Eight positional isomers of 61,6m-di-O-α-d-mannopyranosyl-cyclomaltooctaose (γCD) (m=2–5) and 6-O-α-(n-O-α-d-mannopyranosyl)-d-mannopyranosyl-γCD (n=2, 3, 4, and 6) in a mixture of products from γCD and d-mannose by condensation reaction of α-mannosidase from jack bean were isolated by HPLC. The structures of four isomers of 6-O-α-(n-O-α-d-mannopyranosyl)-d-mannopyranosyl-γCD were elucidated by NMR spectroscopy. On the other hand, four positional isomers of 61,6m-di-O-α-d-mannopyranosyl-γCD were determined by LC–MS analysis of degree of polymerization of the branched oligosaccharides produced by enzymatic degradation with bacterial saccharifying α-amylase (BSA), and combination of BSA and glucoamylase. Similarly cyclomaltodextrin glucanotransferase also digested these isomers.  相似文献

The reactions of Fe(CO)3(R-DAB; R1, H(4e)) (1a: R = i-Pr, R1 = H; 1b: R = t-Bu, R1 = H; 1c: R = c-Hex, R1 = H; 1e: R = p-Tol, R1 = H; 1f: R = i-Pr, R1 = Me) with Ru3(CO)12 and of Ru(CO)3(R-DAB; R1, H(4e)) (2a: R = i-Pr, R1 = H; 2d: R = CH(i-Pr)2, R1 = H) with Fe2(CO)9 in refluxing heptane both afforded FeRu(CO)6(R-DAB; R1, H(6e)) (3) in yields between 50 and 65%.The coordination mode of the ligand has been studied by a single crystal X-ray structure determination of FeRu(CO)6(i-Pr-DAB(6e)) (3a). Crystals of 3a are monoclinic, space group P21/a, with four molecules in a unit cell of dimensions: a = 22.436(3), b = 8.136(3), c = 10.266(1) Å and β = 99.57(1)°. The structure was refined to R = 0.049 and Rw = 0.052 using 3045 reflections above the 2.5σ(I) level. The molecule contains an FeRu bond of 2.6602(9) Å, three terminally bonded carbonyls to Fe, three terminally bonded carbonyls to Ru and bridging 6e donating i-Pr-DAB ligand. The i-Pr-DAB ligand is coordinated to Ru via N(1) and N(2) occupying an apical and equatorial site respectively (RuN(1) = 2.138(4) RuN(2) = 2.102(3) Å). The C(2)N(2) moiety of the ligand is η2-coordinated to Fe with C(2) in an apical and N(2) in an equatorial site (FeC(2) = 2.070(5) and FeN(2) = 1.942(3) Å).The 1H and 13C NMR data indicate that in all FeRu(CO)6(R-DAB(6e)) complexes (3a to 3f) exclusively η2-CN coordination to the Fe atom and not to the Ru atom is present irrespective of whether 3 was prepared by reaction of Fe(CO)3(R-DAB(4e)) (1) with Ru3(CO)12 or by reaction of Ru(CO)3(R-DAB(4e)) (2) with Fe2(CO)9. In the case of FeRu(CO)6(i-Pr-DAB; Me, H(6e)) (3f) the NMR data show that only the complex with the C(Me)N moiety of the ligand σ-N coordinated to the Ru atom and the C(H)N moiety η2-coordinated to the Fe atom was formed. Variable temperature NMR experiments up to 140 °C showed that the α-diimine ligand in 3a is stereochemically rigid bonded.FeRu(CO)6(R-DAB(6e)) (3a and 3e) reacted with allene to give FeRu(CO)5(R-DAB(4e))(C3H4) (4a and 4e). A single crystal X-ray structure determination of FeRu(CO)5(i-Pr-DAB(4e))(C3H4) (4a) was performed. Crystals of 4a are triclinic, space group P1, with two molecules in a unit cell of dimensions: a = 9.7882(7), b = 12.2609(9), c = 8.3343(7) Å, α = 99.77(1)°, β = 91.47(1)° and γ = 86.00(1)°. The structure was refined to R = 0.028 and Rw = 0.043 using 4598 reflections above the 2σ(I) level. The molecule contains an FeRu bond of 2.7405(7) Å and three terminally bonded carbonyls to iron. Two carbonyls are terminally bonded to the Ru atom together with a chelating 4e donating i-Pr-DAB ligand [RuN = 2.110(1) (mean)]. The allene ligand is coordinated in an η3-allylic fashion to the Fe atom while the central carbon of the allene moiety is σ-bonded to the Ru atom (FeC(14) = 2.166(3), FeC(15) = 1.970(2), FeC(16) = 2.127(3) and RuC(15) = 2.075(2) Å). The 1H and 13C NMR data show that in solution the coordination modes of the R-DAB and the allene ligands are the same as in the solid state.Thermolysis reactions of 3a with R-DAB or carbodiimides gave decomposition and did not afford C(imine)C(reactant) coupling products. Thermolysis reactions of 3a with M3(CO)12 (M = Ru, Os) and Me3NO gave decomposition. When the reaction of 3a with Me3NO was performed in the presence of dimethylacetylenedicarboxylate (DMADC) the known complex FeRu(CO)4(i-Pr-DAB(8e))(DMADC) (5a) was formed in low yield. In 5a the R-DAB ligand is in the 8e coordination mode with both the imine bonds η2-coordinated to iron. The acetylene ligand is coordinated in a bridging fashion, parallel with the FeRu bond.  相似文献

