Assessing immunological function in toxicological studies of avian wildlife   总被引:5,自引:2,他引:3  
Laboratory and field studies have demonstrated that the immunesystem is sensitive to environmental contaminants. Testing protocolshave been developed to screen for immunotoxic effects and elucidatemechanisms of toxicity in laboratory rodents. Similar methodshave been applied to wildlife species in captivity and the wild.Several epizootics in wildlife have been associated with elevatedexposure to contaminants. This paper discusses immunotoxicologicaltechniques used in studies of avian wildlife. Measurements ofimmunological structure include peripheral white blood cellcounts and the mass and cellularity of immune organs such asthe thymus, spleen, and bursa of Fabricius. While contaminantscan alter these measures of immunological structure, such measuresdo not directly assess how the immune system functions, i.e.,responds to specific challenges. The two most commonly usedin vivo immune function tests in birds are the phytohemagglutinin(PHA) skin response for T cell-mediated immunity and the sheepred blood cell (SRBC) hemagglutination assay for antibody-mediatedimmunity. In vitro tests of immune function in avian wildlifeinclude proliferation of lymphocytes in response to variousmitogens and phagocytosis of fluorescent particles by monocytes.While optimization of in vitro techniques for wildlife speciesis often time-consuming, these assays usually require only asingle blood sample and can elucidate mechanisms of toxicity.In immunological studies of wildlife, investigators should considerfactors that may influence immune responses, including age,body condition, date, developmental stage of the immune system,and time required for the progression of immune responses.  相似文献

Predation risk may affect the allocation priorities of limitingresources by potential prey. Investment in immune function shouldreceive reduced priority, when hosts are exposed to predatorsbecause of the costs of immune function. We tested this hypothesisby randomly exposing adult house sparrows, Passer domesticus,to either a cat, Felis catus, or a rabbit, Oryctolagus cuniculus,for 6 h while assessing their ability to raise a T-cell–mediatedimmune response to a challenge with phytohemagglutinin. Sparrowsexposed to a cat had a significant reduction of, on average,18% and 36% in T-cell response in two different experimentscompared with sparrows that were exposed to a rabbit. In a fieldexperiment with a barn owl, Tyto alba, or a rock dove, Columbalivia, placed next to a nest-box during laying, we found a meanreduction in T-cell–mediated immune response of 20%. Inmales, the reduction in cell-mediated immune response owingto cat exposure increased with increasing size of the badge,which is a secondary sexual character, but only during the breedingseason. In a third experiment, house sparrows were either exposedto a barn own, T. alba, or a rock dove, C. livia, and developmentof malarial infections was recorded during the following 6 weeks.Individual sparrows exposed to a predator had a higher prevalenceand intensity of Haemoproteus malarial infection than did controlindividuals. Therefore, exposure to predators reduced theirability of hosts to cope with parasitism mediated through effectson immune function.  相似文献

