青霉菌胞外半纤维素酶的固定化研究   总被引:2,自引:0,他引:2  
青霉菌m8胸外半纤维素酶经过离子交换和分子筛层析两步纯化,纯化的酶固定在戊二醛交联的壳聚精上,残活力为45.6%。固定化酶的最适pH为3.6,最适温度为65℃,且固文化酶在65~75℃活力都较高。该酶的耐热性比较强,固定化酶热稳定性优于原酶;以半纤维素为底物,固定化酶的表观米氏常数为3.58×10-2g/L。  相似文献

壳聚糖固定化德氏根霉脂肪酶的研究   总被引:4,自引:0,他引:4  
研究了壳聚糖吸附和戊二醛交联对脂肪酶固定化条件,在室温条件下将0.4g酶粉溶于pH6.0缓冲液中,加入10g壳聚糖,摇匀,再加入浓度为0.6%戊二醛交联6h,得到固定化酶,酶活力回收率约为54.2%。固定化酶的半失活温度比游离酶的高,半失活温度由游离酶的47℃提高到100℃,最适反应温度由40℃上升至80℃,最适pH由6下降到5.5,固定化酶K’m值由游离酶的Km 50mg/mL增加到56mg/mL。该固定化脂肪酶用于酯的合成;在80℃条件下经过10批次连续水解植物油反应,固定化酶的活力仍保持在82.6%以上。  相似文献

从青霉菌m8提取出半纤维素酶,将其固定在用戊二醛交联的壳聚糖载体上.0.5 g壳聚糖与4%的戊二醛结合固定2.5 mg蛋白质,酶活回收率为45.6%. 原酶的最适pH为4.6,固定化酶为pH 3.6.原酶的最适温度为55℃,固定化酶在60~75℃都具有较高活性.固定化酶的耐热性优于原酶. 以半纤维素为底物,固定化酶的表观Km值略低于原酶,前者为5.0×10-2 g/L,后者为3.58×10-2 g/L.  相似文献

为探讨木聚糖酶被固定化后的酶活力变化 ,采用盐析、离子交换和分子筛层析方法对链霉菌胞外木聚糖酶进行了纯化 ,并采用DNS方法对固定化酶的性质进行了研究。结果如下 :粗酶液被纯化了 30 .5倍 ,比活力达 4 5 7.5 ,活力回收 4 2 .6 %。纯化后的酶固定在戊二醛交联的壳聚糖上 ,残活力为 4 1.8%。固定化酶的最适pH为 6 .0 ,最适温度为 5 5℃ ,且固定化酶在 6 5 -75℃活力都较高。该酶的耐热性比较强 ,固定化酶热稳定性优于原酶 ;以木聚糖为底物 ,固定化酶的表观米氏常数为 0 .83× 10 -2g/L。因此 ,固定化的木聚糖酶优于原酶  相似文献

微生物酶拆分方法生产D-泛酸的手性中间体D-泛解酸内酯   总被引:13,自引:0,他引:13  
筛选到一株产D-泛解酸内酯水解酶的串珠镰孢霉菌(Fusarium moniliforme SW-902)。产酶条件研究表明,用甘油作碳源,蛋白胨作氮源,初始pH8.0,温度26℃,摇瓶培养3d,产酶量最高。在60L和1000L发酵罐中通风发酵45-47h,产酶量为6-8g干菌体/L,D-泛解酸内酯水解酶酶活力达到0.87-0.92IU/g干菌体。该酶的最适反应温度为55℃,最适反应pH为7.0-7.5。在酶不对称水解泛解酸内酯过程中,对溶液加酶量5%-10%,底物浓度10%-20%,控制水解率20%-30%,水解效果最好。  相似文献

研究壳聚糖吸附和戊二醛交联对木聚糖酶固定化条件 .将酶液加入到经醋酸溶液处理过的脱乙酰壳聚糖的pH 4 8的悬液中 ,加入浓度为 0 3%~ 0 4 %的戊二醛溶液 ,室温下 ,8h后得到固定化酶 .固定化酶的半失活温度比游离酶高 ,由 5 1℃升至 71℃ ,Km 值由游离酶的 1 2mg ml增加到1 5mg ml ,最适反应温度也由 5 5℃增加到 71℃ ,而最适反应pH由 4 6下降到 3 8.该固定化木聚糖酶可用于制造低聚木糖 .经过 10次连续应用实验后 ,该固定化酶的活力保持 81%  相似文献

用聚乙烯醇凝胶包埋具有高活力天冬氨酸酶的大肠杆菌(Escherichia coli)No.1细胞。该酶的表现活力高达1638 00u/g湿细胞,酶活力的回收率为97.5%。固定化细胞和游离细胞天冬氨酸酶的最适pH均为8.0,最适温度分别为40—45℃和40—55℃。二价金属离子Mn2+、Mg2+、Ca2+和Fe2+对热钝化的天冬氨酸酶活力具有保护作用。在37—45℃下,两种细胞的热稳定性相同。二者在pH6.0的柠檬酸缓冲液中比较稳定。固定化细胞在1mol/L、pH8.0的底物溶液(内含Mn2+1mmol/L)中于4℃冰箱保存6个月,天冬氨酸酶的活力保持不变。用固定化细胞柱连续生产L-天冬氨酸,底物转化为产物的转化率达95%以上;产物的总 收率为91.1%。固定化细胞柱连续运转40天,天冬氨酸酶活力仍保持最初酶活力的90%。  相似文献

对谷胱甘肽硫转移酶的固定化、游离酶和固定化酶的酶学特性进行了研究,通过试验,确定谷胱甘肽硫转移酶的最佳固定化条件为先用2%壳聚糖吸附酶,然后再加戊二醛交联,交联用戊二醛浓度为1.2%,交联时间6h;游离酶的最适温度为45—55℃,最适pH值为6.5-7.0:固定化酶的最适温度为45-50℃,最适pH值为7.0;游离酶和固定化酶的最适酶促反应时间为30min。  相似文献

链霉菌胞外木聚糖酶经过盐析、离子交换和分子筛层析纯化,粗酶液被纯化了32.5倍,比活力达498u/mg,活力回收46.6%。纯化后的酶固定在戊二醛交联的壳聚糖上,酶活回收率为42.8%。固定化酶的最适pH为6.0,最适温度为60℃,且固定化酶在65~75℃活力都较高。该酶的耐热性比较强,固定化酶热稳定性优于原酶;以木聚糖为底物,固定化酶的表观米氏常数为0.93×10-2g/L。  相似文献

将巨大芽孢杆菌胞外青霉素酰化酶通过共价键连接到醋酸纤维素载体上,制成的固定化青霉素酰化酶的表观活力达2000 u/g左右(PDAB法)。水解lO%(w/v)的青霉素G钾盐落液,使用30批,保留活力70%以上。6-氨基青毒烷酸(6-APA)总收率平均达88.37%。固定化青霉素酰化酶水解青霉素G的最适pH为9.95,最适温度为55℃,表观米氏常数为1.093×10-2mol/L,在pH 5.8-10.7,温度45℃以下酶的活力稳定。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献
