Ligands containing the 2-organochalcogenomethylpyridine motif with substituents in the 4- or 6-position of the pyridyl ring, R4,R6-pyCH2ER1 [R4 = R6 = H, ER1 = SMe (1), SeMe (2), SPh (6), SePh (7); R4 = Me, R6 = H, ER1 = SMe (3), SPh (8), SePh (9); R4 = H, R6 = Me, ER1 = SMe (4), SPh (10), SePh (11); R4 = H, R6 = Ph, ER1 = SMe (5), SPh (12), SePh (13)] are obtained on the reaction of R4,R6-pyMe with LiBun followed by R1EER1. On reaction with PdCl2(NCMe)2, the ligands with a 6-phenyl substituent form cyclopalladated species PdCl{6-(o-C6H4)pyCH2ER1-C,N,E} (5a, 12a, 13a) with the structure of 13a (ER1 = SePh) confirmed by X-ray crystallography; other ligands form complexes of stoichiometry PdCl2(R4,R6-pyCH2ER1). Complexes with R6 = H are monomeric with N,E-bidentate configurations, confirmed by structural analysis for 3a (R4 = Me, ER1 = SMe), 7a (R4 = H, ER1 = SePh) and 9a (R4 = Me, ER1 = SePh). Two of the 6-methyl substituted complexes examined by X-ray crystallography are oligomeric with trans-PdCl2(N,E) motifs and bridging ligands, trimeric [PdCl2(μ-6-MepyCH2SPh-N,S)]3 (10a) and dimeric [PdCl2(μ-6-MepyCH2SePh-N,Se)]2 (11a). This behaviour is attributed to avoidance of the Me···Cl interaction that would occur in the cis-bidentate configuration if the pyridyl plane had the same orientation with respect to the coordination plane as observed for 3a, 7a and 9a [dihedral angles 8.0(2)-16.8(2)°]. When examined as precatalysts for the Mizoroki-Heck reaction of n-butyl acrylate with aryl halides in N,N-dimethylacetamide at 120 °C, the complexes exhibit the anticipated trends in yield (ArI > ArBr > ArCl, higher yield for electron withdrawing substituents in 4-RC6H4Br and 4-RC6H4Cl). The most active precatalysts are PdCl2(R4-pyCH2SMe-N,S) (R = H (1a), Me (3a)); complexes of the selenium containing ligands exhibit very low activity. For closely related ligands, the changes SMe to SPh, 6-H to 6-Me, and 6-H to 6-Ph lead to lower activity, consistent with involvement of both the pyridyl and chalcogen donors in reactions involving aryl bromides. The precatalyst PdCl2(pyCH2SMe-N,S) (1a) exhibits higher activity for the reaction of aryl chlorides in Bun4NCl at 120 °C as a solvent under non-aqueous ionic liquid (NAIL) conditions.  相似文献

We have studied internal electron transfer during the reaction of Saccharomyces cerevisiae mitochondrial cytochrome c oxidase with dioxygen. Similar absorbance changes were observed with this yeast oxidase as with the previously studied Rhodobacter sphaeroides and bovine mitochondrial oxidases, which suggests that the reaction proceeds along the same trajectory. However, notable differences were observed in rates and electron-transfer equilibrium constants of specific reaction steps, for example the ferryl (F) to oxidized (O) reaction was faster with the yeast (0.4 ms) than with the bovine oxidase (~ 1 ms) and a larger fraction CuA was oxidized with the yeast than with the bovine oxidase in the peroxy (PR) to F reaction. Furthermore, upon replacement of Glu243, located at the end of the so-called D proton pathway, by Asp the PR → F and F → O reactions were slowed by factors of ~ 3 and ~ 10, respectively, and electron transfer from CuA to heme a during the PR → F reaction was not observed. These data indicate that during reduction of dioxygen protons are transferred through the D pathway, via Glu243, to the catalytic site in the yeast mitochondrial oxidase. The kinetics of single-electron injection into the oxidized nonrelaxed state (OH → EH transition) of the aberrant ba3 cytochrome oxidase from Thermus thermophilus, noted for its lowered efficiency of proton pumping, was investigated by time-resolved optical spectroscopy. Two main phases of intraprotein electron transfer were resolved. The first component (τ ∼ 17 μs) reflects oxidation of CuA and reduction of the heme groups (low-spin heme b and high-spin heme a3 in a ratio close to 50:50). The subsequent component (τ ∼ 420 μs) includes reoxidation of both hemes by CuB. This is in significant contrast to the OH → EH transition of the aa3-type cytochrome oxidase from Paracoccus denitrificans, where the fastest phase is exclusively due to transient reduction of the low-spin heme a, without electron equilibration with the binuclear center. On the other hand, the one-electron reduction of the relaxed O state in ba3 oxidase was similar to that in aa3 oxidase and only included rapid electron transfer from CuA to the low-spin heme b. This indicates a functional difference between the relaxed O and the pulsed OH forms also in the ba3 oxidase from T. thermophilus.  相似文献

