Most of a protein surface is potentially antigenic, consisting of numerous overlapping domains each complementary to antibody-combining sites. These domains may include peptide sequences that are demonstrably antigenic but only when antibodies from the appropriate host individuals and species are used. Methods for locating antigenic peptide sequences are described in which hydrophilic polyamide supports are used for peptide synthesis, then solid-phase radioimmunoassay with antisera and protein A. Most antigenic domains, however, comprise amino acid side chains contributed by two or more nearby polypeptide chains. Such domains can be identified by comparing the cross-reactivities of groups of very closely related proteins towards monoclonal antibodies raised to one of them. Such studies, using myoglobins, have identified a number of residues not previously shown to be antigenic and have provided a guide for the choice of synthetic peptides which are likely to carry several immunodominant side chains. One such peptide corresponding to residues (72–89) of beef myoglobin has been shown, using CD and antibodies to the parent protein, to have interesting conformational and antigenic properties. In this perspective we discuss that bacterial genomes have optimized during evolution to control a range of cytoplasm, from immediately after cell division to a maximum amount/volume present just prior to DNA replication and subsequent cell division. The genetic expansion of bacteria via evolution may be limited to a genome size:cytoplasm amount/volume ratios and energetics that have been selected for during 3.6-4 billion years of evolution on the Earth. The optimal genome size is one that is relatively constant, but also has some plasticity for evolutionary change (via gene transfer) and mutational events, and can control a range of cytoplasm during the cell cycle.  相似文献

ABSTRACT. 1. A laboratory technique for measuring the number of pollen grains consumed by thrips is described.
2. Thrips imaginis Bagnall and Thrips obscuratus (Crawford) (Thripidae) were studied particularly on pollen of the kiwifruit Actinidia deliciosa (A. Chevalier) Liang & Ferguson in New Zealand, and Echium plantagineum L. in Australia.
3. Mean daily feeding rates (in grains per thrips per day) ranged from 29 to 843, with an individual rate as high as 1626 for T.imaginis larvae II on E. plantagineum.
4. The time taken to feed on a single grain was proportional to grain volume, and decreased with temperature.
5. Daily feeding rates were significantly different between pollens, and were higher for smaller grains. The total volume of pollen contents consumed and the total time spent ingesting this volume per thrips per day may be constant with respect to pollen species.
6. Daily feeding rates were equivalent to 0.2–0.7% of the average total pollen production of a flower per thrips per day.
7. Extrapolation of the daily feeding rates suggests that pollen damage by thrips could sometimes be reducing crop yield or plant fitness.  相似文献

The monoclonal antibody 3D6 reacts with the T-cell receptor (Tcr) on the T-leukemic line HPBALL and with 2–13% of human peripheral blood T lymphocytes. In this study Vα and Vβ expression in a panel of T-cell populations and clones expressing the 3D6 epitope was determined by Southern and northern hybridization analysis. The results demonstrate that these 3D6-positive T cells, regardless of CD4/CD8 phenotype or function, express a gene of the Vβ5 family, also expressed by HPBALL. No correlation of the HPBALL Vα gene. The results demonstrate that 3D6 recognizes an epitope solely on the Tcrβ chain and that the use of this β chain, together with an appropriate Vα, can impart a diverse pattern of reactivity to a T cell.  相似文献

The development of educational material as part of major changes in the service of the diabetic clinic in Glostrup University Hospital turned out to be a very rewarding process for the multidisciplinary team. This material includes age-appropriate goals and guidelines for teaching and learning about diabetes, and advice on coping with diabetes in hand-outs designed for diabetic educators, parents/adults and teenagers. The paper briefly describes the developmental process, the concepts and designs of the material, and makes recommendations on how it can be applied. Part of the material was later translated into Polish and Russian in connection with educational projects in these countries, and the material as a whole was recently incorporated into the Novo Care website at www.d4pro.com - Childhood Diabetes. Growing Internet access by diabetes patients, their relatives and health care professionals creates almost overwhelming possibilities, but also brings up many new questions. The development of diabetes educational material of the more 'traditional' type is discussed with regard to content, ideas and design, and focuses on how these concepts and projects can be influenced and adapted across different countries by Internet communication.  相似文献

It is commonly supposed that the contribution of a bond to protein or nucleic acid stability is equal to the in situ stability of the bond itself. This is not true for the noncovalent bonds that stabilize molecular folding. In general, a bonding interaction contributes a free energy increment to protein or nucleic acid stability that is larger, an enthalpy increment that is smaller, and entropy and heat capacity increments that are more positive than the corresponding bond parameter.  相似文献

1. This paper introduces a new approach for setting streamflow-based river ecosystem management targets and this method is called the 'Range of Variability Approach' (RVA). The proposed approach derives from aquatic ecology theory concerning the critical role of hydrological variability, and associated characteristics of timing, frequency, duration, and rates of change, in sustaining aquatic ecosystems. The method is intended for application on rivers wherein the conservation of native aquatic biodiversity and protection of natural ecosystem functions are primary river management objectives.
2. The RVA uses as its starting point either measured or synthesized daily streamflow values from a period during which human perturbations to the hydrological regime were negligible. This streamflow record is then characterized using thirty-two different hydrological parameters, using methods defined in Richter et al . (1996). Using the RVA, a range of variation in each of the thirty-two parameters, e.g. the values at ± 1 standard deviation from the mean or the twenty-fifth to seventy-fifth percentile range, are selected as initial flow management targets.
3. The RVA targets are intended to guide the design of river management strategies (e.g. reservoir operations rules, catchment restoration) that will lead to attainment of these targets on an annual basis. The RVA will enable river managers to define and adopt readily interim management targets before conclusive, long-term ecosystem research results are available. The RVA targets and management strategies should be adaptively refined as suggested by research results and as needed to sustain native aquatic ecosystem biodiversity and integrity.  相似文献

