Genetic control of aluminium tolerance in rye (Secale cereale L.)   总被引:4,自引:0,他引:4  
 Aluminium (Al) tolerance in roots of two cultivars (“Ailés” and “JNK”) and two inbred lines (“Riodeva” and “Pool”) of rye was studied using intact roots immersed in a nutrient solution at a controlled pH and temperature. Both the cultivars and the inbred lines analysed showed high Al tolerance, this character being under multigenic control. The inbred line “Riodeva” was sensitive (non-telerant) at a concentration of 150 μM, whereas the “Ailes” cultivar showed the highest level of Al tolerance at this concentration. The segregation of aluminium-tolerance genes and several isozyme loci in different F1s, F2s and backcrosses between plants of “Ailés” and “Riodeva” were also studied. The segregation ratios obtained for aluminium tolerance in the F2s analysed were 3 : 1 and 15 : 1 (tolerant : non-tolerant) while in backcrosses they were 1 : 1 and 3 : 1. These results indicated that Al tolerance is controlled by, at least, two major dominant and independent loci in rye (Alt1 and Alt3). Linkage analyses carried out between Al-tolerance genes and several isozyme loci revealed that the Alt1 locus was linked to the aconitase-1 (Aco1), nicotinamide adenine dinucleotide dehydrogenase-2 (Ndh2), esterase-6 (Est6) and esterase-8 (Est8) loci, located on chromosome arm 6RL. The order obtained was Alt1-Aco1-Ndh2-Est6-Est8. The Alt3 locus was not linked to the Lap1, Aco1 and Ndh2 loci, located on chromosome arms, 6RS, 6RL and 6RL respectively. Four strains of the phytopathogenic bacteriumCorynebacterium fascians (Tilford)Dowson were selected from our isolates and deposited in the Czechoslovak National Collection of Type Cultures (CNCTC) of the Institute of Hygiene and Epidemiology in Prague. Two very virulent (Cor 83/82 “UPR” and Cor 82/81 “UP”) and one avirulent (Cor 81/80 “CP1b”) pelargonium strains, producing acid from rhamnose, were isolated from fasciations onPelargonium zonale W. One avirulent celery strain ofC. fascians (Cor 80/80 “CF4a”) was isolated from a root expiant ofApium graveolens L. growing on a nutrient mediumin vitro. Morphological, cultural, physiological and biochemical characteristics of these selected Czechoslovak isolates were compared with the American patented strain cotype ATCC 12974.  相似文献

