The binding of one fluorine including triazole (C10H9FN4S, FTZ) to bovine serum albumin (BSA) was studied by spectroscopic techniques including fluorescence spectroscopy, UV–Vis absorption, and circular dichroism (CD) spectroscopy under simulative physiological conditions. Fluorescence data revealed that the fluorescence quenching of BSA by FTZ was the result of forming a complex of BSA–FTZ, and the binding constants (K a) at three different temperatures (298, 304, and 310 K) were 1.516?×?104, 1.627?×?104, and 1.711?×?104?mol L?1, respectively, according to the modified Stern–Volmer equation. The thermodynamic parameters ΔH and ΔS were estimated to be 7.752 kJ mol?1 and 125.217 J?mol?1?K?1, respectively, indicating that hydrophobic interaction played a major role in stabilizing the BSA–FTZ complex. It was observed that site I was the main binding site for FTZ to BSA from the competitive experiments. The distance r between donor (BSA) and acceptor (FTZ) was calculated to be 7.42 nm based on the Förster theory of non-radioactive energy transfer. Furthermore, the analysis of fluorescence data and CD data revealed that the conformation of BSA changed upon the interaction with FTZ.  相似文献

Study on the binding properties of helicid by pepsin systematically using multi-spectroscopic techniques and molecular docking method, and these interactions comprise biological recognition at molecular level and backbone of biological significance in medicine concerned with the uses, effects, and modes of action of drugs. We investigated the mechanism of interaction between helicid and pepsin by using various spectroscopic techniques viz., fluorescence spectra, UV–Vis absorption spectra, circular dichroism (CD), 3D spectra, synchronous fluorescence spectra and molecular docking methods. The quenching mechanism associated with the helicid–pepsin interaction was determined by performing fluorescence measurements at different temperatures. From the experimental results show that helicid quenched the fluorescence intensity of pepsin via a combination of static and dynamic quenching process. The binding constants (Ka) at three temperatures (288, 298, and 308 K) were 7.940?×?107, 2.082?×?105 and 3.199?×?105 L mol?1, respectively, and the number of binding sites (n) were 1.44, 1.14, and 1.18, respectively. The n value is close to unity, which means that there is only one independent class of binding site on pepsin for helicid. Thermodynamic parameters at 298 K were calculated as follows: ΔHo (??83.85 kJ mol?1), ΔGo (??33.279 kJ mol?1), and ΔSo (??169.72 J K?1 mol?1). Based on thermodynamic analysis, the interaction of helicid with pepsin is driven by enthalpy, and Van der Waals’ forces and hydrogen bonds are the main forces between helicid and pepsin. A molecular docking study further confirmed the binding mode obtained by the experimental studies. The conformational changes in the structure of pepsin was confirmed by 3D fluorescence spectra and circular dichroism.  相似文献

The interaction between thiamine hydrochloride (TA) and bovine serum albumin (BSA) was investigated by fluorescence, FTIR, UV–vis spectroscopic and cyclic voltammetric techniques under optimised physiological condition. The fluorescence intensity of BSA is gradually decreased upon addition of TA due to the formation of a BSA–TA complex. The binding parameters were evaluated and their behaviour at different temperatures was analysed. The quenching constants (Ksv) obtained were 2.6 × 104, 2.2 × 104 and 2.0 × 104 L mol?1 at 288, 298 and 308 K, respectively. The binding mechanism was static-type quenching. The values of ΔH° and ΔS° were found to be 26.87 kJ mol?1 and 21.3 J K?1 mol?1, and indicated that electrostatic interaction was the principal intermolecular force. The changes in the secondary structure of BSA upon interaction with TA were confirmed by synchronous and 3-D spectral results. Site probe studies reveal that TA is located in site I of BSA. The effects of some common metal ions on binding of BSA–TA complex were also investigated.  相似文献

