Metabolism of 6-nitrochrysene by intestinal microflora.   总被引:3,自引:3,他引:0       下载免费PDF全文
Since bacterial nitroreduction may play a critical role in the activation of nitropolycyclic aromatic hydrocarbons, we have used batch and semicontinuous culture systems to determine the ability of intestinal microflora to metabolize the carcinogen 6-nitrochrysene (6-NC). 6-NC was metabolized by the intestinal microflora present in the semicontinuous culture system to 6-aminochrysene (6-AC), N-formyl-6-aminochrysene (6-FAC), and 6-nitrosochrysene (6-NOC). These metabolites were isolated and identified by high-performance liquid chromatography, mass spectrometry, and UV-visible spectrophotometry and compared with authentic compounds. Almost all of the 6-NC was metabolized after 10 days. Nitroreduction of 6-NC to 6-AC was rapid; the 6-AC concentration reached a maximum at 48 h. The ratio of the formation of 6-AC to 6-FAC to 6-NOC at 48 h was 93.4:6.3:0.3. Interestingly, compared with results in the semicontinuous culture system, the only metabolite detected in the batch studies was 6-AC. The rate of nitroreduction differed among human, rat, and mouse intestinal microflora, with human intestinal microflora metabolizing 6-NC to the greatest extent. Since 6-AC has been shown to be carcinogenic in mice and since nitroso derivatives of other nitropolycyclic aromatic hydrocarbons are biologically active, our results suggest that the intestinal microflora has the enzymatic capacity to generate genotoxic compounds and may play an important role in the carcinogenicity of 6-NC.  相似文献

A colony of mice that do not harbor lactobacilli in their digestive tracts but whose intestinal microflora is otherwise functionally similar to that of conventional animals was derived. Methods used to reconstitute the intestinal microflora of the mice included inoculation of the animals with cultures of specific microbes, noncultivable microbes attached to epithelial cells, and cecal contents from conventional mice treated with chloramphenicol. Twenty-six microflora-associated characteristics were monitored by using relatively simple tests to determine the microflora status of the mice.  相似文献

Effects of zinc-smelter emissions on forest soil microflora.   总被引:11,自引:0,他引:11  
Within 2 km of a zinc (Zn) smelter in Palmerton, Pennsylvania, near the Lehigh Water Gap, up to 13.5% Zn by weight has been measured in the O2 horizon of the soil, and up to 8% Zn in the A1 horizon. The total numbers of bacteria, actinomycetes, and fungi (measured by dilution plate counts) were greatly reduced in the most severely Zn-contaminated soils compared with control soils. The reduction of microbial populations may be a partial cause of the decreased rate of litter decomposition at Lehigh Gap. Growth of most bacteria from control sites was reduced by 100 to 200 muM Zn, most actinomycetes by 100 muM Zn, and most fungi by 100 to 1000 muM Zn in thin-Pablum extract agar (TPab). All the tested actinomycetes and non-spore-forming bacteria isolated from Zn-contaminated Lehigh Gap soils were Zn-tolerant, growing normally in media containing 600-2000 muM Zn. Most fungi, regardless of source, were capable of at least 50% of normal growth at 700 muM Zn. Zinc-tolerant bacteria, actinomycetes, and fungi were readily isolated from low-Zn soils, suggesting that selection for Zn tolerance may proceed rapidly. Acidophilic Mortierella species have been selectively eliminated near the smelter, apparently because of elevated soil pH. Peryronellaea glomerata (Corda) Goidanich and Coniothyrium spp. were found only in the high-Zn soils.  相似文献

