3-Chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone (MX) was detected by gas chromatography/mass spectrometry in drinking water samples from 3 locations in the U.S.A., and also in a chlorinated humic acid solution. MX appears to account for a significant proportion of the mutagenicity of these samples, as measured in the Ames test using strain TA100 without metabolic activation. Studies on recovery of MX from spiked water samples by XAD-2/8 resin adsorption/acetone elution indicated that sample acidification prior to resin adsorption was essential to the effective recovery of MX. The stability of MX in aqueous solution was pH and temperature dependent. At 23 degrees C the order of stability, based on persistence of mutagenic activity was found to be: pH 2 greater than pH 4 greater than pH 8 greater than pH 6. The half-life at pH 8 and 23 degrees C was 4.6 days. One of the degradation products has been tentatively identified as 2-chloro-3-(dichloromethyl)-4-oxo-2-butenoic acid, an open form of MX which appears to be in the "E" configuration. Overall, these results suggest that MX is formed during water chlorination as a result of reaction of chlorine with humic substances, and that a substantial fraction of the MX formed is likely to persist throughout the distribution system.  相似文献

The removal of phenol (Ph) and 2-chlorophenol (2-CPh) from aqueous solution by native and heat inactivated fungus Funaliatrogii pellets were investigated. The effects of contact time, solid/liquid ratio, optimum pH and temperature on the phenols removal capacity by the pellets were established. The removal efficiency of phenols increased significantly with increasing biomass dose. The optimum pH was detected to be 8.0. The second-order equations are described and evaluated on the basis of a comparative estimation of the corresponding coefficients. The phenol removal equilibrium isotherm was modeled by the Langmuir equations. The enthalpy change values were obtained between −7.62 and −10.64 kJ/mol. This indicated that the uptake of phenols either on native or heat inactivated fungal pellets was based on a physical adsorption process.  相似文献

The present study involves an investigation of various treated fungal biomasses of Aspergillus niger for the removal of thallium from aqueous solutions. Batch pH and kinetic studies were carried out to examine the effects of pH and contact time on the adsorption process. Among various pH values studied, the optimum pH was found to be between 4 and 5. The equilibrium time for Tl adsorption was found to be 6h and the rate of Tl adsorption was rapid in the initial hours. Both Lagergren's pseudo first-order model and Ho's pseudo second-order model well described the reaction kinetics. Batch adsorption experiments conducted at room temperature (22+/-1 degrees C) showed that the adsorption pattern followed the Freundlich isotherm model. Column studies using iron oxide-coated immobilized fungal biomass showed lower adsorption capacities compared to batch studies.  相似文献

The responses of second-stage juveniles (J2) of Meloidogyne incognita race 3 to calcium alginate pellets containing hyphae of the nematophagous fungi Monacrosporiura cionopagum, M. ellipsosporum, and Hirsutella rhossiliensis were examined using cylinders (38-mm-diam., 40 or 72 mm long) of sand (94% <250-μm particle size). Sand was wetted with a synthetic soil solution (10% moisture, 0.06 bar water potential). A layer of 10 or 20 pellets was placed 4 or 20 mm from one end of the cylinder. After 3, 5, or 13 days, J2 were put on both ends, on one end, or in the center; J2 were extracted from 8-ram-thick sections 1 or 2 days later. All three fungal pellets were repellent; pellets without fungi were not. Aqueous extracts of all pellets and of sand in which fungal pellets had been incubated were repellent, but acetone extracts redissolved in water were not. Injection of CO₂ (20 μl/minute) into the pellet layer attracted J2 and increased fungal-induced mortality. In vials containing four randomly positioned pellets and 17 cm³ of sand or loamy sand, the three fungi suppressed the invasion of cabbage roots by M. javanica J2. Counts of healthy and parasitized nematodes observed in roots or extracted from soil indicated that, in the vial assay, the failure of J2 to penetrate roots resulted primarily from parasitism rather than repulsion. Data were similar whether fungal inoculum consisted of pelletized hyphae or fungal-colonized Steinernema glaseri. Thus, the results indicate that nematode attractants and repellents can have major or negligible effects on the biological control efficacy of pelletized nematophagous fungi. Factors that might influence the importance of substances released by the pellets include the strength, geometry, and duration of gradients; pellet degradation by soil microflora; the nematode species involved; and attractants released by roots.  相似文献

