Summary The genomes of the phylogenetically related but morphologically distinct bacteriophages LP52 and theta () were compared by electron microscopic heteroduplex analysis. The heteroduplex maps were aligned with known restriction maps. In the heteroduplices of LP52 DNA (63.8 kb) with the DNA of the lytic phage c (65.9 kb) the tracts of homologous DNA cover about 50% of the genome length and are interspaced by four large and ten smaller non-base-paired regions. The largest block of non-homologous DNA (18.9 kb), represents the right-hand end and there is an unmatched piece of DNA at the left-hand end as well. Most of the heterology is due to substitution resulting in the conservation of the total length of DNA; the three insertions/deletions amount to less than 3.2% of the genome length. Heteroduplices between the DNAs of phage LP52 and the temperate phage 1 (65.0 kb) resembled those of LP52: c except for the absence of minor loops. Heteroduplex c:1 displayed about 9% heterology in seven separate loops which coincided with sections of diversity on the restriction maps; 4.8% of 1 DNA did not hybridize with either c or LP52 DNA.  相似文献

Summary Clear-plaque phage c, attacking bacitracin-producing strains of B. licheniformis, yields spontaneous temperate mutants at high frequency; the temperate mutants fall into several classes phenotypically different in plaque morphology and properties of lysogenised bacteria. The most common phenotype 3 has DNA restriction fragment patterns identical with those of the parent c; some less common temperate forms, i.e. 1 and 2, produce different restriction fragment patterns, sugesting that a part of the original c DNA has been reorganized or replaced by some foreign genetic material. The changed fragment pattern remains stable upon subsequent passaging of the phage or of the lysogenic bacteria. Neither class of temperate phage mutants gives clearplaque revertants at measurable frequency. Lysogenisation of bacteria with any class of temperate phage confers immunity to all temperate forms and to c; virulent mutants vir, which plate with 100% efficiency on lysogens for 1 and 2 but not for 3, occur in stocks of c at a frequency of 10–7. The mutation from c to vir is not accompanied by any change of the restriction fragment patterns of DNA.  相似文献

Summary We studied visual optics using ophthalmoscopy in six species of coraciform birds, five species from the family Alcedinidae (kingfishers) and one from the family Meropidae (bee-eaters). All six species had large angular separations between the two foveae of one eye (angle ); angle was greater than 40° in all cases, the largest separation so far reported for any group of vertebrates. In all kingfishers, but not in the bee-eater, the plane containing the projections of both foveae in one eye (angle ) was rotated from the horizontal plane so that the projection of the monocular fovea was lower than the projection of the binocular fovea.Retinal ganglion cell isodensity maps were obtained for the sacred kingfisher (Halcyon sancta) and laughing kookaburra (Dacelo gigas). These maps were constructed in the usual way for the peripheral regions of the retina. For the high-density, multi-layered, central region of the retinal ganglion cell layer, we used a combined retinal wholemount-cross-sectional technique. It was discovered that the horizontal streak, formed by the elliptically shaped contours of retinal ganglion cell isodensity, deviated inferiorly at its nasal extent from the line of the foveae. This deviation had the same sign, and slightly greater magnitude, as the rotation (angle ) observed ophthalmoscopically when the eyes were in their primary position.Our new observations provide new insights into the functional significance of the bifoveate visual organization. In particular, the relationship between angle and angle suggests one strategy which could be used by kingfishers to maximize their visual capabilities when hunting from a perch above the substrate.  相似文献

Summary The semitendinosus muscle of the rabbit is composed of a homogeneous fiber population. Fiber typing was carried out by means of light and electron microscopy according to which this muscle exhibits structural features that are either characteristic for both red and white fibers, or that do not allow for any classification according to the A-, B-, C-fiber system. Hence the long-held assumption that the semitendinosus muscle of the rabbit represents a classical paradigm of a red muscle should be revised in light of the present results.  相似文献

Summary Natural recombinant genomes between several, phenotypically distinct forms of phages and were isolated and characterized by DNA restriction fragment mapping and electron microscopic heteroduplex analysis. The phenotypes of the recombinants were correlated with the physical maps of the genomes, and several genetic functions were therfore defined and mapped. All genes necessary for the assembly of infectious virus particles map in a contiguous tract of DNA comprising about 20 kb, or nearly one third of the genome length. No DNA homology occurs within these domains of the two genomes, so that homologous recombination does not take place here and phenotypic mixing of the phages is eo ipso excluded. Other regions of heterology contain regulatory genes responsible for the lytic or temperate character of the phages, and for exclusion of phage by .  相似文献