A male child, maintained in a controlled environment, was monitored each month for bacteria, yeasts, and filamentous fungi recovered from the mouth, nasal passages, feces, and nine body surface sites. Three natural microbial categories became apparent. Incident microorganisms were recovered from within the isolator but did not establish permanent residence. Of the 53 incident types isolated, 20 were filamentous fungi and 4 were yeasts. Some genera, such as Fusobacterium, Lactobacillus, Neisseria, and Rothia, which were commonly found in the reference group, did not become permanent inhabitants. Transient microorganisms were repeatedly recovered but could not be demonstrated within the isolated environment at the end of the study. The loss of only a few of the 19 transient species could be associated with antimicrobial therapy. Permanent microorganisms consisted of Pencillium citrinum and 17 bacterial types, of which alpha-hemolytic streptococci, Staphylococcus edpidermidis subgroups II and V, Micrococcus groups 1 and 2, Clostridium bifermentans, and Propionibacterium acnes were recovered throughout the entire 9 years of the study. The number of CFUs recovered from each sample type was generally not unlike that from the reference group of healthy male adults. Also, the number of different aerobic species recovered from the feces was within the normal range of that of the reference group. In contrast, the number of different species recovered from all other samples was less than that commonly found in the reference group.  相似文献

The effects of Bifidobacterium longum feedings for five weeks on the fecal microflora, water contents, pH values, ammonia concentration, and beta-glucuronidase activity were investigated in five healthy human volunteers. Although numbers of major bacterial groups of the fecal microflora were not changed by the bifidobacteria feedings, a remarkably decreasing number of lecithinase-negative clostridia was observed. The percentage of lecithinase-negative clostridia and bacteroides to the total bacterial numbers isolated were decreased during the feedings and numbers of C. paraputrificum and C. innocuum were reduced. A significant reduction of fecal pH values for the last week of the feeding was observed. Ammonia concentration and beta-glucuronidase activity in the feces during the feedings were significantly lower than those before or after the feedings. The oral supplement of B. longum may be introduced to improve the fecal properties such as fecal ammonia concentration and beta-glucuronidase activity, but not the composition of fecal flora.  相似文献

Summary The microfloras in grass-legume silage of high and low quality have been investigated using small laboratory silos of special design.The development of the silage microfloras was followed by analyses at intervals by determination of the occurrence of the main groups bacteria and fungi as well as of lactic acid and proteolytic bacteria.Nearly all organisms occuring were isolated and the occurrence of the single types estimated.Physiological characteristics of the isolates such as acid production, proteolytic activity and antibioses against certain test organisms were investigated.The buffer capacity of chopped and macerated plant material before and after fermentation was determined.These studies are still in progress. Seventy-two pregnant and 88 non-pregnant women were examined to see whether the periurethral region had been colonised with group B streptococci (Streptococcus agalactiae), enterococci, and Gram-negative rods belonging to the Enterobacteriaeceae. A semi-quantitative method was used for periurethral sampling, and paired urethral swabs were also collected to compare the isolation rates of group B streptococci from the two sites and with the two sampling methods. A higher isolation rate was found with periurethral sampling. Most specimens showed no or scanty growth of Gram-negative rods. Pregnancy was often associated with heavy growth of enterococci. Sampling performed during menstruation and while oral contraceptives were being used produced high isolation rates of group B streptococci. These results seem to suggest that the periurethral area might protect against genital colonisation with group B streptococci as it does against urinary tract infection and that hormonal factors influence the carriage of these organisms.  相似文献

Protease activities in human ileal effluent and feces were compared by using a variety of native and diazotized protein substrates. In many cases the diazotized proteins had altered susceptibilities to hydrolysis compared with the native proteins. Proteolytic activity was significantly greater than (P less than 0.001) in small intestinal effluent than in feces (319 +/- 45 and 11 +/- 6 mg of azocasein hydrolyzed per h per g, respectively). Moreover, fecal proteolysis was qualitatively different in that ileal effluent did not hydrolyze the highly globular protein bovine serum albumin, whereas all fecal samples tested degraded this substrate. Inhibition experiments provided further evidence that fecal protease activity differed from that in the small intestine. Physical disruption of fecal bacteria released large quantities of proteases, indicating that the lysis of bacteria in the colon may contribute to the extracellular proteolytic activity in feces. Protease inhibition studies with washed fecal bacteria showed that they produced serine, cystine, and metalloproteases, and experiments with synthetic p-nitroanilide substrates indicated that low levels of trypsin- and chymotrypsin-like activities were associated with whole cells. An elastase-like enzyme was bound to the outer membranes of some fecal bacteria.  相似文献

