Koziol, M. J., Shelvey, J. D. and Beedham, B. 1986 Uptake ofS'v anions and the induction of gaseous emissions in leaf segmentsof SO2-sensitiveand resistant genotypes of Lolium perenne L.—J.exp.Bot. 37: 556–571. At pH 3?0 and over a range of concentrations from 5 to 100 molm–3 NaHSO2 the SO2 genotype (S23) took up less and showed a lower metabolic thresholdfor the induction of gaseous emissions than the SO2 genotype(BR). Emission of H2S from leaf segments of S23 was inducedat an estimated sulphite concentration of 605 ng g–1 fr.wt. compared with 932 ng g–1 fr. wt. in BR. Although theuptake of sulphite by leaf segments incubated on NaHSO3 solutionsat pH 7?0 was similar to that of leaf segments incubated atpH 3?0, very little ethane, ethylene or H2S were emitted. Afree radical scavenging mechanism is proposed to explain theseobservations. At pH 6?0 and concentrations of NaHSO3 equivalent to low tomoderate exposure concentrations of gaseous SO2 BR took up moresulphite than S23. Uptake in both genotypes was partially inhibitedby CCCP and by pre-loading the leaf segments with sulphate,indicating an active uptake mechanism transporting both sulphateand sulphite. The purpose of this study was to examine the effect of prolongedbed rest (BR) on the peak isometric force(Po) and unloaded shorteningvelocity (Vo)of single Ca2+-activated musclefibers. Soleus muscle biopsies were obtained from eight adult malesbefore and after 17 days of 6° head-down BR. Chemicallypermeabilized single fiber segments were mounted between a forcetransducer and position motor, activated with saturating levels ofCa2+, and subjected to slacklength steps. Vowas determined by plotting the time for force redevelopment vs. theslack step distance. Gel electrophoresis revealed that 96% of the pre-and 87% of the post-BR fibers studied expressed only the slow type Imyosin heavy chain isoform. Fibers with diameter >100 µm made uponly 14% of this post-BR type I population compared with 33% of thepre-BR type I population. Consequently, the post-BR type I fibers(n = 147) were, on average, 5%smaller in diameter than the pre-BR type I fibers(n = 218) and produced 13% lessabsolute Po. BR had no overalleffect on Po per fibercross-sectional area(Po/CSA), even though halfof the subjects displayed a decline of 9-12% inPo/CSA after BR. Type Ifiber Voincreased by an average of 34% with BR. Although the ratio of myosinlight chain 3 to myosin light chain 2 also rose with BR, there was nocorrelation between this ratio andVo for either thepre- or post-BR fibers. In separate fibers obtained from the originalbiopsies, quantitative electron microscopy revealed a 20-24%decrease in thin filament density, with no change in thick filamentdensity. These results raise the possibility that alterations in thegeometric relationships between thin and thick filaments may be atleast partially responsible for the elevatedVo of the post-BRtype I fibers.

This paper elucidates the effects of chloroflurenol (morphactin,IT 3456) treatment on the kinetics of 14C-IAA transport throughZea mays L. (cv. Orla-266) coleoptile segments. Maximum inhibitionof transport was achieved when chloroflurenol remained in contactwith the tissue segments for at least 20 min or more. The treatment,without materially affecting the 14C-IAA-transport polarity,or uptake, significantly reduced the velocity from 20.5 mm h–1to 8.79 mm h–1 and also the intensity from 919 (ct/min)h–1 to 413 (ct/min) h–1.  相似文献

The transport of [2-14C]IAA and its derived products has beeninvestigated in segments of the leaf sheath base of Echinochloacolonum. Segments orientated vertically in the normal uprightposition transport 14C basipetally at rates approximately 10times higher that acropetally. In horizontal segments the basipetalrates are lower, being only 1–4 times higher than acropetalrates. Segments orientated vertically in the inverted positiontransport 14C basipetally at approximately 1/10 of the acropetalrate. 14C in the receiver discs from vertically orientated segmentswas found to be present predominantly as [2-14C]IAA and itsderived metabolite. The levels of each of these compounds from upright and invertedvertical segments followed the same pattern as that of theirtotal 14C content.  相似文献