Summary As an introduction the two interpretations of the chyle from milk ducts and milk vessels of certain plants are pointed out. Under the phase microscope the chyle appears comparable to the content of vegetal vacuoles, and like this it is surrounded by a very thin mantle of plasm. From rheological experiments with the emulsoid it follows its Non-Newtonian flow behaviour. There are studied several measurements of the dependence of viscosity on the shear modulus , the degree of dispersity and dilution, the temperature and the dispersal portion, and the dependence on concentration is to assess for the viscosity and for the yield stress . Despite of the Non-Newtonian flow character of the chyles these are not to co-ordinate to the protoplasm but owing to the histologic finds and because of the behaviour of the vacuolar sap ofNitella cells they are compared with the content of vacuoles.  相似文献

Alterations inN- andO-linked glycosylation affect cell surface expression and antigenicity of recombinant glycophorin A expressed in transfected Chinese hamster ovary (CHO) cells. To understand these effects further, glycophorin A was purified by immunoaffinity chromatography from transfected wild type and glycosylation deficient CHO cells. TheO-glycans were characterized both biochemically, using gel filtration and high performance anion exchange chromatography, and immunologically, using carbohydrate specific monoclonal antibodies to probe Western blots. TheO-glycans of human erythrocyte glycophorin A consist mainly of short oligosaccharides with one, two, or three sialic acid residues linked to a common disaccharide core, Gal1-3GalNAc1-Ser/Thr, with the disialylated structure being the most abundant. With the exception of the trisialylated derivative, the same structures were found on recombinant glycophorin A expressed by wild type CHO cells. However, in contrast to human crythrocyte glycophorin A, the monosialylated oligosaccharide was the most abundant structure on the recombinant protein. Furthermore, recombinant glycophorin A was shown to express a small amount of the Tn antigen (GalNAc1-Ser/Thr). Recombinant glycophorin A had the sameO-glycan composition, whether purified from clones expressing high or moderate levels of the recombinant glycoprotein. This indicates that the level of expression of the transfected glycoprotein did not affect itsO-glycan composition. Deletion of theN-linked glycosylation site at Asn26, by introducing the Mi.I mutation (Thr28 Met) by site-directed mutagenesis, did not markedly affect theO-glycan composition of the resulting recombinant glycoprotein expressed in wild type CHO cells. This demonstrates that the presence or absence of theN-glycan did not influenceO-linked glycosylation of the recombinant glycoprotein. Finally, theO-glycans on recombinant glycophorin A expressed in the Lec 2 and Lec 8 glycosylation deficient CHO cells were characterized. TheO-glycans on Lec 2 cell glycophorin A were predominantly Gal1-3GalNAc1-Ser/Thr (T antigen), while those on Lec 8 glycophorin A were exclusively GalNAc1-Ser/Thr (Tn antigen). These results will lead to a better understanding of the cell biology and immunology of this important human erythrocyte glycoprotein.  相似文献

Photosynthesis-irradiance (P-E) curves are widely used to describe photosynthetic efficiency and potential. Contemporary models assume maximal photosynthetic quantum yield () at low irradiances. But P-E observations made with both oxygen evolution and carbon uptake techniques show that this is not always the case. Using new and published data in conjunction with modeling exercises, we demonstrate that regardless of the mechanism there can be reductions in at low irradiances that are not readily observable using conventional P-E analyses. We also show that analytical errors, such as inaccurate estimation of dark oxygen consumption or carbon uptake, can markedly affect the structure of -E curves with negligible effect on P-E curve structure. Whether from respiration `corrections' or other mechanisms, these deviations in at low light levels from the maximum quantum yield of photosynthesis (max) can lead to significant errors (> 50%) in the estimation of the linear portion of the P-E curve and ultimately max. Non-linear models of P-E, such as the rectangular hyperbola, quadratic, exponential and hyperbolic tangent that are commonly used to estimate the initial slope () of the P-E curve assume that is maximal at low light levels and therefore can err in the estimation of max when is reduced at low light levels. Using a diverse data set of 622 P-E curves with a total of 7623 points, we show that although model skills are high (r 2 = 0.96 ± 0.05, 0.97 ± 0.04, 0.97 ± 0.04 and 0.97 ± 0.04, respectively), a large fraction of the model-predicted max differ by greater than 10% from true max values (91%, 50%, 82% and 46%, respectively). Data from these observations and modeling exercises lead us to suggest that max be determined by directly estimating the true maximum of a -E curve rather than using the more conventional methodology employing the initial slope of the P-E curve.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

