A series of experiments were carried out to construct an energy budget for juvenile thick lipped mullet, Crenimugil labrosus Risso. A partial factorial experimental design was used to examine the effects of temperature, fish size and meal size on growth. The maximum ration that the fish were able to ingest completely per day was found to be 0·8, 1·4 and 2·3% wet body weight (b.w.) at 13,18 and 23°C, respectively. Ingested maintenance requirements (M.R.) were estimated to be 137, 205 and 288 cal fish-1 day-1 at 13, 18 and 23°C, respectively. At 18deg; C, M.R. varied as 25 W1.04 cal day-1, where W= fish weight (g). Growth rate increased with increasing temperature. Maximal conversion efficiency was 21–24% and was achieved closer to the maximum ingested ration with increasing temperature. The relationship between respiration rate and W at 18deg; C for 3-20 g fish is described by: respiration rate (ml O2 h-1) = 0·128 W0.976 The energy cost of apparent specific dynamic action at 18deg; C was found to vary between 5·1% and 23·6% of the calorific value of the ingested meal (1% wet b.w.) , mean (± S.E.)=10·2 ± 2·0%. Post mortem analyses of groups of fish fed 0·2, 0·8 and 1·5% wet b.w. meals showed a significant increase in total lipid and a significant decrease in water content with increasing ratio size. A negative correlation was found between body water content and total lipid (and calories). The mean assimilation efficiency (±s.e.) for 5–10 g mullet at 18deg; C was 73·9 ± 3·6%. The observations reported in this study were brought together to construct an energy budget for juvenile C. labrosus which was found to give a reliable prediction (within 10%) of energy demand and growth under the prevailing experimental conditions. Both gross (K1) and net (K2) growth efficiencies, based on energy values, increased with increasing ratio size up to satiation and were independent of temperature. The maximum values of K1 and K2 observed were 0·33 and 0·46, respectively. The third order efficiency (K3) appeared to be independent of temperature and ration size; mean values ranged between 0·66 and 0·84.  相似文献

Diatoms in the top metre of lake sediment have been studied for indications or recent changes in the environment of Loch Leven. The diatoms indicate a eutrophic, alkaline lake with Melosira italica ssp. subarctica, Tabellaria Flocculosa and Stephanodiscus astraea the dominant taxa at the base of the 1-m section. There is then an appeance of Melosira ambigua and Stephanodiscus hantzschii suggesting a richer environment. Post A.D. 1816 changes in taxa suggest possible correlation with the lowering of lake level in 1830 and the dominance of Diatoma elongatum, Cyclotella pseudostelligera, Stephanodiscus astraea var. minutula, S. tenuis, S. hantzschii, and Synedra rumpens and var. soctica in zone 4 agrees with present-day plankton records.  相似文献

This is the first study investigating the plant–herbivore interaction between Sarpa salpa, which has overgrazed seagrass transplants in Portugal, and the seagrasses Cymodocea nodosa, Zostera marina and Zostera noltii, which have been considered for restoration. When offered the choice between the three seagrasses in outdoor tanks, adult S. salpa clearly preferred Z. noltii. Testing the seagrasses separately, mean ± s.d. feeding rates ranged from 21 ± 11 g seagrass fresh mass kg?1 fish mass day?1 for Z. marina to 32 ± 9 g seagrass fresh mass kg?1 fish mass day?1 for C. nodosa and 40 ± 11 g seagrass fresh mass kg?1 fish mass day?1 for Z. noltii (temperature = 16° C). Food‐processing rate in S. salpa did not differ between seagrasses, and there was no evidence of a regulation of processing rate according to food intake. Seagrasses differed substantially in nitrogen content and C:N, with C. nodosa containing the highest nitrogen content and lowest C:N (2·5 ± 0·1% and 14·0 ± 1·0), followed by Z. noltii (2·1 ± 0·1% and 17·0 ± 1·0) and Z. marina (1·4 ± 0·1% and 26·0 ± 2·0). Food‐processing rate in S. salpa and the nutritional value of the seagrasses were not correlated with the observed feeding preference and rate. The study suggests that C. nodosa and Z. marina are less at risk of overgrazing by S. salpa and might thus be preferable to Z. noltii for seagrass restoration in areas with noticeable abundances of this fish.  相似文献