The movement of kinetin has been investigated in roots of Zeaseedlings, grown in the dark, using kinetin-8-14C. Segments (10 and 11 mm) with and without an apex were used andthe following results obtained: The transport of kinetin is polarized basipetally. Kinetin-8-14C is transported in roots without vascular tissue,but the amount transported is always lower than in roots withvascular tissue. Thin-layer chromatography showed that the molecule of kinetinis metabolized in the root segments, adenine being one of theproducts.  相似文献

Experiments were done to determine if the spontaneous recoveryof non-growing segments of corn root (Zea mays L.) from excisioninjury is dependent on auxin. Washing the segments with 5 runindoleacetic acid (IAA) for 2 to 4 hours gave a small but significantincrease in K+ (86Rb) influx, used here as a parameter reflectingrecovery of electrogenie H+-efflux pumping. This promotive effectwas obtained only after an hour of washing, and was sustainedby 100 nm gibberellic acid (GA3). Any early responses to auxinwere obscured by an adverse reaction of the root cells to externalIAA which resulted in a transitory inhibition of H+ pumpingand K+ influx. Pretreatment of excised root tips with 10 µM IAA in thegrinding medium protected a plasmalemma-enriched fraction ofthe microsomes during isolation, giving increased uncoupler-sensitiveATPase activity. Non-growing root tissue thus shows three responses to auxin:an adverse reaction at the outer surface of the plasmalemmawhich blocks H+ pumping; a protective or restorative effecton the H+-ATPase; an increased capacity for K+ influx duringthe developmental phase of washing, which is augmented by thepresence of GA3. The calabrese cultivar Brassica oleracea var. italica cv. GreenComet was used in a study of the effects of exogenous hormoneson the growth and differentiation of seedling organs in vitro.Four types of explants were tested: hypocotyl segments, rootsegments, primary leaf discs and cotyledon discs. These explantswere incubated on media containing factorial combinations ofBAP x IBA, BAP x NAA, KN x IBA and KN x NAA (all at 0, 0.1,10 and 10.0mg l–1). Hypocotyls were the most regenerativeexplants; shoot production was favoured by cytokinin: auxinratios greater than one and was decreased by IBA at 10 mg l–1when callus was produced. Shoot formation from root explantsoccurred either in the absence of hormones or with low concentrations;no shoot was produced when any hormone was present at 10 mgl–1. In contrast, shoot production from primary leaf diseswas favoured by high concentrations of both auxin and cytokininwith the combination of BAP and IBA the most effective. Shootproduction from cotyledon discs was sporadic with no consistentresponse on any auxin/cytokinin combination. After further experimentson the optimization of hormone concentration, the followingcombinations were chosen as allowing reliable regeneration:0.1 mg l–1 BAP+0.1mg l–1 IBA for hypocotyl segments,0.075 mg l–1 KN +0.025 mg l–1 IBA for root segments,and 5.0 mg l–1 BAP+5.0 mg l–1 IBA for leaf discs. The effect of brassinolide (BR) on the reproductive developmentof higher plants was investigated using three different experimentalsystem. Direct application of BR to the staminate inflorescenceof Luffa cylindrica induced bisexual and pistillate flowers,and eventually caused the inflorescence to develop into a shootthat was similar to the main shoot. However, the activity ofBR on this sex modification was inferior to that of N6-benzylaminopurine. BR slightly hastened the flowering of nonvernalized radish plants,Raphanus satiuus cv. Miyashige-sofuto, whereas no such effectwas observed in vernalized plants. GA3 promoted flowering bothin nonvernalized and vernalized plants. Pretreatment of 2?0 mm segments of etiolated zucchini (Cucurbitapepo L.) hypocotyl with cycloheximide (CH) or 2-(4-methyl-2,6-dinitroanilino)-N-methylpropionamide(MDMP) eliminated the stimulation by N-1-naphthylphthalamicacid (NPA) of net uptake of [1-14C]indol-3yl-acetic acid ([1-14C]IAA),but had relatively little effect on the net uptake of IAA inthe absence of NPA. The efflux of [1-14C]IAA from preloadedsegments was not substantially affected by inhibitor pretreatmentin the absence of NPA, but CH pretreatment significantly inhibitedthe reduction of efflux caused by NPA. Pretreatment with CHor MDMP did not affect net uptake by segments of the pH probe[2-14C]5,5-dimethyl-oxazolidine-2,4-dione ([2-14C]DMO), or thenet uptake of [14C]-labelled 3-O-methylglucose ([14C]3-0-MeGlu),suggesting that neither inhibitor affected intracellular pHor the general function of proton symporters in the plasma membrane.Both compounds reduced the incorporation of label from [35S]methionineinto trichloroacetic acid (TCA)-insoluble fractions of zucchinitissue, confirming their inhibitory effect on protein synthesis. The steady-state association of [3H]IAA with microsomal vesiclesprepared from zucchini hypocotyl tissue was enhanced by theinclusion of NPA in the uptake medium. The stimulation by NPAof [3H]IAA association with microsomes was substantially reducedwhen the tissue was pretreated with CH. However, CH pretreatmentdid not affect the level of high affinity NPA binding to themembranes indicating that treatments did not result in lossof NPA receptors. It is suggested that the auxin transport site on the effluxcarrier system and the receptor site for NPA may reside on separateproteins linked by a third, rapidly turned-over, transducingprotein. Ethylene production by IAA-treated mung bean hypocotyl segmentsunder various oxygen levels in the ambient atmosphere was examined.Rate of ethylene production was dependent upon oxygen levels,and gave a sigmoidal curve against oxygen levels. Tissue segmentspreincubated with IAA in low oxygen levels (1–10% O2 inN2) produced ethylene without a lag period at a rate higherthan that by control tissue segments preincubated in air, whenthey were exposed to a high oxygen level (air, 21% O2). Theeffect of cycloheximide on tissue segments transferred froma low oxygen level to air was not much different from that onethylene production by control tissue segments previously incubatedin air. Incorporation of U-14C-leucine into the protein fractionby tissue segments placed in nitrogen was negligible, but thatin 2% oxygen was 10 to 14% of that in air. It was concluded that oxygen was an essential factor for boththe induction process of the ethylene producing system and thesynthesis of ethylene, and that although synthesis of ethyleneis dependent upon oxygen levels, formation of the ethylene producingsystem proceeded even under low oxygen levels. To obtain information about the biosynthesis of brassinosteroids(BRs) in tomato shoots, we examined endogenous BRs by gas chromatography-massspectrometry. We identified two C28 BRs, namely, castasteroneand 6-deoxocastasterone, and a C27 BR, 28-norcastasterone. Callose formation in phloem tissue was promoted by 0.5% eosinin 1 cm segments taken from first year Willow (Salix viminalisL.) shoots and by localized heat treatments in intact secondyear shoots. The eosin did not cause any change in the incorporationof 14C from labelled sucrose into an insoluble fraction butless of the label: from UDP-glucose labelled in the glucosemoiety was incorporated when eosin was present. Localized heattreatments of intact second-year tissue which was translocating[14C]sucrose failed to cause an increase in the amount of labelin an insoluble fraction in the zone where callose formationwas stimulated. The results indicate that the callose formedrapidly in response to stress and wounding is not derived directlyfrom translocated sucrose. An alternative possible origin, fromsugar nucleotides released from ruptured plastids, is discussed.  相似文献

