 The growth responses of lentil (Lens esculenta L. cv. Laird) and two wheat cultivars (Triticum aestivum L. cv. Laura and Neepawa) to Glomus clarum NT4 in soil containing indigenous arbuscular mycorrhizal fungi (AMF) and fertilized with phosphorus at different (0, 5, 10, 20 ppm) levels was studied in a growth chamber. Soil was inoculated with a monospecific culture of G. clarum NT4 to provide an inoculant:indigenous AMF ratio of ca. 1 : 100. The shoot and root growth, and AMF colonization levels of NT4-inoculated lentil were significantly (P≤0.05) greater than the appropriate control plants in the unfertilized soil at 48 days after planting (DAP). At 95 DAP, NT4 inoculation had significantly increased the shoot dry weight (P≤0.08) and AMF colonization (P≤0.05) of lentil plants receiving 5 mg P kg–1 soil, whereas 20 mg P kg–1 soil reduced the shoot growth of NT4-inoculated plants. The NT4 inoculant had no effect (P≤0.05) on shoot P content, but increased (P≤0.08) the P-use efficiency of lentil plants receiving 5 mg P kg–1 soil. In contrast to the inoculant's effect on lentil, NT4 generally had no positive effect on any of the parameters assessed for wheat cv. Laura at any P level at 48 or 95 DAP. Similarly, there was no positive effect of NT4 on shoot or root growth, or AMF colonization of wheat cv. Neepawa plants at any P level at 48 DAP. However, NT4 inoculation increased the grain yield of Neepawa by 20% (P≤0.05) when fertilized with 20 mg P kg–1 soil. This yield increase was associated with a significant (P≤0.05) reduction in root biomass and a significant (P≤0.05) increase in the grain P content of inoculated plants. Thus, NT4 appears to have a preference for the Neepawa cultivar. Our results show that lentil was more dependent on mycorrhizae than wheat and responded to an AMF inoculant even in soil containing high levels of indigenous AMF. It might, therefore, be possible to develop mixed inoculants containing rhizobia and AMF for field production of legumes. Accepted: 22 February 1997  相似文献

K20 is a novel amphiphilic aminoglycoside capable of inhibiting many fungal species. K20's capabilities to inhibit Fusarium graminearum the causal agent wheat Fusarium head blight (FHB) and to this disease were examined. K20 inhibited the growth of F. graminearum (minimum inhibitory concentrations, 7.8–15.6 mg L?1) and exhibited synergistic activity when combined with triazole and strobilurin fungicides. Application of K20 up to 720 mg L?1 to wheat heads in the greenhouse showed no phytotoxic effects. Spraying wheat heads in the greenhouse with K20 alone at 360 mg L?1 lowered FHB severity below controls while combining K20 with half–label rates of Headline (pyraclostrobin) improved its disease control efficacy. In field trials, spraying K20 at 180 mg L?1 and 360 mg L?1 combined with half-label rates of Headline, Proline 480 SC (prothioconazole), Prosaro 421 SC (prothioconazole + tebuconazole), and Caramba (metconazole) reduced FHB indices synergistically. In addition, the K20 plus Proline 480 SC combination reduced levels of the mycotoxin deoxinivalenol by 75 % compared to the control. These data suggest that K20 may be useful as a fungicide against plant diseases such as FHB particularly when combined with commercial fungicides applied at below recommended rates.  相似文献

Summary Shool cultures of apple cv. Pinova were contaminated with faint pink pigmented yeast. Yeast isolates were identified as Rhodotorula slooffiae with standard physiological methods and molecular analysis. Growth of isolated yeasts was tested against different fungicides. The following fungicides inhibited the growth of yeast isolates, and were not phytotoxic to apple shoots at concentrations lower than the minimal phytotoxic concentrations (MPC): ProClin? 300, mancozeb, triforine, myclobutanil, thiabendazole, mancozeb+zoxamid, and silver nitrate. Some fungicides inhibited growth of yeasts, but were phytotoxic. These included miconazole, PPM, copper sulfate, potassium sorbate, and cycloheximide. Benomyl was not phytotoxic, but was effective only at high doses. Decontamination of shoots was achieved using a combination of two treatments. Shoots were first soaked in half-strength Murashige and Skoog (MS) liquid medium containing silver nitrate (588μM) and Silvet 77 (0.01%) for 1–2h, and then transferred to a solidified MS medium containing both mancozeb (15mgl−1) and thiabendazole (40mgl−1).  相似文献

