Population genetic studies carried out on penaeid shrimps have disclosed different patterns of population subdivision, revealing new aspects of shrimp biology as well as the effects of historical contingency molding those patterns. However, the stability of observed allele frequencies over time still remains untested. The objective of this article is to show the analysis of the temporal variation of allozymes in a shrimp species inhabiting Cuba which proves that the genetic structure of this species could significantly change in time. The study involves four populations of Farfantepenaeus notialis sampled in a period of 8 years. The significant statistics obtained from partitions observed in 1995 were not detected in 2003 (as suggested by AMOVA and F(ST)), whereas temporal genetic differentiation and heterozygosity became highly significant. The results strongly suggest that the effect of migrations could be the cause for the loss of F. notialis genetic structure in 2003. It is therefore imperative to call attention on the vulnerability of these populations when facing unstable environmental and habitat conditions. 相似文献

Common groundsel, Senecio vulgaris (Asteraceae), is a highly selfing semelparous ephemeral weed that belongs to the few plant species in central Europe capable of growing, flowering and fruiting all year round. In temperate climates, flowering S. vulgaris cohorts were found to appear up to three times per year. Using amplified fragment length polymorphism (AFLP) molecular markers we examined temporal genetic differentiation among spring, summer and autumn cohorts at each of seven sites located in two regions in Switzerland. Strong genetic differentiation among cohorts may indicate the existence of seasonal races of S. vulgaris, reproductively isolated by nonoverlapping flowering phenologies. Analysis of molecular variance (amova) revealed that < 2.5% of the AFLP variation resided among cohorts within sites, whereas there was significant genetic differentiation among plants from different sites (15.6%) and among individuals within cohorts (81.9%). Significant genetic differentiation was also observed between the two regions. Isolation-by-distance was found on a regional scale, but not on a local scale. Gene flow was estimated to be approximately 15-fold higher among cohorts within sites than among sites. We further found, on average, similar levels of genetic diversity within the three seasonal cohorts. The results of this study demonstrate that season of growth represents a weak barrier for genetic exchange among S. vulgaris populations and does not affect molecular variance. Therefore, there is no evidence for the existence of seasonally specialized races of S. vulgaris. We discuss some implications of the results for the biological control of S. vulgaris using a native rust fungus. 相似文献

We investigated clonal population structure and genetic variation in Quercus havardii (sand-shinnery oak), a deciduous rhizomatous shrub that dominates vegetation by forming uninterrupted expanses of ground cover over sandy deposits on the plains of western Texas, western Oklahoma, and eastern New Mexico. Isozyme electrophoresis (15 loci coding 11 enzymes) was used to recognize and map clones arrayed in a 2000-m transect (50-m sample intervals) and a 200 × 190 m grid (10-m sample intervals). Ninety-four clones were discovered, 38 in the transect and 56 in the grid, resulting in an estimated density of ~15 clones per hectare. Clones varied greatly in size (~100-7000 m), shape, and degree of fragmentation. The larger clones possessed massive interiors free of intergrowth by other clones, while the smaller clones varied in degree of intergrowth. The population maintained substantial levels of genetic variation (P = 60%, A = 2.5, H(exp) = 0.289) comparable to values obtained for other Quercus spp. and for other long-lived perennials. The population was outcrossing as evidenced by conformance of most loci to Hardy-Weinberg expected genotype proportions, although exceptions indicated a limited degree of population substructuring. These data indicate that despite apparent reproduction primarily through vegetative means, Q. havardii possesses conventional attributes of a sexual population. 相似文献

