) -Lys(8) -OH 1, by incorporating furanoid sugar amino acids (SAAs) 2-4 into the molecule. The furanoid SAAs 2-4 were used as dipeptide isosteres to replace Tyr(3) -Pro(4) or Pro(4) -Thr(5) in sequence 1. The resulting analogs 5-9 were tested for their anti-cancer activities in vitro, following the standard MTT assay on a panel of human cancer cell lines. One of the potent analogs, 6a was tested in vivo for tumor regression on primary colon tumor xenografted nude mice. Our experimental results suggest that many of these analogs show either retention or enhancement of biological activity.  相似文献

Tracer experiments with 14C-labelled precursors in Iris × hollandica cv. Wedgwood, Roseda lutea L. and Reseda Odorata L. have demonstrated that 3-(3-carboxyphenyl)alanine and 3-(3-carboxy-4-hydroxyphenyl)alanine can be derived from the corresponding pyruvic acids, presumably by unspecific trans-aminations, and that (3-carboxyphenyl)glycine and (3-carboxy-4-hydroxy-phenyl)glycine can be derived from the corresponding phenylglyoxylic acids The glycine derivatives are derived from the alanine derivatives, and the corresponding mandelic acids are intermediates in these transformations. The corresponding phenylacetic acids are incorporated only slightly into the glycine derivatives, indicating that oxidation at the benzylic position in the C6–C3 compounds takes place early in the transformation. The corresponding cinnamics acids are not metabolized at all in the plants.  相似文献

d-Erythrosyl aziridines were obtained from d-erythrosyl triazoles either by photolysis or through diazirine intermediates. These were found to undergo rich, high yielding chemistry by reaction with protic acids (HCl, BiI3/H2O and trifluoroacetic acid) leading to two types of furanoid sugar α-amino acids, and polyhydroxylprolines. Based on experimental evidence, reaction mechanisms have been proposed for the syntheses.

Based on a CSD search, a meta‐analysis of 1179 structures of 19 natural amino acids H3NCαH(R)C′(O)O and their derivatives H3NCαH(R)C′(O)O(H/R/M), protonated, esterified, or coordinated at the carboxylic group, shows that the chirality chain with its two steps, established in the preceding paper for alanine, can be extended to natural amino acids. High diastereoselectivities are observed in the induction from the L configuration at Cα to the ?ψ and +ψ conformations, which in turn distort the planar carboxylic group CαC′(Ocis)Otrans to asymmetric flat tetrahedra, showing that the chirality chain is an integral part of natural amino acids.  相似文献

Phenolic acids derived from vegetables, fruits and beverages are considered abundant sources of natural antioxidants consumed in the human diet. In addition to having well-known antioxidant activity, phenolic acids also exhibit pro-oxidant activity under selected conditions. We hypothesized that the availability of extracellular H2O2 derived from phenolic acid autoxidation will diffuse across cell membranes to participate as a messenger molecule to activate intracellular redox signaling in response to oxidative stress. We report on the relative activity of structurally different phenolic acids to generate specific changes in the extracellular - intracellular H2O2 flux that induces intracellular redox signaling corresponding to a function to reduce intracellular oxidative stress. HyPer-3 methodology was used to measure increases in intracellular H2O2 in differentiated Caco-2 intestinal cells in response to phenolic acid autoxidation and changes in extracellular H2O2 production. The potential for different phenolic acids to autoxidize and generate H2O2 was dependent on the structure and concentration of phenolic acid. Activation of nuclear factor erythroid 2-related factor (Nrf2) cell signaling was enhanced (p < 0.05) by phenolic acid induced H2O2 production, and mitigated when present along with catalase (p < 0.05), or, alternatively by blocking aquaporin 3 (AQP3) function (p < 0.05) using DFP00173 as the AQP3 inhibitor. The relative capacity of phenolic acids to generate H2O2 via autoxidation was structure specific and corresponded to the level of Nrf2 cell signaling in differentiated Caco-2 epithelial cells. The Nrf2-Keap1 response paralleled the extent of reduced oxidative stress observed in differentiated Caco-2 cells determined by dichloro-dihydro-fluorescein diacetate (DCFH-DA) assay.  相似文献

