Dynamics of growth and oxygen consumption during ontogenesis of insects with direct (striped shield bug Graphosoma lineatum L.) and indirect (cabbage moth Mamestra brassicae L.) development have been compared. The correlation between a character of energy metabolism alteration and peculiarities of development of the insects has been shown. Cyclic decrease of oxygen consumption during molt and sharp dropping during metamorphosis have been observed in insects with indirect development. The decrease of oxygen consumption has been observed in insects with direct development only during molts. The coefficient a of allometric dependence of oxygen consumption on body weight of imago for cabbage moth was two times higher than that for striped shield bug.  相似文献

Lactic acid bacteria (LAB) are generally sensitive to hydrogen peroxide (H2O2), Lactobacillus sakei YSI8 is one of the very few LAB strains able to degrade H2O2 through the action of a heme-dependent catalase. Lactobacillus rhamnosus strains are very important probiotic starter cultures in meat product fermentation, but they are deficient in catalase. In this study, the effect of heterologous expression of L. sakei catalase gene katA in L. rhamnosus on its oxidative stress resistance was tested. The recombinant L. rhamnosus AS 1.2466 was able to decompose H2O2 and the catalase activity reached 2.85 μmol H2O2/min/108 c.f.u. Furthermore, the expression of the katA gene in L. rhamnosus conferred enhanced oxidative resistance on the host. The survival ratios after short-term H2O2 challenge were increased 600 and 104-fold at exponential and stationary phase, respectively. Further, viable cells were 100-fold higher in long-term aerated cultures. Simulation experiment demonstrated that both growth and catalase activity of recombinant L. rhamnosus displayed high stability under environmental conditions similar to those encountered during sausage fermentation.  相似文献

A protocol for in vitro propagation of the wild germander (Teucrium polium L.) was developed. In vitro plants were developed from ex vitro axillary buds. Then, shoot tips were excised and established on Murashige and Skoog medium. Proliferation of shoots was tested with different levels of 6-furfurylaminopurin, 6-benzyladenine, or thiadiazuron. The highest proliferation of T. polium was obtained when 6-benzyladenine and 6-furfurylaminopurin were used at 2.0 and 1.6 mg l−1, respectively. Thiadiazuron gave the lowest response for shoot proliferation. Rooting was experimented at different levels of Indol-3-butric acid, Indol-3-acetic acid, or 1-naphthaleneacetic acid. 1-Naphthaleneacetic was the only growth regulator which promoted root induction. Rooted plants were acclimatized successfully with 75% survival and grown in the greenhouse. In vitro- and in vivo-grown plants were analyzed for essential oil production. In vitro-grown T. polium on MS medium supplemented with 6-benzyladenine and 1-naphthaleneacetic gave higher oil yield than that grown on hormone-free Murashige and Skoog medium. In vivo (wild)-grown T. polium produced different oil yield when collected in different months (April and October). β-caryophyllene, used as a marker compound in the essential oil, was identified and quantified by gas chromatography (GC) analysis. Gas chromatography/mass (GC-MS) spectrometry analysis was also used to identify other components of in vitro cultures and to compare with in vivo-grown plants.  相似文献

We have recently indicated the plant growth promoting activities of Pseudomonas sp. as well as their alleviating effects on some soil stressors such as salinity. This is because in recent years, biological fertilizers have received special attention by scientists in sustainable agriculture. Accordingly, it is pertinent to specify the beneficiary level of such soil bacteria on plant growth including phosphorous (P) uptake. Hence, the objectives were to determine: (1) the plant growth promoting effects of the tested Pseudomonas sp., and (2) its combined effects with different P fertilization rates on the nutrient uptake (N, P, and K) and yield of wheat (Triticum aestivum L.) under greenhouse and field conditions. The experiments were factorially arranged on the basis of a completely randomized block design with three replicates and were conducted at the Research Farm of Agriculture and Natural Resources Research Center of Khorasan, Mashhad, Iran. P was fertilized at three levels including 0, 25 and 50 kg/ha P2O5. Pseudomonas sp. including Pseudomonas fluorescens 153, P. fluorescens 169, P. putida 4, and P. putida 108 were tested. Activities such as production of ACC deaminase and IAA-like products, as well as P solubilization were among the most important activities of the tested Pseudomonas sp. Such bacterial effects greatly enhanced wheat growth and yield under greenhouse and field conditions. The results also showed that the effects of Pseudomonas sp. on wheat nutrient uptake and the effects of bacteria as well as P fertilization on wheat yield were significant. P. putida 108 was the most effective strain enhancing wheat P uptake and grain yield under greenhouse (96 and 58%) and field (80 and 37%) conditions, respectively. Hence, although Pseudomonas sp. could be a suitable replacement for high P fertilization, however, the optimum wheat yield resulted when the bioinoculants are combined with 50% (25 kg/ha P2O5) P fertilization. Optimization of conditions for hydroquinone biotransformation into its β-d-glucoside, arbutin, in agitated shoot cultures of Ruta graveolens L. and Hypericum perforatum L. allowed us to obtain a maximum content of this important therapeutic and cosmetic product of 7.8 and 7.2% (dry weight), respectively. These contents are higher than respective values required for standardization of known arbutin-containing plant raw materials according to the European Pharmacopoeia and national pharmacopoeias of European countries.  相似文献

