Background/Objective

Describing transmissibility parameters of past pandemics from diverse geographic sites remains critical to planning responses to future outbreaks. We characterize the transmissibility of influenza A(H1N1)pdm09 (hereafter pH1N1) in South Africa during 2009 by estimating the serial interval (SI), the initial effective reproductive number (initial Rt) and the temporal variation of Rt.

Methods

We make use of data from a central registry of all pH1N1 laboratory-confirmed cases detected throughout South Africa. Whenever date of symptom onset is missing, we estimate it from the date of specimen collection using a multiple imputation approach repeated 100 times for each missing value. We apply a likelihood-based method (method 1) for simultaneous estimation of initial Rt and the SI; estimate initial Rt from SI distributions established from prior field studies (method 2); and the Wallinga and Teunis method (method 3) to model the temporal variation of Rt.

Results

12,360 confirmed pH1N1 cases were reported in the central registry. During the period of exponential growth of the epidemic (June 21 to August 3, 2009), we simultaneously estimate a mean Rt of 1.47 (95% CI: 1.30–1.72) and mean SI of 2.78 days (95% CI: 1.80–3.75) (method 1). Field studies found a mean SI of 2.3 days between primary cases and laboratory-confirmed secondary cases, and 2.7 days when considering both suspected and confirmed secondary cases. Incorporating the SI estimate from field studies using laboratory-confirmed cases, we found an initial Rt of 1.43 (95% CI: 1.38–1.49) (method 2). The mean Rt peaked at 2.91 (95% CI: 0.85–2.91) on June 21, as the epidemic commenced, and Rt>1 was sustained until August 22 (method 3).

Conclusions

Transmissibility characteristics of pH1N1 in South Africa are similar to estimates reported by countries outside of Africa. Estimations using the likelihood-based method are in agreement with field findings.  相似文献

Background

The emergence in 2014 and persistence of African Swine Fever (ASF) in Lithuania has been linked to infected wild boar movement and close contact with the carcasses of other infected wild boars. Over time the number of reported cases of ASF in wild boars gradually increased, but no detailed epidemiological data has been available. Therefore, the objective of the present study was to determine ASF virus prevalence in wild boars and domestic pigs during the 2014–2017 period and further explore the current geographical distribution of the virus.

Results

Our study results show that ASF virus prevalence in hunted wild boars using PCR analysis increased from 0.83% (95% CI 0.69–0.98) to 2.27% (95% CI 2.05–2.48) from 2014 to 2016 respectively. However, there was a dramatic jump in the number of ASF positive wild boars cases in 2017 resulting in prevalence of 12.39% (95% CI 11.91–12.86) (p <?0.05).The average prevalence of ASF-specific antibodies in wild boar population during years 2014–2017 was 0.45% (95% CI 0.39–0.51) based on ELISA test results.Prevalence of ASF virus in domestic pigs ranged from 0.24% (95% CI 0.17% - 0.32) in 2015 to 2.74% (95% CI 2.33% - 3.15) in 2017. The average seasonal prevalence of ASF virus in pigs was statistically significant (p?<?0.05) and ranged from 0% in spring to 3.68% (95% CI 3.32–4.05) in summer. Correlation between the pig density and number of recorded pig ASF cases in affected regions was only found in 2017 (R =?0.78, p?<?0.05). No correlation was detected between the wild boar density and number of recorded pig or wild boar ASF - positive cases.

Conclusions

This study provides the first results of ASF virus prevalence changes in Lithuania during the 2014–2017. The overall results confirm the relatively high prevalence of ASF virus in wild boar that was gradually increasing from 2014 to 2017. In the last year of study, the number of ASF positive cases in both domestic pigs and wild boars had unexpectedly increased several times. A better understanding of current status of the disease will enable better control and prevent further spread of ASF virus in Western Europe.

Hepatitis A virus (HAV) is a food-borne enteric virus responsible for outbreaks of hepatitis associated with shellfish consumption. The objectives of this study were to determine the thermal inactivation behavior of HAV in blue mussels, to compare the first-order and Weibull models to describe the data, to calculate Arrhenius activation energy for each model, and to evaluate model efficiency by using selected statistical criteria. The times required to reduce the population by 1 log cycle (D-values) calculated from the first-order model (50 to 72°C) ranged from 1.07 to 54.17 min for HAV. Using the Weibull model, the times required to destroy 1 log unit (tD = 1) of HAV at the same temperatures were 1.57 to 37.91 min. At 72°C, the treatment times required to achieve a 6-log reduction were 7.49 min for the first-order model and 8.47 min for the Weibull model. The z-values (changes in temperature required for a 90% change in the log D-values) calculated for HAV were 15.88 ± 3.97°C (R2, 0.94) with the Weibull model and 12.97 ± 0.59°C (R2, 0.93) with the first-order model. The calculated activation energies for the first-order model and the Weibull model were 165 and 153 kJ/mol, respectively. The results revealed that the Weibull model was more appropriate for representing the thermal inactivation behavior of HAV in blue mussels. Correct understanding of the thermal inactivation behavior of HAV could allow precise determination of the thermal process conditions to prevent food-borne viral outbreaks associated with the consumption of contaminated mussels.  相似文献

