Cyanobacteria have two protochlorophyllide (Pchlide) reductasescatalyzing the conversion of Pchlide to chloro-phyllide, a keystep in the biosynthetic pathway of chlorophylls (Chls); a light-dependent(LPOR) and a light-independent (DPOR) reductase. We found anopen reading frame (ORF322) in a 2,131-bp EcoRI fragment fromthe genomic DNA of the cyanobacterium Plectonema boryanum. Becausethe deduced amino acid sequence showed a high similarity tothose of various plant LPORs and the LPOR activity was detectedin the soluble fraction of Esche-richia coli cells over-expressingthe ORF322 protein, ORF322 was defined as the por gene encodingLPOR in P. boryanum. A por-disrupted mutant, YFP12, was isolatedby targeted mutagenesiss to investigate the physiological importanceof LPOR. YFP12 grew as well as wild type under low light conditions(10-25 µE m–2 S–1). However, its growth wassignificantly retarded as a result of a significant decreasein its Chl content under higher light conditions (85-130 µEm–2 s–1). Furthermore, YFP12 stopped growing andsuffered from photobleaching under the highest light intensity(170 µE m–2 s–1). In contrast, a chlL-dis-rupted(DPOR-less) mutant YFC2 grew as well as wild type irrespectiveof light intensity. From these phenotypic characteristics, weconcluded that, although both LPOR and DPOR contribute to Chlsynthesis in the cells growing in the light, the extent of thecontribution by LPOR increases with increasing light intensity;without it, the cells are unable to grow under light intensitiesof more than 130 µ Em–2s-. 1) With Chlorella ellipsoidea cells, in the presence of 5x10–6M DSPD, or in its absence, the amounts of 14CO2 incorporatedin P-esters, serine-plus-glycine and alanine were larger underred light than under blue light, whereas blue light specificallyincreased 14CO2-incorporation in aspartate, glutamate, malateand fumarate (blue light effect). The amount of total 14C fixedunder blue or red light was greatly decreased by the additionof DSPD. When the concentration of DSPD was raised to 5x10–4M, practically no radioactivity was found, under blue or redlight, in aspartate, glutamate and fumarate. Radioactivity inalanine was greatly increased. Effects of higher concentrationof DSPD are explained as due to the inhibition of PEP carboxylaseactivity in Chlorella cells. 2) The percentage incorporation of 14C into aspartate and theother compounds mentioned above, under near infra-red illuminationwas significantly smaller than that under blue light and wasalmost equal to that under red light. Rhodotorula minuta cells, which have only traces of carotenoidswhen grown in the dark, started carotenoid production with theonset of illumination and the amount increased almost linearlyuntil 70 hr then remained constant thereafter when incubationwas continued under illumination, with the number of cells continuingto increase. The rate of carotenoid production [Vc (µgg–1 hr–1)] depended on the intensity of light [I(ergcm–2 sec–1)], with the relationship of Vc=0.74 logI–1.46. The final carotenoid content [C(µg g–1)]of cells incubated under continuous light was also controlledby the light intensity [I], with the relationship of C=52 logI–81. Control of carotenoid production by light occursas a two-phase process consisting of a temperatureindependentphotochemical reaction and light-independent biochemical reactions. A comparison of photosynthesis-irradiance response curves (PEresponse curves) obtained through fast repetition rate (FRR)fluorometry and radiocarbon (14C) tracer method was made inthe chlorophyte, Dunaliella tertiolecta, grown under differentirradiance conditions. In FRR-based PE response curveexperiments, actinic light provided by white light-emittingdiodes (LEDs) was increased gradually from 0 to 1500 µmolquanta m–2 s–1 and the rate of photosyntheticelectron transport was determined at each light level. Short-termexperiments (20 min) of 14C-based PE response curvewere carried out with an improved photosynthetron, which containswhite LEDs as the light source. Irrespective of growth irradiance,the ratios of FRR to 14C-based initial slopes were almost uniform.The ratios of FRR- to 14C-based maximum rates were 25–36%higher than those of FRR- to 