Tomato plants were grown under light intensities of 36 or 90 W m−2 [photosynthetically active radiation (PAR)], and then the light intensity was changed to 36, 90 or 180 W m−2 for 8 h to investigate the effect of temporary changes in light intensity on the carbon budget of photoassimilates from the third leaf using a 14CO2 steady-state feeding method. In the plants that were raised under 90 W m−2, the photosynthetic rate increased when the light intensity was increased to 180 W m−2, whereas no increase occurred in the plants that were raised under 36 W m−2. Although the total amount of carbon fixed during the 8-h light period showed a large difference between plants grown at the two initial light intensities, the proportion of carbon exported during the light period did not differ apparently, irrespective of the change in light intensity. However, the amount of carbon exported during the time course was higher in plants that were raised under 90 W m−2 than those raised under 36 W m−2, irrespective of the change in light intensity. The partitioning pattern of 14C-photoassimilates was not changed by the change in light intensity, irrespective of whether the light intensity was increased or not. However, the amount of 14C-photoassimilates accumulated in each part differed according to the two initial light intensities. The carbon transport from a source leaf was also investigated through a quantitative analysis of carbon balance.  相似文献

The transport and accumulation of 2-[14C]-IAA applied to the apex of cuttings of Pisum sativum L. cv. Alaska was greater in cuttings from stock plants grown under 38 W m−2 than 16 W m−2. Accumulation of 14C in the base of the cuttings from the highest level of irradiance was correspondingly more significant. The level of irradiance to the stock plants greatly affected the rate of accumulation, while the light conditions during IAA transport had a minor effect. The amount of IAA reaching the base of the cuttings increased with increasing concentration of IAA in the treatment solution, but the percentage of applied IAA reaching the base decreased.
The relative chromatographic partition of ethanol-extractable 14C showed that, after 12 h of IAA-transport, the amount of 2-[14C]-IAA was higher in the base of cuttings from 38 W m−2 than in those from 16 W m−2. After a further 12 h of transport the relative amounts of 2-[14C]-IAA in the two types of cuttings were reduced to the same lower level.
A possible role of an irradiance-mediated difference in the topographic distribution of IAA in the base of pea cuttings on the subsequent adventitious root formation is discussed.  相似文献

Rooting ability was studied for cuttings derived from pea plants ( Pisum sativum , L. cv. Alaska) grown in controlled environment rooms. When the cuttings were rooted at 70 μmol m−2 s, 1 (photosynthetic photon flux density) or more, a stock plant irradiance at 100 μmol m−2 s−1 decreased rooting ability in cuttings compared to 5 μmol m−2, s−1, However, cuttings rooted at 160 μmol m−2 s−1 formed more roots compared to 5 (μmol m−2 s−1. Although a high irradiance increased the number of roots formed, it could not overcome a decreased potential for root formation in stock plants grown at high irradiance. Light compensation point and dark respiration of cuttings decreased by 70% during the rooting period, and the final levels were strongly influenced by the irradiance to the cuttings. Respiratory O2 uptake decreased in the apex and the base of the cutting from day 2 onwards, whereas a constant level was found in the leaves. Only the content of extractable fructose, glucose, sucrose and starch varied during the early part of the rooting period. We conclude that the observed changes in the cuttings are initiated by excision of the root system, and are not involved in the initiation of adventitious roots.  相似文献