The infrared and Raman spectra of all 2,5-disubstituted C18 furanoid fatty esters were studied. Where the furan system was located at the end of the alkyl chain or conjugated to the carbomethoxy group, such positional isomers could be readily identified through their unique absorption features in the infrared and Raman spectra. Other positional isomers of the series gave less differentiating spectral absorption characteristics.  相似文献

2-Hydroxy acids were believed to be absent in algae until this study, in which the analysis of microalgae belonging to Chlorophyta (Chlamydomonas reinhardtii and Chlorella pyrenoidosa), Rhodophyta (Cyanidium caldarium M-8 and Cyanidium caldarium RK-1) and Cyanophyta (Anbaena variabilis, Anacystis nidulans, Oscillatoria species and Phormidium foveolarum) is reported. 2-Hydroxy adds with carbon chain lengths of C16-C26, were found in all the algal samples studied, ranging in concentrations from 4.0 to 320μg/g dry alga. The dominant constituents are 2-hydroxyhexadecanoic, 2-hydroxynonadecanoic, 2-hydroxyhexacosanoic and a branched 2-hydroxy-C19 acid. The distribution patterns of the acids differed significantly among the algal samples. Hence 2-hydroxy acids may be useful for the classification of algal species as well as being an important source of 2-hydroxy acids in the natural environment.  相似文献

—Purified myelin incorporated l -[14C]leucine and l -[14C]lysine into myelin proteins in an enzymatic process similar to that of renal brush border membranes. The system was not inhibited by cycloheximide or puromycin or by pretreatment with ribonuclease; the reaction was inhibited by cetophenicol. ATP was an effector, shifting the optimal pH from 7.2 to 8.3. In the presence of ATP, myelin was less dense in a sucrose gradient. Ammonia was released from the membrane during the incorporation of amino acids. Myelin preloaded with cold leucine did not incorporate [14C]leucine but did incorporate [14C]lysine; there was no cross inhibition between the two amino acids. The incorporation was into or onto proteins of the Wolfgram proteolipid fraction of myelin. The incorporation was of the high affinity type with a Km of 10?7m and was restricted to the natural amino acids.  相似文献

Addition to Bacillus acidocaldarius of acids which can act as primers for fatty acid synthesis promote the synthesis of corresponding fatty acids competitively. The effective acids are n?C5 to -?7 (not C4 or C8), iso- and anteiso-C, and ?C, (not C4), and a range of cyclic acids from cyclobutylacetic and cyclopentanecarboxylic to cycloheptylacetic. New non-natural ω-cyclobutyl-, ω-cyclopentyl-, and ω-cycloheptyl-fatty acids are obtainable. The range of acceptable primers and the range of fatty acids produced therefrom indicate, respectively, the substrate specificities of the transacylase which introduces acyl species into fatty acids synthesis and the one which removes them. The specificity of the primer transacylase may be similar to that in some rumen anaerobes.  相似文献

Condensation of 1,2,3,4,6-penta-O-acetyl-á-l-idopyranose (1) with phenol yielded phenyl 2,3,4,6-tetra-O-acetyl-α- (2) and α-l-idopyranoside (4). Deacetylation of 2 and 4 afforded phenyl α and β-l-idopyranosides (3 and 5), respectively, the structures of which were verified by periodate oxidation studies. A platinum-catalyzed oxidation of 3 and 5 produced the amorphous phenyl α- and β-L-idopyranosiduronic acids (9 and 11), respectively, which were isolated as the crystalline cyclohexylammonium salts. Phenyl β- and α-d-glucopyranosiduronic acids are apparent minor byproducts of the catalytic oxidations, resulting from an inversion at C-5. p-Nitrophenyl α-d-mannopyranosiduronic acid and p-nitrophenyl α- and β-d-galactopyranosiduronic acids are also described.  相似文献