14C-based initial slopes. The relationshipbetween electron transport and carbon assimilation was non-linearwith increasing discrepancy towards high actinic light. Thisnon-linear relationship between FRR- and 14C-based estimatesis primarily due to the effect of physiological processes stimulatedat high levels of light, such as cyclic electron flow and theMehler reaction. The results of this study indicate that theFRR fluorometry can be used as a good indicator of photosyntheticrates from low to middle light levels, but becomes increasinglyquestionable as the maximum photosynthetic rate is approached.The degree to which this relationship is further affected bynutrient-status warrants investigation.  相似文献

Changes in the activity of cytochrome c oxidase (EC 1.9.3.1 [EC] ,Cyt-oxidase) in response to growth conditions were studied withthe cyanophyte Synechocystis PCC 6714 in relation to changesin PSI abundance induced by light regime for photosynthesis.The activity was determined with the Vmax of mammalian cytochromec oxidation by isolated membranes. The activity of glucose-6-phosphate(G-6-P):NADP+ oxidoreductase (EC 1.1.1.49 [EC] ) was also determinedsupplementarily. Cyt-oxidase activity was enhanced by glucoseadded to the medium even when cell growth maintained mainlyby oxygenic photosynthesis. G-6-P:NADP+ oxidoreductase was alsoactivated by glucose. The enhanced level of Cyt-oxidase washigher under PSII light, which causes high PSI abundance, thanthat under PSI light, which causes low PSI abundance. The levelwas intermediate under hetetrotrophic conditions. Although theactivity level was low in cells grown under autotrophic conditions,the level was again lower in cells grown under PSI light thanunder PSII light. The change of Cyt-oxidase activity in responseto light regime occurred in the same direction as that for thevariation of PSI abundance. A comparison of the activity and properties of the enzyme phosphoenolpyruvatecarboxylase (PEPC) was made for plants of Sedum telephium L.grown under low (70 µmol m–2 s–1) or high(500µmol m–2 s–1) PPFD and subjected to varyingdegrees of water stress. Under well-watered conditions onlyplants grown under high PPFD accumulated titratable acidityovernight and the extractable activity of PEPC was almost 2-foldhigher in these plants than in plants grown under low PPFD.Increasing drought stress resulted in a substantial increasein the activity of PEPC extracted both during the light anddark periods and a decrease in the sensitivity to inhibitionby malic acid. The magnitude of these changes was determinedby the severity and duration of drought and by light intensity.A comparison of the kinetic properties of PEPC from severelydroughted plants revealed that plants droughted under high PPFDhad a lower Km for PEP than plants under low PPFD. Additionof 2·0 mol m–3 malate resulted in an increase inthe Km for PEP, with plants draughted under low PPFD havinga significantly higher Km in the presence of malic acid comparedto those under high PPFD. Response to the activator glc-6-P,which lowered the Km for PEP, also varied between plants grownunder the two light regimes. Under well-watered conditions PEPCextracted from plants under high PPFD was more sensitive toactivation by glc-6-P than those under low PPFD. After the severedrought treatment, however, the Km for PEP in the presence ofglc-6-P was similar for enzyme extracted from plants grown underboth light regimes. Soluble sugars and starch were depletedovernight and were both possible sources of substrate for PEPC.With increasing drought, however, the depletion of starch relativeto soluble sugars increased under both light regimes. The propertiesof PEPC and the characteristics of carbohydrate accumulation/depletionare discussed in relation to the regulation of CAM in S. telephiumgrown under different light and watering regimes. When seedlings of Cuscuta japonica were grown with Vigna radiata(the host plant) in a flower pot for 6 d under white light andthen irradiated with far-red or blue light (ca. 6 µmolphotons m–2 s–1), the seedlings parasitized V. radiata.However, no parasitism of the seedlings was observed under redor white light or in darkness. The parasitic behavior of