The photophysiological properties of strain RCC 237 belonging to the marine picoplanktonic genus Picochlorum, first described by Henley et al., were investigated under different photon flux densities (PFD), ranging from 40 to 400 μmol photons· m?2·s?1, mainly focusing on the development of the xanthophyll cycle and its relationship with the nonphotochemical quenching of fluorescence (NPQ). The functioning of the xanthophyll cycle and its photoprotective role was investigated by applying a progressive increase of PFD and using dithiotreitol and norflurazon to block specific enzymatic reactions in order to study in depth the relationship between xanthophyll cycle and NPQ. These two processes were significantly related only during the gradually increasing light periods and not during stable light periods, where NPQ and zeaxanthin were decoupled. This result reveals that NPQ is a photoprotective process developed by algae only when cells are experiencing increasing PFD or in response to stressful light variations, for instance after a sudden light shift. Results showed that the photobiological properties of Picochlorum strain RCC 237 seem to be well related to the surface water characteristics, as it is able to maintain its photosynthetic characteristics under different PFDs and to quickly activate the xanthophyll cycle under high light.  相似文献

Diatom cell quantity and their biochemical composition vary among species and are greatly affected by harvest stage or culture conditions. This study compares growth pattern, cell attachment, and biochemical composition of four diatoms suitable for abalone post-larvae: Navicula incerta, Proschkinia sp., Nitzschia sp., and Amphora sp. The four diatoms were grown in F/2 medium at 28.5?±?1.4°C, under 62?±?8?μmol?photons?m?2?s?1, at different original inoculating densities (0.05?×?106, 0.10?×?106, and 0.25?×?106?cells?mL?1) and were harvested in log and stationary phase of growth for biochemical analysis. Total protein, carbohydrate, lipid, and ash composition, as well as fatty acid composition, were determined. All diatoms grew better when inoculated at 0.10?×?106?cell?mL?1 with Proschkinia sp. reaching the highest cell density of 6.56?×?106?cells?mL?1 in log phase. Amphora sp. had the highest cell attachment capacity when inoculated at 0.10?×?106?cell?mL?1 (11,580?cells?mm?2), whereas N. incerta had the lowest (7,750?cells?mm?2). Protein and lipid (percent dry weight) contents were generally highest in cells during log phase of growth; Amphora sp. in log phase of growth had the highest lipid content of 9.74% DW, whereas significant differences in carbohydrate between the two growth phases were only observed for Proschkinia sp. Besides, all diatoms had higher energy contents in log phase of growth. There were no significant differences in ash content among the four diatoms. Polyunsaturated fatty acid (PUFA) content ranged from 23.25% to 38.62% of the total fatty acids, and the four diatoms tested were richer in n-3 PUFA than in n-6 PUFA. All the diatoms had significant quantities of 20:5n-3 (EPA) (between 12.69% and 17.68% of TFA), and Proschkinia sp., in log phase of growth, had the highest quantity of arachidonic acid (20:4n-6; ARA). The results highlight the influence of culture conditions and harvest protocols on diatom nutritive value and enabled a preliminary approach towards the selection of novel diatom species.  相似文献

Filter fractionated picophytoplankton from Antarctic coastal waters (summer 2001) represented only 7–33% of total phytoplankton, even though total stocks were low (average Chl = 0.32 μg l−1, range = 0.13–1.03 μg l−1). Though all cells passed a 2 μm filter, electron microscopy revealed most cells were over 2 μm, principally Parmales, Phaeocystis sp., and small diatoms. CHEMTAX analysis of HPLC pigment data suggested type 8 haptophytes (e.g. Phaeocystis sp. plus Parmales and pelagophytes) contributed 7–58% of picoplanktonic chlorophyll a, type 6 haptophytes (e.g. coccolithophorids) 18–59%, diatoms 0–18% (mostly type 2 diatoms, e.g. Pseudonitzschia sp., 0–15%), prasinophytes 0–17%, with cell fragments of cryptophytes 0–40%, and dinoflagellates 0–11%. Only stocks of type 8 haptophytes and prasinophytes differed significantly due to successional changes. Zeaxanthin concentrations exceeded estimates from previous cyanobacterial counts and may derive from non-photosynthetic bacteria.  相似文献