Abstract

The interaction between Tb(IV)-NR complex and herring sperm DNA in buffer solution of Tris-HCl was investigated with the use of acridine orange(AO) as a spectral probe. The binding modes and other information were provided by the UV–spectrophotometry and fluorescence spectroscopy. The thermodynamic functions expressed that the binding constants of Tb(IV)-NR complex with DNA was Kθ298.15K = 4.03?×?105?L·mol?1, Kθ310.15K =1.30?×?107?L·mol?1, and the ΔrGθ m 298.15?K=?3.20?×?104 J·mol?1. The scatchard equation suggested that the interaction mode between Tb(IV)-NR complex and herring sperm DNA is electrostatic and weak intercalation bindings. FTIR spectroscopy results also indicate that there is a specific interaction between the Tb(IV)-NR complex and the A and G bases of DNA.  相似文献

Cystatins, known for their ubiquitous presence in mammalian system are thiol protease inhibitors serving important physiological functions. Here, we present a variant of cystatin isolated from brain of Capra hircus (goat) which is glycosylated but lacks disulphide bonds. Caprine brain cystatin (CBC) was isolated using alkaline treatment, ammonium sulphate fractionation (40–60%) and gel filtration chromatography on Sephacryl S-100HR column with an overall yield of 26.29% and 322-fold purification. The inhibitor gave a molecular mass of ~44 kDa as determined by SDS-PAGE and gel filtration behaviour. The Stokes radius and diffusion coefficient of CBC were 27.14 Å and 8.18 × 10?7 cm2 s?1, respectively. Kinetic data revealed that CBC inhibited thiol proteases reversibly and competitively, with the highest inhibition towards papain (Ki = 4.10 nM) followed by ficin and bromelain. CBC possessed 34.7% α-helical content as observed by CD spectroscopy. UV, fluorescence, CD and FTIR spectroscopy revealed significant conformational change upon CBC-papain complex formation. Isothermal titration calorimetry (ITC) was used to measure the thermodynamic parameters – ΔH, ΔS, ΔG along with N (binding stoichiometry) for CBC-papain complex formation. Binding stoichiometry (N = .97 ± .07 sites) for the CBC-papain complex indicates that cystatin is surrounded by nearly one papain molecule. Negative ΔH (?5.78 kcal mol?1) and positive ΔS (11.01 cal mol?1 deg?1) values suggest that the interaction between CBC and papain is enthalpically as well as entropically favoured process. The overall negative ΔG (?9.19 kcal mol?1) value implies a spontaneous CBC-papain interaction.  相似文献

Abstract

This research is focussed on kinetic, thermodynamic and thermal inactivation of a novel thermostable recombinant α-amylase (Tp-AmyS) from Thermotoga petrophila. The amylase gene was cloned in pHIS-parallel1 expression vector and overexpressed in Escherichia coli. The steady-state kinetic parameters (Vmax, Km, kcat and kcat/Km) for the hydrolysis of amylose (1.39?mg/min, 0.57?mg, 148.6?s?1, 260.7), amylopectin (2.3?mg/min, 1.09?mg, 247.1?s?1, 226.7), soluble starch (2.67?mg/min, 2.98?mg, 284.2?s?1, 95.4) and raw starch (2.1?mg/min, 3.6?mg, 224.7?s?1, 61.9) were determined. The activation energy (Ea), free energy (ΔG), enthalpy (ΔH) and entropy of activation (ΔS) at 98?°C were 42.9?kJ mol?1, 74?kJ mol?1, 39.9?kJ mol?1 and ?92.3 J mol?1 K?1, respectively, for soluble starch hydrolysis. While ΔG of substrate binding (ΔGE-S) and ΔG of transition state binding (ΔGE-T) were 3.38 and ?14.1?kJ mol?1, respectively. Whereas, EaD, Gibbs free energy (ΔG*), increase in the enthalpy (ΔH*) and activation entropy (ΔS*) for activation of the unfolding of transition state were 108, 107, 105?kJ mol?1 and ?4.1 J mol?1 K?1. The thermodynamics of irreversible thermal inactivation of Tp-AmyS revealed that at high temperature the process involves the aggregation of the protein.  相似文献

