A new antagonistic strain of actinomycete, designated AP19-2, was isolated from the feces of giant pandas inhabiting the Foping National Nature Reserve in China. Cultural characteristic studies strongly suggested that this strain is a member of the genus Streptomyces. The nucleotide sequence of the 16S rRNA gene of strain AP19-2 evidenced profound similarity (97-99%) with other Streptomyces strains. Two pure active molecules were isolated from a fermentation broth of Streptomyces sp. strain AP19-2 via extraction, concentration, silica gel G column chromatography, and HPLC. The chemical structures of the two related compounds (referred to as chromomycin A2 and chromomycin A3) were established on the basis of their Infrared spectra (IR), High Resolution Electrospray Ionization Mass Spectrometry (HR-ESI-MS), and Nuclear Magnetic Resonance (NMR) data, and by comparison with published data.  相似文献

Nineteen triterpenoids including two new tirucallane ones, fisuphanins A and B, were isolated from the whole plant of Euphorbia fischeriana Steud. The new structures were elucidated on the basis of extensive spectroscopy techniques including infrared (IR), High Resolution Mass Spectrometry (HRMS), and Nuclear Magnetic Resonance (NMR). The major marine sponge phospholipids 1,2-di-(5Z,9Z)-5,9-hexacosadienoyl phosphatidylcholine (PC) and phosphatidyl-ethanolamine (PE) hardly incorporate cholesterol into their liposomal bilayers, as reported earlier. Our previous studies indicated that their synthetic short chain (C18-C24) analogs with the same double bond pattern readily incorporated cholesterol, thus demonstrating the importance of the chain length. In order to investigate the possible role of the unusual delta 5,9 diunsaturation 1,2-di-(6Z,9Z)-6,9-hexacosadienoyl phosphatidylcholine and phosphatidylethanolamine were synthesized and their thermotropic behavior studied. Both analogs shows a transition endoterm at 45 degrees C, while the natural 1,2-di-(5Z,9Z)-5,9-hexacosadienoyl PC and its PE counterpart exhibited it at 42 degrees C. A partial incorporation of cholesterol into liposomal bilayers of 1,2-di-(6Z,9Z)-6,9-hexacosadienoyl PC was observed. Our results suggest that while the chain length is the predominant factor in the interactions of these phospholipids with sterols, the double bond location may also play a contributing role.  相似文献

Two distinct electroantennographycally active (EAG-active) components, A and B, and a weakly active component C were found in a solvent extract from virgin females of the Ishigaki strain of the tussock moth, Orgyia postica (Walker). Components A, B, and C were found in the extract of the females at 4.0, 0.5, and 4.0 ng/female respectively. Components A, B, and C were identified as (6Z,9Z,11S,12S)-11,12-epoxyhenicosa-6,9-diene [(11S,12S)-1: posticlure], (6Z)-henicos-6-en-11-one (2), and (6Z,9Z)-henicosa-6,9-diene (3), respectively. Component B was absent in the extract from the Okinawa strain, in which components A and C were present at 2.0 and 1.5 ng/female respectively. (11S,12S)-1 and the racemic mixture showed attractiveness for both the Okinawa and Ishigaki strains, whereas (11R,12R)-1 did not. The addition of 2 significantly reduced the trap catches with (11S,12S)-1 on the Okinawa strain which lacked 2, while there was no significant inhibitory effect on the Ishigaki strain. The addition of 3 to (11S,12S)-1 did not significantly affect trap catches at Ishigaki or Okinawa. This confirmed that the attractant pheromone of O. postica of the Ishigaki strain is also (11S,12S)-1.  相似文献