Combining different sources of information is essential for a complete understanding of the process of genetic differentiation between species. The Iberian and North African wall lizard ( Podarcis ) species complex has been the object of several studies regarding morphological and mitochondrial DNA variation but, so far, no large-scale survey of nuclear variation within this group has been accomplished. In this study, ten polymorphic allozyme loci were studied in 569 individuals collected across the Iberian Peninsula and North Africa. The obtained data were analysed using both conventional population genetic tools and recent Bayesian model-based clustering methods. Our results show that there are several well-differentiated entities corroborating the major splits observed in mtDNA analyses. These groups correspond not only to the fully recognized species Podarcis bocagei , Podarcis carbonelli , and Podarcis vaucheri but also to multiple forms within the polytypic Podarcis hispanica , all of which have a similar level of differentiation to that observed between the acknowledged species. However, relationships between forms are weakly supported both by population and individual clustering methods, suggesting a scenario of a rapid diversification that contrasts to the clear bifurcating model assumed from previous mtDNA analyses. Individual multilocus analyses report few individuals misassigned or apparently admixed, some of which are most likely explained by the persistence of high levels of ancestral polymorphism. A survey described genetically encoded enzyme variation in 197 adult Argentinian hake, Merhccius hubhsi , collected from 10 sample sites on the Argentinian continental shelf. Four of 11 muscle enzyme loci were polymorphic ( PCM *, G3PDH *, EST-1 * and EST-2 *). Genotypes showed no significant difference between males and females. Only one of 37 tests of departure from Hardy-Weinberg equilibrium showed a significant difference. The allele frequency data were tested for genetic heterogeneity among locations. The overall significance of the total samples and among those from Patagonia Central was primarily due to heterogeneity at the EST-1 * locus. The implications of these findings in a population structure context are discussed. 相似文献

Buffalograss, Buchloë dactyloides, is widely distributed throughout the Great Plains of North America, where it is an important species for rangeland forage and soil conservation. The species consists of two widespread polyploid races, with narrowly endemic diploid populations known from two regions: central Mexico and Gulf Coast Texas. We describe and compare the patterns of allozyme and RAPD variation in the two diploid races, using a set of 48 individuals from Texas and Mexico (four population samples of 12 individuals each). Twelve of 22 allozyme loci were polymorphic, exhibiting 35 alleles, while seven 10-mer RAPD primers revealed 98 polymorphic bands. Strong regional differences were detected in the extent of allozyme polymorphism: Mexican populations exhibited more internal gene diversity (He= 0.20, 0.19) than did the Texan populations (He= 0.08, 0.06), although the number of RAPD bands in Texas (n= 62) was only marginally smaller than in Mexico (n= 68). F-statistics for the allozyme data, averaged over loci, revealed strong regional differentiation (mean FRT=+ 0.30), as well as some differentiation among populations within regions (mean FPR=+ 0.09). In order to describe and compare the partitioning of genetic variation for multiple allozyme and RAPD loci, we performed an Analysis of Molecular Variance (AMOVA). AMOVA for both allozyme and RAPD data revealed similar qualitative patterns: large regional differences and smaller (but significant) population differences within regions. RAPDs revealed greater variation among regions (58.4% of total variance) than allozymes (45.2%), but less variation among individuals within populations (31.9% for RAPDs vs. 45.2% for allozymes); the proportion of genetic variance among populations within regions was similar (9.7% for RAPDs vs. 9.6% for allozymes). Despite this large-scale concordance of allozyme and RAPD variation patterns, multiple correlation Mantel techniques revealed that the correlations were low on an individual by individual basis. Our findings of strong regional differences among the diploid races will facilitate further study of polyploid evolution in buffalograss. 相似文献

利用聚丙烯酰胺凝胶电泳方法,分析了我国沿海中部(江苏、山东和浙江)海滨沙滩珊瑚菜(Glehnia littoralis Fr.Schmidt ex Miq.)7个居群8种酶系统19个位点的等位基因遗传变异特征,结果表明居群内多态位点比率平均为82.4%,每一位点平均等位基因数为2.77,有效等位基因数为2.24,固定指数F的平均值为-0.091。珊瑚菜居群基因多样性80.9%产生于居群内,19.1%产生于居群间。居群间的遗传距离平均为0.317,遗传一致性为0.728,居群内维持着较高水平的遗传多样性。据此,对珊瑚菜的渐危机理进行了讨论。 相似文献