Cytochrome bd is a terminal component of the respiratory chain of Escherichia coli catalyzing reduction of molecular oxygen to water. It contains three hemes, b558, b595, and d. The detailed spectroelectrochemical redox titration and numerical modeling of the data reveal significant redox interaction between the low-spin heme b558 and high-spin heme b595, whereas the interaction between heme d and either hemes b appears to be rather weak. However, the presence of heme d itself decreases much larger interaction between the two hemes b. Fitting the titration data with a model where redox interaction between the hemes is explicitly included makes it possible to extract individual absorption spectra of all hemes. The α- and β-band reduced-minus-oxidized difference spectra agree with the data published earlier ([22] J.G. Koland, M.J. Miller, R.B. Gennis, Potentiometric analysis of the purified cytochrome d terminal oxidase complex from Escherichia coli, Biochemistry 23 (1984) 1051-1056., and [23] R.M. Lorence, J.G. Koland, R.B. Gennis, Coulometric and spectroscopic analysis of the purified cytochrome d complex of Escherichia coli: evidence for the identification of “cytochrome a1” as cytochrome b595, Biochemistry 25 (1986) 2314-2321.). The Soret band spectra show λmax = 429.5 nm, λmin ≈ 413 nm (heme b558), λmax = 439 nm, λmin ≈ 400 ± 1 nm (heme b595), and λmax = 430 nm, λmin = 405 nm (heme d). The spectral contribution of heme d to the complex Soret band is much smaller than those of either hemes b; the Soret/α (ΔA430A629) ratio for heme d is 1.6.  相似文献

The kinetics of the oxidation of fully-reduced ba3 cytochrome c oxidase from Thermus thermophilus by oxygen were followed by time-resolved optical spectroscopy and electrometry. Four catalytic intermediates were resolved during this reaction. The chemical nature and the spectral properties of three intermediates (compounds A, P and O) reproduce the general features of aa3-type oxidases. However the F intermediate in ba3 oxidase has a spectrum identical to the P state. This indicates that the proton taken up during the P → F transition does not reside in the binuclear site but is rather transferred to the covalently cross-linked tyrosine near that site. The total charge translocation associated with the F → O transition in ba3 oxidase is close to that observed during the F → O transition in the aa3 oxidases. However, the PR → F transition is characterized by significantly lower charge translocation, which probably reflects the overall lower measured pumping efficiency during multiple turnovers.  相似文献

The membrane-bound enzyme cytochrome c oxidase, the terminal member in the respiratory chain, converts oxygen into water and generates an electrochemical gradient by coupling the electron transfer to proton pumping across the membrane. Here we have investigated the dynamics of an excess proton and the surrounding protein environment near the active sites. The multi-state empirical valence bond (MS-EVB) molecular dynamics method was used to simulate the explicit dynamics of proton transfer through the critically important hydrophobic channel between Glu242 (bovine notation) and the D-propionate of heme a3 (PRDa3) for the first time. The results from these molecular dynamics simulations indicate that the PRDa3 can indeed re-orientate and dissociate from Arg438, despite the high stability of such an ion pair, and has the ability to accept protons via bound water molecules. Any large conformational change of the adjacent heme a D-propionate group is, however, sterically blocked directly by the protein. Free energy calculations of the PRDa3 side chain isomerization and the proton translocation between Glu242 and the PRDa3 site have also been performed. The results exhibit a redox state-dependent dynamical behavior and indicate that reduction of the low-spin heme a may initiate internal transfer of the pumped proton from Glu242 to the PRDa3 site.  相似文献

