Geotropically induced phase-shifts in circumnutations of Heliauthus annuus hypocotyls are studied. - Theoretical deductions from a previously developed theory for geotropical movements are made for the case of a short gravitational stimulation in different phase positions of an oscillating hypocotyl. - Experimental studies are performed to test the theoretical results. - The conclusions are drawn, that the theory mentioned can satisfactorily describe geotropically induced phase-shifts. A comparison with phase-shifts in photoperiodic systems is made.  相似文献

The movement of auxin in Phaseolus vulgaris roots has been examined after injection of IAA?3H into the basal root/hypocotyl region of intact, dark-grown seedlings. Only a portion of the applied IAA?3H was transported unchanged to the root tip. The major part of the chromatographed, labelled compounds translocated to the roots was indole-3-acetylaspartic acid (IAAsp) and an unidentified compound running near the front in isopropanol, ammonia, water. The velocity of the auxin transport (7.2 mm per hour) was calculated from scintillation countings of methanol extracts from serial sections of the root. An accumulation of radioactive compounds in the extreme root tip, was observed 5 h after the injection of IAA. The influence of exogenous IAA on the geotropical behaviour of the bean seedling roots was examined. Pretreated roots were stimulated for 5 min in the horizontal position and then rotated parallel to the horizontal axis of the klinostat for 60 or 90 min. The resulting geotropic curvature of IAA-injected and control roots showed significantly different patterns of development. When the stimulation was started 5 h after application of the auxin, the geotropic curvature became larger in roots of the injected plants than in the controls. If, however, the translocation period was extended to 20 h the geotropic curvature was significantly smaller in the roots of the injected plants. The auxin injection did not significally affect the rate of root elongation. The change in geotropical behaviour of the roots is interpreted as a result of the influence of the conversion products of the applied IAA on the geotropical responsiveness.  相似文献

Summary Horizontally-placed segments of Avena sativa L. shoots show a negative geotropic response after a period of 30 min. This response is based on cell elongation on the lower side of the leaf-sheath base (pulvinus). Triticum aestivum L., Hordeum vulgare L. and Secale cereale L. also show geotropic responses that are similar to those in Avena shoots. The pulvinus is a highly specialized organ with radial symmetry and is made up of epidermal, vascular, parenchymatous and collenchymatous tissues. Statoliths, which are confined to parenchyma cells around the vascular bundles, sediment towards the gravitational field within 10–15 min of geotropic stimulation. Collenchymatous cells occur as prominent bundle caps, and in Avena, they occupy about 30% of the volume of the pulvinus. Geotropic stimulation causes a 3- to 5-fold increase in the length of the cells on the side nearest to the center of the gravitational field. Growth can also be initiated in vertically-held pulvini by the application of indole-3-acetic acid, 1-naphthaleneacetic acid or 2.4-dichlorophenoxyacetic acid. 2.3.5.-triiodobenzoic acid interferes with growth response produced by geotropic stimulation as well as with the response caused by auxin application. Gibberellic acid and kinetin have no visible effect on the growth of the pulvinus. Polarization microscopy shows a unique, non-uniform stretching of the elongating collenchymatous cells. Nonelongated collenchymatous cells appear uniformally anisotropic. After geotropic stimulation or auxin application, they appear alternately anisotropic and almost isotropic. A theory for the geotropic movements of the Heliunthus unnuus hypocotyl has been given earlier by Johnsson et ul. In the present paper this theory is tested by extensive experiments. The magnitude as well as the rate of geotropic curvature are found to be in agreement with the theoretical predictions. Further, the geotropic reaction time, defined as the time between the start of geotropic stimulation and the start of the geotropic reaction is determined as a function of the stimulation angle with respect to the plumb line. A slight variation found in the reaction time is included in the theory.  相似文献

The equation for the quantum transitions (spontaneous and stimulated) of membrane dipoles is solved for the various forms of time-varying stimulation in nerve. From the condition of ever-increasing dipole population in the upper state, the threshold for excitation is determined in each case. The results obtained are in agreement with the established facts. The optimum frequency for stimulation is given asv 0=0.0615/T 2 whereT 2 is the dipole relaxation time. The feature of the theory is that the mathematical formulation is based upon a physical mechanism and the results can thus provide some understanding in the observed phenomena.  相似文献

