Gill and liver microsomal Na+/K+-adenosine triphosphatase (ATPase) activities and plasma levels of 3,5,3'-triiodo-L-thyronine (T3) were measured in rainbow trout (100-300 g) immersed in a freshwater solution of T3 for 6 or 7 days at 11 degrees C. Ambient T3 (1.25 or 2 micrograms T3/100 ml H2O) elevated plasma T3 within a physiologic range; an ambient concentration of 10 micrograms T3/100 ml produced supranormal plasma T3 levels. All T3 treatments depressed gill ATPase to a similar degree. Liver ATPase was lower than gill ATPase and was elevated by a physiologic T3 treatment.  相似文献

Summary Daily intramuscular injection of cortisol (4 mg kg–1 body weight) in rainbow trout,Salmo gairdneri, for 10 days caused significant increases in the number and individual apical surface area of gill chloride cells per mm2 of filament epithelium. Concomitantly, whole body influxes of sodium (Na+) and chloride (Cl) increased. Acute (3 h) intra-arterial infusion of cortisol did not affect whole body Na+ or Cl influx. A significant correlation was observed between both Na+ and Cl influxes and the fractional apical surface area of filament chloride cells in control, sham (saline-injected) and experimental (cortisol-injected) fish. The chloride cells displayed similar ultrastructural modifications in trout undergoing cortisol treatment as in trout transferred to ion-deficient water. These findings suggest the existence of structure/function relationships in which branchial chloride cell morphology is an important determinant of Na+ and Cl transport capacity. We conclude that chronic cortisol treatment enhances whole body Na+ and Cl influxes by promoting proliferation of branchial chloride cells. The results of correlation analysis indicate that the chloride cell is an important site of NaCl uptake in freshwater rainbow trout.  相似文献

Synopsis Chronically starved rainbow trout (Salmo gairdneri) showed a significant fall in liver size, total liver glycogen, liver glycogen concentration and plasma glucose levels. Liver lipid concentration did not differ significantly from controls although total liver lipid reserves fell during the first 40 days of starvation but had partly recovered after 65 days of starvation. Plasma cortisol and T3 levels did not show consistent changes concomitant with food deprivation over the 65 day period of the experiment. However, plasma T4 levels in fish starved for 40 or 65 days were significantly lower than comparably fed animals. The involvement of T4 in intermediate metabolic processes in salmonids is discussed.  相似文献

The effect of the adenosine triphosphate analog, 6,6'-dithiobis(inosinyl imidodiphosphate), (sIMP-PNP)2, was tested on the ouabain-sensitive (Na+ + K+)-ATPase (ATP phosphohydrolase, EC 3.6.1.3) and the ouabain-insensitive Mg2+ - ATPase in microsomes prepared from gill tissue of sea water-adapted rainbow trout, Salmo gairdneri. The (Na+ + K+)-ATPase was completely inhibited by low concentrations of (sIMP-PNP)2 (6 micrometer) but the Mg2+ - ATPase was unaffected by the inhibitor at concentrations as high as 28 micrometer, supporting the suggestion that the two activities represent separate enzymes. The specificity of inactivation could be demonstrated both at a physiological temperature (13 degrees C) and at 37 degrees C. The rates of inactivation were similar at both temperatures. Inactivation of the (Na+ + K+)-ATPase by (sIMP-PNP)2 was reversed by dithiothreitol, suggesting that the inhibitor forms a mixed disulfide with sulfhydryl groups on the enzyme. The inability of substrate (either ATP or its analog, adenyl-5'-yl imidodiphosphate) to protect against inactivation suggests that (sIMP-PNP)2 is reacting with sulfhydryl groups which are not associated with the active site.  相似文献

Na+/K+-ATPase (EC 3.6.1.3) is an important membrane-bound enzyme. In this paper, kinetic studies on Na+/K+-ATPase were carried out under mimetic physiological conditions. By using microcalorimeter, a thermokinetic method was employed for the first time. Compared with other methods, it provided accurate measurements of not only thermodynamic data (deltarHm) but also the kinetic data (Km and Vmax). At 310.15K and pH 7.4, the molar reaction enthalpy (deltarHm) was measured as -40.514 +/- 0.9kJmol(-1). The Michaelis constant (Km) was determined to be 0.479 +/- 0.020 mM and consistent with literature data. The reliability of the thermokinetic method was further confirmed by colorimetric studies. Furthermore, a simple and reliable kinetic procedure was presented for ascertaining the true substrate for Na+/K+-ATPase and determining the effect of free ATP. Results showed that the MgATP complex was the real substrate with a Km value of about 0.5mM and free ATP was a competitive inhibitor with a Ki value of 0.253 mM.  相似文献