Feeding rates of Brachionus plicatilis were studied for two types of food — algae Monochrysis lutheri and baker's yeast Saccharomyces cerevisae. The main regularities of changes in filtration rate and ration were studied in small culture volumes (1 ml) for adult amictic females depending on food concentration (1, 2, 4, 8 and 16 · 106 cells · ml−1), ambient temperature (16 and 26 °C), and salinity (5, 10, 15, 20, 25 and 30 ppt). B. plicatilis ration did not depend on the salinity, but was largely determined by temperature and food concentration. It was found that at 16 and 26 °C the dependence of the ingestion rate (ration) on food concentration differed greatly. A hypothesis was suggested to explain this phenomenon. A critical concentration of both types of food at which the increase in the rotifer ration ceased is 4 · 106 cells · ml−1. This is the minimum “background” food concentration for B. plicatilis mass cultivation. The average rations measured at the concentration of M. lutheri and S. cerevisae of 4 · 106 cells · ml−1 where 1.3 ± 0.1 and 4.8 ± 1.3 μg dry weight. · ind−1 · day−1 at 26 °C and 0.54 ± 0.1 and 1.9 μg d. w. · ind−1 · day−1 at 16 °C, respectively. The rations obtained in the laboratory were corrected for the conditions of rotifer commercial production in the open field in summer time. The correct values were 0.86 and 0.72 μg d. w. · ind−1 · day−1 for algae and yeast, respectively.  相似文献

This study was undertaken to examine the long‐term effects of photoperiod, temperature and their interaction on growth, gill Na+,K+‐ATPase (NKA) activity, seawater tolerance and plasma growth‐hormone levels in Atlantic salmon Salmo salar pre‐smolts and smolts. The fish (mean ± s.e . initial body mass = 15·9 ± 0·4 g) were reared on two photoperiods (continuous light, LL, and simulated natural photoperiod, LDN, 60° 25′ N) and two temperatures (8·3 and 12·7° C) from June to May of the following year. Mean body mass was affected by photoperiod, temperature and their interactions. Both temperature groups on LL developed peak levels in gill NKA activity from October to November, 4–5 months prior to the natural season for the parr–smolt transformation. Fish at 12° C showed peak levels in NKA activity 4–6 weeks before the fish at 8° C. Fish in all four experimental groups showed maximum NKA activity within a similar size range (113–162 g). The present findings further indicate that smoltification in S. salar is to some extent driven by size, and that S. salar will develop smolt characteristics, e.g. a marked increase in NKA activity, within a similar size range. Faster‐growing S. salar will, thus, reach this size threshold at a relatively younger age.  相似文献

Three diatom species, Stephanodiscus hantzschii (Ehr.) Grun., Asterionella formosa Hass. and Fragilaria crotonensis Kitt. Hass. were isolated from Lake Maarsseveen where they are dominant and show a successional sequence. The physiological responses of each species to temperature and limitation by silicon and phosphorus were determined over the temperature range of 5° to 20° C using short-term batch culture methods. Stephanodiscus hantzschii had higher maximum growth rates than the other two species at all temperatures, and the maximum growth rates of all species increased with increasing temperature. Temperature affected not only maximum growth rates but also half-saturation constants (Ks) and the minimum cell quotas. S. hantzschii had low silicon requirements for growth under Si-limiting conditions, and A. formosa and F. crotonensis had higher and nearly identical silicon requirements. The Ks values for silicon for S. hantzschii were essentially constant from 5° to 20° C but varied greatly for the other two species. A. formosa had the lowest requirements for growth under phosphorus limitation, F. crotonensis was intermediate and S. hantzschii had the highest growth requirements for phosphorus. The K1 values for phosphorus were constant over the temperature range for both A. formosa and F. crotonensis and were much higher and variable for S. hantzschii. Nutrient competition experiments were performed in continuous cultures at four temperatures and various Si:P ratios. The results generally, but not always, confirmed the predictions based on the Monod relationships for each species. Results not in agreement with predictions were usually because of similar physiological properties of A. formosa and F. crotonensis or because of decreased loss rates for F. crotonensis due to wall growth. In cultures with all three species phosphorus-limited (Si:P > 75), A. formosa often dominated as predicted, although F. crotonensis was sometimes the most abundant species. As predicted, S. hantzschii never dominated at high Si:P ratios. At intermediate Si:P ratios when A. formosa and F. crotonensis were both Si-limited and S. hantzschii P-limited, all three species coexisted because A. formosa and F. crotonensis have almost identical silicon requirements, although sometimes F. crotonensis was more abundant than predicted. At 10°C the results agreed best with the predictions; A. formosa dominated at high Si:P ratios and S. hantzschii dominated as predicted at low Si:P ratios when all three species were Si-limited.  相似文献

