Chicken feather keratin was treated with lime (calcium hydroxide) to obtain a liquid product rich in amino acids and polypeptides that can be used as an animal feed supplement. The effect of treatment conditions and the properties of the soluble keratin were studied. At high temperatures (150 degrees C), 80% of feather keratin was solubilized within 25 min, whereas a relatively longer reaction time (300 min) is needed at moderate temperatures (100 degrees C). After 3h of hydrolysis at 150 degrees C, 95% of feather keratin was digested. For the recommended conditions (100 degrees C, 300 min, and 0.1g Ca(OH)(2)/g dry feather), after lime treatment, about 54% of calcium can be recovered by carbonating. In rumen fluid, ammonia production from soluble keratin was similar to that of soybean and cottonseed meals and was greatly less than that of urea, indicating that no ammonia toxicity will result from cattle being fed soluble keratin.  相似文献

Hydrolysis of biomass waste (such as fish waste, chicken waste, hair and feather) to produce amino acids was studied in sub-critical water, with reaction temperatures from 180 to 320 degrees C and reaction pressures from 3 to 30 MPa. The product of amino acid was determined by Amino Acid Analyzer (BioLC), and 18 kinds of amino acid were obtained. The results show that the controlling of reaction atmosphere, pressure, temperature and time of hydrolysis is very important to obtain high yield of amino acid; most of amino acids reached maximum yield at reaction temperature range of 200-290 degrees C and reaction time range of 5-20 min. There are obvious changes of amino acids yield at reaction pressures of 6-16 MPa and reaction temperature around 260 degrees C, owing to the homogeneousness of the first two phases of water in the formation of vapor and liquid. There are different yields of the same amino acid in different reaction atmospheres (e.g. air, carbon dioxide and nitrogen).  相似文献

Value-added subcritical water hydrolysate from rice bran and soybean meal   总被引:2,自引:0,他引:2  
New value-added product was derived from agricultural by-products: rice bran and soybean meal by means of subcritical water (SW) hydrolysis. The effect of temperature (200-220 degrees C), reaction time (10-30 min), raw material-to-water weight ratio (1:5 and 2:5), was determined on the yields of protein, total amino acids, and reducing sugars in the soluble products. The suitable hydrolysis time was 30 min and the proper weight ratio of the raw material-to-water was 1:5. The reaction temperature suitable for the production of protein and amino acids was 220 degrees C for raw and deoiled rice bran, 210 degrees C for raw soybean meal, and 200 degrees C for deoiled soybean meal. The products were also found to have antioxidant activity as tested by ABTS(.)(+) scavenging assay. In addition, sensory evaluation of milk added with the hydrolysis product of deoiled rice bran indicated the potential use of the product as a nutritious drink.  相似文献

Chlorosubstitution reactions occur readily during HCl hydrolysis of delta- and epsilon-hydroxynorleucines (Hnle), the products of deamination of poly-L-lysine by nitrite at low pH. During amino acid analysis, chloronorleucines elute as new peaks after delta- and epsilon-Hnle. To determine if other hydroxyamino acids undergo similar changes during hydrolysis, they were subjected individually to HCl hydrolysis conditions with and without added phenol. Amino acid analyses indicated that terminal hydroxy groups on linear side chains undergo reactions during HCl hydrolysis; the products appear as new peaks which may be chloroderivatives. In contrast, no new peaks are observed in HCl hydrolysates of delta-hydroxylysine or amino acids with beta-hydroxy groups (beta-hydroxynorvaline, serine, and threonine). Phenol did not protect linear amino acids from reactions during HCl hydrolysis but did prevent loss of the cyclic amino acids tyrosine, hydroxyproline, and 3,4-dihydroxyphenylalanine. Although the gamma-hydroxy group of homoserine would be expected to undergo reaction, HCl catalyzes its cyclization to form homoserine lactone instead.  相似文献

Dry mixtures of glycine and succinic acid as well as of glycine, phenylalanine and succinic acid were exposed to UV-irradiation and heating up to 170 degrees in atmospheres of O2 and N2. Under these conditions, synthesis of substances with peptide bonds was observed. Irradiation or heating of the first mixture results in synthesis of a substance which is water soluble and capable of dialysis, exhibiting therefore low degree of polymerization. This substance contains COOH-groups, but lacks free NH2-groups. The latter reveal themselves during hydrolysis which brings about the appearance of the initial compounds. In the second mixture, which contains an aromatic amino acid, several products are synthesized some of them being presented by the insoluble and non-dialysed precipitate (polymer). Among soluble products, peptide-like substances (without free NH2-groups) and dipeptides were found. UV-irradiation causes photolysis of phenylalanine which is mixed with succinic acid; as the result, tyrosine, alanine and asparagic acid are found among the products synthesized. These amino acids are not formed during heat-induced synthesis. Abiogenic synthesis takes place more readily in the presence of O2 than in a N2 atmosphere. Irradiation of aqueous solution of the product synthesized resulted in the photolysis of the latter. The results obtained indicate that during abiogenic period, synthesis of peptides and peptide-like substances from dry mixtures of amino acids and organic acids induced by UV-irradiation and heat might take place on the litosphere.  相似文献