We investigated variation in the primary sex ratio within andbetween 14 populations of roe deer (Capreolus capreolus) inrelation to maternal body condition. The sex ratio was increasinglymale biased as average maternal body weight decreased. Thisrelationship did not vary according to the population consideredand was not affected by the litter size produced. This relationshipwas also apparent within populations. These results indicatethat, where environmental conditions are limiting, roe doestend to produce male-biased litters. Dispersal is more commonand occurs at an earlier age among male juveniles in this species,particularly as density increases and resources become increasinglyscarce. Thus, we suggest that where females experience environmentalstress, they tend to produce male kids to avoid potential futurelocal resource competition posed by female offspring. Knowledge of the role of origin‐related, environmental, sex, and age factors on host defence mechanisms is important to understand variation in parasite intensity. Because alternative components of parasite defence may be differently sensitive to various factors, they may not necessarily covary. Many components should therefore be considered to tackle the evolution of host–parasite interactions. In a population of barn owls (Tyto alba), we investigated the role of origin‐related, environmental (i.e. year, season, nest of rearing, and body condition), sex, and age factors on 12 traits linked to immune responses [humoral immune responses towards sheep red blood cells (SRBC), human serum albumin (HSA) and toxoid toxin TT, T‐cell mediated immune response towards the mitogen phytohemagglutinin (PHA)], susceptibility to ectoparasites (number and fecundity of Carnus haemapterus, number of Ixodes ricinus), and disease symptoms (size of the bursa of Fabricius and spleen, proportion of proteins that are immunoglobulins, haematocrit and blood concentration in leucocytes). Cross‐fostering experiments allowed us to detect a heritable component of variation in only four out of nine immune and parasitic parameters (i.e. SRBC‐ and HSA‐responses, haematocrit, and number of C. haemapterus). However, because nestlings were not always cross‐fostered just after hatching, the finding that 44% of the immune and parasitic parameters were heritable is probably an overestimation. These experiments also showed that five out of these nine parameters were sensitive to the nest environment (i.e. SRBC‐ and PHA‐responses, number of C. haemapterus, haematocrit and blood concentration in leucocytes). Female nestlings were more infested by the blood‐sucking fly C. haemapterus than their male nestmates, and their blood was less concentrated in leucocytes. The effect of year, season, age (i.e. reflecting the degree of maturation of the immune system), brood size, position in the within‐brood age hierarchy, and body mass strongly differed between the 12 parameters. Several studies have highlighted the association between birdsong and parasite load, but there is no evidence regarding therelationships among pathogens and alarm or distress calls, whichare used in antipredator strategies. We analyzed the associationbetween virus infection and the distress calls of lesser short-toedlark (Calandrella rufescens), addressing the relationships betweencall acoustic properties, presence of poxvirus lesions, andother measurements of bird health (body condition, T-cell–mediatedimmune response, heterophils-to-lymphocytes ratio [H/L ratio],and blood parasites). The study was carried out in Fuerteventura(Canary Islands), where 55% of lesser short-toed larks wereinfected. Pox infection was associated with changes in the spectrotemporalstructure of lark distress calls and affected the condition-dependentnature of these signals. Virus bearers uttered significantlyshorter and lower pitched calls than virus-free birds, givingeven shorter calls when subject to greater physiological stress(higher H/L ratio), whereas virus-free individuals did the opposite.Call harshness was positively correlated with bird immune andbody condition, independently of virus infection, whereas callpulse rate decreased in stressed birds. We hypothesize thathealthy birds might reveal their ability to bear the costs ofantipredator defense by means of long, harsh, and fast modulateddistress calls. Infection not only affects individual stateand morphology by means of gross lesions but also may altera signal used in an antipredator context, thus potentially reducingbird fitness both directly (through disease) and indirectly(through increased predation).  相似文献

The purpose of this study was to examine whether immune neutralizationof muscle-produced insulin-like growth factor I (IGF-I) would preventan appropriate anabolic response to refeeding in diabetic rats. MaleSprague-Dawley rats were made diabetic by partial pancreatectomy andwere randomly assigned to be either control-fed, fasted, orfasted-refed (n = 7-8 per group). Diabetes decreased rates of protein synthesis and increased rates of protein degradation in incubated epitrochlearis muscles (P < 0.05). In both groups of rats, fasting lowered protein synthesis andincreased proteolysis and subsequent refeeding returned both parameters to near basal values (P < 0.05). Neutralization ofmuscle IGF-I by the addition of IGF-I antibody to the incubation mediumreduced protein synthesis an average of 22% for all groups(P < 0.05). However, rates of protein degradation werenot affected. In nondiabetic rats, refeeding increased proteinsynthesis in both control and antibody-treated muscles(P < 0.05). Refeeding also increased protein synthesisin the control muscles from diabetic rats (P < 0.01).In contrast, muscles from diabetic rats that were incubated withanti-IGF-I did not increase protein synthesis in response to refeeding.These data suggest that immune neutralization of muscle IGF-I inhypoinsulinemic rats negated the ability of endogenous IGF-I to promoteprotein synthesis and thereby prevented an appropriate anabolic response.