We investigated the interacting surface between a short curarimimetic toxin and a muscular-type nicotinic acetylcholine receptor, looking for the ability of various biotinylated Naja nigricollis alpha-neurotoxin analogues to bind simultaneously the receptor and streptavidin. All these derivatives, modified at positions 10 (loop I), 27, 30, 33, 35 (loop II), 46, and 47 (loop III) or the N-terminal (erabutoxin numbering), still shared high affinity for the receptor, and in the absence of receptor they all bound soluble streptavidin. However, the proportion of the toxin-receptor complex that bound to streptavidin-coated beads, varied both with the location of the modification and with the length of the linker between biotin and the toxin. In the receptor-toxin complex, the concave side of loops II and III was not accessible to streptavidin, unlike the N terminus of the toxin and, to a certain extent, loop I. On the convex face, loop III was the most accessible, whereas the tip of loop II, especially Arg-30, seemed to be closer to the receptor. The present data demonstrate that short toxins neither penetrate deeply into a crevice as proposed earlier nor lie parallel to the receptor extracellular wall. These data also suggest that they may not lie strictly perpendicular to the cylindrical wall of the receptor. These results fit nicely with three-dimensional models of interaction between long neurotoxins and their receptors and support the idea that short and long curarimimetic toxins share a similar overall topology of interaction when bound to nicotinic receptors.  相似文献

Although cladistic analysis has been used to compare hypotheses of relationships among early hominids, the outcomes of different studies have depended entirely on the assumptions made by different investigators. Problems include the close genetic relationship of early hominid taxa, small fossil sample sizes, possible correlations among characters, and a lack of understanding about the evolutionary factors affecting characters. This study investigates the interaction of some of these problems affecting early hominid phylogenetics. Monte Carlo simulations of character state evolution in closely related taxa demonstrate that the sample sizes and close genetic relationships of early hominids do not permit cladistic analyses to obtain unequivocal results. Even with unrealistically good assumptions about the evolutionary dynamics affecting characters, the probability of the most parsimonious hypothesis being true is unacceptably small. In the face of these problems, even phylogenetic statements that are supported by a strong consensus of cladistic studies may nevertheless be in error, and such errors are likely to confound the placement of new specimens and taxa. Advancement in our knowledge of hominid phylogeny can depend only on a fuller understanding of the natural history and evolutionary dynamics of traits.  相似文献

Estimating rates of movement among populations is never simple, and where young animals cannot all be captured at their birth sites, traditional field methods potentially underestimate dispersal rates. Genetic assignment tests appear to hold promise for detecting 'precapture' dispersal, and recent evidence suggests that even on the scale of dispersal between populations, genetic parentage analyses can also be informative. Herein, we examine the performance of both types of analysis with data from a 17-year study of dispersal in banner-tailed kangaroo rats Dipodomys spectabilis. We compare estimates of precapture dispersal from (i) the commonly used parentage analysis program cervus (ii) a pedigree-reconstruction program, MasterBayes, that combines genetic with spatial and other nongenetic information and (iii) genetic assignment procedures implemented by the program geneclass2, with (iv) rates of dispersal observed through recapture of a subset of animals initially marked shortly after weaning. geneclass2 estimates a larger proportion of precapture dispersers than MasterBayes, but both approaches as well as those based on field data alone, suggest that approximately 10% of adults in local populations are immigrants and that interpopulation dispersal is slightly female-biassed. All genetic procedures detect precapture dispersal between populations, but dispersers identified by MasterBayes are particularly compatible with what is independently known about body mass at dispersal, dispersal distance and distance between parents. Parentage analyses have considerable potential to infer the value of this otherwise elusive demographic parameter when most candidate parents can be genotyped and when nongenetic information, especially the distance separating candidate mothers and fathers, can be incorporated into the procedure.  相似文献

Juxtacrine signalling is the process of cell communication in which ligand and receptors are both anchored in the cell membrane. We develop three mathematical models for this process, involving different mathematical representations of the dynamics of membrane-bound ligand and free and bound receptors, within an epithelial sheet. We consider the dynamics of this system following a localized disturbance, such as would be provided by a source of ligand or by the generation of a free edge via wounding. We study the ability of the juxtacrine mechanism to transmit a signal away from this disturbance, and show analytically that the spatial half-life of the signal can in fact be arbitrarily large. This result is quite general, since we use a generic reaction kinetic scheme; the key assumption is that ligand and receptor production are both upregulated by binding. Moreover, the result applies to all three of our model formulations. We conclude by discussing applications of the result to the particular case of the transforming growth factor alpha binding to epidermal growth factor receptor in epidermal wound healing.  相似文献

How do several characteristics adapt to gravity while mutually influencing each other? Our study addresses this issue by focusing on the terrestrial gastropod shell. The geometric relationship between the spire index (shell height/diameter) and outline (cylindricality) is theoretically estimated. When the shell grows isometrically, a high-spired shell becomes conical in shape and a low-spired shell becomes cylindrical in shape. A physical model shows that the lowest- and highest-spired shells are the most balanced. In addition, a cone shape is the most balanced for a low-spired shell, and a column shape is the most balanced for a high-spired shell. Spire index and cylindricality measured for freshwater gastropods follow the relationship estimated by the model, whereas those for terrestrial gastropods deviate from this relationship. This translates to a high shell being more cylindrical than a flat shell, except in the case of extremely high or low shells. This suggests that the shape of the most balanced shells (lowest and highest shell heights) is constrained by coiling geometry but that relatively unbalanced shells (intermediate shell heights) do not follow a coiling geometry, as a result of adaptation to enable the snail to carry its shell more effectively.  相似文献