The resting state of wild-type nitrogenase MoFe protein exhibits an S=3/2 electron paramagnetic resonance (EPR) signal originating from the FeMo cofactor, the enzymes active site. When nitrogenase turns over under CO, this signal disappears and one (sometimes two) of three new EPR signals, which also arise from the FeMo cofactor, appears, depending on the CO concentration. The appearance and properties of these CO-inducible EPR signals, which were also generated with variant MoFe proteins (R96Q, R96K, Q191K, R359K, R96K/R359K, R277C, R277H, and nifV) that are impacted around the FeMo cofactor, have been investigated. No new CO-induced EPR signals arise from any variant, suggesting that no new CO-binding sites are produced by the substitutions. All variant proteins, except R277H, produce the lo-CO signal; all, except Q191K, produce the hi(5)-CO signal; but only two (R96Q and nifV) exhibit the hi-CO signal. FeMo cofactors environment clearly dictates which CO-induced EPR signals are generated; however, none of these EPR signals correlate with CO inhibition of H2 evolution observed with some of these variants. CO inhibition of H2 evolution is, therefore, due to CO binding to a different site(s) from those responsible for the CO-induced EPR signals. Some resting-state variants have overlapping S=3/2 EPR signals, whose intensities simultaneously decrease under turnover conditions, indicating that all FeMo cofactor conformations are catalytically active. Moreover, these variants produce a similar number of hi(5)-CO signals after turnover under CO to the number of resting-state S=3/2 signals. The FeMo cofactor associated with the hi(5)-CO signal likely contains two bridging CO molecules.  相似文献

Summary The results of previous behavioral studies can be so interpreted that the prey-catching behavior in the toad is elicited if there is a local motion restricted with-in a small part of the visual field, while it is suppressed if there is a global motion over a large part of the visual field. This has led us to design experiments to answer a specific question (yet a very essential one for understanding neural processes underlying this behavior): Are there local motion detectors in the toad's visual system that are not activated by global motion over a large part of the visual field but are activated by local motion confined within a smaller part of it? The present study showed that (1) the majority of the toad's tectal neurons exhibit properties of the local motion detectors as defined above, and (2) these properties can be explained from the receptive field structure revealed in the present experiments. Based on these results, we suggest that the tectal local motion detectors are essential for the detection and localization of small moving prey-objects in the natural environment while ignoring the large moving objects or the self-induced motion of the visual field.Abbreviations ERF excitatory receptive field - G1-5 group 1–5 neurons  相似文献

To assess the role interleukins and mitogens play in regulating immunoglobulin (Ig) gene expression via the Ig enhancer and promoter, transgenic mice carrying two different Ig gene regulatory regions were generated. One, EkCAT, contains the Ig heavy chain enhancer (E) and the light chain promoter driving the chloramphenicol acetyltransferase (CAT) gene. In the other, EkCAT, CAT is under the control of the promoter alone. E and relative activity were assessed by CAT assay. In EkCAT mice, low CAT expression was consistently found in spleen, bone marrow, mesenteric lymph node, and thymus but not in brain, lung, or kidney. In EkCAT mice, CAT expression was detectable just above background in lymphoid tissues, suggesting a basic level of tissue specificity in the absence of the enhancer. Whole spleen cell cultures prepared from the mice were treated with lymphokines and mitogens. Lipopolysaccharide (LPS), concanavilin A (Con A), interleukin 6 (IL-6), and interferon- (IFN-) increased CAT expression to varying extents in cells derived from EkCAT mice but not in spleen cells prepared from EkCAT mice. Thus, the presence of E, in addition to the promoter, is essential for the stimulation of CAT expression mediated by these factors. B cells from EkCAT mice were separated by density into populations of small and large cells. In untreated small B cells, no CAT expression was detected and only addition of LPS resulted in an increase in CAT expression. In large B cells, CAT was expressed at a low level without addition of exogenous factors. Incubation with LPS, IL-6, Con A and IFN- caused CAT expression to increase several-fold. The possibility of solving the mass balances to a multiplicity of substrates within a CSTR in the presence of a chemical reaction following Michaelis-Menten kinetics using the assumption that the discrete distribution of said substrates is well approximated by an equivalent continuous distribution on the molecular weight is explored. The applicability of such reasoning is tested with a convenient numerical example. In addition to providing the limiting behavior of the discrete formulation as the number of homologous substrates increases, the continuous formulation yields in general simpler functional forms for the final distribution of substrates than the discrete counterpart due to the recursive nature of the solution in the latter case.List of Symbols C{N. Summary To identify the mildew resistance locus Mla in barley with molecular markers, closely linked genomic RFLP clones were selected with the help of near-isogenic lines having the Pallas and Siri background. Out of 22 polymorphic clones 3 were located around the Mla locus on chromosome 5 with a distance of 5.1 + 2.9 cM (MWG 1H068), 4.2±1.7 cM (MWG 1H060) and 0.7 ± 0.7 cM (MWG 1H036), respectively. The polymorphic clone MWG 1H036 displayed the same RFLP pattern in both Pallas and Siri near-isogenic lines and in different varieties digested with six restriction enzymes possessing the same mildew resistance gene. The alleles of the Mla locus were grouped in 11 classes according to their specific RFLP patterns; 3 of these groups contain the majority of Mla alleles already used in barley breeding programs in Europe.  相似文献

