Objective: Obesity is less prevalent in Asian subjects with type 2 diabetes mellitus (T2DM) in contrast to Caucasians. Whether higher axial bone mineral density (BMD) often reported in T2DM is independent of body mass index (BMI) has not been clearly shown. BMD characterization in T2DM patients with hip fractures has also not been performed. We compared the BMD of Asian diabetic and nondiabetic patients with new hip fractures and explored how BMD was influenced by BMI.Methods: We included 255 diabetic and 148 nondiabetic patients. BMD adjusted for age; BMI; race; sex; renal function; and use of statins, proton pump inhibitors, steroids, anticonvulsants, and calcium and/or vitamin D supplements were compared between the groups. We were particularly interested in the BMD comparison between underweight diabetics and nondiabetics with hip fractures.Results: The presence of T2DM was associated with higher BMD (g/cm2) at the femoral neck (0.527 ± 0.103 vs. 0.491 ± 0.102, P<.01) and lumbar spine [LS] (0.798 ± 0.147 vs. 0.723 ± 0.156, P<.01). This association persisted after adjustment for multiple confounding variables including BMI. The age-, BMI-, and sex-adjusted LS BMD was higher in underweight (BMI <18.5 kg/m2) diabetics compared to similar weight nondiabetics (0.733 ± 0.126 vs. 0.649 ± 0.131 g/cm2, P = .014).Conclusion: T2DM is independently associated with higher axial BMD in patients with new hip fractures. Objective

Study the effect of the 3:7 ratio of Astragalus total saponins and Curcumin on the model of diabetic nephropathy rats, and explore its mechanisms.

Methods

Diabetic nephropathy rats model was established by high-fat and high-sugar feed feeding combined with streptozotocin (STZ) injection in sublingual vein. Measured fasting blood glucose of rats on the 10, 20 and 30th day, and measured urine protein content in urine of rats on 30th days. Two hours after the last administration, measured glycated serum protein (GSP), insulin antibody (IA), triglyceride (TG), total cholesterol (TC), low density lipoprotein (LDL), high density lipoprotein (HDL), malondialdehyde (MDA), insulin, superoxide dismutase (SOD), glutathione (GSH), urea nitrogen (BUN), creatinine (Cr) in the serum and calculated the renal index of rat. Take the viscera of pancreas and kidney, and HE staining, so as to observe pathological changes.

Result

Astragalus total saponins and Curcumin 3:7 compatibility each dose group can significantly reduce the diabetic nephropathy rats blood glucose of 30th days, significantly reduce the level of GSP, IA, TG, TC, LDL (P?<?0.01), and reduce MDA levels with different degrees (P?<?0.01 or P?<?0.05), and significantly increase the level of insulin (P?<?0.01), increase the level of HDL, SOD and GSH with different degrees (P?<?0.01 or P?<?0.05 or P?>?0.05); Astragalus total saponins and Curcumin 3:7 compatibility each dose group also can decrease renal index, UN, and Cr levels with different degrees and improve the pathological changes of pancreatic tissue and kidney tissue in diabetic nephropathy rats with different degrees (P?<?0.01 or P?<?0.05 or P?>?0.05).

Conclusion

The 3:7 ratio of Astragalus total saponins and Curcumin can achieve the treatment and protection effects on diabetic nephropathy rats by improve the glycometableolism, insulin resistance, lipid metableolism, oxidative stress levels, and pathological changes.  相似文献

The effects of trace elements of gallium (Ga), germanium (Ge), and strontium (Sr) on ovariectomized (OVX) osteopenic rats were studied in this paper. The urine calcium content, serum calcium, and phosphorus contents, bone mineral content, mineral dissolution, and mechanical strength of the osteopenic rats were analyzed respectively. After the rats were fed with Ga, Ge, and Sr diet for 8 weeks, respectively, the urine calcium content decreased (P?<?0.01). Plasma calcium and phosphate concentrations decreased in the order of OVX group?>?Ge group?>?Sr group?>?Ga group?>?Sham group. Mineral content increased in the order of OVX group?<?Ge group?<?Sr group?<?Ga group?<?Sham group. A dramatic decrease in calcium solubility was found both in the gallium and strontium treated animals (P?<?0.05). However, the same result did not occur in germanium treated groups. The data provide an important proof of concept that gallium and strontium might be a new potential therapy for the management of postmenopausal osteoporosis in humans.  相似文献

