The objective of this study was to determine the biphasic viscoelastic properties of human temporomandibular joint (TMJ) discs, correlate these properties with disc biochemical composition, and examine the relationship between these properties and disc dynamic behavior in confined compression. The equilibrium aggregate modulus (HA), hydraulic permeability (k), and dynamic modulus were examined between five disc regions. Biochemical assays were conducted to quantify the amount of water, collagen, and glycosaminoglycan (GAG) content in each region. The creep tests showed that the average equilibrium moduli of the intermediate, lateral, and medial regions were significantly higher than for the anterior and posterior regions (69.75±11.47 kPa compared to 22.0±5.15 kPa). Permeability showed the inverse trend with the largest values in the anterior and posterior regions (8.51±1.36×10?15 m4/Ns compared with 3.75±0.72×10?15 m4/Ns). Discs were 74.5% water by wet weight, 62% collagen, and 3.2% GAG by dry weight. Regional variations were only observed for water content which likely results in the regional variation in biphasic mechanical properties. The dynamic modulus of samples during confined compression is related to the aggregate modulus and hydraulic permeability of the tissue. The anterior and posterior regions displayed lower complex moduli over all frequencies (0.01–3 Hz) with average moduli of 171.8–609.3 kPa compared with 454.6–1613.0 kPa for the 3 central regions. The region of the TMJ disc with higher aggregate modulus and lower permeability had higher dynamic modulus. Our results suggested that fluid pressurization plays a significant role in the load support of the TMJ disc under dynamic loading conditions. 相似文献

The use of a total joint replacement surgery has been growing in a manner which is poorly understood. This growth is more likely to be limited by saturation of the available medical services than by a limit to the number of people who could benefit from the treatment. Present indications are that services will be saturated in 1974 or 1975, after which 4600 extra people a year will go on to the waiting lists if demand continues as at present. Stability can then be achieved by either raising the clinical criteria for surgical treatment or making about 300 extra orthopaedic beds available each year, buffered from the demands of acute trauma admissions. 相似文献

Copies of death certificates were provided by the Registrar General for all deaths attributed to asthma in persons aged 5 to 34 years which were registered in England and Wales in the last quarter of 1966 and the first quarter of 1967. Information was obtained from the relevant general practitioners about 177 of the 184 subjects, and necropsy data were obtained for 113 of the 124 cases in which a post-mortem examination was known to have been made. Ninety-eight per cent. of the subjects for whom evidence was obtained were known to have been suffering from asthma, and signs of severe asthma (overdistended lungs and small bronchi plugged with mucus) were found in 91% of necropsies (57% of all deaths). Evidence that death might have been due to any other pathological condition was rare. Death was sudden and unexpected in 81% of the subjects (137 out of 171), and 59% of all deaths were referred to coroners. In 39% of cases (67 out of 171) the practitioner had not regarded the patient as suffering from severe asthma in his terminal episode. Corticosteroids and sympathomimetic preparations were the only drugs to have been used by a large proportion of patients. Two-thirds of the patients had received corticosteroids before the terminal episode, but detailed information about their use provided no suggestion that excess use could have been responsible for any large proportion of the deaths. Eighty-four per cent. of the patients were known to have used pressurized aerosol bronchodilators, and several instances of their use in excess were described. Routine inquiries about their use in the hours immediately preceding death were not made, and further evidence is required before their effect can be assessed adequately. 相似文献

Retaining the first terms of asymptotic expansions and assuming zero gradient of the contact pressure at the contact edge in the perpendicular direction to the edge, Ateshian et al. (1994, J. Biomechanics 27, 1347-1360; 1992, Adv. Bioeng. ASME 22, 191-194) have presented an asymptotic axially symmetric and plane strain solutions to the contact problems of the two thin biphasic cartilage layers in the synovial joint (with the synovial fluid film in between already depleted), subjected to a sudden normal load. Both the immediate and an early time response to a step loading have been analysed. The present note shows that the contact width thus obtained immediately after the load application differs from the numerical values for a dry frictionless contact of incompressible single-phase elastic layers available in the literature. The difference increases with the decreasing contact width-to-layer thickness ratio. It is proposed to improve the above solutions by releasing zero pressure gradient condition at the contact edge and calculating the instantaneous contact widths from the equations proposed for the corresponding case by Matthewson (1981, J. Mech. Phys. Solids 29, 89-113) and Meijers (1967, Appl. Sci. Res. 18, 353-383) that approximate better the numerical values. The instantaneous rate of change in the contact width is then obtained for a step load varying in time. 相似文献

