Harpin的表达及其诱导抗烟草花叶病毒感染的活性 总被引:7,自引:0,他引:7
采用人工分段合成梨火疫病细菌(Erwinia amylovora)Harpin基因A、B、C、D片段,然后连接构建全长harpin基因,克隆到pET-23(+)表达载体中,在E.coli BL21中表达Harpin蛋白.通过His@Bind Resin Ni++ 层析柱纯化,获得Harpin蛋白.经SDS-PAGE和Western blot分析,该蛋白分子量为45 kDa.采用叶片穿刺法,5μL 30μg/mL Harpin能诱发烟草叶片的过敏反应.采用叶脉注射法,5μL 30μg/mL Harpin能诱发辣椒叶片的过敏反应.将烟草和辣椒用30μg/mL Harpin喷雾处理5d后,接种烟草花叶病毒(Tobacco mosaic virus, TMV),观察植株病症,并采用ELISA检测TMV含量.结果表明经Harpin处理的烟草和辣椒对TMV侵染有一定的抑制作用. 相似文献

目的:克隆烟草富含甘氨酸RNA结合蛋白基因(glycine-rich RNA-binding protein,GRRBPs)并进行原核表达,为制备抗体和研究烟草抗逆性分子机理打下基础。方法:从总RNA中反转录扩增并克隆烟草富含甘氨酸RNA结合蛋白NtRGP-1a和NtRGP-3全长cDNA,将cDNA序列克隆到表达载体pGEX4T-1上,构建了重组表达质粒pGEX4T-1/NtRGP-1a、pGEX4T-1/NtRGP-3,转化大肠杆菌rosetta,IPTG诱导表达,GSTrap 4B亲和层析柱对重组蛋白进行纯化。结果:重组蛋白在大肠杆菌中可以高效表达,SDS-PAGE显示其相对分子质量与预计大小一致,蛋白纯度占总蛋白的95%以上。结论:成功克隆和表达了烟草富含甘氨酸RNA结合蛋白NtRGP-1a和NtRGP-3基因序列,为制备抗体和烟草抗逆性分子机理等进一步的研究奠定了基础。 相似文献

脱水素在胚胎发育后期累积,外源脱落酸(ABA)、低温、干旱和其他一些环境条件下能诱导脱水素的产生,尽管植物在脱水条件下脱水素广泛存在于细胞中,但其生化功能仍不清楚.为研究小麦在不同时期脱水素基因的表达情况和生物学功能及抗体制备,以小麦幼芽为材料,经干旱胁迫处理后,提取总RNA,通过RT-PCR得到小麦类脱水素基因片段(WZY1-1),再连接至克隆载体PUCM-T,并成功构建重组表达质粒PET-32a( )-wzy1-1,将阳性重组质粒转化于受体菌BL21(DE3)感受态细胞中,经IPTG诱导表达,进行表达产物的聚丙烯酰胺凝胶电泳(SDS-PAGE)检测.结果表明,表达蛋白位于37ku处,小麦类脱水素基因获得高效表达.表达蛋白经Ni2 琼脂糖凝胶亲和层析和透析袋电洗脱法纯化后,对兔子进行免疫,制备的抗血清通过ELISA检测到较高的多克隆抗体效价.蛋白质印迹结果显示,利用纯化的蛋白质制备的兔抗血清可以很好地和所表达的蛋白质带特异性结合,且郑引1号小麦幼苗进行干旱处理,提取粗蛋白,SDS-PAGE,蛋白质印迹检测显示,在分子质量28ku处出现特异的蛋白质条带,这说明所制备的抗血清可以与小麦叶片所表达的dehydrin蛋白特异性结合,证明其具有良好的免疫原性. 相似文献

ΔfosB是fosB基因的自然截短型,稳定存在于许多组织中,在脂肪细胞和成骨细胞的形成和分化中起到重要作用。ΔFosB蛋白可能与钙在骨和乳腺中的代谢有关,并且调控钙从骨向乳腺转移的信号通路。将奶山羊的ΔfosB基因亚克隆到pET32a载体得到pET32a-ΔfosB原核表达载体,IPTG诱导其在大肠杆菌中表达,纯化融合蛋白免疫兔子制备多克隆抗体。ELISA检测显示抗体效价达1:51200,Western blotting结果显示制备的抗体能特异性检测原核表达的ΔFosB蛋白以及在HEK-293细胞中表达的ΔFosB蛋白。进一步的组织差异表达检测表明ΔFosB在山羊的乳腺、骨髓、脑髓、肌肉、心脏、肝和肺组织中均有表达。 相似文献

体外实验研究表明配子生成素结合蛋白1(GGNBP1)可能与GGN1相互作用形成睾丸特异性复合物,在精子生成过程中发挥作用.从小鼠睾丸总RNA中反转录扩增Ggnbpl全长cDNA,构建表达质粒,在大肠杆菌中表达GGNBP1,经聚丙烯酰胺凝胶纯化后免疫新西兰白兔,制备兔多抗血清.镍离子金属螯合柱纯化表达的GGNBP1蛋白,与NHS活化基团交联,制备GGNBP1抗体亲和层析柱,纯化GGNBP1多抗.在293FT细胞中瞬时表达Myc-GGNBP1融合蛋白,用于Mvc单抗验证GGNBP1抗体特异性,结果证明获得了特异性的GGNBP1抗体.分别制备小鼠脑、心、肺、肝、脾、肾、肌肉、卵巢、睾丸和子宫组织匀浆,用GGNBP1抗体进行Western印迹分析,结果仅在睾丸组织匀浆中检测到GGNBP1特异性条带,证明GGNBP1是睾丸特异性表达蛋白. 相似文献

