A human/mouse hybridoma was developed which has the property of secreting a human bone resorbing factor similar or identical to the osteoclast activating factor (OAF) isolated from human tonsil lymphocytes. Mouse plasmacytoma cells negative for OAF production were fused with an enriched subpopulation of human tonsil lymphocytes that had been activated with phytohemagglutinin (PHA) to produce OAF (G. E. Nedwin, M. A. Mohler, and R. A. Luben, submitted for publication). Culture supernatants from mixed hybridomas contained a bone resorbing protein shown to cause the release of 45Ca from previously labeled mouse calvaria. The bone resorbing activity from these hybridomas was inhibited by the presence of OAF-specific monoclonal antibodies. Several hybridomas retained OAF production following limited dilution cloning. One clone, CD6.20, showed a biphasic dose-response curve for bone resorption similar to that of purified OAF from PHA-activated human tonsil lymphocytes. OAF production in the CD6.20 cell line has been retained for over 100 passages. Karyotype analysis of this cell shows the presence of human chromosomes 10 and 18 and the X chromosome. 相似文献

A macromolecular material that enhances the translocation, or binding, of already "activated" receptor-glucocorticoid complex to nuclei in the presence of 5 mM ATP was separated from the cytosol of rat liver by DEAE-cellulose column chromatography with about 0.025 M NaCl. The molecular weight of the material was about 93,000 +/- 4,900, as determined by agarose gel filtration. After incubation at 60 degrees C for 15 min, this material still had activity to increase the nuclear binding, but on boiling for 15 min it lost its activity. 相似文献

The effects of monosaccharides on the cytotoxic activity of cytotoxic T lymphocytes (CTL) and three cloned long-term cytotoxic T-lymphocyte lines (CTLL) are compared. Uncultured CTL and clones CTLL-A2 and CTLL-A11 were derived from the peritoneal cavity of C57BL/6 mice immunized against the H-2Dd determinants on the BALB/c sarcoma Meth A. Clone CTLL-R5 was derived from spleen of (BALB/c X C57BL)F1 mice immunized against a unique determinant on the BALB/c radiation-induced leukemia RL male 1. The cell-surface phenotype of the clones is Lyt-1+,2+,3+. Cytotoxic activity of CTLL-A2 and CTLL-R5 as determined by a 4-hr 51Cr-release assay was inhibited over 50% by 1 mM 2-deoxy-D-glucose. CTLL-A11 and the uncultured cytotoxic T cells were more resistant to inhibition by 2DG (40% at 20 mM). Surprisingly, it was found that the addition of D-mannose, D-galactose, D-glucose, L-fucose, alpha-methyl-D-mannose, and N-acetyl-D-glucosamine also inhibited, in a dose-related manner, the cytotoxicity of CTLL-A2 and CTLL-A11. CTLL-R5 showed a more restricted inhibition pattern: only D-mannose and D-galactose were inhibitory. The mechanism of inhibition remains to be clarified. 相似文献

L4-PHA (L4) and E4-PHA (E4) lectins isolated from Phaseolus vulgaris have different mitogenic properties. The mechanisms of the differences in mitogenic behavior were sought in the interaction of lectin, lymphocyte subsets, and T-cell growt factor (TCGF) also known as interleukin 2 (IL-2). TCGF activity in culture supernatants ( L4S ; E4S ) from L4- and E4-stimulated, freshly isolated lymphocytes was assayed as stimulation of DNA synthesis in TCGF-dependent continuous T-cell cultures (CTC). E4S contained less TCGF than did L4S . Addition of partially purified TCGF does not increase the stimulation of fresh lymphocytes by L4 or E4. L4 and E4 equally stimulate both helper (OKT4+) and suppressor (OKT8+) cells. The ability of L4 to further stimulate CTC is slowly lost (15 greater than 30 greater than 45 days). It is concluded that production of TCGF is not rate limiting in E4 and L4 stimulation of lymphocytes. The growth of CTC, which requires the presence of TCGF, remains sensitive to, but not dependent on, L4 for at least 30 days. 相似文献