On the Acaciin     
Synthesis of N-2,2,3,3-tetrafluoropropionyl-amino acids (abbreviated as tetrafluoropropionyl- or Tfp-amino acids) by the reaction of amino acids with tetrafluoropropionyl anhydride was described, Racemization was observed to occur to a considerable extent during the process of the introduction of this group. Optically active Tfp-amino acids were able to be prepared free from racemization by transesterification of methyl tetrafluoropropionate with amino acid tert-butyl esters, followed by treatment of the resulting Tfp-amino acid tert-butyl esters with trifluoroacetic acid. Some Tfp-dipeptide esters were prepared from the corresponding dipeptide tert-butyl esters in this way. This group was cleavable by mild alkaline hydrolysis or by reduction with sodium borohydrde as well as the Tfa-group.  相似文献

The objectives of this study were to evaluate the dialysed quantities of amino acids and minerals (calcium, phosphorus, magnesium, iron and zinc) in two types of infant formulas, one with a basis of native cow milk proteins and the other highly hydrolysed, to evaluate the possible application of Miller's in vitro method in amino acid and mineral availability studies. The percentage of dialysis differed between proteins. The hydrolysis treatment applied to the proteins significantly increased the dialysis percentage of almost all the amino acids. The dialysability of all the minerals was statistically greater in the formula made with a basis of hydrolysed proteins. Miller's method was seen to be of use for showing the effect which the hydrolysis treatment has on the availability of amino acids. Despite its limitations of having to be carried out in vitro, the study suggests that the nutritional value in relation with amino acids, minerals and trace elements differs between the both types of formulas.  相似文献

The reactivity of the imino acids formed in the D- or L-amino acid oxidase reaction was studied. It was found that: (1) When imino acids reacted with the alpha-amino group of glycine or other amino acids, transimination yielded derivatives less stable to hydrolysis than the parent imino acids. In contrast, when imino acids reacted with the epsilon-amino group of lysine or other primary amines, transimination yielded derivatives more stable to hydrolysis than the parent imino acids. (2) Imino acids react rapidly with hydrazine and semicarbazide, forming stable hydrazones and semicarbazones. At pH 7.7, the rate of reaction of the imino acid analogue of leucine with semicarbazide was 10(4) times greater than that of the corresponding keto acid. The reaction of imino acids with these reagents is rapid enough to permit one to follow spectrophotometrically the amino acid oxidase reaction. Imino acids also reacted with cyanide to yield stable adducts. (3) The rate of hydrolysis of the imino acid analogue of leucine was independent of pH above pH 8.5. At lower pH values, the rate of hydrolysis increased with decreasing pH. At 25 degrees C and in the absence of added amino compounds, this imino acid had a half-life of 22 s at pH 8.5. Two methods are described for determining protein by measurement of amino acids released by acid or base hydrolysis. One achieves sufficient sensitivity to measure as little as 3 ng of protein by reaction of the amino acids with orthophthalaldehyde and mercaptoethanol in a small volume, followed by dilution in 0.5 n NaOH for fluorometric measurement of the product. The other method is somewhat more sensitive. Released l-glutamate is determined enzymatically by reaction with glutamic dehydrogenase (EC 1.4.1.2) and NAD+; the NADH produced is amplified by enzymatic cycling. To avoid the use of sealed tubes and minimize dangers of contamination, hydrolysis is carried out in small droplets in “oil wells.”  相似文献

经酸水解,利用氨基酸自动分析仪测定干制加工前后的苦瓜中总氨基酸和游离氨基酸含量。结果表明干制前后的苦瓜在氨基酸质量分数上有明显差别,总氨基酸质量分数分别为11.99%和10.87%,游离氨基酸分别为2.36%和0.70%。结论:干制前后的苦瓜中氨基酸质量分数差别明显,其中碱性氨基酸质量分数的变化尤其显著,总氨基酸中精氨酸质量分数下降50%,在游离氨基酸中下降78%;同时还原糖质量分数也由干制前的4.86%下降为干制后的1.86%,说明苦瓜干制过程中发生了美拉德反应,造成了氨基酸和还原糖含量的下降。  相似文献