Uptake and assimilation of nitrogen and phosphorus were studied in Olisthodiscus luteus Carter. A diel periodicity in nitrate reductase activity was observed in log and stationary phase cultures; there was a 10-fold difference in magnitude between maximum and minimum rates, but other cellular features such as chlorophyll a, carbon, nitrogen, C:N ratio (atoms) · cell?1 were less variable. Ks values (~2 μM) for uptake of nitrate-N and ammonium-N were observed. Phosphorus assimilated · cell?1· day?1 varied with declining external phosphorus concentrations; growth rates <0.5 divisions · day?1 were common at <0.5 μM PO4-P. Phosphate uptake rates (Ks= 1.0–1.98 μM) varied with culture age and showed multiphasic kinetic features. Alkaline phosphatase activity was not detected. Comparisons of the nutrient dynamics of O. luteus to other phytoplankton species and the ecological implications as related to the phytoplankton community of Narragansett Bay (Rhode Island) are discussed.  相似文献

In roach Rutilus rutilus growth ceases below a temperature threshold of 12° C. This cessation of growth is accompanied by a reduction in feeding. Do roach decrease feeding in the cold because of reduced energy demand, caused by the decelerating effect of low temperature on metabolism and growth, or is feeding directly limited by low temperatures, leading to reduced growth rates? It was found that at low temperatures the intake and digestion of food may be limited by reduced activities of digestive enzymes. Trypsin, amylase and γ‐glutamyl transferase showed a negative compensation with respect to temperature, resulting in very low activities at acclimation temperatures of ≤12° C. Trypsin activity, falling from 400·5 ± 131·2 U g?1 fresh mass of the gut at 27° C to 12·5 U g?1 fresh mass at 4° C, displayed the strongest linear correlation with growth rates, suggesting that trypsin activities may set a limit to growth in the low temperature range. If protein digestion is limiting at low temperatures, this should be reflected in reduced concentrations of amino acid in the white muscle. The size of the total amino acid pool was not affected by temperature acclimation and ranged between 19·2 ± 6·2 and 25·2 ± 3·6 µmol g?1 fresh mass of the white muscle. A decrease, however, was found of several amino acids, mainly of threonine and glutamine, in the low temperature range. Low concentrations of the essential amino acid threonine (0·14 ± 0·03 µmol g?1 fresh mass at 12° C and 0·12 ± 0·05 µmol g?1 fresh mass at 4° C) were probably due to nutritional or digestional limitations and may therefore have resulted from reduced trypsin activity in the cold. The non‐essential amino acid glutamine, however, can be endogenously synthesized and its low level observed at 4° C (0·16 ± 0·09 µmol g?1 fresh mass) was not necessarily a result of low trypsin activities. It is more likely that low temperatures impair glutamine synthesis. The possibility that glutamine concentrations may be down regulated under conditions when anabolic processes are not advantageous is discussed.  相似文献