Summary A bacterium capable of utilizing -pinene as a sole carbon and energy source was isolated from soil. This strain, named strain S201-1, which was identified as Pseudomonas maltophilia on the basis of its taxonomical properties, accumulated limonene, borneol, camphor, perillic acid, and 2-(4-methyl-3-cyclohexenylidene) propionic acid from -pinene in the culture broth. It was demonstrated that -pinene, -pinene, borneol, camphor, and a number of p-menthane derivatives were oxidized by this strain. Relations between the protonation of -pinene and the formation of the products by the microbe are discussed.  相似文献

Male Schizocosa ocreata wolf spiders court females with synchronous visual and seismic displays. We tested whether male S. ocreata modify their courtship in relation to light environment, and associated utility of the visual components. Males were generally more active and more likely to perform the major courtship element (jerky tapping) when in the light. One courtship element (arching) was only observed in the light while another (vertical leg-extend) was only observed in the dark. Courtship in the dark retained visual components, suggesting spiders cannot disengage these components of multi-modal display even when superfluous. Once initiated, there was no evidence that performance rate or time spent in each courtship element differed in the light and in the dark.  相似文献

Summary The production of sex pheromones responsible for the induction of the sexual agglutination ability in the mutants of the mating type locus, mata1, mat1 and mat2, was examined. mata1 cells behaved just like wild-type MATa cells in the production of a pheromone and responsiveness to pheromone. On the other hand, mat1 cells showed neither a nor ability in the production of and the agglutination ability induction by sex pheromones. Cells carrying mat2 secreted a pheromone but not pheromone and showed the ability to inactivate pheromone. Summary Within the Theory of Dual Radiation Action, the heuristically useful function,(), that two energy depositions, a distance apart, will result in observable damage can be written in terms of two more fundamental quantities: One,s(), describes the structure of the sensitive matrix of the cell. The other,g(), describes the probability that two elementary injuries, a distance apart, will combine to produce observable damage. A priori, the known enhancement exhibited byy() at nanometer separations could be due to an enhancement ofs(),g() or both. For the endpoint of chromatid exchanges in Chinese hamster cells,() has been evaluated from experimental data ands() from a model system. From theseg() has been estimated and is roughly constant below 1 µm, decreasing rapidly at larger separations. Summary Crude extracts from -lysogens treated with mitomycin C were prepared, and immunity repressor levels in the extracts were assayed by the binding activity specific to DNA immunity region. It has been shown that while the repressor levels in the extracts from C600(+) are reduced after mitomycin C treatment, the levels in C600recA(+) and C600 C72(+) which have defects in lifting the immunity are not affected by the treatment. The repressor levels in the extracts prepared from C600 T44(+) after temperature shift up, whose prophage is inducible at high temperature, are also reduced. From the study of chloramphenicol effect, it was indicated that de novo protein synthesis is required for the inactivation of the repressor in C600(+) by mitomycin C, but not in T44(+) by high temperature.  相似文献

In natural populations of the anaerobic phototrophic bacterium Chromatium weissei, many cells support a prokaryotic epibiont. This epibiont appears in several forms, all from the life cycle of a single species. A typical epibiont consists of one to five flattened coccoid cells stacked one above the other, perpendicular to the C. weissei surface. The cells at the proximal and distal ends of the stack are 0.6 m in diameter and 0.8 m in length; mid-stack cells are slightly shorter. A typical three or four cell stack is 2 m in length. Small mesosome-like inclusions in the distal cell are involved in the development of droplet shaped cells which are released from the end of each stack. These specialised droplet cells probably transfer to new hosts when C. weissei cells collide, thereafter developing into new epibiont stacks. It is likely that the epibiont grows heterotrophically using the substantial production of dissolved organic carbon within the dense plates of photosynthesising C. weissei which develop naturally. Thus the epibiont uses its unusual method of growth and dispersion to maintain position in the microbial plate upon which it depends.  相似文献
