Lipid metabolism was investigated during the reproductive cycle of Labidura riparia (Pallas). The lipid classes and their constitutive fatty acids present in hemolymph and ovaries were measured using thin‐layer chromatography and gas‐liquid chromatography. In the hemolymph, total lipids increase steadily from the previtellogenic period to vitellogenic arrest. These lipids are predominantly diacylglycerols and phospholipids. In the ovaries, total lipids increase during vitellogenesis then decrease during the vitellogenesis arrest period. The major lipids are triacylglycerols, followed by phospholipids. In both hemolymph and ovaries, all lipid classes contained variable proportions of seven main fatty acids: the saturated fatty acids myristic acid (14:0), palmetic acid (16:0), and stearic acid (18:0); the monounsaturated fatty acids palmitoleic acid (16:1) and oleic acid (18:1); and the polyunsaturated fatty acids linoleic acid (18:2) and linolenic acid (18:3). Unsaturated fatty acids predominate throughout the reproductive cycle. The percentage compositions of total and triacylglycerol fatty acids do not change markedly during the reproductive cycle in hemolymph nor in ovaries, with 18:2, 18:1 and 16:0 fatty acids being the major components. However, for diacylglycerols and phospholipids, the proportions of fatty acids vary systematically. For phospholipids during the vitellogenesis period, 18:2 increases considerably whereas other fatty acids decrease; for diacylglycerols, these fatty acids vary in the reverse way.  相似文献

During the reproductive cycle of the female Labidura riparia, cytological observations show cyclical modifications of lipid droplets in the periovarian adipocyte. Fat body lipids and their constitutive fatty acids are analyzed. The lipids are predominantly triacylglycerols, which increase after adult ecdysis during vitellogenic and non-vitellogenic periods. Small amounts of diacylglycerols and phospholipids are found. Diacylglycerols increase during vitellogenesis and decrease during the non-vitellogenic period. Cytological modifications of lipid droplets are probably related to diacylglycerol fluctuations. Gas-liquid chromatography of fatty acid methyl esters shows oleic acid to be the predominant fatty acid in total lipids and triacylglycerols; unsaturated acids are approximately twice as abundant as saturated acids all along the reproductive cycle. Fatty acid composition of diacylglycerols and phospholipids differs from triacylglycerols and total lipids composition. Palmitic, stearic, oleic and linoleic acids represent the major fatty acids; their relative amounts vary during the different periods of the reproductive cycle. The moult induces important variations in the concentrations and fatty acid composition of lipid classes during the moulting cycle of the male crab Pachygrapsus marmoratus. Phospholipids are the predominant lipid class. The high levels in total saturated fatty acids but also in oleic and palmitic acids result of the semi-terrestrial ethology of this grapsidae. The hepatopancreas presents the most notable variations in the lipid concentration during the moulting cycle. Transport of hepatopancreatic lipids through the hemolymph to the periphery occurs during the end of premolt, just before the new tissues constitution.  相似文献

Changes in lipid composition are observed in the ovary, the hepatopancreas, the haemolymph and the muscle of the female Penaeus indicus Milne Edwards, during the increase of the gonad-somatic index (GSI). Vitellogenesis is characterized by a large accumulation of lipids in the ovary, which seems to be practically linear when expressed as a function of GSI, if there is no interference from moulting. Phospholipids and triacylglycerols are the major lipid fractions in the ovary, but some classes of minor importance, like wax esters, hydrocarbons, and glyceryl ethers, are also incorporated. These last components may play a rôle in the maturation process.The lipid reserves of the hepatopancreas contribute partially to the formation of vitellin, but direct input of lipids from feeding seems to be preponderant. An intraovarian synthesis of lipids may be possible, but probably restricted, considering the low concentrations of free fatty acids and partial acylglycerols found in the ovary. Phospholipids are the main components involved in the increase in lipid concentration of the haemolymph during vitellogenesis. This increase, and the potential rise in the turnover of circulating lipids, suggest a deep alteration in the lipid metabolism of the females during the breeding period.  相似文献