A method was designed to produce rapidly (10–14 days) and directly (without intermediate callus) whole plants of Digitaria sanguinalis with a high yield without subculture. These plants developed from new structures, designated “pseudo-embryogenic structures”, initiated only 1 week after the culture of tranverse thin cell layers (tTCLs), i.e., thin stem sections, on a Murashige and Skoog medium containing 3% sucrose and a combination of a low concentration of 2,4-dichlorophenoxyacetic acid (1 μm) and a high concentration of 6-benzylaminopurine (10 μm). The fresh weight of plants regenerated per tTCL on gelrite was 6 times higher than with agar and 30 times higher than with agarose. The changes of dominance rank among female Japanese monkeys of the Koshima group over a period of 29 years from 1957 were studied. The dominance rank order was relatively stable in the early population growing phase, while large scale-changes of dominance rank order occurred successively in the phase of population decrease brought about by the severe control of artificial feeding after 1972. Nevertheless, the rank order of several females of the highest status was stable. Furthermore, the reproductive success of these highest status females was high (Mori, 1979a;Watanabe et al., in prep.). Divergence of the dominance rank order fromKawamura's rules (Kawamura, 1958) was observed in the following respects: (1) Some females significantly elevated their rank depending on the leader males. (2) If mothers died when their daughters were still juveniles or nulliparous, the dominance rank of some of these offspring females was significantly lower than the mother's one. However 55% of daughters which lost their mothers at a young age inherited the mother's rank. (3) Dominance among sisters whose mother had died when at least one of the daughters was under 6 years old followed the rule of youngest ascendancy in 60% (Kawamura, 1958), and in 80% when both of the daughters were nulliparous at the mother's death. The mean rate of aggressive interactions for each female with subordinates to her was calculated by dividing the total aggressive interactions between the female in question and her subordinates by the number of subordinate females to the female in question. A female which showed a high rate of aggressive interactions with her subordinates was categorized as an “Attacker”, and a female showing a lower rate was categorized as a “Non-attacker”. Similarly, categories of “Attacked”, and “Non-attacked” were distinguished by using the rate of aggressive interactions with dominant females. Several females which were once categorized in one category in a year were repeatedly categorized in the same category over different years. The “Attacked” tended to be females of higher rank, and “Non-attackers” tended to be females of lower rank. “The second-higher-status females”, were “Attacked”, and their rank was unstable. In particular, females of lower rank within the lineage of the highest rank suffered this kind of severe status. Most of the daughters of these females showed a sharp drop of rank, and died when they were still at a young age, i.e. “the second-higher-status females” displayed low fitness. “Non-attackers” were significantly “Non-attacked”; i.e. they were females which showed a non-social attitude. Females which underwent a drop of rank tended to be “Non-attackers”. The most important factor which determined the females' rank was the memory of their dominance relations under the influence of their mother [dependent rank (Kawai, 1958)] in their early life during development. This finding corresponds well with the results in baboons obtained byWalter (1980); the target females of aggressive interactions by adolescent females were determined by the rank of the mothers when these adolescent females were born.  相似文献

The level of the three main polyamines putrescine, spermidine, and spermine and the biosynthetic enzyme arginine decarboxylase (ADC) decreased in Helianthus annuus L. seedlings subjected to increasing (50, 100, and 150 mm) NaCl concentrations. The pattern of polyamines in control plants increased during the initial 72 h and then reached a plateau. The putrescine level showed an increase of 370% after 72 h of development. The lower salt treatment slightly diminished the overall polyamine content. The highest NaCl concentration (150 mm) induced a strong putrescine diminution (from 381 to 78.9 nmol g−1 FW) at 72 h whereas a small decrease in ADC activity was detected. ODC was detected in neither control nor treated plantlets during the experimental period. The level of spermidine also decreased, but the magnitude of the decay was less pronounced than putrescine. The fact that ODC was not detected and ADC activity followed a pattern similar to that of putrescine led us to suppose that the variation in putrescine content could be attributed entirely to the decrease in ADC activity. α-Difluoromethylarginine and α-difluoromethylornithine (ADC and ODC inhibitor, respectively) did not inhibit but delayed the onset of germination of sunflower seeds, and α-difluoromethylornithine increased the content of spermidine and spermine. Summary In the sera of humans and various animals there are two different non-specific inhibitors (α-and β-inhibitors) which may be specifically abolished by two substances (α-and β-“enzymes”) both present in filtrates ofV. cholerae. This neutralization is necessary for the rapid classifying of influenza virus strains with the aid of the haemagglutination inhibition test. The egg line strains of the A- and A′-groups are only sensitive to β-inhibitor, while the mouse line strains of the same groups are only sensitive to the α-inhibitor. This does not apply to strains of the B group, where mouse as well as egg lines are sensitive to both inhibitors. With the aid of isolated β-inhibitor (easily to be prepared from cattle serum), it is possible to decide in a quick manner whether or not a strain from the A- or A′-group has previously been adapted to the mouse. The electrophoretic patterns of 15 protein systems codified for 20 genetic loci were investigated using horizontal electrophoresis. A total of 150 blood samples, from five species of the genusCallithrix were analyzed. Polymorphic variation was observed in 10 out 20 loci analyzed. The genotypic distributions are in Hardy-Weinberg equilibrium. The average heterozygosity (H) varied from 1% to 5%, similar to those observed for other Neotropical primates. The genetic distance coefficients revealed a phylogenetic separation of these species into two groups: (1) “argentata” (C. humeralifer andC. emiliae); (2) “jacchus” (C. jacchus, C. penicillata, andC. geoffroyi). This arrangement is according to the taxonomic arrangement proposed byHershkovitz (1977),de Vivo (1988), andMittermeier et al. (1988). The results in each group are compatible with the subspecies values recorded for the Platyrrhini. These values showed that:C. humeralifer andC. emiliae are subspecies ofC. argentata;C. jacchus, C. penicillata, andC. geoffroyi are subspecies ofC. jacchus. These results also suggest thatC. j. geoffroyi is the “jacchus” group taxon, most similar genetically to the “argentata” group.  相似文献

