Hydrolysis of plant seed gums by microwave irradiation   总被引:5,自引:0,他引:5  
Under microwave irradiation (MW), the seed gums, guar and Ipomoea quamoclit were hydrolyzed to constituent monosaccharides and oligosaccharides in very mild conditions and short reaction time. Under MW both the seed gums could be completely hydrolyzed using very dilute acid (0.00625N H2SO4) within two minutes. Hydrolysis occurs in 2 min and 20 s even in absence of acid under the MW irradiation. Thus hydrolytic fragmentation under MW provides an efficient tool in structural elucidation of polysaccharides.  相似文献

The seed gum solutions of Ipomoea purga, Ipomoea palmata, Ipomoea dasysperma, Cyanaposis tetragonolobus (Guar gum) and Crotolaria medicaginea were microwave (MW) irradiated and their degradation to oligo and monosaccharides was investigated. The gum solutions were fragmented into oligosaccharides/constituent monosaccharides depending upon the length of MW exposure in presence of catalytic amount of mineral acid or even when no acid was used. A mechanism for the microwave induced hydrolytic degradation of the seed gums has been proposed. The MW exposure time required for the partial and complete degradation of the gums was found dependent on the types of the linkages and degree of the branching present in the gums.  相似文献

Using a catalytic amount of potassium persulfate (1.48 x 10(-4)M), eight different seed gums were fully hydrolyzed on alumina support under microwave irradiation. The hydrolysis time varied between 1.33 and 2.33 min depending upon the seed gum structure. The used solid support could be easily separated from the hydrolyzates and recycled. However, under microwave field in an aqueous medium, the same amount of persulfate was unable to hydrolyze the seed gums. Solid-supported microwave hydrolysis has been compared with the microwave-enhanced aqueous hydrolysis (using K2S2O8 or 0.1N H2SO4) and also with the conventional hydrolysis procedures.  相似文献

Several techniques are available to study gene function, but many are less than ideal for soybean. Reverse genetics, a relatively new approach, can be utilized to identify novel mutations in candidate genes; this technique has not produced an allelic variant with a confirmed phenotype in soybean. A xyloglucan (XG) was extracted from the seeds of Hymenaea courbaril (Jatobá) harvested from trees growing at different locations in Brazil. They were from Foz do Chopin Forest Reserve, State of Paraná (HC-I), Cuiaba (HC-II) and Sinope/State of Mato Grosso (HC-III), Natal/State of Rio Grande do Norte (HC-IV), Fortaleza/State of Ceará (HC-V) and Tamarindus indica (TM) from Conceição de Almeida/State of Bahia, Brazil. It was submitted to SEC-MALLS analysis and its intrinsic viscosity determined. When a solution of XG was heated to 85 °C for 2 h, there was a reduction of aggregation and the XG from the seeds of H. courbaril had a higher value of C when compared with that of from T. indica, probably due to the presence of an unique oligossacharide series present in the H. courbaril samples (XXXXG). This gives rise to a more rigid molecule when compared with the XG from T. indica.  相似文献

Twenty cyclopropenes were prepared and their anti-ethylene activity was evaluated in the fruit ripening and plant senescence bioassays. The treatment of banana fruits with developed antagonists led to delay of fruit ripening. Some antagonists were capable to extend exhibition life of cut mini carnation flowers as well as delay senescence of bean leaves. The potency of ethylene antagonists declined with increase in molecular weight and reduction in their solubility in water, irrespective of bioassay used. Aims: The aim of this work was to analyse the coagulant and antibacterial activities of lectin isolated from Moringa oleifera seeds that are used for water treatment. Methods and Results: The water‐soluble M. oleifera lectin (WSMoL) was separated from nonhemagglutinating components (NHC) by chitin chromatography. WSMoL fluorescence spectrum was not altered in the presence of ions that are often present in high concentrations in polluted waters. Seed extract, NHC and WSMoL showed coagulant activity on a turbid water model. Both NHC and WSMoL reduced the growth of Staphylococcus aureus, but only WSMoL caused a reduction in Escherichia coli. WSMoL was also more effective in reducing the growth of ambient lake water bacteria. Conclusions: Data obtained from this study indicate that WSMoL is a potential natural biocoagulant for water, reducing turbidity, suspended solids and bacteria. Significance and Impact of the Study: Moringa oleifera seeds are a material effective in the treatment of water.  相似文献