1. Growth rates of seven species of planktonic algae were determined in culture over a range of temperature from 2 to 35 °C. Additional observations on growth and viability were made for 13 species in the temperature range 20–35 °C. 2. There was a wide range of growth rates between species at their optimal temperatures, from 1.7 divisions day?1 (Asterionella formosa) to 0.3 divisions day?1 (Ceratium furcoides). 3. There were considerable differences between species for growth at low and high temperature. Certain algae, including the diatom A. formosa and the flagellates Cryptomonas marssonii, Dinobryon divergens and Eudorina unicocca var. unicocca, had growth rates of 0.4 divisions day?1 or more at 5 °C. The cyanophyte Tychonema (formerly Oscillatoria) bourrellyi, the xanthophyte Tribonema sp., the desmid Staurastrum cingulum and the large dinoflagellate C. furcoides grew poorly or not at all at this temperature. All 21 species tested could grow at 25 °C, but many – including most of the diatoms, some cyanophytes, and all the flagellates – failed to grow persistently at 30 °C. Only Aphanizomenon flosaquae survived with moderate increase at 35 °C, a lethal temperature for the other species. 4. The applicability was considered of proposed quantitative formulations of the rate‐temperature relationship. Simple exponential relationships applied only to very limited lower ranges of temperature. The relationship proposed by B?lehrádek was a better fit over a wider temperature range, but still excluded rate‐decline at high temperature. 5. The interspecific differences found are of potential significance for restrictions in natural distributions associated with season, altitude (especially above 500 m) and latitude.  相似文献

Telemetered heart rate (fH) was examined as an indicator of activity and oxygen consumption rate (VO2) in adult, cultivated, Atlantic salmon, Salmo salar L. Heart rate was measured during sustained swimming in a flume for six fish at 10° C [mean weight, 1114 g; mean fork length (f. l.), 50·6 cm] and seven fish at 15° C (mean weight, 1119 g; mean f. l., 50·7 cm) at speeds of up to 2·2 body lengths/s. Semi–logarithmic relationships between heart rate and swimming speed were obtained at both temperatures. Spontaneously swimming fish in still water exhibited characteristic heart rate increases associated with activity. Heart rate and Vo2 were monitored simultaneously in a 575–1 circular respirometer for six fish (three male, three female) at 4° C (mean weight, 1804 g; mean F. L., 62· cm) and six fish (three male, three female) at 10° C (mean weight, 2045 g; mean f. l., 63·2 cm) during spontaneous but unquantified activity. Linear regressions were obtained by transforming data for both fH and Vo2 to log values. At each temperature, slopes of the regressions between fH and Vo2 for individual fishes were not significantly different, but in some cases elevations were. All differences in elevation were between male and female fish. There were no significant differences in regression slope or elevation for fish of the same sex at the two temperatures and so regressions were calculated for the sexes, pooling data from 4 and 10° C. There was no significant difference in the mean ± S. D. Vo2 between the sexes at 4° C (male, 66·0 ± 59·6 mgO2 kg?1 h?1; female, 88·0 ± 60·1 mgO2 kg?1 h?1) or 10° C (male, 166·2 ± 115·4 mgO2 kg?1 h?1; female, 169·2 ± 111–1 mgO2 kg?1h?1). Resting Vo2 (x?± s. d.) at 4°C was 36·7 ± 8.4 mgO2 kg?1 h?1, and 10° C was 72·8 ± 11·9 mgO2 kg?1 h?1. Maximum Vo2 (x?± S. D.) at 4° C was 250·6 ± 40·2 mgO2 kg?1 h?1, and at 10° C was 423·6 ± 25·2 mgO2 kg?1 h?1. Heart rate appears to be a useful indicator of metabolic rate over the temperature range examined, for the cultivated fish studied, but it is possible that the relationship for wild fish may differ.  相似文献

This study provides the first measurements of the standard respiration rate (RS) and growth dynamics of European sardine Sardina pilchardus larvae reared in the laboratory. At 15° C, the relationship between RS (µl O2 individual?1 h?1) and larval dry mass (MD, µg) was equal to: RS = 0·0057(±0·0007, ± s.e.)·MD0·8835(±0·0268), (8–11% MD day?1). Interindividual differences in RS were not related to interindividual differences in growth rate or somatic (Fulton's condition factor) or biochemical‐based condition (RNA:DNA).  相似文献