Externally supplied [3H]myo-inositol and [14C]glucose were incorporatedin cell-wall fractions of segments of etiolated squash hypocotyl.The extent of incorporation of [14C]glucose into cell-wall fractionswas very much greater than that of [3H]myo-inositol. Radioactivityfrom [14C]-glucose was effectively incorporated into hemicelluloseB and cellulose fractions and was incorporated uniformly intohexose, pentose and uronic acid residues, but radioactivityfrom [3H]myo-inositol was incorporated predominantly into uronicacid and pentose residues in the pectin and hemicellulose Bfractions. Exogenously applied ABA significantly suppressed the elongationof segments of squash hypocotyl and the incorporation of radioactivityfrom [l4C]glucose and [3H]myo-inositol into the segments. Furthermore,ABA significantly inhibited the distribution of incorporatedradioactivity from [14C]glucose into the cellulose fraction,but did not affect distribution into the pectic fraction. Bycontrast, ABA only slightly inhibited the distribution of theincorporated radioactivity from [3H]myo-inositol into the pecticfraction. These results suggest that most of the cell-wall polysaccharidesin segments of squash hypocotyl are synthesized via the UDP-sugarpathway, and that ABA significantly inhibits the synthesis ofcellulose but not the synthesis of pectic polysaccharides whenABA suppresses the elongation of the segments. 以佛手山药块茎、叶片、茎段为外植体, 探讨了其组织培养技术。结果表明:块茎培养以暗培养MS+6-BA1.0 mg·L-1+NAA0 1 mg·L-1效果较好;叶片诱导的适宜培养基为MS+ 6-BA0.5~1.0 mg·L-1+NAA2.0 mg·L-1, 暗培养;茎段培养都是光培养,无节茎段以MS+6-BA1.0 mg·L-1+NAA0.5 mg·L-1较好;带节茎段的初代培养则以MS+6-BA0.5~1.0 mg·L-1+NAA0.1 mg·L-1效果较好,继代增殖培养基为MS+6-BA0.5 mg·L-1+NAA0.1 mg·L-1,生根培养基为1/2MS +NAA0.5 mg·L-1。  相似文献