In vitro regeneration of black nightshade (Solanum nigrum L.) plants was achieved through callus-mediated shoot organogenesis followed by 30 d indoor ex vitro adaptation to nutritional stress under environmental ambience and thereafter 6-d outdoor acclimatization in pots prior to field establishment. Relevant physiological parameters including pigment content, chlorophyll a fluorescence, net photosynthetic rate (P N), transpiration rate (E), and stomatal conductance (g s) of in vitro-regenerated plants were investigated during the course of ex vitro adaptation. During the first 4 d of indoor transplantation to potting substrate, there was a marginal reduction in the leaf chlorophyll and carotenoid contents but P N and E were strongly reduced. The stomatal conductance and E/P N ratio were significantly higher in plants up to 20 d of indoor adaptation than those of comparable age grown naturally from seeds. The shape of the OJIP fluorescence transient varied significantly with acclimatization, and the maximum change was observed at 2.0 ms. The 2.0 ms variable fluorescence (V j), 30 ms relative fluorescence (M 0), photon trapping probability (TR0/Abs), and photosystem II (PSII) trapping rate (TR0/RC) showed initial disturbance and subsequent stabilization during 30 d of indoor acclimatization. Energy dissipation (DI0/RC) and electron transport probability (ET0/TR0) showed an initial phase of increase during the 4 d after plants were transplanted outdoors. During the 6-d outdoor acclimatization after transfer of plants to soil, no significant change in total chlorophylls and carotenoids, E, and g s were observed, but P N improved after reduction on the first d. The OJIP-derived parameters experienced change on the first d but were stabilized quickly thereafter. There was no significant difference between outdoor acclimatized plants and those of the seed-grown plants of comparable age with respect to photosynthetic and fluorescence parameters. Direct transfer of plants without indoor acclimatization, however, showed a completely different trend with respect to P N, E, and OJIP fluorescence transients. The bearing of this study on optimizing micropropagation is discussed.  相似文献

Picris hieracioides var. japonica (Asteraceae), which grows in occasionally disturbed habitats such as riverbanks, is rarely observed under dense vegetation. We examined the effect of the experience and timing of receiving leaf-transmitted light in gap-detecting seed germination in this plant. Seeds under unfiltered light, which simulated the light conditions of seeds on the soil surface in a canopy gap, germinated at a constant temperature of 20°C. However, most seeds in darkness, which simulated the light conditions of seeds buried in the soil without receiving leaf-transmitted light, germinated under temperature fluctuations of over 4°C. Seeds in darkness after receiving leaf-transmitted light for 1 week, which simulated the light conditions of seeds buried in the soil after receiving leaf-transmitted light, germinated under temperature fluctuations of over 8°C. Finally, seeds under continuous leaf-transmitted light, which simulated the light conditions of seeds on the soil surface below preexisting vegetation, germinated under temperature fluctuations of over 12°C. Seeds that experience unfiltered light, which suggests that they are in a gap, should not delay germination. In contrast, seeds that have received leaf-transmitted light should delay germination until the vegetation above is removed. Seeds exposed to leaf-transmitted light required larger temperature fluctuations in darkness than did untreated seeds, and seeds under continuous leaf-transmitted light required the largest temperature fluctuations. The various germination reactions to each gap signal in P. hieracioides var. japonica seeds allow the more reliable detection of gaps for subsequent seedling establishment. The requirement for gap signals that created high precision of timing in the germination process results in the germination of this species only in gaps. Therefore, P. hieracioides var. japonica is rarely found under dense vegetation.  相似文献