In response to a critique byFerguson (1989),Leonard (1991) reiterates most of his original arguments for supporting “Australopithecus afarensisJohanson, White, andCoppens, 1978 as a single species. He disregards the principle of morphological equivalence by comparing the dental metrics and morphology of a hominid with those of species of the Pongidae, which do not correspond with the degree of variation in hominids, instead of with those of species of the Hominidae. He fails to refute clear evidence that the range of variation of dental metrics and morphology in “A. afarensis” exceeds that seen in species of the Hominidae. On the basis of extreme variation, “A. afarensis” is, therefore, interpreted as representing a composite species.  相似文献

Summary Hassell andVarley described a negative relationship between the Nicholsonianarea of discovery and parasitoid density that permits coexistence of two or more parasitoid species and yields stability. They ascribed the relationship to “mutual interference” among searching parasitoid adults, leading to a “decreased efficiency”. This matter has recently been studied in more detail byHassell and coworkers. Negative relationships can occur for several reasons. In the present paper it is shown that the negative relation occurs under certain assumptions about the way parasitoid eggs are distributed among hosts. Several “contagious” (or “clustered”) distributions were studied. Those assuming variation in host susceptibility or accessibility (negative binomial and added zeros distributions) yielded negative relations. Those assuming variation in parasitoid aggressiveness and fecundity (Neyman Type A distributions) yielded variable results depending on how the distributions of parasitoids per host and of eggs per parasitoid were combined. It was found that, when analyzing actual data, sampling fluctuations can introduce strong negative relations either with or without “contagion”. In all instances, the relation betweenarea of discovery and parasitoid density is affected by host density and is, in general, not linear. The foregoing findings raise serious questions about thearea of discovery concept. An alternative approach is suggested. The feeding process of Japanese monkeys on soy beans which were scattered over a sandy beach on Koshima islet was studied. Younger monkeys were able to pick up more beans when 8 kg of beans were divided and given two times (“two times feeding”) than when the whole amount (8 kg) of beans was given at one time (“one time feeding”). The effect of saturation of the food intake capacity in younger monkeys at the first feeding in “two times feeding” did not appear at the second feeding one hour later. The minutely intake of soy beans (feeding speed) for each age class was analyzed. The decline of feeding speed in adult females after the peak in “one time feeding” was not related to the decline in density of beans on the ground, and this decline was caused by saturation of their food intake capacity. Adult females were divided into four classes according to their dominance rank order: high, lower-high, higher-low, and low classes. The total amount of intake in “one time feeding” was far larger in the high class than in any of the other classes. The total amount of feeding in the first feeding of “two times feeding” increased in accordance with rise in the dominance rank class, and there was no relation to rank and total feeding amount in the second feeding of “two times feeding.” Differences existed in the process of feeding between the rank classes. The feeding speed of the low class was as high as that of the high class on the curve of minutely intake, while the low class stopped feeding much earlier than the high class. The lower-high class displayed a low feeding speed, and stopped feeding the latest. The order of the duration to stay and to feed in the feeding area was lower-high > high > higher-low > low, and this order did not change under the three different feeding conditions, “one time feeding,” and the first and second feedings of “two times feeding.” Adolescent females tended to stay the longest duration in the feeding area among all age classes. Both the lower-high class females and adolescent females had an unstable social status in the Koshima group, and their social status affected their feeding behaviors. The feeding behaviors were similar in attitude depending on social status, and are considered to be maintained for a fairly long time. The feeding strategy of the lower-high class, in staying a longer duration in the artificial feeding area, and departing later, may be effective under the artificial feeding conditions, but it may be a bad strategy in a natural habitat where the food is not so clumped as in artificial feeding, and where choice of other food patches is possible. The above results agree well with previous reports for the Koshima group, indicating that the rank of the lower-high class females was unstable (Mori et al., 1989), and that their reproductive success was low (Watanabe et al., 1992).  相似文献

