Velvet bean (Mucuna pruriens L.) has been found that the degree of suppression on the lettuce root growth by velvet bean was less on the 3D-clinorotation. The number and growth of adventitious root in velvet bean differed among the clinostated and control group. L-DOPA (L-3,4-dihydroxyphenylalanine) is known to be the major substance in the allelopathy of velvet bean plant, released from its root. Since L-DOPA is a precursor of melanin pigment, and is easily converted to melanin by oxidation, locality of L-DOPA production in the plant body can be seen through pigmentation. The amount of L-DOPA was analyzed by HPLC and LC-ESI/MS. The distribution of L-DOPA in the root was different among the ground control condition and pseudo-microgravity.  相似文献

Phaseolus vulgaris L. cv. Kinghorn Wax seedlings, supplied with nutrient solution containing either 0 or 5 mM nitrate as sole N source, were exposed to 0.25 μl/l NO2 for 6 hr each day for 10 days at continuous photosynthetic photon flux (PPF) of 100, 300, 500 or 700 μmol m−2 sec−1. There was a significant interaction of PPF and nitrate. Shoot and root dry weights increased with increasing PPFs only when nitrate was supplied. The main effects of NO2 on plant growth were significant; none of the interactions involving NO2 were significant. Exposure to NO2 decreased shoot and root dry weight in both the presence and absence of nutrient N and at all PPF levels. All interactions were significant for in vitro leaf nitrate reductase activity (NRA), which increased markedly at PPFs above 100 μmol m−2 sec−1 when nitrate was supplied. Treatment with NO2 strongly inhibited enzyme activity in the presence of nitrate, particularly at the 300 μmol m−2 sec−1 PPF level. These experiments demonstrated that PPF level does not modify the effect of NO2 on growth but does have a major effect on NRA and on NO2 effects on NRA in the presence of nutrient nitrate.  相似文献

对黄土区核桃(Juglans regia L.)-(玉米+大豆)、李子(Prunus salicina)-(玉米+大豆)农林复合系统小气候效应的研究表明,在玉米和大豆的花期,农林间作系统与对照地相比可以降低农田地面温度1.7-1.9℃,减低风速55%-67%,提高相对湿度6.9%-8.4%,降低光合有效辐射强度13.9%-24.5%和大气CO2浓度5.3-10.9μmol.mol-1。本研究中,单作大豆叶片光合速率午间变化主要受到非气孔的限制作用。逐步回归分析结果表明,光合有效辐射强度和大气CO2浓度是影响林下作物光合速率的主要环境因子。林下小气候可能是作物避免"午休"现象的主要原因。  相似文献

Photosynthetic rates and photosynthate partitioning were studied in three-week-old soybean [Glycine max (L.) Merr. cv. Williams] plants exposed to either ambient (35 Pa) or elevated (70 Pa) CO2 in controlled environment chambers. Ambient CO2-grown plants also were given a single 24 h treatment with 70 Pa CO2 1 d prior to sampling. Photosynthetic rates of ambient CO2-grown plants initially increased 36% when the measurement CO2 was doubled from 35 to 70 Pa. Photosynthetic rates of the third trifoliolate leaf, both after 1 and 21 d of elevated CO2 treatment, were 30 to 45% below those of ambient CO2-grown plants when measured at 35 Pa CO2. These reduced photosynthetic rates were not due to increased stomatal resistance and were observed for 2 to 8 h after plants given 1 d of CO2 enrichment were returned to ambient CO2. Initial and total ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) activities, percent activation, Rubisco protein, soluble protein and leaf chlorophyll content were similar in all CO2 treatments. Quantum yields of photosynthesis, determined at limiting irradiances and at 35 Pa CO2, were 0.049±0.003 and 0.038±0.005 mol CO2 fixed per mol quanta for ambient and elevated CO2-grown plants, respectively (p<0.05). Leaf starch and sucrose levels were greater in plants grown at 70 than at 35 Pa CO2. Starch accumulation rates during the day were greater in ambient CO2-grown plants than in plants exposed to elevated CO2 for either 1 or 21 d. However, the percentage of C partitioned to starch relative to total C fixed was unaffected by 1 d of CO2 enrichment. The above results showed that both photosynthetic and starch accumulation rates of soybean leaflets measured at 35 Pa CO2 were temporarily reduced after 1 and 21 d of CO2 enrichment. The soybean aphid, Aphis glycines Matsumara, was discovered in the United States in the summer of 2000. Since that initial discovery, the aphid has spread across northern soybean production regions. In 2001, we examined the physiological responses of soybeans to low aphids densities (fewer than 50 aphids/leaf). In this study, we determined photosynthetic rates, leaf fluorescence responses, and photosynthetic responses to variable carbon dioxide and light levels. In addition, analyses for chlorophyll content and stable carbon isotope ratios were used to differentiate potential differences in stomatal versus mesophyll limitations to photosynthesis. We observed rate reductions of up to 50% on infested leaflets, including lealets with no apparent symptoms of aphid injury (such as chlorosis). Differences in fluorescence data indicated that photoelectron transport was not impaired. These results indicate that substantial physiological impact on soybean is possible even at low aphid densities. Also, the conventional view of aphid injury acting through reductions in chlorophyll content and light-harvesting reactions of photosynthesis is not supported by our findings in this system.  相似文献

