Summary The effect of methanol on the ability of several strains of Aspergillus to produce citric acid from galactose has been investigated. In the absence of methanol, very little production (less than 1 g/l) was observed. In the presence of methanol (final concentration 1% v/v), however, citric acid production and yeilds were increased considerably. Strong relationships were observed between citric acid production and the activities of the enzymes 2-oxoglutarate dehydrogenase and pyruvate carboxylase in cell-free extracts. During citric acid production, in the presence of methanol, the activity of 2-oxoglutarate dehydrogenase was low and that of pyruvate carboxylase high. In the absence of methanol, where little citric acid was produced, the reverse was true. It is suggested that the presence of methanol may increase the permeability of the cell to citrate, and the cell responds to the diminished intracellular level by increasing production via repression of 2-oxoglutarate dehydrogenase.  相似文献

An experimental rat model of aluminum accumulation in the brain was developed to aid in determining neurotoxity of aluminum (Al). Al was administered orally, intravenously, and intraperitoneally, in the absence or presence of citric acid or maltol. Oral administration of Al hydroxide [Al (OH)3] or aluminum chloride (AlCl3) with citric acid for 7 wk was not found to increase brain Al levels. Similarly, a single intravenous injection of AlCl3 in the presence or absence of either citric acid or maltol did not alter brain Al levels after 48 h. Only daily intraperitoneal injections of AlCl3 (8 mg Al/kg body weight) and an equimolar amount of maltol over a 14-d period enhanced accumulation of Al in rat brain. No significant increases were observed for the experimental groups receiving intraperitoneal AlCl3 alone or with citric acid. This result suggests that the chemical form of Al strongly influences its bioavailability and that intraperitoneal administration of the Al-maltol complex appears to be useful in creating subacute model of Al accumulation in brain tissue.  相似文献

A solid state fermentation method was used to utilise pineapple, mixed fruit and maosmi waste as substrates for citric acid production using Aspergillus niger DS 1. Experiments were carried out in the presence and absence of methanol at different moisture levels. In the absence of methanol the maximum citric acid was obtained at 60% moisture level whereas in the presence of methanol the maximum citric acid was obtained at 70% moisture level. The stimulating effect of methanol was less at lower moisture level. The inhibitory effect of metal ions was also not observed and maximum citric acid yield of 51.4, 46.5 and 50% (based on sugar consumed) was obtained from pineapple, mixed fruit and maosmi residues, respectively.  相似文献

Summary A new mutant strain,Aspergillus niger GS-III, showing resistance to manganese ions inhibition of citric acid fermentation on a sugarcane molasses containing medium was induced fromAspergillus niger KCU 520, a high citric acid-yielding strain. In submerged, surface or continuous cultures in the presence of manganese ions concentration upto 1.5 ppm the mutant strain yielded citric acid about 90 KgM–3 . The citric acid yield was comparable to that obtained with the parental strain KCU 520 in the absence of manganese ions, but it was atleast 3-fold higher than that obtained by the latter in the presence of manganese ions. The mutant strain immobilized in calcium alginate beads was used in combination with surface-stabilized cultures for about 36-days in a continuous flow horizontal fermenter without any apparent loss in citric acid productivity. These results indicate that the manganese-resistant mutant is stable and may be used in the presence of sufficient manganese ions concentration (1.5 ppm) in the fermentation medium. This capability of the mutant strainA. niger GS-III has been correlated with greatly reduced levels (about one-thirds) of the NADP+ -isocitric dehydrogenase, one of the control points for citric acid accumulation.  相似文献

Leuconostoc oenos M isolated from wine did not grow in the absence of glucose and it was clearly stimulated by the presence of L-malic and citric acids in synthetic medium with different glucose concentrations. In basal medium, D-glucose and L-malic and citric acids were simultaneously consumed. L-Malic acid was metabolized at a higher rate than glucose and citric acid. When the organic acids were completely consumed only 50% of the glucose was utilized. In basal medium 1 mmol of D-lactic acid was produced per mmol of glucose consumed and the amount of ethanol formed was higher with acetate present in the medium. L-Malic acid was completely recovered as L-lactic acid, and in the presence of L-malic acid a carbon imbalance from glucose to D-lactic acid was observed. In the presence of citric acid the amount of D-lactic acid formed was directly proportional to glucose-citrate utilization and acetic acid and ethanol were produced.  相似文献

It was discovered that the addition of 10 g/l acetate to a medium containing 30 g/l sunflower oil caused a drastic increase in citric acid production by Yarrowia lipolytica UOFS Y-1701 i.e. from 0.5 g/l in the absence of acetate to 18.7 g/l in the presence of acetate. Similarly, the ratio of citric acid:isocitric acid increased significantly from 1.7:1 in the absence of acetate to 3.7:1 in the presence of acetate after 240 h of growth.  相似文献

