Two experiments were conducted to determine whether the increased serum LH which occurs within 12 hr after a luteolytic dose of PGF2alpha is dependent upon changes in progesterone or estradiol secretion. In the first experiment, exogenous progesterone abolished the increase in serum LH caused by a subcutaneous injection of 25 mg PGF2alpha in diestrous heifers, but not in ovariectomized heifers. In the second experiment, progesterone pessaries were removed at 6 hr after a subcutaneous injection of 25 mg PGF2alpha. LH remained at pre-PGF2alpha values while the pessaries were in place, but began to increase within 1 hr after they were removed. Blood estradiol also remained at pre-PGF2alpha values until the pessaries were removed, and began to increase at 2 hr after pessary removal. We conclude that the increase in serum LH within 12 hr after PGF2alpha treatment in diestrous cattle is dependent upon withdrawal of progesterone; it is not due to increased serum estradiol.  相似文献

The objective of this study was to evaluate protocols for synchronizing ovulation in beef cattle. In Experiment 1, Nelore cows (Bos indicus) at random stages of the estrous cycle were assigned to 1 of the following treatments: Group GP controls (nonlactating, n=7) received GnRH agonist (Day 0) and PGF2alpha (Day 7); while Groups GPG (nonlactating, n=8) and GPG-L (lactating, n=9) cows were given GnRH (Day 0), PGF2alpha (Day 7) and GnRH again (Day 8, 30 h after PGF2alpha). A new follicular wave was observed 1.79+/-0.34 d after GnRH in 19/24 cows. After PGF2alpha, ovulation occurred in 19/24 cows (6/7 GP, 6/8 GPG, 7/9 GPG-L). Most cows (83.3%) exhibited a dominant follicle just before PGF2alpha, and 17/19 ovulatory follicles were from a new follicular wave. There was a more precise synchrony of ovulation (within 12 h) in cows that received a second dose of GnRH (GPG and GPG-L) than controls (GP, ovulation within 48 h; P<0.01). In Experiment 2, lactating Nelore cows with a visible corpus luteum (CL) by ultrasonography were allocated to 2 treatments: Group GPE (n=10) received GnRH agonist (Day 0), PGF2alpha (Day 7) and estradiol benzoate (EB; Day 8, 24 h after PGF2alpha); while Group EPE (n=11), received EB (Day 0), PGF2alpha (Day 9) and EB (Day 10, 24 h after PGF2alpha). Emergence of a new follicular wave was observed 1.6+/-0.31 d after GnRH (Group GPE). After EB injection (Day 8) ovulation was observed at 45.38+/-2.03 h in 7/10 cows within 12 h. In Group EPE the emergence of a new follicular wave was observed later (4.36+/-0.31 d) than in Group GEP (1.6+/-0.31 d; P<0.001). After the second EB injection (Day 10) ovulation was observed at 44.16+/-2.21 h within 12 (7/11 cows) or 18 h (8/11 cows). All 3 treatments were effective in synchronizing ovulation in beef cows. However, GPE and, particularly, EPE treatments offer a promising alternative to the GPG protocol in timed artificial insemination of beef cattle, due to the low cost of EB compared with GnRH agonists.  相似文献