Rainbow trout (Salmo gairdneri) were exposed to pH 5.0-5.1, 6.6 and/or calcium-enriched freshwater for 14 days. Hematocrit, gill Ca2+-ATPase enzyme activities, gill osmotic water inflow, plasma calcium and osmolarity were measured. No significant changes in plasma calcium ion levels were found. The typical increase in hematocrit usually associated with exposure of fish to acidified water was not found in the present study and is discussed. Plasma osmolarity decreased in fish exposed to calcium-enriched freshwater (60 mg Ca2+ X 1(-1) ) in comparison to fish exposed to control freshwater conditions (2 mg Ca2+ X 1(1) ), irrespective of the pH level. Gill Ca2+-ATPase enzyme activities were measured for both low affinity (3 mM Ca2+) and high affinity (100 microM) activity. Exposure of rainbow trout to low pH (pH 5.0-5.1) did not affect the specific activity of Ca2+-ATPase enzyme. However, low affinity Ca2+-ATPase activity in fish exposed to calcium-enriched freshwater did show a significant reduction. The increase in gill osmotic water permeability in fish exposed to calcium-enriched freshwater is interpreted as a result of the increase in osmolarity of the ambient media.  相似文献

Rainbow trout (Salmo gairdneri) given a single intraperioneal injection of propanolol (a β-adrenoceptor inhibitor) had plasma triiodothyronine (T3) levels which were significantly (P < 0.01) lower than in control fish 8 hr after the injection, but not after 24hr; thyroxine (T4) levels and T4/T3 ratios were not significantly affected.2. Isoproterenol (a β-adrenoceptor stimulator) had no effect on T4, T3 or T4/T3 ratios.3. A single intraperitorial injection of TSH and prolactin given together induced a significant increase (P < 0.05) in plasma T3 levels 24 hr after the injection.4. TSH and prolactin given to propanol-treated fish caused a further significant increase in T3 levels compared with TSH + prolactin-, TSH-, prolactin-, or solvent-injected groups (P < 0.01).5. The data suggest that the inhibition of peripheral monodeiodination of T4 to T3 by propranolol, well documented in mammals, does not occur in rainbow trout. On the contrary, T3 levels after propranolol administration are increased in conditions where thyroid simulation occurs.  相似文献

Two semisynthetic diets differing in the levels of pantothenic acid (PA) supplementation (0 or 40 mg kg−1 diet) were fed to rainbow trout, Salmo gairdneri Richardson, initially weighing 0.7 g fish−1. Each diet was fed to two tanks of fish and, at the start of the feeding trial, each tank contained 200 animals. The experiment was conducted for 28 days during which time, every 2 days, gill tissue was sampled and food consumption was determined. Lamellar hyperplasia was first detected in the gills of deficient fish on the 12th day sample, while reduced feed intake was first manifest at 16 days. Hyperplasia first appeared in the distal regions of the gill filaments, but the lesion rapidly progressed in a proximal direction and 35 of 40 fish examined on days 22–28 exhibited hyperplasia on more than 75% of the filament surface. Gill lamellar hyperplasia is a sensitive indicator of PA deficiency in the rainbow trout. Moreover, the lesion is specific to PA deficiency since its developmental pattern differs histologically from the lamellar hyperplasia of the non-nutritional gill diseases.  相似文献

The adrenergic innervation of structures in the gills of brown and rainbow trout was studied with catecholamine fluorescence histochemistry. In the arterio-arterial vascular pathway, there was an innervation of the afferent and efferent lamellar arterioles, but the afferent and efferent filamental arteries and the secondary lamellae were devoid of any fluorescent nerve fibres. In S. trutta only, there was an additional innervation of the afferent and efferent branchial arteries and the base of the efferent filamental artery. The innervation of the arterio-venous vascular pathway was similar in both trout species. Many fluorescent nerve fibres were found on nutritive arterioles in the gill arch and interbranchial septum, and in the core of each filament between the surface epithelium and the wall of the filament venous sinus. No fluorescent nerve fibres were observed at the origins of the capillaries arising from the efferent filamental artery. The sympathetic nerve supply is provided to the gills mainly through the posttrematic nerve, with an occasional small contribution through the pretrematic nerve. The presence of adrenergic nerves in the gills is discussed in relation to the regulation of blood flow through the arterio-arterial and arterio-venous pathways.  相似文献