The rate of emergence of micropredatory gnathiid isopods from the benthos, the proportion of emerging gnathiids potentially eaten by Labroides dimidiatus, and the volume of blood that gnathiids potentially remove from fishes (using gnathiid gut volume) were determined. The abundance (mean ±s.e .) of emerging gnathiids was 41·7 ± 6·9 m?2 day?1 and 4552 ± 2632 reef?1 day?1 (reefs 91–125 m2). The abundance of emerging gnathiids per fish on the reef was 4·9 ± 0·8 day?1; but excluding the rarely infested pomacentrid fishes, it was 20·9 ± 3·8 day?1. The abundance of emerging gnathiids per patch reef was 66 ± 17% of the number of gnathiids that all adult L. dimidiatus per reef eat daily while engaged in cleaning behaviour. If all infesting gnathiids subsequently fed on fish blood, their total gut volume per reef area would be 17·4 ± 5·6 mm3 m?2 day?1; and per fish on the reefs, it would be 2·3 ± 0·5 mm?3 fish?1 day?1 and 10·3 ± 3·1 mm3 fish?1 day?1 (excluding pomacentrids). The total gut volume of gnathiids infesting caged (137 mm standard length, LS) and removed from wild (100–150 mm LS) Hemigymnus melapterus by L. dimidiatus was 26·4 ± 24·6 mm3 day?1 and 53·0 ± 9·6 mm3 day?1, respectively. Using H. melapterus (137 mm LS, 83 g) as a model, gnathiids had the potential to remove, 0·07, 0·32, 0·82 and 1·63% of the total blood volume per day of each fish, excluding pomacentrids, caged H. melapterus and wild H. melapterus, respectively. In contrast, emerging gnathiids had the potential of removing 155% of the total blood volume of Acanthochromis polyacanthus (10·7 mm LS, 0·038 g) juveniles. That L. dimidiatus eat more gnathiids per reef daily than were sampled with emergence traps suggests that cleaner fishes are an important source of mortality for gnathiids. Although the proportion of the total blood volume of fishes potentially removed by blood‐feeding gnathiids on a daily basis appeared to be low for fishes weighing 83 g, the cumulative effects of repeated infections on the health of such fish remains unknown; attacks on small juvenile fishes, may result in possibly lethal levels of blood loss.  相似文献

The growth of Nile tilapia Oreochromis niloticus (0·02–20·00 g) was measured when fed to excess during the hours of light, following their exposure to five thermal regimes fluctuating around the thermal optimum for growth (Topt = 30° C) over the diel cycle of day (light, L) and night (dark, N), i.e. 27° C(L):33° C(N), 28·5° C(L):31·5° C(N), 30° C(L):30° C(N), 31·5° C(L):28·5° C(N) and 33° C(L):27° C(N) (two replicates per treatment, six weeks' rearing, growth measurements at weekly intervals). A model constructed with a stepwise multiple‐regression analysis accounted for 87·4% of the variation of the specific growth rate (G, % M day?1) from the variations of wet mass (M), the extent of the thermal fluctuation (FT) and their interactions, i.e. log10G = 1·7686 ? 0·2136 log10M + 0·0806 [log 10M× log 10 (1 + FT)] ? 0·0394 [log10M× log 10 (1 + FT)]2. Based on this model, the thermal fluctuation that produces the fastest growth ( ,°C) decreases in a curvilinear way, from 5·1° C at 20 mg to c. 0·7° C at 20 g. Thermal regimes that produce the slowest growth also produce the highest size heterogeneity. Functional hypotheses behind the size‐dependent effects of thermal fluctuations are discussed, together with their implications in natural habitats and aquaculture systems with in different contexts of food availability.  相似文献

Duckweed, Lemna gibba, was grown in 12 m2 shallow ponds in the Negev desert, during 12 months of continuous cultivation, beginning April 1984. Average monthly growth rates varied with the season of the year. The lowest daily yield, 2·6±0·4 g dry weight m−2 day−1, was obtained during January. Highest daily yields, 7·9±2·6 g dry weight m−2 day−1 and 7·0±1·2 g dry weight m−2 day−1, were obtained during September and May. A 35% decline of the yield was seen during midsummer (July), 4·8±1·2 g dry weight m−2 day−1. The average rate for the year was 5·15±1·7 g dry weight m−2 day−1. The protein content of the plants ranged from 30 to 38% per unit dry weight.Growth performance is discussed in relation to the prevailing climatic conditions.  相似文献