In the central nervous system, three enzymes belonging to the serine hydrolase family are thought to regulate the life time of the endocannabinoid 2-arachidonoylglycerol (C20:4) (2-AG). From these, monoacylglycerol lipase (MAGL) is well characterized and, on a quantitative basis, is the main 2-AG hydrolase. The postgenomic proteins α/β-hydrolase domain containing (ABHD)6 and ABHD12 remain poorly characterized. By applying a sensitive fluorescent glycerol assay, we delineate the substrate preferences of human ABHD6 and ABHD12 in comparison with MAGL. We show that the three hydrolases are genuine MAG lipases; medium-chain saturated MAGs were the best substrates for hABHD6 and hMAGL, whereas hABHD12 preferred the 1 (3)- and 2-isomers of arachidonoylglycerol. Site-directed mutagenesis of the amino acid residues forming the postulated catalytic triad (ABHD6: S148-D278-H306, ABHD12: S246-D333-H372) abolished enzymatic activity as well as labeling with the active site serine-directed fluorophosphonate probe TAMRA-FP. However, the role of D278 and H306 as residues of the catalytic core of ABHD6 could not be verified because none of the mutants showed detectable expression. Inhibitor profiling revealed striking potency differences between hABHD6 and hABHD12, a finding that, when combined with the substrate profiling data, should facilitate further efforts toward the design of potent and selective inhibitors, especially those targeting hABHD12, which currently lacks such inhibitors.  相似文献

6(I),6(IV)-Di-O-[α-l-fucopyranosyl-(1→6)-2-acetamido-2-deoxy-β-d-glucopyranosyl]-cyclomaltoheptaose (βCD) {6(I),6(IV)-di-O-[α-l-Fuc-(1→6)-β-d-GlcNAc]-βCD (5)} and 6-O-[α-l-fucopyranosyl-(1→6)-2-acetamido-2-deoxy-β-d-glucopyranosyl]-βCD {6-O-[α-l-Fuc-(1→6)-β-d-GlcNAc]-βCD (6)} were chemically synthesized using the corresponding authentic compounds, bis(2,3-di-O-acetyl)-pentakis(2,3,6-tri-O-acetyl)-βCD as the glycosyl acceptor and 2,3,4-tri-O-benzyl-α-l-fucopyranosyl-(1→6)-3,4-di-O-acetyl-2-deoxy-2-(2,2,2-trichloroethoxycarbonylamino)-d-glucopyranosyl trichloroacetimidate as the fuco-glucosaminyl donor. NMR confirmed that α-l-Fuc-(1→6)-d-GlcNAc was bonded by β-linking to the βCD ring. To evaluate biological efficiency, the biological activities of the new branched βCDs were examined. The cell detachment activity of 5 was lower than that of 6 in real-time cell sensing (RT-CES) assay, indicating that 5 has lower toxicity. In SPR analysis, 5 had a higher special binding with AAL, a fucose-recognizing lectin. These results suggest that 5 could be an efficient drug carrier directed at cells expressing fucose-binding proteins.  相似文献

The synthesis of new zirconium and hafnium mixed ligand phthalocyanine complexes PcM(β-ketoester)2, where M-Zr (IV), Hf (IV); Pc - the dianion of phthalocyanine, and β-ketoester - the out planed ligand, is reported. The obtained complexes are characterized by 1H NMR, IR, UV-Vis spectroscopy and cyclic voltammetry. 1H NMR and elemental analysis confirm the substitution of two Cl atoms for two β-ketoester fragments to the central atom of the macrocycle. The data of 1H NMR, UV-Vis spectroscopy have allowed us to conclude that two β-ketoester ligands are in the cis geometry to the phthalocyanine plane. X-ray crystallography for bis(isopropyl 3-oxobutanoato)hafnium(IV)phthalocyanine confirms this conclusion. The central macrocycle of the phthalocyanine ligand is not exactly planar (deviations from the least-square plane exceed 0.15 Å) and has the conformation of an essentially flattened crown. The Hf(1) atom is 1.349(3) above this least-square plane. Cyclic voltammetry investigation shows that the introduction of two β-ketoester ligands to the central atom of phthalocyanine complex leads to both chemical and electrochemical stabilization of the whole Pc system.  相似文献