Some exotic plants are able to invade habitats and attain higher fitness than native species, even when the native species are closely related. One explanation for successful plant invasion is that exotic invasive plant species receive less herbivory or other enemy damage than native species, and this allows them to achieve rapid population growth. Despite many studies comparing herbivory and fitness of native and invasive congeners, none have quantified population growth rates. Here, we examined the contribution of herbivory to the population dynamics of the invasive species, Lespedeza cuneata, and its native congener, L. virginica, using an herbivory reduction experiment. We found that invasive L. cuneata experienced less herbivory than L. virginica. Further, in ambient conditions, the population growth rate of L. cuneata (λ = 20.4) was dramatically larger than L. virginica (λ = 1.7). Reducing herbivory significantly increased fitness of only the largest L. virginica plants, and this resulted in a small but significant increase in its population growth rate. Elasticity analysis showed that the growth rate of these species is most sensitive to changes in the seed production of small plants, a vital rate that is relatively unaffected by herbivory. In all, these species show dramatic differences in their population growth rates, and only 2% of that difference can be explained by their differences in herbivory incidence. Our results demonstrate that to understand the importance of consumers in explaining the relative success of invasive and native species, studies must determine how consumer effects on fitness components translate into population-level consequences. Selection on traits conferring reduced predation may be opposed by selection on other traits associated with reproduction. Here, we examined the hypothesis that traits associated with reproduction in Gammarus pulex are driven by predation. We studied G. pulex originating from ponds with two different kinds of predator regimes: (1) ponds with fish—often large, non-gap-limited predators and (2) ponds without fish where invertebrates are the dominant predators—often small, gap-limited predators with a much more restricted prey size range. We examined the body size of males and females in G. pulex amplexus pairs originating from fish and fishless ponds. We also examined, in the laboratory, their mating success, the number of offspring per female and offspring mortality under different rearing conditions, with or without fish cue. Mating success, defined as the percentage of amplexus pairs that produced live offspring, was higher for G. pulex from fishless ponds independent of rearing condition. Individuals from fish ponds were larger and they produced a higher number of offspring which tended to be related to female body size. Offspring mortality was higher in populations from fish ponds compared to populations from fishless ponds. Despite the higher offspring mortality, females from fish ponds had a higher number of offspring alive after 13 weeks, which is the approximate time it takes for G. pulex to reach maturity. Our data imply that no trade-off between reducing body size to reduce mortality caused by fish and maximising reproductive success exist in G. pulex from fish ponds. The strategy with many offspring may be the correct strategy in fishponds where predation pressure generally is higher than in fishless ponds.  相似文献

As a step toward greater understanding of the genetics of verticillium wilt resistance in plants, we report the sequencing of a candidate wilt resistance gene, mVe1, from the mint diploid model species, Mentha longifolia (Lamiaceae). mVe1 is a putative homolog of tomato (Solanum lycopersicum L.) verticillium wilt (Ve) resistance genes. The mVe1 gene has a coding region of 3,051 bp. The predicted mVe1 protein contains a leucine-rich repeat domain, a common feature of plant disease resistance proteins. We compared 13 mVe1 alleles from three mint species. These alleles shared 96.2–99.6% nucleotide identity. We analyzed four M. longifolia populations segregating with respect to mVe1 alleles and wilt resistance versus susceptibility and found one association between mVe1 genotype and wilt phenotype. We conclude that mVe1 may play a role in mint verticillium wilt resistance, but variation for resistance in our segregating progenies is likely polygenic. Therefore, further investigations of mVe1 and identification of additional candidate genes are both warranted.  相似文献