The binary mixture of the hydrocarbons (3Z,6Z,9Z)-3,6,9-nonadecatriene and (6Z,9Z)-6,9-nonadecadiene is a sex attractant for males of Alsophila quadripunctata Esp. (Lepidoptera, Geometridae). Triene/diene ratios from 1:1 to 1:0.01 attracted males in the field, the ratios 1:1 and 1:0.7 were most effective. The triene on its own also attracted males, but in significantly lower numbers than the 1:1 mixture. In a dosage test the highest dose, 1800 g of 1:0.8 [triene:diene] mixture applied to rubber caps was most effective. This is the first report of a geometrid sex attractant with field activity containing more than one component.
Zusammenfassung Die binäre Mischung der Kohlenwasserstoffe (3Z,6Z,9Z)-3,6,9-Nonadecatrien [ZZZ3,6,9-19:H] und (6Z,9Z)-6,9-Nonadecadien [ZZ6,9-19:H] in den Verhältnissen 1:1 bis 1:0.01 erwies sich im Freiland als Sexuallockstoff für Männchen von Alsophila quadripunctata (Geometridae, Lepidoptera). Dabei besaß die 1:1-Mischung die beste Fängigkeit, das Nonadecatrien allein lockte schlechter, während das C19-Alkadien unwirksam war. Ein Dosiswirksamkeitstest ergab bei Köderbeladungen von 1.8 mg (Trien:Dien + 1:0.8) die besten Fangergebnisse. Im Verlauf der Untersuchungen wurden weitere zwölf geometrische-oder positionsisomere C19-Polyenkohlenwasserstoffe getestet, wobei keine der geprüften Verbindungen Attraktant-oder Inhibitorwirkung zeigte.

The synthesis and thermotropic properties of 1,2-di-(9Z)-9-tetracosenoylphosphatidylcholine [delta 9-PC(24:1,24:1), 1], 1,2-di-(5Z)-5-tetracosenoylphosphatidylcholine [delta 5-PC(24:1,24:1), 2], and 1,2-di-(15Z)-15- tetracosenoylphosphatidylcholine [delta 15-PC(24:1,24:1), 3] are reported. Liposomes prepared from these phospholipids differ from those of the natural sponge phospholipids, 1,2-di-(5Z,9Z)-5,9-hexacosadienoylphosphatidylcholine (4a) and the corresponding ethanolamine (4b), both of which virtually exclude cholesterol from their bilayers. The behavior of 1 and 2 is similar to that of 1,2-di-(6Z,9Z)-6,9-hexacosadienoylphosphatidylcholine (5), which exhibits a partial molecular interaction with cholesterol. In the case of 3, cholesterol appears to interact with the saturated acyl chain regions of this phospholipid in a manner similar to that of its interaction with DPPC acyl chains. This study delineates the effect of the double-bond location in long fatty acyl chains of phospholipids on their interactions with cholesterol.  相似文献

The study evaluates the impact of sewage sludge on OECD - Organization for Economic Cooperation and Development and vegetable soil phytotoxicity, measured using three test species: Lepidium sativum, Sinapis alba and Sorghum saccharatum, and total and TEQ PCDD/PCDF (toxic equivalency polychlorinated dibenzo-p-dioxins/polychlorinated dibenzofurans) soil concentration, measured using HRGC/HRMS - High Resolution Gas Chromatography/High Resolution Mass Spectrometry. It also evaluates the effect of zucchini and cucumber cultivation during 5-weeks period on mitigating these parameters. The application of 3, 9 and 18 t/ha of sewage sludge gradually increases the phytotoxicity of both OECD and vegetable soil. In the case of OECD soil, the highest roots growth inhibitions were observed for S. alba (73%, 86% and 87%, respectively) and the lowest for S. saccharatum (7%, 59% and 70%), while in vegetable soil inhibitions were averagely 25% lower. Sludge application also led to a 38% (3 t/ha), 169% (9 t/ha) and 506% (18 t/ha) increase in PCDD/PCDF concentration, and the TEQs were augmented by 15%, 159% and 251%. Both soil phytotoxicity and total and TEQ PCDD/PCDF concentrations were diminished as a result of zucchini and cucumber cultivation. The maximum reduction of soil phytotoxicity (83%) was observed as an effect of cucumber cultivation, while zucchini was 11% less effective. Zucchini, in turn, was more efficient in PCDD/PCDF removal (37% reduction), followed by cucumber (24%). Such differences were not observed in the case of TEQ reductions (68% and 66% for zucchini and cucumber cultivation, respectively).  相似文献