Development and acclimation of energy transduction were studied in seedlings of Chenopodium rubrum L. ecotype selection 184 (50° 10' N; 105° 35' W) in response to photomorphogenic and photoperiodic treatments. Dark respiration and photosynthetic capacity [nmol O2 (pair of cotyledons)−1 h−1] were measured with an oxygen electrode. Changes in chloroplast ultrastructure were analyzed concomitantly. After germination, seedlings were grown at constant temperature either in darkness or in continuous light (white, red, far-red and blue) or were subjected to diurnal cycles of light/dark or changes in light quality. Dark respiration was low in far-red light treated seedlings. In red light treated seedlings dark respiration was high and the mean value did not depend on fluence rate or photoperiod. Blue light stimulated transitorily and modulated dark respiration in photoperiodic cycles. Photosynthetic capacity was reduced by far-red light and increased by red light. In response to blue light photosynthetic capacity increased, with indications of a requirement for continuous energy input. Phytochrome and a separate blue light receptor seemed to be involved. In continuous red light a clear cut circadian rhythm of dark respiration was observed. Blue light had a specific effect on chloroplast structure.  相似文献

Arabidopsis thaliana (L.) Heynh. race Columbia plants were grown in red. blue, red + far-red, blue + far-red and various light mixtures of red + blue + far-red light under 14 h light/10 h dark photoperiods. Each single light source and light mixture maintained a constant irradiance (50 μmol m−2 s−1) and the mixtures of red + blue + far-red maintained a constant ratio of red/far-red light, but varied in the ratio of blue to red + far-red light. Depending on the method used for calculation, values of the fraction of phytochrome in the far-red absorbing form (Pfr/Ptot) for these light mixtures were either constant or decreased slightly with increasing percentage of blue light in the mixtures. Arabidopsis flowered early (20 days) in blue, blue + far-red and red + far-red light and late (55 days) in red light. In mixtures of red + blue + far-red light, each of which established a nearly constant Pfr/Ptot flowering was in direct relation to time and irradiance level of blue light. Leaf area and petiole length were also correlated with blue light irradiance levels.  相似文献

The K+ (86Rb+) uptake and the growth of intact wheat seedlings ( Triticum aestivum L. cv. GK Szeged) grown in 0.5 m M CaCl2 solution and of seedlings grown on wet filter paper in Petri dishes were compared under different experimental conditions. Aeroponic (AP) and hydroponic (HP) conditions brought about striking differences in the growth of the roots, whereas the shoot growth was not influenced. The dry weight of the roots was higher for the AP plants than for the HP plants. The AP grown seedlings exhibit a low rate of K+ uptake, which seems to be a passive process. The effect of 2, 4–dinitrophenol (2, 4–DNP) clearly shows the absence of an active component of the K+ uptake in roots grown in air with a high relative humidity. In plants grown under AP conditions the effect of Ca2+ on the K+ uptake is unfavourable, i.e. there is an inhibition (negative Viets effect). Results relating to the effect of 2,4–DNP suggest that the "negative Viets effect" is a feature of the passive K+ uptake. The data suggest that the AP growth conditions play a very important role in the induction and/or development of the ion transport system(s), which becomes impaired under the AP conditions.  相似文献

Triacylglycerols occur in both the endosperm and embryo of Euphorbia lambii seeds. Upon germination, the amount of these neutral lipids in the endosperm decreased with 1.06 mg fatty acid day-1. The embryo contained 1.4 mg fatty acids in the triacylglycerols and this value declined slowly to 0.4 mg seedling-1 during the 8 day period of endosperm depletion. Radioactive acetate was rapidly taken up by the cotyledons of intact seedlings, translocated throughout the entire seedling, and up to 10.5% of the 14C proceeded to the sterols and latex triterpenols. Maximum uptake values of 1.4 μmol seedling-1 day-1 of acetate were measured. Acetate uptake and subsequent incorporation into sterols and triterpenols decreased substantially in the presence of increasing amounts of sucrose (up to 0.3 M). Traces of acetate did not effect [14C]-sucrose uptake and corresponding synthesis of [14C]-sterols and triterpenols, but increased concentrations of acetate (0.05 M and up) reduced both uptake of sucrose and its conversion into unsaponifiable lipids.
The uptake capacity of the cotyledons for [14C]-glycerol exceeded the daily production in the endosperm, but only a small amount of label proceeded to the sterols and triterpenols. [14C]-Triacylglycerols were never detected in the seedling, regardless of the labeled substrate used. Although acetate is an efficient precursor in triterpenol and sterol synthesis, the uptake capacity of the cotyledons for this metabolite is too small in relation to the daily production of water soluble substrates in the endosperm. If acetate is released by the endosperm, only a marginal contribution towards triterpenol and sterol synthesis in the seedling is to be anticipated from this substrate.  相似文献