Phenoxyalkanoic acids are a widely used class of herbicides. This work employed high-resolution 13C NMR to study the structural changes induced by humic substances and horseradish perodixase on 2,4-dichorophenoxyacetic acid (2,4-D) 13C-labelled in the side chain. NMR spectra showed that humic substances chemically catalyze abiotic splitting of [13C]2,4-D into 2,4-dichlorophenol and [13C]acetic acid at pH 7 but not at pH 4.7. Peroxidase did not catalyze the oxidative degradation of [13C]2,4-D at any pH tested and inhibited the effect of humic substances. Catalytic degradation by humic substances was attributed to free-radical reactions enhanced by the stereochemical contribution of large conformational structures formed by heterogeneous humic molecules at neutral pHs. Inhibition of 2,4-D degradation when humic substances were combined with peroxidase was explained by modification of both chemical and conformational humic structure due to peroxidase-promoted oxidative cross-coupling among humic molecules. Our findings show for the first time that the abiotic degradation of 2,4-D is catalyzed by dissolved humic substances at neutral pH. The effects of humic substances and low pH on short‐term Cd uptake by Pseudokirchneriella subcapitata (Korshikov) Hindak and Chlamydomonas reinhardtii Dang were investigated under defined exposure conditions. The uptake experiments were run in the presence of either a synthetic organic ligand (nitrilotriacetate) or natural organic ligands (Suwannee River fulvic or humic acid). An ion‐exchange method was used to measure the free Cd2+ concentrations in the exposure solutions. At pH 5, measured free Cd2+ concentrations agreed with estimations made using the geochemical equilibrium model WHAM, but at pH 7 the model overestimated complexation by both Suwannee River fulvic and humic acids compared with the ion‐exchange measurements. Consistent with the metal internalization step being rate limiting for overall short‐term uptake, intracellular Cd uptake was linear for exposure times less than 20 min at pH 5 or pH 7 for both algal species. After taking into account complexation of Cd in solution, Suwannee River humic substances had no additional effects on cadmium uptake at pH 7, as would be predicted by the free ion model. This absence of effects other than complexation persisted at pH 5, where the tendency of humic substances to adsorb to the algal cell surface is favored. Changes in pH strongly influenced Cd uptake, with the intracellular flux of Cd being at least 20 times lower at pH 5 than at pH 7 for P. subcapitata. Our results support models such as the free ion model or the biotic ligand model, in which humic substances act indirectly on Cd uptake by reducing the bioavailability of Cd by complexation in solution.  相似文献

The mycelia pellets of Penicillium simplicissimum impregnated with powdered biochar (MPPSIPB) were synthesized and applied to remove chromium (VI) from aqueous solution. The effects of pH, MPPSIPB dosage, initial Cr(VI) concentration, and contact time were investigated via batch experiments. Results indicated that the percentage removal of Cr(VI) was significantly dependent on the pH of the solution. Ten grams mycelial pellets and 0.2 g powdered biochar could form the most stable pellets. The maximum value of biosorption of Cr(VI) was 28.0 mg/g. Scanning electron microscopy (SEM) analysis showed that the mycelia pellets of Penicillium simplicissimum had abundant filamentous network, which entrapped powdered biochar firmly. Fourier transform infrared (FTIR) analysis suggested that O?H, N?H, C?H, C?O, and C?OH groups from MPPSIPB were involved in chromium binding and the subsequent reduction. Kinetic studies indicated that the pseudo-second-order equation fit best for Cr(VI) removal from aqueous solution. Freundlich isotherm was found to apply better for the adsorption equilibrium data with respect to the Langmuir isotherm. Furthermore, MPPSIPB can be separated from aqueous solution completely by filtration. Both experimental study and modeling results indicated that MPPSIPB exhibited remarkable affinity for chromate and had a potential application in Cr(VI) removal from water.  相似文献

建立白腐真菌J610-D与苋菜红的共培养体系,研究培养参数对染料脱色降解的影响。结果表明:(1)在含缓冲成分的系统中,菌种J610-D在pH值4.5的培养液比pH值3.0有利于反应进行,浅层培养较深层培养好,缓冲成分差别的作用不明显。(2)在不舍缓冲成分的系统中,生长与反应阶段培养液均为pH值4.5时,效果最佳;生长于pH值4.5,反应在pH值3.0脱色率和降解率高于生长及反应都在pH值3.0的情况,但仍有25%~40%的苋菜红牢固地结合在菌体上;pH值4.5生长,pH值6.0反应,导致反应启动滞后约4~6d,最终结果尚好。  相似文献

