Several lichen compounds, i.e. lobaric acid (1), a β-orcinol depsidone from Stereocaulon alpinum L., (+)-protolichesterinic acid (2), an aliphatic -methylene-γ-lactone from Cetraria islandica Laur. (Parmeliaceae), (+)-usnic acid (3), a dibenzofuran from Cladonia arbuscula (Wallr.) Rabenh. (Cladoniaceae), parietin (4), an anthraquinone from Xanthoria elegans (Link) Th. Fr. (Calaplacaceae) and baeomycesic acid (5), a β-orcinol depside isolated from Thamnolia vermicularis (Sw.) Schaer. var. subuliformis (Ehrh.) Schaer. were tested for inhibitory activity on platelet-type 12(S)-lipoxygenase using a cell-based in vitro system in human platelets. Lobaric acid (1) and (+)-protolichesterinic acid (2) proved to be pronounced inhibitors of platelet-type 12(S)-lipoxygenase, whereas baeomycesic acid (5) showed only weak activity (inhibitory activity at a concentration of 100 μg/ml: 1 93.4±6.62%, 2 98,5±1.19%, 5 14.7±2.76%). Usnic acid (3) and parietin (4) were not active at this concentration. 1 and 2 showed a clear dose–response relationship in the range of 3.33–100 μg/ml. According to the calculated IC50 values the highest inhibitory activity was observed for the depsidone 1 (IC50=28.5 μM) followed by 2 (IC50=77.0 μM). The activity of 1 was comparable to that of the flavone baicalein, which is known as a selective 12(S)-lipoxygenase inhibitor (IC50=24.6 μM). 相似文献

Six species of lichens, such as Usnea florida, Usnea barbata, Usnea longissima, Usnea rigida, Usnea hirta and Usnea subflorida, were collected from different areas of Anatolia (district of Antalya, Karabük, Qankiri, Giresun and Trabzon) in Turkey. Their usnic acid amounts in acetone extracts were determined by HPLC. In addition, antimicrobial activities of these extracts were determined against Escherichia coli (ATCC 35218), Enterococcus faecalis (RSKK 508), Proteus mirabilis (Pasteur Ens. 235), Staphylococcus aureus, Bacillus subtilis and Bacillus megaterium. It was shown that with increasing amount of usnic acid, the antimicrobial activity increased. Usnic acid contents of Usnea species varied between 0.22-6.49% of dry weight. 相似文献

The widespread secondary metabolite usnic acid, a dibenzofuran derivative, is the principal acetone-soluble compound in the lichen Flavocetraria nivalis. Seasonal variation in concentrations were studied in four populations of this lichen, three from Arctic-alpine habitats in the Northern Hemisphere, and one from Patagonian heathland in the Southern Hemisphere. Usnic acid is produced in large amounts, making up between 4% and 8% of thallus dry weight. Large seasonal variation is seen, with a trend towards peak levels in late spring and early summer, and generally low levels during autumn and winter. However, at an Arctic steppe in Central West Greenland, remarkably high levels were also detected during late autumn and early winter. Comparisons with environmental data using model selection procedures show that usnic acid levels of three of the populations are positively correlated with time of season, as measured by the proximity in time to nearest summer solstice, solar radiation levels, and temperature conditions. All these three variables are intercorrelated, thus indicating the same overall trend. For the three driest sites, precipitation rates are included in the models that best explain the variation in usnic acid. However, the explanatory powers of the models are generally low, partly due to high variation between thalli growing together and sampled at the same time. This is the first attempt to compare statistically seasonal variation in usnic acid concentrations and environmental variables, and thus also the first time it is shown that the concentration in various populations of the same lichen species shows different types of correlation with seasonal climatic changes. 相似文献