Cryptomonas erosa Skuja, a planktonic alga, was grown in batch culture at different combinations of light intensity and temperature, under nutrient saturation. Growth was maximal (1.2 divisions · day?1) at 23.5 C and 0.043 ly · min?1, declining sharply with temperature (0.025 divisions-day?1 at 1 C). With decreasing temperature, the cells showed both light saturation and inhibition at much reduced light intensities. At the same time the compensation light intensity for growth declined towards a minimum of slightly above 0.4 × 10?4 ly · min?1 (~1 ft-c) at 1 C or <0.1 ly · day?1 (PAR). Cell division was more adversely affected by low temperature than carbon uptake, and the resulting excess production of photosynthate was both stored and excreted. Extreme storage of carbohydrates resulted in cell volumes and carbon content ca. 22 and 30 × greater, respectively, than the maxima observed for cells incubated in the dark, whereas, at growth inhibitory light levels, as much as 57% of the total assimilated carbon was excreted. A marked increase in cell pigment was observed at the lowest light levels (<10?3 ly · min?1), at high temperature. The growth response of C. erosa in culture provides insight into the abundance and distribution of cryptomonads and other small algal flagellates in nature.  相似文献

The growth of sibling dorada Brycon moorei (Characidae) housed individually in small enclosures (0·25 l; 27·0° C, range ±0·5° C; 12L:12D) from the egg stage was examined at regular intervals until 36 days after hatching (dah) and compared with their behaviour. From 1 to 8 dah, when cannibalism is intense among B. moorei raised in groups, there was no significant increase of size heterogeneity among isolated fish (c.v . of total length of 3·1 and 3·6%, at 1 and 8 dah, respectively) and no primacy of early size differences either. These results suggest that cannibals of B. moorei raised in groups are not natural‐born killers with greater growth capacities than others. Size heterogeneity among isolated fish increased significantly first when B. moorei were weaned on formulated feed (8–15 dah), then again from 24 to 36 dah when the average growth rate was half as fast as before (c. 0·5 v. 1·0 mm day?1), despite fish consistently feeding. During both periods, there was a significant, positive relationship between individual growth and aggression or boldness. These results suggest that (1) boldness can favour the transition to a new food type and (2) fish exhibited a variable responsiveness to spatial restriction in small enclosures, which may have been alleviated in some individuals by establishment of territorial behaviour, as suggested by their enhanced aggression.  相似文献

In the autumn of 2007, marine mucilage caused by the diatom Coscinodiscus granii occurred in the central area of Ariake Sound, Japan, and resulted in damage to fishery. To elucidate the mechanism underlying the outbreak of marine mucilage, we examined the effect of temperature on cell growth and production of transparent exopolymer particles (TEPs) in a culture of this species. Growth and TEP production of C. granii are influenced by temperature. The maximum growth rate (1.63 divisions day−1) and cell yield (1,280 cells mL−1) at all temperatures were obtained at 30°C. Optimal growth rates (>1.15 divisions day−1: ca. 70% of maximum) and cell yield (>900 cells mL−1: ca. 70% of maximum) were observed at temperatures of 25–30°C. TEP production by C. granii depended on whether volume- or cell-related values were considered. The maximum volume-normalized increase rates and concentrations of TEP at all temperatures were observed at 25°C. However, when production rates and concentrations of TEP were normalized to cell numbers, optimal values were measured at 10–15°C. In Ariake Sound, when marine mucilage caused by C. granii occurred, the temperature ranged from 25.0 to 25.4°C. This suggests that growth conditions of C. granii are important factors for production of marine mucilage.  相似文献

