Verticillium wilt of lettuce caused by Verticillium dahliae can cause severe economic damage to lettuce producers. Complete resistance to race 1 isolates is available in Lactuca sativa cultivar (cv.) La Brillante and understanding the genetic basis of this resistance will aid development of new resistant cultivars. F1 and F2 families from crosses between La Brillante and three iceberg cultivars as well as a recombinant inbred line population derived from L. sativa cv. Salinas 88 × La Brillante were evaluated for disease incidence and disease severity in replicated greenhouse and field experiments. One hundred and six molecular markers were used to generate a genetic map from Salinas 88 × La Brillante and for detection of quantitative trait loci. Segregation was consistent with a single dominant gene of major effect which we are naming Verticillium resistance 1 (Vr1). The gene described large portions of the phenotypic variance (R 2 = 0.49–0.68) and was mapped to linkage group 9 coincident with an expressed sequence tag marker (QGD8I16.yg.ab1) that has sequence similarity with the Ve gene that confers resistance to V. dahliae race 1 in tomato. The simple inheritance of resistance indicates that breeding procedures designed for single genes will be applicable for developing resistant cultivars. QGD8I16.yg.ab1 is a good candidate for functional analysis and development of markers suitable for marker-assisted selection.  相似文献

Verticillium wilt (VW) is a widespread and serious potato (Solanum tuberosum) disease caused by the soilborne fungi Verticillium dahliae and V. albo-atrum. Breeding for VW resistance in potato is challenging due to ambiguous symptom expression, a lack of high throughput screening techniques, and variability in colonization by the fungus among and within plants. Genetic studies have identified major genes that confer resistance in diploid Solanum chacoense (V c ) and interspecific hybrids (V w and V t ). However, to date, these genes have not been used to develop molecular markers for the identification of resistant clones. Tomato Ve1 and Ve2 gene sequence information was used to amplify candidate Ve gene orthologs from both resistant and susceptible diploid potato hybrids. A CAPS marker was generated to track VW resistance in a backcross population segregating for resistance. The marker was also tested for its usefulness in other breeding lines. Our results indicate that this marker is effective for selection of the V w gene in segregating breeding populations.  相似文献

Improving genetic resistance is a preferred method to manage Verticillium wilt of cotton and other hosts. Identifying host resistance is difficult because of the dearth of resistance genes against this pathogen. Previously, a novel candidate gene involved in Verticillium wilt resistance was identified by a genome-wide association study using a panel of Gossypium hirsutum accessions. In this study, we cloned the candidate resistance gene from cotton that encodes a protein sharing homology with the TIR-NBS-LRR receptor-like defence protein DSC1 in Arabidopsis thaliana (hereafter named GhDSC1). GhDSC1 expressed at higher levels in response to Verticillium wilt and jasmonic acid (JA) treatment in resistant cotton cultivars as compared to susceptible cultivars and its product was localized to nucleus. The transfer of GhDSC1 to Arabidopsis conferred Verticillium resistance in an A. thaliana dsc1 mutant. This resistance response was associated with reactive oxygen species (ROS) accumulation and increased expression of JA-signalling-related genes. Furthermore, the expression of GhDSC1 in response to Verticillium wilt and JA signalling in A. thaliana displayed expression patterns similar to GhCAMTA3 in cotton under identical conditions, suggesting a coordinated DSC1 and CAMTA3 response in A. thaliana to Verticillium wilt. Analyses of GhDSC1 sequence polymorphism revealed a single nucleotide polymorphism (SNP) difference between resistant and susceptible cotton accessions, within the P-loop motif encoded by GhDSC1. This SNP difference causes ineffective activation of defence response in susceptible cultivars. These results demonstrated that GhDSC1 confers Verticillium resistance in the model plant system of A. thaliana, and therefore represents a suitable candidate for the genetic engineering of Verticillium wilt resistance in cotton.  相似文献