The purpose of this study was to elucidate the binding of paeonol to human serum albumin (HSA) through spectroscopic methods. The fluorescence quenching of HSA by paeonol was a result of the formation of the HSA–paeonol complex with low binding affinity (K = 4.45 × 103 M?1 at 298 K). Thermodynamic parameters (ΔG = –2.08 × 104 J·mol?1, ΔS = 77.9 J·mol?1·K?1, ΔH = 2.41 × 103 J·mol?1, kq = 9.67 × 1012 M?1·s?1) revealed that paeonol mainly binds HSA through hydrophobic force following a static quenching mode. The binding distance was estimated to be 1.91 nm by fluorescence resonant energy transfer. The conformation of HSA was changed and aggregates were formed in the presence of paeonol, revealed by synchronous fluorescence, circular dichroism, Fourier transform infrared spectroscopy, three‐dimensional fluorescence spectroscopy, and resonance light scattering results.  相似文献

Kinetic studies of the reduction of ferrioxamine B (Fe(Hdesf)+) by Cr(H2O)62+, V(H2O)62+, and dithionite have been performed. For Cr(H2O)62+ and V(H2O)62+, the rate is ?d[Fe(Hdesf)+]/dt = k[Fe(Hdesf)+][M2+]. For Cr(H2O)62+, k = 1.19 × 104 M?1 sec?1 at 25°C and μ = 0.4 M, and k is independent of pH from 2.6 to 3.5. For V(H2O)62+, k = 6.30 × 102 M?1 sec?1 at 25°C, μ = 1.0 M, and pH = 2.2. The rate is nearly independent of pH from 2.2 to 4.0. For Cr(H2O)62+ and V(H2O)62+, the activation parameters are ΔH = 8.2 kcal mol?1, ΔS ?12 eu and ΔH = 1.7 kcal mol?1, ΔS = ?40 eu (at pH 2.2) respectively. Reduction by Cr(H2O)62+ is inner-sphere, while reduction by V(H2O)62+ is outer-sphere. Reduction by dithionite follows the rate law ?d[Fe(Hdesf)+]/dt =kK12[Fe(Hdesf)+][S2O42?]12 where K is the equilibrium constant for dissociation of S2O42? into SO2? radicals. The value of k at 25°C and μ = 0.5 is 2.7 × 103 M?1 sec?1 at pH 5.8, 3.5 × 103 M?1 sec?1 at pH 6.8, and 4.6 × 103 M?1 sec?1 at pH 7.8, and ΔH = 6.8 kcal mol?1 and ΔS = ?19 eu at pH 7.8.  相似文献

The interactions of L-aminoglucosidic stereoisomers such as rhodostreptomycins A (Rho A) and B (Rho B) with cations (Mg2+, Ca2+, and H+) were studied by a quantum mechanical method that utilized DFT with B3LYP/6-311G**. Docking studies were also carried out in order to explore the surface recognition properties of L-aminoglucoside with respect to Mg2+ and Ca2+ ions under solvated and nonsolvated conditions. Although both of the stereoisomers possess similar physicochemical/antibiotic properties against Helicobacter pylori, the thermochemical values for these complexes showed that its high affinity for Mg2+ cations caused the hydration of Rho B. According to the results of the calculations, for Rho A–Ca2+(H2O)6, ΔH = ?72.21 kcal?mol?1; for Rho B–Ca2+(H2O)6, ΔH = ?72.53 kcal?mol?1; for Rho A–Mg2+(H2O)6, ΔH = ?72.99  kcal?mol?1 and for Rho B–Mg2+(H2O)6, ΔH = ?95.00  kcal?mol?1, confirming that Rho B binds most strongly with hydrated Mg2+, considering the energy associated with this binding process. This result suggests that Rho B forms a more stable complex than its isomer does with magnesium ion. Docking results show that both of these rhodostreptomycin molecules bind to solvated Ca2+ or Mg2+ through hydrogen bonding. Finally, Rho B is more stable than Rho A when protonation occurs.