We assessed the validity of two gekkonid species, Gekko yakuensis and G. hokouensis, in southern Japan. We first assigned all 398 specimens into 18 samples merely on the basis of localities. By conducting significance test for deviations of genotype frequencies from Hardy‐Weinberg at 11 allozyme loci, we checked the reproductive unity of constituents in each of those local samples, and where necessary, rearranged them into subsamples on the basis of genetic markers so that we recognized minimum reproductively cohesive units. We then compared allele frequencies among all samples and subsamples examined. Results clearly indicated that all but two can be classified into two groups that can be discriminated from each other by remarkable allele frequency differences at four diagnostic loci, and by large genetic distances even between sympatric subsamples. Observations of morphological features of the samples and subsamples confirmed that the two groups correspond to G. yakuensis and G. hokouensis, supporting validities of these two species. Allele frequency comparisons, however, also revealed that the remaining two samples, both from southern Kyushu, possessed ‘marker alleles' of both G. yakuensis and G. hokouensis at all four diagnostic loci. These samples thus were considered to represent populations that have been derived through hybridization of the two species. Detailed analyses for genetic structures demonstrated that all hybrid genotypes in the two samples are post‐Fi generations with only one individual resulting from the back‐cross with a pure line population of G. yakuensis. This finding negates the possibility that the hybrid populations are maintained by a constant supply of newly produced Fj hybrids, but suggests that the hybrid genotypes constitute stable breeding populations. This implies that the genealogical independence of G. yakuensis and G. hokouensis in several other sympatric areas has been maintained by operations of some isolation mechanisms at a pre‐mating phase. Investigations of the morphological variation in each sample or subsample revealed that although the two species can be externally largely discriminated from each other by slight modifications of the currently used diagnoses, it is difficult to detect their hybrids based solely on the morphological features. 相似文献

Knowledge of the degree of temporal stability of population genetic structure and composition is important for understanding microevolutionary processes and addressing issues of human impact of natural populations. We know little about how representative single samples in time are to reflect population genetic constitution, and we explore the temporal genetic variability patterns over a 30-year period of annual sampling of a lake-resident brown trout (Salmo trutta) population, covering 37 consecutive cohorts and five generations. Levels of variation remain largely stable over this period, with no indication of substructuring within the lake. We detect genetic drift, however, and the genetically effective population size (N(e)) was assessed from allele-frequency shifts between consecutive cohorts using an unbiased estimator that accounts for the effect of overlapping generation. The overall mean N(e) is estimated as 74. We find indications that N(e) varies over time, but there is no obvious temporal trend. We also estimated N(e) using a one-sample approach based on linkage disequilibrium (LD) that does not account for the effect of overlapping generations. Combining one-sample estimates for all years gives an N(e) estimate of 76. This similarity between estimates may be coincidental or reflecting a general robustness of the LD approach to violations of the discrete generations assumption. In contrast to the observed genetic stability, body size and catch per effort have increased over the study period. Estimates of annual effective number of breeders (N(b)) correlated with catch per effort, suggesting that genetic monitoring can be used for detecting fluctuations in abundance. 相似文献

