Changes of metabolic activities during dormancy and breaking of dormancy in the cherry flower buds of early blooming (EB) cultivar ( Prunus avium L. cv. Coeur de Pigeon) and late blooming (LB) cultivar ( Prunus serrulata Lindl. cv. Kwanzan) were determined. The LB buds had higher polyamines, protein and 1-(malonylamino) cyclopropane-1-carboxylic acid (MACC) content than the EB buds. During the dormant state, the DNA, RNA, protein and polyamines in the EB buds were low but increased slowly and steadily, whereas those in the LB buds remained at a consistently higher level. The transition from dormancy to the active state in both cultivars was characterized by a sharp increase in DNA, RNA, protein, polyamines, S-adenosyl-methionine (SAM), 1-aminocyclopropane-1-carboxylic acid (ACC) and MACC. After initial swelling and development of flowers, the levels of all these components decreased. Polyamine and ethylene biosyntheses did not seem to be competing for their common substrate, SAM, during flower bud development.  相似文献

The pathway of ethylene biosynthesis was examined in two lower plants, the semi-aquatic ferns Regnellidium diphyllum Lindm. and Marsilea quadrifolia L. As a positive control for the ethylene-biosynthetic pathway of higher plants, leaves of Arabidopsis thaliana (L.) Heynh. were included in each experiment. Ethylene production by Regnellidium and Marsilea was not increased by treatment of leaflets with 1-aminocyclopropane-1-carboxylic acid (ACC), the precursor of ethylene in higher plants. Similarly, ethylene production was not inhibited by application of aminoethoxyvinylglycine and -aminoisobutyric acid, inhibitors of the ethylene biosynthetic enzymes ACC synthase and ACC oxidase, respectively. However, ACC was present in both ferns, as was ACC synthase. Compared to leaves of Arabidopsis, leaflets of Regnellidium and Marsilea incorporated little [14C]ACC and [14C]methionine into [14C]ethylene. Ethylene evolution from tobacco leaves irradiated with UV-B   总被引:8,自引:0,他引:8  
Seedlings of Nicotiana tabacum L. (cv. Petit Havana SR1) were grown in the presence or absence of ultraviolet-B (UV-B, 290–320 nm) irradiation. The evolution of ethylene from the leaves, the content of 1-aminocyclopropane-1-carboxylic acid (ACC), an endogenous precursor of ethylene, and the activity of ACC synthase, a rate-limiting step in the production of ethylene, were increased by UV-B irradiation. The time course of these increases was parallel with the emergence of damage that was estimated by measuring the chlorophyll (Chl) content and the leakage of ions from leaf cells. Treatment of leaves with aminoethoxy-vinyl-glycine (AVG), a specific inhibitor of ACC synthase, reduced the extent of damage caused by UV-B. These results suggest that ethylene acts on certain processes to cause damage in tobacco leaves irradiated with UV-B. Electronic Publication  相似文献

The rise in ethylene production accompanying the respiration climacteric and senescence of cut carnation flowers (Dianthus caryophyllus L. cv. White Sim) was associated with a 30-fold increase in the concentration of 1-aminocyclopropane-1-carboxylic acid (ACC) in the petals (initial content 0.3 nmol/g fresh weight). Pretreatment of the flowers with silver thiosulfate (STS) retarded flower senescence and prevented the increase in ACC concentration in the petals. An increase in ACC in the remaining flower parts, which appeared to precede the increase in the petals, was only partially prevented by the STS pretreatment. Wheat (Triticum aestivum L. cv. Jubilar) coleoptile segments convert 1-aminocyclopropane-1-carboxylic acid (ACC) to ethylene. This process is totally inhibited by nitrogen atmosphere and severely inhibited by free radical scavengers (sodium benzoate, ferulic acid), inhibitors of reactive -SH groups ( p -chlormercuribenzoate, iodoacetate), CoCl2 and EDTA. Indole-3-acetic acid, aminoethoxyvinyl glycine, cycloheximide, actinomycin D, pyridoxal phosphate and NADH have no effect on ACC conversion to ethylene. Some in vivo characteristics of this conversion suggest that it could be catalyzed by peroxidase. However, isoperoxidase B1 isolated from wheat seedlings was not able to catalyze in vitro conversion of ACC to ethylene under a wide range of reaction conditions. Therefore, it is concluded that peroxidase is not directly involved in ethylene biosynthesis.  相似文献

