Macrocentrus grandii which develop within Nosema pyrausta-infected larvae of the European corn borer, Ostrinia nubilalis, develop direct systemic infections from the ingestion of spores at the time of larval emergence from the host. Infections adversely affect pupal development and adult longevity. Infected females are unable to transmit the microsporidian to additional corn borer hosts. Pathogen development in the parasite host appears identical to its development in the corn borer host and mature spores show no morphological differences in size or shape when observed at the ultrastructural level. The prevalence of infection in natural parasite populations is 53.8% and closely parallels the 56.7% prevalence of infection in corn borer populations. Results suggest N. pyrausta may play a significant role in limiting M. grandii populations when levels of N. pyrausta in corn borers are high.  相似文献

Nosema pyrausta (Paillot) (Microsporida: Nosematidae) is an obligatory intracellular parasite of the European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae). This pathogen is maintained in natural populations of O. nubilalis by both horizontal and vertical transmission. The impact of N. pyrausta on fecundity of adults and survival of larvae has been well documented in laboratory and field research. In an extensive study covering a 6 year period at one site, we described the effect of N. pyrausta within O. nubilalis populations in a continuous corn following corn ecosystem. We documented the presence of the pathogen through all life stages of O. nubilalis (egg, larva, pupa, adult), by collecting throughout the crop season and examining each insect stage in the laboratory for the frequency of infection with N. pyrausta. The percentage of infection with N. pyrausta and magnitude of the O. nubilalis population fluctuated throughout generation 1 and generation 2. Both horizontal and vertical transmission played a role in maintaining N. pyrausta in the population in both generations. There were strong correlations between percentage adults with N. pyrausta and percentage larvae with N. pyrausta, and between percentage eggs with N. pyrausta and percentage larvae with N. pyrausta. There was a weak correlation between percentage adults with N. pyrausta and percentage eggs with N. pyrausta. The percentage of insects infected with N. pyrausta was always lowest in the egg.  相似文献

In field populations of the European corn borer, Ostrinia nubilalis, there is a significant inverse correlation between the prevalence of corn borer infection with the microsporidian Nosema pyrausta and the prevalence of parasitism by the introduced braconid Macrocentrus grandii where infections with N. pyrausta exceed 45%. This relationship occurs geographically and from year to year. Corn borer infection with N. pyrausta, the source of infection for M. grandii, is significantly related to corn borer density in the cornfield. These findings strongly suggest that N. pyrausta has a significant adverse affect on field populations of M. grandii and may help explain the diminishing role of this and other introduced parasites as natural controls of the corn borer in the United States.  相似文献

Intraplant epizootics of entomopathogens,Beauveria bassiana (Balsamo) Vuillemin, andNosema pyrausta (Paillot) were studied in a corn,Zea mays L., agroecosystem. Egg masses of the European corn borer,Ostrinia nubilalis (Hübner), infected withN. pyrausta were placed on midwhorl-stage corn plants. Conidia ofB. bassiana were applied in an aqueous suspension. Frass from the initial insects remaining within the plants was contaminated with sufficientN. pyrausta spores to infest 80% of the filial generation in each year of a 2-yr study. Viability ofN. pyrausta within the frass was monitored throughout the winter. Potential for impact of this inoculum on the filial generation is discussed. Conidia from cadavers of the European corn borer that were killed by the initial inoculum ofB. bassiana and/or conidia from the initial inoculum ofB. bassiana significantly reduced tunneling by the filial generation of the European corn borer. However, neither the concentration ofB. bassiana nor the age of the larvae exposed toB. bassiana, had any significant (P<.05) effect on tunneling by the 2nd-generation larvae. This paper reports the results of research only. Methods of preserving Nosema pyrausta spores were developed by subjecting laboratoryreared, nondiapausing larvae of the European corn borer, Ostrinia nubilalis, infected with N. pyrausta to different techniques of lyophilization and vacuum drying. Such material was still viable when it was stored in an airtight container and held at ?12°C for 12 months, or when it was stored in a vacuum desiceator and held at 22–24°C for 6 months.  相似文献