Life history theory predicts a trade-off between number and quality of offspring. Reduced quality with increasing brood size may arise from a decrease in body condition or in immunocompetence that would be important in fighting off virulent parasites by immunologically naive offspring. We tested the effect of rearing conditions on immune function of nestling great tits (Parus major) by reducing or increasing broods by two hatchlings. In the middle of the nestling period (on day 8), nestlings from enlarged broods developed lower T cell responses [as measured from the cutaneous swelling reaction to injection with phytohaemagglutinin (PHA)] and tended to have lower total leukocyte and lymphocyte concentrations in their peripheral blood than nestlings from reduced broods. Brood size manipulation affected the PHA response of nestlings most strongly in small clutches, suggesting that nestling immune function was dependent on their parents’ condition, as estimated by original clutch size. Intra-brood differences in nestling mortality were unrelated to immune parameters, but nestlings in broods without mortality had a stronger PHA response, higher concentration of lymphocytes and higher body mass on day 15 than nestlings in broods with mortality. These results support the prediction that the immune function of altricial birds is affected by rearing conditions, and that growth and immune parameters are related to inter-brood differences in nestling survival. Parasites often exert strong selection pressures on their hosts that have evolved anti-parasite defences to counter the negative effects of parasites. We studied the relationship between intensity of parasitism, one aspect of host immune response, and host reproductive success, using the house martin bug Oeciacushirundinis and its house martin Delichonurbica host as a model system. Experimental manipulation of parasite load of nests during laying of the first clutch altered the intensity of parasitism. Parasites reduced the reproductive success of their hosts measured in terms of body condition and survival of nestlings. Host immune response, measured as the concentration of gammaglobulins and total plasma proteins, was positively associated with parasite reproduction, estimated as the number of juvenile parasites, but was only weakly related to the intensity of adult parasites. The concentration of gammaglobulins was negatively related to nestling body mass, implying a trade-off between immune function and body condition. We tested the novel hypothesis that arrival date in migratorybirds represents a reliable indicator of male quality that canbe used by females as a cue in extrapair mating decisions. Secondarysexual characters are often condition-dependent, and competitionfor early arrival leads to condition-dependent migration. Hence,both secondary sexual characters and arrival date are predictedto be condition-dependent indicators of male phenotypic quality.We studied the relationship between expression of a secondarysexual character, arrival date, and condition, respectively,and extrapair paternity in a Spanish population of barn swallows,Hirundo rustica. By using microsatellite markers to determinepaternity, we showed that 17.8% of all offspring (N = 674) and32.4% of all broods (N = 170) were due to extrapair paternity.Quasi-parasitism (in which the male nest owner fathered theoffspring, but the eggs were laid by another female) occurredin 2.6% of all nestlings and 2.9% of all broods. Individualswere consistent in the frequency of extrapair paternity amongfirst, second, and third broods. Males with long outermost tailfeathers, arriving early and in prime body condition, had littleextrapair paternity in their nests. This was also the case whencontrolling for the confounding effects of male age. Partialcorrelation analysis was used to investigate the direct andindirect effects of tail length, arrival date, and body conditionon extrapair paternity. Body condition accounted for most ofthe variance in extrapair paternity, whereas tail length andarrival date accounted for a smaller proportion of the variance.Body condition was strongly correlated with tail length andarrival date. However, because females cannot directly assesscondition or arrival date (males arrive before females), femalesmay obtain an indirect measure of condition and migration abilityfrom tail length and other phenotypic traits of males. Thissuggests that extrapair paternity depends on the effects ofcondition, through its indirect effects on arrival date, taillength, and other variables.  相似文献

Male secondary sexual characters may have evolved as intra-or intersexual signals of male phenotypic or genetic quality.In birds, singing performance may have the function to honestlyreveal health and vigor of individual males. Infectious diseasesand poor body conditions would therefore be expected to negativelyinfluence singing performance. Since bird pathogens are knownto elicit both a humoral and a cell-mediated immune response,it can be predicted that a negative relationship exists betweensinging performance and activity of the immune system. Thisprediction was tested for the first time in this correlationalstudy. The relationships between song rate and features andhematological variables (concentration of leukocytes in peripheralblood, ratio of gamma-globulins to total plasma proteins, bloodcell sedimentation rate, hematocrit) and body condition wereanalyzed in a population of bam swallows (Hirundo rustica).Song rate was negatively correlated with lymphocyte concentrationand with the ratio of gamma-globulins to plasma proteins. Spectrographicanalysis showed that features of song were not significantlycorrelated with hematological variables or body condition. Thelevel of circulating testosterone was not correlated with songrate nor hematological variables. This study is the first toshow a correlation between a bird's singing performance andhematological profile and suggests that song rate of male barnswallows may reflect their health status. Song in this speciesmight thus have evolved because it allows prospecting femalesto assess aspects of phenotypic and/or genetic quality of potentialmates  相似文献

The expression of sexual ornaments has been suggested to reliablyindicate individual quality, such as the ability to cope withparasites and diseases. The Immunocompetence Handicap Hypothesis(IHH) states that testosterone-dependent ornaments honestlysignal such quality because of physiological costs associatedwith testosterone, such as impaired immune function. We testedpredictions of the IHH both correlatively and experimentallyin red grouse Lagopus lagopus scoticus. Male grouse exhibitsupra-orbital red combs whose size is testosterone-dependent.We found that comb size was not correlated to infection intensityby two parasites (coccidia and the nematode Trichostrongylustenuis), but it was significantly positively correlated withcondition and T-cell-mediated immunity (the ability to mounta primary inflammatory response). We manipulated testosteroneby means of implants and re-caught males after a month to investigatethe effects on comb size, condition, immunity, and parasiteload. Males implanted with testosterone had increased comb size,lost more condition, and had lower T-cell-mediated immunitythan control males. Increased testosterone also resulted ina significant increase in coccidia infection intensity but hadno effect on T. tenuis burden. The results are consistent withpredictions of the IHH and suggest that comb size honestly indicatesimmunocompetence and males' ability to cope with certain parasites.Females could thus benefit from choosing mates based on theexpression of this sexual trait.  相似文献