Hyadaphis foeniculi (Passerini) (Hemiptera: Aphididae) is a cosmopolitan species and the main pest of fennel in northeastern Brazil. Understanding the relationship between temperature variations and the population growth rates of H. foeniculi is essential to predict the population dynamics of this aphid in the fennel crop. The aim of this study was to measure the effect of constant temperature on the adult prereproductive period and the life table fertility parameters (infinitesimal increase ratio (rm), gross reproduction rate (GRR), net reproduction rate (R0), finite increase ratio (λ), generation time (GT), the time required for the population to double in the number of individuals (DT), and the reproduction value (RVx)) of the fennel pest H. foeniculi. The values of lx (survival of nymphs at age x) increased as the temperature rose from 15 to 28°C and fell at 30°C, whereas mx (number of nymphs produced by each nymph of age x) increased from 15 to 25°C and fell at 28 and 30°C. The net reproduction rates (R0) of populations of H. foeniculi increased with temperature and ranged from 1.9 at 15°C to 12.23 at 28°C for each generation. The highest population increase occurred with the apterous aphids at 28°C. The rate of population increase per unit time (rm) (day) ranged from 0.0033 (15°C) to 0.1995 (28°C). The highest values of rm were recorded at temperatures of 28°C and 30°C. The rm values were a good fit to the models tested, with R2 > 0.91 and R2 adj > 0.88. The models tested (Davidson, Sharpe and DeMichele modified by Schoolfield et al., Logan et al., Lamb, and Briere et al.) were very good fits for the rm values observed, with R2 > 0.91 and R2 adj > 0.88. The only exception was the Davidson model. Of the parameters studied, the reproductive capacity was higher in the apterous aphids, with the unique exception of daily fecundity at 28°C, which was higher in the alate aphids of H. foeniculi. Parameters relating to the age-specific fertility table for H. foeniculi were heavily influenced by temperature, with the highest biotic potential and population growth capacity found at 34°C. Therefore, the results obtained in this study could be of practical significance for predicting outbreaks of fennel aphids and improving the management of this aphid in fennel crops.  相似文献

Background

Accurately assessing the transmissibility and serial interval of a novel human pathogen is public health priority so that the timing and required strength of interventions may be determined. Recent theoretical work has focused on making best use of data from the initial exponential phase of growth of incidence in large populations.

Methods

We measured generational transmissibility by the basic reproductive number R0 and the serial interval by its mean Tg. First, we constructed a simulation algorithm for case data arising from a small population of known size with R0 and Tg also known. We then developed an inferential model for the likelihood of these case data as a function of R0 and Tg. The model was designed to capture a) any signal of the serial interval distribution in the initial stochastic phase b) the growth rate of the exponential phase and c) the unique combination of R0 and Tg that generates a specific shape of peak incidence when the susceptible portion of a small population is depleted.

Findings

Extensive repeat simulation and parameter estimation revealed no bias in univariate estimates of either R0 and Tg. We were also able to simultaneously estimate both R0 and Tg. However, accurate final estimates could be obtained only much later in the outbreak. In particular, estimates of Tg were considerably less accurate in the bivariate case until the peak of incidence had passed.