Salicylic acid (SA) and related phenolic compounds can exert control over stomatal opening and previous work in our laboratory has shown that chronic injection of SA increases the photosynthetic rate of corn. The work reported in this paper investigated the effects of foliar applied SA, acetyl salicylic acid (ASA) and gentisic acid (GTA) on photosynthetic rates and growth of soybean (a C3 plant) and corn (a C4 plant) under greenhouse conditions. In general, the tested compounds enhanced photosynthetic rates in both soybean and corn. Stomatal conductance and transpiration were also increased. These compounds do not alter chlorophyll content. In some cases treatment with these compounds resulted in increased leaf areas and plant dry mass, however, plant height and root length were not affected.  相似文献

The effect of certain simple and cost-effective processing methods on the nutritional and anti-nutritional properties of seed materials of an under-utilized food legume, Mucuna pruriens (L.) DC. var. utilis (Wall. ex Wight) Baker ex Burck (velvet bean, VB), collected from Valanadu, Kerala, India was analyzed in experiment 1. The raw VB seeds were found to contain appreciable levels of crude protein (263.2 g/kg dry matter (DM)); ether extract (79.6 g/kg DM); crude fiber (95.8 g/kg DM) and ash content (38.4 g/kg DM). Among the different treatments used, soaking in sodium bicarbonate solution + autoclaving was more effective in reducing maximum levels of various anti-nutritional compounds of VB seeds. Furthermore, in experiment 2, the effect of inclusion of different levels of velvet bean meal (VBM; subjected to soaking in sodium bicarbonate solution + autoclaving) as an alternative protein source in poultry feed on the growth performance of commercial-type broiler birds was investigated. The results indicate that the inclusion of VBM up to the 40% level exhibited better growth performance of the broiler birds such as feed intake, body weight gain, feed conversion ratio and protein efficiency ratio in both the starter and finisher phases.  相似文献

The plant rhizosphere is an important soil ecological environment for plant-microorganism interactions, which include colonization by a variety of microorganisms in and around the roots that may result in symbiotic, endophytic, associative, or parasitic relationships within the plant, depending on the type of microorganisms, soil nutrient status, and soil environment. Rhizosphere competence may be attributable to the differences in the extent of bacterial attachment to the root surface. We present results of the effect of various factors on the attachment to bean (Phaseolus vulgaris) and soybean (Glycine max) roots of some bacterial species of agronomic importance, such as Rhizobium tropici, Rhizobium etli, Ensifer fredii (homotypic synonym Sinorhizobium fredii), and Azospirillum brasilense; as well as the attachment capability of the plant growth promoting rhizobacteria Pseudomonas fluorescens and Chryseobacterium balustinum. Additionally, we have studied various bacterial traits, such as autoaggregation and flagella movements, which have been postulated to be important properties for bacterial adhesion to surfaces. The lack of mutual incompatibility between rhizobial strains and C. balustinum has been demonstrated in coinoculation assays.  相似文献

Constitutive (i.e., preformed) parameters of Mexican bean beetle (MBB) resistance were demonstrated in PI 227687 soybeans grown under highly controlled and reproducible biotron environmental conditions which favored optimal plant growth and development. Constitutive antifeedants in PI 227687 against the beetle were variously soluble in hexane, ethyl acetate and 100% methanol. Antifeedants also remained in the aqueous residue after the sequential organic extractions. PI 227687 and MBB-susceptible Davis soybean lines each showed characteristic temporal patterns of L-phenylalanine ammonia-lyase activity. Healthy PI 227687, but not healthy Davis, also showed a characteristic temporal pattern of L-tyrosine ammonia-lyase activity. Activities of the above two enzymes are positively correlated with plant phenylpropanoid metabolism. PI 227687 and Davis leaves did not differ in content of total phenols. Products from phenylpropanoid metabolism apparently contribute to PI 227687 resistance to MBB feeding.
Résumé Les paramètres de la résistance spontanée (c.a.d. non induite) du soja à E. varivestis ont été examinés sur des PI 227687 cultivés dans des conditions contrôlées et reproductibles d'un phytotron assurant une croissance et un développement optimaux. Les antiappétants de PI 227687 contre E. varivestris présentaient différentes solubilités dans l'hexane, l'acétate, d'éthyle et le méthanol à 100%. Les produits du métabolisme phénylpropanoïde des plantes, y compris les phytoalexines, sont fortement solubles dans un ou plusieurs de ces solvants. Les antiappétants sont restés dans le résidu aqueux après les extractions organiques séquentielles. Les PI 227687 sains, ainsi que les Davis susceptibles ont présenté les caractéristiques temporelles de l'activité tyrosine-amonia lyase. Les niveaux d'activité de ces deux enzymes sont très significativement corrélés avec le métabolisme phenylpropanoïde de la plante. Les teneurs en phénols totax de PI 227687 et Davis ne présentaient pas de différences. Les résultats semblent compatibles avec la présence des produits aglycônes du métabolisme phénylpropanoïde comme paramètres spontanés principaux de la résistance de PI 227687 à E. varivestis.

