The effects of serotonin (5-hydroxytryptamine) on ventilation were investigated by continuous measurements of intrabuccal pressure in unrestrained eel. Intravenous administration of 5-hydroxytryptamine (30 g·kg-1) caused a large increase in ventilatory frequency (+100%) and amplitude (+140%). The 5-hydroxytryptamine-induced hyperventilation was blocked by the 5-HT3-receptor antagonists metoclopramide (1.0 mg·kg-1) or MDL72222 (1.0 mg·kg-1), and was insensitive to the 5-HT1/2-receptor antagonist methysergide (3.0 mg·kg-1) and to the 5-HT4-receptor antagonist DAU 6285 CL (3.0 mg·kg-1). The hyperventilatory response to 5-hydroxytryptamine could be mimicked by the 5-HT3 receptor agonist 1-phenylbiguanide (300 g·kg-1). The chromosomes of the European eel Anguilla anguilla have been analyzed with a replication banding technique from lymphocyte cultures treated with 5-BrdU. This technique allows us to identify with high resolution the individual chromosome pairs and to differentiate classes of chromatin by the order of replication. The replication banding obtained on the chromosomes of European eel can be related with the structural bands described in this species.  相似文献

Summary The ultrastructure of coronet cells of the saccus vasculosus has been studied in specimens of Anguilla anguilla (L.) at different stages of its life cycle. At all the stages observed coronet cells are composed of a basal and an apical part, the latter bearing globules with primary vesicles. In the larva (a marine form) and in the fully metamorphosed small eel at the time of entry into freshwater the narrow lumen and the vesicles within the apical globules are filled with electron-dense material. In forms in which adaptation to freshwater has occurred, the saccus lumen appears expanded, the apical globules are better developed, and the electron-dense material has disappeared. It is suggested that the two situations observed represent different functional states of the organ, in relation to different conditions of environmental salinity.The authors gratefully acknowledge the contribution of Dr. G. We assessed the effect of eel (Anguilla anguilla) removal from three sites of a Cantabrian stream upon its subsequent densities. In the first sample (Sept. 1986) numbers and densities were estimated as 43, 45 and 84 ind and 3490, 3030 and 3750 ind ha −1. Removal of these eels reduced the subsequent numbers and densities which, except on two occasions, were never reached again during the two years (eleven estimates) of study. Highest densities were recorded in the uppermost site in May and July, 1987, coincident with a strong drought and the lowest densities occurred in 1988 during a normal wet year. We hypothesize first that, because of a selective underground homing behaviour of eels, electro-fishing is inefficient and results in underestimates of the population. Second, seasonal variations of water discharge and droughts may not influence the homing behaviour of'eels until a threshold of dryness is reached. If this occurs, eels abandon their refuges and move towards the stream bottom. It seems that in Arroyo Chabatchos this threshold was exceeded in the summer of 1987 when the highest densities were estimated. The re-colonization of these sites experimentally depleted of eels, is a slow procces that lasts for, at least, two years.  相似文献