Three kinds of copper(II) azide complexes have been synthesised in excellent yields by reacting Cu(ClO4)2 · 6H2O with N,N-bis(2-pyridylmethyl)amine (L1); N-(2-pyridylmethyl)-N′,N′-dimethylethylenediamine (L2); and N-(2-pyridylmethyl)-N′,N′-diethylethylenediamine (L3), respectively, in the presence of slight excess of sodium azide. They are the monomeric Cu(L1)(N3)(ClO4) (1), the end-to-end diazido-bridged Cu2(L2)2(μ-1,3-N3)2(ClO4)2 (2) and the single azido-bridged (μ-1,3-) 1D chain [Cu(L3)(μ-1,3-N3)]n(ClO4)n (3). The crystal and molecular structures of these complexes have been solved. The variable temperature magnetic moments of type 2 and type 3 complexes were studied. Temperature dependent susceptibility for 2 was fitted using the Bleaney-Bowers expression which led to the parameters J = −3.43 cm−1 and R = 1 × 10−5. The magnetic data for 3 were fitted to Baker’s expression for S = 1/2 and the parameters obtained were J = 1.6 cm−1 and R = 3.2 × 10−4. Crystal data are as follows. In cytochrome c oxidase, oxido-reductions of heme a/CuA and heme a3/CuB are cooperatively linked to proton transfer at acid/base groups in the enzyme. H+/e cooperative linkage at Fea3/CuB is envisaged to be involved in proton pump mechanisms confined to the binuclear center. Models have also been proposed which involve a role in proton pumping of cooperative H+/e linkage at heme a (and CuA). Observations will be presented on: (i) proton consumption in the reduction of molecular oxygen to H2O in soluble bovine heart cytochrome c oxidase; (ii) proton release/uptake associated with anaerobic oxidation/reduction of heme a/CuA and heme a3/CuB in the soluble oxidase; (iii) H+ release in the external phase (i.e. H+ pumping) associated with the oxidative (R  O transition), reductive (O  R transition) and a full catalytic cycle (R  O  R transition) of membrane-reconstituted cytochrome c oxidase. A model is presented in which cooperative H+/e linkage at heme a/CuA and heme a3/CuB with acid/base clusters, C1 and C2 respectively, and protonmotive steps of the reduction of O2 to water are involved in proton pumping.  相似文献

N,N′-Bis[allylamino]glyoxime, N,N′-bis[anilino]glyoxime, and N,N′-bis[1,2,3,4-tetrahydro-5-naphthalenamino]glyoxime have been prepared from corresponding amines and (E,E)-dichloroglyoxime. These ligands gave orange-red compound with NiCl2 in less acidic medium (pH ∼ 5) that are bis(E,E-dioximato)nickel(II) complexes {[(E,E)-Ni(HL)2]} (1a-3a) and green compounds in acidic medium (pH ∼ 2) that are tris(E,E-dioximato)nickel(II) dichloride complexes {[(E,E)-Ni(LH2)3]Cl2} (1b-3b). The crystal structures of all complexes have been determined by X-ray diffraction on a single crystal. The study of absorption spectra of these two types of complexes shows that they may be converted to each other by addition of acids (1a-3a) or bases (1b-3b) and there is no way for the amphi form.  相似文献

A new cis-dioxomolybdenum complex MoO2(DMLA)2 (DMLA = N,N-dimethyllactamide) has been synthesized and characterized by X-ray crystallography, H NMR and IR spectroscopies and electronic structure calculations at DFT/B3LYP level. This compound (chemical formula C10H20MoO6N2) crystallizes in the orthorhombic space group P212121 with Z = 4, a = 6.9357(2) ?, b = 11.8761(4) ?, c = 17.7251(5), V = 1460.00(8) ?3 and renders a slightly distorted octahedral structure with two long Mo-O bonds (2.253(3) ? and 2.257(3) ?) trans to each of the MoO groups and with two short Mo-O bonds of 1.942(3)4 ? cis to them. The MoO bond length are 1.715(3) and 1.704(3) ?). Each lactamide ligand is bidentate; they are coordinated in their deprotonated form with the carbonyl oxygen occupying a position trans to the MoO moiety while the deprotonated hydroxyl oxygen is located cis to them. Structural characterization is complemented by DFT/B3LYP calculations.  相似文献

The powerful technique of energy diagrams has been used to analyze the mechanism for proton pumping in cytochrome c oxidase. Energy levels and barriers are derived starting out from recent kinetic experiments for the O to E transition, and are then refined using general criteria and a few additional experimental facts. Both allowed and non-allowed pathways are obtained in this way. A useful requirement is that the forward and backward rate should approach each other for the full membrane gradient. A key finding is that an electron on heme a (or the binuclear center) must have a significant lowering effect on the barrier for proton uptake, in order to prevent backflow from the pump-site to the N-side. While there is no structural gating in the present mechanism, there is thus an electronic gating provided by the electron on heme a. A quantitative analysis of the energy levels in the diagrams, leads to Prop-A of heme a3 as the most likely position for the pump-site, and the Glu278 region as the place for the transition state for proton uptake. Variations of key redox potentials and pKa values during the pumping process are derived for comparison to experiments.  相似文献

