The involvement of Na+, K+, Cl- or Ca2+ in the regulation of salinity stress-induced proline accumulation via the inhibition of the activity of proline dehydrogenase (PDH; EC 1.4.3.1), a catabolic enzyme of proline, was investigated in the marine green macroalga Ulva fasciata Delile. After 6 h of exposure to elevated artificial seawater (ASW) salinity, adjusted either by increasing the NaCl content in 30 ASW (a change in ion ratio) or by concentrating ASW (a constant ion ratio), the contents of Na+, K+ and Cl- linearly accumulated with increasing salinity from 30-90 (parts per thousand); the accumulation pattern of each ion was similar between the two treatments. An increase in NaCl content in ASW induced proline accumulation, but decreased both the PDH activity and the total water-soluble Ca2+ contents, while concentrated ASW had no effect. As compared to a constant value at 30, both the contents of total and water-soluble CA2+ and the activity of PDH decreased 1 h after exposure to 60 (adjusted by increasing NaCl content in 30 ASW) and concomitantly the content of seawater Ca2+ increased, while proline accumulated after 3 h. The addition of 15 mM ethylene glycol-bis-(2-aminoethyl ether) N,N,N-tetraacetic acid (EGTA) in 60 ASW (adjusted by increasing the NaCl content in 30 ASW) enhanced both the proline accumulation and the decrease in the content of total and water-soluble cellular Ca2+ and the activity of PDH; the effects of EGTA were reversed by 10 mM CaSO4. These results indicate that a loss of cellular Ca2+ is associated with the NaCl induction of proline accumulation via an inhibition of PDH activity in U. fasciata.  相似文献

An increase of cytosolic Ca2 in the unicellular green alga Eremosphaera viridis activities Ca2-dependent K channels causing a hyperpolarization of the plasma membrane. Data from parallel calcium, and potential measurements were combined with I/V relationships. E. 1987.Comparison of the effects of root temperature on nitrate andammonium nutrition of oilseed rape (Brassica napusL.) in flowingsolution culture. II. Cation-anion balance.—J. exp. Bot.38: 1589-1602. The effects of root temperature and form of N nutrition (NH4or NOJ) on the mineral composition of the plant, the balanceof inorganic cation-anion uptake and on the apparent net effluxof H +/OHions from the roots were studied with 49-d-oldoilseed rape (Brassica napusL. cv. Bien venu) in flowing solutionculture. Plants were pre-treated for 14 d at a root temperatureof 5 °C prior to constant root temperatures of 3, 7, 11or 17°C for 14 d, with a common shoot temperature of 20/15°Cday/night. Nitrogen was supplied as NH+44 or NO3 at 10mmol m3. Values of Q10 (7-17°C) for mean unit absorptionrates of all the major nutrient ions (K+ , Mg++ , NH+4, SO4,H2PO4, NO3), except Ca++, were > 2.0 over the first 5 d oftreatment but thereafter were < 1.5; the apparent effectof temperature on uptake rates diminished with time. Under NH+4nutrition, inorganic cation uptake (Mg+ + + K++Ca+ + +NH+4)exceeded inorganic anion uptake (SO44+ H2PO4) over 14d at all temperatures, with the proportion of cation uptakeas NH4 remaining constant (0.67-0-68) irrespective of root temperature.The net efflux of H + from the roots approximately balancedNH+4 uptake (1:1) over 14 d at each temperature and also balancedthe difference between the total uptake of inorganic cationsand inorganic anions. Under NO3 nutrition, the sum of the netefflux of OH and the change in the carboxylate contents of plantsover 14 d approximately balanced the sum of NO3 and SO4reduced in the plant. The majority of the negative charge associatedwith the reduction of NO3 and SO4 was apparently effluxedas OH, but this fraction was lower at low root temperatures.The results are discussed in terms of mechanisms that have beenproposed to regulate the internal pH of plants. The FV channel dominates the ion conductance of the vacuolar membrane at physiological Ca2+ concentrations. Patch-clamp measurements on whole barley (Hordeum vulgare) mesophyll vacuoles and on excised tonoplast patches showed small differences in a selectivity sequence NH4+ > K+ Rb+ Cs+ >Na+ >Li+. Less permeant cations decreased the open probability. The FV channel allows the uptake of small monovalent cations especially NH4+ into the vacuole.  相似文献