We tested the influence of population density and of drought intensity (measured as the Gaussen Index in spring and summer of the year of birth) on winter body mass, hind foot length, and body condition of roe deer fawns. Body mass decreased with increasing density and increased with increasing Gaussen Index in summer, in a similar way for both males and females. Hind foot length of males showed the same response. On the other hand, hind foot length of females decreased with increasing density only after dry summers, hence when environmental conditions were very harsh. Body condition was affected neither by density nor by drought intensity. Our results indicate that body mass and size are much better indicators of phenotypic quality than body condition in roe deer. The sex-specific responses of body size to environmental conditions could correspond to a differential allocation in favour of daughters by heavier than average roe deer mothers.  相似文献

Traits related to fitness are often pleiotropically linked orotherwise constrained in their expression. Organisms thereforetrade between fitness components such as number and viabilityof their offspring. The physiological mechanisms mediating suchtrade-offs, however, have been poorly investigated. We manipulatedbrood size and satiation of nestling barn swallows, Hirundorustica, to simulate the effect of two kinds of natural stresses,i.e., long-term intense competition in a large brood and acutefood deprivation, and we measured their effect on body condition,T cell–mediated immune response, and corticosterone, themain hormone mediating the adrenocortical stress-response. Broodenlargement increased corticosterone levels compared with thosefor brood reduction, and brood enlargement depressed immuneresponse, body mass, and condition. Corticosterone levels markedlyincreased after food deprivation. Immune response negativelycovaried with corticosterone levels measured after long-termstress. Hence, living in a crowded nest and with food deprivationelicited a stress response mediated by corticosterone, and depressedan important component of offspring fitness such as T cell–mediatedimmunity. The negative covariation between circulating corticosteroneand immunity suggests that the trade-off between offspring numberand quality is mediated by variation in plasma levels of corticosterone,which has immunosuppressive effects.  相似文献

Carotenoid pigments are responsible for many examples of sexuallyattractive red, orange, and yellow coloration in animals andplay an important role in antioxidant and immune defenses. Becausevertebrates cannot synthesize carotenoids, limited dietary availabilitymay impose a trade-off between maintaining ornamental colorationand health. We used an experimental approach to test the carotenoidtrade-off hypothesis in the fighting fish Betta splendens, byexamining whether carotenoid allocation strategies differ amongconspecifics that exhibit a gradient of body coloration fromblue to red. We found that male redness is underlain by carotenoidsand that females preferred to associate with red males overblue males, suggesting a sexually-selected advantage to beingred. Moreover, we found strong experimental support for thecarotenoid trade-off hypothesis, as individuals that variedin color did not appear to allocate carotenoids equally to bothimmune response and coloration. Redder fish given supplementalcarotenoids increased in both immune response (to a phytohemagglutinationchallenge) and redness compared with controls. In contrast,bluer fish given supplemental carotenoids did not become morered but instead benefited immunologically more so than eithercontrol or redder supplemented fish. These results enhance ourunderstanding of the evolution and plasticity of carotenoidmobilization and utilization pathways in animals.  相似文献

An efficient immunity is necessary for host survival, but entails energetic costs. When energy is limited, immunocompetence and body condition should co-vary positively among individuals and, depending on body condition, individuals should allocate more either in innate immunity or in adaptive response. We tested whether immune phenotype depends on body condition in large mammals, using data from two contrasted populations of roe deer Capreolus capreolus in France. Roe deer living at Chizé, a forest with poor habitat quality, were expected to show lower values for body condition and immune parameters than roe deer at Trois Fontaines, a forest with high habitat quality. From 285 blood samples collected between December 2009 and March 2011, we measured seven metabolic parameters and ten immunological parameters. A Principal Component Analysis showed that all indicators of body condition co-varied positively and were lowest at Chizé. Several immunological indicators correlated to body condition and differed between Trois Fontaines and Chizé. However, high body condition was not associated to a high average level of immunocompetence, but instead to high levels of indicators of acute inflammatory innate response, while low body condition was associated to high levels of monocytes and lymphocytes, possibly reflecting adaptive immunity. Limited data suggest that the difference between populations was not related to the presence of specific parasite species, however parasite exposure and stress have to be investigated to gain a more complete understanding of the determinants of immunity.  相似文献