Aims Plant roots store large amount of terrestrial carbon, but little is known about humus formation processes during the decomposing root litter. Compared with coarse roots, fine roots have greater nutrients, which may be favorable to humus formation. The objective of the study was to examine how root diameters affect their humus formation processes. Methods In this study, in order to examine the accumulation of humic acid and fulvic acid of three root diameter classes (0-2, 2-5 and 5-10 mm) of two subalpine tree species (Abies faxoniana and Picea asperata) on the eastern Qinghai-Xizang Plateau of China, a two-year field experiment was conducted using a litter-bag method. Air-dried roots of A. faxoniana and P. asperata were placed in litterbags and incubated at 10 cm of soil depth in October 11th, 2013. Duplicate litter bags were collected in May (late winter) and October (late in the growing season) of 2014 and 2015, respectively. Concentrations of humic acid and fulvic acid were measured, and net accumulations were calculated for different periods. Important findings The concentrations of humic acid and fulvic acid were significantly influenced by root diameter that humic acid and fulvic acid decreased with increase in root diameter. Root diameter had significant effects on the net accumulation of humic acid, but not for the accumulation of fulvic acid. However, there were no significant differences in both humic acid and fulvic acid between A. faxoniana and P. asperata roots. Regardless of tree species, humic acid degraded during the winter but accumulated during the growing season. After two years of decomposition, the net accumulations of humic acid in 0-2, 2-5 and 5-10 mm roots were 8.0, 10.8 and 7.6 g·kg-1 for P. asperata and 15.2, 8.0 and 7.8 g·kg-1 for A. faxoniana, respectively. Conversely, the degradation of fulvic acid in 0-2, 2-5 and 5-10 mm roots were 178.0, 166.0 and 118.0 g·kg-1 for P. asperata and 170.0, 160.0 and 128.0 g·kg-1 for A. faxoniana, respectively. Our results suggest that diameter-associated variations in substrate quality could be an important driver for root litter humification in this subalpine forest. Moreover, diameter effect is dependent on decomposition period in this specific area.  相似文献

采取液体深层发酵的方法 ,研究了不同碳氮比对薄盖灵芝 [Ganodermacapense (Lloyd)Teng]液体培养的影响。结果表明 ,在氮量固定的情况下 ,随碳量的增加 ,薄盖灵芝菌丝体干重逐渐增大 ,各处理发酵液的pH值呈“上升→下降→平稳”的变化趋势 ;当碳量固定时 ,各处理的pH值变化基本呈“双峰”型。不同碳氮比对薄盖灵芝液体发酵影响较大 ,通过对菌丝生长量、pH值的测定以及对发酵液及菌球培养性状的观察 ,认为碳氮比在 2 0∶1~ 4 0∶1(C3~C7)对薄盖灵芝的液体发酵均较适宜。其中以 30∶1(C5 )为最适宜 ,在此条件下 ,终止发酵的pH值为 3 4 ,菌球数量虽为 113个 10mL发酵液 ,但菌丝体干重为 6 83 4mg dL。发酵液澄清度高 ,呈真溶液状 ,菌球大小均匀。  相似文献

AIMS: The objective of this work was to investigate the morphological and rheological properties in submerged culture of the three different basidiomycetes Phellinus (P. baumii, P. gilvus and P. linteus) that produce pharmacologically important exopolysaccharides (EPS). METHODS AND RESULTS: In flask cultures, pH proved to be a critical factor affecting mycelial growth, morphological change and EPS production. The macroscopic morphologies observed under different pHs in flask cultures were also comparable: i.e. starfish-like pellets with a lesser extent of free mycelium appeared in P. baumii, whereas smooth pellets with higher amounts of free mycelium were observed in P. gilvus and P. linteus. The pelleted fermentations were further characterized in a 5-l stirred-tank fermenter by image analysis with respect to mean diameter, core area and pellet circularity. Phellinus baumii showed the largest pellet size (5.2 mm in diameter), whereas P. linteus had extremely small and spherical pellets. The culture broth of P. baumii and P. gilvus yielded extremely high apparent viscosities, ranging from 5 to 7 Pa s. CONCLUSIONS: Three important species of Phellinus showed significantly different morphological and rheological properties. The morphological variation of the three Phellinus species was closely linked to EPS productivity and the apparent viscosity of the whole broth. SIGNIFICANCE AND IMPACT OF THE STUDY: The morphological change in the three species of Phellinus was a good indicator for identifying cell activity for EPS production. Our finding may be beneficial for further optimization of other fungal fermentation processes for large-scale production of EPS.  相似文献