Abstract A synthesis of the epiphytic lichen vegetation on larch in the Italian Alps is presented. Vegetation types are described on the basis of a numerical classification of 291 relevés. They can be compared to a few well-known phytosociological unions found in other parts of the Alps. Some communities are restricted to subalpine areas with continental climate. Despite the homogeneity of the substrate, the flora is rich, with 98 species, several of which are rare, and hence of interest from a conservational point of view. 相似文献

Activities of aminopropyltransferases (spermidine synthase and spermine synthase) were inhibited by aurintricarboxylic acid (ATA). Spermidine synthase was slightly more sensitive to the inhibitor than spermine synthase. These inhibitions were not prevented by 0.15 M NaCl. Inhibition by ATA of spermidine synthase was ‘uncompetitive’ with respect to putrescine and that of spermine synthase was ‘non-competitive’ with respect to spermidine. When the amount of spermidine synthase or spermine synthase was varied, inhibition ratio hardly changed on either case implying no appreciable interaction between ATA and these enzymes. 相似文献

The effect of various culture conditions on growth, lipid production and fatty acid composition in Mucor rouxii and Mucor sp.1b were studied. Total lipid production was higher in media containing potassium nitrate for both the cultures (30%) and cultures grown on plant seed oil produced more than 44% lipid. Among the carbon sources tested, γ-linolenic acid (GLA) production was maximal in cultures grown on glucose. The major fatty acids produced by these two cultures were palmitic, stearic and oleic acids. Levels of GLA in M. rouxii and M. sp.1b was in the range of 3–17% under different culture conditions. Lactose was a poor promoter for biomass and lipid production in both cultures. No GLA was found in fungal cultures grown on sesame oil. The optimal conditions for the production of GLA was standardised in these cultures. 相似文献

Abstract Two isolates of Sclerotinia sclerotiorum , the highly aggressive (B24) and the weakly aggressive (SS41), were grown on liquid media containing one of the following carbon sources: purified cell walls obtained from onion or sunflower, pectin, polygalacturonic acid, carboxymethylcellulose, xylan or arabinogalactan. Isolates were equally able to utilize these substrates for mycelial growth but differed in their ability to utilize them for oxalate production. B24 produces oxalic acid always to a substantial extent, SS41 only in traces. The poor ability to produce oxalic acid by SS41 seems to be due to a lower efficiency in the synthetic pathway. 相似文献

The effects of linoleic acid and bovine serum albumin on hybridoma cell growth and antibody production were investigated. In dish cultivation, linoleic acid on its own promoted cell growth when used at concentrations below 50 mg L–1, but strongly inhibited growth at a concentration of 100 mg L–1 on more. However, linoleic acid bound to bovine serum albumin did not inhibit cell growth, even at a concentration as high as 100 mg L–1. Also, linoleic acid did not affect the specific antibody production rate, with or without bovine serum albumin. In order to elucidate the enhancement of antibody production by bovine serum albumin, fractions were prepared by ultrafiltration (98% molecular weight cut-offs, 50,000 and 17,000) and the effects of the fractionation on antibody production were studied in batch cultivation. The high-molecular-weight fraction (50,000) promoted antibody production whereas the low-molecular-weight fraction (17,000) inhibited it. In continuous cultivation, the high-molecular-weight fraction was also found to enhance antibody production. 相似文献

目的 探索一种新的胚胎大鼠腹侧中脑黑质细胞的混合培养方法,以获得具有高比例多巴胺神经元的原代细胞体系,有利于在体外条件下进行帕金森病(Parkinson's disease,PD)的发病机制和防治的研究.方法 分离E15 (Embryonic Day 15)SD胎鼠中脑黑质区域组织,分散为单细胞后,分别用DMEM/F12+ 10%FBS含血清培养基和Neurobasal+ N1无血清培养基设置不同组别进行培养,通过免疫组织化学方法检测在不同培养条件下细胞的生长状态及多巴胺能神经元的比例.结果 DMEM/F12+ 10% FBS与Neurobasal+ N1先后一周交替换液的的培养体系中获得的TH阳性神经元占神经元的比例可达31%左右,明显高于单独使用DMEM/F12+ 10% FBS培养组(约10%).结论 DMEM/F12+ 10% FBS与Neurobasal +N1先后一周交替换液是一种黑质神经元和胶质细胞混合培养并能获得高比例多巴胺神经元的有效培养方法. 相似文献

