在室内研究核型多角体病毒(HaSNPV)感染对棉铃虫Helicoverpa armigera(Htibner)幼虫同类相残行为的影响。结果显示:感病棉铃虫随感病程度的加重,越容易被健康棉铃虫残食,而自然死亡的感病棉铃虫、冻死的感病棉铃虫和冻死的健康棉铃虫三者被健康棉铃虫残食的百分率无显著差异。表明感病棉铃虫和病虫尸体更易于被健康棉铃虫残食,是由于棉铃虫体力减弱而失去反击能力,不是由于病毒本身的影响。以健康棉铃虫、感病棉铃虫为残食者,冻死的病虫为被残食者,相残率无显著差异,表明病毒并未改变棉铃虫残食同类的天性。残食病虫的健康棉铃虫的化蛹率和羽化率均低于正常的健康棉铃虫,残食者为相残行为付出了很高的代价。  相似文献

Egg cannibalism by Helicoverpa armigeraHübner (Lepidoptera: Noctuidae) larvae was studied in the laboratory and in the field. In laboratory experiments, first instars were exposed to increasing densities of H. armigera eggs on sorghum, Sorghum bicolor (L.) Moench, and pigeonpea, Cajanus cajan (L.) Millsp. The number of eggs eaten per larva increasedsignificantly as egg availability increased on both sorghum and pigeonpea. In small cages 21–37% of eggs were eaten on sorghum, 4–12%on pigeonpea. Plant feeding declinedsignificantly on both sorghum and pigeonpea asegg density increased. Cannibalism was greateron sorghum than on pigeonpea while plantfeeding was greater on pigeonpea than onsorghum. Only around 8% of eggs were eaten inlarger cages with sorghum. The response toincreasing egg availability in all experimentswas linear. Immunoassay with an anti-vitellinmonoclonal antibody showed that egg cannibalismoccurs on pigeonpea under field conditions.Seven percent of all larvae had egg protein intheir gut. Cannibalism may make a significantcontribution to H. armigera populationsuppression.  相似文献

A precise bioassay method, which is not limited by lack of field applicability, as are peroral administration techniques, is described. Purified nuclear polyhedrosis virus (NPV) suspensions were assayed against third and fourth instar Heliothis armigera larvae to provide standards for additive and field testing. Third instar larvae proved to be approximately one hundred times more susceptible to the NPV disease than fourth instar larvae. The minimum time to mortality was 4 days.  相似文献

American bollworm (ABW), Helicoverpa armigera (Hubner), is considered as a major pest of cotton, Gossypium hirsutum, all over the globe. Due to its destructive feeding nature and continuous consumption of the same chemicals, it devolved resistant against many insecticides. Therefore, a combined application of bio- and synthetic-pesticide need to evaluate against this pest. The entomopathogenic viruses like nuclear polyhedrosis virus (NPV), a member of baculoviruses, can be the potential candidates for better control against ABW. The present study was conducted to assess the comparative efficacy of NPV and Spinosad 240SC (with the concentration of 250 mL · ha ̄?) against ABW in the controlled environment. The ABW was treated with different concentrations of NPV and Spinosad separately and in a combination of NPV with 0.1% Spinosad. The results revealed that highest concentrations showed highest mortality (95%) followed by 95%, 92%, 84%, 82% and 78% mortality at 1 × 109, 1 × 108, 1 × 107, 1 × 106 and 1 × 105 POBs, respectively. Spinosad when mixed in diet give 100% mortality at 0.8% followed by 50.87%, 42.10%, 29.82%, 26.31% and 22.80% mortality at 0.4%, 0.2%, 0.1%, 0.5% and 0.025% respectively. The results of this study revealed that microbial control of ABW through NPV is an effective tool. The repeated use of synthetic pesticides caused the resurgence of many insect pests, and this study results would provide useful insight to build a framework for future investigations for the management of many major insect pests.  相似文献

1. Experiments were carried out to investigate the incidence of cannibalism throughout the larval development of the noctuid moth Spodoptera frugiperda, and to examine the risk of infection from consuming conspecifics infected with a nuclear polyhedrosis virus (SfNPV). 2. Cannibalism was observed commonly even when food was not limiting, but occurred more frequently at low food quantities and/or high rearing densities. The sex of the larvae had no effect on the incidence of cannibalistic behaviour, however the probability of cannibalism occurring was affected by larval stage. The frequency of cannibalism was significantly higher among fifth- and sixth-instar larvae than among earlier instars, and larvae were more likely to consume younger conspecifics than larvae of the same stage. 3. Fifth-instar larvae offered fourth-instar victims fed equally on healthy larvae, virus-infected larvae (2 days post-infection), uninfected corpses, and virus-killed corpses (6 days post-infection). Horizontal transmission of SfNPV was only recorded in larvae offered virus-killed corpses, however, and total mortality in this treatment was only 32%. 4. In a similar experiment, fourth-instar larvae avoided cannibalising virus-killed corpses. Horizontal transmission of SfNPV was recorded in fourth-instar larvae that consumed 2-day post-infected larvae. The low incidence of cannibalism observed in fourth-instar larvae, however, suggests that this is unlikely to provide an important route for the transmission of SfNPV.  相似文献