Biflavonoids from the leaves of Wollemia nobilis (Wollemi Pine) have been investigated and compared with data on other members of the family Araucariaceae. Based on 1H and 13C NMR as well as High Resolution Mass Spectrometry (HRMS) the structures of seven isolated biflavonoids were fully determined. Although all of them were previously characterised especially at Agathis and in lesser degree at Araucaria genera this is the first report on their occurrence at Wollemi Pine, one of the world's oldest and rarest plants.  相似文献

A method for the determination of 8-oxo-2'-deoxyguanosine and 8-oxo-2'-deoxyadenosine in DNA and urine by High Performance Liquid Chromatography (HPLC)-Tandem Mass Spectrometry is described. For the urine samples there is no sample preparation except for addition of buffer and internal standards followed by redissolvation of precipitate containing 8-oxo-2'-deoxyguanosine and a centrifugation step before the samples are injected onto the HPLC column. The detection limit for 8-oxo-2'-deoxyguanosine and 8-oxo-2'-deoxyadenosine is approximately 0.3 nM corresponding to 7.5 fmol injected. Long runs, that is, > 50 samples, can be analyzed with only minimal loss of sensitivity. The concentrations excreted into urine samples from humans are between 1 and 100 nM for 8-oxo-2'-deoxyguanosine and below 0.3 nM for 8-oxo-2'-deoxyadenosine. In calf thymus DNA levels down to about 1 oxidized guanosine and adenosine per 10(6) unmodified bases can be detected. High levels of 8-oxo-2'-deoxyguanosine were found, 30 per 10(6) 2'-deoxyguanosine, levels of 8-oxo-2'-deoxyadenosine are at or below the detection limit. These findings indicate that High Performance Liquid Chromatography-Tandem Mass Spectrometry is a highly sensitive and specific method for analysis of oxidative DNA modifications in tissue as well as for analysis of excretion of oxidized nucleotides into urine that ensures a minimum artifact formation.  相似文献

以固相萃取为预处理手段,用高效液相色谱 串联四极杆质谱联用技术,针对澳大利亚南威尔士州畜牧业废水中的丙酸睾酮等13种类固醇化合物含量建立了分析方法.采用大气压化学电离源,在正离子模式下,对色谱条件和质谱条件进行优化,其中,丙酸睾酮等7种化合物以质子化的准分子离子峰[M+H]+、另6种化合物以产生了脱去水的离子峰[M+H-H2O]+为母离子进行二级质谱扫描,以最大丰度确定定量离子对.结果表明:该方法所建立的13种化合物的9点标准曲线的线性相关范围为1~1000 ng·ml-1,在该范围内,相关系数均>0.9990;各化合物的平均回收率在83.75%~111.50%,相对标准差2.02%~14.21%;除美雌醇和雌三醇的灵敏度相对较低,检测限高于15 ng·ml-1外,其余物质的检测限均低于5 ng·ml-1;实际样品测定时,不同处理流程中各化合物的浓度均能得到较好体现,该方法能满足检测要求.  相似文献

Field trials were carried out in order to evaluate some of the factors affecting male response of the almond seed wasp, Eurytoma amygdali Enderlein, to traps baited with the alkadienes (Z,Z)-6,9-tricosadiene and (Z,Z)-6,9-pentacosadiene, the male attractant of this species. The trials were conducted in Latomia, Microthivae, Kanalia and Sykourion (Thessaly, Central Greece) during the 2004 growing season. In all regions, captures in traps baited with a 7:3 ratio of the above blend significantly and positively correlated with male emergence from infested almonds that had been placed in cages in the experimental fields. Traps baited with the 7:3 pheromone ratio were significantly more effective in capturing males than traps baited with the 1:1 ratio, supporting the hypothesis that (Z,Z)-6,9-tricosadiene is the main component of this attractant. Of four trap types tested, Rebel and Lasiotraps attracted significantly more males than Delta and Funnel traps. Height had no effect on trap efficiency; however, males were captured earlier in the day in traps placed 2.3 m above grown than in traps placed 1.3 m above grown. Most male activity was recorded between 10 and 12 h, but captures were continuously recorded during the entire monitoring interval from 6 to 17 h.  相似文献