The present work evaluated low-cost protocols for timed artificial insemination (TAI) in beef cattle. In Experiment 1, cycling nonlactating Nelore cows (Bos indicus, n=98) were assigned to the following groups: GnRH-PGF (GP) and GnRH-PGF-GnRH (GPG), whereas cycling (n=328, Experiment 2) or anestrus (n = 225, Experiment 3) lactating (L) cows were divided into 3 groups: GP-L, GPG-L and GnRH-PGF-Estradiol benzoate (GPE-L). In Experiment 4, lactating cows (n=201) were separated into 3 groups: GP-L, GPE-L and G/2PE-L. Animals from Experiment 1, 3 and 4 were treated (Day 0), at random stages of the estrous cycle, with 8 microg of buserelin acetate (GnRH agonist) intramuscularly (i.m.), whereas in Experiment 2 half of the cows received 8 and the other half 12 microg of GnRH (i.m.). Seven days later (D 7) all animals were treated with 25 mg of dinoprost trometamine (PGF2alpha, i.m.) except those cows from the G/2PE-L group which received only 1/2 dose of PGF2alpha (12.5 mg) via intravulvo-submucosa (i.v.s.m.). After PGF2alpha injection the animals from the control groups (GP and GP-L) were observed twice daily to detect estrus and AI was performed 12 h afterwards. The cows from the other groups received a second GnRH injection (D 8 in GPG-L and d9 in GPG groups) or one injection of estradiol benzoate (EB, 1.0 mg, D 8 in GPE-L group). All cows from GPG and GPG-L or GPE-L groups were AI 20 to 24 or 30 to 34 h, respectively, after the last hormonal injection. Pregnancy was determined by ultrasonography or rectal palpation 30 to 50 days after AI. In the control groups (GP and GP-L) percentage of animals detected in heat (44.5 to 70.3%) and pregnancy rate (20 to 42%) varied according to the number of animals with corpus luteum (CL) at the beginning of treatment. The administration of a second dose of GnRH either 24 (Experiment 2) or 48 h (Experiment 1) after PGF2alpha resulted in 47.7 and 44.9% pregnancy rates, respectively, after TAI in cycling animals. However, in anestrus cows the GPG treatment induced a much lower pregnancy rate (14.9%) after TAI. The replacement of the second dose of GnRH by EB (GPE-L) resulted in a pregnancy rate (43.3%) comparable to that obtained after GnRH treatment (GPG-L, 47.7%, Experiment 2). Furthermore, the use of 1/2 dose of PGF2alpha (12.5 mg i.v.m.s., Experiment 4) resulted in pregnancy rate (43.5%) similar to that observed with the full dose (i.m.). Both protocols GPG and GPE were effective in synchronizing ovulation in cycling Nelore cows and allowed approximately a 45% pregnancy rate after TAI. Additionally, the GPE treatment is a promising alternative to the use of GPG in timed AI of beef cattle, due to the low cost of EB when compared to GnRH agonists.  相似文献

A total of 402 two suckled postpartum beef cows at 2 locations (CSU and ECRC) were utilized in 3 trials to evaluate the effectiveness of a combination melengestrol acetate (MGA)-PGF2alpha estrus synchronization system in spring-calving cows. The cows were allocated by days post partum, body condition score (1 = emaciated; 9 = obese), sire breed, and dam age at the beginning of treatment to 1 of 2 treatments within location and trial: MGA-PGF2alpha (0.5 mg MGA/head/d for 14 d with 25 mg of PGF2alpha injected 17 d after MGA withdrawal), and unsynchronized controls. All cows were observed for estrus at 12-h intervals for at least 5 d post injection. Cows observed in standing estrus were inseminated 12 to 18 h later. There was a location effect on response to treatment that was attributed to differences in body condition score between locations so data were analyzed within a location. Body condition score at the CSU location was 5.7 compared with 4.0 at the ECRC location. The CSU MGA-PGF2alpha treated cows had higher (P < 0.05) 5-day estrus and 5-d pregnancy rates (78.6 and 61.0%, respectively) than the CSU controls (11.1 and 6.9%, respectively). Similar results for 5-d estrus and pregnancy rates but of decreased magnitude were also observed for the ECRC MGA-PGF2alpha treated (31.6 and 21.4%, respectively) cows compared with that of the ECRC controls (11.9 and 8.5%, respectively). The CSU MGA-PGF2alpha treated cows had higher (P < 0.05) 25- and 60-d pregnancy rates (82.5 and 94.8%) than the CSU controls (65.3 and 87.5%). The 25- and 60-d pregnancy rates were similar between the ECRC MGA-PGF2alpha treated cows and ECRC control cows. The MGA-PGF2alpha estrus synchronization system appears to contribute to pregnancy early in the breeding season in postpartum beef cows, although its effectiveness is limited by cow body condition.  相似文献