The wheat seedling endonucleases WEN1 and WEN2 dependent on Mg2+, Ca2+, and S-adenosyl-L-methionine (SAM) and sensitive to the substrate DNA methylation status have an expressed processing action. The enzymes hydrolyze DNA at a few subsequent stages: first, they split λ phage DNA specifically at CNG-sites (WEN1) with liberation of large fragments; second, they hydrolyze these fragments to 120–140 bp oligonucleotides that finally are hydrolyzed to very short fragments and mononucleotides. Initial stages of DNA hydrolysis may proceed in the absence of Mg2+, but subsequent hydrolysis stages are very strongly stimulated by Mg2+. It cannot be ruled out that modulation of enzymatic activity with Mg2+ and probably with DNA fragments formed is associated with reorganization of the structure of eukaryotic (wheat) endonucleases with respective changes in their catalytic properties and site specificity of action. Michaelis constant value for WEN1 endonuclease on hydrolysis of methylated λ phage DNA containing Cm5CWGG and Gm6ATC sites is four-fold lower compared with that observed on hydrolysis of unmethylated λ phage DNA. This may indicate that affinity of WEN1 enzyme to methylated DNA is higher than that to unmethylated DNA. In the presence of SAM, the Michaelis constant for WEN2 on the DNA hydrolysis stage characterized by formation of 120–140 bp fragments is decreased, but for WEN1 it is increased by 1.5–2.0-fold. This means that SAM inhibits WEN1 but stimulates WEN2. Thus, wheat endonucleases WEN1 and WEN2 differ significantly in affinities to substrate DNAs with different methylation status, in velocities of DNA hydrolysis, and time of production of DNA fragments of similar length. It seems that the investigated plant endonucleases can hydrolyze DNA in the nucleus as well to both large and very short fragments including mononucleotides, that is, in particular, essential for utilization of cell nucleic acid material during apoptosis.  相似文献

The effects of the strobilurin fungicides Juwel, Juwel Top and Amistar on the deoxynivalenol contamination of winter wheat was studied in field experiments. In general, the application of strobilurins during stem elongation and inflorescence emergence of wheat resulted in increased deoxynivalenol contents in kernels as compared with the untreated control. This stimulating effect can be reversed by a following azole fungicide applied within a time period of one week during the stages of flowering.  相似文献