seedlingsof C. japonica was observed even if an acrylic rod was usedas a substitute for the host plant. Upon incubation under far-redlight, the seedling twined tightly around the rod and developedhaustoria towards it. Haustoria also developed when apical andsubapical regions of seedlings were held between two glass platesthat were about 0.7 mm apart and were irradiated with far-redlight. However, no haustoria were induced by either the holdor irradiation alone. These results indicate that parasitismof Cuscuta japonica is controlled by the cooperative effectsof two physical signals, far-red light and appropriate tactilepressure. Chlorella cells incubated in the dark longer than 12 hr showedpronounced blue light-induced 14CO2 fixation into aspartate,glutamate, malate and fumarate (blue light effect), whereasthose kept under continuous light showed only a slight bluelight effect, if any. 2) During dark incubation of Chlorellacells, phosphoenolpyruvate carboxylase activity and the capacityfor dark 14CO2 fixation decreased significantly, whereas ribulose-1,5-diphosphatecarboxylase activity and the capacity for photosynthetic 14CO2fixation (measured under illumination of white light at a highlight intensity) did not decrease. 3) In cells preincubatedin the dark, intracellular levels of phosphoenolpyruvate and3-phosphoglycerate determined during illumination with bluelight were practically equal to levels determined during illuminationwith red light. 4) The blue light effect was not observed incells incubated widi chloramphenicol, indicating that blue light-inducedprotein synthesis is involved in the mechanism of the effect. [14C]-assimilation rates were measured on cultures of two unicellulargreen algae (Chlamydomonas sp. and Oocystis sp.) as a functionof light intensities (saturation curves), under steady lightand also under rapidly alternating high and low light intensities.Assimilation rates vary according to the frequency of the intermittentlight regime and it falls under two categories: (1) at 0.1 and0.2 Hz, the assimilation rate is equal to the average of therates observed at high and low light intensities under steadylight, and (2) under 1.0, 1.6 and 10 Hz the assimilation rateis equal to the rate observed under a mean steady irradiance.Moreover, the range of assimilation rates at a given frequencydepends on the difference between the high and the low intensities.Batch cultures of Oocystis sp. have been grown under intermittentlight of 0.1, 1.0 and 10.0 Hz (same mean intensity). Growthrate under intermittent light of 0.1 Hz is –40% lowerthan the control under steady light. Photosynthetic potential(PBmax)and efficiency () change with the growth stages of thecultures. At the end of the logarithmic growth phase, both photosyntheticparameters are maximum at 1.0 Hz and minimum at 0.1 Hz. Averagecell concentrations of chlorophyll a increase as the frequencyof the light regime decreases. During the log phase, concentrationof carotenoids relative to chlorophyll a increases at 1.0 Hz,decreases at 0.1 Hz, and remains constant at 10.0 Hz. Chlorella pyrenoidosa can utilize sodium acetate as a carbonsource for growth in the light. Growth proceeds under aerobicconditions both in the presence and in the absence of carbondioxide, but under anaerobic conditions only in its presence.The assimilation of acetate does not result from oxidation tocarbon dioxide followed by photosynthetic fixation because theproducts of 14C-acetate assimilation are different from theproducts of 14CO2 fixation in the presence of unlabelled acetate. In aerobic conditions 10-6 M DCMU induces a pattern of acetateassimilation in the light similar to that in the dark. Thus,in the presence of DCMU in the light, less acetate carbon isincorporated into cells, particularly into lipids, polysaccharide,and protein, and more is released as carbon dioxide than inits absence. The effect of 4 x 10-3 M MFA on acetate assimilationin the presence of 10-6 M DCMU is the same in light and dark.Acetate assimilation is unaffected by desaspidine and sodiumbisulphite. The mean generation time of C. pyrenoidosa growing on acetatein the light under aerobic conditions is 20 hours. When 10-5M DCMU is added the mean generation time is 60 hours, the sameas that for Chlorella growing on acetate in the dark. The activityof the enzymes of the glyoxylate cycle, isocitrate lyase (E.C.4.1.3.1.)and malate synthetase (E.C.4.1.3.2.) is repressed in the light,but activity of both enzymes increases markedly when DCMU isadded.  相似文献

