Ionic responses to alteration in external and internal pH were examined in an organism from a marine-like environment. Vacuolar pH (pHv) is about 4.9–5.1, constant at external pH (pHo) 5–8, while cytoplasmic pH (pHc) increases from 7.3 to 7.7. pHc regulation fails above pHo 9, and this is accompanied by failure of turgor regulation. Na+ increases above pHo 9, while K+ and Cl decrease. These changes alone cannot however explain the alterations in turgor. The mayfly Leptophlebia marginata was exposed to different concentrations of Fe2+ or Pb2+ at pH 4.5 and pH 7.0. The effects of the metals on escape behavior and survival of the mayflies were investigated during an exposure of 120 hours.
  1. Whole-body metal loads (Fe; Pb) of the mayflies increased in a dose-dependent way at both pH levels. A significant effect of pH on metal concentration in the mayflies was only found for Pb (p < 0.001).
  2. In terms of mortality, both metals were more toxic at pH 4.5 than at pH 7. The 96 h-LC50 values for Fe were 106.3 mg Fe l-1 at pH 7 and 89.5 mg Fe l-1 at pH 4.5. Those for Pb were > 5 mg Pb l-1 at pH 7 and 1.09 mg Pb l-1 at pH 4.5.
  3. The mayflies lost their escape behavior, when exposed to the metals, the effects being more pronounced at low than at circumneutral pH for both metals (p < 0.05). The 96 h-EC50 values for Fe were 70.0 mg Fe l-1 at pH 7 and 63.9 mg Fe l-1 at pH 4.5.

Several pentavalent antimony compounds have been used for the treatment of leishmaniasis for decades. However, the mechanism of these antimony drugs still remains unclear. One of their targets is thought to be trypanothione, a major low molecular mass thiol inside the parasite. We show that pentavalent antimony (SbV) can be rapidly reduced to its trivalent state by trypanothione at mildly acidic conditions and 310 K (k=4.42 M–1 min–1 at pH 6.4), and that SbIII can be bound to trypanothione to form an SbIII-trypanothione complex. NMR data demonstrate that SbIII binds to trypanothione at the two thiolates of the cysteine residues, and that the binding is pH dependent and is strongest at biological pH with a stability constant logK=23.6 at 298 K (0.1 M NaNO3). The addition of low molecular monothiol ligands such as glutathione and cysteine to the SbIII-trypanothione complex results in the formation of a ternary complex. Thiolates from both trypanothione and monothiol bind to the SbIII center. The formation of the ternary complex is important, as the antileishmanial properties of the drugs are probably due to a complex between of SbIII-trypanothione and enzymes. Although thermodynamically stable, the complex is kinetically labile and the free and bound forms of thiolates exchange on the 1H NMR timescale. Abstract. Survival and death of Chara internodal cells were investigated in one of the alkali metal salts KCl, some of the alkali earth metal salts CaCl2, Ca(NO3)2, MgCl2, Mg(NO3)2, SrCl2, Sr(NO3)2, BaCl2 and Ba(NO3)2, potassium phosphate pH buffer solution (pH 7.0), Tris-maleate pH buffer solution (pH 7.0), HEPES (N-2-hydroxyethylpiperazine-N′-2-ethanesulphonic acid)-KOH (pH 7.0) pH buffer solution, calcium buffer solutions, and deionized water. Most of the internodal cells died within a day or a few days in KCl, MgCl2, Mg(NO3)2, BaCl2 and Ba(NO3)2 solutions of higher concentrations, calcium buffer solutions of pCa 6.0, 10.0 mol m-3 potassium phosphate pH buffer solution and 10.0 mol m-3 Trismaleate pH buffer solution. However, all of the internodal cells survived more than 10 d in deionized water, 80.0 mol m-3 CaCl2, 80.0 mol m-3 Ca(NO3)2, 80.0 mol m-3 SrCl2, 80.0 mol m-3 Sr(NO3)2 calcium buffer solutions of pCa 4.0 and pCa 5.0, and 10.0 mol m-3 HEPES-KOH (pH 7.0) pH buffer solution. Addition of Ca2+ or Sr2+ to K+, Mg2+ and Ba2+ salt solutions increased the survival rates of the internodal cells. Calcium release from the internodal cell wall was measured in deionized water, KCl, NaCl, MgCl2, CaCl2, SrCl2 and BaCl2 solutions. Except in deionized water and CaCl2 solution, most of the calcium binding to the cell wall was released within one or a few hours in respective electrolyte solutions. Thus, survival and death of the internodal cells in the electrolyte solutions tested were interpreted in terms of the calcium release from the cell wall and the cell membrane, and intrinsic ability of Sr2+ to maintain the cell membrane normal.  相似文献