To better understand the impact of ocean acidification (OA) and changes in light availability on Southern Ocean phytoplankton physiology, we investigated the effects of pCO2 (380 and 800 µatm) in combination with low and high irradiance (20 or 50 and 200 µmol photons m?2 s?1) on growth, particulate organic carbon (POC) fixation and photophysiology in the three ecologically relevant species Chaetoceros debilis, Fragilariopsis kerguelensis and Phaeocystis antarctica. Irrespective of the light scenario, neither growth nor POC per cell was stimulated by OA in any of the tested species and the two diatoms even displayed negative responses in growth (e.g. C. debilis) or POC content (e.g. F. kerguelensis) under OA in conjunction with high light. For both diatoms, also maximum quantum yields of photosystem II (Fv/Fm) were decreased under these conditions, indicating lowered photochemical efficiencies. To counteract the negative effects by OA and high light, the two diatoms showed diverging photoacclimation strategies. While cellular chlorophyll a (Chl a) and fucoxanthin contents were enhanced in C. debilis to potentially maximize light absorption, F. kerguelensis exhibited reduced Chl a per cell, increased disconnection of antennae from photosystem II reaction centers and strongly lowered absolute electron transport rates (ETR). The decline in ETRs in F. kerguelensis might be explained in terms of different species‐specific strategies for tuning the available flux of adenosine triphosphate and nicotinamide adenine dinucleotide phosphate. Overall, our results revealed that P. antarctica was more tolerant to OA and changes in irradiance than the two diatoms, which may have important implications for biogeochemical cycling.  相似文献

The facultative, heterotrophs Cyclotella cryptica (sclone WT-1-8) and Coscinodiscus sp. were selectively isolated from coastal waters by dark incubation of organically enriched solid medium. C. cryptica grows in the dark with glucose and galactose, and Coscinodiscus sp. with glucose. Clone WT-1-8 of C. cryptica grows about twice as fast with glucose as a previously studied clone (0-3A). In the dark with, 5 × 10?6 M glucose C. cryptica divides every 3 days, while with 5 × 10?5 M glucose Coscinodiscus sp. divides every 10 days. Heterotrophic growth of either diatom for 1 year does not cause a major reduction in carbon, nitrogen, chlorophyll a, and chlorophyll c contents, or in photosynthetic ability, compared to light-grown cells. It is possible that facultative heterotrophy is of ecological benefit to these diatoms, probably for slow growth and survival during extended periods of dim light or darkness.  相似文献

Diatoms are a group of highly abundant and diverse aquatic algae species. They contain high lipid content along with many bioactive compounds that can be exploited for biotechnological applications. Despite these attractive attributes, diatoms are underrepresented in production projects due to difficulties in their cultivation. To optimize the growth of three freshwater diatom isolates, Cyclotella sp., Synedra sp. and Navicula sp., an orthogonal assay on N, P, Si and Fe, as well as temperature and pH, was designed using traditional single‐factor tests. We also studied the effect of using nanosilica as an alternate Si source on growth and found that the diatom isolates studied achieved their highest growth rates under different combinations of nutrient and environmental conditions. Silica had the greatest influence on growth, followed by phosphate and iron. The optimized growth conditions for Synedra sp. were N: 30 mg L?1, P: 3 mg L?1, Si: 14.8 mg L?1, Fe: 0.448 mg L?1, temperature 25°C and pH 8. For Navicula sp.: N: 20 mg L?1, P: 2.5 mg L?1, Si: 19.7 mg L?1, Fe: 0.112 mg L?1, temperature 30°C and pH 7.5–8. For Cyclotella sp.: N: 20 mg L?1, P: 2.5 mg L?1, Si: 19.7 mg L?1, Fe: 0.448 mg L?1, temperature 30°C and pH 7.5–8. Nano silica negatively affected growth in Navicula sp. and Cyclotella sp., but no such effect was observed in Synedra sp. Fatty acid profiling showed C16:0, C16:1(n ? 7), C18:0 and C20:5(n ? 3) as major fatty acids, with no significant differences in fatty acid methyl ester profiles between traditional and modified media. This work gives us a new insight into the growth requirements of freshwater diatom species, which are less studied than marine species.  相似文献