Thymocytes, isolated 6 days following subcutaneous (sc) transplantation of BALB/c MOPC-315 plasmacytoma into F1 (BALB/c × C57BL) hybrid mice, when injected sc into normal syngeneic mice, caused the development of a solid sc tumor. The cells of the newly developed tumor were of a mixed population of θ+F1 (BALB/c × C57BL) and θ? BALB/c cells (approximately 1:1), which represents a new type of mixed T cell-plasma cell neoplasm. Efforts were made to isolate the transformed thymocytes (θ+) from the plasmacytoma (θ?) cells in the new tumor, exploiting differences in their surface properties. Treatment of the mixed tumor cell population with peanut agglutinin (PNA) revealed that only the T tumor cells were agglutinated. The agglutinated cells were recovered after dispersing the clumps with d-galactose (0.15 M) and consisted of 95% θ+ cells. The PNA-agglutinated cells were found to induce a similar tumor (85% θ+ cells) when injected sc into F1 (BALB/c × C57BL) mice. 相似文献

A mutant cell line derived from Chinese hamster fibroblasts is described which is defective in oxidative energy metabolism. Glucose is continuously required in the medium. As a result of a block in the Krebs cycle, these cells are auxotrophs for carbon dioxide and asparagine. Several experiments support our conclusion that the mutant cells lack appreciable levels of succinate dehydorgenase activity. Other components of the electron transport chain appear to be fully functional, although there is the possibility that electron transport and oxidative phosphorylation are uncoupled. 相似文献

The inhibitory effects of seven compounds (hydrocortisone, histamine, interferon, isoproterenol, norepinephrine, serotonin, and γ-aminobutyric acid) were tested in phytohemagglutinin (PHA)-stimulated cultures of human peripheral blood mononuclear cells as a function of PHA concentration. The cultures stimulated by suboptimal levels of PHA were much more sensitive to inhibition by the added compounds, with the curve of percentage inhibition versus concentration of inhibitor shifted to the left by greater than two orders of magnitude in all cases. At the lower doses of mitogen, concentrations of hydrocortisone, histamine, interferon, and isoproterenol which approach the concentrations of these or analogous compounds in vivo caused significant and substantial inhibition of the mitogen response. 相似文献

Chemotactic bacteria, such as E. coli, detect changes in the chemical composition of the environment. Addition of an attractant or repellent leads to an immediate response, characterized by a change in the swimming behavior of the cells--a process known as sensory excitation. However, the response gradually disappears with time, despite the continued presence of the chemical--a process known as sensory adaptation. We report here the behavior of a class of nonchemotactic mutants (cheX) that can carry out sensory excitation but are defective in the process of sensory adaptation. These mutants are also defective in the ability to carry out a protein methylation reaction which has previously been implicated in the adaptation process (Goy, Springer and Adler, 1977). The results presented here establish a firm relationship between the methylation reaction and sensory adaptation. 相似文献

Splenic leucocyte effector cells from 3-month-old C3H/HeJ mice which were antibody negative for influenza A virus were found to be spontaneously cytotoxic for mouse L929 fibroblasts infected with influenza virus, but not for uninfected cells. The cytotoxic activity was significantly enhanced if the effector cells were preincubated with splenic leucocytes from older 6- or 9-month-old antibody-negative mice before being added to the target cells. Enhancement appeared to be due to a cooperative effect between leucocytes from mice of different ages. Preincubation of effector cells with autologous sera markedly suppressed spontaneous cytotoxicity, but the suppression was significantly less with sera from 7- or 10-month-old mice than with sera from 3-month-old mice. Most cytotoxic activity was observed with a nylon wool-adherent subpopulation of leucocytes, although some cytotoxicity remained in the non-adherent fraction. The results suggested that spontaneous cytotoxicity was due to two subpopulations of cells, possibly adherent monocyte-macrophages and nonadherent natural killer cells. 相似文献

Dialyzed fetal calf serum (FCS) was a poor source of serum supplement for in vitro cytotoxic T lymphocyte (CTL) generation. Serum dialysate or biotin fully restored dialyzed FCS to activities comparable to FCS. It was concluded that the active principal in serum dialysate was biotin because its further dialysis was prevented by addition of avidin, a biotin binding protein. Avidin inhibited CTL generation only when added during the early stages of mixed lymphocyte cultures, whereas biotin could restore activity even if added at a later time. When FCS enriched in a fatty acid mixture, or in palmitic acid alone, was used as the serum supplement, avidin-mediated inhibition of CTL generation was markedly reduced. Avidin also inhibited CTL generation in cultures containing killed macrophages as the stimulating cell, and supplemented with Con-A-induced spleen cell supernatant, a source of helper factor(s). These experiments suggest that fatty acid biosynthesis and the attendant synthesis of structural lipids of appropriate fatty acid composition play a prominent role in the generation of CTL 相似文献