The seasonal histories and phenological relationships of European corn borer,Ostrinia nubilalis (Hübner), and its 2 parasitoids,Macrocentrus grandii (Goidanich) andEriborus terebrans (Gravenhorst) were studied in southcentral Minnesota. Both parasitoids overwintered in mature borer larvae, broke diapause, completed development, and emerged at the same time as did borer adults. Thus the 1st generation parasitoids coincided with the peak abundance of their preferred larval instars of the 1st host generation. Both parasitoids had a 2nd generation, matching the bivoltinism ofO. nubilatis in Minnesota. The activity of 2nd generationE. terebrans was before the peak 2nd generation borer moth flight and was not synchronized with the peak abundance of 2nd generation borer larvae. The peak activity of 2nd generationM. grandii occurred after the peak 2nd generation borer moth flight and was fully synchronized with the peak abundance of 2nd generation borer larvae. Thus,M. grandii has both generations synchronized with the host seasonal history, and was the more effective of the 2 parasitoids.  相似文献

This paper is the first report of the impact on the bacterial community in the midgut of the Asian corn borer (Ostrinia furnacalis) by the chitinase from the transgenic Trichoderma strain. In this study, we detected a change of the bacterial community in the midgut of the fourth instar larvae by using a culture-independent method. Results suggested that Proteobacteria and Firmicutes were the most highly represented phyla, being present in all the midgut bacterial communities. The observed species richness was simple, ranging from four to five of all the 16S rRNA clone libraries. When using Trichoderma fermentation liquids as additives, the percentages of the dominant flora in the total bacterial community in larval midgut changed significantly. The community of the genus Ochrobactrum in the midgut decreased significantly when the larvae were fed with the fermentation liquids of the transgenic Trichoderma strain Mc4. However, the Enterococcus community increased and then occupied the vacated niche of the Ochrobactrum members. Furthermore, the Shannon–Wiener (H) and the Simpson (1-D) indexes of the larval midgut bacterial library treated by feeding fermentation liquids of the transgenic Trichoderma strain Mc4 was the lowest compared with the culture medium, fermentation liquids of the wild type strain T30, and the sterile artificial diet. The Enterococcus sp. strain was isolated and characterized from the healthy larvae midgut of the Asian corn borer. An infection study of the Asian corn borer larvae using Enterococcus sp. ACB-1 revealed that a correlation existed between the increased Enterococcus community in the larval midgut and larval mortality. These results demonstrated that the transgenic Trichoderma strain could affect the composition of the midgut bacterial community. The change of the midgut bacterial community might be viewed as one of the factors resulting in the increased mortality of the Asian corn borer larvae.  相似文献

The effects of juvenile hormone (JH) on the larval diapause of the European corn borer, Ostrinia nubilalis, were investigated. The larval period was prolonged and the development of gonads was retarded if non-diapause larvae were treated with JH. Pupation of diapausing larvae was accelerated by allatectomy. We tested the combined effect of the fungus Beauveria bassiana and the microsporidium Nosema pyrausta on the European corn borer larvae, Ostrinia nubilalis, in the laboratory. The first instar of O. nubilalis larvae was the most sensitive to the B. bassiana infection followed by the fifth, second, third, and fourth instar (LC50s were 4.91, 6.67, 7.13, 9.15, and 6.51 × 105 conidia/ml for the first to fifth instars, respectively). Mortality of each instar increases positively with concentration of conidia. When B. bassiana and N. pyrausta were used in combination, mortality increased significantly in all instars. Relative to the B. bassiana treatment alone, the B. bassiana + N. pyrausta treatment decreased the LC50s by 42.16%, 37.63%, 21.60%, 27.11%, and 33.95% for the first to fifth instars, respectively. The combined effects of the two pathogens were mostly additive. However, at the two highest concentrations the pathogens interacted synergistically in the first and second instar. Individuals that survived the B. bassiana and B. bassiana + N. pyrausta treatments and developed into adults had significantly shorter lifespans and females oviposited fewer eggs than non-exposed insects. The effects on the longevity and the egg production were most pronounced at high concentration of B. bassiana conidia.  相似文献

In 1974, an application of the microsporidan,Nosema pyrausta (Paillot, 1927)Kotlan, 1928, with a back-pack type sprayer (22.5×107 spores/plant) to whorl stage maize infested with European corn borers,Ostrinia nubilalis (Hübner) reduced the number of larvae/plant by 48.1% and produced an infection of 15.3×104 spores/mg of larval weight in 62.1% of the collected larvae. In 1975, applications of 24.3×107 spores/plant to similar maize, in 2 separate tests, reduced the number of larvae/plant by 18.8 and 43.8% and caused an infection of 14.3 and 19.1×104 spores/mg of larval weight in 65.9 and 63.3% of the collected larvae. Also, in 1975, applications of 24.3×107 spores/plant to pollen shedding maize in 2 separate tests reduced the number of larvae/plant by 17.2 and 14.1% and caused an infection of 24.3 and 27.2×104 spores/mg of larval weight in 99.2 and 95.2% of the collected larvae.  相似文献