探讨了橘青霉 (Penicilliumcitrinum)呈球状体生长的条件和形成的机理。发现在培养液初始pH为 7.0 ,孢子年龄为16~ 2 0天 ,孢子悬液浓度为 10 5个·mL-1,表面活性剂Tween80或TritonX - 10 0质量浓度为 1g/L ,摇床转速为 140r/min的条件下 ,于 2 5℃培养 5天 ,可形成大小适中 ,有一定机械强度 ,光滑均匀的菌丝球。孢子的聚集在菌丝球的形成中起了重要作用。用此菌丝球吸附质量浓度为 5 0mg·L-1的 1 氨基 2 萘酚 4 磺酸水溶液模拟处理染料废水 ,吸附率达到 6 2 .4%。  相似文献

This paper presents the adsorption of humic acid from aqueous solution onto crosslinked chitosan derivative (carboxymethylchitosan), formed by additionless irradiation technique. The surface charge and swelling properties of crosslinked samples were investigated. The adsorption of humic acid onto crosslinked carboxymethylchitosan was carried out by the batch method at room temperature, and it was found to be strongly pH-dependent. Maximum amount of humic acid was adsorbed under acidic conditions at the optimum pH value of 3.5. Adsorption kinetic studies indicated the adsorption process was transport-limited at the same pH. The adsorption isotherm analysis data under various initial humic acid concentrations confirms that experimental data fitted well into the Langmuir equation. X-ray photoelectron spectroscopy (XPS) revealed that the amino groups of carboxymethylated chitosan were protonated, suggesting the formation of organic complex between the protonated amino groups and humic acid. From these preliminary evaluations, it was concluded that crosslinked carboxymethylated chitosan derivatives have a great potential in water treatment for the removal of humic acid and other polarized or electrically charged species.  相似文献

从南京某石化厂排污口附近采集土样,以菲为碳源的选择性培养基分离筛选到一株菲高效降解菌F10a,根据形态和生理生化特性初步鉴定为芽孢杆菌属,并对其降解菲的特性及各种影响因素进行了研究.结果表明,F10a在50 mg·L-1的条件下,28 ℃振荡培养27 h,菲的降解率达到98.12%;静置培养8 h,菲的降解率达到98.7%.pH值分别为、6、8时,F10a对菲具有良好的降解效能;pH值为10时F10a不生长.Zn2+与Pb2+的存在不影响F10a的降解效能,Cu2+可以延缓菲的降解,Cr2+对F10a有毒性.F10a在菲浓度为200 mg·L-1时,28 ℃振荡培养8 h,降解率为99.6%.菲的降解程度与细菌数量的增长呈正相关关系.  相似文献

This study investigated the effects of surfactant additives and medium pH on mycelial morphology and exopolysaccharide (EPS) production in liquid culture of a valuable medicinal fungus Cordyceps sinensis Cs-HK1. In the medium containing 20 g l−1 glucose and 6 g l−1 peptone as the sole nitrogen source, the Cs-HK1 fungal mycelia formed smooth and spherical pellets about 1.8-mm mean diameter. The mycelial pellets became less uniform at pH (4.0–5.0) lower than the optimum (pH 6.0) or turned to filamentous form at higher pH (8–9). Surfactants added to the medium inhibited pellet formation, resulting in smaller and looser pellets. Tween 80 exhibited a remarkable promoting effect on EPS production, increasing the EPS yield more than twofold at 1.5% (w/v), which was most probably attributed to the stimulation of EPS biosynthesis and release from the fungal cells by Tween 80.  相似文献

对生防链霉菌Ⅲ-61产生抗真菌活性物质的摇瓶发酵工艺进行了研究。利用正交试验设计优化了发酵培养基组分,其最适配方为黄豆粉1.5%,蛋白胨0.3%,蔗糖1.0%,淀粉1.3%,磷酸二氢钾0.02%,硫酸镁0.025%,氯化钠0.5%,配咸水溶液,调pH至7~7.4,加碳酸钙1%。通过单因素试验,筛选获得了最优培养条件组合:液体种龄24h,接种量5%~10%,500mL摇瓶培养基装量为80mL,摇床转速240r/min,培养温度31℃,发酵周期96~120h。此优化的发酵培养基与发酵条件的组合昕得菌株Ⅲ-61发酵液对主要靶标黄瓜灰霉病菌的抑菌圈直径达49.5mm,较优化前提高了45.59%。  相似文献