Thermodynamic parameters for the reduction of ferrioxamine E as calculated from redox potentials determined at four different temperatures were found to be ΔH=7.1±3.4 kJ mol?1 and ΔS=?146 J mol?1 K?1. The negative entropy value is large, because the decrease in the charge at the metal center and an increase in its ionic radius force the structure of the complex to become less rigid and resemble the desferrisiderophore. The hydrophilic groups of the system are now (relatively more) available for solvent interaction. Thus, a large negative entropy change accompanies the reduction of the complex. Kinetics of reduction of ferrioxamine by VII, CrII, EuII, and dithionite were measured at different temperatures and by dithionite at different pH values. The CrII and EuII reactions proceed by an inner‐sphere mechanism and have second‐order rate constants at 25° of 1.37×104 and 1.23×105 M ?1 s?1, respectively. For the VII reduction, the corresponding rate constant was 1.89×103 M ?1 s?1. The activation parameters for the VII reduction were ΔH = 8.3 kJ mol?1; ΔS = ?154 J mol?1 K?1. These values are indicative of an outer‐sphere mechanism for VII reduction. The reduction by dithionite is half order in dithionite concentration indicating that SO . is the sole reducing species. log of reduction rate constants of different trihydroxamates by this reductant were correlated with their respective redox potentials, and the variation was found to be in approximate correspondence with the expectations of Marcus relationship.  相似文献

Ultraviolet-visible (UV-vis) spectra, fluorescence spectra, electrochemistry, and the thermodynamic method were used to discuss the interaction mode between the inclusion complex of hematoxylin with β-cyclodextrin and herring sperm DNA. On the condition of physiological pH, the result showed that hematoxylin and β-cyclodextrin formed an inclusion complex with binding ratio nhematoxylin:nβ-cyclodextrin = 1:1. The interaction mode between β-cyclodextrin-hematoxylin and DNA was a mixed binding, which contained intercalation and electrostatic mode. The binding ratio between β-cyclodextrin-hematoxylin and DNA was nβ-cyclodextrin -hematoxylin:nDNA = 2:1, binding constant was K? 298.15K = 5.29 × 104 L·mol?1, and entropy worked as driven force in this action.  相似文献

Protein‐nucleic acids binding driven by electrostatic interactions typically are characterized by the release of counter ions, and the salt‐inhibited binding association constant (Ka) and the magnitude of exothermic binding enthalpy (ΔH). Here, we report a non‐classical thermodynamics of streptavidin (SA)–aptamer binding in NaCl (140–350 mM) solutions near room temperatures (23–27 °C). By using isothermal titration calorimetry (ITC) and circular dichroism (CD)/fluorescence spectroscopy, we found that the binding was enthalpy driven with a large entropy cost (ΔH ?20.58 kcal mol?1, TΔS ?10.99 kcal mol?1, and Ka 1.08 × 107 M?1 at 140 mM NaCl 25 °C). With the raise of salt concentrations, the ΔH became more exothermic, yet the Ka was almost unchanged (ΔH ?26.29 kcal mol?1 and Ka 1.50 × 107 M?1 at 350 mM NaCl 25 °C). The data suggest that no counter Na+ was released in the binding. Spectroscopy data suggest that the binding, with a stoichiometry of 2, was accompanied with substantial conformational changes on SA, and the changes were insensitive to the variation of salt concentrations. To account for the non‐classical results, we propose a salt bridge exchange model. The intramolecular binding‐site salt bridge(s) of the free SA and the charged phosphate group of aptamers re‐organize to form the binding complex by forming a new intermolecular salt bridge(s). The salt bridge exchange binding process requires minimum amount of counter ions releasing but dehydration of the contacting surface of SA and the aptamer. The energy required for dehydration is reduced in the case of binding solution with higher salt concentration and account for the higher binding exothermic mainly. The reaction of Ru(XTPP)(DMF)2, where XTPP is the dianion of para substituted tetraphenylporphyrins and X is MeO, Me, H, Cl, Br, I, F, with O2 and CO were studied in DMF. The process was found to be first-order in metalloporphyrin, first-order in molecular oxygen and carbon monoxide, and second-order overall. Second-order rate constants for the CO reaction ranged from 0.170 to 0.665 M?1 s?1 at 25°C, those for the O2 reaction from 0.132 to 0.840 M?1 s?1 at 25°C. Similar activation parameters (ΔHCO± = 87 ± 1 kJ mol?1, ΔSCO± = 22 ± 4 JK?1 mol?1; ΔHO2± = 81 ± 1 kJ mol?1, and ΔSO2± = 11 ± 5 JK?1 mol?1) were found within each series. Reactivities of X substituted metalloporphyrins were found to follow different Hammett σ functions. The CO reactions correlated with σ? following normal behavior; the O2 reactions correlated with σ8° indicating O2 is π-bonded in the transition states. Ellagic acid (EA), a natural polyphenol evidence several pharmacological benefits. The binding profile of EA with human serum albumin (HSA) has been explored and investigated by Isothermal titration calorimetry (ITC), circular dichroism (CD) spectroscopy, time-correlated single-photon counting (TCSPC), absorbance spectroscopy, steady-state fluorescence spectroscopy, and modelling studies. The ITC data analysis revealed the binding Constant (Ka), ΔH, ΔS and ΔG values to be 15.5×104M?1, ?116.2±18.1 Kcal mol?1, ?366 cal mol?1K?1 and ?7.13 Kcal mol?1 respectively with a unique binding site at HSA. EA effectively quenched the intrinsic fluorescence of HSA by static quenching, whereas TCSPC data also revealed association of dynamic quenching also. Thermodynamic analysis confirmed that hydrophobic and mainly hydrogen bonding interaction played important role in stabilizing the HSA-EA complex. It further dictates the binding reaction to be enthalpy driven. The secondary structure of HSA was altered upon binding with EA. CD spectroscopic data indicated the fraction of alpha helicity to be decreased from 52% to 40% upon binding to EA. This study will provide an insight on evaluation of this bioactive interaction during transport and releasing efficiency at the target site in human physiological system since HSA is the most important carrier protein in blood serum.  相似文献