The haptophyte microalga Tisochrysis lutea was heterotrophically grown in F2 medium with different combinations of pH and salinity. Growth, oil content and fatty acids (FAs) profile were determined under each set of conditions. The salinity was adjusted using NaCl at concentrations of 0.4, 0.6, 0.8, or 1.0 M, while pH was adjusted at 7, 8, or 9, and heterotrophic growth was performed using organic carbon in the form of sugar cane industry waste (CM). Fatty acid methyl esters (FAMEs) were identified by gas chromatography. The results showed that pH of 8.0 was the optimal for dry weight and oil production, regardless of the salinity level. At pH 8.0, growth at a salinity of 0.4 M NaCl was optimal for biomass accumulation (1.185 g L-1). Under these conditions, the maximum growth rate was 0.055 g L-1 d-1, with a doubling time of 17.5 h and a degree of multiplication of 2.198. Oil content was maximal (34.87%) when the salinity was 0.4 M and the pH was 9.0. The ratio of saturated to unsaturated FAs was affected by the pH value and salinity, in that unsaturated FAs increased to 58.09% of the total FAs, considerably greater than the value of 40.59% obtained for the control (0.4 M NaCl and pH 8.0).  相似文献

One-year-old Prunus avium L. were grown under greenhouse conditions in a Countesswells soil in all combinations of 2 pH and 2 P levels. The soil, obtained from a long-term liming and fertilizer experiment, provided pH values throughout the experiment of 3.75–3.99 (pH 1) and 4.81–5.41 (pH 2). The P treatments had 0.43% acetic acid extractable P of 31–44 g g-1 (P1) and 145–173 g g-1 (P2). The trees were harvested 92 (H1), 134 (H2), and 168 (H3) days after initiation of growth.Top (leaf+new stem) dry weight was significantly increased for pH 2 and P2 at H2 and H3. P2 also increased leaf weight (H1), the weight of the original stem-root (H2 and H3), and root length but decreased root diameter at both soil pHs (H2 and H3). Total tree uptake of N, P, K, Ca, and Mg was also increased by pH-P combinations which had significantly greater dry matter production and root length. Total Mn uptake decreased at pH2. Root nutrient inflows (uM m-1 day-1) were increased for Ca at pH2 and for P at P2. Mn inflow decreased at pH2 and at pH1 P2 although the increased root length associated with the latter treatmen resulted in increased total tree Mn uptake. In general, high nutrient inflows occurred in all trees at H1 and in severely stunted trees at pH1 P1; both had larger than average root diameters.  相似文献

31P-Nuclear-magnetic-resonance spectra of maize (Zea mays L.) root tips, that had been induced to extrude large amounts of H+ in response to fusicoccin (FC) in the presence of potassium salts, indicate that the cytoplasmic pH does not become higher than that of controls. In fact, the cytoplasmic pH may become slightly (approx. 0.1 pH unit) lower in cells extruding H+. Estimations of the buffer capacity of the cells show that without active intracellular pH regulation, H+ extrusion caused by FC would cause the intracellular pH to rise by at least 0.6 pH unit h-1. The intracellular pH of the halotolerant green algae Dunaliella tertiolecta, was determined by the distribution of 5,5-dimethyl-2(14C)-oxalolidine-2,5-dione (DMO) between the cell and the surrounding medium. 5,5-dimethyl-2(14C)oxalolidine-2,4-dione was not metabolized by the algal cells. The intracellular pH of Dunaliella tertiolecta was 6.8 in the dark and 7.4 in the light. During a salt stress, after two hours, the intracellular pH was increased by 0.2 pH units in both light and dark. The salt stressed cells maintained a constant pH of about 7.5 over the pH range of 6.5 to 8.5. Because of the relatively low permeability coefficient of the plasma membrane for DMO, this technique does not permit rapid pH determinations during the induction period after a salt stress. Experiments with free cell biomass (cells + exopolysaccharides) ofRhizobium BJVr 12 (mungbean isolate) showed that amount ofCr3+ ion sorbed is influenced by the amount of biomass toCr3+ concentration ratio and time of contact. A ratio of 0.5 gfresh biomass to 10.0 ml 5.03 ppm Cr3+ sorbed 0.0275 mg Crequivalent to an uptake of 2.86 mg Cr g-1 dry biomass and 1.0g: 10.0 ml sorbed 0.0366 mg Cr equivalent to an uptake of 1.9 mg Crg-1 biomass. Immobilized