Agar blocks that contacted the upper sides of tips of horizontally-orientedroots of Zea mays contain significantly less calcium (Ca) thanblocks that contacted the lower sides of such roots. This gravity-inducedgradient of Ca forms prior to the onset of gravicurvature, anddoes not form across tips of vertically-oriented roots or rootsof agravitropic mutants. These results indicate that (1) Cacan be collected from mucilage of graviresponding roots, (2)gravity induces a downward movement of endogenous Ca in mucilageoverlying the root tip, (3) this gravity-induced gradient ofCa does not form across tips of agravitropic roots, and (4)formation of a Ca gradient is not a consequence of gravicurvature.These results are consistent with gravity-induced movement ofCa being a trigger for subsequent redistribution of growth effectors(e.g. auxin) that induce differential growth and gravicurvature. Phosphate uptake by excised roots of sunflower (Helianthus annuus)was determined by the disappearance of phosphate from the externalsolution and by the accumulation of phosphate labelled with32P. Over a 24 h period it was observed that net phosphate uptakedeclined to zero whilst uptake of 32P continued unabated. Theelectrical PD of the cortical cell membranes declined in parallelwith net phosphate uptake and it was found that both could berestored by creating a pH gradient across the plasmalemma. Chloride concentrations in longitudinal files of cells acrossthe root of Commelina communis have been determined. Vacuolarsap was taken from the root using a microsampling techniqueand chloride concentration determined on nanolitre samples byelectrometric microtitration. No radial gradient in vacuolar chloride was observed, eitherfor roots grown in a nutrient solution containing a low level,or for those grown in a solution containing a high level, ofchloride. Vacuolar electrical potentials were also determined,on attached and excised roots. The PD was found not to varysignificantly across the root from epidermis to pericycle despitethe PD in attached roots being 50 mV more negative than thatin excised roots. These results are discussed in relation tothe mechanism of ion transport across the root.  相似文献

The effect of high pH on the morphology and anatomy of the rootsof lupin (Lupinus angustifolius L. cv. Yandee) and pea (Pisumsativum L. cv. Dundale) was examined in buffered solution. Themorphology and anatomy of lupin roots were markedly altered,and root growth was reduced by increasing solution pH from 5·2to 7·5, whereas pea roots were unaffected. In lupin roots,pH 7·5 caused disintegration of the root surface andimpaired root hair formation. Lupin roots grown at pH 7·5also had decreased cell lengths but increased cell diameterin both the epidermis and the cortex in comparison to rootsgrown at pH 5·2. High pH reduced cell volume greatlyin the epidermis, to a lesser extent in the outer cortex andnot at all in the inner cortex. The intracellular pH of an acidophilic unicellular alga, Cyanidiumcaldarium, was determined as a function of external pH by 31Pnuclear magnetic resonance. The algal cells incubated underaerobic conditions or under anaerobic and illuminated conditionsmaintained the intracellular pH in the range from 6.8 to 7.0even when the external pH was changed from 1.2 to 8.4. Underanaerobic and dark conditions, however, the intracellular pHacidified at the acidic pH region of the external medium. Theacidified intracellular pH reversibly returned to neutral eitheron aeration or illumination. The results indicate that, in Cyanidiumcells growing in extremely acidic environments, an active H+efflux (H+ pump) which depends on metabolic activity (respirationor photosynthesis) is essential to maintain the intracellularpH at a constant physiological level against the passive H+leakage due to the steep pH gradient across the cell membrane. Membrane transport pathways mediatingtranscellular secretion of urate across the proximal tubule wereinvestigated in brush-border membrane vesicles (BBMV) isolated fromavian kidney. An inside-positive K diffusion potential induced aconductive uptake of urate to levels exceeding equilibrium.Protonophore-induced dissipation of membrane potential significantlyreduced voltage-driven urate uptake. Conductive uptake of urate wasinhibitor sensitive, substrate specific, and a saturable function ofurate concentration. Urate uptake was trans-stimulated byurate and cis-inhibited by p-aminohippurate (PAH). Conductive uptake of PAH was cis-inhibited by urate.Urate uptake was unaffected by an outward -ketoglutarate gradient. In the absence of a membrane potential, urate uptake was similar in thepresence and absence of an imposed inside-alkaline pH gradient or anoutward Cl gradient. These observations suggest a uniporter-mediated facilitated diffusion of urate as a pathway for passive efflux acrossthe brush border membrane of urate-secreting proximal tubule cells.