Two potential biological control agents of cucumber powdery mildew (Sphaerotheca fuliginea),Verticillium lecaniiandSporothrix rugulosa,were tested under glasshouse conditions. Two experiments were carried out. In the first experiment, two cucumber varieties with different levels of resistance, cv Corona (susceptible) and cv Flamingo (partially resistant), were used.Verticillium lecaniicontrolled the mildew better thanS. rugulosa.On cv Flamingo,V. lecaniicould keep the mildew severity below 15% infected leaf area for 9 weeks after inoculation withS. fuliginea.Treatment by Hora Oleo 11E, alone or as an additive toV. lecanii,was as good as a fungicide treatment. In the second experiment, weekly and biweekly treatments withV. lecaniiwere compared on cv Flamingo. Germin-like proteins (GLPs) play important roles in plant disease resistance but are rarely reported in cotton. We compared the expression of GLPs in Verticillium dahliae inoculate G. hirsutum (susceptible) and G. barbadense (resistant) and enriched 11 differentially expressed GLPs. 2741 GLP proteins identified from 53 species determined that GLP probably originated from algae and could be classified into 7 clades according to phylogenetic analysis, among which Clade I is likely the most ancient. Cotton GLP (two allopolyploids and two diploids) genes within a shared clade were highly conserved. Intriguingly, clade VII genes were mainly located in gene clusters that derived from the expansion of LTR transposons. Clade VII members expressed mainly in root which is the first battle against Verticillium dahlia and could be induced more intensely in G. barbadense than G. hirsutum. The GLP genes are resistant to Verticillium dahliae, which can be further investigated against Verticillium wilt.  相似文献

Several bacterial and fungal strains have been evaluated as biocontrol agents (BCAs) against Verticillium dahliae. In these studies, the BCAs were applied as a root drenching inoculum; however, this application method may have an adverse effect on the native, beneficial for the plants, microbial community. In the present study, it was evaluated whether endophytic application by stem injecting a conidial suspension of the non pathogenic Fusarium oxysporum F2 strain, isolated from a V. dahliae suppressive compost amendment, could reduce significantly Verticillium wilt symptom development in eggplants. It was revealed that stem injection of F2 seven days before transplanting the seedlings to soil infested by V. dahliae microsclerotia resulted in reduced disease severity compared to the control treatment. To visualise F2 ramification into the plant vascular system eggplant stems were injected with an EGFP transformed F2 strain. It was shown that F2 colonises effectively the plant vascular tissues over a long period of time as it was assessed by F2 DNA levels. In parallel, qPCR analysis showed that the application of F2 reduced significantly the amount of V. dahliae DNA in the stem tissues compared to the control treatment.  相似文献

Six cultivars and breeding lines of potato (Solanum tuberosum) differing in susceptibility to verticillium wilt caused by Verticillium dahliae were studied with respect to quantitative and qualitative differences in the bacterial flora of their soil and rhizosphere-rhizoplane. Although, no association was observed between the types of bacteria that inhabited the soil or roots of wilt resistant and susceptible cultivars, quantitative differences were evident. These differences provide the first direct evidence that potato genotypes can influence bacterial populations. Bacterial populations were 9–25-fold higher on roots than in the adjacent soil. As the plants aged, the total number of rootcolonizing bacteria increased between 15 and 245%. Pseudomonas spp. were the most abundant microbes in the soil and rhizosphere-rhizoplane. The bacteria antagonistic to V. dahliae in vitro were identified as members of the genera Bacillus, Pseudomonas, Flavobacterium, and Gluconobacter. A statistically significant trend was evident toward the association of antagonistic bacteria with the more resistant potato cultivars.  相似文献