A method for the determination of the mycotoxin citrinin (CT) in rye, wheat and barley is described. The proposed method is based on ethyl acetate extraction, solid phase clean-up (SPE) on aminopropyl columns and reversed phase high performance liquid chromatography with fluorescence detection (RP-HPLC-FLD). The limits of detection and quantification of CT amounted to 0.6–0.9 μg/kg and 1.7– 3.3 μg/kg with mean recovery rates in the range of 77–92% (RSD 4.8–5.5%). A field experiment was conducted for 2 years on an alkaline calcareous seleniferous soil to study the effect of different levels of gypsum (0.2 – 3.2 t ha−1) applied to wheat only in the first year on Se accumulation by wheat (Triticum aestivum L.) – rice (Oryza sativa L.) cropping sequence. With gypsum application, grain yield of both rice and wheat crops increased by 0.4 – 0.5 t ha−1; the increase in straw yield was 0.4 – 1.1 t ha−1. Significant reduction in Se accumulation by wheat was observed with gypsum application up to 0.8 t ha−1 and its residual effect was evident on the following crops for 2 years. Reduction in Se accumulation varied from 53 to 64% in wheat grain, 46 to 49% in wheat straw, 35 to 63% in rice grain and 36 to 51% in rice straw with an application of gypsum at 0.8 t ha−1. A corresponding increase in S concentration was observed. In the gypsum-treated plots, the ratio of S:Se increased by 6 – 8 times in wheat and 3 – 6 times in rice. Reduction in Se accumulation by crop plants through gypsum application may help in lowering the risk of Se over-exposure of animals and humans that depend on diet materials grown on high selenium soils. Natural Occurrence of Mycotoxins in Staple Cereals from Ethiopia   总被引:1,自引:0,他引:1  
The occurrence of mycotoxins in barley, sorghum, teff (Eragrostis tef) and wheat from Ethiopia has been studied. Samples were analyzed for aflatoxin B1 (AFB1), ochratoxin A (OTA), deoxynivalenol (DON), nivalenol (NIV) and zearalenone (ZEN) using high performance liquid chromatography (HPLC) and for fumonisins (FUM) using enzyme linked immunosorbent assay (ELISA). AFB1 and OTA were detected in samples of all the four crops. AFB1 was detected in 8.8% of the 352 samples analyzed at concentrations ranging from trace to 26 μg kg−1. OTA occurred in 24.3% of 321 samples at a mean concentration of 54.1 μg kg−1 and a maximum of 2106 μg kg−1. DON occurred in barley, sorghum and wheat at 40–2340 μg kg−1 with an overall incidence of 48.8% among the 84 mainly ‘suspect’ samples analyzed; NIV was co-analyzed with DON and was detected at 40 μg kg−1 in a wheat sample and at 50, 380, and 490 μg kg−1 in three sorghum samples. FUM and ZEN occurred only in sorghum samples with low frequencies at concentrations reaching 2117 and 32 μg kg−1, respectively. The analytical results indicate higher mycotoxin contamination in sorghum, which could be related to the widespread storage of sorghum grain in underground pits leading to elevated seed moisture contents. A field experiment was carried out to investigate the effects of presoaking the wheat grains (Triticum aestivum L.) in 33 or 66 mM NaCl and indolyl-3-acetic acid (IAA at 50 g m−3), gibberellic acid (GA3 at 100 g m−3) or kinetin (100 g m−3) on some tolerance criteria in wheat flag leaf at different stages of development. At various stages of flag leaf development pretreatment with 33 or 66 mM NaCl decreased degree of succulence (particularly 66 mM), relative growth rate, net assimilation rate, relative water content, K+ content and K+/Na+ ratio and at the same time induced accumulation of abscisic acid and Na+. In the majority of cases grain pretreatment with GA3 or kinetin and to a lesser extent with IAA alleviated either partially or completely the deleterious effect of salinity on the above mentioned parameters.  相似文献

Adult rove beetles (Staphylinidae) were sampled every 7–14 days from one winter wheat field located in each of the four major wheat growing regions of Oklahoma during the 1999–2000 and 2000–2001 growing seasons. The number of cereal aphids per tiller, wheat plant growth stage, and wheat tiller density also were estimated. A total of 12 genera representing 13 species of beetles were collected from the field. The density of rove beetles was generally low, ranging from 0.003 beetles per m2 in fall to 0.106 beetles per m2 in spring. Rove beetle communities differed among seasons. After accounting for the effect of season, there was no statistically significant association between rove beetle community structure and field location, aphid density, wheat plant growth stage, or wheat plant density. Most rove beetle species showed no association with a particular season, however, Aleochara notula Erichson, Lathrobium sp., and Oxypoda sp. were present predominantly in fall, while Bisnius inquitus Erichson was associated with winter. Oxypoda sp. was the most abundant rove beetle in winter wheat fields in spring and was relatively abundant in winter, but was not collected from wheat fields in fall. Tachyporus jocosus Say was present in wheat fields during all seasons. T. jocosus was the most abundant rove beetle species in the winter wheat fields in fall and winter and was the second most abundant species during spring.  相似文献