The regulation of salt absorption in the sea water cell intestine was studied by evaluating the effects of theophylline, 8 Br cyclic adenosine monophosphate, 8 Br cyclic guanosine monophosphate, atriopeptin III, porcine vasoactive intestinal peptide and prostaglandin E 1 on the short-circuit current, the transepithelial voltage difference and conductance and on the dilution potentials. It was shown that theophylline increased the transepithelial conductance and reduced the magnitude of the dilution potentials, indicating that the drug increase the anion conductance of the tight junctions. In addition its inhibitory effect on short-circuit current and transepithelial voltage difference suggests that theophylline also affects the transcellular transport mechanisms. It was shown that 8 Br cyclic guanosine monophosphate and 8 Br cyclic adenosine monophosphate affect transcellular mechanisms underlying Cl transport since both compounds reduced short-circuit current and transepithelial voltage difference; however, cyclic adenosine monophosphate is less effective since unlike cyclic guanosine monophosphate, even at maximal concentration, it was not able to completely abolish transepithelial voltage difference and short-circuit current. The effects of cyclic guanosine monophosphate and cyclic adenosine monophosphate were not additive even if cyclic guanosine monophosphate may produce further inhibition of ion transport in 8 Br cyclic adenosine monophosphate-treated tissues. In addition, cyclic guanosine monophosphate but not cyclic adenosine monophosphate reduced the magnitude of the dilution potentials, suggesting that cyclic guanosine monophosphate acts also on the paracellular pathway. Rat atriopeptin III, a peptide known to increase cyclic guanosine monophosphate cellular levels, behaved like 8 Br cyclic guanosine monophosphate since it lowered the dilution potentials and reduced short-circuit current and transepithelial voltage difference to near zero values, suggesting that the hormone modulates both paracellular and transcellular transport mechanisms, probably acting on the Na-K-2Cl cotransport. Agents acting via cyclic adenosine monophosphate, like porcine vasoactive intenstinal peptide and prostaglandin, behaved like 8 Br cyclic adenosine monophosphate. Summary Eels were adapted to black- or white-coloured backgrounds and the pituitary glands were prepared for light and electron microscopy. Immunocytochemical staining was used to study the distribution of the neurohypophysial melanin-concentrating hormone in the neurointermediate lobe. The hormone was located in small, elliptical, electron-opaque neurosecretory granules, measuring approximately 120×90 nm. The neurones terminated on blood vessels in the centre of the neurohypophysis and on the basement membrane separating neural and intermediate lobe tissues. The results of both light and electron immunocytochemistry and of radioimmunoassay are consistent with a higher rate of hormone release from eels adapted to white backgrounds than from those adapted to black backgrounds. In addition to this, when fish that had been adapted to white tanks were transferred to black tanks, there was an accumulation of irMCH in the gland and an increased numerical density of secretory granules at nerve terminals. These results reinforce the proposal that MCH is released during adaptation to a white background, to cause melanin concentration and to inhibit MSH release, and that its release is halted in black-adapted fish.  相似文献

The effects of serotonin on continuously recorded cardiac parameters (heart rate, cardiac output, cardiac stroke volume), ventral and dorsal aortic blood pressures, branchial and systemic vascular resistances were investigated in the European eel in vivo. Intravenous administration of serotonin (30 g · kg–1) caused a marked bradycardia (45%) and a simultaneous decrease in cardiac output (50%), ventral (35%) and dorsal (50%) aortic blood pressures. Branchial resistance was markedly increased (60%) and systemic resistance decreased (30%). Cardiac stroke volume remained unchanged. The effects of serotonin on cardiac mained unchanged. The effects of serotonin on cardiac parameters were suppressed either by methysergide or a bilateral section of the cardiac vagus. Bradycardia could then be regarded as the consequence of a vagal mechanism triggered by serotonin action on central methysergide-sensitive serotonergic receptors. No inotropic effect of serotonin was observed. This lack of myocardiac contractility modification is discussed. The serotonin-mediated branchial vasoconstriction was attenuated by vagotomy, whereas the residual increase in branchial resistance (40%) was suppressed by methysergide. The serotonin-mediated branchial vasoconstriction could be the consequence of both a passive mechanism (compliance) caused by the decrease in cardiac output and an active mechanism involving methysergide-sensitive serotonergic receptors of the branchial vasculature. A possible involvement of this vasomotor effect in gill oxygen uptake is discussed. Summary The parapineal organ of the glass eel (elver) consists of approximately 400 cells and is situated to the left of the connection of the pineal stalk to the third ventricle. A conspicuous nerve tract containing approximately 350 fibers arises from the parapineal organ and runs in spatial relationship to the habenular commissure toward the left habenular nucleus. The dominating cell type of the parapineal organ of the elver is a neuron (sensory neuron) of small diameter provided with atypical cilia (9×2+0, or rarely 8×2+0 types). Well-developed photoreceptor outer segments are lacking, and no interstitial cells of ependymal type have been observed with certainty in the parapineal organ. The axonal processes from the nerve cells form the tract leaving the parapineal organ.The pineal organ proper of the elver consists of photoreceptor cells with well-developed outer segments, interstitial cells of ependymal type, and ganglion cells. Axons from the latter form the pineal tract, which leaves the pineal organ and runs in close contact with the subcommissural organ toward the posterior commissure. The proximal part of the pineal stalk contains only a few photoreceptor cells the outer segments of which are less developed than those of the pineal body and the distal part of the pineal stalk.  相似文献