The contents of the lower alimentary tract from rats fed a semisynthetic, pectin-supplemented diet showed increased nitrate reductase activity and an increase in the amount of luminal contents in the intestine and cecum. Nitrate reductase activity was associated with the insoluble fraction of the gut contents which was sedimented by centrifugation (5,100 X g,20 min) and was abolished after treating the animals with streptomycin, neomycin, and bacitracin for 7 days. The pectin-dependent increase in cecal size and microbial nitrate reduction were reversed when animals were transferred from a pectin-supplemented onto a control semisynthetic diet. Polygalacturonic acid (pectic acid) was without effect on either cecal size or cecal microbial nitrate reductase activity. The studies demonstrate that pectin influences microbial metabolism in the alimentary tract.  相似文献

Developmental changes in the activities of bacterial nitrate reductase, nitroreductase and beta-glucuronidase and their response to fermentable dietary fibre, were investigated in caecal contents from suckling mice (2-week-old) and in mice aged 4-24 weeks fed either a purified fibre-free diet or that diet supplemented with 5% (w/w) pectin. There was no apparent age-related trend common to the three enzymes studied. Nitrate reductase activity in the mice fed the fibre-free diet did not markedly alter with age. Pectin administration, however, was associated with a significant increase in nitrate reductase activity, particularly in 4-week-old mice. Nitroreductase activity exhibited an overall upward trend in mice from 2 to 12 weeks and thereafter decreased. Caecal beta-glucuronidase activity in mice increased sharply between 2 weeks and 4 weeks of age, thereafter not changing significantly until the 24th week. Pectin feeding had no consistent effect on activities either of nitroreductase or beta-glucuronidase. The changes in enzyme activities with age were not related to the concentration of bacteria in the caecum, which was highest in the 2-week-old mice. We conclude that the weaning is a period in which marked changes in caecal bacterial enzyme activities can occur.  相似文献

Изуались состав и некотоые свойства бактериальной ?лоры, выделенной с поверхности корней, из ризос?еры и из свободной ночвы в начальных ?азах роста ншепицы.. Выделевные бактерии были разделены на групиы по своим мор?ологическим свойствам, по требованиям кпитанию, по интенсивности роста и по некоторым биохимическим свойствам. Было установлено, что существуют различия между представителями бактериальной ?лоры с поверхности корпей, из ризос?еры и из свободной почвы в начальных ?азх развития пшепицы, что касается мор?ологических тиов, требований к литанию и активности роста. И что касается родов бактерий, представленных в этих трех зонах, заметно проявился э??ект ризос?еры. Сравнение свойств бактериальной ?лоры семян и почвы, в которой лшенида выращивалась, показало, что бактерии семян так же, как и бактерии почвы, могут переходит на корни, но ?изиологически более активные почвенные бактерии получают преимущество и оказывают главное влияние на соств популяции ризос?еры.  相似文献

The bacterial flora of colonic aspirates taken after oral administration of a polyethylene glycol-electrolyte lavage solution to patients undergoing colonoscopy was analyzed by standard bacteriological methods and DNA hybridization techniques. The data were compared with those for normal feces collected from the same patients the day before treatment. The results indicate that the bacterial composition of feces differs from individual to individual but is not altered by the oral intake of a polyethylene glycol-electrolyte lavage solution. This pilot study demonstrates that routine colonoscopy aspirates taken after standard colonoscopy preparation accurately reflect the microflora of feces and therefore may be used in large-scale epidemiological studies.  相似文献