Two routes for the preparation of the title compound (6) have been developed. Reaction of equimolar quantities of 2-aminobenzaldehyde, pentane- 2,4-dione and 1,2-diaminoethane yields the ligand 6 and 2-methyl-3-acetylquinoline as side product. The compound 6 was obtained in high yield in a one- step condensation of 2-aminobenzaldehyde with 1-amino-4-methyl-3-azahept-4-ene-6-one (5). Studies on the condensation of 5 with various 2-aminobenz- aldehyde derivatives revealed that the yield of unsymmetrical ligand is evidently influenced by the acidic properties of the hydrogen atom (of atoms) of the ring substituent in the ortho position to the carbonyl group. Copper(II), nickel(II) and palladium(II) complexes of 6 have been prepared and characterized. Spectroscopic data of nickel and palladium complexes are consistent with their planar structure. A superhyperfine splitting due to nitrogen is observed in the EPR spectrum of the copper complex in spite of the presence of azomethine hydrogen atom.  相似文献

The mRNA levels of neuropoietic cytokines, ciliary neurotrophic factor (CNTF), leukemia inhibitory factor (LIF), and interleukin-6 (IL-6), and their receptor components (CNTFR, LIFR, IL-6R, and gp130) were examined in seventy-six patients with various peripheral neuropathies to determine the extent of expression of these cytokines and receptors, and their relationship to nerve fiber pathology and cell infiltration in the diseased nerves. The CNTF mRNA levels were significantly decreased in the diseased nerves and were correlated to residual myelinated fiber population. In contrast, the mRNA levels of LIF, IL-6 and the ligand-binding receptor components (CNTFR, LIFR and IL-6R) were elevated to variable extent in the diseased nerves. The CNTFR, LIFR, and IL-6R mRNA levels showed a weak positive correlation with the extent of demyelinating pathology and their levels were related to each other. Moreover, the CNTF and LIF mRNA levels were inversely proportional to the extent of macrophage invasion, whereas the CNTFR and IL-6R mRNA expressions were correlated to the increase in macrophage infiltration. The neuropoietic cytokine family and its receptor expressions in the diseased human nerves are regulated by an underlying pathology-related process rather than type of diseases, and could play a role in peripheral nerve regeneration and repair.  相似文献

The number of species in the freshwater mussel genus Cristaria Schumacher, 1817 recognized from Far East Russia has varied over the last several decades. While some authors consider the occurrence of only one species, Cristaria plicata (Leach, 1815), widespread in East Asia, others, recognize two separate species Cristaria herculea (Middendorff, 1847) and Cristaria tuberculata Schumacher, 1817 from Far East Russia, distinct from C. plicata. For the present study, freshwater mussels, identified as C. herculea, were collected in the Upper Amur basin (Transbaikalia, Russia). The shell morphology and the whole soft body anatomy were analysed in detail and compared with previously published information on other Cristaria spp.. Additionally, a cytochrome oxidase subunit 1 (CO1) gene fragment was sequenced from foot tissue samples of selected animals, collected from the same region, and compared with published data. Based upon morphological similarities of glochidia and adult morphology and anatomy as well as the mitochondrial DNA sequence analysis, we consider C. herculea as a synonym of C. plicata. Further analysis of Far East Russia C. herculea and C. tuberculata specimens using both molecular and morphological characters should be carried in the future to enhance our knowledge about the taxonomy within the Cristaria genus. Moreover, a comprehensive revision of the genus Cristaria is needed, restricting the type locality and comparing topotypic specimens for both C. plicata and C. tuberculata, and including all recognized Cristaria species.  相似文献

The levels of expression of Gsα(short and long), Gα(olf) and Gβ(common) subunits, and calcium-sensitive adenylyl cyclases isoforms (AC1, 5/6, and 8) in human brain cortical and caudate membranes were quantified by western blot analysis in order to establish their contribution to the patterns of AC functioning. Both areas expressed Gsα(long) (52 kDa) with values ranging from about 1400 ng/mg of membrane protein in cerebral cortex to close to 600 ng/mg of membrane protein in caudate nucleus. In contrast, Gsα(short) and Gsα(olf) were expressed separately, Gsα(short) in cortical membranes with values around 500 ng/mg of membrane protein and Gα(olf) in caudate membranes with values around 1300 ng/mg of membrane protein. Quantitative measurements of Gβ, revealed a similar expression level in cortical and caudate membranes (5444±732 versus 5511±394 ng/mg protein; p=0.966). The B(max) values of GTPγS-dependent [(3)H]-forskolin binding show the following descending order: rat striatal membranes>rat cortical membranes=human caudate membranes>human cortical membranes. Therefore, as measured immunochemically and by [(3)H]-forskolin binding, there seems to be a vast excess of Gsα subunits over catalytic units of AC. The highest levels of AC5/6 expression were detected in caudate membranes. AC8 was little expressed, and there were no significant differences in the relative values between both human brain regions. Finally, the levels of the AC1 isoform were significantly lower in caudate than in cortical membranes. It is concluded that these stoichiometric data contribute nonetheless to explain the significant differences observed in signalling capacities through the AC system in both human brain regions.  相似文献