Effect of stage of the estrous cycle at the time of prostaglandin F(2alpha) (PGF(2alpha)) injection on subsequent reproductive events in beef females was studied in four trials involving 194 animals. Cycling animals were given two injections of 25 mg PGF(2alpha) 11 days apart or, in some cases, the interval was altered to allow the second injection to fall on a specific day of the cycle. Day of estrous cycle at time of the second injection was determined by estrous detection. Interval from the second PGF(2alpha) injection to the onset of estrus (interval to estrus) was shorter (P<.01) in heifers than in cows. Both cows and heifers injected on days 5 to 9 (early cycle) had a shorter (P<.01) interval to estrus (estrus = day 0) than did those injected on days 10 to 15 (late cycle). Conception rate was lower (P<.05) for early-cycle heifers than for late-cycle heifers inseminated by appointment at 80 hours. There was no significant difference in conception rate of early-or late-cycle heifers or cows inseminated according to estrous detection or early- or late-cycle cows inseminated at 80 hours. Progesterone concentrations in blood samples collected in heifers at 4-hour intervals after the second PGF(2alpha) injection on either day 7 or day 14 declined linearly (P<.05) through 36 hours. Day of the estrous cycle at PGF(2alpha) injection had no effect on rate of progesterone decline, even though heifers injected on day 7 had a shorter (P<.05) interval to estrus. All animals whose cycle length was not affected by the second PGF(2alpha) injection were treated on days 5 through 8 of the cycle, indicating that PGF(2alpha) was less effective in regressing the corpus luteum between days 4 and 9 of the cycle than later in the cycle.  相似文献

An experiment was conducted to study an estrous synchronization regimen that involved the use of prostaglandin F(2alpha) (PGF(2alpha)) alone or in combination with estradiol benzoate (EB) and appointment breeding. Fifty-three registered Angus yearling heifers and 167 registered Angus cows (3 to 9 yr of age) were given two injections of PGF(2alpha) 11 d apart. Forty-eight hours after the second injection of PGF(2alpha') a random sample consisting of 117 cows and heifers was injected with EB in sesame seed oil. All females in the herd were artificially inseminated 80 h after the second injection of PGF(2alpha). Nearly equal percentages (25.1 vs 25.6%; P = 0.93) of treated (EB) and control (no EB) females conceived at the appointment breeding. Use of EB tended to reduce (P = 0.06) natural service conception rate (83.4 vs 93.1% for EB and control groups, respectively). Estrous synchronization treatment did not affect interval from Day 1 of the breeding season to calving.  相似文献