Infection with Nosema pyrausta Paillot lengthens developmental period of Bt-susceptible Ostrinia nubilalis (Hübner) to a similar extent as feeding on Cry1Ab-incorporated diet in Cry1Ab-resistant O. nubilalis, and these two factors combined lengthen developmental period further than either alone. Resistant O. nubilalis mating with infected susceptible, or infected resistant partners would produce partially- and fully-resistant offspring, respectively, infected with N. pyrausta. To investigate the impacts on the progeny of such matings, test crosses were set up to produce partially- and fully Cry1Ab-resistant O. nubilalis offspring transovarially infected and not infected with N. pyrausta, which were exposed to Cry1Ab toxin at doses of 0, 3, or 30 ng/cm2 for 7 days. Transovarial infection with N. pyrausta significantly decreased 7 day survival of partially and fully-resistant O. nubilalis feeding on 30 ng/cm2 Cry1Ab. In addition, N. pyrausta infection delayed larval development (as measured by weight) of partially- and fully-resistant O. nubilalis feeding on 3 and 30 ng/cm2 Cry1Ab. Impacts of natural enemies on target pests may have the potential to impact evolution of resistance. N pyrausta-infected O. nubilalis are more strongly affected by feeding on Bt, and would be less likely to survive to adulthood to pass on resistance to the next generation. This indigenous microsporidium may work to delay evolution of resistance in O. nubilalis by lowering their ability to survive on Bt.  相似文献

The organization of the retrocerebral gland system in larvae of six species of Lepidoptera belonging to the family Pyralidae was compared using light and electron microscopy. We have demonstrated for the first time the presence of separate corpora cardiaca and corpora allata in the following economically important borers: the southwestern corn borer, Diatraea grandiosella, the sugar cane borer, Diatraea saccharalis, the European corn borer, Ostrinia nubilalis, and the rice stalk borer, Chilo plejadellus. In these species a long nervus corporis allati (ca. 300 μm) runs from the corpus cardiacum to the corpus allatum which is attached to the duct of the mandibular gland.The identity of the corpora allata of D. grandiosella was confirmed by transplantation. Corpora allata removed from pre-diapausing larvae and implanted into the haemocoele of early last stage non-diapausing larvae led to a high incidence of supernumerary larval rather than pupal ecdyses.  相似文献

The high dose/refuge strategy for delaying evolution of resistance to Bt maize [Zea mays L. (Poaceae)] relies on random mating between resistant European corn borers, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae), and susceptible O. nubilalis from the refuge. However, differences in developmental rate caused by feeding on Bt maize, or infection with the microsporidium Nosema pyrausta Paillot (Microsporida: Nosematidae) may result in assortative mating. Developmental delays and mortality caused by infection with N. pyrausta and feeding on Bt maize were quantified alone and in combination in Cry1Ab‐resistant and susceptible O. nubilalis. Feeding on Cry1Ab‐incorporated diet significantly increased number of days from hatch to pupation and decreased survival in the resistant population. Infection with N. pyrausta increased mortality and lengthened development in both the resistant and susceptible populations. The combination of Cry1Ab‐incorporated diet and infection with N. pyrausta in resistant O. nubilalis lengthened development and increased mortality to a greater extent than either factor alone. Greater larval delays of resistant O. nubilalis feeding on Bt maize could lead to temporal isolation from adults emerging from refuge maize. The resulting assortative mating would hasten the evolution of resistance. Developmental delays caused by infection with N. pyrausta may increase the likelihood of mating between resistant and infected susceptible adults emerging from refuge maize, producing infected offspring that are also more susceptible to Bt maize.  相似文献

Fumidil B suppressed the level of Perezia pyraustae infection in the larvae of the European corn borer, Ostrinia nubilalis, while they were feeding on diet containing the drug. After feeding ceased and the gut was emptied of diet containing Fumidil B, the level of infection again increased. Drug levels of 1000 and 1500 ppm in the meridic diet significantly reduced the mean level of P. pyraustae in the adult stage as compared with lower drug levels of 200 and 500 ppm. The level of infection increased throughout the pupal and adult stages, but during the pupal stage it increased much faster in the female than in the male. The rapid increase in the level of infection in the female is discussed in relation to transovariole transmission to the next generation.  相似文献