The aim of the work was to find the optimal photon irradiance for the growth of green cells of Haematococcus pluvialis and to study the interrelations between changes in photochemical parameters and pigment composition in cells exposed to photon irradiances between 50 and 600?µmol?m?2?s?1 and a light:dark cycle of 12:12?h. Productivity of cultures increased with irradiance. However, the rate of increase was higher in the range 50–200?µmol??2?s?1. The carotenoid content increased with increasing irradiance, while the chlorophyll content decreased. The maximum quantum yield of PSII (Fv/Fm) gradually declined from 0.76 at the lowest irradiance of 50?µmol??2?s?1 to 0.66 at 600?µmol??2?s?1. Photosynthetic activity showed a drop at the end of the light period, but recovered fully during the following dark phase. A steep increase in non-photochemical quenching was observed when cultures were grown at irradiances above 200?µmol??2?s?1. A sharp increase in the content of secondary carotenoids also occurred above 200?µmol?m?2?s?1. According to our results, with H. pluvialis green cells grown in a 5-cm light path device, 200?µmol??2?s?1 was optimal for growth, and represented a threshold above which important changes in both photochemical parameters and pigment composition occurred.  相似文献

Daily integrals of photosynthesis by a cyanobacterial bloom in the Baltic Sea, during the summer of 1993, were calculated from the vertical distributions of light, temperature and the organisms in the water column and from photosynthesis/irradiance curves of picoplanktonic and diazotrophic cyanobacteria isolated from the community. The distribution of chlorophyll a in size-classes <20?µm and >20?µm was monitored over 9 days that included a deep mixing event followed by calm. Picocyanobacteria formed 70% of the cyanobacterial biomass and contributed 56% of the total primary production. Of the filamentous diazotrophs that formed the other 30%, Aphanizomenon contributed 28% and a Nodularia-containing fraction 16% of the primary production. For the whole population there was little change in standardized photosynthetic O2 production, which remained at about 31?mmol?m?2 before and after the mixing event. There were differences, however, between the classes of cyanobacteria: in picocyanobacteria primary production hardly changed, while in Aphanizomenon it increased by 2.6 and in Nodularia it fell below zero. Total phytoplankton photosynthesis was strongly dependent on total daily insolation with the compensation point at a photon insolation of 22.7?mol?m?2?d?1. Similar analyses of N2 fixation showed much less dependence on depth distribution of light and biomass: Aphanizomenon fixed about twice as much N2 as Nodularia their; their fixation exceeded their own N demand by about 12%. Together, these species contributed 49% of the total N demand of the phytoplankton population. Computer models based on the measured light attenuation and photosynthetic coefficients indicate that growth of the cyanobacterial population could occur only in the summer months when the critical depth of the cyanobacteria exceeds the depth of mixing.  相似文献

The Bitter Lakes are the most significant water bodies of the Suez Canal, comprising 85% of the water volume, but spreading over only 24% of the length of the canal. The present study aims at investigation of the trophic status of the Bitter Lakes employing various trophic state indices, biotic and abiotic parameters, thus reporting the health of the Lake ecosystem according to the internationally accepted classification criteria’s. The composition and abundance of phytoplankton with a dominance of diatoms and a decreased population density of 4315–7376?ind. l?1 reflect the oligotrophic nature of this water body. The intense growth of diatoms in the Bitter Lakes depends on silicate availability, in addition to nitrate and phosphate. If the trophic state index (TSI) is applied to the lakes under study it records that the Bitter Lakes have an index under 40. Moreover, in the total chlorophyll-a measurements of 0.35–0.96?µg?l?1 there are more indicative of little algal biomass and lower biological productivity. At 0.76–2.3?µg?l?1, meanwhile, the low quantity of Phosphorus is a further measure of low biological productivity. In the Bitter Lakes, TN/TP ratios are high and recorded 147.4, and 184.7 for minimum and maximum ratios, respectively. These values indicate that in Bitter lakes, the limiting nutrient is phosphorus and confirm the oligotrophic status of the Bitter Lakes. The latter conclusion is supported by Secchi disc water clarity measurements, showing that light can penetrate, and thus algae can photosynthesize, as deep as >13?m. This study, therefore, showed that the Bitter Lakes of the Suez Canal exhibit oligotrophic conditions with clear water, low productivity and with no algal blooming.  相似文献