The length of root hairs of wheat (Triticum aestivum L.) growingin Long Ashton nitrate nutrient solution was affected mainlyby pH and the concentrations of calcium and nitrate. Root hairlength was decreased by high pH (> 7.0) and by low calciumconcentrations but was increased by nitrate. Buffering at pH6.5 was found to affect the relationship between nutrient solutionstrength and the length of root hairs. The interactions betweencalcium, nitrate and buffering were determined. Phosphate concentrationdid not affect the length of root hairs of wheat, although lowconcentrations have been reported to stimulate root hair growthin other plants. Techniques have been developed for continuous recording of electricalpotential difference in maize root preparations. In this waythe continuous p.d. profile along the lateral axis of the rootcould be obtained. The features of the profile are resolvedin terms of the root morphology. Analysis of the p.d. profile indicates (a) that the phases withinthe root are electrically negative with respect to the bathingmedium, (b) that the vacuoles are positive relative to the cytoplasm,i.e. a p.d. exists across the tonoplast, (c) that all cytoplasmicphases are equipotential, which indicates a continuity withinand between the cortical and stelar symplasms, (d) that theextracellular space of the root is presumably only slightlyinfluenced by the concentration of the external medium sincethe p.d. profile within the root does not react to changes ofconcentration in the external medium, (e) that the potentialof the stelar vacuoles is negative relative to the corticalone and this is consistent with the accumulation of surplusions in the stelar vacuoles. The implications of the present findings to the elucidationof ion transport mechanism across the root are briefly discussed.  相似文献

We studied the response of maize (Zea mays L. cv. Anjou 256)to a simultaneous, but separated supply of ammonium and nitrate(localized supply, LS). A split-root system was used to supplyhalf of the roots with ammonium and the other half with nitrate.A homogeneously distributed supply of both nitrogen forms (HS)was the control treatment. Seedlings were grown for 12 d fromthe two-leaf to the three-leaf stage in hydroponics at threepH levels (4, 5·5 and 7). The total N concentration was3 mol m-3. The split-root system was established by removingthe seminal root system and using only four nodal roots perplant. Total root length and root surface area were recordedautomatically with a modified Delta- T area meter. Other morphologicalroot traits (such as main axis length and diameter, number,density, and length of laterals) were recorded manually. Uptakeof ammonium and nitrate was measured by the depletion of thenutrient solution. As compared with LS, HS was superior in shootand root DM, total root length and root surface area, ammoniumand nitrate uptake and shoot nitrogen concentration, irrespectiveof pH level. This indicates that, also under field conditions,mixed ammonium and nitrate fertilization is only beneficialto plant growth if both N forms are evenly distributed in thesoil. At both HS and LS, ascending pH increased the ammonium:nitrateuptake ratio. The hydrostatic pressure gradient across the root cortex inducedby transpiration was simulated by raising the pressure on themedium surrounding the roots of detopped tomato (Lycopersicumesculentum) and castoroil plants (Ricinus communis).Applyinga pressure of 2 atm. resulted in a doubling of the sodium fluxfrom medium to xylern (table I). This flux depended on a maintainedsuply of Na in the medium (fig.1) indicating that there wasa small but readily available storage and exchange capacityfor Na in the root tissues. In the presence of metabolic inhaibitorsthe Na flux was considerablyu reduced and equalled the fluxof mannitol (talble II) believed to be a passive mass flow.The Na flux under a pressure gradient appeared therefore tobe metabollism-facilitated. Metabolic inhibition after the rootshad been allowed to take up Na led to a large flux of sodiuminto the xylem (Fig.2). This was considered to come from a largeunavailable store in the roots. Metabolism-facilitated processes also appeared to predominatein the flux of calcium to the xylem of Ricinus, buit not inthe flux through tomato roots. The location of the centripetal metabolism-facilitated transferand the available and unavailable stores is discussed in relationto the effects of pressure gradients.  相似文献