Increasing studies have shown protective effects of intermittent hypoxia on brain injury and heart ischemia. However, the effect of intermittent hypoxia on blood glucose metabolism, especially in diabetic conditions, is rarely observed. The aim of this study was to investigate whether intermittent hypoxia influences blood glucose metabolism in type 1 diabetic rats. Streptozotocin-induced diabetic adult rats and age-matched control rats were treated with intermittent hypoxia (at an altitude of 3 km, 4 h per day for 3 weeks) or normoxia as control. Fasting blood glucose, body weight, plasma fructosamine, plasma insulin, homeostasis model assessment of insulin resistance (HOMA-IR), pancreas β-cell mass, and hepatic and soleus glycogen were measured. Compared with diabetic rats before treatment, the level of fasting blood glucose in diabetic rats after normoxic treatment was increased (19.88?±?5.69 mmol/L vs. 14.79?±?5.84 mmol/L, p?<?0.05), while it was not different in diabetic rats after hypoxic treatment (13.14?±?5.77 mmol/L vs. 14.79?±?5.84 mmol/L, p?>?0.05). Meanwhile, fasting blood glucose in diabetic rats after hypoxic treatment was also lower than that in diabetic rats after normoxic treatment (13.14 ± 5.77 mmol/L vs. 19.88 ± 5.69 mmol/L, p<0.05). Plasma fructosamine in diabetic rats receiving intermittent hypoxia was significantly lower than that in diabetic rats receiving normoxia (1.28?±?0.11 vs. 1.39?±?0.11, p?<?0.05), while there were no significant changes in body weight, plasma insulin and β-cell mass. HOMA-IR in diabetic rats after hypoxic treatment was also lower compared with diabetic rats after normoxic treatment (3.48?±?0.48 vs. 3.86?±?0.42, p?<?0.05). Moreover, intermittent hypoxia showed effect on the increase of soleus glycogen but not hepatic glycogen. We conclude that intermittent hypoxia maintains glycemia in streptozotocin-induced diabetic rats and its regulation on muscular glycogenesis may play a role in the underlying mechanism.  相似文献

To determine whether leptin receptor (LEPR) 223A>G polymorphism has an effect on the plasma leptin levels and the macroangiopathic complications in type 2 diabetes mellitus (T2DM). The genotypes and allelic frequencies of the LEPR 223A>G were examined with polymerase chain reaction and restriction fragment length polymorphism in 301 patients with T2DM and 172 unrelated healthy subjects. The plasma concentrations of leptin were determined in all subjects. The mean plasma leptin levels in the T2DM group were significantly higher than that of controls and the plasma levels of leptin were higher in diabetic patients with macroangiopathy than in patients without macroangiopathy (P < 0.05). The genotype (GG, AG and AA) distribution of 223A>G polymorphism was 58.3, 32.5, and 9.2% in diabetic patients with macroangiopathy, 75.3, 22.1, and 2.6% in patients without macroangiopathy, and 70.3, 27.5, 2.2% in controls respectively, a significant difference was found between diabetic patients with and without macroangiopathy (P < 0.05). The frequency of the allele A was higher in patients with macroangiopathy than in patients without macroangiopathy (25.6 vs. 16.3%; P < 0.05). Moreover, the plasma leptin levels were markedly higher in patients with AA genotype than those with AG or GG genotype in patients with macroangiopathy (P < 0.05). The LEPR 223A>G gene polymorphism associated with a predisposition to increased plasma leptin levels could constitute a useful predictive marker for diabetic macroangiopathy.  相似文献