Summary and conclusions S.paracitrovorus does not readily dissimilate citric acid in the absence of sugar but does attack citric acid relatively vigorously in the presence of small quantities of glucose or lactose. The effect of glucose and lactose in initiating the dissimilation of citric acid is catalytic.The sugars which act catalytically are themselves fermented to approximately equimolar quantities of carbon dioxide, ethyl alcohol and lactic acid. The dissimilation of a combined substrate of citrate and glucose forms, in addition, acetic acid, acetylmethylcarbinol, 2,3-butylene glycol and under certain conditions, pyruvic acid which acts as an intermediate compound. Pyruvate is dissimilated to products similar to those from a fermentation of citrate plus glucose.The reactions ofKrebs' citric acid cycle apparently do not apply to the dissimilation of citric acid byS.paracitrovorus because the fermentation of citric acid proceeds anaerobically, consumes little oxygen aerobically and is not inhibited by arsenite.Inasmuch as milk contains lactose, the fermentation of citric acid in milk byS.paracitrovorus may be catalyzed as shown in these studies.Journal paper No.J711 of the Iowa Agricultural Experiment Station, Project 451.  相似文献

Effects of cadmium on the sorption of citric acid In isolatedxylem cell walls were Investigated. 2.5 nM to 9.5 mM [1.5–14]crticacid solutions were perfused through columns of xylem cell wallmaterial, isolated from tomato plants (Lycoperslcon esculentumMill, cv. Tiny Tim). The anion exchange potential of the column was estimated byamino acid analysis as approximately 46 meq dm whereas the apparentanion exchange capacity (AEC) was estimated as 1.65±0.1810–4(citric acId units). This low AEC was attributed toa ‘zipper’ effect, a mutual screening of fixed Rand A+ charges. Pre-loading with 115Cd2+ did not affect citric acid sorption,indicating the absence of Cd-effects on the availability offixed A+ charges, and the absence of the formation of effectiveR-Cd2+ and Donnan tree space (DFS) (Cd(cit)H2]+ complexes. Simultaneous application of both citric acid and 115Cd2+,45Ca2+or 28Mg2+ resufted in increased sorption of citric acid, probablydue to capacity improvement rather than changes in valence-dependentanion sorption; this may be due to the presence of bulk (M(cit)H2]+,held in the column as [M(cit)H2]+ after protonation in the DFS.Sorption of citric acid was greatest in the presence of Ca2+which was discussed in the light of the differences betweenCa, Cd and Mg in their characteristics as co-ordinative M-complexes of citric acid. The overall results indicate the potentialimportance of the presence of metal ions for the xylem transportbehaviour of organic acids in plants. The effect of citrate on production of diacetyl and acetoin by four strains each of heterofermentative and homofermentative lactic acid bacteria capable of utilizing citrate was studied. Acetoin was quantitatively the more important compound. The heterofermentative bacteria produced no acetoin or diacetyl in the absence of citrate, and two strains produced traces of acetoin in its presence. Citrate stimulated the growth rate of the heterofermentative lactobacilli. Acidification of all heterofermentative cultures with citric acid resulted in acetoin production. Destruction of accumulated acetoin appeared to coincide with the disappearance of citrate. All homofermentative bacteria produced more acetoin and diacetyl in the presence of citrate than in its absence. Two types of pyruvate kinases were found in Fusarium oxysporum. One type (inducible) was present mainly during the early stages of growth on glucose or sucrose and displayed Michaelis-Menten kinetics with respect to phosphoenolpyruvate and adenosine diphosphate. The major type (constitutive) was present under all conditions of growth and displayed in the absence of potassium ions, a sigmoidal substrate saturation curve when phosphoenolpyruvate was used as the variable substrate. In the presence of potassium ions the saturation curve for phosphoenolpyruvate exhibits a plateau at half-maximal velocity. The effects of various metabolites on the activity of the inducible and constitutive kinases were also studied. Fructose-1,6-diphosphate, cyclic AMP, acetyl CoA, tryptophan, and phenylalanine had no effect on the activity of the enzymes. Citrate was a potent inhibitor of the constitutive pyruvate kinase activity and increased the sigmoidicity of the saturation curve for phosphoenolpyruvic acid. In the presence of K+, the bimodal plot observed in the absence of citrate gradually changed to a hyperbolic shape as the concentration of citric acid was increased. In the presence of K+ and ADP as the variable substrate citric acid converted the hyperbolic plot to a sigmoidal one. Citrate had no effect on the inducible enzyme.  相似文献