Summary In the initial phase of the geotropical reaction of the Chara rhizoid the growth difference postulated by Sievers (1967c) between the physically upper, slightly subapical flank and the lower one is demonstrated. In horizontal exposure the growth of the extreme cell apex is continued, while the growth of the lower flank is inhibited and that of the upper one is promoted. In the end phase the cell apex shows a damped oscillation until it finally reaches the vertical growth direction. The statoliths follow the oscillating growth of the cell tip from one flank to the opposite one until they are statistically equally redistributed in their normal position.—In vertical exposure under reduced turgor pressure the statoliths fall down into the extreme cell apex, where they inhibit the growth of this part of the cell wall, while the subapical wall grows transversally.—It is concluded that the statoliths inhibit the growth of the cell wall area which they cover.—The physical phase of the reaction chain, the susception, is the gravity-induced downward displacement of the statoliths. The physiological phase starts with the diversion of the acropetal transport of the Golgi vesicles to the upper part of the cell, which is caused by the block of statoliths (perception). The greater rate of vesicle incorporation into the upper flank in comparison to the lower one causes the subapical growth difference which results in the curvature (reaction).—In the case of the Chara rhizoid Golgi- and statolith-apparatus function as a self-regulating cellular system.

Summary Growth is initiated in segments of the leaf sheath base of Triticum aestivum by gravitational stimulation and by incubation in buffer solutions of pH 3. Both responses involve only an increase in segment length, are rapidly terminated on removal of the stimulus, have a similar Q10 and are dependent upon cell turgor. They differ, however, in that the response to acid solutions is rapid and unaffected by anoxia. Acid-induced growth can be stopped and started repeatedly by changing the pH, an increase in pH from 3 to 5 or 7 being sufficient to terminate the response. The maximum growth induced by low pH is not increased by simultaneous stimulation by gravity.  相似文献

In experiments on isolated medullo-spinal preparations from 2- to 3-day-old rats, we recorded the spontaneous phasic respiratory activity (RA) from filaments of the ventral spinal C 1-C 4 roots and studied modifications of this activity after applications of vasopressin and electrical stimulation of the n. trigeminus. Vasopressin added to the superfusate in minute concentrations (threshold concentration about 1 nM) increased the frequency of the phasic RA; higher concentrations (100 nM or more) induced the development of a tonic activity in the cervical ventral roots. Electrical stimulation of the n. trigeminus by a series of threshold current pulses inhibited the development of the next in turn RA volleys; similar stimulation, but applied in the course of vasopressin application, resulted in generation of inspiratory volleys in response to each stimulating series. The mechanisms of facilitative influences of vasopressin on the RA generation and effects evoked by trigeminal stimulation are discussed.  相似文献

The tentacle withdrawal reflex of the terrestrial snail Helix aspersa was studied in vitro. The reflex is evoked by mechanical stimulation of the nose. Lesion experiments showed that 45% to 75% of the response amplitude is attributable to peripheral pathways alone. The central contribution increases with increasing stimulus intensity.Repeated stimulation produced pure habituation at low stimulus strengths, and habituation mixed with intrinsic sensitization (warm-up effect) at high stimulus strengths. The simultaneous occurrence of habituation and sensitization is consistent with the dual process theory of plasticity. Additional results differentiate the two processes. Habituation can occur without the CNS, whereas intrinsic sensitization requires the CNS. Also, the two processes are differentially effective in their influences on response amplitude and duration: habituation is more effective in determining response amplitude, while sensitization is more effective in determining response duration.Although the establishment of sensitization requires the CNS, 81% of the memory for intrinsic sensitization was localized to the periphery, by lesion experiments. Extrinsic sensitization, caused by stimulation of the medial lip nerve, had similar behavioural effects and a similar memory locus. Hypocotyl sections from dark-grown mung bean (Vigna radiata (L.) Wilczek) seedlings grown without boron exhibit little or no increase in the translocation of fluorescein in phloem cells following gravitational stimulation or red irradiation. In contrast, pretreatment of the hypocotyl sections with low concentrations of boric acid before red irradiation or gravitational stimulation results in a rapid increase in fluorescein movement. Mung bean root tips without boron fail to remain attached to a negativelycharged glass surface after several minutes of red irradiation. A low boron concentration in the solution induces tip attachment. The suggestion is made that boron is required to stabilize a positive electrostatic charge in the plasma membrane that is generated by the actions of phytochrome and gravity.  相似文献

Gravity is the one constant, ubiquitous force that has shaped life on Earth over its 4.8 billion years of evolution. But the sheer inescapability of Earth’s gravitational pull has meant that its influence on Earth’s organisms is difficult to study. Neutralization of the gravity vector (so‐called simulated microgravity) by random movement in three‐dimensional space is the best option for Earth‐based experiments, with spaceflight alone offering the possibility to assess the effects of an extremely reduced gravitational field (microgravity). However, the technical constraints associated with spaceflight introduce complications that can compromise the interpretation of microgravity experiments. It can be unclear whether changes detected in these experiments reflect additional spaceflight‐related stresses (temperature shifts, vibrational effects, radiation exposure, and so on) as opposed to the loss of gravitational force per se. In this issue, Herranz et al. (2010) report a careful study in which the effects of simulated and actual microgravity on gene expression in Drosophila melanogaster were compared and the effects of the flight‐associated stresses on the microgravity responses were investigated. A striking finding emerged. The additional stresses associated with the spaceflight experiment altered the response to microgravity. Despite controlling for the effects of these stresses/constraints, the group found that responses to microgravity are much stronger in the stressed/constrained background than in its absence. This interaction of gravity with other environmental influences is a novel finding with important implications for microgravity research and other situations where multiple stress factors are combined.  相似文献