An experiment was conducted to determine the effect of dietary copper (Cu) on mineral profile, hematological parameters, and lipid metabolism in lambs. Eighteen Zandi male lambs (approximately 3 months of age; 17.53?±?1.6 kg of body weight) were housed in individual pens and were assigned randomly to one of three treatments. Treatments consisted of (1) control (no supplemental Cu), (2) 10 mg Cu/kg dry matter (DM) from copper sulfate (CuS), and (3) 10 mg Cu/kg DM from Cu proteinate (CuP). The Cu concentration was 8.2 mg/kg DM in the basal diet. Blood was sampled from the jugular vein at the beginning of the study (enrollment, before feeding Cu supplement) and at days 25, 50, and 70 of experiment. The amounts of total serum glucose, urea nitrogen, calcium, phosphorus, iron, copper, zinc, and lipids and hematological parameters were measured. Average daily gain and feed efficiency were improved (P?<?0.05) with Cu supplementation and were better for the lambs fed diet supplemented with CuP. The concentrations of serum Ca, P, and Zn were not affected by source of Cu in the diet. However, Fe concentration was lower (P?<?0.01) in the Cu-supplemented groups. Experimental treatment had no significant effects on the hematological parameters. The serum glucose concentration was not affected by treatments. However, the urea nitrogen concentrations were significantly affected (P?<?0.05) by added Cu and was lower for CuP group as compared to the lambs in the CuS and control groups. Addition of Cu had no influence (P?>?0.05) on the serum triglyceride concentration, but lambs fed with CuP supplement had lower (P?<?0.05) serum cholesterol than the CuS and control animals. These results indicated that CuP supplemented at 10 mg/kg DM improved gain and enhanced the efficiency of nitrogen in male lambs.  相似文献

Diabetes mellitus (DM) is known to impair many physiological functions. Some reports claim that medicinal plants can reduce these alterations caused by DM. The aim of this study was to investigate the therapeutic potential of aqueous-methanol extracts of Urtica dioica, Thymus vulgaris (TV), Myrtus communis (MC), Scolymus hispanicus (SH) and Cinnamomun zeylanicum (CZ) on streptozotocin (STZ)-induced type 1 DM in rats. Diabetes was induced via a single i.p. injection of STZ (65 mg/kg body weight). After 1 week to allow for development of diabetes, each plant extract was administered to diabetic rats separately at a dose of 100 mg/kg body weight daily for 28 days. The results showed that only SH extract significantly (P < 0.05) amended fasting blood glucose level. The lipid profile was ameliorated especially by supplementations of TV, MC and CZ extracts. Almost all plant extract treatments markedly (P < 0.05) increased reduced glutathione content and decreased lipid peroxidation levels of erythrocyte, plasma, retina and lens tissues. They also significantly (P < 0.05) amended erythrocyte catalase activity, levels of marker serum enzymes (except amylase), urea and blood urea nitrogen when compared to diabetic rats treated with nothing. Furthermore, none of the plant extracts counteracted body weight loss of diabetic rats. Our data revealed that the aforementioned plant extracts have remarkable potential to counteract DM-caused alterations, probably through their antioxidant and free radical-defusing effects.  相似文献

The study was conducted to determine the effects of iron glycine chelate (Fe-Gly) on growth, tissue mineral concentrations, fecal mineral excretion, and liver antioxidant enzyme activities in broilers. A total of 360 1-day-old commercial broilers (Ross?×?Ross) were randomly allotted to six dietary treatments with six replications of ten chicks per replicate. Broilers were fed a control diet with no Fe supplementation, while five other treatments consisted of 40, 80, 120, and 160?mg Fe/kg diets from Fe-Gly, and 160?mg Fe/kg from ferrous sulfate, respectively. After a 42-day feeding trial, the results showed that 120 and 160?mg Fe/kg as Fe-Gly improved the average daily gain (P?<?0.05) and average daily feed intake (P?<?0.05) of broilers (4?C6?weeks). Addition with 120 and 160?mg Fe/kg from Fe-Gly and 160?mg Fe/kg from FeSO4 increased Fe concentration in serum (P?<?0.05), liver (P?<?0.05), breast muscle (P?<?0.05), tibia (P?<?0.05), and feces (P?<?0.01) at 21 and 42?days. There were linear responses to the addition of Fe-Gly from 0 to 160?mg/kg Fe on Fe concentration in serum (21?days, P?=?0.005; 42?days, P?=?0.001), liver (P?=?0.001), breast muscle (P?=?0.001), tibia (P?=?0.001), and feces (21?days, P?=?0.011; 42?days, P?=?0.032). Liver Cu/Zn superoxide dismutase activities of chicks were increased by the addition of 80, 120, and 160?mg Fe/kg as Fe-Gly to diets at 42?days. There were no differences in liver catalase activities of chicks among the treatments (P?>?0.05). This study indicates that addition with Fe-Gly could improve growth performance and iron tissue storage and improves the antioxidant status of broiler chickens.  相似文献