Changes in the PSII fluorescence upon shift of light qualitywere studied with the red alga Porphyridium cruentum IAM R-1and supplementarily with P. cruentum ATCC 50161, the cyanophytesSynechocystis spp. PCC6714 and PCC6803 and Synechococcus sp.NIBB1071. When Porphyridium cruentum grown under a weak redlight (PSI light) preferentially absorbed by Chl a was illuminatedwith a weak orange light (PSII light) mainly absorbed by phycobilisomes(PBS), a change of PSII fluorescence at room temperature wasinduced. The ratio of Fvm (Fm— Fo) to Fm was reduced rapidlyaccompanying the increase in Fo (T1/2 ca. 3 min). The effectsof DCMU and 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinoneindicated that the fluorescence change is induced when plastoquinonepool is highly reduced. The fluorescence change after a shortPSII light illumination was reversible; it rapidly recoveredin the dark (T 1/2 ca. 3 min). The reversibility was graduallyreduced and disappeared after 40 h under PSII light accompanyingdecrease in PSII activity per PBS down to almost 50%. Sincethe pattern of the fluorescence change resembles that observablewhen PSII is photoinactivated, PSII light probably induces thephotoinactivation of PSII, possibly reversibly at first andirreversibly after prolonged illumination. Such a rapid fluorescencechange was insignificant in Synechocystis sp. either PCC6714or PCC6803. Only a slow and small decrease in Fvm/Fm level appearedafter prolonged PSII light illumination (the reduction of PSIIactivity per PBS was around 20%). In Porphyridium, shift fromPSII light to PSI light caused a rapid and chloramphenicol-sensitiveFvm/Fm elevation during the first 10 h while the increase inPSH activity per PBS was only 10% of that before the light shift.Then, a gradual elevation followed up to the level at the steadystate under PSI light. A similar rapid increase in Fvm/Fm wasobserved with Synechocystis PCC6714, in which the synthesisof PSII is not regulated, suggesting that a rapid increase inFvm/Fm does not reflect the acceleration of the synthesis ofPSII. Results were interpreted as that (1) PSII light causesphotoinactivation of PSII. Such a photoinactivation is markedin Prophyridium cells grown under PSI light. (2) In Porphyridium,changes in the abundance of PSII upon shift of light qualityare largely attributed to the photoinactivation of this type. The Cyt f and P700 contents in leaves of three Sorghum, varietieswere measured, in relation to their carbon assimilation, underdifferent light intensities during growth. At the maximum irradiationused (1,800 µE m–2 s–1) the ratio of P700to Cyt f was close to unity, whereas under low irradiation (450µE m–2 s–1) the ratio of P700 to Cyt f rangedfrom two to three. A strikingly positive correlation existedbetween the P700 contents of the leaves and their rates of carbondioxide fixation, dry matter production and Cyt f contents,only when the plants were grown under high light intensities.The P700 content of the leaves in plants grown under low irradiationwas unrelated to the contents of Cyt f. Thus, at a high lightintensity there is a close relationship between the Cyt f andP700 levels, but at low intensities the amounts of electroncarriers and the reaction centre are independent. Maximal l4CO2-fixation in spinach occurs in the middle of thepalisade mesophyll [Nishio et al. (1993) Plant Cell 5: 953],however, ninety percent of the blue and red light is attenuatedin the upper twenty percent of a spinach leaf [Cui et al. (1991)Plant Cell Environ. 