The rate of destruction of hemagglutinins and infectivity of Newcastle disease virus was determined over a temperature range of 37.8 to 60 C. From the calculated values of deltaH and deltaS, it was concluded that inactivation of the hemagglutinating activity and viral infectivity was due to protein denaturation.  相似文献

The action of DNase I on transforming DNA, both adsorbed to marine sediment and in solution, was investigated. DNase I reduced the transformation frequencies of free DNA more than of adsorbed DNA. Changes in salt concentration or pH did not have a significant influence on the DNA-sediment complex. Soil components other than organic materials and clay minerals can bind DNA and retard its enzymatic degradation.  相似文献

The effect of Mg2+ on the thermal inactivation and unfolding of calf intestinal alkaline phosphatase has been studied at different temperatures and Mg2+ concentrations. Increasing the Mg2+ concentration in the denatured system significantly enhanced the inactivation and unfolding of the enzyme during thermal inactivation. The analysis of the kinetic course of substrate reaction during thermal inactivation showed that at 47°C the increased free Mg2+ concentration caused the inactivation rate to increase. Increasing the temperature strengthened the effect of Mg2+ on the thermal inactivation. Control experiment showed that this is not due to salt effect. The time course of fluorescence emission spectra showed that the emission maximum for Mg2+-containing system was always higher than that of Mg2+-free system, and the higher temperature enhanced this difference. In addition, Mg2+also enhanced the unfolding rate of the enzyme at 47°C. The potential biological significance of these results are discussed.  相似文献

The thermal stability of Newcastle disease virus has been characterized in terms of the rate constants for inactivation of hemagglutinating activity (HA), neuraminidase activity (NA), and infectivity. Inactivation of HA results in the concomitant loss of NA. Infectivity, however, is much more thermolabile. Disintegration of the virus particle is not responsible for the identical rate constants for inactivation of HA and NA, nor is their parallel inactivation uncoupled in envelope fragments produced by pretreating the virus with phospholipase-C. The data indicate that a common envelope factor(s) can influence the thermal stability of both activities.  相似文献

Active immunization is effective in the prophylaxis of Pseudomonas septicemia in burned mice. Vaccines were prepared from bacterial cells and growth medium of Verder's 10 different O serological types of Pseudomonas aeruginosa strains, as well as from Escherichia coli and Proteus mirabilis. Mice given a tail burn could be significantly protected against a local Pseudomonas challenge by both specific and, to a lesser extent, by nonspecific Pseudomonas vaccines prepared either from bacterial cells or from the medium in which they were grown. The vaccine was effective when administered prior to or after thermal trauma. After a more extensive rump burn, the protective effect of a specific vaccine given after thermal injury was significant only when the challenge was postponed until 4 days postburn; the level of protection was less than in the mice with smaller burns.  相似文献

The half-life of trehalase and invertase at 65 and 60 C was found to be much greater when intact ascospores of Neurospora tetrasperma were heated, as compared with extracts. By contrast, no protection was afforded these enzymes when they were heated in intact conidia and mycelium of N. crassa or N. tetrasperma. The protective effect of ascospores for trehalase was further investigated by heating ascospore extracts before and after dialysis. The removal of small molecules by dialysis lowered the heat resistance of trehalase significantly in such extracts. When the dialysate from extracts of mycelium, conidia, or ascospores was added to dialyzed enzyme extracts, that from ascospores was by far the most active. However, the same dialysates had only a small protective effect on invertase. The addition of ashed dialysates did not protect trehalase, and trehalose and glucose protected less effectively than the dialysate.  相似文献

Selective binding of [3H]bremazocine and [3H]-ethylketocyclazocine to kappa-opioid receptor sites in frog (Rana esculenta) brain membranes is irreversibly inactivated by the sulfhydryl group alkylating agent N-ethylmaleimide (NEM). Pretreatment of the membranes with kappa-selective compounds [ethylketocyclazocine (EKC), dynorphin (1-13), or U-50,488H] but not with [D-Ala2,N-Me-Phe4,Gly5-ol]enkephalin (DAGO; mu specific ligand) or [D-Ala2,N-Me-Phe4,Gly5-ol]enkephalin (DADLE; delta specific ligand) strongly protects the binding of the radioligands against NEM inactivation. These results provide more evidence for the existence of kappa-opioid receptors in frog brain. The relatively high concentrations of NEM that are needed to decrease the specific binding of [3H]bremazocine together with the observation of an almost complete protection of its binding sites by NaCl suggest that bremazocine may act as an opioid antagonist in frog brain.  相似文献