The recombinant ovalbumin produced in Escherichia coli was purified from the cytoplasmic fraction and analyzed for its chemical and conformational properties. The recombinant ovalbumin displayed almost exactly the same circular dichroism and intrinsic tryptophan fluorescence spectra as egg white ovalbumin. As in the egg white protein, four cysteine sulfhydryls and one cystine disulfide were contained in the recombinant protein, according to the results of amino acid analyses; the disulfide bond was found by a peptide mapping analysis to correspond to the native cystine, Cys73-Cys120. According to a gel electrophoresis analysis, the presence of the disulfide bond was accounted for by specific oxidation of the corresponding cysteine residues during purification of the cytoplasmic protein. Unlike the identity in the conformational and peptide structures, none of the post-translational modifications (N-terminal acetylation, phosphorylation, and glycosylation) that are known with egg white ovalbumin were detected in the recombinant protein. The recombinant ovalbumin was transformed into a thermostabilized form in a similar manner to the transformation of egg white protein into S-ovalbumin; alkaline treatment increased the temperature for thermostability by 8.7 degrees C. These data strongly suggest that the post-translational modifications of ovalbumin are not related to the formation mechanism for S-ovalbumin. 相似文献

A substantial explication of the edge effect has been attempted by use of capture-recapture data for a vole population (Microtus montebelli), gathered intwo plots of 100×100 m or less during 12 days, cheked twice daily, in August 1970; the sample was quite sufficient for the aim. The edge effect as guessed by increased catch per trap is usually suspected to ensue from range-settlers in the outside boundary strip of a plot and immigrants. But by a theoretical analysis I could attain a tentative conclusion that no increased catch per trap will occur unless any invasion takes place. Then it follows that, apart from the effect of invasion, the role of the adjoining outside settlers in the edge effect is essentially required to be studied in the light of knowledge on the truth of size and shift in home range. The variation in range behavior for 183 adult voles, captured 6 times or more, could be grouped into eight types, of which the range-conservative type possessed 52% of the sample and the group of the type was justly utilized for giving averages of range size. Besides, it was seen from the observed frequency of types that a considerable number of immigrants onto the census plot were induced perhaps being allured by trap baits, but the majority of them proved to be assigned to the voles that have their ranges inside the assessment line ofDice ; the rest referable to effective immigrants was only a few (7%). I could perceive no reason such as disproves the idea ofDice 's additional boundary strip. Viewed from maps of ingress shift of ranges, the effect of ingress must have been greater in the outer trap rows than in the inner within the plot, so that it might well be called edge effect in general; such effect, however, is seen gradually diminishing toward the center, and hence it is almost unlikely that one should find any clear-cut intra-plot assessment lines demarcating such an inner square as quite free from edge effects. Averages of observed range length and width (ORL and ORW), as reliable measures for the true range size, were determined from the above group of specimens; as a result, the remarkable concept of elliptic range shape was established by regarding ORL as long axis and ORW as short one, and, directly from these averages, the mean range sizes worked out at 0.04 for females and 0.09 for males in acreage which proved to be surprisingly well agreeable with those of isotope-revealed ranges for voles given byGodfrey (1954) andAmbrose (1969). The catchability for marked voles () was estimated by the maximum likelihood method by use ofJolly 's formulae (1965), but that for unmarked ones () was made by the regression census formula; as a result it was shown that the population was clearly of π>p type and that the trap-experience that voles underwent one month or more ago can make them retain as high catchability as π. 相似文献

We replaced the Escherichia coli lexA gene by a segment of DNA coding for resistance to spectinomycin and streptomycin. The use of this segment expands the range of selectable markers usable for allele replacement. A high incidence of unexpected metal sensitivity was found in patients with metal-to-metal (McKee) hip arthroplasties. Patients with metal-to-plastic (Charnley) prostheses had no greater incidence of metal sensitivity than a control group awaiting operation. IntroductionUse of disease-modifying anti-rheumatic drugs (DMARDs) in rheumatoid arthritis (RA) may prevent joint damage and potentially reduce joint replacement surgeries. We assessed the association between RA drug use and joint replacement in Quebec, Canada.MethodsA cohort of new-onset RA patients was identified from Quebec’s physician billing and hospitalization databases from 2002–2011. The outcome was defined using procedure codes submitted by orthopedic surgeons. Medication use was obtained from pharmacy databases. We used alternative Cox regression models with time-dependent variables measuring the cumulative effects of past use during different time windows (one model focussing on the first year after cohort entry) for methotrexate (MTX), and other DMARDs. Models were adjusted for baseline sociodemographics, co-morbidity and prior health service use, time-dependent cumulative use of other drugs (anti-tumor necrosis factor [anti-TNF] agents, other biologics, cyclooxygenase-2 inhibitors [COXIBs], nonselective nonsteroidal antiinflammatory drugs [NSAIDs], and systemic steroids), and markers of disease severity.ResultsDuring follow-up, 608 joint replacements occurred among 11,333 patients (median follow-up: 4.6 years). The best-fitting model relied on the cumulative early use (within the first year after cohort entry) of MTX and of other DMARDs, with an interaction between MTX and other DMARDs. In this model, greater exposure within the first year, to either MTX (adjusted hazard ratio, HR = 0.95 per 1 month, 95 % confidence interval, 95 % CI 0.93-0.97) or other DMARDs (HR = 0.97, 95 % CI 0.95-0.99) was associated with longer time to joint replacement.ConclusionsOur results suggest that longer exposure to either methotrexate (MTX) or other DMARDs within the first year after RA diagnosis is associated with longer time to joint replacement surgery.