The interaction of the nerve growth factor with the neurotubule protein has been studied with the aim of elucidating the nature of the large complexes that they form when incubated together and the factors that control this event. The results show that the binding of nerve growth factor to tubulin is followed by the formation of large structures that, in certain experimental conditions, accelerate the rate of tubulin polymerization to form microtubules or catalyze their assembly in conditions where this process does not occur spontaneously. The formation of large nerve growth factor-tubulin complexes starts to occur only at a molar ratio of 1.0–1.5 NaCl or GTP strongly inhibit this process without a detectable effect on NGF binding. Two hypotheses are postulated to explain these finding. Firstly, that tubulin has two sites with different affinity for nerve growth factor and the polymerization occurs only when the second NGF molecule has interacted with the microtubule protein. Alternatively, free tubulin in solution is the limiting factor of the polymerization by hindering a site of tubulin-factor complexes present in solutio at a 1 : 1 molar ratio. In both cases, GTP, Na+ or H+ will affect the formation of large unsoluble, tubulin-NGF complexes, by changing their conformation or by decreasing electrostatic interactions. 相似文献

Probabilistic automata are compared with deterministic ones in simulations of growing networks made of dividing interconnected cells. On examples of chains, wheels and tree-like structures made of large numbers of cells it is shown that the number of necessary states in the initial generating cell automaton is reduced drastically when the automaton is probabilistic rather than deterministic. Since the price being paid is a decrease in the accuracy of the generated network, conditions under which reasonable compromises can be achieved are studied. They depend on the degree of redundancy of the final network (defined from the complexity of a deterministic automaton capable of generating it with maximum accuracy), on the "entropy" of the generating probabilistic automaton, and on the effects of different inputs on its transition probabilities (as measured by its "'capacity" in the sense of Shannon's information theory). The results are used to discuss and make more precise the notion of biological specificity. It is suggested that the weak metaphor of a genetic program, classically used to account for the role of DNA in specific genetic determinations, is replaced by that of inputs to biochemical probabilistic automata. 相似文献

When the surfactant mixture Bio-Solv-BBS-3 is added to a scintillation solvent it acts as a primary scintillator in response to β emissions (and Compton electrons from γs). The fluorescence excitation threshold is higher and fluorescence yield is lower than those of the primary scintillators usually employed in scintillation counting. Presence of a surfactant in a sample containing 14C or more energetic βs will be counted at higher efficiency than would be indicated by a quench correction curve (efficiency vs sample channels ratios or external standard channels ratios) derived from standards not containing surfactant. 相似文献

Human plasma Factor II has been purified approximately 800-fold by a combination of barium citrate adsorption, ion-exchange chromatography and preparative polyacrylamide gel electrophoresis. The procedure is relatively simple and results in excellent yields of purified Factor II essentially free of Factor X activity. The purified factor behaved as a single component by analytical polyacrylamide gel disc electrophoresis at pH 8.9. No Factor V, VII or IX activity was detected in the purified Factor II. Its molecular weight was 7200±3000 as determined by analytical ultracentrifugation, electrophoresis in the presence of sodium dodecyl sulfate and gel filtration on Bio-Gel P-200. An apparent molecular weight of 90 000–100 000 was observed on calibrated columns of Sephadex G-100, G-150, and G-200. The specific activity of human factor II was approximately 1300 N.I.H. units/mg as determined by the two-stage assay and 7 Ortho units/mg by the one stage assay. The purified protein contained by weight 2.8% neutral hexose, 2.3% sialic acids and 3.1% hexosamines. 相似文献

Copper has been demonstrated in the feet of slugs exposed to the molluscicide copper sulfate. Using the scanning electron microscope coupled with X-ray microanalysis, copper was localized to the epithelium of the foot. Under the transmission electron microscope the subcellular distribution of copper was obtained by precipitating the molluscicide with potassium ferrocyanide. The reaction product was both electron opaque and insoluble in the preparative media. The precipitate was localized in the zonula adhaerens and septate junctions between adjacent epithelial cells. Reaction product was also present in some of the pinocytotic vesicles of the epithelial cells. The identity of the reaction product was confirmed using X-ray microanalysis. 相似文献