Xyloglucan nonasaccharide (XG9) is recognized as an inhibitorof 2,4-D-induced long-term growth of segments of pea stems.In the presence of 10–5 M 2,4-D, inhibition by 10–9M XG9 of elongation of third internode segments of pea seedlingswas detected within 2 h after the start of incubation, in someexperiments. Analysis by double-reciprocal (Lineweaver-Burk)plots of elongation in the presence of various concentrationsof 2,4-D, with or without XG9, gave parallel lines, indicatingthat XG9 inhibited 2,4-D-induced elongation in an uncompetitivemanner. XG9 did not influence the 2,4-D-induced cell wall loosening.Thus, XG9 does not fulfill the proposed definition of an "antiauxin". XG9 at 10–11 to 10–6 M did not influence IAA-inducedelongation of segments from pea third internodes, azuki beanepicotyls, cucumber hypocotyls, or oat coleoptiles. Inhibitionof IAA-induced elongation by XG9 was not observed even whenthe segments from pea or azuki bean were abraded. Furthermore,fucosyl-lactose at 10–11 to 10–4 M did not affectthe IAA-induced elongation of segments of pea internodes orof azuki bean epicotyls. XG9 may be incapable of inhibitingthe IAA-induced cell elongation (especially in oat) or, alternatively,the endogenous levels of XG9 may be so high that exogenouslyapplied XG9 has no inhibitory effect on IAA-induced elongation. The patterns of uptake of indol-3yl-acetic acid (IAA-2-14C)by etiolated stem segments of varying lengths have been examined,employing tissues excised from (a) the first and third internodesof Pisum sativum, (b) the top and base of the hypocotyl of Gossypiumhirsutum, and (c) the mesocotyl of Avena sativa. For all species,concentrations (10–5–10–3 M) and times upto 24 h, there is a steady accumulation of radioactivity inthe segments. For equal volumes of tissue uptake is inverselycorrelated with segment length but for extending tissues theinitial enhanced extension growth is independent of length;that is there is no direct linkage between the rate of extensionand auxin content. Comparisons between segments with free andsealed ends established that over 24 h some 57–73 percent of the IAA enters via the cut surfaces. Initially, thepercentage is greater; expressed as a rate per unit of surfacethe differences between cut and epidermal surfaces can reach28-fold. The rate of entry through the epidermal surface islinearly proportional to the external concentration but thisdoes not hold for cut surfaces. The addition of streptomycin,synthalin, cetyltrimethylammoniumbromide (CTAB), and chitosanto the external medium does not promote uptake of IAA by Pisumsegments; indeed synthalin is markedly inhibitory. With Gossypiumsynthalin causes little inhibition. Larger depressive effectswere induced for entry via the cut surfaces. On entry the IAAis rapidly metabolized and the rate of conversion is higherfor segments with sealed ends. These findings are discussedin relation to (a) differences in the mechanisms determiningthe uptake of IAA and other auxins, (b) cell extension and thedistribution of auxin in the tissues.  相似文献