The European eel (Anguilla anguilla L.) is distributed in coastal and inland habitats all over Europe, but spawns in the Sargasso Sea and is thus affected by both continental and oceanic factors. Since the 1980s a steady decline has been observed in the recruitment of glass eels to freshwater and in total eel landings. The eel is considered as critically endangered on the International Union for the Conservation of Nature and Natural Resources Red List of species. The Skagerrak beach seine survey from Norway constitutes the longest fishery-independent dataset on yellow/silver eels (starting in 1904). The Skagerrak coastal region receives larvae born in the Sargasso Sea spawning areas that have followed the Gulf Stream/North Atlantic Drift before they penetrate far into the North Sea. The Skagerrak coastal time series is therefore particularly valuable for exploring the impacts of oceanic factors on fluctuations in eel recruitment abundance. Analyses showed that Sargasso Sea surface temperature was negatively correlated with eel abundance, with a lag of 12 years revealing a cyclic and detrimental effect of high temperatures on the newly hatched larvae. The North Atlantic Oscillation index and inflow of North Atlantic water into the North Sea were negatively correlated with eel abundance, with a lag of 11 years. Increased currents towards the North Atlantic during high North Atlantic Oscillation years may send larvae into the subpolar gyre before they are ready to metamorphose and settle, resulting in low recruitment in the northern part of the distribution area for these years. The Skagerrak time series was compared with glass eel recruitment to freshwater in the Netherlands (Den Oever glass eel time series), and similar patterns were found revealing a cycle linked to changes in oceanic factors affecting glass eel recruitment. The recent decline of eels in the Skagerrak also coincided with previously documented shifts in environmental conditions of the North Sea ecosystem.  相似文献

Summary In the neuro-intermediate lobe (NIL) of the eel, Anguilla anguilla, a specific formaldehyde-induced fluorescence, indicating a catecholamine (CA) innervation, has been demonstrated in the neural lobe processes. Microspectrofluorimetric analyses and pharmacological treatments indicate noradrenaline or dopamine or both to be responsible for the fluorescence.The fluorescence in the NIL has displayed a definite tendency toward variation during the adaptation to a white and to a black background. The highest amounts of fluorescence were generally found in animals adapted to a black background, especially when adapted for a rather long period, and in animals recently transferred to a white background. The lowest amounts of fluorescence were generally found in animals adapted to a white background.This and the result of injections of CA-depleting drugs suggest that the monoaminergic nerves are active when the animal is on a white background, inhibiting the MSH release directly or indirectly or both, or in co-operation with other factors.Specific green fluorescent structures were also found in other parts of the neural lobe supplying the pars distalis.In some pharmacologically untreated specimens and in animals treated with CA-depleting drugs, the intermedia cells fluoresced. Zusammenfassung Das Flimmerepithel von Anguilla anguilla besteht aus 4 Zellarten: Flimmerzellen, Stützzellen, Basalzellen und Schleimbecherzellen. Flimmerzellen enthalten im oberen Zelldrittel zahlreiche Mitochondrien und tragen an ihrer Oberfläche bis zu 140 Kinocilien. Die Basalkörper dieser Kinocilien haben lange Wurzelfilamente, von denen ein Teil ins Zellinnere zieht; der andere Teil verläuft parallel zur Oberfläche und verbindet benachbarte Basalapparate. — Ein Übergangsepithel verknüpft das Flimmerepithel mit dem Riechepithel. Im Riechepithel finden sich außer den Zellarten des Flimmerepithels die Rezeptoren. Bei einheitlichem Aufbau des Zellkörpers lassen sich aufgrund rein morphologischer Unterschiede der Vesiculae olfactoriae 3 Rezeptortypen unterscheiden: 1. Cilien-Rezeptor, 2. Mikrovilli-Rezeptor und 3. Pfriem-Rezeptor. — Der Cilien-Rezeptor trägt unterhalb der Vesicula olfactoria in einer Einschnürung 4–8 sensorische Cilien, die alle auf gleicher Höhe entspringen. Zwei gegenüberliegende sensorische Cilien schließen einen konstanten Winkel von 60° ein. — Der Mikrovilli-Rezeptor trägt auf seiner abgerundeten Vesicula olfactoria 30 bis 60 Mikrovilli von 0,1 m Dicke und bis zu 5 m Länge. Der Mikrovillus wird von einem zentralen, 160 Å weiten, Tubulus durchzogen. Unterhalb der Vesicula olfactoria liegen mehrere Centriolen. Die Rezeptornatur dieser Zellen wird durch ein Axon unterstrichen. — Der Pfriem-Rezeptor besitzt eine 0,8 m breite und bis zu 4 m lange Vesicula olfactoria ohne sensorische Cilien und ohne Mikrovilli. Im Lumen der Vesicula olfactoria befinden sich neben Neurotubuli auch Fibrillen von 40–50 Å Durchmesser, die gebündelt auftreten. An der Basis des Köpfchens liegen mehrere Centriolen.

