Gibberellic-acid (GA3)-induced hypocotyl elongation of intact lettuce (Lactuca sativa L.) seedlings was inhibited by colchicine (4×10-4 M) but not by lumicolchicine (4×10-4 M). In excised lettuce hypocotyls, GA3 (10-5 M) increased respiration over water controls, while both colchicine and lumicolchicine alone, or in combination with GA3, reduced respiration. Microtubules were present in the hypocotyls of lumicolchicine-treated seedlings but absent in those treated with colchicine. It is suggested that lumicolchicine is a useful drug to discriminate between the metabolic and microtubule-mediated processes in cell morphogenesis.  相似文献

Seedlings of Pinus contorta were grown for 16 wk without ectomycorrhizas in the greenhouse at three levels of irradiance (100, 210 and 470 μEm-2s-1) and three levels of ammonium nitrate (3, 62 and 248 mg litre-1 of N). Measurements at 5, 10 and 16 wk of age indicated that plants receiving the highest levels of irradiance and nitrogen had significantly larger biomass, a higher total foliar N and P content and net photosynthesis per unit leaf area than those at lower levels of each factor. Although the root/shoot ratio also increased from low to high irradiance at each harvest, nitrogen application resulted in an increased ratio from 3 to 62 mg litre-1 N, but decreased ratio at 248 mg litre-1 N.  相似文献

Changes in soluble carbohydrate composition and concentration in leavesand petioles of watermelon (Citrullus lanatus (Thunb)Matsum and Nakai cv. Sugar Baby) seedlings during early stages ofphytochrome-regulated petiole elongation were investigated. Watermelon seedlingswere grown in a controlled environment with 350 molm–2 s–1 photosynthetically activeradiation (PAR) during a 12-h photoperiod. Low intensity end-of-day(EOD) light treatments (for 15 min) of red (R), far-red (FR) and FRfollowed by R (FR/R) were initiated when the seedlings were 14 days old.Seedling growth, and soluble carbohydrate concentration and composition inleaves and petioles were determined after 3 and 6 days of EOD light treatments.The EOD FR increased the petiole length and dry mass partitioned to petioles asearly as 3 days into the treatment. This increased petiole dry mass inFR-treated plants was accompanied with an increase in reducing sugar (glucoseand fructose) concentration in the petioles. Although both leaves and petiolesshowed this effect, the relative increase was greater in petioles than leaves.While the most abundant sugars in petioles were fructose and glucose, thepredominant sugars in leaves were sucrose, raffinose, and stachyose. Thephotoreversion of FR induced changes in growth and sugar concentrations by Rindicates the involvement of phytochrome in these processes.  相似文献

Deacclimation of Scots pine ( Pinus sylvestris ) in northern Finland (65°N) was studied from the perspective of protein metabolism, which was related to nitrogen fertilization. Two-year-old seedlings were fertilized with calcium ammonium nitrate (0, 442 and 884 kg N ha−1) in the summer prior to the study. Needles were harvested for analyses at 3-week intervals during the natural deacclimation period from mid-March to the beginning of June 1998. Deacclimation was followed by monitoring various physiological variables: freezing resistance decreased and osmotic potential increased during the experiment and needle dry weight increased from bud flushing onwards. The concentration of soluble proteins in needles was higher in fertilized seedlings but decreased transiently in the 884 kg N ha−1 fertilized seedlings before budbreak. The abundance of several small polypeptides (17–32 kDa) decreased in the spring. A 60-kDa protein, identified by immunoblotting as a dehydrin, was detected in all treatments. The quantity of this dehydrin decreased with resumption of growth, along with the appearance of 50- and 56-kDa dehydrins. The concentration of these dehydrins decreased during dehardening more rapidly in fertilized seedlings than in the control plants. The fertilized seedlings started to grow earlier than the unfertilized plants. In conclusion, nitrogen fertilization provided good reserves for new growth but did not affect deacclimation of pine needles.  相似文献