Hydraulic properties of entire root systems and isolated rootsof three contrasting sugarcane clones were evaluated using transpiration-induceddifferences in hydrostatic pressure across intact root systems,root pressure-generated xylem sap exudation, and pressure-fluxrelationships. Regardless of the measurement technique employed,the clones were ranked in the same order on the basis of theirleaf area–specific total root system hydraulic conductance(Croot). All methods employed detected large developmental changesin Grootroot with maximum values occurring in plants with approximately02 m2 total leaf area. Genotypic ranking according to Groot,was reflected as a similar ranking according to root length-specifichydraulic conductance (L) of individual excised roots. Genotypicdifferences in Groot and L were consistent with anatomical characteristicsobserved in individual roots. Patterns of Groot, during soildrying and following re-irrigation suggested that the declinein Groot, observed during soil drying occurred within the rootsrather than at the soil–root interface and may have beencaused in part by xylem cavitation in the roots. The light-induced H+ efflux observed at acidic pH in Cyanidiumcells was shown to be an active H+ transport depending on theintracellular ATP produced by cyclic photo-phosphorylation.Triton X-100 was found to act as an effective uncoupler in intactCyanidium cells without collapsing the pH gradient across theplasma membrane. Triton X-100 at 0.015% significantly reducedthe intracellular ATP levels, stimulated the p-BQ, Hill reactionand completely inhibited the light-induced H+ efflux. Inhibitionof the H+ efflux by Triton X-100 correlated well with the depressionof the apparent rale of light-induced ATP synthesis as wellas the decrease in the intracellular ATP level in light. The light-induced H+ efflux was completely inhibited by diethylstilbestrol,a specific inhibitor of plasma membrane ATPase, without anychanges in the intracellular ATP level, thereby suggesting theparticipation of the plasma membrane ATPase in the light-inducedH+ efflux. 1The data in this paper are included in the Ph. ATPase activity was measured in crude homogenates of guard cellprotoplasts of Commelina communis L. using a linked enzyme assay.A low level of azide-sensitive ATPase activity was detectedwith a pH optimum of 6.8. This activity was stimulated by 0.01%(v/v) Triton X-100, and the pH optimum shifted to pH 7.4. Nitrate-sensitiveATPase activity was measured in the presence of azide and showeda pH optimum around pH 8.0. Proton pumping activity in a mixedpopulation of vesicles from GCP was monitored using fluorescencequenching of quinacrine. Mg-ATP dependent proton pumping wasobserved at pH 8.0, but not at pH 6.6. The activity at pH 8.0was inhibited by nitrate and DCCD but not vanadate. These dataindicate that activity of the tonoplast proton pump was beingmeasured. There was, however, no evidence for a tonoplast cation(K+)/proton antiporter under these assay conditions as potassiumdid not reduce the initial rate of pH gradient formation orincrease the rate of collapse of a pre-formed gradient afterinhibition of the pump. The influx and efflux of Na+ across the plasma membrane of Characorallina and Chara longifolia were examined under mild saltstress conditions. Na+ influx was found to be rapid in bothspecies with the freely exchangeable cytoplasmic Na+ cominginto isotopic equilibrium with external 22Na+ within 1 h ofexposure to isotope. Cytoplasmlc Na+ concentration and Na+ influxwere greater in C. corallina than in C. longifolla under thesame conditions. Na+ influx across the tonoplast was much lowerthan the flux across the plasma membrane. Na+ efflux was stimulatedat pH 5 relative to pH 7 by 218% in C. coralllna and 320% inC. longifolia. In both species externally applied Li+ inhibitedNa+ efflux at pH 5 but not at pH 7. Na+ etflux was not significantlyinhibited by amiloride. Sodium efflux from 22Na+-loaded root tips root tips of Hordeumvulgare L. was markedly increased by replacing 10mM Na2SO4 inthe washing solution by K2SO4 with the same electrical conductivity.This increase was inhibited by both an uncoupler and an inhibitorof oxidative phosphorylation but not by ouabain. Potassium ionsdid not enhance Na+ efflux in the presence of a rapidly absorbedcounter anion, such as Cl, instead of . Efflux of 22Na+ could also be enhanced by a low pH in theabsence of K+; this was prevented by uncouplers, but not byan inhibitor of the mitochondrial ATPase. It seems that K+ indirectly enhances Na+ efflux. It is suggestedthat metabolic K+ uptake in excess of the counter anion resultsin a proton gradient across the plasmalemma (acid outside) inducingH+/Na+ antiport.  