In the presence of insoluble metal oxides (industrial filter dust, zinc oxide, synthetic mixture of metal oxides), Penicillium simplicissimum developed the ability to excrete considerable amounts of citric acid (>100 mM). Parallel with the increase of citric acid concentration in the culture broth, zinc was solubilized from zinc oxide. The adsorption of filter dust onto the mycelium (the pellets formed were less than 1 mm in diameter) was required for not only the citric acid excretion but also the leaching of zinc. When the filter dust was replaced with a synthetic mixture of metal oxides or with zinc oxide in combination with trace elements, levels of adsorption and citric acid production were observed to be similar to those in experiments where industrial filter dust was used. The two most important properties of the filter dust were its heavy-metal content and its buffering capacity. These properties were simulated by adding heavy metals in soluble form (as chlorides, sulfates, or nitrates) or soluble buffers to the medium. Both heavy metals and buffers were not able to induce a citric acid efflux. As with citric acid production by Aspergillus niger, the addition of manganese lowered citric acid excretion (by 40% with metal oxide-induced citric acid efflux and by 100% with urea-induced citric acid efflux). Copper antagonized the effect of manganese. The mechanism for the bulk of citric acid excretion by P. simplicissimum, however, seemed to be different from that described for citric acid accumulation by A. niger. Because of the inefficiency of metals in solubilized form and of soluble buffers to induce a strong citric acid efflux, adsorption of an insoluble metal compound (zinc oxide) turned out to be essential. Surface phenomena possibly involving the plasma membrane H+-ATPase are thought to participate in the induction of citric acid excretion by P. simplicissimum in the presence of industrial filter dust.  相似文献

AIMS: The goal of this study was to examine the growth of Oenococcus oeni in the presence of phenolic compounds under wine conditions and to see how these compounds affect bacterial metabolism. METHODS AND RESULTS: Phenolic compounds have been added to a basal medium that simulates the composition of wine. Fifty milligrams per litre or more of phenolic compounds stimulated bacterial growth. Oenococcus oeni seemed to use citric acid and trehalose, if they were present, before glucose and fructose. Citrate was completely exhausted in three days and the yield of acetate was higher when phenolic compounds were present. CONCLUSIONS: Phenolic compounds reduced the rate of sugar consumption and enhanced citric acid consumption, increasing the yield of acetic acid. SIGNIFICANCE AND IMPACT OF THE STUDY: This study allows a better knowledge of co-metabolism of citric acid and sugars by O. oeni in the presence of phenolic compounds of wine.  相似文献

Copper and cadmium inhibited the growth as well as citric acid production (depending on the heavy metal concentrations) by citric-acid-producingAspergillus niger. Activity of citrate synthase was connected with citrate synthesis in the absence as well as in the presence of heavy metals. The activity of aconitase, and both NAD- and NADP-isocitrate dehydrogenases was strongly inhibited by copper. The contents of DNA and proteins in the cells decreased but the contents of lipids and polysaccharides increased considerably in the presence of both heavy metals.  相似文献

Anaerobic growth of Aerobacter aerogenes on citrate as a carbon source required the presence of Na(+). The growth rate increased with increasing Na(+) concentration and was optimal at 0.10 m Na(+). The requirement was specific for Na(+), which could not be replaced by K(+), NH(4) (+), Li(+), Rb(+), or Cs(+). K(+) was required for growth in the presence of Na(+), the optimal K(+) concentration being 0.15 mm. Enzyme profiles were determined on cells grown in three different media: (i) intermediate Na(+), high K(+) concentration, (ii) high Na(+), high K(+) concentration, and (c) high Na(+), low K(+) concentration. All cells contained the enzymes of the citrate fermentation pathway, namely, citritase and the Na(+)-requiring oxalacetate (OAA) decarboxylase. All of the enzymes of the citric acid cycle were present, except alpha-ketoglutarate dehydrogenase which could not be detected. The incomplete citric acid cycle was, in effect, converted into two biosynthetic pathways leading to glutamate and succinate, respectively. The specific activities of citritase and OAA decarboxylase were lowest in medium (i), and under these conditions the activity of OAA decarboxylase appeared to be limited in vivo by the availability of Na(+). Failure of A. aerogenes to grow anaerobically on citrate in the absence of Na(+) can be explained at the enzymatic level by the Na(+) requirement of the OAA decarboxylase step of the citrate fermentation pathway and by the absence of an alternate pathway of citrate catabolism.  相似文献

Abstract Immunoblotting profiles of whole or protease-K-digested organisms with homologous antisera demonstrated the presence of a characteristic ladder pattern of smooth LPS in Treponema phagedenis . Periodic acid silver staining of SDS-PAGE gels confirmed these findings. However, when heterologous or homologous serum was reacted with Treponema pallidum , no such pattern or cross-reactions were observed. The significance of apparent absence of LPS in T. pallidum is discussed.  相似文献

The response to anther culture, of six genotypes of Brussels sprouts was tested on six media which included two levels of 2,4-D and the presence or absence of silver nitrate. The presence of silver was usually beneficial, and with some genotypes had a very large effect. Increasing 2,4-D could be beneficial in the absence of silver nitrate, but was sometimes detrimental in the presence of silver. Replacing agar with highly purified agarose was not particularly beneficial. Genotype, medium and genotype × medium interactions were all significant factors, with genotype being the most important.  相似文献