Seasonal changes in the fatty acid composition of neutral and polar lipids were measured in the ovary, liver, white muscle, and adipopancreatic tissue of northern pike. The role of environmental and physiological factors underlying these changes was evaluated. From late summer (August–September) to winter (January–March), the weight percentage of n-3 polyunsaturated fatty acids (especially 22:6n3) declined significantly in the neutral lipids of all somatic tissues examined. However, large quantities of n-3 polyunsaturated fatty acids accumulated in the recrude cing ovaries during fall and the weight percentage of n-3 polyunsaturated fatty acids in ovary polar lipids also increased significantly. Additionally, the n-3 polyunsaturated fatty acid content of somatic polar lipids increased significantly during fall due to increases in the total polar lipid content of the somatic tissues. This suggests that during fall n-3 polyunsaturated fatty acid are diverted away from somatic neutral lipids and thereby conserved for use in ovary construction and for incorporation into tissue polar lipids. The percentage of n-3 polyunsaturated fatty acid in ovary neutral lipids also declined during fall and early winter, perhaps as an adaptation to conserve these fatty acids for storage in oocyte polar lipids and later incorporation into cellular membranes of the developing embryo. Reductions in the n-3 polyunsaturated fatty acids content of somatic and ovarian neutral lipids during fall were compensated for specifically by increases in the percentage of monounsaturated fatty acids rather than saturated fatty acids. This suggests that the ratio of saturated to unsaturated fatty acids in pike neutral lipid, is regulated physiologically, and hence may influence the physiological functioning of these lipids. During fall and early winter the percentage of saturated fatty acids declined significantly in the polar lipids of all tissues examined. This change was consistent with the known effects of cold acclimation on the fatty acid composition of cellular membranes. As the ovaries were recrudescing from September to January, liver polar lipids exhibited significant decreases in the percentage of total polyunsaturated fatty acids and n-3 polyunsaturated fatty acids and increases in monounsaturated fatty acids, and acquired a fatty acid composition very similar to that of ovary polar lipids. Therefore, seasonal changes in the percentage of polyunsaturated and monounsaturated fatty acids in liver polar lipids probably reflect the liver's role in vitellogenesis rather than the effects of temperature on membrane fatty acid composition. During fruit development of oil palm (Elaeis guineensis) oil deposition in the mesocarp startedca12–13 weeks after flowering (WAF) and continued until the fruit ripened at 20 WAF. Over the next 1–2 weeks oil continued to be deposited but the fruit became loose and readily detached from the bunch. The lipids extracted at this stage contained over 50 % free fatty acids andca6%, polar lipids. The major fatty acids in the storage triacylglycerols were 16:0,18:1 and 18:2. The fatty acid composition of the neutral lipid classes and polar lipids during oil deposition were similar except that the latter also contained a high proportion of 18:3. Longer chain acids (20:3 and 22:0) were detected in certain lipid classes at 8 and 12 WAF.  相似文献

Chlorella ellipsoidea Gerneck (IAM C-27) was synchronously grown, and cells at an intermediate stage in the ripening phase of the cell cycle were hardened at 3°C for 48 hours. A nonpolar lipid which increased greatly during hardening was analyzed by gas-liquid chromatography. Palmitic, oleic, linoleic, and linolenic acids were the main components of the lipid. Electron micrographs revealed the appearance of lipid bodies in hardened cells. When formation of free fatty acids and lipid bodies was inhibited with cycloheximide, oligomycin, and 3-(3,4-dichlorophenyl)-1,1-dimethylurea, the development of a high level of hardiness was always inhibited. However, the converse results were not always realized. Cells hardened in the dark in the absence of glucose developed a measurable hardiness in spite of their failure to form free fatty acids. The appearance of lipid bodies was invariably accompanied by the formation of the fatty acids. In pulse-labeling with [14C]NaHCO3 for 4 minutes at zero time and at the 12th hour of hardening, initial incorporation rates of 14C into total lipids of whole cells and the cellular membrane fraction were significantly higher than that into free fatty acids. These results suggest that, although fatty acids are inserted into membrane lipids during hardening, the accumulation of free fatty acids and the appearance of lipid bodies per se are not involved in the development of frost hardiness.  相似文献