Spiny-surfaced species of Prorocentrum form harmful algal blooms, and its taxonomic identity is obscure due to the size and shape variability. Molecular phylogenies reveal two major clades: one for P. cordatum with sequences mainly retrieved as P. minimum, and the other for P. shikokuense with sequences also retrieved as P. dentatum and P. donghaiense. Several closely related clades still need to be characterized. Here, we provide nuclear SSU and LSU rRNA genes, and nuclear ITS region (ITS1-5.8S gene-ITS2) sequences of the strain CCMP3122 isolated from Florida (initially named P. donghaiense) and strains Prorocentrum sp. RCC6871–2 from the Ross Sea, Antarctica. We describe Prorocentrum thermophilum sp. nov. based on the strain CCMP3122, a species also distributed in the open waters of the Gulf of Mexico, New Zealand, and the Arabian Gulf; and Prorocentrum criophilum sp. nov. based on the strain RCC6872, which is distributed in the Antarctic Ocean and Arctic Sea. Prorocentrum thermophilum is roundish (~14 μm long, ~12 μm wide), with an inconspicuous anterior spine-like prolongation under light microscopy, valves with tiny, short knobs (5–7 per μm2), and several (<7) large trichocyst pores (~0.3 μm) in the right valve, as well as smaller pores (~0.15 μm). Prorocentrum criophilum is round in valve view (~11 μm long, 10 μm wide) and asymmetrically roundish in lateral view, the periflagellar area was not discernible under light microscopy, valves with very tiny, short knobs (6–10 per μm2), and at least 12 large pores in the right valve. Other potentially undescribed species of spiny-surfaced Prorocentrum are discussed.  相似文献

Abstract

In this study, a diesel oil-degrading bacterium was isolated from an oilfield water injection (water-bearing formations, 1,205?m depth) in Algeria. The bacterial strain, designated NL1, was cultivated on diesel oil as sole carbon and energy sources. Molecular analyses of the 16S rRNA gene sequence (KY397882) placed NL1 strain closely related to distinct cultivated species of the Delftia genus. Optimal diesel oil biodegradation by Delftia sp NL1 strain occurred at pH 11, 40?°C, 2?M NaCl and initial hydrocarbon concentration of 5% (v/v) as sole carbon source. GC-MS analyses evidenced that strain Delftia sp NL1 was able to degrade more than 66.76% of diesel oil within only 7?days. On the other hand, and in the same conditions, biosurfactant production by Delftia sp NL1 was also evaluated evidencing high emulsifying capacity (E24 = 81%), ability to lower the surface tension of growing media (with the value of 25.7?mN m?1), and production of glycolipids (8.7?g L?1) as biosurfactants. This research presents indigenous strain Delftia sp NL1 for diesel degradation and synthesis of biosurfactant in extreme conditions. In this sense, strain NL1 is a good candidate for possible in situ oil recovery and in wastewater treatment in refineries and oil terminals in petroleum industry.  相似文献