The role of ethylene in the control of senescence of both petals and unpollinated carpels of pea was investigated. An increase in ethylene production accompanied senescence, and the inhibitors of ethylene action were effective in delaying senescence symptoms in different flower verticils. Pollination did not seem to trigger the senescence syndrome in the corolla as deduced from the observation that petals from pollinated and unpollinated flowers and from flowers whose carpels had been removed senesced at the same time. A cDNA clone encoding a putative ethylene-response sensor (psERS) was isolated from pea flowers, and the pattern of expression of its mRNA was studied during development and senescence of different flower tissues. The levels of psERS mRNA paralleled ethylene production (and also levels of 1-aminocyclopropane-1-carboxylic acid oxidase (ACO) mRNA) in both petals and styles. Silver thiosulfate treatments were efficient at preventing ACO and psERS mRNA induction in petals. However, the same inhibitor showed no ability to modify expression patterns in pea carpels around the anthesis stage, suggesting different controls for ethylene synthesis and sensitivity in different flower organs. The increase in ethylene formation and in 1-aminocyclopropane-1-carboxylic acid (ACC) content in flavedo tissue of grapefruit (Citrus paradisi Macfad. cv. Ruby Red) in response to excision was markedly inhibited by exogenous ethylene. Ethylene treatment inhibited the synthesis of ACC, but increased the tissue's capability to malonylate ACC to N-malonyl-ACC, resulting in further reduction in the endogenous ACC content. The development of extractable ACC-malonyl-transferase activity in the tissue was markedly promoted by treatment with exogenous ethylene. Isolated membranes from the petals of senescing carnation flowers (Dianthus caryophyllus L. cv. White-Sim) catalyze the conversion of 1-aminocyclopropane-1-carboxylic acid (ACC) to ethylene. A microsomal membrane fraction obtained by centrifugation at 131,000 g for 1 h proved to be more active than the membrane pellet isolated by centrifugation at 10,000 g for 20 min. The ethylene-producing activity of the microsomal membranes is oxygen-dependent, heat-denaturable, sensitive to n-propyl gallate, and saturable with ACC. Corresponding cytosol fractions from the petals are incapable of converting ACC to ethylene. Moreover, the addition of soluble fraction back to the membrane fraction strongly inhibits the ACC to ethylene conversion activity of the membranes. The efficiency with which isolated membranes convert ACC to ethylene is lower than that exhibited by intact flowers based on the relative yield of membranes per flower. Using cotyledonary segments of cocklebur ( Xanthium pennsylvanicum Wallr. ) seeds, the inhibitory effect of α-aminoisobutyric acid (AIB) on ethylene production was compared with that of propyl gallate and CoCl2. Of these inhibitors only AIB was effective in causing the accumulation of endogenous free 1-aminocyclopropane-l-carboxylic acid (ACC) in the tissue. The degree of inhibition of ethylene production by AIB decreased markedly with increasing concentrations of pre-loaded ACC, while the inhibition by propyl gallate and CoCl2 changed little. Kinetic analysis showed that AIB competitively inhibited the conversion of pre-loaded ACC to ethylene, but propyl gallate and CoCl2 did not. Short-chain organic acids and analogues of AIB, such as acetic, propionic, butyric and cyclopropanecarboxylic acids, did not inhibit ethylene production by the segments. Thus, additional support for the competitive mode of inhibitory action of AIB on the conversion of free ACC to ethylene was provided.