Endophytes, bacterial, fungal or viral, colonize plants often without causing visible symptoms. More important, they may benefit host plants in many ways, most notably by preventing diseases caused by normally virulent pathogens. Craigella tomatoes (Lycopersicon esculentum Mill.) can be infected with Verticillium dahliae Kleb., either race 1 (Vd1) or a non-host isolate Dvd-E6 resulting in susceptibility or tolerance, respectively. The present study sought to determine whether Dvd-E6 is endophytic and can protect tomato against Vd1. The total amount of Verticillium in stems and roots was determined by quantitative PCR; the relative amounts of Vd1 and Dvd-E6 were assessed by restriction fragment polymorphism. When Dvd-E6 infects before or together with Vd1, Vd1 is excluded almost completely from the root but, when Vd1 infects first, Dvd-E6 can compete on an equal basis. Previous studies suggested that Dvd-E6 suppresses symptom-related genes, raising the possibility that Dvd-E6 simultaneously induces tolerance to Vd1. This does not seem to be entirely the case since the minimal symptoms following Vd1 infection of Dvd-E6 tolerant Craigella result, at least in part, from restricted Vd1 colonization. Furthermore, when Vd1 and Dvd-E6 are cultured on PDA plates alone or together, the growth rates are similar and neither is inhibitory to the other. Dvd-E6 does not outgrow or inhibit Vd1, in vitro. The protective effect apparently requires interplay between Dvd-E6 and the plant. Expression analyses of tomato genes involved in resistance and defence support this interpretation.  相似文献

The need for a new analytical approach was encountered in the course of characterizing newly developed tomato lines resistant to late blight. Late blight resistant tomato lines were created in independent breeding programs using the accession Solanum pimpinellifolium L. (formerly Lycopersicon pimpinellifolium (L.) Miller) L3708 as the source of the resistance. However, initial field observation suggested that the late blight resistance in the lines produced by two independent breeding programs differed. Possible causes included a partial transfer of the late blight resistance derived from S. pimpinellifolium L3708 or the possibility of race specificity of this resistance. A crucial issue was determining the most appropriate and robust analytical method to use with data from laboratory analyses of the responses of nine tomato lines against five P. infestans isolates. Prior analysis by standard ANOVA revealed significant differences across tomato lines but could not determine whether the disease responses in the CLN-R lines were different from those of the heterozygous F1 hybrids, created by crossing susceptible tomatoes with the fixed CU-R lines. A different analytical method was needed. Therefore, sporangia numbers/leaflet and diseased area data were analyzed using a half-normal probability plot and regression analysis. The results of this analysis show its utility for genetic or pathology studies. Considering only populations of the uniform tomato lines, this method confirms the results obtained by using a standard ANOVA, but provides a clearer demonstration of the distributions of the individuals within the populations and how this distribution impacts variance and the difference among the populations. This method also allows a joint analysis of the uniform lines with an additional population that is less uniform, because it is segregating. Such an analysis would be invalid using a standard ANOVA. The results of this joint analysis determined that the additional population was divergent from the fixed CU-R lines, and, against some isolates, against the CLN-R lines as well. Half-normal probability plot analysis method would be applicable more broadly beyond analysis of disease resistance data. It could be useful for data from populations that are not normally distributed, for traits which are affected by epistatic gene action, and could be useful for selection of extremes.  相似文献

Summary Organic (e.g. chitin, green manure, cellulose) amendments to soil induced quantitative and qualitative changes in the rhizosphere microflora of antirrhinum plants infected withVerticillium dahliae Kleb. Whereas reduction in disease severity occurred with chitin and green manure amendments, an increase in disease severity was observed with the application of cellulose. Ethylene evolved during compatible or susceptible disease interactions may hasten and/or worsen disease symptom development; if so, the prevention of disease-response ethylene should reduce disease symptoms. We have examined the effects of reduced ethylene synthesis on Verticillium wilt (causal organism, Verticillium dahliae) of tomato by transforming tomato with ACC deaminase, which cleaves ACC, the immediate biosynthetic precursor of ethylene in plants. Three promoters were used to express ACC deaminase in the plant: (i) CaMV 35S (constitutive expression); (ii) rolD (limits expression specifically to the site of Verticillium infection, i.e. the roots); and (iii) prb-1b (limits expression to certain environmental cues, e.g. disease infection). Significant reductions in the symptoms of Verticillium wilt were obtained for rolD- and prb-1b-, but not for 35S-transformants. The pathogen was detected in stem sections of plants with reduced symptoms, suggesting that reduced ethylene synthesis results in increased disease tolerance. The effective control of formerly recalcitrant diseases such as Verticillium wilt may thus be obtained by preventing disease-related ethylene production via the tissue-specific expression of ACC deaminase.  相似文献