Pea plants ( Pisum sativum L. ev. Greenfeast) were grown for 2 to 3 weeks in while (˜ 50 μmol photons m−2 s−1; 400–700 nm) or green (˜ 30 μmol photons m−2 s −1 400–700 nm) light (16 h day/8 h night), with or without far-red light. Supplementary far-red light decreased leaf area and increased internodal length in both white and green light, demonstrating that phytochrome influenced leaf size and plant growth. However, there was no effect of far-red light on chlorophyll a /chlorophyll b ratios, chlorophyll-protein composition, the stoichiometry of electron transport complexes or photosynthetic function of isolated thylakoids. These results suggest that phytochrome is ineffective in modulating the composition and function of thylakoids in pea plants grown at low irradiance. One possible explanation of the ineffectiveness of phytochrome on thylakoids is discussed in terms of the drastic attenuation of red relative to far-red light in green tissue.  相似文献

Pinus banksiana seedlings were grown for 9 months in enclosures in greenhouses at CO2 concentrations of 350 or 750 μmol mol−1 with either low (0.005 to 0. 3 W m−2) or high (0.25 to 0. 90 W m−2) ultraviolet-B (UV-B) irradiances. Total seedling dry weight decreased with high UV treatment but was unaffected by CO2 enrichment. High UV treatment also shifted biomass partitioning in favor of leaf production. Both CO2 and UV treatments decreased the dark respiration rate and light compensation point. High UV light inhibited photosynthesis at 350 but not at 750 μmol mol−1 CO2 due to a UV induced increase in ribulose-1, 5-bisphosphate carboxylase/oxygenase efficiency and ribulose-1, 5-bisphosphate regeneration. Stomatal density was increased by high UV irradiance but was unchanged by CO2 enrichment.  相似文献

The interactive effects of ozone and light on leaf structure, carbon dioxide uptake and short-term carbon allocation of sugar maple ( Acer saccharum Marsh.) seedlings were examined using gas exchange measurements and 14C-macroautoradiographic techniques. Two-year-old sugar maple seedlings were fumigated from budbreak for 5 months with ambient or 3 × ambient ozone in open-top chambers, receiving either 35% (high light) or 15% (low light) of full sunlight. Ozone accelerated leaf senescence, and reduced net photosynthesis, 14CO2 uptake and stomatal conductance, with the effects being most pronounced under low light. The proportion of intercellular space increased in leaves of seedlings grown under elevated ozone and low light, possibly enhancing the susceptibility of mesophyll cells to ozone by increasing the cumulative dose per mesophyll cell. Indeed, damage to spongy mesophyll cells in the elevated ozone × low light treatment was especially frequent. 14C macroautoradioraphy revealed heterogeneous uptake of 14CO2 in well defined areole regions, suggesting patchy stomatal behaviour in all treatments. However, in seedlings grown under elevated ozone and low light, the highest 14CO2 uptake occurred along larger veins, while interveinal regions exhibited little or no uptake. Although visible symptoms of ozone injury were not apparent in these seedlings, the cellular damage, reduced photosynthetic rates and reduced whole-leaf chlorophyll levels corroborate the visual scaling of whole-plant senescence, suggesting that the ozone × low light treatment accelerated senescence or senescence-like injury in sugar maple.  相似文献