Incorporation of tritium from tritiated water into lipid fractions was measured in isolated hepatocytes from rainbow trout (Salmo gairdneri) acclimated to 5 degrees C and 20 degrees C. Hepatocytes from cold-acclimated trout exhibited significantly higher rates of tritium incorporation into both fatty acid and sterol fractions at assay temperatures of 15 degrees C and 20 degrees C than did hepatocytes from warm-acclimated trout. Tritium incorporation into the fatty acid fraction was nearly temperature independent in hepatocytes from warm-acclimated trout (Q10 = 1.39) but markedly temperature dependent (Q10 = 2.63) in hepatocytes from cold-acclimated trout; in contrast, rates of sterol synthesis were more temperature dependent in warm-acclimated trout. At 5 degrees C, fatty acid lipogenesis comprised a significantly greater percentage of the total tritium incorporation in hepatocytes from warm-acclimated trout and the percentage of total lipogenesis attributable to fatty acids decreased significantly in warm-acclimated trout as the assay temperature increased; the opposite trends were observed in cold-acclimated trout.  相似文献

Young rainbow trout, Salmo gairdneri (Richardson) were injected intraperitoneally with tritiated thymidine, and killed at intervals between 2 hr and 16 days after inoculation. Labelled epithelial cells were first detected autoradiographically along the base of gill lamellae. Epithelial cells proliferated here and then migrated toward the tips of the lamellae. Uniform labelling along the length of the filaments at the base of lamellae indicated that cells were dividing at a constant rate. Transverse sections of filaments showed that epithelial proliferation was also uniform across the base of the lamellae. The interior of the lamellae often had labelled pillar cells, indicating that these cells also divide. The high intensity of the label in animals killed 16 days after inoculation with tritiated thymidine suggests that division probably occurs slowly, less than once every 16 days.  相似文献

The kidney is an organ playing an important role in ion regulation in both freshwater (FW) and seawater (SW) fish. The mechanisms of ion regulation in the fish kidney are less well studied than that of their gills, especially at the level of transporter proteins. We have found striking differences in the pattern of Na+/K+/2Cl- cotransporter (NKCC) expression between species. In the killifish kidney, NKCC is apically localized in the distal and collecting tubules and basolaterally localized in the proximal tubules. However, in the SW killifish gill, NKCC is basolaterally co-localized with Na+/K+-ATPase, whereas in FW, NKCC immunoreactivity is primarily apical, although still colocalized within the same mitochondria-rich cell with basolateral Na+/K+-ATPase. Rainbow trout kidney has NKCC only in the apical membrane of the distal and collecting tubules in both environments, with no signal being detected in the proximal tubule. On the other hand, in the trout gill, NKCC is found basolaterally in both FW and SW environments. An important observation is that, in the gills of rainbow trout, the trailing edge of the filament possesses mostly Na+/K+-ATPase-positive but NKCC-negative mitochondria-rich cells, whereas in the region between and at the roots of the gill lamellae, most mitochondria-rich cells exhibit both Na+/K+-ATPase- and NKCC-positive immunoreactivity. These results suggest that the differential localization of transporters between the two species represents differences in function between these two euryhaline fishes with different life histories and strategies. Synopsis The relationship between standing in a dominance hierarchy and physiological stress was studied in rainbow trout. Individual fish were assigned relative dominance ranks, based on behavioral observations in a large, simulated stream tank. These ranks were compared to histometric measures of interrenal cell activity. Fish, isolated individually in the stream tank had significantly lower levels of interrenal activity than fish from the crowded holding tank. Groups of fish in the stream tank formed stable, linear dominance hierarchies. Interrenal activity correlated inversely with dominance rank, with the exception that top ranking fish had higher activity than expected. Possible cause and effect relationships are discussed.  相似文献

Interspecific differences in the stress response of fish may be due, in part, to differences in the sensitivity of target tissues to cortisol. The relative response of brown and rainbow trout to a standardized dose of cortisol was assessed by monitoring condition (K factor), the number of circulating lymphocytes and mortality due to disease, following cortisol treatment. Cortisol implantation resulted in a significant decline in K factor and number of circulating lymphocytes in immature brown trout, but not in immature rainbow trout, despite plasma cortisol levels being similar in both cases. Cortisol implantation in mature brown and rainbow trout significantly increased the mortality rate due to bacterial and fungal infection compared with control fish. Furthermore, the mortality rate due to disease was significantly greater in brown trout than rainbow trout, despite both groups receiving similar doses of steroid.  相似文献