The porcine pancrease lipase was immobilized by entrapment in the beads of K-carrageenan and cured by treatment with polyethyleneimine (PEI) in the phosphate buffer. The retention of hydrolytic activity of lipase and compressive strength of the beads were examined. The activity of free and immobilized lipase was assessed by using olive oil as the substrate. The immobilized enzyme exhibited a little shift towards acidic pH for its optimal activity and retained 50% of its activity after 5 cycles. When the enzyme concentration was kept constant and substrate concentration was varied the Km and Vmax were observed to be 0.18 × 10−2 and 0.10, and 0.10 × 10−2 and 0.09 respectively, for free and for entrapped enzymes. When the substrate concentration was kept constant and enzyme concentration was varied, the values of Km and Vmax were observed to be 0.19 × 10−7 and 0.41, and 0.18 × 10−7 and 0.41 for free and entrapped enzymes. Though this indicates that there is no conformational change during immobilization, it also shows that the reaction velocity depends on the concentration. Immobilized enzyme showed improved thermal and storage stability. Hydrolysis of olive oil in organic–aqueous two-phase system using fixed bed reactor was carried out and conditions were optimized. The enzyme in reactor retained 30% of its initial activity after 480 min (12 cycles). 相似文献

Mechanical properties of a polyacrylamide gel with reversible DNA crosslinks are presented. In this system, three DNA strands replace traditional chemical crosslinkers. In contrast to thermoset chemically crosslinked polyacrylamide, the new hydrogel is thermoreversible; crosslink dissociation without the addition of heat is also feasible by introducing a specific removal DNA strand. This hydrogel is characterized by a critical crosslink concentration at which gelation occurs. Below the critical point, a characteristic temperature exists at which a transition in viscosity is observed. Many species of marine algae have life cycles that involve multipleseparate, free-living phases that frequently differ in ploidylevels. These complex life cycles have received increasing scientificattention over the past few decades, due to their usefulnessfor both ecological and evolutionary studies. I present a synthesisof our current knowledge of the ecological functioning and evolutionaryimplications of the isomorphic, biphasic life cycles commonlyfound in many species of marine algae. There are both costsand benefits to life cycles with 2 morphologically similar butseparate, free-living phases that differ in ploidy levels (haploidsand diploids). Evolutionary theory predicts that the existenceof subtle yet important differences between the phases may bewhat allows these life cycles to be maintained. Different phasesof the same species can vary in abundance, in demographic parameterssuch as mortality and fecundity, in their physiology, and intheir resistance to herbivory. In order to determine whether there is a genetic component to hip or knee joint failure due to idiopathic osteoarthritis (OA),
we invited patients (probands) undergoing hip or knee arthroplasty for management of idiopathic OA to provide detailed family
histories regarding the prevalence of idiopathic OA requiring joint replacement in their siblings. We also invited their spouses
to provide detailed family histories about their siblings to serve as a control group. In the probands, we confirmed the diagnosis
of idiopathic OA using American College of Rheumatology criteria. The cohorts included the siblings of 635 probands undergoing
total hip replacement, the siblings of 486 probands undergoing total knee replacement, and the siblings of 787 spouses. We
compared the prevalence of arthroplasty for idiopathic OA among the siblings of the probands with that among the siblings
of the spouses, and we used logistic regression to identify independent risk factors for hip and knee arthroplasty in the
siblings. Familial aggregation for hip arthroplasty, but not for knee arthroplasty, was observed after controlling for age
and sex, suggesting a genetic contribution to end-stage hip OA but not to end-stage knee OA. We conclude that attempts to
identify genes that predispose to idiopathic OA resulting in joint failure are more likely to be successful in patients with

Actomyosin, myosin, and actin from different sources are adsorbed, apparently as a monolayer, by polystyrene particles teins for 1 mg of Lytron were about 10-7 liters mol-1, while heterogeneity indices (alpha) varied from 0.70 to 1.0 presumably as a function of spontaneous aggregation in the liquid phase. Adsorption was irreversible. Orientation of absorbed molecules permitted association of bound muscle actin with platelet or muscle myosin. The association constant of the former reaction was 2.78 times 10-6 liters mol-1. Enzymatic properties of adsorbed actomyosin, Mg2+ATPase activity was abolished, but association of myosin with bound actin, or association of actin with bound myosin was accompanied by restoration of Mg2+ATPase activity. Every subunit of F-actin strands, unless F-actin had been fully depolymerized to G-actin, could bind myosin and activate Mg2+ATPase activity. Immunogenic characteristics of muscle myosin were enhanced by Lytron adsorption. Elicited antibodies showed selective specificity for an antigenic determinant located near or at the actin combining site of muscle myosin. Antibodies did not react with actomyosin. Antibodies prevented association of actin with muscle myosin because they inhibited both superprecipitation and development of Mg2+ATPase activity. 相似文献