Cyanophora paradoxa Korshikov synchronized autotrophically in a light-dark regime of 14 h light and 10 h dark divides in the last two hours of the dark period. The division rate of the free-living blue-green alga, Synechococcus leopoliensis Raciborski, at identical culture conditions (24°C; 32 W m−2) is only slightly lowered in the light period. The comparison of thylakoid differentiation in the endocyanelles of Cyanophora paradoxa and in Synechococcus leopoliensis during the light-dark regime yields (1) the same ensemble of pigment-protein complexes in both organisms, (2) comparable syntheses of chlorophyll and phycobilins of Cyanophora paradoxa grown under 32 W m−2 and of Synechococcus leopoliensis grown under light intensities below 9.2 W m−2, and (3) identical photosynthetic oxygen evolution during the light period of the light-dark regime with minima at the beginning, in the middle (6th–7th h), and at the end of the light period. In both organisms this stage-specific oxygen evolution is inhibited by treatment with chloroamphenicol. Cycloheximide, however, causes no significant alterations. Results are discussed in view of the endosymbiotic theory.  相似文献

The effects of UV-C (254 nm), UV-A (365 nm) and broad-band UV (280–380 nm) on guard cells of Vicia faba L. cv. Long Pod were investigated in the presence of white light (450 μmol m−2 s−1). UV-C (7 μmol m−2 s−1) was found to cause leakage of 86Rb+ from guard cells, while UV-A (0.3 μmol m−2 s−1) stimulated increased uptake in these cells. A relatively small stimulatory effect was observed by broad-band UV (3 μmol m−2 s−1) during the first 30 min of irradiation with an apparent equilibration of influx and efflux thereafter. Leakage of 86Rb+ from guard cells continued despite the removal of UV-C and an increase in the amount of white light from 450 to 1500 μmol m−2 s−1, suggesting that membranes were irreversibly damaged. Irradiation of guard cells with UV-C for 30, 45 and 90 min indicated that these cells began to be affected already by 30 min UV-C irradiation.  相似文献

Summary. Soluble potassium concentrations were determined for the slightly vacuolated, unicellular, walled alga Chlorella emersonii. Sap of cells grown in 1 mol m−3 NaCI contained 140 mol m−3 K+ and sap of cells grown in 125, 200, and 335 mol m−3 NaCI contained 160-180 mol m−3 K +.
The possible regulation of K + concentrations by a system of lurgor and volume maintenance was investigated by supplying 3-0-methylglucose. This solute accumulates to 85-230 mol m−3 in C. emersonii , but is not metabolized. Accumulation of 3-0-methylglucose increased the volume of cells grown at both low and high NaCI by about 10%. Furthermore, accumulation of 3-0-methylglucose also increased turgor pressures of cells grown in 1 and 125 mol m−3 NaCI by 0.3 and 0.2 MPa, respectively. (Similar measurements were not attempted for cells grown in 200 and 335 mol m−3 NaCI, because of the insensitivity of available methods to measure turgor pressure of cells exposed to high external osmotic pressures.)
At all NaCI concentrations, the K + concentrations of cells which had accumulated 3-0-methylglucose were only 10-20 mol m−3 lower than K+ in cells which had not been supplied with 3-0-methylglucose. In contrast, accumulation of 3-0-methylglucose greatly decreased concentrations of the endogenous osmotic solutes, proline and sucrose, which accumulated in cells grown in 125 mol m−3 and higher NaCI concentrations.
It is concluded that K+ concentrations in Chlorella emersonii are not controlled by a system of turgor and volume maintenance.  相似文献