The two fungicides azoxystrobin and fenpropimorph are used against powdery mildew and rust diseases in wheat (Triticum aestivumL). Azoxystrobin, a strobilurin, inhibits fungal mitochondrial respiration and fenpropimorph, a morpholin, represses biosynthesis of ergosterol, the major sterol of fungal membranes. Although the fungitoxic activity of these compounds is well understood, their effects on plant metabolism remain unclear. In contrast to the fungicides which directly affect pathogen metabolism, benzo(1,2,3) thiadiazole-7-carbothioic acid S-methylester (BTH) induces resistance against wheat pathogens by the activation of systemic acquired resistance in the host plant. In this study, we monitored gene expression in spring wheat after treatment with each of these agrochemicals in a greenhouse trial using a microarray containing 600 barley cDNA clones. Defence-related genes were strongly induced after treatment with BTH, confirming the activation of a similar set of genes as in dicot plants following salicylic acid treatment. A similar gene expression pattern was observed after treatment with fenpropimorph and some defence-related genes were induced by azoxystrobin, demonstrating that these fungicides also activate a defence reaction. However, less intense responses were triggered than with BTH. The same experiments performed under field conditions gave dramatically different results. No gene showed differential expression after treatment and defence genes were already expressed at a high level before application of the agrochemicals. These differences in the expression patterns between the two environments demonstrate the importance of plant growth conditions for testing the impact of agrochemicals on plant metabolism.  相似文献

Zearalenone is a mycotoxin mainly produced by severalFusarium species, which are known to colonize grains in temperate climates. The purpose of the study is to provide a reliable isotope dilution method for the quantification of this mycotoxin. A derivative of the analyte to be used as standard is obtained by reaction with acetic anhydride, which is available in two pure isotopic forms, a protonated (“light”) and a hexadeuterated (“heavy”). The derivatized standards are added to the matrix split intwo parts. Then, the derivatization procedure is repeated on both matrices derivatizing the part containing the “heavy” labelled standard with the “light” acetic anhydride and the part containing the “light” labelled standard with the “heavy” acetic anhydride. Both extracted mixtures are analyzed by LC/MS, monitoring the “light” and the “heavy” labelled analytes and using the former as standard for the latter in one case and viceversa in the other case. The effect of applying Lactobacillus buchneri (LB), alone or in combinations with L. plantarum (LP) and yeasts at ensiling, on the ensiling fermentation and aerobic stability of wheat and sorghum silages was studied under laboratory conditions. Treatments comprised LB, LP, yeasts, LB + yeasts, LP + yeasts, LB + LP and B-589 (a lactic acid bacterial strain isolated from wheat silage in Israel) alone. The treatments were also applied to sterilized aqueous extracts of wheat which were incubated at 30°C for 10 days. The pH of all treatments was below 4.0 already on day 4 of the experiment. Silages treated with LB had higher acetic acid concentrations than those treated with LP: 32–34 vs 16–18, and 28–34 vs 4–7 g kg−1 in the experiments with wheat and sorghum, respectively. Similar results were obtained in wheat extracts. In the aqueous phase, marked differences in pH decrease were noticed among the treatments: 4.4 in LB, 6.0 in the yeast, and 3.7 in LP and B-589 (from day 3 and onwards). In both crops LB resulted in aerobically stable silages when applied alone or with LP and yeasts, whereas LP resulted in unstable silages upon aerobic exposure; the stability of the LB-treated silages is attributed to the higher acetic acid concentrations. Summary Thidiazuron (TDZ) is a substituted phenylurea which has been shown to be an efficacious regulator of in vitro morphogenesis of many dicot plant species. However, information regarding the effect of TDZ on in vitro regeneration of monocot species is limited. We investigated the effects of TDZ on in vitro regeneration of barley (Hordeum vulgare L.) and wheat (Triticum aestivum L.) and found that it promoted shoot regeneration from callus in these two important cereal species. Plant regeneration from calluses derived from immature embryo culture of barley and wheat was observed in regeneration media with a wide range of TDZ concentrations (0.045–45 μM). Shoot regeneration from barley calluses was the highest (38.3% for cv. Golden Promise) at 4.5 μM (1 mg l−1) TDZ, while the optimal TDZ level for wheat regeneration seemed to be 0.9 μM (0.2 mg l−1) (87% for cv. Bob White and 49.4% for cv. Hi Line). Roots developed normally when the regenerated wheat and barley shoots from TDZ-containing media were transferred to the rooting medium. Comparison with other plant growth regulators commonly used in wheat and barley regeneration media suggested that TDZ was among the best for in vitro regeneration of wheat and barley. MgMfs1, a major facilitator superfamily (MFS) gene from the wheat pathogenic fungus Mycosphaerella graminicola, was identified in expressed sequence tag (EST) libraries. The encoded protein has high homology to members of the drug:H(+) antiporter efflux family of MFS transporters with 14 predicted transmembrane spanners (DHA14), implicated in mycotoxin secretion and multidrug resistance. Heterologous expression of MgMfs1 in a hypersensitive Saccharomyces cerevisiae strain resulted in a strong decrease in sensitivity of this organism to a broad range of unrelated synthetic and natural toxic compounds. The sensitivity of MgMfs1 disruption mutants of M. graminicola to most of these compounds was similar when compared to the wild-type but the sensitivity to strobilurin fungicides and the mycotoxin cercosporin was increased. Virulence of the disruption mutants on wheat seedlings was not affected. The results indicate that MgMfs1 is a true multidrug transporter that can function as a determinant of pathogen sensitivity and resistance to fungal toxins and fungicides.  相似文献