基于拟南芥内质网生长素结合蛋白基因的cDNA序列,设计合成了Ap5和Ap3两个引物,应用RT-PCR技术扩增了拟南芥的ABP基因。将该基因克隆在植物表达载体p35SSIN的35S启动子必Nos3‘端之间,得到植物表达载体p35SE。通过农杆菌介导的方法对烟草SR1进行了转化,由分子杂交等检测证明,生长素结合蛋白基因已在烟草中表达,同时转基因烟草后代对生长素的敏感性明显增加。 相似文献

血浆视黄醇结合蛋白(Retinol binding protein,RBP)是体内将视黄醇从肝脏转运至靶组织的特异载体蛋白。最近研究发现在盐度升高时肾脏RBP蛋白表达量下降。为了进一步研究香鱼RBP基因mRNA和蛋白表达与盐度应激相关性,从香鱼、肝脏cDNA文库中获得RBP基因cDNA序列。香鱼RBP基因mRNA在肝脏中表达量最高,肾、肠、脑和鳃中表达次之。实时荧光定量RT-PCR结果显示,盐度升高时,RBP基因mRNA表达在不同组织中呈不同下降趋势,其中渗透压调节相关组织鳃、肾中,表达量下降最显著。Western blot实验证实,盐度升高时,香鱼血清RBP蛋白表达量也显著下降。揭示了RBP可能在香鱼盐度适应中有重要作用。 相似文献

目的:为方便实验室工作中对HIV-1 B′/C亚型Rev蛋白的检测,制备相应的Rev蛋白及其抗体.方法:将我国HIV-1 B′/C亚型流行株的rev基因按大肠杆菌优势密码子进行改造后人工合成,在原核系统中与pET30a(+)载体中的His·Tag、Trx·Tag及S·Tag进行融合表达,目的蛋白经Ni2+金属螯合层析柱纯化后用于免疫家兔,制备多克隆抗体.结果与结论:合成基因在原核系统中融合表达得到相对分子质量约18×103的融合蛋白,目的蛋白的表达量约占菌体总蛋白量的36%;用纯化后的融合蛋白免疫家兔,制备了多克隆抗体,Western印迹及间接免疫荧光检测结果显示,获得的多克隆抗体与HIV-1 B′/C亚型的Rev蛋白能产生特异性反应,可用于检测HIV-1 B′/C亚型Rev蛋白的表达. 相似文献

Background
Smoking and passive smoking are collectively the biggest preventable cause of death in Bangladesh, with major public health burden of morbidity, disability, mortality and community costs. The available studies of tobacco use in Bangladesh, however, do not necessarily employ nationally representative samples needed to monitor the problem at a national scale. This paper examines the prevalence and patterns of tobacco use among adults in Bangladesh and the changes over time using large nationally representative comparable surveys.Methods
Using data from two enumerations of the International Tobacco Control (ITC) Bangladesh Project conducted in 2009 and 2012, prevalence estimates are obtained for all tobacco products by socio-economic determinants and sample types of over 90,000 individuals drawn from over 30,000 households. Household level sample weights are used to obtain nationally representative prevalence estimates and standard errors. Statistical tests of difference in the estimates between two time periods are based on a logistic regression model that accounts for the complex sampling design. Using a multinomial logit model, the time trend in tobacco use status is identified to capture the effects of macro level determinants including changes in tobacco control policies.Results
Between 2009 and 2012, overall tobacco use went down from 42.4% to 36.3%. The decline is more pronounced with respect to smokeless tobacco use than smoking. The prevalence of exclusive cigarette smoking went up from 7.2% to 10.6%; exclusive bidi smoking remained stable at around 2%; while smoking both cigarette and bidi went down from 4.6% to 1.8%; exclusive smokeless tobacco use went down from 20.2% to 16.9%; and both smokeless tobacco use and smoking went down from 8.4% to 5.1%. In general, the prevalence of tobacco use is higher among men, increases from younger to older age groups, and is higher among poorer people. Smoking prevalence is the highest among the slum population, followed by the tribal population, the national population and the border area population, suggesting greater burden of tobacco use among the disadvantaged groups.Conclusions
The overall decline in tobacco use can be viewed as a structural shift in the tobacco market in Bangladesh from low value products such as bidi and smokeless tobacco to high value cigarettes, which is expected with the growth in income and purchasing power of the general population. Despite the reduction in overall tobacco use, the male smoking prevalence in Bangladesh is still high at 37%. The world average of daily smoking among men is 31.1%. The Tobacco Control Act 2005 and the Amendment have yet to make a significant impact in curbing tobacco usage in Bangladesh. The findings in this paper further suggest that the tobacco control policies in Bangladesh need to include targeted interventions to restrain the use of particular types of tobacco products among specific demographic and socio-economic groups of the population, such as smoked tobacco among men, smokeless tobacco among women, and both smoked and smokeless tobacco among those living in rural areas, those in low socio-economic status and those belonging to the tribal and the slum population. 相似文献14.

Polyribosomes have been isolated from tobacco leaves. Upon infection with TMV, a new polyribosome has appeared which is specific for infection, and TMV-antigenic protein is formed on this polyribosome. This new polyribosome has an S-value of 360–380. From these results, it is suggested that this TMV specific polyribosome is an aggregate of 60–80 ribosomes which is bound by TMV-RNA as messenger, and that TMV-RNA is translated polycistronically. 相似文献