Photosynthetic response of Eragrostis tef to temperature   总被引:1,自引:0,他引:1  
Photosynthetic characteristics of leaves of tef, Eragrostis tef (Zucc.) Trotter, plants, grown at 25/15°C (day/night), were measured at temperatures from 18 to 48°C. The highest carbon exchange rates (CER) occurred between 36 and 42°C. and averaged 27 μmol m−2 s−1. At lower or higher temperatures, CER was reduced, but the availability of CO2 to the mesophyll, measured as internal CO2 concentration, was highest when temperatures were above or below the optimum for CER. In addition, CER and stomatal conductance were not correlated, but residual conductance was highly correlated with CER (r = 0.98). In additional experiments, relative 13C composition for leaf tissue grown at 25, 35 and 45°C averaged -14.4 per mille, confirming that tef is a C4 grass species. Dry matter accumulation was higher at 35 than at 25, and lowest at 45°C. Leaf CER rates increased hyperbolically with increased light when measured from 0 to 2000 μmol m−2 s−1 PPFD. The highest CER, 31.8 μ-mol m-2 s−1, occurred at 35°C and 2000 μmol m−2 s−1 PPFR. At high light, CER at 25 and 35°C were nearly equal because of higher stomatal conductance at 25°C. Residual conductance was, however, clearly highest at 35°C compared to 25 and 45°C treatments. Stomatal conductance and residual conductance were not correlated in either set of experiments, yet residual conductance was always highest when temperatures were between 35 and 42°C across experiments, suggesting that internal leaf photosynthetic potential was highest across that temperature range.  相似文献

The development of proteinase inhibitors as potential insect control agents has been constrained by insect adaptation to these compounds. The velvet bean caterpillar (Anticarsia gemmatalis) is a key soybean pest species that is well-adapted to proteinase inhibitors, particularly serine-proteinase inhibitors, which are abundant in the caterpillar host. The expression of diverse proteolytic enzymes by gut symbionts may allow the velvet bean caterpillar to circumvent proteinase inhibitors produced by the host plant. In this study, we characterized the proteolytic activity of the four nonpathogenic species of gut bacteria isolated from the velvet bean caterpillar—Bacillus cereus, Enterococcus gallinarum, Enterococcus mundtii and Staphylococcus xylosus. Two proteinase substrates, N-α-benzoyl-l-Arg-p-nitroanilide (l-BApNA) and N-α-p-tosyl-l-Arg methyl ester (l-TAME) and five proteinase inhibitors [aprotinin, E-64, ethylenediamine tetraacetic acid (EDTA), pepstatin and N-α-tosyl-l-lysine chloromethyl ketone (TLCK)] as well as CaCl2, pH and temperature profiles were used to characterize the expressed proteolytic activity of these bacterial strains in vitro. Kinetic parameters for proteolytic activity were also estimated. The results of these experiments indicated that serine- and cysteine-proteinase activities were expressed by all four gut bacteria symbionts of the velvet bean caterpillar. The cysteine- and serine-proteinase activities of these gut symbionts were distinct and different from that of gut proteinases of the caterpillar itself. This finding provides support for the potential involvement of gut symbionts in the mitigation of the negative effects of serine-proteinase inhibitors in the velvet bean caterpillar.  相似文献

Iron deficiency (ID) disrupts brain dopamine (DA) and norepinephrine (NE) metabolism including functioning of monoamine transporters and receptors. We employed caudate microdialysis and no net flux (NNF) in post-weaning rats to determine if ID decreased the extraction fraction ( E d). Five micromolar quinpirole, a dopamine D2 receptor agonist, resulted in 80% decrease in extracellular DA and 45% higher E d in control animals. The D2 agonist had no effect on E d in ID animals despite a reduction in basal DA. DAT mRNA levels were reduced by 58% with ID, while DAT protein in ventral midbrain and caudate and membrane associated DAT were also reduced by ID. Carbidopa/ l -DOPA was administered to determine if elevated extracellular DA in ID was due to increased release. The DA response to l -DOPA in ID rats was 50% smaller and delayed, whereas the NE response was threefold higher. The caudate concentration of NE was also elevated in ID. Elevated dopamine-β-hydroxylase activity in ID provides a tentative explanation for the increased NE response to l -DOPA. These experiments provide new evidence that ID results in altered synthesis and functioning of DAT and perhaps suggests some compensatory changes in NE metabolism.  相似文献