Cogongrass is considered to be one of the ten most troublesome and problematic weedy species in the world. This species is found throughout tropical and subtropical regions, generally in areas disturbed by human activities. Over 100 common names have been associated with cogongrass, including japgrass, speargrass, alang-alang, and bladygrass. Although this species has several commercial uses, the problems associated with its weediness far outweigh most positive benefits. Cogongrass is a major impediment to reforestation efforts in southeast Asia, the number one weed in agronomic and vegetable production in many parts of Africa, and is responsible for thousands of hectares of lost native habitat in the southeastern U.S. Biologically, cogongrass possesses several features that foster its spread and persistence. Management efforts for cogongrass consist of an integrated approach with several control strategies. In agronomic production, the use of cover crops is widely successful, but incorporation into the overall production scheme is challenging. Success has been achieved with continuous deep tillage or chemical applications, but long-term eradication/suppression must employ sustainable revegetation strategies.  相似文献

Synthesis of complexes cis,cis-WVOXL (X=Cl, NCS), cis,trans-WVOXL (X=Cl, OPh, SPh) and cis,trans-WVIE2L (E2=O2, OS, S2) of the title ligand LH2 are reported. cis,cis-WVOCIL crystallises in space group P21/c with a=13.6541(9) Å, b=7.1555(11) Å, c=18.198(2) Å, β=95.294(6)°, V=1770.4(3) Å3 and Z=4 while the cis,trans isomer crystallises in space group P21/n with a=10.361(3) Å, b=14.141(4) Å, c=12.213(5) Å, β=102.56(3)°, V=1747(2) Å3 and Z=4. cis,trans-WVIS2L crystallises in space group P21/n with a=10.645(2) Å, b=13.929(2) Å, c=12.189(2) Å, β=103.14(2)°, V=1760(1) Å3 and Z=4. A short CH3···Cl distance of 3.067(7) Å and an acute OWCl angle of 94.1(2)° are seen in cis,cis-WVOClL, which converts to the cis,trans form on heating in MeCN. The latter isomer features a CH3···Cl distance of 3.38(2) Å and an OWCl angle of 105.1(8)°. Electrochemical and EPR data are reported. In particular, cis,trans-WVIE2L may be reduced to [WVE2L]. EPR properties of these anions and those of complexes WVOXL are discussed in the context of WV centres in tungsten enzymes.  相似文献

This computational study is intended to shed light on the crystalline and molecular structure, together with the hydrogen bonding (H-bonding) differences between two forms of native cellulose. DFT calculations were carried out to characterize the 17O, 1H and 13C nuclear magnetic resonance (NMR) parameters in cellulose Iα and Iβ with the B3LYP functional employing the 6–311++G7 and 6–31+G1 basis sets. Geometry optimization revealed that the average HB length is shortened by 0.01–0.08 Å when the chains are aligned, whereas the average bond angle increases by about 4–8° exhibiting the enhancement of HB strength. For the isolated cellotetramer chains, the isotropic 17O–H chemical shifts were plotted as a function of HB length. Our results indicated that as the HB length in cellotetramer Iα increases, the 17O–H chemical shift isotropy increases, but this parameter changes in the opposite direction for the other structure. Moreover, B3LYP/6–311++G7 calculations reveal that there is an acceptable correlation between the calculated 13C chemical shifts of the two structures and their experimental values.  相似文献

  • 1.1. Metabolic rates (ml O2/mg/hr) of three geographically separated populations of the carabid beetle Calathus melanocephalus L. (Finse and Je 10y, Norway and Drenthe, The Netherlands) were measured and compared by ANCOVA.
  • 2.2. No significant relationship (P > 0.05) between metabolic rates and body weight or sex of the animals were found.
  • 3.3. Individuals mostly acclimated to low temperatures by increased metabolic rates and in the opposite direction to higher temperatures. Individuals collected in early summer also showed higher metabolic rates than those caught later in the autumn.
  • 4.4. Contradicting the theory of metabolic cold adaptation, beetles from The Netherlands had the highest metabolic rates, beetles from Finse intermediate rates and beetles from Jeløy the lowest rates.
  • 5.5. No significant relation were found between geographical origin of the beetles and their respective chill-coma temperature.