The plum cankerworm moth, Cystidia couaggaria couaggaria (Geometridae: Ennominae), is a defoliator of Chinese plum trees (Prunus mume). The pheromone components of the female were analyzed by gas chromatography (GC) with an electro-antennographic (EAG) detector and GC coupled with mass spectrometry. The crude pheromone extract included several EAG-active components, i.e., trienyl, dienyl, and saturated hydrocarbons, with a C21-C25 straight chain. The characteristic mass spectra indicated the unsaturated hydrocarbons to be (3Z,6Z,9Z)-3,6,9-trienes and (6Z,9Z)-6,9-dienes. In the fields, mixtures of the synthetic C<21 and C<23 trienes in a ratio of 2:3 and 1:4 successfully attracted males of this diurnal species during daytime. While the male antennae responded to the C25 triene and saturated hydrocarbons, their synergistic effects were not observed on the male attraction in the fields. Addition of the C21 diene interestingly inhibited the activity of the triene mixture. Males of Cystidia truncangulata, a sympatric diurnal congener of C. c. couaggaria, showed similar EAG responses to the unsaturated hydrocarbons, but no C. truncangulata males were attracted by the lures tested for C. c. couaggaria males, indicating that the identified hydrocarbons comprised the species-specific pheromone of C. c. couaggaria females.  相似文献

随着模式植物拟南芥和水稻基因组测序相继完成, 使植物基因组学研究成功迈入到功能基因组学研究的时代。这为蛋白质组学产生及其发展奠定了坚实的基础。文章重点介绍了蛋白质组学的概念、产生背景和蛋白质组学的关键技术。蛋白质组学的关键技术包括双向电泳、高效液相色谱、蛋白芯片、质谱技术、蛋白质组学的相关数据库、定量蛋白组技术、蛋白复合体标签亲和纯化技术和酵母双杂交系统。同时对当前蛋白质组技术面临的挑战和发展前景进行了讨论。  相似文献

从大苞藤黄枝叶的混合粉碎物中分离到11个化合物,运用光谱手段分别鉴定为neobractatin(1),brasixanthone B (2),5-O-methylxanthone V1 (3),10-O-methylmacluraxanthone (4),isobractatin (5),xanthone V1(6),xerophenone A (7),xerophenone B (8),bractatin (9),macluraxanthone (10)和3-O-methylneobractatin (11).本文首次应用超高效液相色谱-质谱联用技术分离了异构体7和8并测定了其精确分子量.其中化合物6~8为首次从该植物中发现.  相似文献

The Japanese giant looper, Ascotis selenaria cretacea, is a serious defoliator of tea gardens in Japan. The females produce racemic (Z,Z)-6,9-cis-3,4-epoxynonadecadiene (epo3,Z6,Z9-19:H, main component) and (Z,Z,Z)-3,6,9-nonadecatriene (Z3,Z6,Z9-19:H, minor component). The orientation of the males to the synthetic pheromone placed in a trap was strongly disrupted by Z3,Z6,Z9-19:H or a mixture of its monoepoxy derivatives (epoxydiene mixture, EDM) impregnated in septa and placed around the trap. Based on this result, polyethylene tubes containing Z3,Z6,Z9-19:H or EDM were prepared and effect of these dispensers was examined in a field. Disruption of male orientation to synthetic pheromone traps was achieved in orchards permeated with Z3,Z6,Z9-19:H at dispenser density of 3000 and 5000 tubes ha–1 (release rate: 0.55–0.61 mg day–1 tube–1) and with EDM at every tested dose, 250–5000 tubes ha–1 (release rate: 0.25–0.39 mg day–1 tube–1). Furthermore, disruption of mating in tethered females was observed in these orchards; particularly, the mating was perfectly inhibited in the areas treated with EDM at 3000 and 5000 tubes ha–1. This is the first formulation for the mating disruption of a geometrid pest.  相似文献