A Forssman antigen (GalNAc1-3GalNAc1-3Gal1-4Gal1-4Glc1-1Cer)-binding lectin has been purified from velvet bean (Mucuna derringiana) seeds by a combination of affinity chromatography and reversed phase HPLC. This lectin agglutinates both native and trypsin-treated sheep erythrocytes as well as trypsinized rabbit erythrocytes, but neither native rabbit nor human erythrocytes, irrespective of blood group type. SDS-PAGE and gel filtration chromatography reveal the lectin to be a homodimer consisting of two 54 kDa subunits linked by non-covalent bonds. The results obtained by quantitative precipitation, haemagglutination inhibition and TLC overlay assays indicate that theMucuna lectin specifically recognizes Forssman antigen and Forssman disaccharide (GalNAc1-3GalNAc)-related structures. Trypsin-like proteases from the midgut of Anticarsia gemmatalis Hubner (Lepidoptera: Noctuidae) were purified on an aprotinin-agarose column equilibrated with 0.01 M Tris-HCl containing 5 mM CaCl2 (pH 7.5). The yield was 66.7% with a purification factor of 107 and a final specific activity of 6.88 mM/min/mg protein with the substrate N-alpha-benzoyl-L-Arg-p-nitroanilide (L-BApNA). The purified fraction showed three bands with proteolytic activity and molecular weights of 66,000, 71,000 and 91,000 (sodium dodecyl sulphate (SDS)-polyacrylamide gel electrophoresis (PAGE)). Enzyme specificity assays were carried out using seven synthetic peptides containing 13 amino acid residues, but differing only on the 5th residue (K, R, Y, L, W or P). Peptide cleavage takes place only with amino acids K or R at the 5th position, which is typical of trypsin. The partially purified enzymes hydrolyzed casein and the synthetic trypsin substrates L-BApNA and N-alpha-p-tosyl-L-Arg methyl ester (L-TAME). Higher activity was observed at pH 8.5 and 35 degrees C when using L-BApNA as substrate and at pH 8.0 and 30 degrees C when using L-TAME. Maximum enzyme activity against L-BApNA was obtained with 20 mM CaCl2 in the reaction mixture. The partially purified enzymes showing trypsin activity were sensitive to inhibition by ethylenediaminetetraacetic acid (EDTA), phenylmethyl sulphonyl fluoride (PMSF), N-alpha-tosyl-L-lysine chloromethyl ketone (TLCK), benzamidine and aprotinin. Highest inhibition was obtained with TLCK and benzamidine. KM values obtained were 0.32 mM for L-BApNA and 52.5 microM for L-TAME.  相似文献

Photosynthetic characteristics of ear and flag leaves of wheat species, tetraploid Triticum dicoccoides Kom and hexaploid Bima1, were studied in plants grown under well-watered (WW) and water-stressed (WS) conditions. Compared to ears, flag leaves exhibited higher photosynthetic rate (P N) at the filling stage, but more severe decrease under WS. P N in the tetraploid wheat ear remained higher than that in the hexaploid wheat during the grain-filling stage. Water stress decreased PN in both the organs; this decline was caused by a reduction in Rubisco activity, not by drought-induced stomatal limitation. Tetraploid wheat ears exhibited higher relative water content and water-use efficiency than that of hexaploid wheat, under WS. The change in phosphoenolpyruvate carboxylase activity and carbon isotope composition indicated the absence of C4 metabolism in the ears of both species under both conditions. The improved performance of the tetraploid wheat ears under WS was associated with better water relations.  相似文献

The photosynthetic properties of two commonly used suspension cultured lines, embryogenic and photoautotrophic (PA, SB-1 line) cells of soybean [Glycine max (L.) Merr.] were characterized. We found that compared to the dark green PA cells, the light green embryogenic cells contained fewer and smaller plastids with less-developed thylakoid membranes. The embryogenic cells also contained much lower contents of both chlorophyll and the large subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco; EC 4.1.1.39) protein, an undetectable level of Rubisco small subunit protein, and a very low rate of photosynthesis. While the DNA contents of the nuclear genomes were similar in these two types of cultured cells, the embryogenic cells possessed a markedly lower content of plastid DNA. The 18-year-old PA suspension culture, SB-1, continues to evolve with higher Rubisco and plastid DNA contents than leaves, and with small decreases in nuclear DNA content that appears to mimic changes in chromosome numbers. These findings may prove useful in the application of plastid transformation, particularly when non-leaf or non-green tissues must be used as targets for transformation and plant regeneration.  相似文献