Conclusions

The basic reproductive number and mean serial interval can be estimated simultaneously in real time during an outbreak of an emerging pathogen. Repeated application of these methods to small scale outbreaks at the start of an epidemic would permit accurate estimates of key parameters.  相似文献

Neoseiulus californicus (McGregor) (Acari: Phytoseiidae) is one of the most important and effective predators of Tetranychus spp. (Acari: Tetranychidae). This study analyzed the effects of hexythiazox and spiromesifen resistance on biological characteristics of N. californicus. Pre-oviposition, oviposition, post-oviposition times, adult lifespan, total number of eggs laid per female, net reproductive rate (R0), intrinsic rate of increase (rm), doubling time (DT), mean generation time (T) and finite rate of increase (λ) were compared among three populations: 63.9-fold hexythiazox resistant (HEX14), 53.6-fold spiromesifen resistant (SPR13) and a susceptible base population. Pre-oviposition and oviposition times, mean number of eggs/female, adult lifespan, R0 and rm values were all significantly higher for the two resistant populations than for the susceptible population. Life tables of HEX14, SPR13 and the susceptible population showed that R0 was 35.0, 26.5 and 19.4 (females/female generation), rm was 0.35, 0.32 and 0.30 (females/female/day), DT was 1.92, 2.07 and 2.27 days, T was 9.8, 9.8 and 9.7 days, and λ was 1.43, 1.39 and 1.35 (individuals/female/day), respectively.  相似文献

Andalusia (Southern Spain) is considered one of the main routes of introduction of bluetongue virus (BTV) into Europe, evidenced by a devastating epidemic caused by BTV-1 in 2007. Understanding the pattern and the drivers of BTV-1 spread in Andalusia is critical for effective detection and control of future epidemics. A long-standing metric for quantifying the behaviour of infectious diseases is the case-reproduction ratio (Rt), defined as the average number of secondary cases arising from a single infected case at time t (for t>0). Here we apply a method using epidemic trees to estimate the between-herd case reproduction ratio directly from epidemic data allowing the spatial and temporal variability in transmission to be described. We then relate this variability to predictors describing the hosts, vectors and the environment to better understand why the epidemic spread more quickly in some regions or periods. The Rt value for the BTV-1 epidemic in Andalusia peaked in July at 4.6, at the start of the epidemic, then decreased to 2.2 by August, dropped below 1 by September (0.8), and by October it had decreased to 0.02. BTV spread was the consequence of both local transmission within established disease foci and BTV expansion to distant new areas (i.e. new foci), which resulted in a high variability in BTV transmission, not only among different areas, but particularly through time, which suggests that general control measures applied at broad spatial scales are unlikely to be effective. This high variability through time was probably due to the impact of temperature on BTV transmission, as evidenced by a reduction in the value of Rt by 0.0041 for every unit increase (day) in the extrinsic incubation period (EIP), which is itself directly dependent on temperature. Moreover, within the range of values at which BTV-1 transmission occurred in Andalusia (20.6°C to 29.5°C) there was a positive correlation between temperature and Rt values, although the relationship was not linear, probably as a result of the complex relationship between temperature and the different parameters affecting BTV transmission. Rt values for BTV-1 in Andalusia fell below the threshold of 1 when temperatures dropped below 21°C, a much higher threshold than that reported in other BTV outbreaks, such as the BTV-8 epidemic in Northern Europe. This divergence may be explained by differences in the adaptation to temperature of the main vectors of the BTV-1 epidemic in Andalusia (Culicoides imicola) compared those of the BTV-8 epidemic in Northern Europe (Culicoides obsoletus). Importantly, we found that BTV transmission (Rt value) increased significantly in areas with higher densities of sheep. Our analysis also established that control of BTV-1 in Andalusia was complicated by the simultaneous establishment of several distant foci at the start of the epidemic, which may have been caused by several independent introductions of infected vectors from the North of Africa. We discuss the implications of these findings for BTV surveillance and control in this region of Europe.  相似文献

The reaction of Pt(COD)Cl2, where COD is 1,5-cyclooctadiene, with one equivalent of a diamidato-bis(phosphino) Trost ligand ((R,R)-2 = N,N′-bis(2-diphenylphosphino-1-benzoyl)-(1R,2R)-1,2-diaminocyclohexane, (R,R)-N,N′-bis(2-diphenylphosphino-1-naphthoyl)-(1R,2R)-1,2-diaminocyclohexane, or (±)-N,N′-bis(2-diphenylphosphino-1-benzoyl)-1,2-bis(aminobenzene)) in the presence of base afforded square planar diamidato-bis(phosphino) platinum(II) complexes (R,R)-2-Pt, (R,R)-3-Pt, (±)-4-Pt. Characterization of all complexes included the solution and solid state structure determination of each complex based on multinuclear NMR and X-ray analyses, respectively. Stability of the complexes in acid was examined on addition of HCl to (R,R)-2-Pt in chloroform and compared to the unreactive nature of the similar diamidato-bis(phosphino) complex 1-Pt (= 1,2-bis-N-[2′-(diphenylphosphino)benzoyl]diamino-benzene) in the presence of acid. Protonation of the bound amidato nitrogen atoms of (R,R)-2-Pt was observed along with decoordination of the nitrogen atoms from the platinum(II) center producing (R,R)-2-PtCl2 in quantitative yield by NMR analysis. Confirmation of the product was made on comparison of the NMR spectra to that of authentic (R,R)-2-PtCl2 prepared on reaction of Pt(COD)Cl2 with (R,R)-2 in CH2Cl2 and characterized by single-crystal X-ray diffraction analysis and NMR spectroscopy. Results add to the knowledge of rich coordination chemistry of bis(phosphino) ligands with late transition metals, metal-amidato chemistry, and has implications in catalysis.  相似文献