Mode of action of FC was compared with that of auxin in differentexperimental systems and the following results were obtained.
  1. FC, as well as auxin, primarily induced elongation of the epidermisof pea epicotyl segments, but it also promoted elongation ofthe inner tissue, as judged by its action in split stem tests,elongation of hollow-cylinder segments and elongation of unpeeledand peeled segments.
  2. FC decreased the minimum stress relaxationtime (T0) and increasedthe extensibility (mm/gr) of the epidermalcell wall of peaepicotyl segments, as did auxin.
  3. FC failedto induce expansion growth of Jerusalem artichoketuber sliceswhen given alone or in combination with kinetinor gibberellicacid.
  4. FC at concentrations lower than 10–6 M, when givenwithauxin at concentrations lower than 0.03 mg/liter, promotedelongationof Avena coleoptile segments in an additive manner,to achievethe maximum elongation at higher concentrations.
  5. An antiauxin, 2,4,6-trichlorophenoxyacetic acid, inhibitedtheelongation of Avena coleoptile segments due to auxin butnotthat due to FC.
  6. Nojirimycin, an inhibitor of ß-glycosidases,inhibitedelongation of pea internode segments due not onlyto auxin butalso to FC.
  7. At concentrations more than 10–5MFC promoted root elongationof intact lettuce seedlings, whichwas inhibited by exogenousauxin.
From these results it is concluded that FC and auxin have acommon mechanism, which may involve hydrogen ion extrusion,leading to cell wall loosening and thus cell elongation. Thisgrowth is limited to the extent that the cells are capable ofelongating in response to hydrogen ions. Otherwise there isa definite difference in the mode of actions between FC andauxin, including the nature of cellular receptors for thesetwo compounds. (Received August 29, 1974; )  相似文献

The movement and metabolism of kinetin-8-14C in the mesocotyland coleoptile of Zea mays have been investigated. The segments (10 mm) of coleoptile with and without an apexwere used. The presence of apex decreases the quantity of countswhich move basipetally in the segments. The quantity transported acropetally in the segments (apex incontact with receiver blocks) was about three times higher thanthat transported basipetally (apex in contact with donor blocks). Using 10 mm coleoptile segments excised I mm below the apex,there was no appreciable difference between acropetal and basipetalmovement. The transport of kinetin in the mesocotyl was studied with andwithout vascular tissue. The quantity transported in segmentswithout vascular tissue was always lower than in the segmentswith vascular tissue. The presence of the node between the coleoptile and mesocotyldoes not disturb the movement of kinetin in the basipetal position(node in contact with donor blocks). Thin layer chromatography of plant extracts after a 48-hourtransport period revealed several radioactive substances whichappeared to be adenine with Rf 0.5 and other components appearedat Rf 0.6.  相似文献

Soleus musclefibers from the rat display a reduction in peak power andCa2+ sensitivity after hindlimbsuspension. To examine human responses to non-weight bearing, weobtained soleus biopsies from eight adult men before and immediatelyafter 17 days of bed rest (BR). Single chemically skinned fibers weremounted between a force transducer and a servo-controlled positionmotor and activated with maximal (isotonic properties) and/orsubmaximal (Ca2+ sensitivity)levels of free Ca2+. Gelelectrophoresis indicated that all pre- and post-BR fibers expressedtype I myosin heavy chain. Post-BR fibers obtained from one subjectdisplayed increases in peak power andCa2+ sensitivity. In contrast,post-BR fibers obtained from the seven remaining subjects showed anaverage 11% reduction in peak power (P < 0.05), with each individualdisplaying a 7-27% reduction in this variable. Post-BR fibersfrom these subjects were smaller in diameter and produced 21% lessforce at the shortening velocity associated with peak power. However,the shortening velocity at peak power output was elevated 13% in thepost-BR fibers, which partially compensated for their lower force.Post-BR fibers from these same seven subjects also displayed a reducedsensitivity to free Ca2+(P < 0.05). These results indicatethat the reduced functional capacity of human lower limb extensormuscles after BR may be in part caused by alterations in thecross-bridge mechanisms of contraction.

14C from indol-3-yl-(acetic acid-2-14C) (IAA-14C) was transportedin a weak but definitely polar manner through segments of youngand matured regions of pea roots. Greater quantities of 14C-labelledmaterial moved acropetally than basipetally. Up to 70 per centof radioactivity originally present in donor agar blocks wastaken up by the root segments, but only approximately 2 to 3per cent of this emerged into the receiver agar blocks. Anydifferences in uptake, transport, or binding of auxin were veryslight in the three regions of root studied. The IAA-14C wasmetabolized during passage through the root segments, yieldingtwo principal radioactive products. The identities of thesewere not determined, but they appeared to have auxin activityand may be formed spontaneously, but more slowly, in solutionsof IAA-14C. IAA-14C was transported into receiver blocks morereadily than its radioactive derivatives.  相似文献