Peptic hydrolyzate of soy protein was submitted to the plastein reaction with α-chymotrypsin under the following condition: substrate concentration, 20%; enzyme-substrate ratio by weight, 1/100; reaction pH, 7.0; and reaction temperature, 37°C. The plastein yield resulting from the plastein reaction for 24 hr was found to depend on the degree of hydrolysis of the substrate (per cent ratio between nitrogen amount in 10% trichloroacetic acid soluble nitrogen and that in whole hydrolyzate); the optimum degree of hydrolysis for the highest plastein yield seemed to lie around 80%. A turbidity appeared in the process of the plastein reaction, whose intensity was correlative to the plastein yield. The peptic hydrolyzate of soy protein per se had bitterness and its magnitude decreased with increasing plastein yield.

As a result of the plastein reaction applied for 24 hr to the hydrolyzate whose degree of hydrolysis was 80%, the average molecular weight estimated by the change in amino nitrogen content increased by approximately three times. The molecular weight distribution pattern obtained by gel filtration supported the above result. The total amount of amino acids liberated from the plastein reaction product by its treatment with either leucine aminopeptidase or carboxypeptidase A was significantly less than that liberated from the original hydrolyzate by its similar treatment. This result also supports the formation of higher-molecular protein-like substances by the plastein reaction. Deuteration study followed by IR spectrometry showed the occurrence of peptide bond formation, i.e. decrease in ionized carboxyl group at 1575 cm?1 and increase in deuterated amide at 1450 cm?1, even at the earlier stages of the plastein reaction.  相似文献

Responses of soluble amino acids and organic acids to either ambient (36 Pa) or elevated (100 Pa) CO2 treatments were determined using barley primary leaves (Hordeum vulgare L. cv. Brant). Total soluble amino acids were increased 33% by CO2 enrichment 9 days after sowing (DAS), but a decrease relative to the ambient CO2 treatment was observed with increasing leaf age. Marked declines of glutamine and asparagine were observed under CO2 enrichment, both diurnally and with advancing leaf age. Consequently, total soluble amino acids were 59% lower in the elevated compared to the ambient CO2 treatment 17 DAS. It was likely that chlorosis in response to CO2 enrichment negatively impacted soluble amino acid levels in older barley primary leaves. In contrast to the ambient CO2 treatment, glutamine and most other soluble amino acids decreased as much as 60% during the latter half of a 12 h photoperiod in primary leaves of 13-day-old seedlings grown under enhanced CO2. Malate was decreased about 9 percent by CO2 enrichment and citrate and succinate were increased by similar amounts when measured 9 and 13 DAS. Malate accumulation was also decreased about 20% by CO2 enrichment on a diurnal basis. The onset of CO2-dependent leaf yellowing had much less of an effect on organic acids than on soluble amino acids. This above results emphasized the sensitivity of N metabolism to CO2 enrichment in barley. Increased levels of citrate and succinate in response to CO2 enrichment suggested that the tricarboxylic acid cycle was upregulated in barley by CO2 enrichment. In summary, organic and amino acid levels in barley primary leaves were dynamic and were altered by age, diurnally and in response to CO2 enrichment.  相似文献

1. It has been demonstrated that hair contains lipids which cannot be removed by extensive extraction with chloroform-methanol mixtures. These integral lipids can be extracted only after the hair has been subjected to alkaline hydrolysis. 2. Integral hair lipids include cholesterol sulfate (0.7-2.9 mg/g hair), ceramides (0.6-1.4 mg/g), cholesterol (0.3-1.4 mg/g), fatty alcohols (trace-0.2 mg/g) and fatty acids (2.3-4.0 mg/g). 3. One of the major integral hair lipids, representing 38.4-47.6% of the total fatty acids, is the anteisobranched 18-methyleicosanoic acid. 4. The species examined included human (Homo sapiens), pig (Sus scrofa), dog (Canis familiaris), sheep (Ovis ammon aries) and cow (Bos taurus).  相似文献

Bluegill-degrading bacteria were isolated from various environmental sources. Brevibacillus sp. BGM1 degraded bluegill efficiently at 50 degrees C, and its culture supernatant showed the highest peptide and amino acid concentrations as trichloroacetic acid (TCA) soluble fraction (ASF) (10.7 mg/ml) of all supernatants obtained with bluegill as a substrate. Strain BGM1 secreted a protease(s) into the medium, and the concentration of peptides and amino acids gradually increased. The fertile effect of the degraded bluegill products (DGP) on Brassica rapa was also investigated. The root hair density of B. rapa grown with DGP at a concentration of 30 mug peptides and amino acids/ml was about 1.7 times higher than when grown with the same concentration of undegraded bluegill. DGP was shown to increase root hair numbers and adventitious root formation. The results of this study suggest that a specific peptide(s) for promotion of root hair is produced from the order Perciformes with a protease(s) from BGM1.  相似文献

