为了探讨牦牛适应高海拔低氧环境的基因表达特征与规律,对在高海拔(3 560 m)和低海拔地区(478 m)饲育4个月的2.5~3岁健康雄性麦洼牦牛肺组织进行转录组测序。转录组测序采用Illumina高通量测序平台(HiSeqTM2500/4000)进行,并以qRT-PCR验证差异表达基因的表达量。结果显示,高海拔组牦牛肺脏转录组平均每个测序样本得到约5.76亿条Clean Reads,低海拔组牦牛中得到约6.10亿条Clean Reads,比对到参考基因组上的Reads数分别占91.74%和91.28%以上,共发现了2 047个新转录本。低海拔组与高海拔组牦牛肺脏组织之间共有199个差异表达基因,其中含89个差异上调表达基因和110个差异下调表达基因。所得差异表达基因富集在297个GO条目和146个KEGG通路中,包含62个低氧适应相关的GO条目和35个低氧适应相关代谢通路。其中低氧适应相关GO条目在生物过程、细胞组成和分子功能三种类别中占比最多的分别为细胞粘附、蛋白复合物和钙离子结合。低氧适应相关KEGG通路中占比最多的为肿瘤坏死因子(TNF)信号通路,其次为低氧诱导因子1(HIF-1)信号通路。qRT-PCR验证结果显示,Ⅱ类人类白细胞抗原α链(HLA-DOA、HLA-DRA)、补体因子 (C2)和甘露糖结合凝集素相关丝氨酸蛋白酶1(MASP1)基因的表达量变化与转录组测序结果相符。本研究为全局和深入理解牦牛肺组织转录本表达对高海拔低氧的响应提供了有价值的切入点。 相似文献

麦洼牦牛和九龙牦牛FSHβ基因的PCR-SSCP分析 总被引:1,自引:0,他引:1
运用一对特异性引物对麦洼牦牛、九龙牦牛的FSHβ基因的5’端侧翼区进行了扩增,应用PCR-SSCP方法对其进行了多态性分析。结果表明:在麦洼牦牛的FSHβ基因的5’端侧翼区有AA型、AB型和BB型三种基因型,九龙牦牛的FSHβ基因的5’端侧翼区只检测到了AA型、AB型两种基因型。在两种牦牛品系中,AA基因型频率最高,A等位基因频率均明显高于B等位基因频率。麦洼牦牛和九龙牦牛群体中多态信息含量分别为0.3431、0.1411,麦洼牦牛多态性能高,遗传变异大。 相似文献

心脏作为血液循环最重要的器官在动物环境适应中发挥着重要的作用。心肌细胞是心脏主要的功能细胞,然而高海拔地区牦牛心肌细胞基因表达变化及细胞间通讯关系尚不清楚。为此,本研究以高海拔地区牦牛(青海祁连,海拔4 000 m)和低海拔地区牦牛(青海循化,海拔2 600 m)为研究对象,基于心脏组织10×单细胞转录组测序数据,对心肌细胞和心脏其他细胞进行细胞通讯及配体-靶基因调控预测,同时对高、低海拔牦牛心肌细胞差异表达基因进行功能注释分析,以期探究心肌细胞在牦牛适应高原环境过程中的作用。结果显示,牦牛心肌细胞与内皮细胞、上皮细胞的关联最强,心肌细胞-树突状细胞及心肌细胞-巨噬细胞两个“细胞对”中CD74_COAP和CD74_APP配受体表达量相对较高;配受体活力在TNF信号通路中相对较强;在免疫相关通路发挥重要作用的配体PTPRC、PECAM1、ITGB2、ANXA1、BDNF等对所有“细胞对”影响明显,且PI3K-Akt信号通路在配体-靶基因调控中潜力分值最高;高、低海拔牦牛心肌细胞中差异表达基因功能主要富集在代谢途径,其中氧化磷酸化、糖酵解代谢通路相关的基因表达明显增强。本研究结果提示,牦牛... 相似文献

为了从分子水平上检测麦洼牦牛Bos grunniens乳酸脱氢酶-1(LDH1)的H亚基编码基因Ldhb的多态性,实验提取79头麦洼牦牛基因组DNA,采用PCR-RFLP技术分析Ldhb基因多态性,并测定牦牛背最长肌中肌红蛋白含量。实验建立的牦牛Ldhb的G896A突变位点的PCR-RFLP检测方法,在79头麦洼牦牛中检测到Ldhb-AA和Ldhb-AG两种基因型,其中AG基因型频率为16.46%,并且这些样品的乳酸脱氢酶-1的电泳迁移率快于Ldhb-AA样品。本实验未检测到Ldhb-GG基因型;麦洼牦牛Ldhb基因多态性与背最长肌中的肌红蛋白含量之间未见相关性。 相似文献

