The Barcelona Conference on Epigenetics and Cancer (BCEC) entitled “Epigenetic Mechanisms in Health and Disease” was held in Barcelona, October 26-26, 2017. The 2017 BCEC was the fifth and last edition of a series of annual conferences organized as a joint effort of five leading Barcelona research institutes together with B-Debate. This edition was organized by Albert Jordan from the Molecular Biology Institute of Barcelona (IBMB-CSIC) and Marcus Bushbeck from the Josep Carreras Leukaemia Research Institute (IJC). Jordi Bernués, Marian Martínez-Balbás, and Ferran Azorín were also part of the scientific committee. In 22 talks and 51 posters, researchers presented their latest results in the fields of histone variants, epigenetic regulation, and chromatin 3D organization to an audience of around 250 participants from 16 countries. This year, a broad number of talks focused on the epigenetic causes and possible related treatments of complex diseases such as cancer. Participants at the 2017 BCEC elegantly closed the series, discussing progress made in the field of epigenetics and highlighting its role in human health and disease.  相似文献

The Barcelona Conference on Epigenetics and Cancer (BCEC) entitled “Challenges, opportunities and perspectives” took place November 21–22, 2013 in Barcelona. The 2013 BCEC is the first edition of a series of annual conferences jointly organized by five leading research centers in Barcelona. These centers are the Institute of Predictive and Personalized Medicine of Cancer (IMPPC), the Biomedical Campus Bellvitge with its Program of Epigenetics and Cancer Biology (PEBC), the Centre for Genomic Regulation (CRG), the Institute for Biomedical Research (IRB), and the Molecular Biology Institute of Barcelona (IBMB). Manuel Perucho and Marcus Buschbeck from the Institute of Predictive and Personalized Medicine of Cancer put together the scientific program of the first conference broadly covering all aspects of epigenetic research ranging from fundamental molecular research to drug and biomarker development and clinical application. In one and a half days, 23 talks and 50 posters were presented to a completely booked out audience counting 270 participants.  相似文献

The Barcelona Conference on Epigenetics and Cancer (BCEC) entitled “Beyond Cancer Genomes” took place October 13th and 14th 2016 in Barcelona. The 2016 BCEC was the fourth edition of a series of annual conferences coordinated by Marcus Buschbeck and subsequently organized by leading research centers in Barcelona together with B?DEBATE, a joint initiative of BIOCAT and “La Caixa” Foundation. Salvador Aznar-Benitah, Eduard Batlle, and Raúl Méndez from the Institute for Research in Biomedicine in Barcelona selected the 2016 BCEC panel of speakers. As the title indicates, this year's conference expanded the epigenetic focus to include additional cancer-relevant topics, such as tumor heterogeneity and RNA regulation. Methods to develop therapeutic approaches on the basis of novel insights have been discussed in great detail. The conference has attracted 217 participants from 11 countries.  相似文献

The 2015 BCEC was the third edition of a series of annual conferences jointly organized by 5 leading research centers in Barcelona together with B-Debate, an initiative of BioCat. Luciano Di Croce from the Center for Genomic Regulation and Marcus Buschbeck from the Josep Carreras Leukemia Research Institute put together the scientific program with a particular focus on the role of non-coding RNAs in enhancer regulation, epigenetic control by Polycomb complexes, histone variants, and nuclear organization. An outstanding array of prominent investigators gathered in Barcelona on May 28th and 29th, 2009 for the first Cancer Epigenetics and Biology Symposium to discuss progress in different areas of cancer research. Organized by Dr. Manel Esteller, this Symposium also publicized the launching of the Cancer Epigenetics and Biology Program (PEBC), of which Esteller is the director. The symposium covered a variety of topics, including the use of animal models to study cancer, the study of epigenetic pathways involved in tumorigenesis, and the future of epigenetic therapeutics. Some of the highlights of this meeting are summarized in this report.  相似文献

The Second German-Catalan Workshop on Epigenetics and Cancer was held in Barcelona on November 19–21, 2014. The workshop brought together, for the second time, scientists from 2 German and 2 Catalan research institutions: the DKFZ, from Heidelberg, the CRCME, from Freiburg, and the IMPPC and PEBC/IDIBELL, both from Barcelona. The German-Catalan Workshops are intended to establish the framework for building a Research School to foster collaborations between researchers from the different institutions. Exchange programs for graduate students are among the activities of the future School. The topics presented and discussed in 33 talks were diverse and included work on DNA methylation, histone modifications, chromatin biology, characterization of imprinted regions in human tissues, non-coding RNAs, and epigenetic drug discovery. Among novel developments from the previous Workshop are the report of the epigenetics angle of the Warburg effect and the long-range trans-acting interaction of DNA methylation and of nucleosome remodeling. A shift in the view on DNA methylation became apparent by the realization of the intertwined interplay between hyper- and hypo-methylation in differentiation and cancer.  相似文献

The term epigenetics is defined as inheritable changes that influence the outcome of a phenotype without changes in the genome. Epigenetics is based upon DNA methylation and posttranslational histone modifications. While there is much known about reversible acetylation as a posttranslational modification, research on reversible histone methylation is still emerging, especially with regard to drug discovery. As aberrant epigenetic modifications have been linked to many diseases, inhibitors of histone modifying enzymes are very much in demand. This article will summarize the progress on small molecule epigenetic inhibitors identified by structure- and computer-based approaches.  相似文献