This experiment was conducted to investigate the variation in lipid composition during the ovarian maturation of the crab Eriocheir sinensis. The Chinese mitten-handed crab broodstock was divided into six different maturation periods according to the size and color of ovary. Ovary, hepatopancreas, muscle, and hemolymph of broodstock in different maturation periods were analyzed for total lipid and fatty acids using gas chromatography, and lipid classes by thin-layer chromatography. The ovarian lipid concentration (expressed as percent wet ovarian weight) increased steadily from stage II (5.4%) to stage IV (19.1%), and decreased to the lowest levels after spawning (stage V, 6.6%). The hepatopancreatic lipid concentration (expressed as percent wet hepatopancreatic weight) increased with maturity of the ovaries, reached a maximum at stage III(2) (29.9%), and decreased during the subsequent period to spawning (16.7%). The muscular and hemolymph lipid concentration did not change markedly during the ovarian development. These results suggest the possible movement of hepatopancreatic lipids to the ovaries during the ovarian maturation. Both triacylglycerol and phosphatidylcholine were responsible for the increase in ovarian lipid concentration during sexual maturation. The fatty acids of total lipid, triacylglycerol, and phosphatidylcholine of the ovaries did not vary systematically during the ovarian maturation, but the ratio between n-3PUFA (polyunsaturated fatty acid) and n-6PUFA did change regularly with the ovarian lipid. These suggest that enough PUFA, especially n-3PUFA, should be supplied to the crab during ovarian maturation.  相似文献

By a combination of thin-layer chromatography and gas liquid chromatography, a complete study of the development of the different lipid classes and of their fatty acids, during the development of the fruit of Hedera helix L., the English Ivy, has been achieved. In any part of the fruit observed, at any particular stage, the phospholipids and the neutral lipids are the most abundant lipid classes. They accumulate during the entire process of maturation, whereas significant changes occur in their relative proportions, phospholipids being largely dominant until fruit blackening. The accumulation of fatty acids during maturation is characterized by large amounts of C18:1 in the neutral lipids, especially in the seed, where petroselinic acid (C18:1Δ6) reaches 86% of the total fatty acids. To a smaller extent, the phospholipids also accumulate and thus have the character of reserve molecules. However, their composition remains more stable, which relates them to the “structural lipids” such as galactolipids that maintain their characteristic fatty acid composition, despite the radical changes occurring in the fatty acid metabolism during fruit ripening.  相似文献

The composition of lipids, PLs and FAs in the treegrowing lichenXanthoria parientina, collected during the period from Marchto May, was studied. The major polar lipids found, includingphospholipids, were DGTS, PC, PE, and PG. Polar and PL contentswere also identified, and certain trends in the changing proportionsof PC (increasing from 17.8 to 50.1%) and DGTS (decreasing from27.1 to 12.6%) were determined. The fatty acid composition wasexamined using capillary GC-MS in the neutral, glyco- and PLfractions. Hydroxy acids were detected only in the glycolipidfraction; their seasonal dynamics were also studied. The seasonalchanges occurring in lipid composition due to the temperaturefactor were identified. It was found that X. parientina hada characteristic temperature-based lipid pattern, increasingin its neutral lipid content from 40.6% in March to 52.7% inMay but decreasing in glycolipid from 39.0% in March to 27.0%in May. The physical state of the membrane lipids, as determined by fatty acid composition and environmental temperature, has a marked effect on both the temperature range within which Acholeplasma laidlawii B cells can grow and on growth rates within the permissible temperature ranges. The minimum growth temperature of 8 °C is not defined by the fatty acid composition of the membrane lipids when cells are enriched in fatty acids giving rise to gel to liquid-crystalline membrane lipid phase transitions occurring below this temperature. The elevated minimum growth temperatures of cells enriched in fatty acids giving rise to lipid phase transitions occurring at higher temperatures, however, are clearly defined by the fatty acid composition of the membrane lipids. The optimum and maximum growth temperatures are also influenced indirectly by the physical state of the membrane lipids, being significantly reduced for cells supplemented with lower melting, unsaturated fatty acids. The temperature coefficient of growth at temperatures near or above the midpoint of the lipid phase transition is 16 to 18 kcalmol, but this value increases abruptly to 40 to 45 kcalmol at temperatures below the phase transition midpoint. Both the absolute rates and temperature coefficients of cell growth are similar for cells whose membrane lipids exist entirely or predominantly in the liquid-crystalline state, but absolute growth rates decline rapidly and temperature coefficients increase at temperatures where more than half of the membrane lipids become solidified. Cell growth ceases when the conversion of the membrane lipid to the gel state approaches completion, but growth and replication can continue at temperatures where less than one tenth of the total lipid remains in the fluid state. An appreciable heterogeneity in the physical state of the membrane lipids can apparently be tolerated by this organism without a detectable loss of membrane function.  相似文献