The concentration of free amino acids and total nitrogen was studied in needles, stems and roots of seedlings of Pinus sylvestris L. for five weeks during the second growth period ("summer"). In one group of seedlings the source/sink relation was disturbed through removal of the terminal buds. The seedlings were cultivated in artificial year-cycles in a climate chamber.
Total nitrogen increased in needles and sterns of intact seedlings in the beginning of the "summer" and decreased during shoot growth. In seedlings, from which the buds had been removed, nitrogen remained at high levels in the primary needles and accumulated in steins and roots. The results are consistent with utilization of nitrogen in older needles and in the stem during shoot elongation.
The pool of free amino acids increased in the beginning of the "summer" and decreased after bud break in primary needles, stems and roots. Arginine and glutamine, in the roots also asparagine, were the dominating amino acids (amides included). Together, these compounds (plus glutamate and aspartate) contributed about 90% of the nitrogen in the amino acid pool in all organs. In primary needles and in the stem, arginine predominated at the end of hardening (75–85% of the amino acid nitrogen). Free amino acids contributed at most ca 10% of the total nitrogen in primary needles, where the ratio of free amino acid nitrogen: total nitrogen was highest at the end of dormancy and in the early "summer". Free amino acids accumulated after bud removal in primary needles and especially in stems and roots. Glutamine became relatively more dominant than arginine in the different organs.
The observations are consistent with the role of arginine and glutamine for storage and transport of nitrogen in conifers. Because of the low concentrations of amino acid nitrogen in the primary needles, arginine is not considered a major nitrogen reserve in needles of Scots pine seedlings.  相似文献

Coniferin (coniferyl alcohol-β-d-glucoside) was not detected in pine seeds (Picea abies) but it accumulated in the stems and roots of pine seedlings. Pulse labeling experiments with l-phenylalanine-[U-14C] and 100-day-old pine seedlings in hydroponic solution showed a turnover of coniferin with a half life of about 60 hr. Pulse labeling with 14CO2 and seedlings kept in soil gave a half life of about 120 hr for coniferin. The results indicate that coniferin could be an intermediate of lignin biosynthesis in pine seedlings.  相似文献

Zeatin-9-riboside was identified in shoots and roots of Pinus resinosa by GC-MS analysis of its permethyl derivative. Based on their chromatographic properties on Sephadex LH-20 and C18 HPLC, and their susceptibility to enzymatic degradation, several other cytokinins have been tentatively identified. The basic fraction of both the roots and shoots contained zeatin, whereas the shoots contained dihydrozeatin-O-glucoside and the roots contained zeatin-O-glucoside. Zeatin-9-riboside monophosphate, isopentenyladenosine monophosphate ([9R-5P]iP) and glucosyl phosphate derivatives were detected in the acidic fractions from both roots and shoots. There were equivalent amounts of [9R-5P]iP in both roots and shoots. Tomato is a cold-sensitive crop that is vulnerable to chilling injury. To screen for chilling resistance in tomato mutants, seeds were irradiated with different doses of 60Co-γ and then evaluated according to the five stages of the chilling injury index. Moreover, physiological indexes and observation of the submicroscopic structure by electron microscopy were used to examine cold resistance in the mutants. The physiological index results showed much higher cold resistance in the mutants compared to wild type. The cellular structures of the cold-resistant mutants were more complete than in wild-type plants after chilling treatment. The chloroplasts of the mutants were close to the cell periphery; the double membrane structure of the chloroplast was intact, and well-developed granal stacks were interconnected by stroma. Compared to wild-type plants, the stomatal density of cold-resistant mutants increased considerably, though the stomatal size showed no obvious changes.  相似文献

Loblolly pine (Pinus taeda L.) seedlings were nitrogen fertilized during winter in a bare root forest tree nursery located in the coastal plain of the southeastern United States. Total application rates were 0, 50, 100, and 200 kg/N/ha applied in split applications 4 weeks apart in January and February. Seedlings were lifted and outplanted in March, 4 weeks after the second fertilization and measured at 3 and 6 months after outplanting. No seedling morphological differences were encountered at the time of lifting and outplanting although seedling shoot nitrogen content was 28% greater in the highest fertilization treatment compared to the check. Shoot nitrogen concentrations fell after outplanting regardless of treatment, decreasing from an average of 1.51% across all treatments at the time of planting to 0.64% at 6 months after planting. When measured at 6 months after outplanting, seedling dry weight and height growth after planting was shown to increase by 12% and 24%, respectively, for the high nitrogen treatment. This and other studies across a variety of sites have found positive post-outplanting seedling growth response after nutrient loading in the nursery.  相似文献