The electrochemical behavior of the S,S-bridged adducts of square planar metalladithiolene complexes was investigated by using cyclic voltammetry and electrochemical spectroscopies (visible, near-IR, and ESR). The norbornene-bridged S,S-adduct [Ni(S2C2Ph2)2(C7H8)] (2a; C7H8=norbornene) formed by [Ni(S2C2Ph2)2] (1a) and quadricyclane (Q) was dissociated by an electrochemical reduction, and anion 1a and norbornadiene (NBD) were formed. Q was isomerized to NBD in the overall reaction. The o-xylyl-bridged S,S-adduct [Ni(S2C2Ph2)2(CH2)2(C6H4)] (3a; (CH2)2(C6H4)=o-xylyl) was also dissociated by an electrochemical reduction, and this reaction gave the o-xylyl radical (o-quinodimethane). The reduction of complex 3a in the presence of excess o-xylylene dibromide underwent the catalytic formation of o-quinodimethane. The butylene-bridged S,S-adduct [Ni(S2C2Ph2)2(CH2)4] (4a; (CH2)4=butylene) was stable on an electrochemical reduction. The lifetimes of reduced species of these adducts 2a-4a were influenced by the stability of the eliminated group (stability: NBD > o-xylyl radical (o-quinodimethane) > butylene radical). Therefore, the reduced species are stable in the sequence 4a > 3a > 2a. Although the palladium complex [Pd(S2C2Ph2)2] (1b) was easier to reduce than the nickel complex 1a or the platinum complex [Pt(S2C2Ph2)2] (1c), their S,S-adducts were easier to reduce in the order of Ni adduct > Pd adduct > Pt adduct.  相似文献

In [PtX(PPh3)3]+ complexes (X = F, Cl, Br, I, AcO, NO3, NO2, H, Me) the mutual cis and trans influences of the PPh3 groups can be considered constants in the first place, therefore the one bond Pt-P coupling constants of P(cis) and P(trans) reflect the cis and trans influences of X. The compounds [PtBr(PPh3)3](BF4) (2), [PtI(PPh3)3](BF4) (3), [Pt(AcO)(PPh3)3](BF4) (4), [Pt(NO3)(PPh3)3](BF4) (5), and the two isomers [Pt(NO2-O)(PPh3)3](BF4) (6a) and [Pt(NO2-N)(PPh3)3](BF4) (6b) have been newly synthesised and the crystal structures of 2 and 4·CH2Cl2·0.25C3H6O have been determined. From the 1JPtP values of all compounds we have deduced the series: I > Br > Cl > NO3 > ONO > F > AcO > NO2 > H > Me (cis influence) and Me > H > NO2 > AcO > I > ONO > Br > Cl > F > NO3 (trans influence). These sequences are like those obtained for the (neutral) cis- and trans-[PtClX(PPh3)2] derivatives, showing that there is no dependence on the charge of the complexes. On the contrary, the weights of both influences, relative to those of X = Cl, were found to depend on the charge and nature of the complex.  相似文献

A novel cytochrome ba complex was isolated from aerobically grown cells of the thermoacidophilic archaeon Acidianus ambivalens. The complex was purified with two subunits, which are encoded by the cbsA and soxN genes. These genes are part of the pentacistronic cbsAB-soxLN-odsN locus. The spectroscopic characterization revealed the presence of three low-spin hemes, two of the b and one of the as-type with reduction potentials of + 200, + 400 and + 160 mV, respectively. The SoxN protein is proposed to harbor the heme b of lower reduction potential and the heme as, and CbsA the other heme b. The soxL gene encodes a Rieske protein, which was expressed in E. coli; its reduction potential was determined to be + 320 mV. Topology predictions showed that SoxN, CbsB and CbsA should contain 12, 9 and one transmembrane α-helices, respectively, with SoxN having a predicted fold very similar to those of the cytochromes b in bc1 complexes. The presence of two quinol binding motifs was also predicted in SoxN. Based on these findings, we propose that the A. ambivalens cytochrome ba complex is analogous to the bc1 complexes of bacteria and mitochondria, however with distinct subunits and heme types.  相似文献

Copper(II)-zinc(II) bimetallic imidazolate metal-organic framework compounds of composition CuaZnbIm2(a + b) (Im = C3H3N2), including Cu2ZnIm6 (1), were prepared in high yields from the metal oxides under mild aqueous conditions using a novel acid catalysis method. Mild acidic hydrothermal treatment of paramagnetic 1 (≥120 °C) gave diamagnetic Cu(I)-containing Cu2ZnIm4 (2) in high yield. The formation mechanism of 2 involves electron transfer from Im to Cu(II), with concomitant formation of the unusual cyclotriimidazole, C9H6N6. Air-stable 2, characterized by single-crystal X-ray diffraction, crystallized in the tetragonal space group , with a = b = 10.9623(3), c = 6.3231(4) Å, α = β = γ = 90°, V = 759.86(6) Å3, and Z = 1.  相似文献