cell biomass in ceramic beads and inaquacel (a porous cellulose carrier with a charged surface) were moreefficient than free cell biomass in adsorbing Cr(III). A reduction of49.7percnt; of Cr(III) for free cells, 95.6% for cells immobilized inceramic beads and 94.6% for cells in aquacel was achieved after 48hours under shaken conditions. Sorption capacities of immobilized cellbiomass in ceramic beads and aquacel ranged from 5.01 to 5.06 mg Crg-1 dry cell biomass. The biosorption of Cr3+follows generally the Langmuir and Freundlich models of adsorption at lowCr3+ concentrations. The Langmuir constant for immobilizedcells in ceramic beads are: Q0, 0.065 mmol Crg-1 biomass; b (affinity constant), - 694 lmmol-1 Cr and for cells in aquacel Q, 0.07 mmol Crg-1 biomass; b, - 694 l mmol Cr g-1 Cr. TheFreundlich constants are: K, 0.071 mmol Cr g-1 biomass; n,0.13 g-1 biomass l-1 and for aquacel: K, 0.074mmol g-1 biomass; n, 0.13 g-1 biomass. Biotrapsmade up of immobilized cells in ceramic beads and aquacel were tested foradsorbing Cr(III) using two different flow rates: 0.5 ml/min and 1.5 ml/min.A significantly higher amount of Cr(III) was adsorbed at the lower flow rateof 0.5 ml/min. Biosorption of Cr3+ is competitive. Thetreatment of a waste water sample containing 6.03 ppm Cr3+ andother cations with the biomass reduced the Cr3+ concentrationto that much lower than for the test solution containing only Cr. Recoveryof biosorbed Cr(III) was by treatment at a different pH using dilute HClsolution. Recovery was higher for cells imbibed in ceramic beads thanaquacel. Percentage recoveries for cells in aquacel are 46.4% at pH1.0, 33.0% at pH 3.0 and 6.6% at pH 6.0–7.0. For cellsin ceramic beads, percentage recoveries are: 93.1% at pH 1.0,75.6% at pH 3.0 and 16.4% at pH 6.0–7.0. The regulation of intracellular neuronal pH and pH from the extracellular space was studied in the isolated stomatogastric ganglion of the crab Cancer pagurus. Intracellular neuronal pH was found to be 0.3–0.4 pH units more acidic than the standard bath pH of 7.4 and surprisingly, the extracellular space pH was found to be around 0.1 pH units more alkaline than the bath pH. Extracellular space pH shifts in response to bath pH changes decreased as a function of the depth of the recording site within the ganglion, suggesting the existence of restrictions in the free diffusion of H+. The amplitude of these pHe shifts increased in Na+-free saline or with amiloride, suggesting Na+-dependent regulation of the extracellular space pH. In Na+ free saline or in the presence of amiloride, intracellular pH recovery from an NH4Cl induced acidosis was reduced, and the H+ muffling capacity (cf. Thomas et al. 1991) of the extracellular space was markedly reduced. A field growth chamber study was conducted to determine the effects of ozone and simulated acid rain (SAR) on soil heavy metals. Loblolly pine (Pinus taeda L.), grown in open-top chambers, was exposed to three concentrations of ozone (charcoal filtered air with 0.026 µL O3 L-1, and two non-filtered treatments in which ozone concentrations were 0.074 µL L-1 and 0.147 µL L-1, respectively) and two levels of SAR (pH 3.5 and 5.2). Ozone was applied for 12 h d-1 for 9 months and acid rain deposition was 125 mm event-1. After 9 months exposure, soil pH, organic matter and DTPA-extractable heavy metals (Cd, Pb, Zn, Mn, Fe, Cu) were determined on soil samples collected from exposed chambers at two depths (0–15 cm and 15–30 cm). Simulated acid rain decreased the original soil pH. The concentrations of Cd, Pb and Mn at SAR pH 3.5 were significantly higher than at SAR pH 5.2. Ozone did not affect Zn, Fe and Cu, but a significant interaction between pH and O3 on Mn, Pb and Cd was observed. Due to the poor drainage capacity of this soil, leaching of heavy metals was not observed.  相似文献