将中华稻蝗(Oxya chinensis)注射1.5g/L的马拉硫磷,24h后死亡率为56%。用等位酶电泳确定死亡与存活个体在多态性基因座Pgm和Me上的基因型,并比较各基因型的死亡率差异。结果表明:在Pgm基因座上,马拉硫磷对具有不同基因型的中华稻蝗个体存在选择性致死作用,但是在Me基因座上未观察到类似选择性致死作用。在基因座Pgm上,Pgm-ab 基因型个体死亡率最高(80%),Pgm-bb和Pgm-bc基因型个体死亡率最低(49%),低于平均死亡率。卡方检验表明,基因型Pgm-bb和Pgm-cc的死亡率之间存在显著差异。在存活个体中,等位基因Pgm-b的频率显著增加。基于Roger’s遗传距离的聚类分析表明,马拉硫磷的急性致死作用会导致中华稻蝗种群遗传组成的变化。马拉硫磷是蝗虫防治中常用的杀虫剂,研究显示,不同基因型在死亡率上的差异也可能存在于田间。Abstract: Allozyme electrophoresis was employed to compare the difference in mortality among the genotypes at two polymorphic loci of Pgm and Me of grasshopper Oxya chinensis individuals acutely exposed to 1.5g/L malathion which resulted in 56% mortality in 24 hours. The selective lethal effects were observed among the genotypes at Pgm locus but not at Me locus. It is noted that the genotype Pgm-ab experienced the highest mortality (80%), whereas Pgm-bb and Pgm-bc were 49%, lower than theaverage. The χ2 tests showed significant difference in morality between Pgm-bb and Pgm-cc. After exposure the allele frequency of Pgm-b showed a notable increase among surviving individuals. The cluster analysis based on Roger’s genetic distance indicated that the acute exposure to malathion can cause differentiation in genetic composition at population level in Oxya chinensis. Because malathion is commonly used as the insecticide for grasshopper control, the data obtained in this study suggest that the similar genotype-mortality effects may occur in crop fields. 相似文献

Populations of insect herbivores that feed on several host plant species may experience different selective forces on each host. When the hosts cooccur in a local area, herbivore populations can provide useful models for the study of evolutionary mechanisms in patchy environments. A first step in such a study involves determination of the genetic structure of host adaptation in the region: how is genetic variation for host use structured within and between subpopulations of herbivores on each host? The structure of genetic variation for host use reveals patterns of local adaptation, probable selective consequences of migration between hosts, and the potential for further evolution. To estimate the population structure of host adaptation in a patchwork, 7–11 pea aphid clones were collected at the beginning of the summer from each of two alfalfa and two red clover fields within a very localized area (about 15–20 km2). Using a reciprocal transplant in the field, replicates of these 35 clones were allowed to develop individually on each of the two crops. A complete life table was made for each replicate. Individual fitness was calculated from the life tables as the expected rate of population increase; longevity, age at first reproduction, and total fecundity were also measured for each clonal replicate. Currently, experimental estimates of genetic variation in complete life tables are virtually nonexistent for natural populations, even for single environments (Charlesworth, 1987); field studies are even less common. Because clones from each of two source crops were tested reciprocally on both hosts, variation in relative genotypic fitness on alfalfa and clover could be partitioned among clones within source crops, between fields of the same crop, and between source crops (alfalfa or red clover), providing a view of population structure. Significant clonal variation in relative performance on alfalfa and red clover was found: clones tended to have higher fitness on the crop from which they had been collected (the “home” crop) than they did on the “away” crop, suggesting local adaptation in response to patchy patterns of selection. Clonal variability within collections from the two crops suggests the potential for changes in the genetic constitution of these aphid populations within established fields as a result of clonal selection during the summer season. Significantly negative genetic correlations across crops were found for fitness and its major components. The possibility that these negative cross-environment correlations could act as evolutionary constraints on adaptation to the patchwork is considered. 相似文献

We report data on genetic drift of mitochondrial DNA (mtDNA) haplotypes in a natural brown trout (Salmo trutta) population in Sweden. Large temporal frequency shifts were observed over the 14 consecutive year classes studied. The observed rate of drift was used to estimate the effective size of the population. This effective size applies to the female segment of the population as mtDNA is maternally inherited. The magnitude of mtDNA haplotype frequency change is compared with the corresponding allele frequency changes at 14 allozyme loci in the same population. The female effective size is estimated as 58, which is approximately half the effective size of 97 for the total population (both sexes) previously obtained from the shifts of allozyme allele frequencies. 相似文献