The charcoalified fragment of the dorsiventrally organized, internally stratified presumed green algal lichen Chlorolichenomycites salopensis from the Lower Devonian Lochkovian strata in the Welsh Borderland carries bacterial colonies on the upper surface, i.e. the cortex, and actinobacterial filaments in the medulla underneath the photobiont layer. Moreover relatively thin hyphae of presumed endolichenic fungi were found. As in extant lichens, which are best regarded as consortia with an unknown number of participants, this internally stratified, fossil thallus fragment of a presumed green algal lichen harbours a diverse microbial community. 相似文献

Summary Production of 2,3-butanediol byKlebsiella oxytoca was enhanced in the presence of low levels (<8 g/l) of added sodium lactate. Cell growth was inhibited, however, and essentially stopped above 15 g/l added lactate. Levels of by-products (acetic acid and ethanol) were also higher. With 3 g/l lactate and an initial glucose level of 98 g/l, butanediol concentration and productivity increased 164% with 98% utilization of glucose. With high glucose concentration (219 g/l), addition of 2.64 g/l lactate after the growth phase resulted in 81 g/l butanediol, with a productivity of 0.65 g/l/h and 71% glucose utilization. 相似文献

In vitro and in vivo techniques were used to examine the production and subsequent fate of the endogenous excitotoxin quinolinic acid (QUIN) following administration of its bioprecursor L-kynurenine (KYN). Incubation of liver slices in the presence of 10-1000 microM KYN resulted in a dose- and time-dependent release of QUIN into the incubation medium. Less than 15% of total QUIN produced was recovered from the tissue. In vivo experiments, performed with a microdialysis probe inserted in the jugular vein of anesthetized rats, showed that injection of KYN (20-600 mg/kg, i.v.) causes rapid and dose-dependent increases in the serum level of QUIN. Peak QUIN concentrations in serum dialysates were reached 75 minutes following KYN administration. Longer lasting increases were detected following the administration of pyrazinamide (20 mg/kg, i.p.), an indirectly acting stimulator of QUIN biosynthesis in the periphery. The data demonstrate the feasibility of assessing the mechanisms of QUIN production and disposition in experimental paradigms which can be expected to allow insights into the function and possible dysfunction of QUIN in the brain. 相似文献

Usnic acid (UA), an old antibiotic and one of the first described mitochondrial uncouplers, has demonstrated many beneficial activities, such as antimicrobial, antiviral, antitumour and anti-inflammatory properties. Here, we performed a thorough investigation of effects of usnic acid and its analogues on artificial planar bilayer lipid membrane (BLM), rat liver mitochondria and bacteria. Surprisingly enough, all of the three hydroxyl groups of UA appeared to be involved in its proton-shuttling activity on BLM. We ascribed this fact to an ability of UA to form complexes with calcium ions, aiding it in cycling protons across the membrane. Actually, the addition of calcium ions markedly stimulated the UA-induced electrical current across BLM. By using the calcium ionophore A23187, we proved the involvement of calcium ions in the UA uncoupling action on isolated rat liver mitochondria. The calcium-chelating property of UA was demonstrated here by the method of extracting metal ions into a hydrophobic phase. Modification of any of the hydroxyl groups in UA dramatically reduced not only the UA-induced current across BLM and the UA-mediated calcium extraction, but also the uncoupling activity of UA in mitochondria and the inhibiting effect of UA on the growth of Bacillus subtilis. The ability of UA to cause dissipation of membrane potential in isolated liver mitochondria and bacterial cells was shown here for the first time. In view of the data obtained, the protonophoric activity of UA is considered to make a significant contribution to its antibacterial action. 相似文献