Substituted salicylaldehydes [C6HR1R2R3(CHO)(OH)] react with CoMe3(PMe3)3 to afford 6-coordinate (cis-dimethyl)(2-formyl-phenolato)trans-bis(trimethylphosphine)cobalt(III) compounds Co[C6HR1R2R3(CHO)(O)Me2](PMe3)2 (1: R1 = H; R2 = Me; R3 = tert-Bu; 2: R1, R2 = C6H4; R3 = H). Accordingly, substituted enolated malonic dialdehydes (CHO-CR4CR5-OH) react with CoMe3(PMe3)3 to afford 6-coordinate (cis-dimethyl)(2-formyl-enolato)trans-bis(trimethylphosphine)cobalt(III) compounds Co[(CHO-CR4CR5-O)(Me)2](PMe3)2 (3: R4, R5 = (CH2)2C6H4; 4: R4 = R5 = C6H5). In the molecular structure of 4, the cobalt atom is centred in an octahedral coordination geometry brought about by a six-membered chelate ring (O:O-ligand), cis-dimethyl and trans-trimethylphosphine groups. A reaction mechanism is suggested.  相似文献

Background

The clinical attack rate of influenza is influenced by prior immunity and mixing patterns in the host population, and also by the proportion of infections that are asymptomatic. This complexity makes it difficult to directly estimate R0 from the attack rate, contributing to uncertainty in epidemiological models to guide pandemic planning. We have modelled multiple wave outbreaks of influenza from different populations to allow for changing immunity and asymptomatic infection and to make inferences about R0.

Data and Methods

On the island of Tristan da Cunha (TdC), 96% of residents reported illness during an H3N2 outbreak in 1971, compared with only 25% of RAF personnel in military camps during the 1918 H1N1 pandemic. Monte Carlo Markov Chain (MCMC) methods were used to estimate model parameter distributions.

Findings

We estimated that most islanders on TdC were non-immune (susceptible) before the first wave, and that almost all exposures of susceptible persons caused symptoms. The median R0 of 6.4 (95% credibility interval 3.7–10.7) implied that most islanders were exposed twice, although only a minority became ill in the second wave because of temporary protection following the first wave. In contrast, only 51% of RAF personnel were susceptible before the first wave, and only 38% of exposed susceptibles reported symptoms. R0 in this population was also lower [2.9 (2.3–4.3)], suggesting reduced viral transmission in a partially immune population.

Interpretation

Our model implies that the RAF population was partially protected before the summer pandemic wave of 1918, arguably because of prior exposure to interpandemic influenza. Without such protection, each symptomatic case of influenza would transmit to between 2 and 10 new cases, with incidence initially doubling every 1–2 days. Containment of a novel virus could be more difficult than hitherto supposed.  相似文献

Bis(azido)bis(phosphine)-Pd(II) and -Pt(II) complexes, [M(N3)2L2] {L = PMe3, PEt3, PMe2Ph, dppe = 1,2-bis(diphenylphosphino)ethane}, underwent 1,3-dipolar cycloaddition with organic chiral isothiocyanates (R-NCS: R = (S)-(+)-1-phenylethyl, (R)-(−)-1-phenylethyl, (±)-1-phenylethyl, (S)-(+)-1-indanyl) to give the corresponding tetrazole-thiolato Pd(II) and Pt(II) complexes, trans-[M{S[CN4(R)]}2L2] or [M{S[CN4(R)]}2(dppe)]. Spectroscopic (IR and NMR) and X-ray structural analyses of the products showed that the absolute configuration of the starting organic isothiocyanates is retained throughout the reaction. Further treatments of the isolated tetrazole-thiolato complexes with electrophiles such as HCl or benzoyl chloride produced heterocyclic compounds containing a tetrazole thione or a tetrazolyl sulfide group. In addition, organic tetrazole thiones, [S = {CN4H(R)}] containing a chiral moiety, were prepared from NaN3 and R-NCS in the presence of water.  相似文献

