The natural host range of beetles feeding on broom ( Cytisus scoparius ) and 14 other species (including six other Cytisus species) in the tribe Genisteae was investigated at 39 sites in Spain , Portugal and France in 1989 and 1992 as part of a biological control programme for broom . Data on host - plant associations were analyzed for 36 phytophagous beetle species from 18 sites , and host records were listed for an additional 58 species . Nine species were apparently restricted to the genus Cytisus : Cryptocephalus octoguttatus, Gonioctena olivacea, Bruchidius lividimanus, B. villosus, Exapion elongatissimum, E. fuscirostre, Lepidapion sp . 1 , Polydrusus confluens and Tychius parallelus. These field records suggest a narrower host - plant range for some beetle species than laboratory host - range tests , and may assist in interpreting host - plant associations reported in the literature . Beetle species with a restricted host - plant range were rarely found on related non - host plants .  相似文献

The photoautotrophic culture medium for Cytisus scoparius Link cells was improved. Linsmaier and Skoog medium was modified by adding a 4-fold concentration of phosphate and lowering the auxin concentration (a-naphthaleneacetic acid 10?6M). Concentrations of all the inorganic elements except phosphate in the original basal medium were adequate for the photoautotrophic growth of Cytisus cells. A high concentration of cytokinin compensated for the inhibition of growth caused by a high concentration of auxin.  相似文献

The structure of protein bodies differs in the upper and lower parts of the cotyledons of mature seeds of Cytisus scoparius L. The palisade-mesophyll cells contain essentially homogeneous protein bodies, without globoids, but the protein bodies of the spongy-mesophyll cells are heterogeneous, with numerous globoids. The complete amino acid sequence of 2-acetamido-2-deoxy-D-galactose-binding Cytisus scoparius seed lectin II (CSII) was determined using a protein sequencer. After digestion of CSII with endoproteinase Lys-C or Asp-N, the resulting peptides were purified by reversed-phase high performance liquid chromatography (HPLC) and then subjected to sequence analysis. Comparison of the complete amino acid sequence of CSII with the sequences of other leguminous seed lectins revealed regions of extensive homology. The amino acid residues of concanavalin A (Con A) involved in the metal binding site are highly conserved among those of CSII. A carbohydrate-binding peptide of CSII was obtained from the endoproteinase Asp-N digest of CSII by affinity chromatography on a column of GalNAc-Gel. This peptide was retained on the GalNAc-Gel column and was presumed to have affinity for the column. The amino acid sequence of the retarded peptide was determined using a protein sequencer. The retarded peptide was found to correspond to the putative metal-binding region of Con A. These results strongly suggest that this peptide represents the carbohydrate-binding and metal ion-binding sites of CSII.  相似文献

The fixation of 14CO2 by mature brown stems of the tea plantwas studied by supplying 14CO2 to selected stems of pruned andintact plants for 24 h under field conditions. Utilization of14C assimilates for the production of new shoots was also examined.The photosynthetic nature of the fixation of 14CO2 is demonstrated.The efficiency of this fixation was very low compared with thattaking place in leaves. AIMS: To contribute to the understanding of Cytisus scoparius success at invading and establishing itself in Australia. METHODS AND RESULTS: Root-nodule bacteria isolated from C. scoparius, growing on five different sites and originally introduced to Australia, were compared with isolates from indigenous plants growing in France and isolates from native legumes growing on the same Australian sites as C. scoparius. Small-subunit rDNA from 251 isolates were analysed by PCR-RFLP and representatives from different genospecies were selected for sequencing. Phylogenetic analyses revealed a great diversity of lineages belonging to Bradyrhizobium, with one genospecies being specific for Cytisus both in Australia and in France, Rhizobium and Mesorhizobium and one falling outside the described genera of legume-nodulating bacteria. Principal component analysis showed that the Cytisus Australian rhizobial communities are more similar to each other than to their co-occurring native partners. CONCLUSIONS: Early established rhizobial symbionts may have an increased probability to contribute inoculum for the development of further nodules. SIGNIFICANCE AND IMPACT OF THE STUDY: This is a first report comparing rhizobia nodulating C. scoparius in its native and exotic environments. Cytisus scoparius symbionts were identified outside the Bradyrhizobium genus and a new lineage of legume-nodulating bacteria was identified.  相似文献

Scotch broom (Cytisus scoparius) is a serious weed of natural and pastoral ecosystems and is invasive in many countries worldwide. Eight polymorphic microsatellite loci were developed and characterized for this species. The number of alleles per locus ranged from five to 23. The range of observed heterozygosity was between 0.207 and 0.928. These microsatellite markers will be useful tools for studies of population genetics in the native and invasive range of this species.  相似文献