对棉铃虫Helicoverpa armigera(Hübner)的同类相残进行行为学观测,以分析体重在胜负中作用,以及不同寄主植物情况下,同类相残导致的数量变化。结果表明:棉铃虫同类相残包括试探性攻击、反击、追击、嘶咬、缠绕、取食等过程,其中能否成功缠绕对方是成功的关键。体重在同类相残过程中起到重要的作用,同一龄期,体重大的个体获胜可能性显著大于体重较小的个体。寄主植物的营养状况对同类相残有着显著影响,不适宜寄主植物环境下,同类相残在低龄期即可降低种群数量。  相似文献

All the instars of Heliothis armigera larvae were found to be susceptible to both nuclear polyhedrosis virus (NPV) and granulosis virus (GV). An inverse relationship between mortality and larval age was found in the case of the NPV, while the GV displayed a rather erratic mortality pattern. A degree of maturation immunity against the NPV was found to exist, but the same is not true for the GV. The important role that pupation plays on the effect of a lethal infection is also discussed.  相似文献

A comparative study of Spodoptera nuclear polyhedrosis virus (NPV) and Autographa NPV replication in Spodoptera exigua revealed some cytopathologic differences. Infection with Spodoptera NPV was accompanied by electron-dense intranuclear granules. Autographa infection within the midgut led to secretion within the lumens of the goblet cells of paracrystalline arrays of small, round particles, 9.5 nm in diameter. Autographa virus was also observed in various stages of possible replication within the cytoplasm.  相似文献

The effects of photoperiod and temperature on growth parameters of Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) were investigated under laboratory conditions by exposing its larvae to fluctuating and alternating temperature regimes. Our data made evident that the interplay between photoperiod and temperature has a direct effect on growth parameter responses of H. armigera. However, the type of temperature regime (fluctuating or alternating) may enhance or diminish the effects of photoperiod. With fluctuating temperature, larval developmental time was significantly reduced with increasing photophase, irrespective of the diapause status later in the pupal stage; the lowest value was observed under continuous light conditions. With alternating temperature, larval developmental time was significantly decreased with the reduction in temperature amplitude, and with the coincidence of the thermophase with the scotophase. At both temperature regimes, no significant differences in pupal weight were observed between diapausing and non‐diapausing individuals, although, in most treatments, the diapausing pupae tended to be heavier than the non‐diapausing ones. The increased pupal weight of individuals destined for diapause is probably related to their longer larval developmental time. Knowing the effects of these factors on the variation of H. armigera growth is necessary in explaining phenomena associated with immature stages, and can lead to a more profound understanding of the potential for this insect to evolve in response to environmental changes.  相似文献

Helicoverpa armigera (Hubner) is a pest that poses serious threat to the tomato crop in India. Larvae of this species are susceptible to the nucleopolyhedrovirus (NPV), which has attracted interest as a potential biocontrol agent. Rearing of larvae in natural diet resulted in less number of pupae, while in artificial diet more pupae and healthy adults were obtained. The NPV was tested for its insecticidal action against H. armigera in natural and artificial diets. The bioassay results of inoculated H. armigera nucleopolyhedrosis virus (HaNPV) with different concentrations indicate that the 4.0 g/l dosage caused maximum mortality (70.3% and 60.54%), and minimum mortality 46.83% and 44.08% was recorded in the 0.5 g/l dosage under laboratory and pot culture conditions, respectively. Polyhedral inclusion bodies (PIBs) were estimated using a standard haemocytometer. According to the estimate, purified PIBs were found at a concentration of 1 × 109/ml. The different formulation of NPV, i.e. wettable fine powder with a hue of white, had a particle size of 100 μm with a concentration of 1 × 108/ml. The encapsulated product was found to be in the form of beads and was red in colour with a concentration of 1 × 108 PIBs/ml. The effectiveness of NPV against H. armigera larvae was noted at maximum LC50 values at a concentration of 1 × 109/ml. Qualitative analysis estimates showed that protein polyhedrin and enzyme chitinase had a molecular weight of 33 and 45 kDa, respectively. Viral DNA was isolated from NPV-infected larvae and was then separated using SDS-PAGE. The estimated polyhedrin with a molecular weight of 33 kDa, which is responsible for the increased mortality percent of larvae, was confirmed by the bioassay. Hence it was concluded that NPV is ecofriendly in the management of pests on tomato and other crops.  相似文献