Two new phototrophic consortia, “Chlorochromatium lunatum” and “Pelochromatium selenoides”, were observed and collected in the hypolimnion of several dimictic lakes in Wisconsin and Michigan (USA). The two consortia had the same morphology but different pigment composition. The cells of the photosynthetic components of the consortia were half-moon-shaped. This morphology was used to differentiate them from the previously described motile phototrophic consortia “Chlorochromatium aggregatum” and “Pelochromatium roseum”. These phototrophic cells did not resemble any described unicellular green sulfur bacteria. The predominant pigments detected were bacteriochlorophyll d and chlorobactene for the green-colored “Clc. lunatum”, and bacteriochlorophyll e and isorenieratene for the brown-colored “Plc. selenoides”. Their pigment compositions and the presence of chlorosomes attached to the inner face of the cytoplasmic membrane in both kinds of photosynthetic cells confirmed this new half-moon-shaped morphotype as a green sulfur bacterium. Both consortia were found thriving in lakes with low concentrations of sulfide (< 60 μM), below the layers of “Clc. aggregatum” and “Plc. roseum”. The green consortia were observed in lakes where the oxic-anoxic interface was located at shallow depths (2–7 m), while the brown consortia were found at greater depths (8–16 m). The possible role of polyamines (PAs) in the regulation of silymarin (Sm) production in milk thistle [Silybum marianum (L.) Gaernt] cell suspension cultures was studied in a young cell culture line (H2 line) and in a synchronized cell line (>3 years; H1 line). The effect of two exogenous PAs, putrescine (Put) and spermidine (Spd), and a number of metabolic inhibitors (L-canavanine, DL-α-difluoromethylornithine, methylglyoxal-bis-guanylhydrazone, cyclohexylamine) on the production of Sm during the growth cycle were analyzed. The results suggest that PAs are not directly involved in the Sm synthesis pathway. In our cell culture system, Sm production and PA contents were determined by the age of the suspension culture cells: with increasing age, the suspension culture cells showed a decreasing capacity to reach the stationary phase during prolonged subculture that was associated with a decreased production of Sm, a steady increase in PA content, and a constant Put/Spd ratio. The synchronization of dividing cells from the S. marianum H1 line did not modify this behaviour. In young cell suspensions, maximum Sm production occurred in the stationary phase, concurrent with the cellular PA contents reaching their minimum value. At the start of the stationary phase, the high percentage of cells in the growth phases (G0/G1) and a transient increase in the Put/Spd ratio were accompanied by maximum Sm production and a blockade of cell division.  相似文献

Summary The investigation made byWilkens andTasman (1959) into the immunization of tuberculosis patients with Tetanus Phosphate Toxoid has been extended to a further group of patients and control subjects. The work described here covered a total of 29 controls and 54 tuberculosis patients; the latter were from two different clinics (SAZU and “Zonnegloren”). On the whole, the results previously reported byWilkens andTasman were comfirmed in that tuberculosis patients were found to respond to primary immunization with Tetanus-P.T. (2 injections of 0.5 ml at a one-month interval) by a less marked and slower antitoxin production than the normal controls. The patients treated at SAZU showed a more deviant behaviour in this respect, than did the “Zonnegloren” patients. A number of specifically designed animal experiments showed that administration of PAS had an unfavourable effect on development of immunity after the first P.T. injection. This fact might possibly explain the behaviour seen in the tuberculosis patients. It might also explain the marked difference in immunological reactivity between the SAZU and the “Zonnegloren” patients, because the former chiefly received this agent by intravenous drip, while the latter were given oral medication. It is possible that intravenous administration of PAS produces higher blood concentrations than oral administration.  相似文献