Knowledge of the introduction history of invasive plants informs on theories of invasiveness and assists in the invasives management. For the highly successful invasive shrub Scotch broom, Cytisus scoparius, we analysed a combination of nuclear and chloroplast microsatellites for eight native source regions and eight independent invasion events in four countries across three continents. We found that two exotic Australian populations came from different sources, one of which was derived from multiple native populations, as was an invasive sample from California. An invasive population from New Zealand appeared to be predominantly sourced from a single population, either from the native or exotic ranges. Four invasive populations from Chile were genetically differentiated from the native range samples analysed here and so their source of introduction could not be confirmed, but high levels of differentiation between the Chilean populations suggested a combination of different sources. This extensive global data set of replicated introductions also enabled tests of key theories of invasiveness in relation to genetic diversity. We conclude that invasive populations have similar levels of high genetic diversity to native ranges; levels of admixture may vary across invasive populations so admixture does not appear to have been an essential requirement for invasion; invasive and native populations exhibit similar level of genetic structure indicating similar gene flow dynamics for both types of populations. High levels of diversity and multiple source populations for invasive populations observed here discount founder effects or drift as likely explanations for previously observed seed size differences between ranges. The high levels of genetic diversity, differential and source admixture identified for most exotic populations are likely to limit the ability to source biocontrol agents from the native region of origin of invasive populations.  相似文献

Most of the activity of an α-amylase present in crude pea (Pisum sativum L. cv Laxton's Progress No. 9) leaf preparations cannot be found in isolated pea leaf protoplasts. The same extrachloroplastic α-amylase is present in pea stems, representing approximately 6% of total stem amylolytic activity and virtually all of the α-amylase activity. By a simple infiltration-extraction procedure, the majority (87%) of this α-amylase activity was recovered from the pea stem apoplast without significantly disrupting the symplastic component of the tissue. Only 3% of the β-amylase activity and less than 2% of other cellular marker enzymes were removed during infiltration-extraction.  相似文献

Cloned saplings of beech (7-y-old) were exposed to enhanced UV-B irradiation (+25 %) continuously over three growing seasons (1999–2001). Analysis of CO2 assimilation, variable chlorophyll (Chl) a fluorescence, and pigment composition was performed in late summer of the third growing season to evaluate the influence of long-term elevated UV-B irradiation. This influence was responsible for the stimulation of the net assimilation rate (P N) over a range of irradiances. The increase in P N was partially connected to increase of the area leaf mass, and thus to the increased leaf thickness. Even a higher degree of UV-B induced stimulation was observed at the level of photosystem 2 (PS2) photochemistry as judged from the irradiance response of electron transport rate and photochemical quenching of Chl a. The remarkably low irradiance-induced non-photochemical quenching of maximum Chl a fluorescence (NPQ) in the UV-B plants over the entire range of applied irradiances was attributed both to the reduced demand on non-radiative dissipation processes and to the considerably reduced contribution of the quenching localised in the inactivated PS2 reaction centres. Neither the content of Chls and total carotenoids expressed per leaf area nor the contents of lutein, neoxanthin, and the pool of xanthophyll cycle pigments (VAZ) were affected under the elevated UV-B. However, the contributions of antheraxanthin (A) and zeaxanthin (Z) to the entire VAZ pool in the dark-adapted UV-B treated plants were 1.61 and 2.14 times higher than in control leaves. Surprisingly, the retained A+Z in UV-B treated plants was not accompanied with long-term down-regulation of the PS2 photochemical efficiency, but it facilitated the non-radiative dissipation of excitation energy within light-harvesting complexes (LHC) of PS2. Thus, in the beech leaves the accumulation of A+Z, induced by other factors than excess irradiance itself, supports the resistance of PS2 against combined effects of high irradiance and elevated UV-B.  相似文献

Infection of barley leaves by the fungus Rhynchosporium secalisincreases the degree to which stomata open in the light. Openingis enhanced at CO2, levels between 0 and 4000 parts/106. Theability of stomata to close in the dark, as normal, is retaineduntil an advanced stage of tissue necrosis is reached. Increased stomatal opening is confined to those areas of theleaf which have been colonized by the fungus. Stem assimilation of Spartium junceum and Cytisus scoparius was measured over diurnal cycles during four time periods of 1992 in California. Spartium junceum plants were growing at an inland, coastal mountain site while the C. scoparius plants were growing at a coastal bluff habitat. Both species had positive stem assimilation that resulted in approximately 200 mmol m 2 day-1 carbon dioxide accumulation. Daily carbon gain decreased from spring to fall for S. junceum due to a decrease in shoot water potential. Although C. scoparius had the same relationship between assimilation and shoot water potential as that of S. junceum, carbon gain and water potential did not decrease during the year for C. scoparius. The major limitation for carbon gain of C. scoparius was light intensity because the coastal site was characterized by lower temperature, higher humidity, and more fog than the inland site. Although these species grew in habitats that had a different vapor pressure and temperature, the assimilation response to vapor pressure was similar between the species. Water use efficiency was higher and intercellular carbon dioxide was lower for S. junceum compared to C. scoparius. Although the leaves of both species are ephemeral, the canopy has a constant positive carbon balance because of stem assimilation. The constant carbon gain throughout the year, from stem assimilation, may enhance the growth capacity of these invasive species in disturbed habitats.  相似文献

Studies on exogenous application of phenolic compoundsviz: p-hydroxybenzoic acid, resorcinol and chlorogenic acid each with concentration of 10-4 M are done on the legume (Cajanus cajan (L.)Millsp.) AL-15. The effect of applied phenolic compounds as well as of structural differences in phenols indicate a marked influence of phenolic compounds in regulating growth processes in plants. Fresh and dry mass of various plant parts increased after foliar spray with phenols resulting in an improved harvest index. It is seen that phenols also play an important role in the initiation and development of nodules.  相似文献