Smith, J. R. 1987. Potassium transport across the membranesof Chara. II. 42K fluxes and the electrical current as a functionof membrane voltage.—J. exp. Bot. 38: 752–777. The current required to clamp the trans-membrane voltage ofinternodal cells of Chara australis at different levels wasmeasured simultaneously with either the 42K influx or efflux.Examination of the voltage-dependence of the ratio of the electricalcurrent to the unidirectional tracer fluxes yielded no evidenceof any amplification of the electrical driving force on theK+ ions. There was thus no evidence for the interaction of K+ions with themselves or any other species during their passageacross the membrane. These measurements allow the determinationof , the fraction of the electrical current carried by K+ ions.When the external [K+] = 10 mol m–3, the average valueof was 0?85 for Vm > –125 mV and 07?5 for Vm <–150 mV. When the external [K+] = 0?1 mol m–3, was 0?6 for Vm < –80 mV and 0?1 for Vm > –250mV. It was also found that the conductance associated with K+transport was inhibited by hyperpolarization. The translocation of various anions, cations, and acids acrossthe membrane system of turnip mitchondria has been investigatedby light scattering techniques. The results imply that the membranesof turnip mitochondria are not permeable to choline, K+, Na+,, , , and . It has been established that Cl,, and SCN– passively permeate the mitochondrial membrane. It is probable that permeates as NH3 and that and cross the membrane either as the corresponding acid or on ananion/ hydroxide antiporter. The alkali metals Na+ and K+ equilibrateacross the membrane via specific H+/Na+ and H+/K+ antiporters,having similar activity at neutral pH.  相似文献