Three cassava clones (SOM-1, “05”, and “50”) were cultured in vitro on MS medium plus sucrose (30 g L−1) and myo-inositol (100 mg L−1) without plant growth regulators and with additions of 0 (control), 0.5, 1, 1.5, 2, 2.5, and 3 g L−1 NaCl to test their salt tolerance. The same cassava clones were cultivated in greenhouse conditions on a sandy soil substratum and irrigated with 20% strength Hoagland solution, and additions of 0, 4, and 8 g L−1 of NaCl. Salinity negatively affected the survival, development, leaf water content, and mineral composition (mainly by accumulation of Cl and Na) of both in vitro and ex vitro plants, but with different intensity in each clone. In both conditions of culture (in vitro and ex vitro) clone SOM-1, from a desert arid saline zone of Somalia, was the most tolerant and clone “05”, from a rainy region of Ivory Coast, the most sensitive. Clone “50” tolerance to in vitro salt treatments, although lower, was not significantly different from that of SOM-1 but the ex vitro response was similar to “05”. In general, there was a correlation between in vitro and ex vitro behavior of the cassava plant regarding salt tolerance, which would allow the in vitro culture method to be used for selection of salt-tolerant plants of this crop.  相似文献

Two successive hydroponic experiments were carried out to identify barley varieties tolerant to Cd toxicity via examining Soil–Plant Analyses Development (SPAD) value, plant height, leaves and tillers per plant, root number and volume, and biomass accumulation. The results showed that SPAD values (chlorophyll meter readings), plant height, leaf number, root number and volume, and biomass accumulation of shoot/root were significantly reduced in the plants grown in 20 μM Cd nutrient solution compared with control, and the uptake and translocation of Zn, Mn, and Cu was also strictly hindered. Furthermore, there was a highly significant difference in the reduction in these growth parameters among varieties, and varieties “Weisuobuzhi” and “Jipi 1” showed the least reduction both in the two experiments, suggesting their high tolerance to Cd toxicity, while “Dong 17” and “Suyinmai 2” with the greatest reduction and the toxicity symptoms appeared rapidly and severely, denoting as Cd-sensitive varieties. Significant variety difference in Cd concentration was also found, with Weisuobuzhi containing the highest and Jipi 1 the lowest Cd concentration in shoots.  相似文献

Ten cell lines established from juvenile green sea turtles were tested and evaluated for their cytotoxic responses to four heavy metals: cadmium (Cd), chromium (Cr), zinc (Zn), and copper (Cu). Following a 24-h exposure to these metal salts at selected concentrations, test cells were comparatively characterized by morphology, viability, and proliferation. Experimental results indicated that all these metal salts were cytotoxic to these turtle cell lines at varied concentrations. Calculated 10% and 50% inhibitory concentration (IC10 and IC50) values revealed that the cytotoxicities of Cd and Cr were significantly more potent than the other two metal salts (p < 0.01). Comparative analysis of IC10 values in these ten cell lines showed that turtle lung cells (GT-LG) are the most sensitive cell line to Cd, Cr, Zn, and Cu. Among these turtle cell lines, turtle liver cells (GT-LV) are more tolerant than other cells to Cd, Cr, and Zn, while GT-EYE cells are more tolerant to Cu, as determined by IC50 values. Overall, GT-LG represents the most sensitive cells to heavy metal contamination and may be used for initial environmental monitoring, while the highly tolerant nature of GT-LV and GT-EYE cells to the tested heavy metals suggest their potential use as an emergency last-resort indicator of potential metal-related adverse effect on human health.  相似文献