Wheat–soybean is one of the most dominant cropping systems on the Vertisols of central India. Cultivation of durum wheat in winter season (November to April) has a considerable potential due to congenial climate, while soybean in rainy season (June to October) has witnessed a phenomenal growth in the last two decades in the region. Beside including a legume (soybean) in sequence with a cereal crop (wheat), combined use of available organic sources along with chemical fertilizers may prove beneficial for long-term productivity and sustainability of the system. A long-term experiment was conducted during 1995–2000 on the fine-textured Vertisols at Indore, India to study the effect of combined use of farmyard manure (FYM), poultry manure, vermicompost and biofertilizers (Azotobacter + phosphate solubilizing bacteria) with 0.5 and 1.0 NPK (120 kg N + 26.2 kg P + 33.3 kg K ha−1) on wheat, and residual effect on following soybean. Grain yield of aestivum wheat in the initial 2 years and durum wheat in the later 3 years was significantly increased with 0.5 NPK + poultry manure at 2.5 t ha−1 or FYM at 10 t ha−1 compared with 0.5 NPK alone, and was on par with 1.0 NPK. However, the highest productivity was obtained when these organic sources were applied along with 1.0 NPK. Quality parameters of durum wheat viz protein content, hectolitre weight and sedimentation value showed improvement, and yellow berry content was significantly lower with combined use of NPK + organic sources compared with NPK alone and control. Soybean did not show much response to residual effect of nutrient management treatments applied to wheat. Wheat gave higher profit than soybean, particularly in the later years due to lower grain yields and market price of soybean. However, the superiority of FYM as well as poultry manure along with 1.0 NPK was evident on the overall profitability of the system. Various soil fertility parameters including chemical and biological properties showed conspicuous improvement over the initial status under the treatments of FYM and poultry manure. Sustainability yield index was maximum under 1.0 NPK, followed by 1.0 NPK + poultry manure or FYM. It was concluded that application of available organic sources, particularly FYM and poultry manure along with full recommended dose of NPK fertilizers to wheat was essential for improving productivity, grain quality, profitability, soil health and sustainability of wheat–soybean system.  相似文献