Summary The ciliary and olfactory epithelia of the olfactory folds in Anguilla anguilla were studied with the electron microscope. The ciliary epithelium is composed of ciliary cells, supporting cells, basal cells, and mucous cells. The ciliary cells contain numerous mitochondria in their apical portion and bear up to 140 cilia. The ciliary basal bodies have rootlets, some of which project towards the central part of the cell, and others parallel to the cell surface thereby connecting neighbouring basal bodies. A transitional epithelium is located between the ciliary and olfactory epithelia. The olfactory epithelium is composed of the same 4 cell types of the ciliary epithelium and besides contains three morphologically different receptor cell types: ciliary receptor cells, microvillous receptor cells, and receptors with a single rodshaped free appendage. The ciliary receptors have 4 to 8 sensory cilia which project from below the vesicula olfactoria, each forming a constant angle of about 30° with the vertical cell axis. The vesicula olfactoria of the microvillous receptors bears from 30 to 60 microvilli, each of 0.1 m diameter and up to 5 m length. Each microvillus of this receptor type contains a central tubulus of 160 Å diameter. Few centrioles are located closely to the vesicula olfactoria. The third receptor type, which has neither cilia nor microvilli, is characterised by a single rod-shaped appendage of 0.8 m diameter which projects up to 4 m above the epithelial surface. This appendage contains neurotubules and fibril bundles; some centrioles lie close to the base of the appendage.

Cholinesterase (ChE) activities were characterized in silver European eel, Anguilla anguilla, grown in the brackish lagoon of Comacchio (Italy). All specimens were harvested at the “lavoriero”, a traditional eel trapping weir that captures eels while leaving internal waters at the onset of reproductive migration. To our knowledge, no investigation on ChE was reported in silver eels. Therefore a first characterization of enzyme activity in muscle, brain, liver and plasma of silver eel was carried out, in the presence of different substrates, selective inhibitors, and four pesticides representative of the carbamate and organophosphate classes. Brain and white skeletal muscle showed similar ChE activities, 5- and 10-fold higher than those detected in liver and plasma, respectively. Km values of 0.31 and 0.30 mM, and Vmax values of 40.28 and 35.47 nmol min–1 mg protein–1 were obtained in brain and muscle ChE, respectively. Acetycholinesterase was the predominant ChE form in all tissues, as concluded by comparing the effects of BW 284c51, iso-OMPA and eserine. ChE activities in brain and muscle were significantly inhibited by in vitro treatment with pesticides, with the following order of potency: carbofuran > carbaryl > chlorpyrifos ≥ diazinon.  相似文献

The screening of 2,735 eels from European waters and aquaculture farms was conducted using mitochondrial Cytochrome b and 16S rRNA gene fragments amplified by polymerase chain reaction. Reaction products were either sequenced directly or subjected to analysis using restriction fragment length polymorphism which resulted in species-specific restriction patterns. Beside the expected European eel, Anguilla anguilla (Linnaeus, 1758), the American eel, Anguilla rostrata (Le Sueur, 1817), was also identified in samples from both aquaculture (N = 40 out of 1,025) and from natural waters (N = 44 out of 1,710). The life stages of American eels identified from several German waters draining to either the Baltic Sea and the North Sea ranged from elver to silver eels. This indicates that stocking with glass eels or elvers must have occurred several times most likely in the period from 1998 to 2002. The application of a fast and precise method for species identification and genetic monitoring of eels delivered for stocking is therefore essential for maintaining the autochthonous species composition in future. European silver eels migrate 6000 km to their supposed spawning area in the Sargasso sea. As the eel is fasting, this intense swimming activity is realised only with fat stores, involving mainly red muscle i.e. aerobic metabolism. However, eel migration is performed at depth and thus in cold water, both being known to induce changes in muscle energy metabolism. During migration, white and red muscles can operate together or separately in order to counteract the eventual effects of low temperatures and/or high pressures. We have studied the temperature sensitivity (5, 15, and 25 °C) of aerobic and anaerobic metabolism in both sexes. At the same temperature, migrating eels have a higher basal glycolytic flux. Moreover, there are temperature and sex effects: anaerobic glycolysis (JB) is more sensitive to cold water whereas aerobic (JA) is more affected by warm. Males, which are less sensitive to cold water, also have higher aerobic fluxes than females. As depth corresponds to low temperature, the possibility that males migrate more deeply than females is discussed. In an ecophysiological context, it is interesting to suppose that males and female eels migrate at different depths in order to optimize their energy utilization by aerobic and / or anaerobic pathways.  相似文献