Abstract

Soybean [Glycine max (L.) Merrill] meal is one of the most important plant-based protein sources in the world. Developing cultivars high in seed protein concentration and seed yield is a difficult task because the traits have an inverse relationship. Over two decades ago, a protein quantitative trait loci (QTL) was mapped on chromosome (chr) 20, and this QTL has been mapped to the same position in several studies and given the confirmed QTL designation cqSeed protein-003. In addition, the wp allele on chr 2, which confers pink flower color, has also been associated with increased protein concentration. The objective of our study was to evaluate the effect of cqSeed protein-003 and the wp locus on seed composition and agronomic traits in elite soybean backgrounds adapted to the Midwestern USA. Segregating populations of isogenic lines were developed to test the wp allele and the chr 20 high protein QTL alleles from Danbaekkong (PI619083) and Glycine soja PI468916 at cqSeed protein-003. An increase in protein concentration and decrease in yield were generally coupled with the high protein alleles at cqSeed protein-003 across populations, whereas the effects of wp on protein concentration and yield were variable. These results not only demonstrate the difficulty in developing cultivars with increased protein and yield but also provide information for breeding programs seeking to improve seed composition and agronomic traits simultaneously.

对7个具有代表性种源的白皮松种子的休眠特性研究表明,白皮松种子种皮的透水性在种源间差异明显,表现出随地理纬度的增高而逐渐增大的趋势,分布越是偏北的种源透水性趋于加强。根据种皮的透水性,7个种源可被分为差异明显的2组,组间以秦岭为界。不同种源种子种皮的透气性以外种皮造成的差异为最大,内种皮次之。种子的透气性与透水性表现出的变化趋势相同。不同种源种子浸提液的抑制能力显著不同,而且这种差异随浸提液浓度的  相似文献

This paper describes the genetic control of two new water-soluble proteins in barley. Water-soluble proteins (WSPs) of mature barley seed form part of the albumin/globulin class of seed proteins. They can be extracted from hand-milled grain with water, though some WSPs are more efficiently extracted with a solution of 10 mM dithiothreitol. Polymorphisms for WSPs were detected in isoelectric focusing gels incorporating various ampholine combinations. Two new controlling genes (Wsp4 andWsp5) have been identified and located using wheat/barley chromosome addition lines and barley doubled haploids.Wsp4 is located on chromosome 2 (2H), andWsp5 was found to be tightly linked toWsp2 on the long arm of chromosome 7 (5HL). Segregation of a sixth gene (Wsp6) is also described, but this has not been mapped. Two major initiatives are under way to correct the beta-cell deficit of diabetes: one would generate beta-cells ex vivo that are suitable for transplantation, and the second would stimulate regeneration of beta-cells in the pancreas. Studies of ex vivo expansion suggest that beta-cells have a potential for dedifferentiation, expansion, and redifferentiation. Work with mouse and human embryonic stem (ES) cells has not yet produced cells with the phenotype of true beta-cells, but there has been recent progress in directing ES cells to endoderm. Putative islet stem/progenitor cells have been identified in mouse pancreas, and formation of new beta-cells from duct, acinar and liver cells is an active area of investigation. Peptides, including glucagon-like peptide-1/exendin-4 and the combination of epidermal growth factor and gastrin, can stimulate regeneration of beta-cells in vivo. Recent progress in the search for new sources of beta-cells has opened promising new opportunities and spawned clinical trials.  相似文献

C-α and C-β, previously isolated from seed of Phaseolus coccineus, are shown respectively to be the bis-O-isopropylidene and the 16,17-mono-O-isopropylidene derivatives of ent-6α,7α,16β,17-tetrahydroxykauranoic acid. By GC-MS characterization of the products of acidic, basic and enzymatic hydrolysis, water soluble conjugates of the following compounds have been shown to occur in P. coccineus seed: GA8, GA17, GA20, GA28, ent-6α,7α,13-trihydroxykaurenoic acid, ent-6α,7α,17-trihydroxy-16β-kauranoic acid, ent-6α,7α,16β,17-tetrahydroxykauranoic acid, 7β,13-dihydroxykaurenolide and abscisic acid.  相似文献