The objective of this study was to determine the effects of supplemental chromium as chromium picolinate (CrPic) on productive performance, chromium (Cr) concentration, serum parameters, and colostrum composition in sows. Thirty Yorkshire sows were bred with semen from a pool of Landrace boars. The sows were equally grouped and treated with either a diet containing 0 (control) or 400 ppb dietary Cr supplementation throughout gestation. The sows received the same basal diet based on corn-DDGS meal. Supplemental CrPic increased (P?<?0.05) the sow body mass gain from the insemination to the day 110 of gestation in sows. No differences (P?>?0.50) were observed in the gestation interval, sow mass, and backfat at insemination, after farrowing, at weaning and lactation loss. The number of piglets born alive, piglets per litter at weaning, and litter weaned mass were increased (P?<?0.05) for those supplemented with CrPic compared with the control. However, the total number of piglets born, total born litter mass, average piglet birth body mass, born alive litter mass, and average born alive piglet mass did not differ among the treatments (P?>?0.05). The placental masses of sows were similar among treatments (P?>?0.05). Dietary supplementation with CrPic throughout gestation in sows showed increased (P?<?0.01) concentration of Cr in the colostrum or serum at days 70 and 110. Compared with the control group, dietary supplementation with CrPic throughout gestation in sows decreased (P?<?0.05) the serum insulin concentration, the glucose or serum urea nitrogen concentration at days 70 and 110. However, no differences (P?>?0.05) were observed in total protein concentration among treatments. No differences (P?>?0.05) were observed in total solids, protein, fat or lactose among sows fed the diets supplemented with CrPic compared with the control. Two experiments were conducted to study the effects of dietary manganese levels on growth performance, nutrients digestibility, and N balance of minks during growing period. In experiment 1, 75 healthy male minks (60 days old) were selected and randomly divided into five groups with different types of diet. The diet was supplemented with 0 (control), 50, 100, 300, and 600 ppm of manganese as MnSO4 of dry matter (DM) in basic diet, respectively. From early July to middle September, the results showed that the final body weights of minks were significantly affected by diets (P?<?0.05). Average daily gains (ADG) were significantly higher in the 300-ppm manganese group than those in other groups. The ratio of feed to body weight gain (F/G) was significantly affected by manganese level (P?<?0.05). In experiment 2, 45 male minks (75 days old) with the same body weight were selected from each group of experiment 1 to carry out the nutrient digestion and N-balance tests which lasted for 4 days for the collection of the feces and urine, and the diets and treatment codes were same as in experiment 1. The results showed that no significant differences were found in DM, crude protein (CP), and crude carbohydrate (CC) digestibility among all groups (P?>?0.05), but ether extract (EE) and gross energy (GE) digestibility were all the highest in the 300-ppm group. N intake and fecal N were similar among all groups (P?>?0.05). Urinary N was lower in the 300-ppm group; in contrast, N retention was higher in this group (P?<?0.05). In conclusion of experiment 1 and experiment 2, the diet supplemented with 300 ppm of manganese (as manganese sulfate) could improve the growth performance and increase the EE and GE digestibility of mink during the growing period and moreover reduce the nitrogen emissions to the environment, and the optimal total manganese level in mink’s diet was 409.16 in DM during the growing period.  相似文献