A conjugated hydrolysable ACC was found to be present in abundance in cotyledons of this seed. However, its content in the tissue was hardly affected by treatment with the three inhibitors and by administration of exogenous ACC, suggesting that the conjugated ACC was not directly involved in ethylene production.  相似文献

The cofactor of enzymatic, 1-aminocyclopropane-1-carboxylic acid dependent ethylene formation was concentrated on cation exchange columns. When chelators of cations were added to the homogenates, cofactor activity was lost. Cofactor fractions were partly resistant to oxidation at 600° C. Mn2+ substituted for the cofactor in ethylene formation from 1-aminocyclopropane-1-carboxylic acid by a protein fraction isolated from etiolated pea shoots. In addition, Mn2+ enhanced the stimulatory effect of the concentrated cofactor. The elution volume for the cofactor on a Sephadex G-25 column was lower than that of MnCl2. In paper electrophoresis the cofactor migrated to the cathode at pH 10.8 and 2.2. The RF of cofactor on cellulose plates developed in butanol: acetic acid: H2O was 0.4. After cellulose chromatography, cofactor activity had to be reconstituted by the addition of MnCl2. Chelators, anti-oxidants, and catalase were inhibitors of Mn2+-cofactor-dependent ethylene formation. Hypobaric conditions and treatments with ethylene and the ethylene analogue propylene were used to investigate effects of oxygen and elhylene on 1-aminocyclopropane-1-carboxylic acid (ACC) content, ACC synthase activity and ethylene production of apples ( Malus sylveslris Mill. cv. Golden Delicious). Prcclimacteric apples were stored in air at 6.6 kPa (reduced pressure); 6.6 kPa ventilated with pure O2; 6.6 kPa ventilated with 2600 μl 1−1 C2H4; and in air at 101.3 kPa (atmospheric pressure) for 4 months at 4°C. No ACC synthase activity was detectable in apples stored at 6.6 kPa, whereas ACC synthase activity was induced in apples stored at 6.6 kPa and ventilated with either O2 or C2H4. In a further experiment, preclimacteric apples were stored for 14 days either in air at 20 kPa or at 20 kPa ventilated with pure O2. Both treatments were supplied with 58 500 μl 1−1 propylene from day 0 to day 9 or from day 9 to day 12. Ethylene production of apples treated with propylene from day 0 to day 9 increased earlier than ethylene production of untreated apples. Propylene treatment from day 9 to day 12 did not stimulate ethylene production. Ethylene and propylene induced and stimulated extractable ACC synthase activity and ACC formation of apples. Oxygen enhanced this effect. The results also suggest inhibition of in vivo ACC synthase activity by propylene.  相似文献

The biosynthesis of ethylene was examined in suspension-cultured cells of parsley (Petroselinum hortense) treated with an elicitor from cell walls of Phytophthora megasperma. Untreated cells contained 50 nmol g-1 of the ethylene precursor, 1-aminocyclopropane-1-carboxylic acid (ACC), and produced ethylene at a rate of about 0.5 nmol g-1 h-1. Within 2 h after addition of elicitor to the culture medium, the cells started to produce more ethylene and accumulated more ACC. Exogenously added ACC did not increase the rate of ethylene production in control or elicitor-treated cells, indicating that the enzyme converting ACC to ethylene was limiting in both cases. The first enzyme in ethylene biosynthesis, ACC synthase, was very rapidly and transiently induced by the elicitor treatment. Its activity increased more than tenfold within 60 min. Density labelling with 2H2O showed that this increase was caused by the denovo synthesis of the enzyme protein. Cordycepin and actinomycin D did not affect the induction of ACC synthase, indicating that the synthesis of new mRNA was not required. The peak of ACC-synthase activity preceded the maximal phenylalanine ammonia-lyase (PAL) activity by several hours. Exogenously supplied ethylene or ACC did not induce PAL. However, aminoethoxyvinylglycine, an inhibitor of ACC synthase, suppressed the rise in ethylene production in elicitor-treated cells and partially inhibited the induction of PAL. Exogenously supplied ACC reversed this inhibition. It is concluded that induction of the ethylene biosynthetic pathway is a very early symptom of elicitor action. Ethylene production by intact green bean ( Phaseolus vulgaris L. cv. Limburgse vroege) seedlings was investigated in white light and in darkness. In white light both endogenous and 1-aminocyclopropane-1-carboxylic acid (ACC)-induced ethylene production were stimulated. A decrease in the 1-(malonylamino)cyclopropane-1-carboxylic acid (M-ACC) level and a slight increase in the free ACC concentration could be observed in light. The total amount of endogenous ACC was not changed by light. We related the effect of light to the effect of paraquat on ethylene biosynthesis. Paraquat caused a strong increase of endogenous ethylene production in light. However, the conversion of exogenously applied ACC in light was not influenced by the paraquat treatment, although the presence of the herbicide in the chloroplasts was evident through the inhibition of net photosynthesis. In light, paraquat increased the total ACC content. This was due to an enlargement of the free ACC pool. The effects of white light and paraquat on ethylene biosynthesis can be differentiated from one another: white light exerts its influence on the conversion of ACC to ethylene; it also seems to inhibit