The thermodynamic parameters, ΔH′, ΔG′, and ΔS′, and the stoichiometry for the binding of the substrate 2′-deoxyuridine-5′-phosphate (dUMP) and the inhibitor 5-fluoro-2′-deoxyuridine-5′-phosphate (FdUMP) to Lactobacillus casei thymidylate synthetase (TSase) have been investigated using both direct calorimetric methods and gel filtration methods. The data obtained show that two ligand binding sites are available but that the binding of the second mole of dUMP is extremely weak. Binding of the first mole of dUMP can best be illustrated by dUMP + TSase + H+?(dUMP-TSase-H+). [1] The enthalpy, ΔH1′, for reaction [1] was measured directly on a flow modification of a Beckman Model 190B microcalorimeter. Experiments in two different buffers (I = 0.10 m) show that ΔH1′ = ?28 kJ mol?1 and that 0.87 mol of protons enters into the reaction. Analysis of thermal titrations for reaction [1] indicates a free energy change of ΔG1′ = ?30 kJ mol?1 (K1 = 1.7 × 105 m?1). From these parameters, ΔS1′ was calculated to be +5 J mol?1 degree?1, showing that the reaction is almost totally driven by enthalpy changes. Gel filtration experiments show that at very high substrate concentrations, binding to a second site can be observed. Gel filtration experiments performed at low ionic strength (I = 0.05 m) reveal a stronger binding, with ΔG1′ = ?35 kJ mol?1 (K1 = 1.2 × 106 m?1), suggesting that the forces driving the interaction are, in part, electrostatic. Addition of 2-mercaptoethanol (0.10 m) had the effect of slightly increasing the dUMP binding constant. Binding of FdUMP to TSase is best illustrated by 2FdUMP + TSase + nHH+?FdUMP2 ? TSase ? (H+)nH. [2] The enthalpy for this reaction, ΔH2, was also measured calorimetrically and found to be ?30 kJ mol?1 with nH = 1.24 at pH 7.4 Assuming two FdUMP binding sites per dimer as established by Galivan et al. [Biochemistry15, 356–362 (1976)] our calorimetric results indicate different binding energies for each site. Based on the binding data, a thermodynamic model is presented which serves to rationalize much of the confusing physical and chemical data characterizing thymidylate synthetase.  相似文献