In this review, parity‐violating gravitational potentials are presented as possible sources of both true and false chirality. In particular, whereas phenomenological long‐range spin‐dependent gravitational potentials contain both truly and falsely chiral terms, it is shown that there are models that extend general relativity including also coupling of fermionic degrees of freedom to gravity in the presence of torsion, which give place to short‐range truly chiral interactions similar to that usually considered in molecular physics. Physical mechanisms which give place to gravitational parity violation together with the expected size of the effects and their experimental constraints are discussed. Finally, the possible role of parity‐violating gravity in the origin of homochirality and a road map for future research works in quantum chemistry is presented. We studied the interaction of synaptic effects in efferent (rubro-spinal) neurons (RSN) of the red nucleus (NR) of the cat brain using intracellular recording and different variants of stimulation of the cerebellar nucl. interpositus (NI) and nucl. ventrolateralis (VL) of the thalamus. Combined simultaneous or consequent stimulation of the above two inputs allowed us to use collision of the impulses in pre-synaptic pathways for estimation of the peculiarities of summation of post-synaptic potentials; it was taken into consideration that cerebello-rubral fibers are collaterals of the cerebello-thalamic axons. In the case of relatively low intensities of stimulation of the VL and NI, EPSP evoked by these stimulations in RSN could demonstrate linear summation, which is indicative of the absence of common fibers excited by stimulation of both the thalamic and cerebellar structures and of the absence of interaction of synaptic effects on the membrane of the postsynaptic unit (an RSN). With increased stimulation intensity and definite time relations between the stimuli applied to the above structures, the second EPSP evoked by the two stimuli was significantly suppressed; this resulted from a decrease of the afferent drive coming to the NR due to collision of afferent impulses in presynaptic pathways. When an interstimulus interval was increased, the second EPSP could be facilitated (probably due to generation of repetitive impulses in common presynaptic fibers). Our experiments show that the statement concerning the nature of cerebello-rubral inputs (all these inputs are formed by collaterals of the cerebello-(interposito)-thalamic pathways) should be considered with reservation.  相似文献

The author summarizes the results of many-year application of the model of in vitro sturgeon oocyte maturation for different purposes, such as comparison of gonadotropic activities of different preparations, selection of females for breeding, and studying the effects of different factors in order to improve the breeding technology. Special attention is paid to factors that can affect the results of experiments on hormonal stimulation of in vitro oocyte maturation and ovulation and their interpretation. Two other phenomena are discussed: the inhibitory effect of gonadotropic pituitary hormones on the progesterone-induced in vitro oocyte maturation and the nonhormonal induction of oocyte maturation, further studies of which can elucidate the mechanisms underlying the hormonal regulation of oogenesis in sturgeons.  相似文献

Summary DNA-DNA hybridisation experiments show that chloramphenicol induces a burst of initiation from the oriC region of a dnaA46 mutant of Escherichia coli at 36.5° C but not from the isogenic dnaA + strain. Following this stimulation of initiation, DNA replication proceeds normally towards the terminus. The temporal pattern of the extra initiation is in parallel with the induced stimulation of RNA synthesis caused by chloramphenicol in the same strain. This is consistent with the hypothesis that the stimulation of initiation in the dnaA mutant is the result of the stimulation of the synthesis of an RNA species.  相似文献

A comparison is made between gravity-induced and light-induced transversal electrical potentials in excised coleoptiles of Zea Mays. The potentials are measured with a vibrating condensor method, thus the plants are not touched at the measuring point. It is found that all experimental evidence supports the assumption that the reaction chains leading to the potentials are identical in the two cases, apart of course from the perception stages. This theory is based on the following points which have been experimentally verified:
  • 1 The stimulus-response functions are quite similar. In both cases the voltage is a logarithmic function of the stimulation time (at least for shorter stimulation times). When continuous stimulation is used, the stimulation time must exceed a threshold value to cause a transversal potential.
  • 2 Some inhibitors (DNP and NaN3) diminish both types of potential.
  • 3 A latency time is found in photo-induced potentials as well as in gravity-induced ones. The Q10 for the latency period of the photo-induced potentials is found to be 2.8, which is the same as for the chemical part of the latency period of the gravity-induced potentials.
  • 4 In both cases the potential-generating process is controlled by a Q10 of 1.6–1.8, thus indicating this process as separate from the one mentioned above.
  • 5 The temperature dependence of the maximum voltage follows in both cases the same pattern with a maximum at about 25–30°C.
Some measurements on potential oscillations are described and discussed. The origin of the potentials is discussed.  相似文献