The present population structure of a species reflects the influence of population history as well as contemporary processes. To examine the relative importance of these factors in shaping the current population structure of Littorina keenae, we sequenced 762 base pairs of the mitochondrial ND6 and cytochrome b genes in 584 snails from 13 sites along the northeastern Pacific coast. Haplotype network analysis revealed a 'star-like' genealogy indicative of a recent population expansion. Nested clade and mismatch analyses also supported the hypothesis of sudden population expansion following a population bottleneck during the Last Glacial Maximum. Analysis of molecular variance and pairwise Phi(ST) showed no significant spatial population differentiation from Mexico to Oregon - not even across the recognized biogeographic boundary at Point Conception. This is probably due to high contemporary gene flow during the free-swimming larval stage of this snail. Surprisingly, we found a highly significant temporal population differentiation between a San Pedro sample from 1996 and one from 2005, which gave an estimate of effective population size (N(e)) of only 135. Nearly statistically significant changes in the frequency of a particular haplotype in three other populations over 2-3 years further support Hedgecock's 'sweepstakes' hypothesis. When by chance nearly all of the progeny from an aggregation of highly fecund sisters that possess a rare haplotype successfully recruit to become the next generation, the rare haplotype can become temporarily common across the entire species' range. This modification of the sweepstakes hypothesis can explain why the temporal variation that we observed was much greater than the spatial variation. 相似文献

This work explores the possibility that constraints on genetic variation guide host shifts and are responsible for the evolutionary conservatism of host affiliation in phytophagous insects. To this end, we used full- and half-sib breeding designs to screen two species of the North American bettle genus Ophraella for genetic variation in larval and adult feeding responses to several host plants of other species of Ophraella. All the plants are in the family Asteraceae. In O. conferta, we observed effectively no feeding response, and hence no genetic variation in response, to three of five test plant species; only those plants related to the species' natural hosts evoked genetically variable responses. In O. artemisiae, adults displayed genetic variation in response to a congener of the natural host, but not to two distantly related plants. However, significant variation among full-sib broods in larval feeding suggests the existence of nonadditive genetic variance in feeding response to all five species of test plants—although survival was very low on most of them. The results suggest that patterns of presence versus apparent absence of detectable genetic variation may be related to the chemical similarity of plants to the insects' natural hosts, but not evidently to the phylogenetic history of host affiliation within the genus. Almost all genetic correlations in responses to host plants were not significantly different from zero; the few significant correlations were positive, and negative correlations that might explain host specificity were not found. Our data do not explain why exclusive shifts to new hosts should occur, but the apparent lack of genetic variation in responses to some plants suggests that the direction of host shifts is genetically constrained. 相似文献

In a study of the genetic relationships among 879 anadromous brown trout Salmo trutta from 13 streams at the Island of Gotland, Sweden, using RFLP analysis of a mitochondrial DNA segment (NADH dehydrogenase-1 gene), six haplotypes were detected. Significant genetic divergence was observed among streams as well as between cohorts within streams. Approximately 8–9% of the total variation was due to differences between populations, and 4–5% was explained by differences between cohorts within populations. The female effective population size ( N ef ) was assessed from temporal haplotype frequency differences between consecutive cohorts; the estimated average N ef over all populations was just below 30, suggesting that these populations were effectively quite small. With such small effective sizes the populations are expected to loose genetic variability quickly, but the observed levels do not appear particularly low. This indicates that female migration between streams occurs. The observed level of differentiation does not support the presumption that a particular pre-smolt migratory behaviour observed in Gotland streams, with large portions of fry leaving for the sea soon after hatching, results in a reduced homing ability. From a conservation management perspective the Gotland brown trout streams should be regarded as a population system where the vitality and survival of brown trout in one stream is dependent on the opportunity of contact and exchange of individuals from other streams. 相似文献