The aim of this study was to compare two protocols for estrus synchronization in suckled beef cows over a 2 years period. The population studied consisted of 172 Charolais and 168 Limousin cows from 12 and 14 beef herds, respectively. In each herd, cows were allotted to groups according to parity, body condition score and calving difficulty. Cows in Group 1 (n=174) received PRID on Day-8 with estradiol benzoate (10mg, vaginal capsule), dinoprost on Day-4 (25mg i.m.), eCG on Day 2 (500 IU i.m.). The PRID was removed on Day-2 and cows were inseminated on Day 0, 56 h after PRID was removed. Cows in Group 2 (n=166) received GnRH on Day-10 (100 microg i.m.), dinoprost on Day-3 (25mg i.m.) and GnRH on Day-1 (100 microg i.m.), and were inseminated on Day 0, 16-24h after the last GnRH treatment. Plasma progesterone concentrations were measured to determine cyclicity prior to treatment (Days-20 and -10), to confirm the occurrence of ovulation (Days 0 and 10) and to determine the apparent early pregnancy rate (Days 0, 10 and 24). Pregnancy diagnosis was performed by ultrasonography between Days 35 and 45. The effects of various factors on ovulation, apparent early pregnancy and pregnancy rates were studied using logistic mixed models. There was no significant difference between Groups 1 and 2, respectively, for the cyclicity rate before treatment (80.5% versus 80.1%), for apparent pregnancy rate on Day 24 (62.1% versus 54.8%, P=0.09) and for pregnancy rate on Days 35-45 (53.8% versus 46.3%, P=0.16). Ovulation rate was higher (P<0.01) in Group 1 (90.8%) than in Group 2 (77.1%) and was affected by cyclicity prior to treatment in Group 2 but not in Group 1 (Group 1: 88.2% in anestrous cows versus 91.4% in cyclic cows; Group 2: 45.5% in anestrous cows versus 85.0% in cyclic cows, P interaction=0.05). Apparent pregnancy rates on Day 24 were influenced by the year of study (52.4% versus 68.8%, OR=2.12, P<0.01) and by the cyclicity before treatment (anestrous cows 46.3% versus cyclic cows 61.5%, OR=1.86, P<0.05). Pregnancy rates at 35-45 days were influenced by the year of study (44.2% versus 59.8%, OR=1.92, P<0.01). In conclusion, although pregnancy rates were similar for the two treatments, the combination of GnRH+PGF2alpha+GnRH in suckled beef cows induced a lower rate of ovulation than treatment with PRID+PGF2alpha, particularly in anestrous cows.  相似文献

Henderson and McNatty (Prostaglandins 9:779, 1975) proposed that LH from the preovulatory LH surge attached to receptors on luteal cells and that this attachment might protect the early corpus luteum from PGF2alpha induced luteolysis. To test this hypothesis, experiments were performed on heifers at day 10-12 of the cycle. Both jugular veins were catheterized and infusions of either saline (0.64 ml/min) or LH-NIH-B9 (10 microgram/min; 0.64 ml/min) were given. Saline infusions were from 0-12 h; LH infusions were for 10 h and were preceded by a 2 h saline infusion. All animals were given 25 mg PGF2alpha im at 6 h (6 h into the saline infusion and 4 h into the LH infusion). Blood samples were taken at 0.5 h, 1 h and 4 h intervals from 0-12h, 13-18 h and 12-42 h respectively. Serum was assayed for LH and progesterone by radioimmunoassay methods. Two animals received saline and two received LH in each experiment. Each treatment was replicated 6 times. LH infusion resulted in a mean serum LH of 75 ng/ml compared to 0.90 ng/ml in saline infused animals. This elevation of LH did not alter PGF2alpha induced luteolysis as indicated by decline in serum progesterone. This experiment does not support the hypothesis that the newly formed corpus luteum is resistant to PGF2alpha because of protection afforded by the proestrus LH surge.  相似文献

Synchronization of ovulation in dairy cows using PGF2alpha and GnRH   总被引:2,自引:0,他引:2  
This paper reports a new method for synchronizing the time of ovulation in cattle using GnRH and PGF(2alpha). In Experiments 1 and 2, lactating dairy cows (n=20) ranging from 36 to 280 d postpartum and dairy heifers (n=24) 14 to 16 mo old were treated with an intramuscular injection of 100 mug GnRH at a random stage of the estrous cycle. Seven d later the cattle received PGF(2alpha) to regress corpora lutea (CL). Lactating cows and heifers received a second injection of 100 mug GnRH 48 and 24 h later, respectively. Lactating cows were artificially inseminated 24 h after the second GnRH injection. Ovarian morphology was monitored daily by trans-rectal ultrasonography from 5 d prior to treatment until ovulation. In Experiment 3, the flexibility in the timing of hormonal injections with this synchronization protocol was evaluated by randomly assigning 66 lactating dairy cows to 3 different treatment groups. Lactating cows received the injection of PGF(2alpha) 48 (Group 1), 24 (Group 2), and 0 h (Group 3) prior to the second injection of GnRH, which was administered at the same time in each group to ensure the second injection of GnRH was given when follicles were at a similar stage of growth. In Experiments 1 and 2, the first injection of GnRH caused ovulation and formation of a new or accessory CL in 18 20 cows and 13 24 heifers. In addition, this injection of GnRH initiated or was coincident with initiation of a new follicular wave in 20 20 lactating cows and 18 24 heifers. Corpora lutea regressed after PGF(2alpha) in 20 20 cows and in 18 24 heifers. All cows and 18 24 heifers ovulated a newly formed dominant follicle between 24 and 32 h after the second injection of GnRH. Ten of 20 cows conceived to the timed artificial insemination. In Experiment 3, the conception rate in Groups 1 and 2 were greater than in Group 3, (55 and 46 % vs 11%, respectively). In summary, this protocol could have a major impact on managing reproduction in lactating dairy cows, because it allows for AI to occur at a known time of ovulation and eliminates the need for detection of estrus.  相似文献