相似文献

The physico-chemical properties of iron oxyhydroxide plaquesformed on the roots of Phragmites australis under field andlaboratory conditions were determined using electron microscopyand energy dispersive spectrometry (EDS) analysis. Plaques werepresent as an amorphous coating on roots with an uneven distribution.They were shown to be composed of iron in both the field andlaboratory, and phosphorus was found to be adsorbed onto thesurface of plaques formed in the laboratory. The uptake of copperand manganese in the presence and absence of plaque was investigatedunder two different pH regimes. Concentrations of Cu were lowerin the shoots of P. australis in the presence of plaque (565mg kg-1) than when it was absent (1400 mg kg-1), under growthconditions of higher pH (6.0). The adsorption of Cu and Mn ontothe plaque surface was not the mechanism by which plaque reducedthe uptake of other metals. Alternatively, the plaque may simplyact as a physical barrier. Under conditions of lower pH (3.5)the activity of hydrogen ions at the root surface interferedwith the movement of metals into the root and masked any potentialeffect of iron plaque. Thecharacteristics of L-lactic acid transport across thetrophoblast basal membrane were investigated and compared with those across the brush-border membrane by using membrane vesicles isolated from human placenta. The uptake ofL-[14C]lactic acid into basal membranevesicles was Na+ independent, and an uphill transport wasobserved in the presence of a pH gradient([H+]out > [H+]in).L-[14C]lactic acid uptake exhibitedsaturation kinetics with a Km value of 5.89 ± 0.68 mM in the presence of a pH gradient.p-Chloromercuribenzenesulfonate and-cyano-4-hydroxycinnamate inhibited the initial uptake, whereas phloretin or 4,4'-diisothiocyanostilbene-2,2'-disulfonate did not.Mono- and dicarboxylic acids suppressed the initial uptake. Inconclusion, L-lactic acid transport in the basal membraneis H+ dependent and Na+ independent, as is alsothe case for the brush-border membrane transport, and itscharacteristics resemble those of monocarboxylic acid transporters.However, there were several differences in the effects of inhibitorsbetween basal and brush-border membrane vesicles, suggesting that thetransporter(s) involved in L-lactic acid transport in thebasal membrane of placental trophoblast may differ from those in thebrush-border membrane.

A glasshouse experiment was carried out to investigate the factorscontrolling the abstraction of xylem fluid from its host bythe parasiteStriga hermonthica(Scrophulariaceae).Strigahad amean daily transpiration rate far exceeding that of its hostsorghum (Sorghum bicolor), with infestation byStrigaalso shownto lower the transpiration rate of the host. Stopping the host'stranspiration was shown to decrease the transpiration rate ofthe parasite. Stopping the parasite's transpiration only gavean initial increase in the host's transpiration rate which wasnot sustained. The parasite had a lower water potential thanits host, values being -0.42 MPa and -0.23 MPa, respectively,and an accompanying higher osmotic pressure of 0.68 MPa against0.51 MPa for sorghum. Modifying the water potential gradientby bagging both partners together showed that the differentialin osmotic pressure and water potential was largely maintainedby the parasite's higher rate of transpiration. A favourablewater potential gradient towards the parasite still existedfollowing the cessation of transpiration, this being generatedby the haustorial resistance to hydraulic conductivity whichwas found to be some 1.5–4.5 times greater than that offeredby the parasite shoot.