the malonylation and may act on the formation of ACC itself. The characteristics of the conversion of 1-aminocyclopropane-1-carboxylic acid (ACC) to ethylene by pea (Pisum sativum L.) epicotyls and by pea epicotyl enzyme are compared. Of the four stereoisomers of 1-amino-2-ethylcyclopropane-1-carboxylic acid (AEC), only (1R,2S)-AEC is preferentially converted to 1-butene in pea epicotyls. This conversion is inhibited by ACC, indicating that butene production from (1R,2S)-AEC and ethylene production from ACC are catalyzed by the same enzyme. Furthermore, pea epicotyls efficiently convert ACC to ethylene with a low K m (66 M) for ACC and do not convert 4-methylthio-2-oxo-butanoic acid (KMB) to ethylene, thus demonstrating high specificity for its substrate. In contrast, the reported pea epicotyl enzyme which catalyzes the conversion of ACC to ethylene had a high K m (389 mM) for ACC and readily converted KMB to ethylene. We show, moreover, that the pea enzyme catalyzes the conversion of AEC isomers to butene without stereodiscrimination. In order to understand the physiological significance of the in-vitro lipoxygenase (EC 1.13.11.12)-mediated ethylene-forming system (J.F. Bousquet and K.V. Thimann 1984, Proc. Natl. Acad. Sci. USA 81, 1724–1727), its characteristics were compared to those of an in-vivo ethylene-forming system. While oat (Avena sativa L.) leaves, as other plant tissues, preferentially converted only one of the 1-amino-2-ethylcyclopropane-1-carboxylic acid (AEC) isomers to 1-butene, the lipoxygenase system converted all four AEC isomers to 1-butene with nearly equal efficiencies. While the in-vivo ethylene-forming system of oat leaves was saturable with ACC with a Km of 16 M, the lipoxygenase system was not saturated with ACC even at 10 mM. In contrast to the in-vivo results, only 10% of the ACC consumed in the lipoxygenase system was converted to ethylene, indicating that the reaction is not specific for ethylene formation. Increased ACC-dependent ethylene production in oat leaves following pretreatment with linoleic acid has been inferred as evidence of the involvement of lipoxygenase in ethylene production. We found that pretreating oat leaves with linoleic acid resulted in increased ACC uptake and thereby increased ethylene production. A similar effect was observed with oleic acid, which is not a substrate of lipoxygenase. Since linoleic acid hydroperoxide can substitute for lipoxygenase and linoleic acid in this system, it is assumed that the alkoxy radicals generated during the decomposion of linoleic acid hydroperoxide are responsible for the degradation of ACC to ethylene. Methyl jasmonate (JA-Me), applied to dendrobium and petunia flowers either as an aqueous solution through the cut stem or stigma, or as a gas, accelerated senescence. The rate of appearance of wilting symptoms was directly related to the amount of JA-Me applied to the flowers. JA-Me increased ethylene production by the flowers, irrespective of application method, and this effect was also proportional to the dose of the compound. In both dendrobium and petunia flowers, the JA-Me induced increases in ethylene production and 1-aminocyclopropane-1-carboxylic acid content followed similar patterns. Aminooxyacetic acid, an inhibitor of ACC-synthase, and silver-thiosulfate, an inhibitor of ethylene action, completely inhibited the effects of JA-Me. Changes in the level of 1-(malonylamino)-cyclopropane-1-carboxylic acid (MACC) were determined in 6 winter wheat cultivars during cold hardening at 4°C. The cultivars differed by one degree of frost resistance within the range of degree II to VII of the COMECON scale. The time-course of changes in MACC level showed a similar pattern in all 6 cultivars; i.e. increase till day 6, no changes for the next 10 days, and then a steady decrease till the end of the hardening period. There was little difference between the final and the initial levels. The increase of MACC level, expressed as per cent of the original level, was not directly correlated with either the degree of frost resistance of the actual percentage of survival. In some cultivars. mean errors exceeded the difference in MACC accumulation between cultivars closest on the resistance scale.
The fate of MACC during the second half of hardening and after transfer of plants to 25°C was studied in cultivars Bezostaya and San Pastore. During the second half of the hardening period the level of MACC decreased in the leaves of both cultivars, but increased significantly in the roots. Within two days of transfer of the hardened plants to 25°C, the MACC level in leaves increased again, while that in the roots decreased. This finding, together with the preliminary evidence of very low MACC metabolism, strongly suggest that MACC accumulates in roots during the hardening period and when transferred to 25°C, it moves from roots to leaves.  相似文献

Activity and biochemical characteristic of 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase from pear ( Pyrus communis cv. Blanquilla) was determined. The enzyme showed a low Km (57.5 μM) for ACC and was dependent on O2 (Km 0.44% in atmosphere). It had an absolute requirement for Fe2+, ascorbate and CO2 and was inhibited by α-aminoisobutyric acid (AIB: K1 4.2 m M ) and cobalt. ACC oxidase has an optimum pH of 6.7 and temperature maxima at 28 and 38°C and it is concluded that the activity of ACC oxidase from pear resembles authentic in vivo activity.  相似文献