One hundred and sixty-five suckled postpartum beef cows were utilized to evaluate the effectiveness of 2 estrus synchronization systems for the initiation and synchronization of estrus. The treatment groups consisted of 1) melengestrol acetate (MGA)-PGF2alpha (cows were given 0.5 mg MGA/head/day for 14 d with 25 mg PGF2alpha injected 17 d after the last day of MGA administration); 2) MGA-48-h calf removal (CR)-PGF2alpha (cows were given 0.5 mg MGA/head/day for 14 d with 48-h calf removal starting on the second day after completion of the MGA regimen plus 25 mg PGF2alpha administered 17 d after the last day of MGA); and 3) unsynchronized controls. Cows were assigned to treatments by the numbers of days post partum, body condition, age, and breed of sire. The cows were observed for estrus at 12-h intervals for 5 d after PGF2alpha administration and were artificially inseminated 12 to 18 h after the observed estrus. Both the MGA-PGF2alpha and MGA-CR-PGF2alpha treatments (64.8 and 61.8%) had greater (P < 0.05) 5-d estrus rates than the control treatments (34.5%). The synchronized pregnancy rate was greater (P < 0.05) for the MGA-CR-PGF2alpha than the control treatment.(52.7 vs 30.9%, respectively). The MGA-CR-PGF2alpha cows had a higher 25-d pregnancy rate than either the MGA-PGF2alpha (P < 0.05) or control cows (P < 0.08). Of the anestrous cows at the beginning of treatment, more MGA-CR-PGF2alpha (P = 0.1) and MGA-PGF2alpha cows were cyclic posttreatment than control cows (58.7 and 55.1 vs 44.7%, respectively), suggesting that treatment initiated estrous cycles in only a small number of the anestrous cows. Both MGA-PGF2alpha and MGA-CR-PGF2alpha treatments appear to be effective methods of synchronizing estrus in suckled postpartum beef cows. However, MGA-CR-PGF2alpha was more effective in establishing pregnancy earlier in the breeding season than MGA-PGF2alpha.  相似文献

This study compared the efficacy of two sources of PGF2alpha on the reproductive performance of virgin beef heifers, after synchronization of estrus using melengestrol acetate (MGA) and PGF2alpha. Angus-based heifers (n = 1002) in five herds were fed 0.5 mg per head per day of MGA for 14 days. Nineteen days after the last day of MGA feeding, heifers were randomly assigned to receive (i.m.) either 0.5 mg cloprostenol (n = 504; Estrumate, E) or 25 mg dinoprost tromethamine (n = 498; Lutalyse, L) as a source of exogenous PGF2alpha. Heifers were observed twice daily for 5 days for signs of estrus and artificially inseminated 8-12 h later, except in herd A, wherein animals not detected in estrus by 80 h after PGF2alpha were mass-mated and no longer monitored for signs of estrus. Estrumate and Lutalyse were equally (P > 0.1) effective among all response variables evaluated, including estrus response (E, 89% and L, 86%), conception rate (E, 67% and L, 67%), and synchronized pregnancy rate (E, 61% and L, 57%). Synchrony of estrus was not affected (P > 0.1) by PGF2alpha source, and peak estrus response occurred 60 h post-PGF for both treatments. Conception rate to timed insemination was not different (P > 0.1) among Estrumate- and Lutalyse-treated heifers within herd A (14%, 4/28 and 7%, 2/29, respectively). Herd had a significant (P < 0.05) effect on all indicators of reproductive performance. Conception rates within herds A and D were influenced by technician (P < 0.05), however, this effect was balanced across treatments and no treatment by technician interaction was detected. In conclusion, when administered 19 days after a 14-day MGA feeding period, cloprostenol and dinoprost tromethamine are equally efficacious for synchronous induction of a fertile estrus in virgin beef heifers.  相似文献