Polymorphisms in the gene encoding for A disintegrin and metalloprotease 33 (ADAM33) are closely associated with the risk of bronchial asthma attacks in different populations. We collected blood samples from 248 asthma patients – 130 of the Han ethnic group and 118 of the Mongolian ethnic group – living in the Inner Mongolia region of China, and analyzed the single nuclear polymorphisms (SNPs) of the T1, T2 and V4 loci of the ADAM33 gene using PCR-RFLP (restriction fragment length polymorphism). In addition, we also tested 256 healthy controls (134 and 122 from the Han and Mongolian ethnic groups respectively) for the same SNPs. Three genotypes of the T1, T2 and V4 loci were predominantly detected: while polymorphisms in the T1 locus were significantly associated with asthma risk in both Mongolian and Han ethnicities (P?<?0.05, 1P?<?0.05), that in the V4 locus were relevant only in the Mongolian patients (P?<?0.05, 1P?>?0.05). Litterfall production, decomposition and nutrient use efficiency in three different tropical forest ecosystems in SW China were studied for 10 years. Annual mean litterfall production in tropical seasonal forest (TSF) (9.47?±?1.65 Mg ha?1) was similar to that in man-made tropical forest (MTF) (9.23?±?1.29 Mg ha?1) (P?>?0.05) but both were significantly lower than that in secondary tropical forest (STF) (12.96?±?1.71 Mg ha?1) (P?<?0.05). The annual variation of litterfall was greater in TSF (17.4%, P?<?0.05) than in MTF (14.0%) or STF (13.2%). The annual mean decomposition rate of litterfall increased followed the order of MTF (2.72)?<?TSF (3.15)?<?STF (3.50) (P?<?0.05), which was not correlated with annual precipitation or annual mean temperature, but was rather related to litter quality. The nutrient use efficiency was found to be element-dependent and to vary significantly among the three forest types (P?<?0.05). These results indicate that litterfall production and decomposition rates in different tropical forest systems are related to plant species composition and are influenced strongly by coexisting species and their life stage (age) but less so by the species richness. Constructing multi-species and multistory man-made tropical forest is an effective way to enhance biological productivity and maintain soil nutrients on degraded tropical land.  相似文献

Phytic acid is a major determinant of zinc bioavailability. Little is known about phytic acid intakes or indices of zinc bioavailability in type 2 diabetes mellitus (DM), a condition that predisposes to zinc deficiency. The aim of this cross-sectional study was to measure and explore the relationships among phytic acid intake, zinc bioavailability, and molecular markers of zinc homeostasis in 20 women with DM compared to 20 healthy women. The phytate/zinc, (calcium)(phytate)/zinc, and (calcium + magnesium)(phytate)/zinc molar ratios were used to indicate zinc bioavailability. Plasma zinc concentrations and zinc transporter (ZnT1, ZnT8, and Zip1) gene expression in mononuclear cells were measured. Participants with DM consumed 1,194?±?824?mg/day (mean?±?SD) phytic acid, an amount similar to the intake of healthy women (1,316?±?708?mg/day). Bread products and breakfast cereals contributed more than 40?% of the phytic acid intake in each group. A positive relationship was observed in all participants between phytic acid and dietary fiber (r?=?0.6, P?<?0.001) and between dietary fiber and the (calcium)(phytate)/zinc ratio (r?=?0.5, P?<?0.001). Compared to the healthy group, the messenger RNA ratio of ZnT1 (zinc export) to Zip1 (zinc import) was lower in participants with DM, which may indicate perturbed zinc homeostasis in the disorder. The plasma zinc concentration was not predicted by age, body mass index, health status, zinc bioavailability, or zinc transporter expression. Healthy and diabetic women consume phytic acid in amounts that are likely to decrease the bioavailability of dietary zinc. Recommendations to consume greater amounts of dietary fiber, much of which is associated with phytate, increase the risk of zinc deficiency.  相似文献