The calculation of the size of the “sensitive volume” or “control center” in biological effects of radiations is discussed from the viewpoint of the probabilistic theory of these phenomena based on the concept of random “effective events”. On the bases of that theory, the resistivity of a microorganism to radiation is defined as its “mean life” under a radiation of one roentgen per minute. This mean is calculated for processes with and without recovery. The case of variable sensitivity, as it occurs for instance during mitosis, is discussed in detail. Methods are given to calculate this variability from survival curves or similar experimental data. The theory is applied to experiments of A. Zuppinger on irradiation ofAscaris eggs with X-rays.  相似文献

The contraction of Stentor and Blepharisma, in response to mechanical and electrical stimulation and of Spirostomum in response to mechanical stimulation is described. All three species respond to electrical stimulation by contraction of the cytoplasm, beginning at the anodal end regardless of orientation of the animal. The differences in contractile ability and shapes during contraction are discussed in relation to body form and microanatomy. Stentor and Spirostomum also respond to mechanical stimulation. Dropping a weight on the slide causes contraction of the whole body of Spirostomum, but not of Stentor. Stimulation of the oral region of Stentor by means of a vibrating needle causes a contraction of the entire body, but this sensitivity is limited to the oral region. Blepharisma does not respond to mechanical stimulation. Spirostomum and Stentor undergo rapid spontaneous contractions, but Blepharisma does not contract spontaneously.  相似文献

In 1974, Zimm described a theory which predicts that the sedimentation coefficient of high-molecular-weight DNA will decrease as the rotor speed of measurement increases. In 1979, this theory was revised, and the new formula predicts speed-dependence effects that are substantially smaller than the predictions of the original version. This report describes the results of subjecting both the original and the revised versions of the theory to quantitative tests using a well-defined sucrose-gradient system and a DNA of known molecular weight (T4c DNA). T4c bacteriophage is a mutant, whose DNA contains the unmodified base cytosine, instead of the glucosylated hydroxymethylcytosine characteristic of the T-even bacteriophages, and has a molecular weight of 115 ± 3 × 106. The DNA of the wild-type phage (T4D+) was also used in some experiments. In addition to the quantitative tests, the experiments test for an effect first observed by Rubenstein and Leighton, which showed that the sedimentation coefficient measured for T2 DNA depended on the composition of the centrifuge tube used for the measurement (tube composition effect). It can be inferred from this observation that an interaction occurs between particle and tube wall during sedimentation, and this leads to a reduction in sedimentation velocity independent of the reduction in S described by Zimm's theory. The results show that in the range of 25,000–50,000 rpm, the original but theoretically incorrect form of the theory quite accurately describes the sedimentation behavior of both T4c and T4D+ DNA, although T4D+ was a special case in some respects. The revised (corrected) form of the theory predicts much less of a speed-dependence effect than that actually observed. The discrepancy between corrected theory and observation suggests that other factors (perhaps arising from the use of the swinging bucket rotor geometry) are causing the additional observed reduction in S20,w. However, the experiments show that the tube composition effect does not seem to be one of these.  相似文献

We present a stochastic approach to phase-resetting of an ensemble of oscillators. In order to describe stimulation-induced dynamical phenomena we develop a stochastic model which consists of an ensemble of phase oscillators interacting via random forces. Every single oscillator is submitted to a phase stimulus. The ensemble's dynamics is determined by a Fokker-Planck equation. The stationary states are calculated explicitly, whereas the transients are analysed numerically. If the stimulus of a given (non-vanishing) intensity is administered at a critical initial cluster phase for a critical duration T crit the ensemble's synchronized oscillation is annihilated. A transition from type 1 resetting to type 0 resetting occurs when the stimulation duration exceeds T crit. Stimulation causes a shift of the mean frequency of every single oscillator. This frequency shift is explicitly calculated by deriving the mean first passage time. The model shows that there is a subcritical intensity which is connected with an enhanced vulnerability to stimulation. The desynchronized states, the so-called black holes, are typically associated with a double peak in the ensemble's phase distribution. This is important for analysing experimental data because simple peak-detection algorithms are not able to extract the underlying dynamics.Our results are discussed from the experimentator's point of view so that the insights derived from our model can improve data analysis and design of stimulation experiments.  相似文献