Transgenic corn, Zea mays L., hybrids expressing crystal protein endotoxin genes from Bacillus thuringiensis Berliner are an increasingly popular tactic for managing the European corn borer, Ostrinia nubilalis (Hübner), in North America. O. nubilalis populations also are often vulnerable to the ubiquitous entomopathogenic microsporidium Nosema pyrausta (Paillot). We examined the effect of feeding meridic diet incorporated with purified Cry1Ab on growth, development, and survival of Nosema-infected and uninfected neonate O. nubilalis. Infected larvae developed more slowly than uninfected larvae. Increasing the concentration of Cry1Ab in diet reduced larval development, and this effect was amplified by microsporidiosis. Infected larvae weighed significantly less than uninfected larvae. The relationship among Nosema infection, Cry1Ab concentration, and larval weight was fitted to an exponential function. The LC50 of infected larvae was one-third that of uninfected larvae, indicating that infected larvae are more vulnerable to toxin. This work has implications for resistance management of O. nubilalis and demonstrates that it is important to determine whether N. pyrausta is present when testing susceptibility of larvae to transgenic corn hybrids.  相似文献

The parasite Microplitis croceipes required 1.1 days longer at 26°C to complete development in Heliothis zea larvae than was required for the fungus Nomuraea rileyi to kill the host larvae and sporulate. Host larvae parasitized by M. croceipes or infected with N. rileyi failed to complete a fifth larval molt or pupate. Of the remaining healthy larvae, one-half completed six larval stadia before popation. Larvae parasitized by M. croceipes were predisposed to infection by N. rileyi, but the fungus inhibited development of M. croceipes if host larvae were infected with N. rileyi within 1 day after parasitization.  相似文献

Long-term observations have revealed the factors responsible for periodic rises in abundance of the European corn borer Ostrinia nubilalis Hb. in the Central Chernozem Region of Russia. These factors are sufficient precipitation during several consecutive years and the current decline in agrotechnical soil treatment. The host specialization, biology, and phenology of the corn borer in the region have been clarified. Corn is damaged most intensely among the cultivated plants, and the yellow foxtail Setaria glauca, among the weeds. The effects of precipitation on seasonal development and harmfulness of the corn borer have been demonstrated. Low precipitation during the pest pupation period leads to mass pupal mortality, a shift of the seasonal development onto later dates, and a lower proportion of damaged stems in the crops. Insufficient precipitation during the larval feeding period enhances the effect of pest damage on the plant productivity and results in heavier yield loss. During the corn borer outbreaks the yield loss reached 15% in corn and 11.4% in millet. Depending on the weather conditions, the loss of millet yield due to this pest may be as great as 39% or as low as 3%.

The ability of non‐crop plants to support complete development of insect pests is an important factor for determining the impact of those plants on resistance management programs for transgenic crops. We assessed the effect of one physical factor, plant stem diameter, on the ability of plants to support full development of the European corn borer (ECB), Ostrinia nubilalis Hübner (Lepidoptera: Crambidae), the target pest of transgenic Bt‐corn. In the field, European corn borer larvae were significantly more likely to tunnel and survive in plants with larger stem diameters. Larvae were 40× more likely to survive on corn, the largest plant tested, compared to many of the smaller plants. In the laboratory, larvae were more likely to survive in and less likely to abandon the largest diet‐filled artificial stems that varied only in stem diameter. In conditions simulating those that an ECB larvae would encounter upon abandoning a host, larvae survived up to three weeks and were able to locate corn as a new host with a significantly higher frequency than would be expected if they were foraging randomly. These results indicate that the probability of ECB larval survival to maturity on a plant other than corn is relatively low and thus these smaller stemmed non‐corn plants may not make a substantial contribution to the pool of susceptible adults. Conversely, since more mature larvae are not as susceptible as neonates, any larvae that partially develop on non‐corn plants and subsequently colonize Bt‐corn may not be exposed to a lethal dose of the toxin. Since some proportion of the individuals that survive could be partially resistant heterozygotes the presence of non‐corn host plants could facilitate the development of resistant ECB populations.  相似文献