The commonly used equation, = P - + , which describes thepartitioning of plant water potential, , into components ofhydrostatic pressure, P, osmotic pressure, , and matric potential,, is misleading. The term , which is supposed to show the influenceof a solid phase on , is zero if a consistent definition ofpressure is used in the standard thermodynamic derivation. However,it can be usefully defined by = + D, where D is the osmoticpressure of the equilibrium dialysate of the system. Xyloglucan is a polysaccharide found in the primary cell wallsof all higher plants examined. Its cellulose-like backbone,which is about 0.15 to 1.5 µm long, consists of 300 to3 000 ß-(14)-linked D-glucopyranose residues. About60–75% (or, in grasses, about 30–40%) of the glucoseresidues have side-chains attached to position 6. The majorside-chains are: D-xylopyranosyl--1 -, D-galactopyranosyl-ß-(12)-D-xylopyranosyl--I , L-arabinofuranosyl-(1 -2)-D-xylopyranosyl--1-, and (except in grasses) L.-fucopyranosyl--(1 -2)-D-galactopyranosyl-ß-(1-2)-D-xylopyranosyl--1-. There is some regularity in the distribution of these side-chainsalong the backbone. Xyloglucan plays two very different r?les in the control ofcell growth: (a) as a major building material of the wall [concentrationof xyloglucan in the wall in vivo 10% (w/v)] it probably directlydictates wall extensibility and, therefore, the rate of cellexpansion and (b) it can be broken down to a fucose-containingoligosaccharide which [at a concentration of 0.0000001% (w/v)]exerts a hormone-like anti-auxin effect on growth. In addition,xyloglucan lacking fucose is used by certain dicotyledonousseeds as a food reserve which is mobilized after germination.Xyloglucan is, therefore, the subject of considerable currentinterest in several apparently disparate areas of botany. This work aimed to study the impacts of acquisition and assimilationof various nitrogen sources, i.e. NO3, NH4+ or NH4NO3,in combination with gaseous NH3 on plant growth and acid-basebalance in higher plants. Plants of C3 Triticum aestivum L.and C4 Zea mays L. grown with shoots in ambient air in hydroponicculture solutions with 2 mol m–3 of nitrogen source asNO3, NH4+ or NH4NO3 for 21 d and 18 d, respectively,had their shoots exposed either to 320 µg m–3 NH3or to ambient air for 7 d. Variations in plant growth (leaves,stubble and roots), and OH and H+ extrusions as wellas the relative increases in nitrogen, carbon and carboxylatewere determined. These data were computed as H+/N, H+/C, (C-A)/N,and (C-A)/C to analyse influences of different nitrogen sourceson acid-base balance in C3 Triticum aestivum and C4 Zea maysplants. Root growth in dry weight gain was significantly reduced bytreatment with 320 µg m–3 NH3 in Triticum aestivumand Zea mays growing with different N-forms, whereas leaf growthwas not significantly affected by NH3. In comparison with C3Triticum aestivum, non-fumigated C4 Zea mays had low ratiosof OH/N in NO3–3-grown plants and of H+/N in NH4+- and NH 4NO3-grown plants. Utilization of NH3 from the atmospherereduced both the OHN ratios in NO3 -grown plantsand the H+/N ratio in NH4+ - and NH4NO3 -grown plants of bothspecies. Furthermore, Zea mays had higher ratios of (C-A)/Nin NH4+ - and NH4NO3-grown plants than Triticum aestivum. Thismeans that C4 Zea mays had synthesized more organic anion perunit increase in organic N than C3 Triticum aestivum plants.Within both species, different nitrogen sources altered theratios of (C-A)/N in the order: NH4NO3>NH4+>NO3.Fumigation with NH3 increased organic acid synthesis in NO3- and NH4+ - grown plants of Triticum aestivum, whereas it decreasedorganic acid synthesis in Zea mays plants under the same conditions.Furthermore, these differences in acid-base regulation betweenC3 Triticum aestivum and C4 Zea mays plants growing with differentnitrogen sources are discussed. The euryhaline charophyte Lamprothamnium papulosum (Wallr.)J. Gr. was adapted to media with decreasing salinities rangingfrom 550 to 0 mosmol kg–1. Vegetative plants grown inmedia with osmotic pressures (0) in the range of 550 to 130mosmol kg–1 maintained a constant turgor pressure () at309 + 7 mosmol kg–1. The ions K+, Na+ and Cl–, werethe predominant solutes in the vacuole. Changes in their concentrationsaccount for the variation in internal osmotic pressure (1) with,0. The divalent ions Mg2+, Ca2+ and were also present in significant amounts, but their concentrationsdid not alter with changes in, 0. In cells subjected to hypo-osmotic shock the regulation of was incomplete. The turgor pressure increased from 302 to 383mosmol kg–1. The first rapid response to the sudden decreasein 0 was a loss of K+ and Cl. In contrast to the decreasein ionic concentrations an accumulation of sucrose occurredwhich could account for the increase of . The increase in sucroseconcentration started 24 to 48 h after the downshock and reachedits highest value after 3 to 4 weeks. The sucrose concentrationin the vacuole was up to 320 mol m–3. During this timethe ionic content continued to decrease but did not counterbalancethe sucrose concentration sufficiently to regain the original. High sucrose levels accompanied by an enhanced were also observedduring the period of fructification (sexual reproduction: formationof antheridia and oogonia) in Lamprothamnium kept under conditionsof constant salinity. It is concluded that high sucrose content and elevated arecharacteristic of sexual reproduction in this charophyte. The daily cycle of the transpiration rate, stomatal conductance,water potential, and the concentration of the main osmoticumidentified in ash leaves, malate and mannitol, were monitoredin a field on the Isere river plain. On sunny days, the stomatalconductance tends to remain close to its maximun value allowinga high transpiration rate and diurnal variations in leaf waterpotential, w, which may fall as low as –2 MPa at solarnoon. These variations of w are closely correlated with changesin malate, mannitol and the concentration of the well-knownosmoticum K+, which agree with the involvement of an osmoticadjustment to counteract the evaporative demand during daylighthours. How malate, mannitol and K+ contribute to the osmoticadjustment was analysed subsequently by comparing the solutepotential s, evaluated by the Boyle-Van't Hoff relation, tothe osmotic potential measured by thermocouple psychrometry.These experiments have led us to suspect some errors in themeasurement of , presumably due to experimental artefacts andthe ability of Ca 2+ , present in high levels in leaves, toform chelates with malate once the cells have been decompartmentedby freezing and thawing. Since significant changes of Ca2+ alsooccurred during the diurnal variations of w, the possible mechanismsby which Ca2+ may be implicated in controlling the water statusof the tree are discussed. The dominating mechanism of K+ uniport through the plasmalemma of Coscinodiscus wailesii has been studied in some detail as part of a general study of ionic relations in marine diatoms. Electrical measurements with double-barrelled glass-microelectrodes have been made in intact cells (diameter 100 m) bathed in artificial sea-water in which [K+] has been changed from 3 mM to 100 mM. Using a modified Goldman equation, these results provide an estimate of [K+]i of about 400 mM and a selectivity for K+ over Na+ and CI-, which could spontaneously vary by orders of magnitude and reach values of about 1000. Voltage-clamp experiments have been carried out in these states of high K+ selectivity using bipolar staircase command voltages over a range from -180 to +60 mV. The resulting steady-state current-voltage relationships have inward rectifying sigmoid characteristics with a negative saturation current around -30 nA, and a slope conductance of the order or 1 S at free running voltages <-60 mV. Temporal responses of the clamp currents upon rectangular voltage steps were basically rectangular, i.e. they did not show the familiar relaxation kinetics of voltage-induced activation/inactivation. The sigmoid steady-state current-voltage relationships could not be described by a usual model of constant-field currents through a voltage-gated pore, where the positive current of an inward rectifier would not saturate but vanish. Alternatively, the observed steady-state inward rectifying current-voltage relationship and its changes upon changes in [K+]o, are well described by a three-state reaction cycle for catalysis of K+ translocation with a steady activity.  相似文献