Growth and development of hydroponically grown pea seedlings ( Pisum sativum L. cv. Alaska) were measured using stem and root length as well as number of leaves and lateral roots. The growth was dependent on the presence of cotyledons and was modulated by the irradiance. All plants were grown in a full nutrient solution. If grown at low irradiance (73 μmol m-2s-1) they depended more and for a longer time on the cotyledons than plants grown at high irradiance (220 μmol m-2s-1). Low irradiance caused stem elongation but decreased root length and number of lateral roots as compared to plants grown at high irradiance. The dark respiration of the leaves was measured as oxygen uptake. In plants grown at the low irradiance, excision of the cotyledons caused the rate of oxygen uptake to increase by a factor of three, and the increase was sensitive to cyanide. Decotyledonized plants showed a high respiration rate and a diminished leaf growth for their entire life cycle. CO2 fixation also increased in decotyledonized pea seedlings grown at either irradiance. The mobilization of food reserves from the seeds was positively correlated to seed dry weight, but only if the plants were grown at 73 μmol m-2s-1. Increasing dry weight of the seed enhanced top growth, whereas root growth was depressed, so that top and root responds differently with regard to that part of growth which depends on mobilization of reserves from the seed.  相似文献

Abstract. The rates of uptake of 32P-labelled orthophosphate by whole root systems of young apple trees (M.9 rootslocks and Worcester Pearmain seedlings) were measured in solution culture. Using a solution depletion technique, the 32P-phosphate uptake rates per unit length, surface area or fresh weight of roots were determined as a function of 32P-phosphate concentration in solution at the root surface over the range 0.25–10 mmol m−3. The effect of P concentration within various plant parts on the relation between uptake rate and external P concentration was studied using plants differing in internal P levels.
The apparent minimutn P concentration below which P uptake ceased was of the order of 0.25–0.50 mmol m−3. Fluxes, inflows and unit absorption rates increased approximately proportionately with solution concentration up to 10mmolm−3. Except perhaps in the case of the low-P M.9 plant, there was no evidence of a diminishing returns type of relationship over the range of solution concentrations examined. The threshold P concentration in solution above which uptake rates cease to increase thus appears to be higher for apples than for other species.
At any given P concentration, fluxes, inflows and unit absorption rates were higher for M.9 than for Worcester and for low-P plants than for high-P plants. The difference between plants of different P status was more marked for M.9 and seems to be more closely related to shoot P levels than to root P.  相似文献

To test for the effects of far‐red light on preventing budset in Picea abies , seedlings of six populations originating from latitudes between 67°N and 47°N were grown for 4–8 weeks in continuous incandescent (metal halogen) light at 300 µmol m−2 s−1 and 20°C and then transferred, at the same temperature, to a daily regime of 8 h incandescent light (300 µmol m−2 s−1) followed by 16 h cool white fluorescent light (40 µmol m−2 s−1). (Cool white lamps are deficient in far‐red light, with a R/FR ratio of 7.5 compared with 2.0 for the incandescent lamps.) All the seedlings from 67° and 80% of those from 64° stopped extension growth and set terminal buds within 28 days of the change of regime. The seedlings from 61° and further south continued growing, as did control seedlings from 67° grown as above but with incandescent light at 20 µmol m−2 s−1 replacing cool white illumination. To distinguish between a clinal and ecotypic pattern of variation, the interval between 64° and 59° was investigated by growing populations originating from that area in the same regimes as before. After 28 days in the cool white day‐extension regime, the percentage budset was 86 for the population from 64°, 0 for the population from 59° and 25–50 for the intermediate populations; i.e. the populations showed a clinal variation in requirement for far‐red light according to latitude. Thus northern populations of Picea abies appear to behave as 'light‐dominant' plants for the photoperiodic control of extension growth and budset, whereas the more southern populations behave as 'dark‐dominant' plants.  相似文献