Pigmented tissues from bovine eye were used as a source for isolation of tyrosinase from normal melanocytes. Tyrosinase is highly hydrophobic and the isolation procedure is mainly based on the use of hydrophobic interaction chromatography. The bovine enzyme is, in contrast to the human melanoma tyrosinase, mainly soluble. The predominant part of the ocular enzyme from cow has a molecular weight and isoelectric behavior similar to that of the soluble tyrosinase in the human melanoma cells. The N-terminal amino acid sequence of isolated bovine tyrosinase was determined by automated Edman degradation. The N-terminal amino acid sequence from normal bovine tyrosinase was identical to the sequence of an N-terminal region of mouse melanoma tyrosinase predicted from a c-DNA clone by Kwon et al. (1988). The amino acid sequence of bovine tyrosinase shows homology to that of human tyrosinase (Wittbjer et al., 1989), but three amino acids of the 16 residues determined by us differed. A careful examination of the low-molecular-weight solutes released during mild acid hydrolysis of the lipopolysaccharide of Pseudomonas aeruginosa (N.C.T.C. 1999) revealed the presence of ethanolamine triphosphate. During storage, the compound decomposed to give ethanolamine pyrophosphate, identified in a previous study (Drewry et al., 1971); PP(i) may be a further decomposition product. Evidence for the attachment of ethanolamine triphosphate to a polysaccharide fraction was obtained, but the possibility that some was attached to the lipid A moiety was not excluded. Basic compounds released during the hydrolysis of lipopolysaccharide included amino acids, polyamines and oligopeptides.  相似文献

Amino acids can be converted to peptides by the process of heating if the mixture is dry and if the appropriate proportion of glutamic acid as the dominant monomer is present. Three thermal peptide fractions of the diffusible fraction were isolated in this study. Trifluoracetic acid (TFA) hydrolysis was utilized to open the blocked N-terminal group before performing the dansylation reaction. When the molecular weight of the thermal peptide increased, the proportion of neutral amino acids increased slightly, with a decrease in the concentration of glutamic acid. Cow kappa-casein has been modified by photo-oxidation in the presence of rose bengal and by the chemical reagents diethyl pyrocarbonate, 2-hydroxy-5-nitro-benzyl bromide and iodoacetic acid. Photo-oxidation resulted in the destruction of histidine and tryptophan residues and all of the histidines could be ethoxy-formylated by treatment with diethyl pyrocarbonate. Both procedures caused a loss in the susceptibility of the Phe-Met linkage of kappa-casein to chymosin hydrolysis. Treatment of kappa-casein with 2-hydroxy-5-nitrobenzyl bromide and iodoacetic acid caused the loss of tryptophan and methionine residues respectively but, in both cases, the susceptibility of the modified protein to chymosin hydrolysis remained unaffected. Of the amino acids examined it is concluded that only the histidine residues of cow kappa-casein are important for the hydrolytic action of chymosin and, furthermore, the treatment with diethyl pyrocarbonate suggests that only one of the three histidines plays an essential role.  相似文献

An assay has been developed to monitor the activity of the lytic transglycosylases which does not involve the use of radiolabel. Samples of lytic transglycosylase were incubated with isolated and purified insoluble peptidoglycan as substrate for varying lengths of time. Residual insoluble material was removed by ultracentrifugation in a microfuge and the solubilized components were treated with sodium borohydride prior to acid hydrolysis. The optimal conditions for this acid hydrolysis were established to be incubation at 96 degrees C for 1 h in 6 M HCl, in vacuo. The hydrolyzed samples were subjected to amino acid/sugar analysis by cation-exchange chromatography on a Beckman System Gold amino acid analyzer. To effect a clear resolution of muramic acid from serine and glutamic acid, the equilibration buffer was modified to be composed of 33 mM sodium citrate, pH 3.12. The product of the lyase reaction of the lytic transglycosylases are 1,6-anhydromuramyl residues, which are not reduced by the sodium borohydride treatment. On the other hand, the muramyl residues arising at the reducing ends of peptidoglycan after treatment with muramidases (hydrolyases) are reduced to muramitol residues, which elute from the amino acid analyzer prior to aspartic acid. This assay thus distinguishes the activity of the two enzymes and was applied to determine the initial activities of increasing concentrations of a soluble derivative of lytic transglycosylase B from the opportunistic pathogen Pseudomonas aeruginosa.  相似文献