New enzyme-based method for analysis of water-soluble wheat arabinoxylans   总被引:1,自引:0,他引:1  
Arabinoxylans (AX) are the predominant cell-wall polysaccharides in wheat flour. Water-extractable AX are essential for dough and bread properties and performance. However, there is no specific and accurate way of determining the content and structure of AX. An enzyme-assisted method employing an efficient enzyme mixture for the total hydrolysis of AX was developed in the present work. Enzymatic hydrolysis (EH) is a gentle method during which no unwanted sugar destruction occurs. Following EH, liberated monosaccharides were analysed by gas chromatography (GC) and liquid chromatography using HPAEC-PAD. The results were compared with acid methanolysis (AM) and acid hydrolysis (AH). EH performed better on commercially isolated AX samples than the reference method AM. Its action in the water extract from wheat flour was also more efficient than that of AM and comparable to the efficiency of AH. HPAEC-PAD revealed a significant amount of fructose in the water extract following EH, originating from fructans in wheat flour not detected in the GC analysis. The wheat flour examined contained 0.29% water-extractable AX. The arabinose/xylose ratio was 0.32. The enzyme-based method developed is applicable for comparison of different wheat flours and can be used in evaluating the effect of processing on the content and structure of water-extractable AX.  相似文献

The erythroagglutinating activity of purified Vicia faba lectin was enhanced in the presence of gums; gum guar caused the highest enhancement. Circular dichroism probe demonstrated 40-57% beta-conformation and 4-23% alpha-conformation of the lectin at pH 7.2 depending upon the analytical methods used. The beta-conformations of untreated and modified V. faba lectins were increased in the presence of gums. The mixing of gum guar with lectin and with modified lectin, respectively, led to the highest values of beta-conformational change in the protein molecule, thereby increasing the number of receptor sites of the lectin molecule. The enhancement of the activity of V. faba lectin in the presence of gum guar might be due to the conformational change of the protein molecule.  相似文献

在已发表的“种子地理学”基本理论框架和内涵的基础上, 该文对种子地理学理论进行了补充和修订, 在以前提出的7个方面的研究内容基础上, 补充了土壤种子库的地理学、种子雨的地理学、种子散布方式的地理学三个方面的研究内容, 提出了“土壤种子库的三向(纬度、经度和海拔)梯度变化假说”和“种子散布模式的三向梯度变化假说”。还对以前提出的6个方面的研究内容进行了增补, 在“种子化学性状地理学”方面添加了种子稳定性同位素性状, 在“种子重量的地理学”方面添加了油质体性状, 在“种子形态性状的地理学”方面补充了种子长、种子宽、种子长宽比、种子的附属结构(如果翅的有无以及种子附属物种翅长、种翅宽、种翅长宽比)等性状, 在“花和果实的地理学”方面增添了一些新的关注对象, 如花瓣的长和宽、花萼长和宽, 还有果实的长、宽、高及其比值, 夏季成熟的果实及秋天成熟的果实比例等, 在“种子遗传学性状的地理学”方面添加了DNA含量这个性状, 在“种子细胞性状的地理学”方面增添了淀粉粒。  相似文献

A water-soluble seed gum was isolated from seed endosperm of Cassia javahikai. The acid-catalyzed fragmentation, methylation, selective enzymatic degradation and periodate oxidation suggested a heteropolymeric structure for the polysaccharide. The polysaccharide was shown to have a linear chain of β(1 → 4) linked d-mannopyranosyls units with side chains of α(1 → 6) d-galactopyranosyl units. Grafting of polyacrylamide onto the gum was performed using K2S2O8/ascorbic acid redox system in presence of Ag+ as catalyst at 35 ± 2 °C. The viscosity of the gum solution increased on grafting and the grafted gum was observed to resist biodegradation for more than 256 h. Thermogravimetric analysis revealed that grafted gum was more thermally stable than native gum.  相似文献