The ontogeny of strombid behavior was studied by observing thebehavior of Strombus maculatus veligers collected from the planktonand reared past metamorphosis to adults, and by observing juvenilestrombids collected in the field. Complete adult modal actionpatterns (MAP's) associated with locomotion, feeding, and rightingof overturned shells are performed by S. maculatus juvenilesimmediately after metamorphosis. There are changes in the frequencyof the use of certain MAP's which are associated with variationsin shell shape and size. The unique strombid escape response to molluscivorous gastropods(Conns spp.) is not released until juvenile S. maculatus arethree weeks past metamorphosis and two millimeters in shelllength. At that stage, the complete response is released uponthe first encounter with a predator. Experience with a predatordoes not seem to lower the age or size criteria. During ontogeny there is a trend toward an increasing complexityof behavior which is paralleled by an increasing complexityof neural structure and general morphology. There are majorsteps in the ontogeny of strombid behavior which probably coincidewith neural and morphological stages.  相似文献

Experiments on birds, fish and mammals have shown that adverse conditions during infancy can produce diverse long‐term and delayed deficits during adulthood, prejudicing both the individual and its offspring. Natural selection should prepare animals to cope with adversity of the type, magnitude and timing that commonly occur in their natural habitat, but very little is known about such evolved developmental buffering against natural ‘poor starts’ in life. In two‐chick broods of the blue‐footed booby Sula nebouxii, the junior (younger) chick usually experiences aggressive subordination, reduced nutrition and growth and elevated circulating corticosterone. To test whether this poor start produces long‐term, delayed or intergenerational deficits in body size, body condition or cell‐mediated immune response, we measured 3–8 yr old female breeders banded as chicks, and their infant offspring. Results failed to support our predictions. Compared to former seniors and former singletons (solitary nestlings), former juniors showed no deficit in cell‐mediated immune response at any age. They showed an 8.04% deficit in body condition at age 4–6 yr but this deficit disappeared completely by age 7–8 yr. Furthermore, their offspring showed no deficits in body size, body condition or immune response. Junior chicks are affected by their poor start, but their developmental resilience, also confirmed by studies of post‐fledging survivorship, recruitment, natal dispersal, aggressive nest defense and reproduction, is evidence of evolved developmental buffering against predictable adversity during infancy.  相似文献

Investigations focusing on host–ectoparasite interactions in animals have revealed asymptomatic to severe health and fitness consequences suggesting that species mobilize different interspecific response mechanisms. Fewer studies, however, have examined intraspecific responses to ectoparasitic burdens. In this study, we analyzed host health and fitness responses to increasing ectoparasite burdens along with the presence/absence of hemoparasites of free‐ranging insular rock iguanas (Cyclura cychlura) in The Bahamas. Using hematology, plasma biochemistry, as well as body condition and growth rate comparisons, we failed to find significant associations of tick burdens with annual growth rate, corticosterone, packed cell volume, total white blood cell, and heterophil, monocyte, eosinophil or hemoglobin measures. We did, however, find mixed and significant associations of tick burdens with lymphocyte and basophil counts, heterophil‐to‐lymphocyte ratios, and body condition indices. These associations varied by sex, size, and hemoparasite infection status suggesting that different life stages of iguanas may invest differently in immune responses, and impacts may be modulated based on size and sex of hosts, and coinfection status.  相似文献

Abstract.  1. Immune defence imposes fitness costs as well as benefits, so organisms are expected to optimise, not maximise, their immune responses. This should result in variation in immune responses under varying environmental conditions.
2. Males and females are expected to exhibit different immune responses because life-history differences between the sexes affect optimal immune response. These life-history differences should usually result in a greater female, than male, immune defence. In this study, intra- and inter-sexual variation in one component of immune defence, the encapsulation response, in cabbage white butterflies ( Pieris rapae L.), was examined.
3. Encapsulation decreased with increasing age and in response to reduced diet quality.
4. Contrary to predictions, males generally had greater immune responses than females, although this pattern varied with age.
5. These patterns of inter- and intra-sexual variation in encapsulation may result from resource-based trade-offs with components of reproductive effort and/or because of sexual dimorphism in melanin-based wing patterns.  相似文献