The main glycerolipids (monogalactosyl-, digalactosyl-, sulphoquinovosyl diacylglycerol, phosphatidylglycerol) from five blue-green algae (Microcystis, Anabaena, Nostoc, Oscillatoria, Tolypothrix) were analyzed for fatty acid composition, occurrence of diglyceride species and positional distribution of fatty acids between thesn-1- andsn-2-position of glycerol. In contrast to eucaryotic plants biosynthetically closely related lipids (monogalactosyl-, digalactosyl-, trigalactosyl diacylglycerol) show nearly identical diglyceride moieties, whereas sulphoquinovosyl diacylglycerol and phosphatidylglycerol are separated from galactolipids by composition as well as occurrence of fatty acids. On the other hand the positional distribution of fatty acids in all lipids is controlled exclusively by chain length and not by degree of unsaturation with C18-fatty acids at thesn-1- and C16-fatty acids at thesn-2-position. While the L -configuration of amino acids predominates in all known living systems, D -enantiomers of amino acids have been detected with highly sensitive chromatographic techniques in human physiological fluids. In the present study, the survival of Chinese hamster ovary cells (CHO) and HeLa cells was inhibited by exposure to high concentrations of some D - or L -amino acids. Inhibition of colony formation, though, was not necessarily observed to be chiral-dependent. Some L -amino acids (L AAS) were found to be toxic while other D -amino acids (D AAS) were innocuous in both cultures. This is contradictory to the previous observations that D AAS were generally considered to be harmful. Frequently it was implied, although not experimentally proven, that the L AAS were not toxic. One of the metabolites produced by oxidative deamination of D - or L AAS is hydrogen peroxide (H2O2), a reactive oxygen species (ROS) that is decomposed by catalase. Increased intracellular H2O2 can result in peroxidation of lipids. We measured catalase activity and the lipid peroxide levels (LPO) after incubating cells in either D - or L AAS. The amino acids (AAS) that were found to inhibit colony formation were found to be associated with higher levels of catalase activity and LPO. Therefore, we hypothesize that enhanced ROS generation may be, in part, responsible for the observed toxicity of some amino acids. © 1996 Wiley-Liss, Inc.  相似文献

DL-erythro-2,3-dihydroxyoctadecanoic acid, synthesized according to Palameta and Pro?tenik (Tetrahedron, 19 (1969) 1463) was converted to the R(+)-1-phenylethylamide and the obtained diastereomers resolved by chromatography on silica gel. The enantiomeric erythro-2,3-dihydroxyoctadecanoic acids were recovered by acidic hydrolysis (m.p., 117°, [α]D22 = ±3.28°, [MD22 = ±10.38°). A comparison of the chromatographic mobility of R- and S-1-phenylethylamides and the optical rotation of 2,3-dihydroxy fatty acids from fungi sphingo-lipids shows that the natural occurring enantiomer has (+)D-erythro configuration.  相似文献