Helicoverpa armigera stunt virus (HaSV) is a member of the Tetraviridae family of RNA viruses whose replication and expression strategies are not well understood due to the absence of an in vitro cell culture system. We set out to find such a system for HaSV by screening an array of 13 insect and 1 mammalian cell culture lines with both virus particle infection and genomic RNA transfection. No cell line was found to be permissive for replication, although entry of genomic RNA was verified. The apparent specificity of this virus for its in vivo midgut target site was strongly corroborated by studies involving Northern blots of RNA extracted from infected insects. Only larval midgut RNA showed the presence of virus after hosts were infected per os or by injection which exposed other host cell types to the virus. The absence of replication in cell culture was due to a lack, or presence, of host factors important to replicase activity and also the likely absence of virus particle binding and entry. We thus provide both in vitro- and in vivo-based evidence demonstrating that this virus is extremely specific in the type of cells in which it will initiate an infection.  相似文献

Baculoviruses from Autographa californica (AcNPV-E2) and Spodoptera frugiperda (SfNPV-2) were titered in five insect cell lines: IAL-PID2, IAL-SFD1, IPLB-SF-21AE, TN-368, and IAL-TND1. AcNPV-E2 replicated in all the cell lines while SfNPV-2 did not replicate in the lines TN-368 and IAL-TND1. Further in vivo studies of SfNPV-2 showed the virus was not infectious when fed to Trichoplusia ni larvae per os or when injected into the hemocoel. These data suggest that the barrier to SfNPV-2 infectivity in T. ni is at the cellular level, as opposed to the midgut.  相似文献

The first comprehensive field trial using an insect small RNA virus as a control agent on a cropping system was conducted with the Helicoverpa armigera stunt virus (family Tetraviridae, genus Omegatetravirus, HaSV). The virus was semipurified, quantified, and applied at two rates, 4 x 10(15) and 4 x 10(14) virus particles/ha, with minimal formulation on sorghum against the bollworm Helicoverpa armigera (Hübner). For comparison, a commercial preparation of Helicoverpa zea single-nucleopolyhedrovirus (HzSNPV, Gemstar) was applied at the same time at 9.27 x 10(11) polyhedral inclusion bodies/ha. The HaSV application rates were determined by a novel procedure using laboratory LC50 bioassay data for HaSV and HzSNPV and calibration to the known field application rate of the HzSNPV. The baculovirus and the higher rate of HaSV produced statistically equivalent reductions in the larval populations of around 50% at both 3 and 6 d postapplication (dpa) compared with untreated plots. The 10-fold lower rate of HaSV reduced the larval population by 50% at 3 dpa and approximately 30% at 6 dpa. Persistence of HaSV over a 72-h period was found to be similar to that of HzSNPV, although the amount of HaSV available on the sorghum heads increased at 130 h postapplication, due most likely to dispersal of newly produced virus from cadavers and frass. The results from this trial indicate that HaSV could be used as an effective biopesticide for the control of H. armigera in sorghum and the ramifications for its broader use are discussed.  相似文献

Nuclear polyhedrosis virus ofHelicoverpa armigera (HaNPV) from Madurai (south India) was isolated, purified and subjected to electron microscopical studies. The results indicate that the isolate is a multi embedded form of NPV. Further, STEM observations on polyhedra and polyhedral envelope of the HaMNPV are presented. The restriction pattern and genome size of HaMNPV Madurai isolate are compared with HaNPV isolates reported earlier. A dendrogram is presented to show the relatedness among the isolates.  相似文献

To evaluate the hygienic risk involved in using baculoviruses for insect pest control, safety studies are required. Pigs were chosen as representative test animals of commercial and agricultural importance. The tests were aimed at detecting virus propagation, immune reactions, and signs of acute infection (changes in body temperature and hematology profile, swelling of lymph nodes). Four of five animals inoculated with nuclear polyhedrosis virus showed a slight temperature rise at day 2 postinfection. After day 4 postinfection, no differences between infected animals and controls were observed. In the bioassay, virus activity could be recovered from fecal samples; however, no activity was found in organ extracts. The data did not indicate hygienic risks involved in the application of nuclear polyhedrosis virus, especially that from Mamestra brassicae, in biological pest control.  相似文献

In order to find the cellular interaction factors of the Heliothis armigera nuclear polyhedrosis virus capsid protein VP39, a Heliothis armigera cell cDNA library was constructed. Then VP39 was used as bait. The host actin gene was isolated from the cDNA library with the yeast two-hybrid system. This demonstrated that VP39 could interact with its host actin in yeast. In order to corroborate this interaction in vivo, the vp39 gene was fused with the green fluorescent protein gene in plasmid pEGFP39. The fusion protein was expressed in the Hz-AM1 cells under the control of the Autographa californica multiple nucleopolyhedrovirus immediate early gene promoter. The host actin was labeled specifically by the red fluorescence substance, tetramethy rhodamine isothicyanete-phalloidin. Observation under a fluorescence microscopy showed that VP39, which was indicated by green fluorescence, began to appear in the cells 6 h after being transfected with pEGFP39. Red actin cables were also formed in the cytoplasm at the same time. Actin was aggregated in the nucleus 9 h after the transfection. The green and red fluorescence always appeared in the same location of the cells, which demonstrated that VP39 could combine with the host actin. Such a combination would result in the actin skeleton rearrangement.  相似文献