Fossil species ofMelanopsis from a freshwater formation in the Jordan Valley (near Al-Qarn) were investigated and the deposits containing these species are formally described as Al-Qarn Formation. Four species were found:Melanopsis buccinoidea Olivier,M. tchernovi Heller & Sivan,M. costata Olivier andM. aaronsohni Blanckenhorn.Melanopsis costata was represented by two groups, “stepped” and “non-stepped”, the latter differing in its lower figurativity index. Intermediates were found betweenM. buccinoidea andM. tchernovi; they may be hybrids. TheMelanopsis assemblage bridges the faunal gap, in the Jordan Valley, between the 2 Ma lake of ‘Erq el Ahmar on the one hand and the 0.8–1.7 Ma lake of ‘Ubeidiya on the other. This suggests an early Pleistocene age of about 1.8 million years for the Al-Qarn Formation.  相似文献

Mobile genetic elements constitute a substantial part of eukaryotic genome and play an important role in its organization and functioning. Co-evolution of retrotransposons and their hosts resulted in the establishment of control systems employing mechanisms of RNA interference that seem to be impossible to evade. However, “active” copies of endogenous retrovirus gypsy escape cellular control in some cases, while its evolutionary elder “inactive” variants do not. To clarify the evolutionary relationship between “active” and “inactive” gypsy we combined two approaches: the analysis of gypsy sequences, isolated from G32 Drosophila melanogaster strain and from different Drosophila species of the melanogaster subgroup, as well as the study of databases, available on the Internet. No signs of “intermediate” (between “active” and “inactive”) gypsy form were found in GenBank, and four full-size G32 gypsy copies demonstrated a convergence that presumably involves gene conversion. No “active” gypsy were revealed among PCR generated gypsy ORF3 sequences from the various Drosophila species indicating that “active” gypsy appeared in some population of D. melanogaster and then started to spread out. Analysis of sequences flanking gypsy variants in G32 revealed their predominantly heterochromatic location. Discrepancy between the structure of actual gypsy sites in G32 and corresponding sequences in database might indicate significant inter-strain heterochromatin diversity. [(4-methoxy-4(3-β-d-galactose-4-chlorophenyl)]spiro[1,2-dioxetane-3-1,3-tricyclo[7.3.1.02,7]tridec-2,7-ene] (“sβ-Gal 102”) and sodium [4-methoxy-4(3-β-d-glucuronic acid-4-chlorophenyl)]spiro[1,2-dioxetane-3-1,3-tricyclo[7.3.1.02,7]tridec-2,7-ene] (“sβ-Glucor 102”) are carbohydrate-containing 1,2-dioxetane compounds that produce chemiluminescence upon enzymatic hydrolysis by β-d-galactosidase, and β-d-glucuronidase, respectively. In this study, we have characterized and validated a sensitive detection principle for viable Escherichia coli based on enzymatic cleavage of sβ-Gal 102 and sβ-Glucor 102 (“ColiLight II”). The proposed chemiluminescent assay was optimized with respect to analytical requirements including incubation time, temperature, pH, enzyme induction, and cell permeabilization. The sensitivity and specificity rates of the assay were tested on ten different bacterial genera. The assay was found to be representative based on low coefficients of variations for both accuracy and precision. The analysis time was less than 1 h and the analytical detection limit was 102 to 103 E. coli cells. In combination with membrane filtration and a brief resuscitation step of 4 h, the proposed assay was capable of detecting low concentrations of stressed E. coli in potable water (<30 CFU 100 ml−1). The proposed chemiluminescent enzyme assay may be used for assessing the metabolic activity of E. coli in oligotrophic environments and for early warning detection of low concentrations of E. coli in water for human consumption.  相似文献