The electrical resting potential across the plasmalemma of Lemna gibba L. (G 1) cells is −230 to −250 mV and the diffusion potential in the presence of 1 mol m−3 KCN + 1 mol m−3 salicylhydroxamic acid is about −100 mV. A concentration of 0.01 mol m−3 HgCl2 depolarises the transmembrane electrical potential in a largely reversible way. When the cells after 16 min of HgCl2-application are returned to Hg-free solution, the transmembrane electrical potential is only depolarised by 24 × 13 mV (SD, n = 13) compared with the potential prior to HgCl2 treatment. In contrast, a 16 min pretreatment with HgCl2 followed by a wash with mercury-free solution reduces the transient depolarisations of transmembrane potential observed after addition of 5 mol m−3 D-glncose or 1 mol m−3 L-alaoine to about 60% of controls. These transient depolarisations are due to the onset of solute uptake. Accordingly, HgCl2-pretreatment inhibits uptake of 14C-3-O-methyl- d -glucose by more than 50% and uptake of 14C- l -alanine by more than 70%. Washing with 1 mol m−3 1,4-dithiothreitol does not reverse this inhibition. It is, therefore, concluded that Hg2+ irreversibly binds to essential SH-groups of the H+-hexose and the H+-amino-acid cotransport carriers of Lemna gibba and inhibits these carriers without appreciably affecting the electrogenic proton-extrusion pump.  相似文献

Primary leaves of 4-day-old, dark-grown mung bean [ Vigna radiata (L.) Wilczek cv. Berken] seedlings were exposed to 24 h of white light (200 μmol m−2 s−1) which was terminated by a 15 min, phytochrome-saturating red or far-red light exposure. Phytochrome content (in vivo and in vitro) and leaf area were monitored during the subsequent dark period. Red light treatments resulted in lower phytochrome content and greater leaf expansion than did far-red treatments. Phytochrome accumulation and leaf expansion were less in norflurazon- (no carotenoids and very low Chl) than in tentoxin- (very low Chl) treated leaves. After 3 days of darkness, leaf expansion was about 25% greater and phytochrome content was about 50% less in red- than in far-red-treated leaves of all treatments. These effects generally took longer to develop in norflurazon- than in tentoxin-treated tissues. Norflurazon-treated tissues exposed to long white light periods apparently do not as accurately reflect phytochrome-controlled photomorphogenic events of green tissues as do tentoxin-treated tissues of mung bean seedlings.  相似文献

Five-day-old etiolated radish ( Raphanux salivux L. cv. Saxa) seedlings exposed to white continuous light in the presence of Cd2+ (0.2 mM) showed characteristic changes in their light-harvesting chlorophyll a/b protein complex II after 48 h of greening. The content of its oligomeric supramolecular form was greatly diminished with a concomitant increase in the level of the monomer. The isolation of highly purified light-harvesting chlorophyll a/b protein complex II from control and Cd2+ treated radish cotyledons and a detailed analysis of its structure and composition revealed that first of all, Cd2+ altered the content of the specific phosphatidylglycerol fatty acid - trans -Δ3-hexadecenoic acid, widely accepted as a component responsible for the oligomerization of this chlorophyll-protein complex. This fatty acid in the thylakoid membrane phosphatidylglycerol pool seems to be very sensitive to different environmental stresses lowering its content, which indicates the vital significance of this component for the supramolecular organization and proper functioning of the light-harvesting chlorophyll a/b protein complex II.  相似文献

Partitioning of [14C]-labeled assimilates was studied in relation to photoperiodic floral induction and evocation in one-week-old Pharbitis nil Choisy cv. 'Violet' seedlings. In plants kept under 16 h photoperiods, one 15 h night induced 100% axillary flowering whereas a 24 h night induced both terminal and axillary flowering. A 15 min night break of red light given 8 h after the beginning of the dark period inhibited flowering. Total [14C]-assimilate distribution among major sinks (plumules + epicotyl and roots + hypocotyl) from a single source cotyledon was unchanged by one inductive night; however, import of [14C]-assimilates into shoot apices was increased in induced plants compared to vegegative controls. This increase was several-fold in plants subjected to a 24 h night. N6-Benzyladenine (BA) application to cotyledons or plumules under non-saturating night lengths increased the number of floral buds per plant without affecting the position of the first floral bud (i.e. the speed of induction). The same treatment caused increased label accumulation in induced apices, while it only slightly affected non-induced ones. The mode of action of BA on flowering through growth stimulation and resulting assimilate mobilization is discussed.  相似文献