With the goal of discussing how epigenetic control and chromatin remodeling contribute to the various processes that lead to cellular plasticity and disease, this symposium marks the collaboration between the Institut National de la Santé et de la Recherche Médicale (INSERM) in France and the University of California, Irvine (UCI). Organized by Paolo Sassone-Corsi (UCI) and held at the Beckman Center of the National Academy of Sciences at the UCI campus December 15–16, 2011, this was the first of a series of international conferences on epigenetics dedicated to the scientific community in Southern California. Epigenetics literally means heritable changes in gene expression without any modification in the DNA sequence. The field of epigenetics is revolutionising our understanding of basic fundamental principles behind the normal development and the diseased state of an individual. However, chromatin modifications during infection, wherein the pathogen interacts with its host, received comparatively little attention. Nevertheless, the role of epigenetics in the establishment of infectious diseases by breaching the host defense system is an emerging area of research. Epigenetic regulation impacts differentiation and expression of virulence attributes of a pathogen. For example, antigenic variations in parasites such as Giardia lamblia and Plasmodium falciparum are epigenetically determined. Similarly, chromatin modifying elements have been implicated in fungal morphogenesis and virulence. In particular, chromatin modifying enzymes including histone methyl transferases (KMTs), histone acetyl transferases (KATs), and histone deacetylases (KDACs) have been shown to epigenetically modulate pathogenicity of the human opportunistic pathogen Candida albicans. The significance of chromatin modifications has the potential for explaining the mechanistic basis for distinct lifestyles of the fungus. In this review, we summarize the existing body of evidence that emphasizes the importance of various chromatin modulations involved in providing phenotypic plasticity of the medically important fungal pathogen C. albicans.  相似文献

In the First German-Catalan Workshop on Epigenetics and Cancer held in Heidelberg, Germany (June 17–19, 2013), cutting-edge laboratories (PEBC, IMPPC, DKFZ, and the Collaborative Research Centre Medical Epigenetics of Freiburg) discussed the latest breakthroughs in the field. The importance of DNA demethylation, non-coding and imprinted genes, metabolic stress, and cell transdifferentiation processes in cancer and non-cancer diseases were addressed in several lectures in a very participative and dynamic atmosphere.

The meeting brought together leading figures in the field of cancer epigenetics to present their research work from the last five years. Experts in different areas of oncology described important advances in colorectal, lung, neuroblastoma, leukemia, and lymphoma cancers. The workshop also provided an interesting forum for pediatrics, and focused on the need to improve the treatment of childhood tumors in order to avoid, as far as possible, brain damage and disruption of activity in areas of high plasticity. From the beginning, the relevance of “omics” and the advances in genome-wide analysis platforms, which allow cancer to be studied in a more comprehensive and inclusive way, was very clear. Modern “omics” offer the possibility of identifying metastases of uncertain origin and establishing epigenetic signatures linked to a specific cluster of patients with a particular prognosis. In this context, invited speakers described novel tumor-associated histone variants and DNA-specific methylation, highlighting their close connection with other processes such as cell-lineage commitment and stemness.  相似文献

真核生物基因表达受到染色质结构的调控,组蛋白与DNA的共价修饰构成表观遗传标签,并在植物胁迫应答如防御病原菌侵染过程中起重要作用.病原菌侵染可引起基因组整体DNA甲基化模式变化及胁迫应答基因的位点特异性去甲基化,导致植物抗性基因表达上调或下调,并进一步调控植物对病原菌的胁迫应答;组蛋白去乙酰化酶HDAC通过茉莉酸途径增强植物对病原菌的胁迫应答;此外,染色质重塑复合物Swr1复合体通过识别DNA基元和组蛋白乙酰化修饰状态靶向基因启动子,负调控SA敏感基因.该文从DNA甲基化、组蛋白乙酰化、甲基化修饰,染色质重塑等方面着重阐述植物与病原菌互作过程中发生的主要事件的分子基础及其研究进展.  相似文献

Epigenetics has long been a hot topic in the field of scientific research. The scope of epigenetics usually includes chromatin remodelling, DNA methylation, histone modifications, non‐coding RNAs and RNA modifications. In recent years, RNA modifications have emerged as important regulators in a variety of physiological processes and in disease progression, especially in human cancers. Among the various RNA modifications, m6A is the most common. The function of m6A modifications is mainly regulated by 3 types of proteins: m6A methyltransferases (writers), m6A demethylases (erasers) and m6A‐binding proteins (readers). In this review, we focus on RNA m6A modification and its relationship with urological cancers, particularly focusing on its roles and potential clinical applications.  相似文献

不完全的表观遗传重编程是造成转基因克隆动物效率低下的主要原因,组蛋白修饰作为表观遗传修饰的一个重要部分,可以直接影响克隆胚胎的发育和外源基因的表达情况。TSA(Trichostatin A)作为一种组蛋白去乙酰化抑制剂,可以改变组蛋白的乙酰化水平,促进表观遗传重编程,提高克隆动物的效率。同时TSA能改变染色质结构,使转录因子易于与DNA序列结合,促进外源基因的表达。文章确定了TSA处理转基因猪成纤维细胞和核移植胚胎的最佳条件,分别为250 nmol/L、24 h和40 nmol/L、24 h,通过进一步正交实验发现,TSA同时处理供体细胞和克隆胚胎可以显著的促进核移植胚胎的体外发育。此外,无论TSA处理转基因猪成纤维细胞或核移植胚胎,都可以提高外源基因的表达水平。  相似文献