The extraction equilibrium of the hydronium-uranium(VI)-dicyclohexano-24-crown-8 complex was carried out in the crown ether1,2-dichloroethaneHCl aqueous solution system at different temperatures. The extraction complex has the overall composition (L)2·(H3O+·χH2O)2·UO2Cl42− (L = dicyclohexano-24-crown-8). The values of the extraction equilibrium constants (Kex) increase steadily with a decrease in temperature: 13.5 (298 K), 7.96 (301 K), 4.20 (303 K) and 2.07 (305 K). A plot of log Kex against 1/T shows a straight line. The value of the enthalpy change, ΔH°, was calculated from the slope and equals −212 kJ mol−1. The value of the entropy change, ΔS°, was calculated from ΔH° and Kex and equals −690 J K−1 mol−1, whereas ΔG° = −6.45 kJ mol−1. Comparing these thermodynamic parameters with those of the dicyclohexano-18-crown-6 isomer A [1] (ΔS° = −314 J K−1 mol−1, ΔH° = −101 kJ mol−1 and ΔG° = −8.37 kJ mol−1), it can be seen that ΔH° and ΔS° are more negative for the former than for the latter, and both are enthalpy-stabilized complexes. The molecular structure of the complex has the feature that there are two H5O2+ ions in it, in contrast to the H3O+ ions in the dicyclohexano-18-crown-6 isomer A complex [1]. Each of the H5O2+ ions is held in the crown ether cavity by four hydrogen bonds. The H5O2+ ion has a central bond. The uranium atom forms UO2Cl42− as a counterion away from the crown ether. The formation of this complex is in good agreement with more negative entropy change and less negative free energy change, as mentioned above.  相似文献

Intermolecular interaction study of human serum albumin (HSA) with two anthraquinones i.e. danthron and quinizarin has been performed through fluorescence, UV-vis and CD spectroscopy along with docking analysis. The titration of drugs into HSA solution brought about the quenching of fluorescence emission by way of complex formation. The binding constants were found to be 1.51 × 104 L mol?1 and 1.70 × 104 L mol?1 at λexc = 280 nm while at λexc = 295 nm, the values of binding constants were 1.81 × 104 L mol?1 and 1.90 × 104 L mol?1 which hinted toward binding of both the drugs in the vicinity of subdomain IIA. Different temperature study revealed the presence of static quenching mechanism. Moreover, more effective quenching of the fluorescence emission was observed at λexc = 295 nm which also suggested that both the drug molecule bind nearer to Trp-214. Thermodynamic parameters showed that hydrophobic interaction was the major force behind the binding of drugs. The UV-vis spectroscopy testified the formation of complex in both the systems and primary quenching mechanism as static one. The changes in secondary structure and α-helicity in both the systems were observed by circular dichroism spectroscopy. Furthermore, molecular docking analysis predicted the probable binding site of drugs in subdomain IIA of HSA molecule. The types of amino acid residues surrounding the drug molecule advocated that van der Waals forces, hydrophobic forces and electrostatic forces played a vital role in the stabilization of drug-protein complex formed.  相似文献

Interaction of a tyrosine kinase inhibitor, vandetanib (VDB), with the major transport protein in the human blood circulation, human serum albumin (HSA), was investigated using fluorescence spectroscopy, circular dichroism (CD) spectroscopy, and molecular docking analysis. The binding constant of the VDB–HSA system, as determined by fluorescence quenching titration method was found in the range, 8.92–6.89?×?103?M?1 at three different temperatures, suggesting moderate binding affinity. Furthermore, decrease in the binding constant with increasing temperature revealed involvement of static quenching mechanism, thus affirming the formation of the VDB–HSA complex. Thermodynamic analysis of the binding reaction between VDB and HSA yielded positive ΔS (52.76 J?mol?1 K?1) and negative ΔH (?6.57?kJ?mol?1) values, which suggested involvement of hydrophobic interactions and hydrogen bonding in stabilizing the VDB–HSA complex. Far-UV and near-UV CD spectral results suggested alterations in both secondary and tertiary structures of HSA upon VDB-binding. Three-dimensional fluorescence spectral results also showed significant microenvironmental changes around the Trp residue of HSA consequent to the complex formation. Use of site-specific marker ligands, such as phenylbutazone (site I marker) and diazepam (site II marker) in competitive ligand displacement experiments indicated location of the VDB binding site on HSA as Sudlow’s site I (subdomain IIA), which was further established by molecular docking results.