This study evaluated the effect of exogenous PGF(2)alpha on circulating LH concentrations in postpartum multiparous (n = 32) and primiparous (n = 46) Brahman cows. The cows were randomly allotted within parity and calving date to receive 0, 1, 2 or 3 mg im PGF(2)alpha (alfaprostol)/100 kg body weight (BW), with or without GnRH on Day 30 after calving. Blood samples were collected at weekly intervals from calving through treatment. Serum progesterone concentrations were determined using RIA procedures to assure that only anestrous cows were treated. Sterile marker bulls were maintained with cows on Coastal bermudagrass pastures until the first estrus was detected. Multiparous cows had a shorter (P < 0.05) interval from calving to estrus than did primiparous cows. Serum LH was affected by time (P < 0.0001), PGF(2)alpha dose (P < 0.0002), GnRH (P < 0.0001), parity by PGF(2)alpha dose (P < 0.0003), PGF(2)alpha dose by GnRH (P < 0.0009), parity by GnRH (P < 0.0008), and by parity by PGF(2)alpha dose by GnRH (P < 0.0005). Multiparous cows not receiving GnRH had higher mean serum LH (P < 0.02), LH peak pulse height (P < 0.03), and area under the LH release curve (P < 0.03) compared with primiparous cows. The number of LH pulses/6 h was greater (P < 0.06) in multiparous than primiparous cows, and was greater (P < 0.02) in multiparous cows receiving 3 mg/100 kg BW than in cows receiving 2 mg/100 kg BW, but not in the controls or in cows receiving 1 mg/100 kg BW. Exogenous GnRH resulted in increased (P < 0.0001) serum LH concentrations in all cows, and LH was enhanced (P < 0.0009) by simultaneous treatment with PGF(2)alpha. Primiparous cows had a greater response (P < 0.0005) to PGF(2)alpha and GnRH compared with multiparous cows. Pituitary release of LH in response to GnRH was enhanced by simultaneous exposure to PGF(2)alpha in Day 30 postpartum cows.  相似文献

Two studies were conducted to determine the changes in gonadotropin secretion associated with growth and development of the largest follicle and the ability of the largest ovarian follicle present on Day 5 following estrus to ovulate if luteal regression is induced. In both studies, cows received either saline (i.m.) or prostaglandin F(2)alpha (PGF(2)alpha; 25 mg i.m.) on the fifth day post estrus. Frequency of LH pulses declined (P<0.01) with increasing day of cycle, while pulse amplitude and duration increased (P<0.05) in saline-treated cows. In PGF(2)alpha-treated cows, LH remained as high frequency-low amplitude pulses. Secretory patterns of FSH were similar between the two groups. In Experiment 2, the largest ovarian follicle present was marked around its periphery with sub-epithelial injections of charcoal. In saline-treated cows, the size of the charcoal marked follicles generally decreased, indicating atresia. A corpus luteum was present within the area of a previously marked follicle in three PGF(2)alpha-treated cows. The size of the marked follicles either decreased or increased in the remaining PGF(2)alpha-treated cows, with ovulation occurring at a different site. In summary, PGF(2)alpha-induced luteal regression on the fifth day of estrus subsequently alters the frequency, amplitude and duration of LH pulses, but not FSH pulses, and the largest follicle present on Day 5 either increases or decreases in size or ovulates when PGF(2)alpha is given on Day 5 following estrus.  相似文献