We attempted to evaluate the affinity of the anionic phospholipids to cytochrome c by means of surface plasmon resonance (SPR) technique and to correlate it with the cytochrome c active site alterations and peroxidase activity. Our experiments showed a strong interdependence between the phospholipid fatty acid saturation degree, the active site structure alterations and peroxidase activity of the cytochrome c phospholipid complex. Cytochrome c peroxidase activity and Trp59 fluorescence increase in the sequence of phosphatidyl choline (PC) → phosphatidylserine (PS) → cardiolipin (CL) → phosphatidic acid (PA). The association constant (Ka) increased in the sequence PC → PA → PS → CL. The SPR spectroscopy data shows that Ka is independent of lipid saturation degree, but correlates with phospholipid negative charge value.  相似文献

The reaction of AgX with the diphosphazane ligand, PriN(PPh2)2 (L) gives the polymeric complexes, [Ag2(μ-X)2(μ-L)]n (X = NO31a or OSO2CF31b). Single crystal X-ray analysis of 1a reveals a novel structural motif formed by interlinking of giant 40-membered rings; the diphosphazane ligand L adopts a unique ‘Cs’ geometry. These polymeric complexes exhibit a completely reversible ring-opening polymerization-depolymerization relationship with the dinuclear and mononuclear complexes, [{Ag(μ-L)(X)}2] (X = NO32a, X = OSO2CF32b) and [Ag(κ2-L)2]X (X = NO33a, X = OSO2CF33b).  相似文献

A series of palladium complexes, trans-[1-(R)-pz3,5-Me2]2PdCl2 {R = CH2CONH(2,6-i-Pr2-C6H3) (1b) and 2-(OH)-C6H10 (2b)}, supported over N/O-functionalized pyrazole derived ligands effectively catalyzed the more challenging Suzuki-Miyaura cross-coupling of a variety of activated aryl chlorides with phenyl boronic acid in air in a mixed-aqueous medium (DMF:H2O, v/v = 9:1) in moderate to excellent yields. Besides the commonly encountered Csp2-Csp2 coupling, the 1b and 2b precatalysts also catalyzed the relatively difficult Csp2-Csp3 coupling of benzyl chloride with phenyl boronic acid. The 1b and 2b complexes were synthesized by the direct reaction of the respective N/O-functionalized pyrazolyl ligands, 1a and 2a, with (COD)PdCl2 in 62-66% yields. The stability of the pyrazole-palladium interaction in the 1b and 2b complexes has been attributed to the deeply buried Npyrazole-Pd interaction as evidenced from the density functional theory (DFT) studies.  相似文献

When the iron sulfide complexes (μ-Sx)[CpFe(CO)2]2 (x = 2, 3) are treated with O-alkyl oxalyl chlorides ROCOCOCl the complexes CpFe(CO)2SCOCO2R (1) [R = Me (a), Et (b)] are obtained. Similarly, the complexes CpFe(CO)2SeCOCO2R (2) are obtained from the analogous iron selenide (μ-Se)[CpFe(CO)2]2 reaction with the same reagents. Treatment of the iron selenide with half equivalent of oxalyl chloride produces the dimeric complex [CpFe(CO)2SeCO]2 (3). The new complexes, 1, 2 and 3, have been characterized by elemental analyses, IR and 1H NMR spectroscopy. The solid state structures of 1a, 2a, 3 and [CpFe(CO)2SCO]2 (4) were determined by an X-ray crystal structure analysis.  相似文献

Galactokinase (GALK), a member the Leloir pathway for normal galactose metabolism, catalyzes the conversion of α-d-galactose to galactose-1-phosphate. For this investigation, we studied the kinetic mechanism and pH profiles of the enzyme from Lactococcus lactis. Our results show that the mechanism for its reaction is sequential in both directions. Mutant proteins D183A and D183N are inactive (<10 000 fold), supporting the role of Asp183 as a catalytic base that deprotonates the C-1 hydroxyl group of galactose. The pH-kcat profile of the forward reaction has a pKa of 6.9 ± 0.2 that likely is due to Asp183. The pH-kcat/KGal profile of the reverse reaction further substantiates this role as it is lacking a key pKa required for a direct proton transfer mechanism. The R36A and R36N mutant proteins show over 100-fold lower activity than that for the wild-type enzyme, thus suggesting that Arg36 lowers the pKa of the C-1 hydroxyl to facilitate deprotonation.  相似文献