We report the response of carotenoids and chlorophylls during 120 h time series virus infection experiments of the marine coccolithophorid Emiliania huxleyi (Lohm.) Hay et Mohler culture. The response of individual carotenoids to infection varied: Diatoxanthin (Dtx) increased rapidly relative to chlorophyll-a, whereas diadinoxanthin (Ddx) and β-carotene showed a rapid decrease and fucoxanthin and 19′hexanoyloxyfucoxanthin a slight increase. The response of the individual carotenoids reflects their role in epoxy/de-epoxidation cycling, antioxidant protection, biosynthetic conversion and vulnerability to photooxidative destruction. We observed for the first time the operation of the diadinoxanthin cycle occurring in response to viral infection in E. huxleyi with the de-epoxidation ratio (Dtx / (Dtx + Ddx)) increasing exponentially with time (R2 = 0.92) and decreasing exponentially with FV / FM (R2 = 0.97). Our findings contribute to our understanding of the conversion and fate of key biochemical cell constituents in algae and are important in understanding the physiological stress response to virus infection.  相似文献

The reaction of (COD)PdCl2 (COD = 1,5-cyclooctadiene) with (3-Py)2SiR1R2 (3-Py = 3-pyridyl; R1 = Ph, R2 = Ph (m-pdps); R1 = Ph, R2 = Me (m-pmps)) in acetone affords single crystals consisting of cyclodimers, [PdCl2((3-Py)2SiR1R2)]2, whereas the same reaction in a mixture of dichloromethane and ethanol yields amorphous spheres consisting of cyclotrimers, [PdCl2((3-Py)2SiR1R2)]3. In a boiling chloroform solution, the cyclodimers are completely converted to cyclotrimers. These cyclotrimers, in the 10−60 °C range, are partly returned to cyclodimers. By contrast, the reaction of (COD)PdCl2 with (3-Py)2SiR1R2 (R1 = Bu, R2 = Me (m-pbms); R1 = dodecyl, R2 = Me (m-pddms)) yields amorphous spheres consisting of cyclotrimers irrespective of solvents. Both [PdCl2(m-pbms)]3 and [PdCl2(m-pddms)]3 are initially cyclotrimers in chloroform, but they exist as a mixture of cyclodimers and cyclotrimers in solution in the 10−60 °C range. The metallacycles tend to form cyclodimers in the order m-pdps > m-pmps > m-pbms > m-pddms. The equilibrium between cyclodimers and the cyclotrimers is sensitive to solvent, temperature, and concentration as well as molecular structure.  相似文献

The [PdII{(R,R)-chxn}(OH)2] reagent (chxn = 1,2-diaminocyclohexane) is introduced as a metal probe for the detection of the bidentate chelating sites of a glycose. Two moles of hydroxide per mole palladium support double deprotonation of potentially chelating diol functions at a glycose’s backbone. The individual chelating sites are detected using one- and two-dimensional NMR techniques. At equimolar amounts of palladium(II) and aldose, the metal-binding sites include mostly the hydroxy function at the anomeric carbon atom. Chelators are derived from both the pyranose and the furanose isomers. Most pyranose-based chelators form five-membered chelate rings by using their 1,2-diol function. Though 1,2-diolate bonding is also common to the furanoses, the formation of six-membered chelate rings by 1,3-bonding is more significant for them. Metal-excess conditions provoke mostly bis-bidentate 1,2;3,4-chelation but unusual isomers form also: thus d-xylose is dimetallated in its all-axial β-pyranose form, and erythrose’s dimetallation results in the formation of two isomers of a metal derivative of the open-chain hydrate. The spectroscopic results are supported by crystal-structure determinations on [Pd{(R,R)-chxn}(α-d-Xylp1,2H−2O1,2)]·H2O (Xyl = xylose), [Pd{(R,R)-chxn}(α-d-Ribp1,2H−2O1,2)]·2.25H2O (Rib = ribose), [Pd{(R,R)-chxn}(α-l-Thrf1,3H−2O1,3)]·2H2O (Thr = threose) and [Pd{(R,R)-chxn}(α-d-Eryf1,3H−2O1,3)]·3H2O (Ery = erythrose).  相似文献

In view of the wide applicability and versatility of titanium based Lewis acids in selective organic synthesis including asymmetric synthesis, we have synthesized a family of mono and polyatomic titanium derivatives. The polymetallic complexes prepared are bridged by pyridimine, quinone and triazine based ligands.