基于RNA-Seq高通量测序技术的牦牛卵巢转录组研究:进一步完善牦牛基因结构及挖掘与繁殖相关新基因 总被引:3,自引:0,他引:3
RNA-Seq作为近年来新发展起来的高通量转录组测序技术,为大规模转录组学研究提供了一种全新的且更为有效的方法.目前该技术已广泛应用于转录组学中多方面的研究,尤其近年来,该技术在进一步完善基因结构信息及挖掘新转录本及新基因方面的功能也逐渐受到关注.本研究以牦牛卵巢组织作为研究对象,应用RNA-Seq技术对其进行高通量转录组测序分析,经测序后得到了一个包含26826516条过滤后测序读数,4828772880 bp的卵巢测序文库,比对分析显示,有16992条牦牛基因发生表达,其中3734条基因存在不同类型的可变剪接.功能分析表明,这些表达基因涉及多种GO分类及KEGG通路.进一步分析转录组数据发现,共有7340个基因的5′或3′端在原有基因组的位置基础上发生了延伸,同时还发现了6321个新转录本,定位回基因组预测显示,外显子数量为1~84个,其中2267个新转录本预测具有编码蛋白的能力.比对分析显示,共有1200~4993条新转录本分别与Nt数据库、Nr数据库及SwissProt数据库中的基因比对上,其中与牛相似性基因最多(41.4%),其次为野牦牛(33.0%)、绵羊(6.3%)、人类(2.8%)及小鼠(1.6%)等其他物种.进一步对新转录本进行GO分类注释,结果显示,与繁殖发育相关的GO分类占有较大比例,其中繁殖类别(reproduction)所涉及的新转录本最多.本研究结果为描绘牦牛卵巢正常转录组图谱及进一步探析牦牛繁殖性能提供了基础,同时证实RNA-Seq高通量转录组技术在完善基因结构及挖掘新转录本及新基因方面的具有强大的优势,为进一步完善牦牛基因组结构信息及挖掘潜在的新基因提供了丰富数据. 相似文献

为了探讨生长相关基因对团头鲂生长发育的调控, 研究采用Real-time PCR的方法定量分析了团头鲂6个生长相关基因在其不同生长发育阶段(3、6、12月龄)相关组织(脑、肝脏、肌肉)的表达情况, 并比较了这些基因在生长快和慢两个群体的表达差异. 结果显示: GHRs基因在肝脏与肌肉中的表达量高于脑, 在6月龄表达量高于3月龄与12月龄, 生长快群体中的表达量高于生长慢群体(P0.05); IGFs基因在三个组织中均有表达, 肝脏表达量最高, 生长快群体中的表达量高于生长慢群体(P0.05). MSTN a与MSTN b基因在组织中表达模式存在差异, MSTN a在肌肉中高表达, MSTN b主要在脑与肝脏中表达. HCL聚类结果表明: 除了MSTN a基因外, 其他5个基因在生长差异的两个群体中表达量均分别聚为一支. 不同时期组织表达聚类结果表明, 除了3月龄肝脏与12月龄肌肉组织, 6个生长相关基因在不同时期的同一组织中的表达模式存在相似性. Pearson相关分析显示: GHRs与IGFs呈正相关, MSTN a基因与GHR 2、IGFs基因呈负相关, 相同基因在两个群体中呈极显著相关(P0.01).

摘要:土壤盐渍化是一个世界性的资源问题和生态问题,盐分胁迫几乎会影响棉花所有重要的生命活动,
造成棉花的减产或其他不利影响。本试验通过对14个品棉花种(系)的萌发期耐盐性、芽期耐盐性以及两个耐盐性不同的品种(系)三叶期的耐盐性,对棉花不同时期的耐盐特性进行了研究,结果表明:棉花在不同生育阶段耐盐能力是不同的,棉花盐敏感时间是在萌发出苗期。随着生育期的延长,棉花的耐盐性是逐步提高的,进行耐盐性鉴定的最佳时期在是萌发出苗期。在三叶期,随着盐分浓度的提高,棉花苗受伤害程度逐渐增加,盐敏感性品种受伤害大于耐盐品种,在同样盐浓度胁迫条件下,耐盐性不同的品种各部分受到的伤害不同,耐盐品种(系)中棉所35受伤害程度由高到低依次为:真叶>上胚轴>子叶;盐敏感品种(系)Tamcot CAB-CS受伤害程度由高到低依次为:真叶> 子叶>上胚轴。 相似文献