Forty-day-old seedlings of Atriplex halimus were treated either with NaCl (50, 300 and 550 mM) for the subsequent 30 days or with 15% PEG for the subsequent 10 days. As much as 50 mM of NaCl significantly increased shoot fresh and dry weight and height; nevertheless, 300 or 550 mM NaCl seemed to have no effect. On the other hand, these growth parameters were not affected by drought after 3 or 6 days, but were reduced after 10 days. The gas exchange parameters (photosynthetic rate, stomatal conductance and transpiration rate) were increased by 50 mM NaCl, but decreased by 300 and 550 mM. These parameters were decreased in response to drought only after 10 days of withholding water. In contrast to Na+, K+ was significantly decreased by NaCl but not by drought. The time course effect revealed that phosphoenol pyruvate carboxylase (PEPC) protein was doubled in response to NaCl after 1 and 5 h and continued thereafter, higher than control, while drought had no significant effect. Rubisco seemed unchanged by NaCl or drought. It could be concluded that the decrease in fresh weight might be attributed to the decrease in water content. Moreover, the decrease in photosynthesis could result from a decrease in stomatal conductance, a protective mechanism against water loss to improve water use efficiency. These findings indicate that Atriplex halimus tolerates NaCl and drought through decreasing growth, reducing gas exchange parameters to improve water use efficiency, uptake Na+ and saving, if any, the photosynthetic enzyme particularly PEPC.  相似文献

Respiration and photosynthesis are two important processes in microalgal growth that occur simultaneously in the light. To know the rates of both processes, at least one of them has to be measured. To be able to measure the rate of light respiration of Chlorella sorokiniana, the measurement of oxygen uptake must be fast, preferably in the order of minutes. We measured the immediate post-illumination respiratory O2 uptake rate (OUR) in situ, using fiber-optic oxygen microsensors, and a small and simple extension of the cultivation system. This method enables rapid and frequent measurements without disturbing the cultivation and growth of the microalgae. Two batch experiments were performed with C. sorokiniana in a short light-path photobioreactor, and the OUR was measured at different time points. The net oxygen production rate (net OPR) was measured online. Adding the OUR and net OPR gives the gross oxygen production rate (gross OPR), which is a measure for the oxygen evolution by photosynthesis. The gross OPR was 35–40% higher than the net OPR for both experiments. The respiration rate is known to be related to the growth rate, and it is suggested that faster algal growth leads to a higher energy (ATP) requirement, and as such, respiratory activity increases. This hypothesis is supported by our results, as the specific OUR is highest in the beginning of the batch culture when the specific growth rate is highest. In addition, the specific OUR decreases toward the end of the experiments until it reaches a stable value of around 0.3 mmol O2 h−1 g−1. This value for the specific OUR is equal to the maintenance requirement of C. sorokiniana as determined in an independent study of (Zijffers et al. 2010 (in press)). This suggests that respiration could fulfill the maintenance requirements of the microalgal cells.  相似文献

We conducted a laboratory experiment to investigate the influence of Daphnia infochemicals on growth rate, microcystin production, colony formation and cell size of eight Microcystis strains isolated from two lakes. The strains were characterized genetically by their 16S-23S rDNA ITS sequence. The experiment was composed of four treatments: (1) a control using filtered WC medium, (2) addition of Scenedesmus obliquus culture medium filtrate, (3) addition of Daphnia magna culture medium filtrate and (4) addition of sodium octyl sulphate, a commercially available Daphnia infochemical. Our results showed that sympatric strains differed strongly for the measured functional traits, while no correlations between traits were found. Between-strain differences in growth rate, microcystin production, colony formation and cell size were generally larger than the differences in phenotypes observed between treatments. Despite this, several strains reacted to the infochemicals by changing functional trait values. Daphnia culture medium filtrate and, to a lesser extent, sodium octyl sulphate had a negative influence on the growth rate of half of the strains and stimulated microcystin production in one strain, but the latter effect was not Daphnia-specific as Scenedesmus culture medium filtrate had the same effect. Daphnia culture medium filtrate also induced colony formation in one strain. Our data suggest that Daphnia infochemicals generally have a weak influence on growth rate, microcystin production and colony formation of Microcystis strains as compared to the inter-strain variability, while existing inducible effects are highly strain-specific.  相似文献

The effect of p-nitrophenol (PNP) concentration with or without glucose and yeast extract on the growth and biodegradative capacity of Ralstonia eutropha was examined. The chemical constituents of the culture medium were modeled using a response surface methodology. The experiments were performed according to the central composite design arrangement considering PNP, glucose and yeast extract as the selected variables whose influences on the degradation was evaluated (shaking in reciprocal mode, temperature of 30°C, pH 7 and test time of about 9 h). Quadratic polynomial regression equations were used to quantitatively explain variations between and within the models (responses: the biodegradation capacity and the biomass formation). The coefficient of determination was high (R adjusted2 = 0.9783), indicating the constructed polynomial model for PNP biodegradative capacity explains the variation between the regressors fairly well. A PNP removal efficiency of 74.5% occurred within 9 h (15 mg/L as the initial concentration of PNP with use of yeast extract at 0.5 g/L).  相似文献