The possible interaction of two stresses, UV-B radiation and cadmium, applied simultaneously, was investigated in Brassica napus L. cv. Paroll with respect of chlorophyll fluorescence, growth and uptake of selected elements. Plants were grown in nutrient solution containing CdCl2, (0, 0.5, 2 or 5 M) and irradiated with photosynthetically active radiation (PAR, 400-700 nm, 800 mol m-2 s-1) with or without supplemental ultraviolet-B radiation (UV-B, 280-320 nm, 15 kJ m-2 d-1, weighted irradiance). After 14 d of treatment, the most pronounced effects were found at 2 and 5 M CdCl2 with and without supplemental UV-B radiation. Exposure to cadmium significantly increased the amount of Cd in both roots and shoots. In addition, increases occurred in the concentrations of Fe, Zn, Cu, and P in roots, while K was reduced. In shoots the S content rose significantly both in the presence and absence of UV-B radiation, while significant increases in Mg, Ca, P, Cu, and K occurred only in plants exposed to Cd and UV-B radiation. Manganese decreased significantly under the combined exposure treatment. The rise in S content may have been due to stimulated glutathione and phytochelatin synthesis. Cadmium exposure significantly decreased root dry weight, leaf area, total chlorophyll content, carotenoid content, and the photochemical quantum yield of photosynthesis. As an estimation of energy dissipation processes in photosynthesis, non-photochemical quenching (qNPQ) was measured using a pulse amplitude modulated fluorometer. The qNPQ increased with increasing Cd, while the combination of cadmium and UV-B reduced the qNPQ compared to that in plants exposed only to cadmium or UV-B radiation. The chlorophyll a:b ratio showed a reduction with UV-B at no or low Cd concentrations (0 M, 0.5 M CdCl2), but not at the higher Cd concentrations used (2 M, 5 M CdCl2). Thus in some instances there appeared to be a UV-B and Cd interaction, while in other plants response could be attributed to either treatment alone.Keywords: Brassica napus, cadmium, ultraviolet-B radiation.   相似文献

Fidgeon, C. and Wilson, G. 1988. Uptake and accumulation ofa-naphthalene acetic acid by cell suspensions of Galium mollugoL.—J. exp. Bot. 39: 241-249. Galium mollugo cell suspensions require -NAA for continued growthand cell division. The kinetics of -NAA uptake from the medium(B5) by Galium cells was assessed using 1-14C -NAA in a standardratio of cells to medium (0.25 g: 2.5 cm3). It was found thatthe uptake of -NAA was rapid, over 90% being taken up within4 h. Cells which had accumulated -NAA for 4 h or more did notrelease it back into the medium. It was found that Galium cellsaccumulated -NAA against a significant concentration gradient;suggesting the participation of an active component in the uptakemechanism. The effect of free-space and surface adsorption onthe uptake of -NAA was determined by means of a repeated washtechnique. These two factors were found to be of importanceonly during the first hour of uptake. Neither dead cells norplasmolysed cells absorbed -NAA. It is clear that, in the normal growth cycle, Galium cells cantake up the available -NAA within 3 or 4 h of inoculation andthat this can stimulate a cell division response of 3-4 generationsover the subsequent 14 d. An error occurs in the calibration of xylem pressure potential() against leaf-water potential () when the calibration is madeusing plant material in which the water stress has been inducedartificially after excision. In recent years alternative ways have been proposed to transformmeasurements of leaf water potential, , and relative water content,R*, in order to derive values of osmotic pressure at full turgidityin leaves and shoots, o(when 0). Two types of transformationsare usually considered: 1/ versus R* and versus 1/R*, and linearregression is used to fit the data in the region where turgoris thought to be zero. It appears that when o is estimated bylinear extrapolation of 1/Psi; versus R* then apoplastic watermight not influence the accuracy of o but when the versus \/R*transformation is used apoplastic water causes an underestimateof o. We examine the accuracy of the estimate of o obtainedfrom the two transformations when there are random errors in, systematic errors in , and when the osmotic solutions arenon-ideal. The 1/ versus R* transformation generally producesthe best estimate of 0 by linear extrapolation.  相似文献