Investigations on the absorption of shortchain fatty acids across ruminal epithelium of sheep were performed both in vitro (Ussing chamber technique, using propionic acid representatively for shortchain fatty acids) and in vivo (washed, isolated reticulorumen). A pH-induced, nearly tenfold increase in the concentration of undissociated propionate led to an only twofold increase in mucosal-to-serosal flux of propionate (in vitro). Neither amiloride (1 mmol·l-1, in vitro) nor theophylline (10 mmol·l-1, in vivo), inhibitors of the ruminal Na+/H+ exchanger, exerted any significant influence on propionate fluxes or short-chain fatty acids absorption, respectively. Total replacement of luminal Na+ (by choline) did not alter short-chain fatty acids absorption (in vivo). Mucosal 4,4-diisothiocyanatostilbene-2,2-disulfonic acid (0.1 mmol·l-1) or mucosal nitrate (40 mmol·l-1) markedly reduced propionate net flux (in vitro). Increasing mucosal Cl- concentration brought about a significant drop in mucosal-to-serosal flux of propionate (in vitro) and in short-chain fatty acids net absorption (in vivo), respectively. The results obtained suggest that short-chain fatty acids are absorbed both as anions and as undissociated acids across ruminal epithelium of sheep. The mechanisms by which adult male Schistosoma mansoni transport amino acids have been investigated using radioactive amino acids during 2-min incubation times. The transport constants (Kt) for mediated uptake of glycine, proline, methionine, arginine, glutamate, and tryptophan were calculated to be 0.60-1.05, 1.67-1.98, 2.0, 0.10-0.35, 0.30-0.50, and 0.5-1.0 mM, respectively. Maximal velocities (Vmax) were 5.5–7.5, 25, 6.4, 1.5-2.0, 2.5, and 3.0–6.0 μmoles absorbed/g worm protein/2 min, respectively. Cysteine is taken up solely by diffusion. Proline uptake is unique in that no significant diffusion component was found. The other amino acids studied were absorbed by diffusion as well as by specific transport systems. In the 2-min incubation periods employed glycine, proline, glutamate, and methionine were not significantly metabolized indicating that the uptake studies using these substrates reflect transport. Metabolism of the other amino acids used in these studies was not examined. The specificity of the transport systems was studied by testing the inhibitory effects of various amino acids on the uptake of each of the amino acids studied. The results suggest the presence of at least five transport systems. There is a highly specific transport locus for proline, and one for acidic amino acids. There are probably at least two transport systems, each of broad and overlapping specificity, for most of the neutral amino acids. Basic amino acids also appear to be taken up by complex transport systems, at least one of which overlaps with the neutral sites. The results are discussed with respect to the nutrition of the parasite and the host-parasite relationship.  相似文献

The metabolism of arachidonic and linoleic acids by VX2 carcinoma tissue was determined. Prostaglandin E2 was the major metabolic product of arachidonic acid in the neoplastic tissue. Minor products accounting for 3– 8% of arachidonic acid metabolism were 11-hydroxy-5, 8, 12, 14-eicosatetraenoic acid (11-HETE) and 15-hydroxy-5, 8, 11, 13-eicosatetraenoic acid (15-HETE). Linoleic acid was converted to a mixture of 9-hydroxy-10, 12-octadecadienoic acid (9-HODD) and 13-hydroxy-9, 11-octadecadienoic acid (13-HODD). The conversion of linoleic acid to monohydroxy C-18 fatty acids varied from 40–80% 9-HODD and 20–60% 13-HODD in tumor tissue harvested from different animals. The quantity of monohydroxy C-18 fatty acids biosynthesized by VX2 carcinoma tissue from endogenous linoleic acid equals or exceeds that of prostaglandin E2 biosynthesis from endogenous arachidonic acid. The presence of a hydroxyl group adjacent to a conjugated diene suggest that the monohydroxy C-18 and monohydroxy C-20 fatty acids were formed via the action of lipoxygenase-like enzymes. These lipoxygenase-like reactions are inhibited by indomethacin in a concentration-dependent fashion similar to the inhibition of prostaglandin E2 biosynthesis. The enzymes catalyzing the lipoxygenase-like reactions of linoleic and arachidonic acids are localized in the microsomal fraction of VX2 carcinoma tissue. These data suggest that the lipoxygenase-like reactions are catalyzed by fatty acid cyclooxygenase and that there are two major pathways of fatty acid cyclooxygenase metabolism of polyenoic fatty acids in the neoplastic tissue. One pathway involves the formation of prostaglandin E2 via cyclic endoperoxy intermediates. The second pathway involves the formation of monohydroxy C-18 fatty acids from linoleic acid via lipoxygenase-like reactions.  相似文献