The effect of copper additions (Cu ranging from 0 to 30?µM) on the photosynthesis of three different microalgal biofilms was studied to identify the factors that cause sensitivity differences between benthic and pelagic algae. The response of biofilms which colonized artificial substrata in the River Meuse was compared with those of two laboratory-grown monospecific biofilms, one consisting of the diatom Synedra ulna, and the other composed of a filament-forming cyanobacterium, Oscillatoria sp. The photosynthetic yield ΦII (quantum efficiency of photosystem II) was studied with PAM (Pulse Amplitude Modulated) fluorimetry. S. ulna biofilms appeared to be the most sensitive to Cu, followed by the cyanobacteria, while natural biofilms, dominated by supposedly very sensitive diatom species such as Melosira varians and Diatoma vulgare, were the most resistant to Cu. In the highly productive biofilms, pH is suggested to play a role in lowering toxicity by helping the precipitation of cupric ions. Cu accumulation by the biofilms during the exposure period followed a linear relationship with Cu concentration, saturation not being observed; natural biofilms had an accumulation factor of 1–2.5?×?103 relative to the concentrations in the water, while the diatoms growing unattached to the substratum had a higher concentration factor, up to 4.9?×?103. It was concluded that the physical structure of the biofilm (package of cells and thickness), and not the species composition, was the main factor regulating the sensitivity of the biofilm to Cu toxicity during short-term exposures.  相似文献

Abstract

Growth and production of carotenoid in three Dunaliella species (Dunaliella salina (Dunal) Teodoresco, Dunaliella bardawil Ben-Amotz & Avron and Dunaliella sp.) were investigated using flat-plate photobioreactors in outdoor conditions with two optical paths (3?cm and 5?cm). The experiment was conducted in duplicate and lasted four weeks during which light intensity, temperature, pH and optical density were checked daily. The pigment production (total carotenoid and chlorophyll a) was monitored every two days. To induce an additional stress besides temperature and light intensity, two different salt concentrations were used, i.e. 6% and 8% NaCl. The highest growth in all treatment groups was noticed for Dunaliella sp. followed by D. bardawil and D. salina. D. salina produced a higher content of carotenoid concentrations corresponding to 5?cm/8% and 5?cm/6% groups; 779.102?±?0.434?μg.mL?1 and 694.326?±?0.098?μg.mL?1 were registered at the end of the experiment. The effect of combined iron, silicate, and light co-limitation was investigated in the three diatom species Actinocyclus sp. Ehrenberg, Chaetoceros dichaeta Ehrenberg, and Chaetoceros debilis Cleve, isolated from the Southern Ocean (SO). Growth of all species was co-limited by iron and silicate, reflected in a significant increase in the number of cell divisions compared to the control. Lowest relative Si uptake and drastic frustule malformation was found under iron and silicate co-limitation in C. dichaeta, while Si limitation in general caused cell elongation in both Chaetoceros species. Higher light intensities similar to SO surface conditions showed a negative impact on growth of C. dichaeta and Actinocyclus sp. and no effect on C. debilis. This is in contrast to the assumed light limitation of SO diatoms due to deep wind driven mixing. Our results suggest that growth and species composition of Southern Ocean diatoms is influenced by a sensitive interaction of the abiotic factors, iron, silicate, and light.  相似文献

Light source can affect the stomata opening, photosynthesis process, and pigment content in microalgae cells. In this study, growth rate, chlorophyll a (chl a) content, and electrogenic capability of Desmodesmus sp. A8 were investigated under incandescent and fluorescent lamps. Growth rate, productivity, and chl a content of strain A8 exposed to incandescent light were recorded as 0.092 ± 0.010 day?1, 0.019 ± 0.008 g L?1 day?1, and 15.10 ± 1.40 mg L?1, which decreased to 0.086 ± 0.006 day?1, 0.012 ± 0.004 g L?1 day?1, and 10.06 ± 1.59 mg L?1, respectively, under fluorescent light. The stable current density of bioelectrochemical systems inculcated with strain A8 under incandescent and fluorescent lamps were 249.76 and 158.41 mA m?2 at ?0.4 V vs. Ag/AgCl, coupling with dissolved oxygen within biofilm decreasing from 15.91 to 10.80 mg L?1. This work demonstrated that illuminating microalgae under an incandescent lamp can improve biomass production and electrogenic capabilities.  相似文献