Verticillium dahliae (Vd) is an emerging threat to sugar beet production. Control measures such as fungicides are not available and the utilisation of resistant cultivars is very limited. Hence, we explored the potential of two soil-derived fungal biocontrol agents (BCAs), Fusarium oxysporum F2 (FoF2) and Verticillium tricorpus 1808 (Vt1808), against Verticillium wilt of sugar beet. Pathogenicity tests revealed that Vd caused over 90% disease incidence and severity and led to a significant yield reduction, whereas BCAs neither inhibited nor promoted plant growth. Germination rate was higher in BCA-treated seeds compared to untreated ones. Viability of both BCAs was significantly reduced after six months of storage in liquid methylcellulose formulation (MC), while BCA concentrations remained stable on stored seeds treated with MC. In contrast, Vt1808 produced in sand-rye flour formulation remained 100% viable after storage. Similarly, post-storage analysis of the FoF2 talcum powder formulation revealed improved colony forming units, but increments were not significant. In vitro, both BCAs caused no growth inhibition zones and only insignificant Vd growth reductions were observed. In the greenhouse, soil amendment with a higher dose of FoF2:Vt1808 mixture resulted in substantial reductions of disease severity on the crown (33.3%), disease incidence (55.6%) and disease severity (68.8%) on the beet as well as an improved yield (32.9%). In contrast, seed coating did not reduce symptoms on the crown and on the beet. In the field, both BCAs did not provide significant disease reductions. Nevertheless, a promising result was achieved with the application of FoF2. Despite the need for improvement of the biocontrol activity, the results of this study demonstrate the suitability of the optimised BCA formulations for utilisation in commercial sugar beet production.  相似文献

选育和推广抗病品种是防治陆地棉黄萎病的主要措施,为了早日实现多类型、多区域大面积抗病品种的应用,本研究选取107份遗传背景差异较大的种质,利用河北省农林科学院棉花研究所小安舍试验站黄萎病病圃进行了3年黄萎病抗性重复鉴定。鉴定得到抗病品系8个,占7.5%;耐病品种(系)20个,占18.7%。本研究表明,当前被作为育种亲本的抗病品系还太少,需要深入开展抗病遗传机制,以及与其他经济性状协同改良的关系,为陆地棉抗病育种提供理论指导;达到抗病或接近抗病水平的大部分品种(系)来自于海陆野远缘后代,具有外源基因血统,证明了远缘杂交是陆地棉黄萎病抗性改良的有效手段。  相似文献

为探讨人工巨尾桉林叶片挥发物对周边农作物的化感作用,采用不同质量新鲜巨尾桉叶片及由新鲜叶片提取的桉叶油对玉米、辣椒、西红柿等三种植物种子进行处理,观测其种子萌发和幼苗生长状况。结果表明:(1)在三种被测植物中,桉树叶片挥发物对玉米种子萌发影响最小,对辣椒影响最大;(2)当叶片用量小于200g时,桉树叶片挥发物对三种植物种子萌发影响不明显,当叶片用量达400g时,能完全抑制辣椒、西红柿种子萌发,并能极显著降低玉米种子萌发(P0.01);(3)玉米幼苗芽生长随叶片用量的增加呈现先促进后抑制现象,对芽高、鲜重、干重的促进和抑制均达到显著(P0.05)或极显著水平(P0.01);(4)当叶片用量小于或等于100g时,桉树叶片挥发物对辣椒、西红柿幼苗生长影响不明显,当用量达到200g时则能极显著抑制辣椒、西红柿幼苗生长;(5)桉叶油对三种测试植物的抑制效果与叶片自然挥发物相似,且效果更显著。  相似文献