Estrus synchronization contributes to optimizing the use of time, labor, and financial resources by shortening the calving season, in addition to increasing the uniformity of the calf crop. We determined whether acceptable pregnancy rates could be achieved after synchronization of ovulation and fixed-time artificial insemination (AI) in peripuberal replacement beef heifers using gonadotropin-releasing hormone (GnRH) and PGF2alpha. Crossbred heifers from two herds (MH, n=239; SS, n=330) were wintered at a single location. After a prebreeding examination revealed that 55 heifers had a reproductive tract score (RTS) of 1 (infantile reproductive tracts), they were culled and the remaining heifers were assigned randomly to one of three treatment groups: administration of 25mg PGF2alpha i.m. on Days -12 and 0 followed by estrus detection and insemination between 10 and 14 h after an observed estrus (Control; n=173); administration of 100 microg GnRH i.m. on Day -6, followed by 25 mg PGF2alpha i.m. on Day 0, then fixed-time AI and administration of 100 microg GnRH i.m. on Day +2 (GPG; n=172); and, treatment as for group GPG in addition to administration of 100 microg GnRH i.m. on Day -12 (GGPG; n=169). Bulls were introduced 10 days after AI for 60 days to breed heifers which did not conceive after AI (clean-up bulls). On Days -12, -6, and 0 transrectal ultrasonography was used to monitor ovarian structures in a subset of heifers (30 per treatment). At 30-35 days after AI, ultrasound was used to determine the presence of a viable fetus. Presence of a fetus and stage of pregnancy were determined via palpation per rectum 61-63 days after the conclusion of the breeding season. Heifers in the MH herd (309+/-1.9 kg) were heavier (P<0.001) than those in the SS herd (283+/-1.7 kg) at initiation of the breeding season. Synchronized pregnancy rates were greater (P<0.05) in GGPG (25.4%) and GPG (22.1%) than Control (12.7%) heifers. Pregnancy rates were 9, 21, 32, or 31% for heifers with RTS of 2, 3, 4, or 5, respectively. The average diameter of 22 follicles induced to ovulate in heifers treated with GnRH (GPG and GGPG treatments) was 14.2+/-0.8 mm (range=10.0-23.6 mm). In conclusion, a fixed-time ovulation synchronization program using GnRH and PGF2alpha improved pregnancy rates in peripuberal, lightweight replacement beef heifers.  相似文献

Half-life (t1/2), volume of distribution (Vd) and total body clearance (TBC) of 13,14-dihydro-15-keto PGF2 alpha (PGFM) were measured in order to determine optimal sampling frequency for accurate measurement of PGFM. Three yearling Holstein bulls (349.2 +/- 6.7 kg) and 3 yearling Holstein steers (346.7 +/- 7.0 kg) were utilized in a 3 X 3 Latin square design. Animals were given 0, 25 or 50 micrograms PGF2 alpha I.V.; blood samples collected every 2 min and plasma PGFM determined. The t1/2, Vd and TBC of PGFM were 2.3 +/- .2 min, 43.3 +/- 3.3 liters and 13.7 +/- 1.9 liters/min, respectively and were similar for 25 and 50 micrograms doses. To determine the relationship between endogenous PGFM and LH secretion in bulls, blood samples were collected every 2 min for 12 h in 4 yearling Angus bulls (489.1 +/- 11.6 kg). All animals elicited at least one LH surge and PGFM concentrations were measured in samples coincident with the LH surge. Mean plasma PGFM concentrations were greater prior to the LH surge than during the LH surge. In addition, mean plasma PGFM concentration and frequency of PGFM peaks appeared to increase prior to the LH surge suggesting an association between PGFM and pulsatile LH secretion in the bull.  相似文献