对岭南药材广东紫珠(Callicarpa kwangtungensis)地上部分进行化学成分研究,得到11个萜类化合物,分别鉴定为sambucunlin A(1)、2α-羟基羽扇豆醇(2)、swinhoeic acid(3)、3β-羟基-乌苏烷-11-烯-13β,28-内酯(4)、蔷薇酸(5)、2α,3β,6β,18β,23-pentahydroxy-olean-12-en-28-oic acid(6)、rel-5-(3S,8S-dihydroxy-1R,5S-dimethyl-7-oxa-6-oxobicyclo-oct-8-yl)-3-methyl-2Z,4E-pentadienoic acid(7)、salvionoside B(8)、齐墩果酸(9)、白桦脂酸(10)和α-香树脂醇(11)。其中,化合物1~4和6~8为首次从该属植物中分离得到。在化学成分分离基础上,进一步选择脂多糖(LPS)诱导的RAW 264.7小鼠巨噬细胞炎症模型进行萜类化合物抗炎活性测试。结果表明:化合物3和9具有显著的抗炎活性,对比结构发现,三萜类化合物(3~6、9和11)抗炎活性优于倍半萜类化合物(7和...  相似文献

Top Down analysis revealed that at least fourteen genes encoding histone H2A are coexpressed in HeLa cells. Characterization of these species revealed that all except H2A.Z and H2A.F/Z were alpha-N-acetylated, H2A.O and H2A.C,D,I,N,P were the most abundant, and those exceeding approximately 10% abundance lacked post-translational modifications. This unequivocal identification of H2A forms illustrates the advantages of Top Down Mass Spectrometry and provides a global perspective of H2A regulation through the cell cycle.  相似文献

【目的】采用特征次级代谢产物生物合成的保守功能基因探针,定向分离土壤中产生特征次级代谢产物的菌种资源,借助基因转录分析为导向的培养基优化方法,获得目标次生代谢产物。【方法】首先,根据5种特征次级代谢产物保守的合成功能基因设计简并引物,定向从土壤样品中筛选、分离并纯化菌株。然后,以RT-qPCR为指导开展目标产物的发酵培养基优化;最后,对菌株进行发酵,利用多种色谱技术分离纯化目标天然产物,并结合高分辨质谱与核磁共振等技术对所获得的化合物进行结构鉴定。【结果】从土壤中筛选得到了一株AHBA合酶基因和环氧化酶基因均为阳性的链霉菌菌株(编号为CQ01819),根据转录分析优化发酵培养基,最终从该菌株分离纯化得到了含有AHBA结构单元的丝裂霉素C、聚醚类抗生素莫能霉素A和缬吲霉素。【结论】本研究通过菌株的定向分离纯化,筛选得到了产生预期抗生素的浅紫灰链霉菌菌株CQ01819;基于RT-qPCR指导的发酵培养基优化,确定了菌株的发酵条件;获得发酵粗提物后,采用多种色谱整合技术和光谱分析策略,快速分离并鉴定了目标产物。该研究为目标菌种资源的定向筛选、菌株的发酵条件的快速优化和化合物的定向分离提供了较...  相似文献

The composition of oil gland exudates from two oribatid mites, Trhypochthoniellus sp. and Trhypochthonius japonicus, was studied with reference to the related species Trhypochthoniellus crassus. Trhypochthoniellus sp. contained a mixture of seven compounds; (Z,Z)-6,9-heptadecadiene, geranial, 3-hydroxybenzene-1,2-dicarbaldehyde (γ-acaridial), neryl formate, neral, (Z)-8-heptadecene and geranyl formate in decreasing order of abundance. The profile of the components from T. japonicus consisted of two types depending on the locality of sampling with unknown reason; one possessing a mixture of eight compounds [(Z,E)-farnesal, γ-acaridial, (Z,Z)-6,9-heptadecadiene, (E,E)-farnesal, (Z)-8-heptadecene and geranial in decreasing order] together with two unknown compounds, and the other composed of the same set of compounds together with 2-hydroxy-6-methylbenzaldehyde as the most abundant component. Relative abundance among common components was consistent between the two types of T. japonicus. Profiles of components differed among three species including T. crasus. The phylogenetic relationship between Oribatida and Astigmata was discussed based on secretory compounds commonly distributed between these two suborders. Nine 5 alpha, 8 alpha-epidioxy delta 6 and delta 6,9(11) sterols were identified including the hitherto unknown (22E)-5,8-epidioxy-5 alpha, 8 alpha-stigmasta-6,9(11),22-trien-3 beta-ol 9 from the sea anenome Metridium senile based on 200 MHz 1Hnmr and mass spectral data.  相似文献
