多糖是蛹虫草Cordyceps militaris产生的重要活性成分。为提高其液体发酵胞外多糖的产量,通过单因素轮换法、方差分析、正交试验对蛹虫草菌株YCC-H产胞外多糖的发酵条件进行了优化。单因素结果表明液体发酵最适初始pH为5,最适葡萄糖浓度(质量分数)为6%,最适氮源为酵母膏,最适装液量为80 mL/250 mL,最适接种量(体积分数)为12%;在不同培养条件下菌体生物量变化较大;通过方差分析及正交试验进一步优化,结果显示:装液量(A)、氮源(B)、接种量(C)对蛹虫草产胞外多糖影响程度为B>A>C,最佳条件组合为A2B3C2,即装液量80 mL/250 mL、酵母膏为唯一氮源、接种量(体积分数)为12%,其余因素选择单因素中最佳水平。经验证,该菌株合成胞外多糖的质量浓度达247.06 mg/L,较未优化前胞外多糖的产量23.67 mg/L提高了9.43倍。  相似文献

文章为优化葡萄糖氧化酶产生菌的诱变育种和发酵工艺,将桔青霉CICC 40277作为生产菌株,通过对固、液培养基中菌种的生长和产酶活力进行比较,选取木薯叶为培养基质,经固体培养活化后,菌株的产酶活力最高达到66.48 U/g。然后采用2-脱氧-D-葡萄糖为菌株抑制剂,以甲基红为指示剂设计了一种高效、简便的筛选方法。通过紫外诱变育种,最终筛选得到一株高产菌株Uv-7,其产酶活力达到79.76 U/g,比原始菌株提高了20%。采用单因素试验对Uv-7菌株产葡萄糖氧化酶的最佳菌种活化条件进行优化,得到最适条件为:培养时间144 h、培养温度30℃、接种量20%(V/V);最适培养基组分为:以5.5 g木薯叶为固体基质,按照1∶1(W/V)的固液比添加盐溶液(g/L):MgSO4·7H2O 0.5、Na2HPO4 2.0、KCl 0.3、CaCO3 3.0、葡萄糖30、酵母粉6,当pH为6.0时,菌株经活化后产葡萄糖氧化酶的活性提高到了101.52 U/g,与原始菌株相比提高了52.7%...  相似文献

旨在以枯草芽胞杆菌Bacillus subtilis J为生产菌株,发酵生产β-甘露聚糖酶,通过优化产酶条件,以达到提高β-甘露聚糖酶产量的目的。利用DNS比色法检测β-甘露聚糖酶活力,采用单因素试验,研究碳氮源种类及碳氮源浓度、温度、pH、接种量和装液量对菌株Bacillus subtilis J发酵产β-甘露聚糖酶的影响,结合响应面试验设计确定菌株Bacillus subtilis J发酵产甘露聚糖酶的最优发酵培养条件。单因素试验和响应面试验得到最优的发酵条件为魔芋粉28 g/L,胰蛋白胨21 g/L,K2HPO4 6 g/L,MgSO4·7H2O 1 g/L,温度31℃,pH值8.5,接种量1%(体积分数),装液量50 mL/250 mL,发酵周期24 h。利用优化后的培养基生产β-甘露聚糖酶,其酶活力达到84.38 U/mL,是初始发酵培养基产酶活力的3.36倍。通过对发酵条件的优化,大幅度提高了β-甘露聚糖酶的产量,为其工业生产提供参考。  相似文献

为了对荷叶离褶伞产漆酶条件进行优化,在单因素实验基础上,通过最陡爬坡实验(PB)对培养基8因素进行筛选,获得影响产漆酶的3个显著性因素:葡萄糖,pH和KH2PO4;通过中心组合(CCD)设计及响应面分析确定了最优发酵条件:葡萄糖20.09g/L,酪蛋白1.5g/L,酵母提取物1.5g/L,MgSO4 3g/L,CuSO4 3.75mg/L,KH2PO4 3.97g/L,pH 4.98,VB1 0.1g/L,愈创木酚12mg/L,该条件下,漆酶酶活为829.83U/mL,较未优化对照提高46.6%.  相似文献

【目的】研究产低温脂肪酶菌株CZW001发酵培养基。【方法】在单因素试验的基础上, 采用Plackett-Burman (P-B)设计, Box-Behnken (B-B)设计和响应面试验设计(RSM), 在20 °C、pH 8.0、160?r/min发酵2 d条件下, 对发酵培养基进行优化。【结果】该菌株最适产酶培养基为(g/L): 葡萄糖7.68, 橄榄油21.93, 硫酸铵2.0, 磷酸二氢钾1.0, 硫酸镁0.27, 氯化钙0.3, 氯化钠20.0, 吐温-80 1.0。其最高酶活为62.8 U/mL, 比优化前提高了3.14倍。【结论】通过对产低温脂肪酶菌株CZW001发酵培养基优化研究, 明显提高低温脂肪酶活力。  相似文献