The main objective of the current article is to investigate the diabetic polyneuropathy which represents a major preoccupation within the context of high incidence of diabetes mellitus (DM) and its complications. Moreover, neuropathy may develop despite intensive hyperglycaemic control. The effect of Zn and black grape seed polyphenols (BGSP) in streptozotocin diabetic rats was studied. Zn and BGSP were administered by gavage, daily, for 16 weeks to Wistar rats that have been rendered diabetic by a single i.v. injection of streptozotocin (55 mg/kg body weight). Dysalgesia was investigated under the conditions of nociceptive stimulation through the following tests: the thermoalgesic mechanism through the tail-flick test, the hot plate test and the plantar test, and the mechanoalgesic mechanism through the algesimetric test. Thermal hyperalgesia detected in the diabetic group is significantly reduced (p?<?0.001) through the administration of polyphenols, or even better, of Zn. Diabetes-associated mechanical hyperalgesia decreased significantly (p?<?0.001) probably through the inhibition of the NMDA receptors. Administration of Zn or BGSP to the diabetic group improves glycosylated haemoglobin (HbA1c) values but does not bring them to normal. The present data suggest a favourable effect of Zn and BGSP in inhibiting diabetic complications by several mechanisms.  相似文献

Background

Evidence has shown that endogenous H2S plays an important role in the physiological and pathophysiological processes of many organs. The study aimed to explore whether exogenous H2S has a potential therapeutic effect on a rat ovariectomy-induced model of osteoporosis.

Methods

The OVX osteoporosis model was established in female Sprague-Dawley rats by full bilateral ovariectomy. The rats were randomly divided into four groups, with the two experimental groups receiving an intraperitoneal injection of GYY4137 or sodium alendronate. The level of H2S in the plasma was determined and common laboratory indicators to diagnose osteoporosis, such as alkaline phosphatase (ALP) activity and the levels of osteocalcin (OCN), calcitonin, parathyroid hormone and leptin were measured. The bone mineral density (BMD) of the 4th and 5th lumbar vertebrae was measured using dual-energy X-ray absorptiometry. The maximum stress of femoral fracture was obtained through a three-point bending test of the femur.

Results

The OVX osteoporosis model was successfully established. GYY4137 was injected to increase the level of H2S in the plasma in one group, designated OVX-GYY during the observation period (p?<?0.05). At 12 weeks, the BMD value of the fourth lumbar vertebra in the OVX-GYY group had increased (p?<?0.05). The BMD femur value in the OVX-vehicle group had decreased (p?<?0.05). Bilateral ovariectomy leads to biochemical disorders related to bone metabolism and hormone levels in rat plasma (all p?<?0.05). Ovariectomy also reduced blood calcium, blood phosphate and calcitonin, and increased parathyroid hormone and leptin. The opposite results were obtained for the groups with alendronate sodium or GYY4137 treatment (all p?<?0.05).

Conclusions

Through the slow release of H2S, GYY4137 did an excellent job of simulating endogenous neuroendocrine gaseous signaling molecules. Exogenous H2S had a regulatory effect on osteoporosis in ovariectomized rats, showing potential value for the treatment of human postmenopausal osteoporosis.

The aim of this study was to evaluate the anti-apoptosis effects of resveratrol (RSV) on diabetic rats retinal Müller cells in vivo and in vitro and to further investigate the roles of microRNA-29b (miR-29b)/specificity protein 1 (SP1) in the anti-apoptosis mechanism of RSV. Retina was obtained from normal and diabetic rats with or without RSV (5 and 10 mg/kg/day) treatments at 1–7 months. TdT-mediated dUTP-biotin nick end labeling (TUNEL) and Annexin V/PI staining were used to detect apoptosis. Immunofluorescence was used to assess distribution of SP1 in retina. MiR-29b and SP1 messenger RNA (mRNA) expression was evaluated by quantitative real-time polymerase chain reaction (qRT-PCR). SP1, Bax, and bcl-2 protein expression was evaluated by western blotting. Caspase-3 activity was detected by assay kit. Our study showed that the TUNEL-positive cells were mainly localized in the inner nuclear layer (INL) of retina and RSV administration effectively suppressed streptozotocin (STZ)-induced apoptosis of retinal cells in INL in vivo (P?<?0.001). Our study also showed that RSV administration effectively suppressed high glucose (HG)-induced retinal Müller cells’ apoptosis in vitro (P?<?0.001). Alpha-lipoic acid (ALA) is widely used as an antioxidant for the treatment of diabetes and its complications; however, the pro-oxidant potential of ALA has recently been reported. This study was designed to investigate whether ALA supplementation could have pro-oxidant effects on cardiac tissues in normal and diabetic rats. Diabetes was induced by a single dose of streptozotocin (STZ; 55 mg/kg (intraperitoneal). Diabetic and normal rats were treated with ALA (100 mg kg?1 day?1) for 45 days. ALA supplementation resulted in oxidative protein damage as evident by significant reduction in the cardiac levels of protein thiol in ALA-treated normal rats (P?<?0.01) together with a significant elevation (P?<?0.001) in the plasma levels of advanced oxidation protein products in ALA-treated normal rats and in ALA?+?STZ-diabetic rats compared with the normal control rats. Nicotinamide adenine dinucleotide phosphate (NADPH) oxidase has emerged as the major source of superoxide anion and enhanced oxidative damage in heart failure. ALA supplementation increased the myocardial immunoreactivity of p47phox subunit of NADPH oxidase in both normal nondiabetic and diabetic rats reflecting its pro-oxidant effect. Data showed that ALA supplementation failed to prevent cardiac complications in diabetic rats and led to cardiac toxicity in normal rats as indicated by pathological changes (cellular infiltration, fibrosis, and degeneration) and by the elevation of serum cardiac biomarkers compared with normal controls. The pro-oxidant effects of ALA suggest that careful selection of appropriate doses of ALA in reactive oxygen species-related diseases are critical.  相似文献