Cholera toxin inhibition of progesterone-induced meiosis of Xenopus laevis oocytes in vitro has been correlated with increased cAMP levels. Inhibition of germinal vesicle breakdown (Gvbd) and cAMP increase occurred after a lag period of 2 hr, when cholera toxin was injected, or 4--5 hr, when applied externally. The ability of the maturation-promoting factor (Mpf) to provoke Gvbd when injected into recipient oocytes was found to be dependent upon whether the oocytes had been exposed to cholera toxin alone or to toxin and progesterone. With the former, cAMP levels were elevated and Mpf activity was abolished, whereas with the latter, the increase in cAMP was less pronounced and Mpf activity was observed. Injection of cAMP or its 8-thio derivatives shortly before the appearance of progesterone-induced Mpf abolished Gvbd. If injected earlier or later, no inhibition was observed. In contrast, cholera toxin inhibited maturation even when added several hours before progesterone, suggesting a sustained accumulation of cAMP. No Gvbd occurred when 8-thio-methyl-cAMP was injected together with Mpf. These data suggest that cAMP is involved in the control of the formation/amplification and/or activity of Mpf-a result which may be of general significance in cell division mechanisms. 相似文献

This report examines the histochemical staining patterns, ultrastructure, and cell-surface phenotypes of six antigen-specific T-cell clones. Histochemical analyses indicated that all cell lines expressed alpha-napthyl butyrate esterase characteristic of the monocytic isoenzyme, intense napthol AS-D chloroacetate reactivity characteristic of granulocytes, and were negative for leucoperoxidase, alkaline phosphatase, and Sudan black. Only one clone stained weakly for acid phosphatase. The esterase staining patterns became evident in newly established cell lines after growth for only 1 week in interleukin 2-conditioned medium. Ultrastructurally, the outstanding feature was numerous membrane-bound granules containing a complex-appearing globular material. The cell-surface phenotypes of the lines as determined by protein A-sheep red blood cell rosetting and indirect immunofluorescence was Ly-5+, T200+, Qa-5-, MAC-1-, and Lyt-1+,2,3- for the helper line and Lyt-1-2,3+ for the five cytotoxic lines. By quantitative absorption analyses, low levels of Lyt-1 antigens were detected on all examined cytotoxic lines. The results strengthen the view that long-term T-cell lines can retain normal T-cell characteristics while also expressing markers that are either absent or in undetectable levels on uncultured T lymphocytes. The presence of the esterases may be associated with an expanded functional role in the T-cell lines. 相似文献

Lymphocyte-activating factor (LAF), a mitogen for thymocytes and T lymphocytes, is released into the culture medium by human mononuclear cells and mouse peritoneal exudate cells following treatment with various macrophage stimulants. Experiments were performed to determine if recently described mouse macrophage cell lines released LAF in response to stimulation with bacterial lipopolysaccharide. Four continuous cell lines (P388-D1, J774, WEHI 3, and PU5-1.8) were found to release LAF in serum-free medium following endotoxin stimulation. The results of partial purification indicated that LAF obtained from cell lines had a higher molecular weight and lower isoelectric point than LAF from human mononuclear cells. 相似文献

A human lymphokine, eosinophil stimulation promoter (ESP), was shown to be produced by mononuclear leukocytes in response to non-helminthic antigens. Supernatants of tuberculin-stimulated cultures of mononuclear leukocytes from tuberculinskin test reactive humans contained non-dialyzable ESP activity; ESP could not be demonstrated in the supernatants of tuberculin stimulated cultures from skin test negative donors. Thus, human ESP, like murine ESP, is a soluble lymphokine product of sensitized mononuclear cells. ESP activity was also demonstrated in supernatants of mononuclear leukocyte cultures stimulated with streptococcal antigens. ESP selectively enhanced migration of eosinophils. ESP production could be blocked by puromycin, and its activity was diminished by prior incubation with eosinophil-rich granulocytes. Correlation of antigen stimulated ESP activity with skin test reactivity establishes ESP as another in vitro correlate of delayed hypersensitivity in man. A video method of data collection and analysis is described which greatly facilitates in vitro studies of ESP activity. The method is applicable to the study of other lymphokines which effect cellular migration. 相似文献