Growth yield of the halotolerant bacterium A505 was increased by the supplement of Na+, K+, or Rb+ into the culture media with pH 7.5, and inhibited by Li+ or Cs+. In the presence of less than 0.1 M NaCl or KCl alkaline growth media, pH 9.2 to 9.7, afforded optimal growth of this strain. Intracellular ion content of this microbe changed reflecting on the Na+ or K+ concentration in the media, although it tended to accumulate K+ and extrude Na+ in the media without NaCl supplemented. A 1.2 to 1.4-fold stimulation of in vitro NADH oxidase activity was obtained by supplement of salts, except for LiCl. The rate of NADH oxidation in the absence of salts correlated with the pH and showed clear maxima at pH about 8, irrespective of growth conditions. In the presence of 0.5 M NaCl or KCl, on the other hand, pH dependence was less significant and showed only a flat maximum at pH around 7. The interaction of bovine prothrombin with Ca2+ and Mg2+ ions was investigated by following H+ release as a function of metal ion concentration at pH 6 and pH 7.4 at high and low ionic strength. Prothrombin Ca2+ and Mg2+ binding is characterized by high- and low-affinity sites. M2+ binding at these sites is associated with intramolecular conformational changes and also with intermolecular self-association. The pH dependence of H+ release by M2+ is bell shaped and consistent with controlling pKa values of 4.8 and 6.5. At pH 6 and low ionic strength, both Ca2+ and Mg2+ titrations following H+ release clearly show independent low- and high-affinity binding sites. Laser light scattering reveals that at pH 7.4 and low ionic strength, and at pH 6.0 and high ionic strength, the prothrombin molecular weight is between 73 and 98 kD. At pH 7.4 and high ionic strength, prothrombin is monomeric in the absence of metal ions, but appears to dimerize in the presence of M2+. At pH 6.0 and low ionic strength prothrombin exists as a dimer in the absence of metal ions and is tetrameric in the presence of Ca2+ and remains dimeric in the presence of Mg2+. Understanding the chemical speciation of metals in solution is necessary for evaluating their toxicity and mobility in soils. Soil samples from the Powder River Basin, Wyoming were extracted with distilled deionised H2O. Soil water extracts were subjected to chemical speciation to determine the relative distribution and chemical forms of copper (Cu), zinc (Zn) and lead (Pb) in acidic environments. As pyrite oxidised, the pH decreased from 6.6 to 2.4, concentration of dissolved sulfate (ST) increased from 259 to 4,388 mg L-1 and concentration of dissolved organic carbon (DOC) decreased from 56.9 to 14.4 mg L-1. Dissolved Cu concentrations ranged from 0.06 to 0.42 mg L-1 and dissolved Zn concentrations ranged from 0.084 to 4.60 mg L-1. Dissolved concentrations of Pb were found to be 0.003 to 0.046 mg L-1. Chemical speciation indicated that at near neutral pH, dissolved metal concentration in soil water extracts was dominated by DOC- metal complexes. At low pH, dissolved metal concentration in soil water extracts was dominated by free ionic forms (e.g. Cu2+, Zn2+, Pb2+) followed by ion pairs (e.g. CuSO4 0, ZnSO inf4 sup0 , PbSO in4 sup0 ). Results obtained in this study suggest that as soil pH decreased, the availability and mobility of metal ions increased due to the chemical form in which these metal ions are present in soil solutions.  相似文献

Phosphoglycolate phosphatase (EC 3.1.3.18), isolated from maize leaf bundle sheath, was purified 200-fold to a specific activity of about 99 mol mg-1 protein · min-1. The purification procedure included Sephadex G-75 filtration, and diethylaminoethyl-cellulose and Phospho-Ultrogel A6R chromatography. This partially purified enzyme exhibited optimum activity over a broad pH range, from pH 6.3 to pH 8.0. It displayed a very high degree of specificity for phosphoglycolate and required a divalent cation to be active; Mg2+ was the most effective activator. Saturation curves of the Michaelis-Menten type were observed both with phosphoglycolate (Km=0.57 mmol·l-1) and with Mg2+ (Km=0.015 mmol·l-1). The activation constant for Mg2+ was unchanged when the pH was raised from 7.0 to 8.0. These results indicate that variations of stromal pH and Mg2+ during the darklight transition could not directly modifity the activity of the phosphoglycolate phosphatase in maize bundle-sheath chloroplasts. The undissociated protein showed a pI of 4.95, as determined by isoelectric focusing. For the native phosphatase a molecular mass of about 61 500 Da was estimated by polyacrylamide gradient gel electrophoresis. The subunit was found to have a relative molecular mass of 31 500 Da by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The potential of alginate-immobilized Anabaena doliolum and Chlorella vulgaris was assessed for removal of nutrients (NO inf3 sup- and NH inf4 sup+ ) and metals (Cr2O inf7 sup2- and Ni2+) at different biomass concentrations (0.05, 0.1, 0.25, 0.49 and 1.22 g dry wt l-1) and pH values (4 to 10). Though uptake of all these substances was higher in concentrated algal beads (0.25, 0.49 and 1.22 g dry wt l-1), their rate of uptake was significantly (P<0.001) lower than that of low (0.05 g dry wt l-1) cell density beads. For A. doliolum, there was no significant difference in uptake rates for beads having densities of 0.05 and 0.1 g dry wt l-1. Chlorella vulgaris, however, showed maximum efficiency at 0.1 g dry wt l-1. Uptake of both the nutrients and the metals was maximal at pH 7 followed by pH 8, 6, 9, 10, 5 and 4. Of the different substances (organic acids and divalent cations) used, humic acid was most efficient in decreasing metal uptake. Mg2+ was, however, more efficient than Ca2+ in decreasing Ni2+ uptake. Immobilized algae with a cell density of 0.1 g dry wt l-1 were the most efficient for nutrient and metal removal at pH 6 to 8.  相似文献

The uptake of K+ by the leafy shoots of four submersed higher aquatic plants (Elodea canadensis, Ranunculus aquatilis, R. trichophyllus, and Callitriche hamulata) with different HCO3 - affinity was measured in successive 2-h periods under the conditions of high or low photosynthetic rates (i.e. at pH 7.5 or 10). At pH 7.5 the uptake of K+ by species with the higher HCO3 - affinity (E. canadensis, R. trichophyllus) was significantly faster than that by species with a lower HCO3 - affinity (R. aquatilis, C. hamulata). In the former group of species, the K+ uptake rate at pH 7.5 was 1.7 - 3.5 times higher than at pH 10. At pH 10, the soft-water species, R. aquatilis, had the lowest net photosynthetic rate (PN) of the three HCO3 - users but, in contrast to the relative hard-water species, R. trichophyllus, showed a small K+ efflux (47 nmol kg-1 s-1). Thus, K+ uptake by shoots was not strictly correlated with PN. A significant K+ efflux (73 - 86 nmol kg-1 s-1) occurred from all HCO3 - users in darkness. The relatively low K+ uptake by the strict CO2 user, C. hamulata, was quite independent of PN and light or darkness. It may be suggested that uptake of K+ by shoots of submersed plants depends on their HCO3 - affinity. Glomus aggregatum and Leucaena leucocephala were allowed to interact in a manganese-rich oxisol at pH 4.3–6.0 and at soil P concentrations considered optimal for mycorrhizal host growth and sufficient for nonmycorrhizal host growth. At 0.02 mg P l-1, vesicular-arbuscular mycorrhizal fungal (VAMF) colonization of roots increased as soil pH increased from 4.3 to 5.0. However, VAMF colonization of roots did not respond to further increases in pH. At pH 6.0, growth of mycorrhizal Leucaena observed at 0.02 mg P was comparable with that observed at 0.8 mg P l-1. Increasing P concentration from 0.02 to 0.8 mg P 1-1 increased target soil pH from 4.3 to 4.7 and reduced the concentration of available soil Mn from 15.1 to 1.9 mg 1-1. Thus, the normal plant growth observed at the higher P concentration at pH<5 was mainly due to the alleviation of Mn toxicity as a result of its precipitation by excess P. VAMF colonization levels observed at pH 5.0–6.0 were similar, but maximal plant growth occurred at pH 6.0, suggesting that the optimal pH for mycorrhizal formation was substantially lower than for VAMF effectiveness. The death of the hybridoma VO 208 in a continuous culture at pH 7 and 6.8 was investigated by measuring both the appearance of visible dead cells which do not exclude the trypan blue dye and the release of lactate dehydrogenase (LDH) in the culture medium. The intracellular LDH was found to be completely released either when live cells lysed or when they were transformed into visible dead cells. No significant lysis of blue dead cells could be observed at the two different pH. Using a LDH balance over the culture system, cell lysis was found negligible at pH 7, but accounted for 20% of the total cell death at pH 6.8. A methodology is proposed to evaluate the rate constants of hybridoma lysis and total death. Micropropagated Choisya, Daphne, Delphinium, Hemerocallis, Hosta, Iris and Photinia were found to adjust the pH of Murashige and Skoog's plant tissue culture medium (initial pH 5.6 or 3.5) to different values depending on the species.