A study was conducted using 360 broiler chickens to evaluate the effects of dietary vitamin E (0, 125 and 250 mg/kg), selenium (Se, 0, 0.5 and 1 mg/kg), or their different combinations on immune response and blood biological parameters of broilers raised under either thermoneutral (TN, 23.9 °C constant) or heat stress (HS, 23.9 to 37 °C cycling) conditions. Humoral immunity was assessed by intravenous injection of 7 % sheep red blood cell (SRBC) followed by evaluation of serum for antibody titers in primary and secondary responses. Heterophil to lymphocyte (H/L) ratio also determined as an indicator of stress. Furthermore, at the end of the experiment, birds were bled for determination of some biological parameters. There was a significant reduction in body weight and feed intake, but the feed conversion ratio increased when the birds were exposed to HS (P?<?0.05). Body weight and feed intake were not influenced significantly by dietary vitamin E and Se (P?>?0.05), whereas feed conversion was improved significantly by 125 mg/kg vitamin E (P?<?0.05). The liver and lymphoid organ weights as well as IgM and IgG, antibody titers for primary and secondary antibody responses to SRBC were reduced significantly under HS (P?<?0.05). Heat stress also resulted in a significant increase in H/L ratio (P?<?0.05). Dietary vitamin E resulted in improvement of primary and secondary antibody responses both in TN and HS broilers (P?<?0.05). The HS birds also showed an improved antibody titer in secondary response with high concentration of Se (P?<?0.05). Vitamin E and Se had interactive effects on anti-SRBC titers; however, no consistent differences were found between dietary levels during the study. The presence of heparin and a mixture of penicillamine, hypotaurine, and epinephrine (PHE) solution in the in vitro fertilization (IVF) media seem to be a prerequisite when bovine spermatozoa are capacitated in vitro, in order to stimulate sperm motility and acrosome reaction. The present study was designed to determine the effect of the addition of heparin and PHE during IVF on the quality and penetrability of spermatozoa into bovine oocytes and on subsequent embryo development. Sperm quality, evaluated by the integrity of plasma and acrosomal membranes and mitochondrial function, was diminished (P?<?0.05) in the presence of heparin and PHE. Oocyte penetration and normal pronuclear formation rates, as well as the percentage of zygotes presenting more than two pronuclei, was higher (P?<?0.05) in the presence of heparin and PHE. No differences were observed in cleavage rates between treatment and control (P?>?0.05). However, the developmental rate to the blastocyst stage was increased in the presence of heparin and PHE (P?>?0.05). The quality of embryos that reached the blastocyst stage was evaluated by counting the inner cell mass (ICM) and trophectoderm (TE) cell numbers and total number of cells; the percentage of ICM and TE cells was unaffected (P?>?0.05) in the presence of heparin and PHE (P?<?0.05).