Ammonia has been shown to be virucidal in sludge and NH4Cl solutions, although the rates at which viruses are inactivated have not been thoroughly studied. In the present studies, the kinetics of the poliovirus type 1 (strain CHAT) and bacteriophage f2 inactivation were examined in such a way that the effects of OH and NH4+ could be separated from those of NH3. Purified virus stocks were placed into solutions of NH4Cl and control solutions containing an equivalent concentration of NaCl and incubated at 20°C. The percentage of virus surviving was calculated, and the kinetics were evaluated by constructing semilogarithmic plots of data. At all pH values and NH3 concentrations studied, the kinetics of the inactivation of both viruses were pseudo-first order. OH had no measurable effect on the viruses, whereas the effects of NH4+ and Na+ were similar. A dose-response relationship between NH3 and the viruses was also found. Bacteriophage f2 was approximately 4.5 times more resistant to the effects of NH3 than was poliovirus.  相似文献

Bacteriol. 90:213-219. 1965.-Infection with a measles virus variant resulted not only in formation of polykaryocytes (PK) but also in formation of multicellular immunofluorescent foci (IFF) in which no cytopathic effect could be detected. The ratio of IFF to PK changed from 27 to 4 during the first passage and remained 4 after a second passage. PK were plaques. Plaque assay was linear in the presence of IFF. To investigate the mechanism of PK formation, radioautography was done on cells pulse-labeled with tritiated thymidine before virus multiplication began. The results showed that PK were formed by fusion; there were no PK whose nuclei contained no label, and the proportion of labeled nuclei (32%) and distribution of grain counts was the same in PK as in uninvolved cells, ruling out nuclear replication without concomitant cytoplasmic membrane formation as the mechanism of formation of these PK. Early in PK development, neutral red uptake was markedly increased ("red" plaques). As PK matured, hyperchromicity disappeared ("white" plaques). This sequence provided an index of rate of evolution of PK. Rate of PK maturation was more rapid at 37 than at 32 C.  相似文献

Twenty-four chemical disinfectants considered to be viricidal were tested. Ten disinfectants were not viricidal for vesicular stomatitis virus within 10 min at 20 C when an LD(50) titer of 10(8.5) virus units per 0.1 ml were to be inactivated. Quantitative inactivation experiments were done with acid, alkaline, and a substituted phenolic disinfectant to determine the kinetics of the virus inactivation. Substituted phenolic disinfectants, halogens, and cresylic and hydrochloric acids were viricidal. Basic compounds such as lye and sodium metasilicate were not viricidal.  相似文献

The infectivity of herpes simplex virus type 1 (HSV-1) was inactivated after treatment with either concanavalin A (ConA) or periodate. Phytohemagglutinin, wheat germ agglutinin, pokeweed mitogen, and neuraminidase failed to inactivate the virus. The effect of ConA could be specifically inhibited or reversed by the addition of α-methyl-d-glucoside or α-methyl-d-mannoside. Evidence was obtained that HSV-1 inactivated by ConA could adsorb to host cells. Viral aggregation was not a major mechanism in the inactivation of HSV-1 by ConA. Under the experimental conditions employed, inactivation of HSV-1 was faster by ConA than by antiserum and less temperature dependent. A ConA-resistant fraction was detected which appeared to adsorb less quickly than untreated virus, and penetration of ConA-resistant fraction was strikingly slow. The presence of aggregates in the virus preparation did not appear to account for the ConA-resistant fraction. Inactivation of viral infectivity by ConA was obtained only with enveloped viruses, since HSV-1, HSV-2, pseudorabies, and vesicular stomatitis virus were inactivated and vaccinia and echovirus type 6 were not.  相似文献

Exposure of Venezuelan equine encephalomyelitis (VEE) virus (at -70 C) to 6 × 106 r γ-radiation (60Co) resulted in loss of lethality for young adult mice and guinea pigs, and loss of capacity to produce plaques or cytopathic effects in tissue culture. The suckling mouse was more sensitive for detecting live virus in radiated suspensions than was the adult mouse or guinea pig. Live virus was demonstrable in preparations exposed to 6 × 106 r but not in suspensions exposed to 8 × 106 r and more. The rate of inactivation of VEE virus by γ-radiation was an exponential function of the dosage.  相似文献

Purified Semliki forest virus in aerosols is inactivated rapidly at 40% and above 70% relative humidity. At all humidities tested the decay of virus infectivity runs parallel with the decrease in hemagglutination activity, whereas the biological integrity of the virus ribonucleic acid is preserved. Also, free infectious ribonucleic acid is stable after spraying at all relative humidities. Evidence is presented for the hypothesis that above 20% relative humidity, virus inactivation in aersols is mainly due to surface-dependent factors, damaging the virus coat.  相似文献