14: 493]. In this report, we showed thatgreen light drives 14CO2-fixation deep within spinach leavescompared to red and blue light. Blue light caused fixation mainlyin the palisade mesophyll of the leaf, whereas red light drovefixation slightly deeper into the leaf than did blue light.14CO2-fixation measured under green light resulted in less fixationin the upper epidermal layer (guard cells) and upper most palisademesophyll compared to red and blue light, but led to more fixationdeeper in the leaf than that caused by either red or blue light.Saturating white, red, or green light resulted in similar maximal14CO2-fixation rates, whereas under the highest irradiance ofblue light given, carbon fixation was not saturated, but itasymptotically approached the maximal 14CO2-fixation rates attainedunder the other types of light. The distribution and partitioning of dry matter and photoassimilateof Lolium perenne was investigated under two light regimes providingphotosynthetically active radiation of 350 µmol m–2s–1 (low light treatment) or 1000 µmol m–2s–1 (high light treatment). Plants were grown at specificgrowth conditions in either soil or sand microcosm units tofollow the subsequent release of carbon into the rhizosphereand its consequent incorporation into the microbial biomass(soil system) or recovery as exudates (sand system). Loliun perenne L. (cv.S. 23) was grown on vermiculite in winterin a heated greenhouse for 8 weeks under factorial combinationsof two potassium regimes (nominally 6 parts/106 and 156 parts/106in Hewitt's solution) and three densities of artificially supplementedvisible radiation flux (36.1, 7.3, and 2.2 W m–2). Growthand potassium uptake were studied through the calculation ofvarious growth functions from fitted curves. There was little effect of potassium treatment but the experimentalmaterial responded markedly to light. Leaf-area ratio in thethree treatments showed extreme plasticity in increasing from2–3 x 10–2 through 6 x 10–2 to 8–9 x10–2 m2 g–1 as light intensity decreased. Correspondingdecreases in unit leaf rate, however, caused over-all reductionsin relative growth rate. Specific absorption rates for potassium (AK, dry-weight basis)were strongly reduced at the lower light intensities but alsodisplayed complex ontogenetic drifts. Values of the allometricconstant, k (the ratio of root and shoot relative growth rates),decreased from c. 0.7 at 36.1 W m–2 through c. 0.3 at7.3 W m–2 to a value not significantly different fromzero (P < 0.05) at 2.2 W m–2. In material grown under the two higher light intensities a constantinverse relationship was found between the mass ratio of rootand shoot and the corresponding activity ratio. The resultsconform to this model: Mass ratio = –0.001+45.0 (1/activityratio) where activity ratio is expressed as specific absorptionrate for potassium (in µg g root–1 h–1)/unitshoot rate (rate of increase of whole-plant dry weight per unitshoot dry weight, in mg g shoot–1 h–1). The rate of net photosynthesis (P) of whole plant stands oftomato (Lycopersicon esculentum Mill.), cucumber (Cucumis sativusL.) and sweet pepper (Capsicum annuum L.) was measured in sixlong-term experiments in large greenhouses under normal operatingconditions and CO2-concentrations between 200 and 1200 µmolmol-1. The objective was to quantify the responses to lightand carbon dioxide and to obtain data sets for testing simulationmodels. The method of measuring canopy photosynthesis involvedan accurate estimation of the greenhouse CO2 balance, usingnitrous oxide (N2O) as tracer gas to determine, on-line, theexchange rate between greenhouse and outside air. The estimatedrelative error in the observed P was about ± 10%, exceptthat higher relative errors could occur under particular conditions. A regression equation relating P to the photosynthetically activeradiation, the CO2 concentration and the leaf area index explained83-91% of the variance. The main canopy photosynthesis characteristicscalculated with the fitted regression equations were: canopyPmax 5-9 g m-2 h-1 CO2 uptake; ratio Pmax/LAI 1·5-3 gm-2 h-1; light compensation point 32-86 µmol s-1 m-2;light use efficiency (quantum yield) at low light 0·06-0·10µmol µmol-1 and CO2 compensation point 18-54 µmolmol-1. Cells, of Synechococcus sp. PCC 7002 showed a low oxidationlevel of P700 under a far-red light at 6 W m–2 which inducednearly complete oxidation of P700 in spinach leaves, and a strongerfar-red light was required to observe the oxidation of P700.DCMU did not affect the level of P700+2 but 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinoneinduced the oxidation of P700 under far-red light, indicatingthat the low oxidation level of P700 was due to the donationof electrons to P700+2 from the cytosolic respiratory donorsthrough the intersystem chain at the plastoquinone pool. Theelectron transfer from the cytosolic donors to the intersystemchain was inhibited by HgCl2 but not by antimycin A. The relationship between the rate of CO2 fixation and sucroseconcentration in the leaves of rice (Oryza sativa L.) grownat different light intensities was investigated. Maximum sucrosecontent coincided with maximum rates of CO2 fixation, achievedat a photon flux density of 1600 µmol m–2 s–1.The levels of sucrose and fructose 2,6-bisphosphate were alsocompared in the leaves under different light intensities. Fructose2,6-Msphosphate accumulated during growth at low light. Theactivity of fructose-6-phosphate 2-kinase was high in the leavesgrown at low light while that of fructose-2,6-bisphosphatasewas low. The activities of phosphoglucose isomerase and phospho-glucomutasewere slightly increased by growth at low light The activitiesof UDP glucose pyrophosphorylase were adversely affected invitro with increased concentrations of fructose 2,6-bisphosphatewhile those of sucrose phosphate synthase were moderately affected.Phosphoglucose isomerase and phosphoglucomutase were activatedby fructose 2,6-bisphosphate (8-0 mmol m–3) by 12-15%.The results suggested that low light intensities during growthresult in an accumulation of fructose 2,6-bisphosphate whichmodulates the key enzymes of sucrose biosynthesis thus regulatingcarbon flow under conditions of limited photosynthesis. Changes in intracellular levels of Chl a precursors were examinedin relation to changes in the PSI/PSII stoichiometry in thecyanophyte Synechocystis PCC 6714. Protochlorophyllide (Pchlide)accumulated markedly in cells with a low PSI/PSII stoichiometrygrown under light that is absorbed by Chl a (PSI light) whereasno accumulation occurred in cells with a high PSI/PSII stoichiometrygrown under light absorbed by phycobilisomes (PSII light). Levelsof Pchlide in cells grown under PSI light decreased rapidlyupon a shift to PSII light. The rapid decrease in Pchlide accompanieda transient increase in chlorophyllide a, indicating that reductionof Pchlide was enhanced by shift to PSII light. The action spectrumindicated that the Pchlide decrease upon the shift to PSII lightdepended on excitation of Pchlide, suggesting that the accumulationof Pchllide was due to limited excitation of Pchlide, so thatPchlide photoreduction, under PSI light. However, comparisonof levels of Pchlide and the photosystem complexes in wild-typePlectonema boryanum with those in a mutant that lacked the darkPchlide reductase (YFC 1004) indicated that dark reduction compensatedfor the limited photoreduction under PSI light. Similar compensationby dark reduction was confirmed with Synechocystis PCC 6714.In cultures of Synechocystis under conditions where Pchlidecould not be photoreduced, accumulation of Pchlide and low PSI/PSIIstoichiometry occurred only when cells were illuminated withlight that preferentially excited PSI. The results indicatethat the low PSI/PSII stoichiometry in cells grown under PSIlight is not a result of inefficient synthesis of Chl a witha reduced rate of Pchlide photoreduction. Etiolated Hordeum vulgare (barley) Plants were greened underwhite light or far-red (> 700 nm) light. Exposure to far-redlight inhibited chlorophyll synthesis (especially chlorophyllb) and the development of photosystem II which were seen whengreening took place under white light. Primary leaves were detachedand the labelling of their acyl lipids from [14C]acetate wasstudied under white light or far-red light illumination. Greeningwith far-red light caused a reduction in the radiolabellingof polyunsaturated fatty acids and diacylgalactosylglycerol.Total fatty acid labelling rates were unaffected. Phosphatidylethanolamine,which was normally poorly labelled, accounted for up to 15 percent of the total radioactivity in acyl moieties of lipids inleaves greened with far-red light. The results are discussedin connection with the role that acyl lipids may play in normalthylakoid structure and function.