Aluminum (Al) in the rhizosphere adversely affects plant nutrition and growth. Although many conifer species, and pitch pine (Pinus rigida) in particular, have evolved on acidic soils where soluble Al is often high, controlled environment studies often indicate that Al interferes with seedling growth and nutrient relations. Under normal field conditions, conifer roots grow in a symbiotic relationship with ectomycorrhizal fungi, and this association may modulate the effects of Al on root physiology. To investigate the influence of mycorrhizal infection on Al toxicity, pitch pine seedlings were grown with or without the ectomycorrhizal symbiont Pisolithus tinctorius and were exposed to low levels of Al in sand culture. Aluminum at 50 μM reduced nonmycorrhizal seedling growth and increased foliar Al concentrations, but did not alter photosynthetic gas exchange or other aspects of seedling nutrition. Nonmycorrhizal seedlings exposed to 200 μM Al exhibited decreased growth, increased transpiration rates, decreased water use efficiency, increased foliar Al and Na levels, and reduced foliar P concentrations. Seedlings inoculated with P. tinctorius exhibited unaltered growth, physiological function, and ionic relations when exposed to Al. The fungal symbiont evidently modulated ionic relations in the rhizosphere, reducing Al-P precipitation reactions, Al uptake, and subsequent root and shoot tissue Al exposure.  相似文献

Modern corn ( Zea mays L.) varieties have been selected for their ability to maintain productivity in dense plantings. We have tested the possibility that the physiological consequence of the selection of the modern hybrid, 3394, for increased crop yield includes changes in responsiveness to auxin and light. Etiolated seedlings in the modern line are shorter than in an older hybrid, 307, since they produce shorter coleoptile, mesocotyl, and leaves (blade as well as sheath). Etiolated 3394 seedlings, as well as isolated mesocotyl and sheath segments, were less responsive to auxin and an inhibitor of polar auxin transport, N-1-naphthylphthalamic acid (NPA). Reduced response of 3394 to auxin was associated with less reduction of elongation growth by light (white, red, far-red, blue) than in 307, whereas the activity of polar auxin transport (PAT) and its reduction by red or far-red light was similar in both genotypes. NPA reduced PAT in etiolated 3394 seedlings much less than in 307. A characteristic feature of 3394 plants is more erect leaves. In both hybrids, light (white, red, blue) increases leaf declination from the vertical, whereas NPA reduces leaf declination in 307, but not in 3394. Our results support findings that auxin and PAT are involved in elongation growth of corn seedlings, and we show that light interacts with auxin or PAT in regulation of leaf declination. We hypothesize that, relative to 307, more erect leaves in the modern hybrid may be primarily a consequence of a reduced amount of auxin receptor(s) and reduced responsiveness to light in etiolated 3394 plants. The more erect leaves in 3394 may contribute to the tolerance of the modern corn hybrid to dense planting.  相似文献

Colchicine has been a very useful diagnostic tool to determineif a particular developmental process requires cell divisionor microtubules; however, it produces certain side effects whichmay limit its usefulness. Low concentrations depolarize cellenlargement and higher concentrations actually inhibit cellenlargement; the threshold concentration varies depending onthe tissue. 0.2% (w/w) inhibits solute uptake and respirationin corn seedling roots. Higher concentrations also inhibit DNAand ethylene synthesis. Although ethylene and colchicine cause a similar swelling inthe elongation zone of roots, colchicine does not promote ediylenesynthesis and may even inhibit. In addition, the ethylene antagonist,CO2, does not prevent the depolarization of cell enlargementin corn roots. Thus colchicine induction of swelling in cornroots is not mediated by ethylene. The interaction of kinetin with IAA and GA3 on the elongationof hypocotyl sections of Cucumis sativus L. cv. National Picklingwas studied. Kinetin in the concentration range of 10–7M to 10–4 M markedly inhibited IAA-induced elongation,while in a lower range from 10–10 M to 10–8 M, itsynergistically enhanced IAA-induced elongation. Kinetin alonein this range had no effect. A 5-to 15-min pulse treatment seemsenough to induce the maximum effect for both inhibition andpromotion. Since the magnitude of the maximum inhibition dependedon the concentration and not on the duration of treatment, thereaction in the cell caused by kinetin seemed to be completedwithin a short period. Washing of the sections with distilledwater after kinetin treatment (30 min) did not significantlyeliminate the kinetin effect. This probably indicates that thebinding of kinetin molecules to a supposed acceptor is not reversible.Interaction of kinetin with GA3 in their pretreatment effectson IAA-induced elongation shows that in the inhibitory concentrationrange, the kinetin effect was partly overcome by GA3, and thatin the promotive range, the magnitude of the enhancement wasdetermined by kinetin regardless of the presence of GA3.