Abstract

The study recommends the Artemisia nilagirica (Clarke) Pamp. essential oil (ANEO) as plant-based shelf-life enhancer of millets against fungal, aflatoxin B1 (AFB1) contamination and lipid peroxidation with favourable safety profile. Chemical characterisation of ANEO through GC-MS, recorded 1,5-heptadiene-4-one,3,3,6-trimethyl (32.72%)as the main compound, followed by Artemisia alcohol (13.40%), alpha lonone (4.55%), benzene, methyl (1-methylethyl) (2.97%) and 1-cyclohexene-1-carboxaldehyde,4-(1-methylethyenyl) (2.23%). The mycoflora analysis of millet samples showed Aspergillus flavus strain[LHP(R)-5] as the most AFB1 secreting strain. The ANEO inhibited growth and AFB1 production by the toxigenic strain at 1.4 and 1.0?µL?mL?1, respectively, and also possess broad fungitoxic spectrum. The decrement in membrane ergosterol content, enhanced leakage of cellular Ca2+, K+ and Mg2+ ions along with SEM and TEM study of ANEO-treated cell denotes plasma membrane as antifungal site of action. The ANEO also showed strong antioxidant activity as determined by DPPH? and ABTS?+ assays having IC50 value 2.51 and 1.07?µL?mL?1, respectively. More than 70.78% protection of Ragi samples from fungal contamination was observed during in situ trial. The ANEO showed favourable safety profile with high LD50 value (7528.10?µL?kg?1) for male mice and also exhibited non-phytotoxicity for Ragi seeds germination.  相似文献

Growth, productivity and potential for exploitation of the clam Tawera gayi from shallow waters (3–5 m) of Ushuaia Bay, Beagle Channel were investigated. Mean abundance and biomass in the study area were 1091 ± 737 ind. m?2 and 901.83 g SFWM m?2 (shell‐free wet mass), respectively. Individual growth was described best by the von Bertalanffy growth model with the parameter values H = 28.03 mm, K = 0.288 year?1, t0 = ?0.34 (r2 = 0.83). Annual production of the population was estimated to be 120.45 g SFWM m?2 year?1, corresponding to a production‐to‐biomass ratio (P/B) of 0.134 year?1. The single negative exponential mortality model does not fit the population mortality pattern, but predation by gastropods (Xymenopsis muriciformis, Trophon geversianus, Natica sp.) appears to be the major cause of mortality. These highly mobile predators together with the comparatively slow growth and low turnover of T. gayi in Ushuaia Bay limit its potential for sustainable commercial exploitation.  相似文献

Growth of Codium fragile subsp. tomentosoides (van Goor) Silva in culture depends upon the season of seawater collection. One factor responsible for this variation in growth may be indole-3-acetic acid (IAA). When 10?9 to 10?4 M IAA is added to cultures of Codium fragile, optimum growth is at 10?6 M. The response to exogenous IAA depends upon the time of year when the sea-water is collected. The growth in a range of known IAA concentrations allows the prediction of a seasonal cycle of IAA, or its physiological equivalent, in Rhode Island coastal waters. Such a compound may be an important ecological factor for some algal species.  相似文献

Viable diatom and dinoflagellate resting stages were recovered from sediments in Koljö Fjord on the west coast of Sweden. To determine the maximum survival time of buried resting stages, samples from sediment depths down to 50?cm were incubated at temperatures of 3, 10 and 18?°C. Sediment cores were dated by 210Pb and the age of samples containing viable resting stages was determined using the constant rate of supply model. Dilution cultures of surface sediments allowed semiquantitative estimates of the potential seed bank. Dinoflagellate cysts from species such as Diplopsalis sp., Gymnodinium nolleri, Oblea rotunda and Protoceratium reticulatum were viable down to 15?cm depth, or 37 years old. Spores and resting cells of the diatoms Chaetoceros spp., Detonula confervacea and Skeletonema costatum were viable to over 40?cm depth, and may have been buried for many decades. The seed bank of living resting stages in surficial sediments was found to be rich (c. 57000 diatom resting stages g?1 wet weight and c. 200 dinoflagellate cysts g?1 wet weight), and the percentage of viable resting stages was higher for spore- and cyst-forming species. The oxygen-deficient sediments in Koljö Fjord appear to be a natural conservator of cell viability, a condition not easily simulated in laboratory studies. These results are ecologically important since spores and cysts are a repository of genetic material able to repopulate waters if resuspended and exposed to suitable light, temperature and nutrients.  相似文献