Triglycerides and phospholipids dominate in Baltic herring flesh lipids throughout the plankton growth season though at the same time plankton species and their lipids vary considerably. Wax esters and fatty alcohols do not exist in significant amounts in Baltic herring flesh or plankton. Saturated fatty acids exist in equal proportions in herring and plankton lipids whereas monoenoic acids are more dominant in herring and polyunsaturated acids in plankton lipids, respectively. The component alkyl chains are similar in both fish and plankton fatty acids, thus suggesting the planktonic origin. During the period of abundant availability of plankton the changes in the fatty acids of the herring depot fat are related to the changes in the plankton lipid composition.  相似文献

The spring diatom bloom in Loch Ewe (N.W. Scotland) during 1980 has already been the subject of a detailed study. As an extension of this work the component fatty acids and sterols of the diatom lipids have been analysed throughout the bloom period. The results confirm that detailed changes occur in the lipid composition of diatoms during their normal growth. In addition, the data are discussed in relation to published analyses of diatom lipids and suggestions made as to reasons for the considerable variation in reported fatty acid composition.  相似文献

Microbial fatty acids are an attractive source of precursors for a variety of renewable commodity chemicals such as alkanes, alcohols, and biofuels. Rerouting lipid biosynthesis into free fatty acid production can be toxic, however, due to alterations of membrane lipid composition. Here we find that membrane lipid composition can be altered by the direct incorporation of medium-chain fatty acids into lipids via the Aas pathway in cells expressing the medium-chain thioesterase from Umbellularia californica (BTE). We find that deletion of the aas gene and sequestering exported fatty acids reduces medium-chain fatty acid toxicity, partially restores normal lipid composition, and improves medium-chain fatty acid yields.  相似文献

When heterotrophic cell cultures of red goosefoot (Chenopodium rubrum) turned photoautotrophic, their contents of various glycolipids and phospholipids increased. The total lipids and the individual lipid classes, especially monogalactosyldiacylglycerols, became richer in linolenic and poorer in linoleic acids. When photoautotrophic cell cultures were rendered heterotrophic again a reversal of changes occurred; both the composition of lipids and the patterns of their constituent fatty acids became similar to those of the starting heterotrophic cultures.The results indicate that the biosynthesis of linolenic acid in photoautotrophic cell cultures involves mainly desaturation of linoleic acid and that chain extension of hexadecatrienoic acid is possibly another, though minor pathway. Monogalactosyldiacylglycerols are apparently the substrates preferred for linolenic acid biosynthesis, whereas various phospholipids are the substrates preferred for linoleic acid biosynthesis.During a growth period of 6 weeks, the levels of polyunsaturated fatty acids in the lipids from both heterotrophic and photoautotrophic cell cultures decrease with time, whereas the proportions of palmitic acid increase.  相似文献

Agonistic behaviour and lipid state were examined in tarantula Brachypelma albopilosa females during the foraging period. Modulation of the agonistic behaviour of females was not connected to their body size. Results show that the agonistic pattern of females differed significantly from the predation pattern at the behavioural and lipid levels. Aggressive-foraging females had low predation behaviour. Quantitative lipid changes were observed in relation to agonistic behaviour and predation. The total lipid index was studied by colorimetric methods, and lipid compounds were analyzed by gas chromatography-mass spectrometry in cuticle and hemolymph of females. The lipid components were free fatty acids, methyl esters, cholesterol, and long-chain aliphatic hydrocarbons. Methyl esters were much more abundant in cuticular lipids; unsaturated free fatty acids (linoleic and oleic acids) and methyl esters (methyl linoleate and methyl stearate) predominated in the hemolymph. Spider aggression was positively correlated with lipid concentration (cholesterol, fatty acids, methyl esters and hydrocarbons) in the hemolymph and the levels of cuticular fatty acids. Lipid levels are hypothesized to have evolved as a regulatory factor of predation and agonistic behaviours in tarantula females.  相似文献