【目的】筛选一株海藻酸裂解酶高产菌株,并通过优化产酶条件提高海藻酸裂解酶活性。【方法】以海藻酸钠为唯一碳源的培养基,对福建漳州滨海土壤中的微生物进行筛选和分离,获得海藻酸裂解酶高产菌株;依据形态、生理生化特征及16S rDNA序列分析对目的菌株进行鉴定;然后通过单因素和正交试验对其产酶条件进行优化。【结果】十六烷基吡啶(CPC)染色得到4株透明圈与菌落直径比值(D/d)3的菌株;DNS法测定4菌株发酵液中海藻酸裂解酶活力,其中菌株SH-1的海藻酸裂解酶活性最高,达到315.52 U/mL;经形态、生理生化和16S rDNA测序鉴定,将其命名为Microbulbifer sp. SH-1;通过单因素和正交试验优化,确定该菌株最适产酶培养基为:海藻酸钠10 g/L,NaCl 5 g/L,(NH_4)_2SO_45g/L,MgSO_40.2g/L,K_2HPO_41g/L,FeSO_40.02g/L。对培养条件的进一步优化结果发现,在初始pH 7.5、温度32°C条件下,以1%的接种量将SH-1菌株接入50 mL优化培养基中,240 r/min转速下振荡培养24 h,SH-1菌株产酶最大活性可达757.90 U/mL,比优化前提高了2.4倍。【结论】SH-1最佳产酶条件的建立,为海藻酸裂解酶的大规模制备以及更深层次研究提供了试验基础和理论依据。  相似文献

为了提高野生型坎帕尼亚盐单胞菌株(Halomonas campaniensis sp. XH26)胞内Ectoine的积聚量,选用磁性纳米金属颗粒(Fe3O4 NPs),采用单因素实验、Plackett-Burman设计和响应面法分析菌株胞内的Ectoine积聚量和菌株生长量,并探讨Fe3O4 NPs应用于盐单胞菌发酵的可行性。单因素分析表明Fe3O4 NPs能促进菌株生长和促进菌株Ectoine的积聚,菌株生长对数期是最佳的Fe3O4 NPs添加时期。Plackett-Burman及响应面分析表明:优化条件下(0.05 g/L Fe3O4 NPs、1.53 mol/LNaCl和0.03 mol/LL-谷氨酸钠),摇瓶发酵Ectoine的积聚量可达640.28 mg/L,与野生菌株(391.35 mg/L)相比提高了63.61%。透射电镜显示Fe3...  相似文献

复合菌系WSD-5具有高效的分解能力和产酶能力,以探明WSD-5的协同分解机理和优化高效组合为目的,通过纯培养分离手段,获得了11株细菌和3株真菌。16S rDNA比对结果表明,细菌分别为Pseudomonas sp.、Pseudomonas aeruginosa、Achromobacter sp.、Stenotrophomonas sp.、Bacillus fusiformis、Bacillus cereus、Brevundimonas sp.、Ochrobactrum sp.、Cytophaga sp.、Benzo(a)pyrene-degrading bacter、Flavobacterium sp.的近缘种;26S rDNA比对结果表明3株真菌分别为Pseudallescheria boydii、Coprinus cinereus的近缘种。分离菌株中有4株细菌和3株真菌能在CMC平板上产生透明圈,但以糖化力法测定酶活结果只有3株真菌具有产酶能力。3株真菌的酶活动态测定结果,酶活的高峰均出现在7?14 d,并且呈现多峰变化;3株真菌的酶活种类表现为,滤纸酶活性、纤维素内切酶活性和外切酶活性均以菌株F1最高,分别达到了1.05、5.53和0.56 U/mL,β-葡萄糖甘酶活性和木聚糖酶活性以菌株FC最高,分别达到0.44和58.95 U/mL,其木聚糖酶活为F1最高值的6倍。  相似文献

苏云金芽孢杆菌BRC-LLP29为新型高效杀蚊菌株,应用快速有效的数学统计方法对其杀蚊毒力的发酵培养进行优化.通过单因素筛选确定最佳碳源为葡萄糖、麦芽糖、可溶性淀粉,氮源为typetone、大豆蛋白胨、干酪素;最佳金属离子为Mg2+、Al3+.采用二水平Placlkett-Burman设计对影响毒力的8因素进行显著性筛选,获得培养基成分中3个重要影响因子:葡萄糖、干酪素和Al2(SO43;运用爬坡路径法对这3种因子进行试验,获得3种重要因子的最适浓度范围;通过响应面分析法得到3个重要因子的交互作用和最佳条件,确定BRC-LLP29菌株最佳毒力水平的发酵培养基为:葡萄糖19.8g/L、干酪素28.4g/L、Al2(SO431.2g/L、MgSO4 2g/L、K2HPO4 3g/L、CaCO3 0.5g/L,优化后毒力水平达到致死率61.11%,与响应面数学模型的预测值只有5.91%的误差.发酵条件优化结果表明:发酵温度为31°,发酵初始pH为7.0,摇瓶装量为40mL/250mL三角瓶,每瓶的接种量为3.5%,发酵72h,对致倦库蚊最终致死率达到最高为83.33%.  相似文献

【目的】以发酵液纤溶酶活力为指标,优化海洋来源的链霉菌菌株MY0504的发酵条件。【方法】在菌株生长曲线及单因素试验基础上,采用Plackett-Burman设计筛选影响纤溶酶活性的主要因素,进一步用最陡爬坡试验及Box-Behnken中心组合设计法优化发酵条件。【结果】纤溶酶活性最高的发酵条件为:葡萄糖21.68 g/L,酵母粉25.31 g/L,NaCl5.0 g/L,K_2HPO_4·3H_2O3.0 g/L,MgSO_4·7H_2O 0.5 g/L,FeSO_4·7H_2O 0.02 g/L,装液量50 mL(250 mL摇瓶),接种量10%(体积比),初始pH 7.5,温度24°C,转速200 r/min,培养时间4.5 d。发酵液纤溶酶活性可达2 190.6 U/mL。【结论】确定了MY0504菌株产纤溶酶的最优发酵条件,为该酶的进一步分离纯化及性质研究奠定基础。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