This study aimed to identify proteins secreted by Mycobacterium bovis into culture medium at different stages of bacterial growth. A field strain of M. bovis was grown in Middlebrook 7H9 media and culture supernatant was collected at three-time points representing three different phases of growth (early exponential, late exponential, and stationary phases). Supernatants were double filtered, digested by trypsin and analyzed by LC-MS/MS. The study found 15, 21, and 16 proteins in early, mid and late growth phases, respectively. In total, 22 proteins were identified, 18 of which were reported or predicted to have a cell wall or extracellular localization. To our knowledge, this is the first study to identify proteins secreted into the culture medium by a field strain of M. bovis in three different stages of growth. The dataset generated here provides candidate proteins with the potential for the development of serological diagnostic reagents or vaccine for bovine tuberculosis. The aims of the present work were (1) to determine the influence of plant growth stages on the population size of culturable bacteria and fungi associated with rhizoplane and endo-rhizosphere of maize grown in field and (2) to establish the community structure of total culturable bacteria and fungi. Density, diversity and community structure of culturable rhizoplane and endo-rhizosphere populations at different maize plant growth stages were estimated. Plant development did not have influence on total culturable microflora density but it selectively influenced some bacterial and fungal groups present in the rhizosphere. However, the microbial community structure changed markedly over time. This knowledge is relevant for exploring endophytic rhizobacteria potential in the promotion of plant-growth, protection against pathogens and to detect perturbations in natural agro ecosystems. 相似文献

Roots are a vital organ for absorbing soil moisture and nutrients and influence drought resistance. The identification of quantitative trait loci (QTLs) with molecular markers may allow the estimation of parameters of genetic architecture and improve root traits by molecular marker-assisted selection (MAS). A mapping population of 120 recombinant inbred lines (RILs) derived from a cross between japonica upland rice 'IRAT109' and paddy rice 'Yuefu' was used for mapping QTLs of developmental root traits. All plant material was grown in PVC-pipe. Basal root thickness (BRT), root number (RN), maximum root length (MRL), root fresh weight (RFW), root dry weight (RDW) and root volume (RV) were phenotyped at the seedling (I), tillering (II), heading (III), grain filling (IV) and mature (V) stages, respectively. Phenotypic correlations showed that BRT was positively correlated to MRL at the majority of stages, but not correlated with RN. MRL was not correlated to RN except at the seedling stage. BRT, MRL and RN were positively correlated to RFW, RDW and RV at all growth stages. QTL analysis was performed using QTLMapper 1.6 to partition the genetic components into additive-effect QTLs, epistatic QTLs and QTL-by-year interactions (Q x E) effect. The results indicated that the additive effects played a major role for BRT, RN and MRL, while for RFW, RDW and RV the epistatic effects showed an important action and Q x E effect also played important roles in controlling root traits. A total of 84 additive-effect QTLs and 86 pairs of epistatic QTLs were detected for the six root traits at five stages. Only 12 additive QTLs were expressed in at least two stages. This indicated that the majority of QTLs were developmental stage specific. Two main effect QTLs, brt9a and brt9b, were detected at the heading stage and explained 19% and 10% of the total phenotypic variation in BRT without any influence from the environment. Insecticide protection at the vegetative, reproductive or both vegetative and reproductive (complete) crop growth stages and untreated control was used to assess yield loss due to insect pests at the different growth stages of soybean in Ghana from 2007–2009. The objectives were to determine the economic importance of the two major insect pest guilds in soybean, viz. defoliators and pod feeders, and when to apply control measures for maximum benefit. The defoliators recorded were Podagrica spp., Ootheca mutabilis (Shalberg), Zonocerus variegatus L., Sylepta derogata F., Spodoptera littoralis Boisduval, Amsacta spp. and Helicoverpa armigera Hübner. The pod feeders recorded were the pod-sucking bugs (PSBs) Riptortus dentipes F., Thyanta sp. Aspavia armigera F., Nezara viridula L. and Dysdercus völkeri Schmidt. Generally, insect densities, pod and seed damage were lower while seed yields were significantly greater and similar in plots that were protected at the reproductive stage against PSBs and those protected at both vegetative and reproductive stages. Yield loss ranged between 25.8 and 42.8% in untreated plots, 11.1 and 34.3% in plots that were protected at the vegetative stage, and 5.2 and 11.3% in plots that were protected at the reproductive stage. There was a consistent negative correlation between yield and numbers of PSBs as well as pod and seed damage. These results showed that PSBs that attack soybean at the reproductive stage were the most important insect pests limiting soybean yield in Ghana. 相似文献