Changes in components of leaf water potential during soil waterdeficits influence many physiological processes. Research resultsfocusing on these changes during desiccation of peanut (Arachishypogeae L.) leaves are apparently not available. The presentstudy was conducted to examine the relationships of leaf waterl, solute s and turgor p potentials, and percent relative watercontent (RWC) of peanut leaves during desiccation of detachedleaves and also during naturally occurring soil moisture deficitsin the field. The relationship of p to l and RWC was evaluated by calculatingp from differences in l and s determined by thermocouple psychrometryand by constructing pressure-volume (P-V) curves from the land RWC measurements. Turgor potentials of ‘Early Bunch’and ‘Florunner’ leaves decreased to zero at l of–1.2 to –1.3 MPa and RWC of 87%. There were no cultivardifferences in the l at which p became zero. P-V curves indicatedthat the error of measuring s after freezing due to dilutionof the cellular constituents was small but resulted in artefactualnegative p values. Random measurements on two dates of l, s, and calculation ofp from well-watered and water-stressed field plots consistingof several genotypes indicated that zero p occurred at l of–1.6 MPa. It was concluded that the relationships of p,l, s, and RWC of peanut leaves were similar to leaves of othercrops and that these relationships conferred no unique droughtresistance mechanism to peanut.  相似文献

The natural 15N abundance (15N value) in acetylpropyl derivatives of amino acids and in ethyloxycarbonyl derivatives of polyamines was determined using a gas chromatography/combustion/mass spectrometer-(GC/C/MS). 15N value determined for 12 amino acids and five polyamines by GC/C/MS were identical to those obtained by a direct combustion method using an automatic nitrogen and carbon analysis (ANCA) mass spectrometer, the difference being less than 1.0% in most cases. The GC/C/MS method was used to analyse 15N values in the amino acids and polyamines from root nodules of pea and faba bean and from stem nodules of Sesbania rostrata. The analysis of 15N values revealed that homospermidine had high 15N values, as much as +40%, while the amino acids investigated had 15N values between -3 and +6%, putrescine between +2 and +8%, cadaverine between +1 and +7%, spermidine between -2 and +4%, and spermine between 0 and +6%. The mechanism of 15N enrichment in homospermidine is discussed.  相似文献

With the aim of determining the level at which ageing exerts its effect on the expression of -amylase, GA3 regulation of -amylase production was studied in isolated aleurone layers from aged wheat seeds. GA3-induced -amylase activity was lower in the tissue from aged seeds than in controls. However, the proportion of 35S-methionine incorporated into -amylase was higher in the aged than in control tissue. The pattern of -amylase isoforms was resolved by isoelectric focusing and showed that two isogroups were present with the activity of the high-pI isogroup being higher in the control than in the aged lot. These apparently contradictory results may be explained in terms of differences in isozyme expression. Studies on the expression of -amylase genes indicated a reduction in the level of high-pI mRNA in aged tissue. Dose-response curves showed lower GA3-responsiveness of aleurone layers from aged seeds as compared to the controls. From these results, it is proposed that the diminished capacity of -amylase production in aleurone from aged seeds is apparently due to a decrease in the expression of the high-pI -amylase genes, and this reduction is associated with a decrease in the response to GA3.Key words: Seed ageing, wheat aleurone, gibberellic acid, -amylase isozymes, gene expression.   相似文献

An equation is derived expressing average turgor pressure ofa leaf (p) as a function of relative water content (RWC). Basedon this derivation, the relationships of the bulk elastic modulus(v) and both RWC and p, are formulated and discussed. The bulkelastic modulus (v) becomes zero for p = 0, that is at the turgorloss point for the leaf. At full water saturation the valueof ev is proportional to the water saturation turgor potentialp(max). The factor relating P and v (structure coefficient ,Burstrom, Uhrstr?m and Olausson, 1970) changes only very littlefor values of p, which are not too close to zero. An exampleis given for the calculation from experimental data of the turgorpressure function, the structure coefficient function, and thev function.