In a previous study we showed that estrus synchronization with 2 treatments of PGF2 alpha 13 d apart reduced conception rate at the synchronized estrus and that this reduction occurred mainly in cows in the early luteal phase at the second PGF2 alpha treatment. The objective of the present study was to determine the efficacy of a synchronization regimen in which PGF2 alpha was administered during the mid- to late-luteal phase to cows that had previously been synchronized with progesterone. Spring-calving cows from 6 dairy herds were used in this study. On Day -32 (Day 1 = the start of the breeding season), cows that had calved 2 or more weeks ago were randomly assigned to a synchronization (S, n = 732) or control (C, n = 731) group. Cows in Group S were treated with an intravaginal progesterone device (CIDR) for 12 d from Day -32 to Day -20, while those in Group C were left untreated. Similar percentages of cows in Group S (80.6%) and C (82.9%) had cycled by Day -7. The CIDR treatment synchronized the onset of estrus, resulting in 92.9% of cows in estrus being detected within 7 d after CIDR removal. Cows in Group S that had cycled by Day -7 were treated with PGF2 alpha (25 mg, i.m., Lutalyse) on Day -2. Cows in both groups that were anestrous on Day -7 were treated with a combination of progesterone and estradiol benzoate (EB) to induce estrus and ovulation (CIDR and a 10 mg EB capsule on Day -7, CIDR removal on Day -2, and injection of 1 mg EB 48 h after CIDR removal). The PGF2 alpha treatment synchronized the onset of estrus in 87.5% of the cows. Group S and C cows had similar conception rates to first (61.0 vs 58.3%) and second (58.4 vs 60.9%) AI; similar pregnancy rates over the AI period (82.8 vs 79.2%) and over the whole breeding season (91.9 vs 90.6%); and required a similar number of services per pregnancy to AI (1.7 vs 1.8). The interval from the start of the breeding season to conception for cows conceiving to AI or to combined AI and natural mating was shorter (P < 0.001) by 5.7 and 6.2 d, respectively, for the Group S cows. It is concluded that the treatment regimen tested in the present study achieved satisfactory estrus synchronization, had no detrimental effect on fertility at the synchronized estrus, and shortened the interval from start of the breeding season to conception.  相似文献

The use of CIDR-B or PRID in combination with prostaglandin F2alpha (PGF2alpha) for synchronizing estrus in embryo transfer recipients was evaluated in 2 experiments. In Experiment 1, virgin heifers (n=263) were synchronized using either a PRID (including estradiol benzoate capsule) or a CIDR-B in a combined program in which devices were inserted on Day 1, an injection of prostaglandin was given on Day 6, and devices were withdrawn on Day 7. The interval from device removal to the onset of estrus was significantly shorter for CIDR-B than for PRID-treated animals (50.44 vs 55.50 hours; P<0.003). The CIDR-B treatment resulted in a similar degree of synchrony to the PRID treatment (74.0 vs 70.4%; P=0.68). InExperiment 2, cows (n=95) and heifers (n=93) were allocated at random to be synchronized using a PRID (excluding estradiol benzoate capsule) plus PGF2alpha or a CIDR-B device plus PGF2alpha. The devices were inserted on Day 1, an injection of prostaglandin was given on Day 10 and the devices were removed on Day 12. Estrus was observed earlier following the CIDR-B treatment (43.50 vs 47.04 hours; P=0.01), but the degree of synchrony was similar (76.2 vs 76.3%; P>0.10) for the CIDR-B and PRID-treated animals. In both experiments, there were no significant differences in the proportions of animals observed in estrus, selected as embryo transfer recipients, or which achieved pregnancy consequent on embryo transfer between those synchronized using CIDR-B or PRID regimens. We conclude that the CIDR-B is a suitable device for synchronizing estrus in embryo transfer recipients.  相似文献