The skeletal dry weight of the 4.4 ± 0.2 cm size class of Asterias rubens L. from Kiel Bay in the western Baltic is 0.34 ± 0.08 g. The sum of calcium and magnesium carbonates in the skeleton amounts to 94.0 ± 1.3% while the individual concentrations are 86.9 ± 1.3% CaCO3 and 7.1 ± 0.7% MgCO3. The MgCO3 is 4% lower than expected for a magnesium calcite precipitated under Baltic temperature conditions (8°C). Strontium was not determined but strontium carbonate is known to be in the region of 0.4%. The remainder is organic matter and this gives rise to 2.9 ± 1.3% organic carbon.Animals studied were at the ‘waiting stage’ and their actual growth was minimal. Uptake of 45Ca in the skeleton consists of a fast step followed by a slow step. The fast step is attributed to saturation of exchangeable skeletal pools while the slow step is due to net deposition of CaCO3. Skeletal growth at the waiting stage calculated from the second rate constant was found to be 0.76 μg CaCO3j-mg skeleton?1 · day?1 or 0.09% · day?1 compared with 9.3 μg CaCO3 · mg skeletont1&#x0304; · day?1 or 1.1% · day?1 at log phase. The isotope method is considered superior to size-frequency analysis in that it is capable of detecting differences in growth rate in individuals of the same size class and thus provides an insight into asteroid population structure.  相似文献

Aims: The purification and biochemical properties of the 1,4‐β‐xylosidase of an oenological yeast were investigated. Methods and Results: An ethanol‐tolerant 1,4‐β‐xylosidase was purified from cultures of a strain of Pichia membranifaciens grown on xylan at 28°C. The enzyme was purified by sequential chromatography on DEAE cellulose and Sephadex G‐100. The relative molecular mass of the enzyme was determined to be 50 kDa by SDS‐PAGE. The activity of 1,4‐β‐xylosidase was optimum at pH 6·0 and at 35°C. The activity had a Km of 0·48 ± 0·06 mmol l?1 and a Vmax of 7·4 ± 0·1 μmol min?1 mg?1 protein for p‐nitrophenyl‐β‐d ‐xylopyranoside. Conclusions: The enzyme characteristics (pH and thermal stability, low inhibition rate by glucose and ethanol tolerance) make this enzyme a good candidate to be used in enzymatic production of xylose and improvement of hemicellulose saccharification for production of bioethanol. Significance and Impact of the Study: This study may be useful for assessing the ability of the 1,4‐β‐xylosidase from P. membranifaciens to be used in the bioethanol production process.  相似文献

This study establishes the bioenergetics budget of juvenile whitespotted bamboo shark Chiloscyllium plagiosum by estimating the standard metabolic rate (RS), measuring the effect of body size and temperature on the RS, and identifying the specific dynamic action (RSDA) magnitude and duration of that action in juvenile whitespotted bamboo sharks. The mean ±s .d . (RS) of six fish (500–620 g) measured in a circular closed respirometry system was 30·21 ± 5·68 mg O2 kg?1 h?1 at 18° C and 70·38 ± 14·81 mg O2 kg?1 h?1 at 28° C, respectively. There were no significant differences in RS between day and night at either 18 or 28° C (t‐test, P > 0·05). The mean ±s .d . Q10 for 18–28° C was 2·32 ± 0·06 (n = 6). The amount of oxygen consumed per hour changed predictably with body mass (M; 295–750 g) following the relationship: (n = 40, r2= 0·92, P < 0·05). The mean magnitude of RSDA was 95·28 ± 17·55 mg O2 kg?1 h?1. The amount of gross ingested energy (EI) expended as RSDA ranged from 6·32 to 12·78% with a mean ±s .d . of 8·01 ± 0·03%. The duration of the RSDA effect was 122 h. The energy content of juvenile whitespotted bamboo shark, squid and faeces determined by bomb calorimeter were 19·51, 20·3 and 18·62 kJ g dry mass?1. A mean bioenergetic budget for juvenile whitespotted bamboo sharks fed with squid at 18° C was 100C = 29·5G + 31·9RS+ 28·2RSDA+ 6·7F + 2·1E + 1·6U, where C = consumption, G = growth, F = egestion, E = excretion and U = unaccounted energy.  相似文献