AIMS: To test the anaerobic fungus, Piromyces sp. FNG5, for its tolerance to phenolic monomers released in the rumen by degradation of lignocellulosic poor-quality feeds. METHODS AND RESULTS: Effects of phenolic monomers on biomass and fibrolytic enzyme activities of a pure culture of lignocellulolytic anaerobic fungus (Piromyces sp. FNG5) isolated from faeces of wild nil gai (blue bull, Baselophus tragocamelus) were evaluated. There was a reduction in fungal biomass at 1 mm concentration of catechol with complete inhibition at 10 mm. p-Coumaric acid caused a reduction in biomass at 10 mm and no growth was observed above 20 mm concentration. The fungal isolate could tolerate up to 5 mm of ferulic acid without any reduction in biomass level, and was able to grow to some extent up to the highest level of ferulic acid tested (20 mm). Vanillic acid had no effect on biomass of the fungus even up to 50 mm level. The phenolic monomers varied in their potential to inhibit the secretion of carboxymethyl cellulase, xylanase, beta-glucosidase and acetyl esterase activities with catechol being the most inhibitory and vanillic acid being the least inhibitory. After 14 days of incubation, 38.49-65.14%p-Coumaric acid, 65.22-74.10% ferulic acid and 34.13-66.78% vanillic acid disappeared from the medium under anaerobic conditions. CONCLUSIONS, SIGNIFICANCE AND IMPACT OF THE STUDY: It is concluded that the anaerobic fungus Piromyces sp. FNG5 is tolerant to phenolic monomers and has ability to degrade them. Therefore, such anaerobic fungi may play an important role in fibre degradation in the rumen.  相似文献

本文就小麦全蚀病菌胞外-1,3-葡聚糖酶的产生和部分酶学特性进行了研究。结果表明,小麦全蚀病菌能够产生胞外-1,3-葡聚糖酶。在供试的三种培养基中,最佳产酶培养基为改进的MS培养基。当以改进的MS为基础培养基时,最佳碳源为麦麸皮;最佳氮源为牛肉浸膏;产酶的最适条件为培养基初始pH为6,培养温度为26℃,250ml三角瓶中装培养基量为50ml时接菌量为4块菌饼(直径5mm)。另外,对酶的部分性质的研究结果表明,酶最适作用温度和pH分别为60℃和7.0,在50℃以下以及pH 5.5~7.5范围内稳定。  相似文献

Humic substances represent the main carbon reservoir in the biosphere, estimated at 1600 × 1015 g C. Due to their crucial role in reductive and oxidative reactions, sorption, complexation and transport of pollutants, minerals and trace elements, sustaining plant growth, soil structure and formation, and control of the biogeochemistry of organic carbon in the global ecosystem, humic substances are extremely important to environmental processes. Saprotrophic fungi active in the decomposition process of humic substances include mainly ascomycetes and basidiomycetes, which are both common in the upper layers of soils. White rot and litter decomposing fungi are the most important organisms in the degradation and mineralization of refractory organic matter (OM), whereas ascomycetes are mainly involved in the modification and polymerization of humic substances. The mechanisms of degradation probably involve mainly a variety of non specific oxidizing enzymes. This review provides an overview of the subject, while bridging two main disciplines: soil OM chemistry and fungal microbiology. It is aimed to highlight problems, unsolved questions and hypotheses.  相似文献

This research documents the removal of the dye Gris Lanaset G from aqueous solutions by fungal pellets. Adsorption of the dye by dead biomass pellets of Trametes versicolor was determined and compared with dye removal by enzymatic degradation. Six kinetic equations were fitted to the experimental adsorption data obtained. The results indicate that kinetics such as the Elovich equation, which considers that the rate-controlling step is the diffusion of the dye molecules, show the best fit. Nonlinear Langmuir and Freundlich equations were also fitted into the adsorption data, and it can be concluded that the adsorption equilibrium can be interpreted by the Langmuir isotherm. Adsorption plays an important role in the process of the elimination of color from textile wastewater, although not all of the elimination is due to this physical process when the microorganism is active. The removal of color (around 90%) with active microorganisms is greater than that obtained with the adsorption process.  相似文献