The in vitro competency of mature cereal embryos (winter, spring and durum wheats, oat, barley and triticale) was assessed for direct multiple shoot production on culture media containing the plant growth regulators, thidiazuron (TDZ) and/or 6–benzylaminopurine (BAP). Mature embryos of CDC Dancer oat showed the best response, with 69 shoots per explant on culture medium containing a combination of 4.5 μM TDZ and 4.4 μM BAP. TDZ alone induced about 16 shoots per explant from the oat. Among the wheat genotypes, durum wheat showed the most number of shoots (35) per explant on culture medium containing 4.5 μM of TDZ and 4.4 μM of BAP. With TDZ alone, shoot regeneration for durum wheat ranged from 27–32 shoots per explant. The regeneration frequency from the three winter wheat genotypes ranged from 11–25 shoots per explant and was highest on culture medium containing 9.1 μM TDZ and 4.4 μM BAP. The latter culture medium was also effective for a triticale genotype, inducing 34 shoots per explant. The regeneration from mature embryos of barley genotypes ranged from 5–9 shoots per explant. The mature embryos of all the cereals tested could be used for in vitro regeneration with TDZ and TDZ+BAP combinations.  相似文献

This study provides a first attempt investigation of a serie of studies on the ability of Anthracophyllum discolor, a recently isolated white-rot fungus from forest of southern Chile, for the treatment of soil contaminated with pentachlorophenol (PCP) to future research on potential applications in bioremediation process. Bioremediation of soil contaminated with PCP (250 and 350 mg kg−1 soil) was investigated with A. discolor and compared with the reference strain Phanerochaete chrysosporium. Both strains were incorporated as free and immobilized in wheat grains, a lignocellulosic material previously selected among wheat straw, wheat grains and wood chips through the growth and colonization of A. discolor. Wheat grains showed a higher growth and colonization of A. discolor, increasing the production of manganese peroxidase (MnP) activity. Moreover, the application of white-rot fungi immobilized in wheat grains to the contaminated soil favored the fungus spread. In turn, with both fungal strains and at the two PCP concentrations a high PCP removal (70–85%) occurred as respect to that measured with the fungus as free mycelium (30–45%). Additionally, the use of wheat grains in soil allowed the proliferation of microorganisms PCP decomposers, showing a synergistic effect with A. discolor and P. chrysosporium and increasing the PCP removal in the soil.  相似文献

Since 1999 the harvest of grain in Mecklenburg-Vorpommern has been investigated yearly as to its Fusaria infection rate and contamination with mycotoxins. 376 samples, most of them winter wheat, were tested for the mycotoxins zearalenone and deoxynivalenol in 1999–2003. The investigations were carried out with an ELISA; some results were confirmed by a chromatographic method (HPLC). In 2002 the Fusaria infection rate was graded as moderate (17,4%), in the other years as low (max. 9%). Zearalenone was detected only in 2,7% of the samples (max.236 μg/kg). 9,6% of samples contained deoxynivalenol (max. 1,2 mg/kg). Therefore the mycotoxin contamination in grain, harvested in Mecklenburg-Vorpommern 1999–2003 and the risk for consumers and animals as well is thought to be low. These results should help the farmers to understand that mycotoxins are not necessarily the sole cause of depressions in yield and loss of animals. It should be possible to avoid a false interpretation of mycotoxin findings.

The objectives of this study were to evaluate the fumonisin production by 16 F. verticillioides strains on corn cultures and the effect of quintozene and fludioxonil + metalaxyl-M fungicides on “in vitro” mycelial growth on agar. In addition, the effect of fludioxonil + metalaxyl-M on fumonisin production in defined liquid culture medium was analyzed. Fumonisin B1 levels on corn cultures ranged from 2.41 to 3996.36 μg/g and the F. verticillioides 103F strain produced the highest level (3996.36 ± 390.49 μg/g, P < 0.05). F. verticillioides strains were inoculated in potato dextrose agar with the addition of quintozene (75 to 9,375 μg/ml) and fludioxonil + metalaxyl-M (1.5 + 0.6 to 187.5 + 75 μg/ml) in order to evaluate the effect of these fungicides on “in vitro” mycelial growth. The F. verticillioides strains showed great variability concerning ED50 values, which were below the recommended application dose for quintozene, but above that for fludioxonil + metalaxyl-M. Moreover, fungicide addition to the culture medium increased mean FB1 levels compared to the control, suggesting the importance of focusing on the effect of fungicides on mycotoxin production as well as on the phytopathogen control.  相似文献