Changes in the lipid and fatty acyl compositions of the marine microalga Nannochloropsis oculata Droop were examined during a batch culture growth cycle. During the early phase of batch culture the cellular proportion of triacylglycerols (TAG) increased. This was in addition to the increases in TAG observed in many microalgal species in the stationary-phase. Concomitant increases in the relative proportions of both saturated and monounsaturated fatty acids and decreases in the proportion of polyunsaturated fatty acids in total lipid were also associated with this phase. The separated individual lipid classes were found to have characteristic fatty acyl compositions. The relative proportion of lipid per cell, the relative proportions of the individual lipid classes and the fatty acyl compositions of the individual classes were all subject to variability during the growth cycle. The changing total lipid fatty acyl composition of N. oculata was found to be determined by the proportion of the total lipid present as TAG. The data suggest that the changes observed in the fatty acyl composition of N. oculata are a result of the partitioning of photosynthetically fixed carbon between polar and neutral lipid class biosynthesis and fatty acyl desaturation and elongation pathways. The effect of such a partitioning of carbon is discussed in relation to the effects of environmental variables and growth phase upon the balance of lipid class and polyunsaturated fatty acids (PUFA) synthesis in marine microalgae.  相似文献

The fatty acid distribution of Aerobacter aerogenes was studied by comparing the fatty acid composition of the lipoidal component of the endotoxin (lipid A) with the fatty acids of the readily extractable native lipids and total cellular fatty acids. The results for total cellular fatty acids and readily extractable native lipids were generally similar, but both quantitative and qualitative differences exist. In addition, profound differences between these two fractions and lipid A were observed. These differences included fewer fatty acids and lower concentrations of unsaturated and cyclopropane fatty acids in the lipid A. Hydroxy fatty acids persisted in the lipid A. The significance of these differences with respect to mammalian toxicity of endotoxins is discussed.  相似文献

The fatty acid content and composition of the house cricket Acheta domesticus have been investigated in entire insects at different developmental stages and in selected organs of male and female adults. We have also determined the fatty acid composition of the various lipid classes within extracts of the organs of adult female insects. Fatty acids were analysed by capillary gas chromatography or mass spectrometry as their methyl esters (FAMEs) after direct transesterification of insect material or separated lipid classes.The major esterified fatty acids in all extracts were palmitate (C16:0), stearate (C18:0), oleate (C18:1) and linoleate (C18:2). Levels of esterified fatty acid varied considerably between organs but the fatty acid compositions showed only small variations. The levels of polyunsaturated fatty acids of the C18 series were considerably higher in phospholipid fractions than in other lipid classes. Triacylglycerols formed the major lipid class in ovaries, fat-body and newly-laid eggs, whereas diacylglycerols and phospholipid predominate in the haemolymph. Triacylglycerols, phospholipids, diacylglycerols and free fatty acids were all found in significant amounts in the gut tissue.  相似文献

Seven day old seedlings of Echinochloa crus-galli var. oryzicola (Vasing) had a higher total lipid content when germinated under N2 than in air, although ungerminated seeds contained more lipid than either seedling. The triacylglycerol pool was not depleted under anaerobiosis as it was in air and only air-grown seedlings showed a net increase in free fatty acids and polar lipids. Concentrations of most of the individual acids of the total fatty acid profile declined during germination in air and in the free acid and polar lipid fractions of these seedlings the relative proportion of polyunsaturated fatty acids increased. Compared to air-grown seedlings, ungerminated seeds and N2-grown seedlings had a similar qualitative and quantitative lipid composition. Our results show that mobilization of storage lipids was apparently severely inhibited under anoxia. The importance of lipid metabolism to the germination and growth of Echinochloa during anoxia is discussed in terms of maintaining membrane integrity and serving (indirectly) to reoxidize pyridine nucleotides.  相似文献