There are three most important bacterial causative agents of serious infections that could be misused for warfare purposes: Bacillus anthracis (the causative agent of anthrax) is the most frequently mentioned one; however, Fracisella tularensis (causing tularemia) and Yersinia pestis (the causative agent of plague) are further bacterial agents enlisted by Centers for Disease Control and Prevention into the category A of potential biological weapons. This review intends to summarize basic information about these bacterial agents. Military aspects of their pathogenesis and the detection techniques suitable for field use are discussed.  相似文献

The combined effects of drought and salinity were investigated on the growth, efficiency and competitiveness of three Mesorhizobium sp. strains (ORS 3416, ORS 3428 and ORS 3437) isolated from Acacia senegal (L.) Willd. Sixteen combinations of polyethylene glycol (PEG) 6000 (0, 6, 12, 20%, w/v) and NaCl (0.005, 0.5, 1, 1.5%, w/v) were used to study rhizobial strains growth in screw-capped tubes containing yeast extract mannitol liquid medium. Whatever the combination, the strains grew. However, high concentrations of PEG 6000 and NaCl were depressed on rhizobial growth but strain ORS 3428 was the most tolerant. The effect of rhizobial inoculation on growth of A. senegal plants was also examined under non-limited (100% of water holding capacity, WHC) and water-limited conditions (50% WHC) in the greenhouse for 4 months on non-sterile sandy soil. A significant decrease in the nodulation rate, nodule dry weight, plant height and shoot dry weight was observed under water-limited conditions. Nevertheless, inoculation with the three selected Mesorhizobium strains significantly improved nodulation and A. senegal plant growth under water-limited conditions. Whatever the irrigation condition, strains ORS 3428 and ORS 3437 were more competitive than ORS 3416. These rhizobial strains may prove valuable for inoculation of Acacia senegal plants grown for transplantation to dry saline habitats.  相似文献

 The phylogeny of the genus Gunnera is investigated for the first time. Twelve species representing the six currently recognised subgenera are analysed. Two chloroplast DNA regions, the rbcL gene and the rps16 intron, together provide 46 informative characters out of 2335. A combined analysis of both genes gives four most parsimonious trees, firmly establishing the east South American G. herteri as sister group to the rest of the genus. The African G. perpensa is sister group to two well-supported clades, one including the South American subgenera Misandra and Panke, the other the Australian/New Zealand/Malayan species of subgenera Milligania and Pseudogunnera. Thus, South America is a composite area for Gunnera, showing up at two different levels in the cladogram. Our analysis supports a close biogeographic relationship between Australia and New Zealand. The evolution of some morphological characters is discussed. Lastly, the unusual structure of some of the rbcL sequences is reported. Received July 6, 2000 Accepted October 24, 2000  相似文献

Segregation analysis was performed in the progenies obtained in analyzing crosses of hybrids of spring and winter accessions of rye Secale cereale L. and wild S. montanum subsp. anatolicum (Grossh.) Tzvel. (syn. S. strictum (J. Presl) J. Presl). The test genes controlled the brittle stem (bs), the allelic variants of aromatic alcohol dehydrogenase (Aadh1) and shikimate dehydrogenase (Skdh), and the growth habit (Vrn1). A linkage was observed in the inheritance of the brittle stem and the aromatic alcohol dehydrogenase and shikimate dehydrogenase alloenzymes. The order of genes was established to be bs-Skdh-Aadh1, and the genetic distances were estimated to be bs-(9.0%)-Skdh, bs-(10.8%)-Aadh1, and Skdh-(5.3%)-Aadh1. The recombination coefficient between the Skdh and Aadh1 genes varied from 2.2 to 18.2%, averaging 5.3%. The growth habit was inherited independently of the bs-Skdh-Aadh1 linkage group.  相似文献

The work on the introduction of the zander Sander lucioperca (L.) from Lake Onega to Lake Vygozero is reviewed. A comparative analysis is performed for the biological parameters of zander from these lakes. The zander population from Lake Vygozero differs from that of Lake Onega in its lower growth rate, later maturation, and lower fat content of internal organs; these differences are determined by the limnological parameters and food supplies of both waterbodies.  相似文献