Summary The development of the pituitary gland and its blood vessels is described in rat embryos (gestational day 12 through day 21) injected with India-ink via the umbilical vessels. The vascularization of all components of the pituitary gland develops from the surface network covering the prosencephalic vesicle. However, vascular connections exist between the prospective median eminence and the anterior pituitary gland in the earliest stages examined (day 12) but are not augmented by vessels from the stomatodeal roof until day 13. Primary portal veins appear initially on day 13, the vascularization of the pars distalis is visible on day 15. The Mantelplexus covering the floor of the diencephalon is discernible on day 16. Large-caliber portal veins appear immediately before birth, but otherwise there is no significant change in the vascular pattern during the last five days of gestation. The pars intermedia and the median eminence-pituitary stalk region remain avascular throughout embryonic life.  相似文献

Summary Colour change in the eel resulted in marked ultrastructural changes in the pre-dominating (Type II) secretory cells of the pars intermedia of the pituitary. The effects on these cells of transferring eels from white to black backgrounds for periods of up to 56 days were: a) hypertrophy of the rough endoplasmic reticulum, which increased from 12 to 35% of the cytoplasmic volume; b) loss of secretory granules which decreased from 38 to 5% of the cytoplasmic volume; c) development of a system of fine (25–35 nm) tubules located especially at the secretory poles of the cells but also found in the region of the Golgi apparatus. The tubules were seen to connect with the plasma membrane, with the limiting membrane of the secretory granules, and in one instance to connect a granule with the plasma membrane. After glutaraldehyde fixation at pH 5, electron dense material similar to that found in the secretory granules was observed in the lumen of many of the tubules.The changes that occurred in black background eels are taken to indicate that the Type II cells of the pars intermedia are responsible for MSH secretion, particularly since these changes were reversed by returning eels to white backgrounds. The cytoplasmic tubules found in Type II cells may indicate a process for MSH release which does not involve granule extrusion, but rather direct transport of material from the Golgi apparatus to the cell membrane.The electron microscope facilities used in this investigation were funded by the Medical Research Council.  相似文献

Dispersal, one of the most important processes in population ecology, is an issue linking physiological and behavioral features. However, the endocrine control of animal dispersal remains poorly understood. Here, we tested whether and how thyroid hormones may influence dispersal in glass eels of Anguilla anguilla, by testing their influence on locomotor activity and rheotactic behavior. Glass eels were caught during their estuarine migration and treated by immersion in either a l-thyroxine (T(4)) or a thiourea (TU) solution. As measured by radioimmunoassay, T(4) and TU treatments induced, respectively, increased and decreased whole-body thyroid hormone levels relative to untreated controls. We tested a total of 960 glass eels distributed into control, and T(4) and TU treatment groups, on their swimming behavior in experimental flume tanks equipped with upstream and downstream traps that allowed us to concurrently measure both the locomotor activity and the rheotactic behavior. Compared to controls, locomotor activity significantly increased among the hyperthyroid, T(4)-treated eels, but significantly decreased among the hypothyroid, TU-treated eels. The results on rheotactic behavior suggested a more complex regulatory mechanism, since TU but not T(4) treatment significantly affected rheotactic behavior. The influence of thyroid hormones on locomotor activity suggests a central role for these hormones in the regulation of mechanisms leading to the colonization of continental habitats by glass eels. Thyroid hormones are also implicated in the control of locomotor activity in mammals and migratory behavior in birds, suggesting that these hormones represent conserved, proximate mediators of dispersal in vertebrates.  相似文献
